OBJETIVO: No presente trabalho foi avaliada a toxicidade do H. perforatum administrado a ratas no período de organogênese (9º ao 15º dia de gestação). MÉTODOS: Trinta ratas Wistar inseminadas foram distribuídas aleatoriamente nos grupos controle e tratado, que receberam, respectivamente, 0,5 mL de solução fisiológica e 36 mg/kg de extrato seco de Jarsin diluídos em 0,5 mL de solução fisiológica por gavagem. A toxicidade materna foi avaliada por: consumo de água e ração, peso corporal, piloereção, deambulação, diarréia e ocorrência de mortes. As ratas foram sacrificadas no 21º dia de gestação, quando foram removidos e pesados: rins, fígado e ovários. Foram calculados os índices de implantação e de reabsorção e foi verificado o número médio de fetos por rata. RESULTADOS: Não foram observados sinais clínicos de toxicidade materna e nenhuma das variáveis analisadas apresentou diferenças estatisticamente significativas entre os grupos experimentais. CONCLUSÃO: Na dose administrada e no modelo experimental utilizado, o Hypericum perforatum não apresenta manifestações tóxicas para ratas prenhas no período de organogênese.BACKGROUND: Saint John's wort (Hypericum perforatum) is a medicinal plant used in the treatment of depression and other psychiatric disorders. OBJECTIVE: In the present paper, the toxicity of H. perforatum administered to female rats during organogenesis (9th to 15th day of pregnancy) was evaluated. METHODS: Thirty inseminated Wistar rats were randomly distributed into Control and Treated groups, which received by gavage, respectively, 0.5 ml of saline and 36 mg/Kg body weight of Jarsin dried extract diluted into 0.5 ml of saline. Maternal toxicity was evaluated by means of: water and food intake, body weight, piloerection, walking activity, diarrhea and death. Animals were killed on the 21st day of pregnancy, when kidneys, liver and ovaries were weighed. Implantation and reabsorption indices were calculated, as well as the average number of fetuses per mother. RESULTS: Clinical signs of maternal toxicity were not observed and none of the variables analyzed showed statistically significant differences. CONCLUSION: At the dose administered in the experimental model used, H. perforatum does not seem to be toxic to the mother.

O presente trabalho objetivou avaliar o efeito de diferentes formas de inoculação, no meio nutritivo, de explantes foliares de Eugenia involucrata DC., uma espécie florestal nativa com diversas potencialidades econômicas. Foram avaliadas as posições abaxial, adaxial, com e sem cortes no limbo foliar. Foi utilizado o meio de cultura MS acrescido de 10 µM de ANA isolado ou das combinações, em µM, de 2,4-D e BAP: 5-5 e 5-10. A posição dos explantes afeta a calogênese e a organogênese em segmentos foliares de E. involucrata, sendo mais adequada a abaxial sem cortes na região do limbo. A associação dos reguladores de crescimento 2,4-D + BAP na concentração de 5-10 µM foi mais promissora para a obtenção de calos, especialmente os nodulares, putativos à embriogênese somática. The aim of this research was to evaluate the effect of different forms of leaf explants inoculation in nutritive medium of Eugenia involucrata DC., an native forest species with diverse economic potentials. Inoculations were evaluated at abaxial and adaxial positions, with and without cuts on the leaf surface in the MS nutritive medium supplemented with 10 µM NAA or combination, in µM, of 2.4-D and BAP: 5-5 and 5-10. The explants position inoculation affects the callus induction and organogenesis in E. involucrata leaf segments, being the abaxial the most suitable position, without cuts in the leaf surface. The combination of growth regulators 2.4-D and BAP, at concentrations of 5-10( µM) was most promising of callus obtaining, especially nodular callus, putative at somatic embryogenesis.

Umburana-de-cheiro é uma espécie nativa da Caatinga, amplamente utilizada pela população local, devido o valor madeireiro, ornamental, forrageiro e, principalmente medicinal. Considerando o significativo risco de extinção e a inexistência de estudos relacionados ao cultivo e propagação, objetivou-se com este trabalho estabelecer um protocolo para micropropagação de Amburana cearensis. Para isso, avaliou-se a influência do regulador vegetal 6-benzilaminopurina (BAP) em cinco concentrações (0,0; 2,22; 4,44; 8,88 e 17,76µM) sobre a multiplicação in vitro de diferentes explantes (segmento nodal, ápice caulinar e segmento cotiledonar) e na etapa de enraizamento testou-se diferentes concentrações de ácido indol butírico (AIB) (0,0; 2,5; 5,0 e 10,0µM) na presença e ausência de 1,0g L-1 de carvão ativado. Maior número de brotos (2,36) foi obtido na presença de 4,44µM de BAP, a partir do segmento cotiledonar e maior porcentagem de enraizamento (92,0%) ocorreu na presença de 10,0µM de AIB na ausência de carvão ativado. Não existiram dificuldades para o estabelecimento do protocolo de micropropagação de A. cearensis. Umburana de cheiro is a species native to the Caatinga, widely used by local people, because the value of timber, ornamental, forage and especially medicinal. Considering the significant risk of extinction and the ausence of studies related to the cultive and propagation, the aim of this work was to establish a protocol for micropropagation Amburana cearensis. For this, we evaluated the influence of plant growth regulator BAP at five different concentrations (0.0, 2.22, 4.44, 8.88 and 17.76mM) on the in vitro multiplication of different explants (nodal segments, apex cotyledon and stem segment) and the rooting stage we tested different concentrations of IBA (0.0, 2.5, 5.0 and 10.0mM) in the presence and absence of 1.0g L-1 of activated charcoal. Highest number of shoots (2.36) was obtained in 4.44mM of BAP from cotyledonary segment and higher rooting percentage (92.0%) ocurred in 10.0mM of IBA in the absence of activated charcoal. There were no difficulties to the establishment of micropropagation protocol A. cearensis.

Techniques that may improve the ability to propagation this rootstock are of great interest. The aim of this study was to adjust protocols for in vitro multiplication and regeneration of quince cultivars MC and Adams. The results of this word are presented in chapter. Material used in the experiments was obtained from in vitro propagation in culture medium consisting of MS salts and vitamins supplemented with myo-inositol (100 mg L-1), 6 - benzylaminopurine (BA) (0.3 mg L-1), sucrose (30 g L-1) and agar (8 g L-1). Chapter 1 we used MS medium added agar (8 g L-1) on solidified medium and vermiculite (3 g flask-1) in the liquid medium and the flasks were sealed with aluminum foil, PVC film and polypropylene cap. In Chapter 2 how carbon source in culture medium was added sucrose, fructose or sorbitol at concentrations 0, 15, 30, 45 and 60 g L-1. For in vitro regeneration the Chapter 3 was divided in two experiments. In experiment 1 the culture medium consisted of salts and vitamins MS and SH supplemented with myoinositol (100 mg L-1), sucrose (30 g L-1), agar (8 g L-1), NAA (2 mM) and TDZ at concentrations 0, 1.5, 3, 4.5 and 6 μM. In experiment 2 the culture medium consisted of SH salts and vitamins supplemented with myo-inositol (100 mg L-1), sucrose (30 g L-1), agar (8 g L-1), TDZ (0 , 1.5, 3, 4.5 and 6 μM) combined with NAA or IBA at concentrations of 0, 1.0, 1.5 and 2 μM. All experiments were evaluated after 60 days. Considering the results presented in Chapter 1 is possible to conclude that the use of culture medium solidified with agar and the flasks sealed with aluminum foil or polypropylene cap favored the in vitro multiplication of quince 'MC' and 'Adams'. In the second chapter it was found that the sucrose concentration of 45 g L-1 to cultivar MC and 30 g L-1 to cultivar Adams are the best concentrations and carbon source for in vitro multiplication quince. In the Chapter 3 first verified in experiment 1 that the in vitro regeneration of adventitious shoots quince cultivars MC and Adams was favored by the use of SH culture medium and addition of 4.5 μM TDZ to cultivar MC and 6 μM TDZ for cultivar Adams. And in experiment 2 it was concluded that multiple shoots were formed from the combination of 4.5 μM TDZ and 2 μM NAA for cultivar MC and 6 μM TDZ and 2 μM NAA to cultivar Adams. Pomares de pereira com plantio em alta densidade foram possíveis devido a utilização de portaenxerto de marmeleiro (Cydonia oblonga Mill.), obtendo-se assim plantas de pequeno porte e rápida frutificação, além de proporcionar uniformidade a esses pomares. Técnicas que venham melhorar a capacidade de propagação deste portaenxerto são de grande interesse. O objetivo desse trabalho foi ajustar protocolos de multiplicação e regeneração in vitro de marmeleiro cultivares MC e Adams. Os resultados do trabalho estão apresentados na forma de capítulos. O material vegetal utilizado nos experimentos foi obtido a partir da multiplicação in vitro em meio de cultura básico composto dos sais e vitaminas MS, suplementados com mio-inositol (100 mg L-1), 6- benzilaminopurina (BAP) (0,3 mg L-1), sacarose (30 g L- 1) e ágar (8 g L-1). No capítulo 1, utilizou-se meio básico acrescentado de ágar (8 g L- 1) no meio solidificado e vermiculita (3 g frasco-1) no meio líquido e os frascos foram vedados com papel alumínio, filme PVC e tampa de polipropileno. No capítulo 2, como fonte de carbono no meio de cultura foi utilizado sacarose, frutose ou sorbitol nas concentrações de 0, 15, 30, 45 e 60 g L-1. Para regeneração in vitro o capítulo 3 foi dividido em dois experimentos. No experimento 1 os meios de cultura constituíram-se dos sais e vitaminas MS e SH, suplementados com mio-inositol (100 mg L-1), sacarose (30 g L-1), ágar (8 g L-1), ANA (2 μM) e TDZ nas concentrações de 0, 1,5, 3, 4,5 e 6 μM. No experimento 2 o meio de cultura constituiu-se dos sais e vitaminas SH, suplementado com mio-inositol (100 mg L-1), sacarose (30 g L-1), ágar (8 g L-1), TDZ (0, 1,5, 3, 4,5 e 6 μM) combinado com ANA ou AIB nas concentrações de 0; 1,0; 1,5 e 2 μM. Todos os experimentos foram avaliados após 60 dias. Diante dos resultados apresentados no capítulo 1 é possível concluir que a utilização de meio de cultura solidificado com ágar e os frascos vedados com papel alumínio ou tampa de polipropileno favorecem a multiplicação in vitro de marmeleiro 'MC' e 'Adams'. No capítulo 2 constatou-se que a sacarose na concentração de 45 g L-1 para cultivar MC e 30 g L-1 para cultivar Adams são as melhores concentrações e fonte de carbono para multiplicação in vitro dos portaenxertos de marmeleiro testados. No capítulo 3, primeiro verificou-se no experimento 1 que a regeneração in vitro de brotos adventícios de marmeleiro das cultivares MC e Adams foi favorecida pelo uso do meio de cultura SH e pela adição de 4,5 μM de TDZ para cultivar MC e 6 μM de TDZ para a cultivar Adams. No experimento 2 concluiu-se que houve maior taxa de regeneração a partir da combinação de 4,5 μM de TDZ e 2 μM de ANA para cultivar MC e 6 μM de TDZ e 2 μM de ANA para cultivar Adams.

Garlic cultivars are sexually sterile under standard growth conditions, with direct implications for commercial production costs as well as breeding programs. Garlic is propagated commercially via bulblets, which facilitates disease transmission and virus load accumulation over vegetative generations. Tissue culture produces virus-free clones that are more productive, while keeping the desired traits of the cultivar. Consequently, this technique allows studies of garlic genetics as well as guarantees genetic conservation of varieties. We aimed at analyzing the in vitro regeneration of eight marketable cultivars of garlic using root segments as explants. For each genotype, bulblet-derived explants were isolated and introduced into MS medium supplemented with 2,4-D and 2-iP. Calli were transferred to MS medium supplemented with 8.8 mM BAP and 0.1 mM NAA (regeneration medium A), or with 4.6 mM kinetin alone (regeneration medium B). The calli were then evaluated for regeneration frequency after sixty days of in vitro cultivation. The noble cultivar 'Jonas' presented the highest rates of plant regeneration among the cultivars tested. The medium A, which contained auxin and cytokinin, induced the highest regeneration rates of all cultivars. The process described herein is simple, reproducible and can potentially be used as a tool in molecular breeding strategies for other marketable cultivars and genotypes of garlic. Cultivares de alho são sexualmente estéreis sob condições padrão de cultivo, com implicações diretas nos custos de produção comercial, bem como em programas de melhoramento. O alho é comercialmente propagado por meio de bulbilhos, o que facilita a transmissão de doenças e leva ao acúmulo de cargas virais ao longo das gerações. A cultura de tecidos produz clones livres de vírus que são mais produtivos, mantendo as características desejadas da cultivar. Consequentemente, esta técnica permite estudar a genética do alho, bem como garantir a conservação genética das variedades. Nosso objetivo foi analisar a regeneração in vitro de oito cultivares comerciais de alho usando segmentos de raiz como explante. Para cada genótipo, explantes derivados de bulbilho foram isolados e introduzidos em meio MS suplementado com 2,4-D e 2-iP. Calos foram transferidos para meio MS suplementado com 8,8 mM BAP e 0,1 mM NAA (meio de regeneração A), ou com 4,6 mM cinetina somente (meio de regeneração B). Os calos foram avaliados quanto à frequência de regeneração após sessenta dias de cultivo in vitro. A cultivar nobre 'Jonas' apresentou as maiores taxas de regeneração entre as cultivares testadas. O meio de cultura A, o qual continha auxina e citocinina, induziu as maiores taxas de regeneração em todas as cultivares. O processo aqui descrito é simples, reprodutível e pode ser usado potencialmente como uma ferramenta em estratégias de melhoramento para outras cultivares comerciais e genótipos de alho.

Dissertação de mestrado em Biotecnologia Vegetal, apresentada ao Departamento Ciências da Vida da Faculdade de Ciências e Tecnologia da Universidade de Coimbra. A embriogénese somática é um processo que permite a rápida propagação de espécies economicamente importantes e uma poderosa ferramenta na transformação genética. Uma das fases mais importantes neste processo é a maturação dos embriões somáticos, visto que durante esta fase é comum ocorrer um número significativo de embriões anómalos, o que pode ter implicações na sua conversão em plântulas. A indução da embriogénese somática e o desenvolvimento de embriões de tamarilho (Cyphomandra betacea) foram estudados no Laboratório de Biotecnologia da FCTUC e usados com sucesso na regeneração das plantas a partir de vários tipos de explantes. É um sistema de duas fases no qual a embriogénese somática é induzida em meio MS, na presença de uma auxina e com concentrações elevadas de sacarose, seguindo-se a transferência do tecido embriogénico obtido para um meio sem auxina e com níveis reduzidos de sacarose, de modo a obter embriões somáticos. Trabalhos anteriores demonstraram que optimizando as condições de maturação há um maior rendimento e uma melhor qualidade dos embriões formados. Neste trabalho, pretendeu-se optimizar a maturação de embriões somáticos de tamarilho, testando diferentes tipos de calos embriogénicos em diversas condições de maturação. Calos com diferentes idades, origens e qualidades embriogénicas foram avaliadas, variando a composição do meio de cultura, em particular pela adição de carvão activado, várias concentrações de sacarose e de outros agentes osmóticos para além da sacarose. A manifestação de diferentes capacidades embriogénicas por parte das linhas estudadas perante os tratamentos aplicados, confirmou assim a importância da origem e da idade dos explantes na obtenção de embriões somáticos maduros. O calo induzido a partir de material micropropagado de uma árvore adulta (TJ-CE3) revelou uma ausência de competência embriogénica, que poderá ser justificada pelos baixos níveis endógenos de peróxido de hidrogénio nesta linha de calo (cerca de 6 μmol/l), quando comparados aos detectados em calos embriogénicos (cerca de 12 μmol/l) induzidos na mesma altura, mas a partir de material com origem seminal (linha TV-CE2). De um modo geral, foi a linha TVCE1 a que demonstrou uma resposta melhor, em termos do número de embriões formados, sendo o ABA (2 mg/l, TMA2) o composto mais eficaz, neste tipo de calo, promovendo o desenvolvimento de 204 embriões/100 mg de calo. No caso do TV-CE2, uma linha de calo mais recentemente estabelecida, contrariamente ao esperado, este apresentou respostas inferiores ao TV-CE1, sendo os melhores resultados obtidos para o meio TM2 (50,3 xi embriões somáticos/100 mg de calo). O controlo da maturação e dos processos de conversão através da análise histológica permitiram observar os efeitos das condições de cultura no desenvolvimento dos embriões, em particular no padrão apical-basal e na organização de meristema apical. Observou-se que, mesmo em embriões anómalos o meristema apical se organizou de acordo com o padrão normal, sugerindo que este tipo de embriões, teria potencial para desenvolver de uma forma normal e atingir a conversão em plântulas. Para além dos ensaios de maturação realizados foram testados factores influentes na indução de embriogénese somática como o genótipo, a dimensão do explante e as condições do meio, mais concretamente a sua composição química. Nestes ensaios observaram-se disparidades nas taxas de indução observadas para os 3 genótipos testados e na quantidade de calo observado. Foi o genótipo de tamarilho vermelho que apresentou as taxas de indução mais altas, a rondar os 68% de explantes com tecido embriogénico. A fragmentação dos explantes foliares em secções com 3-10 mm revelou-se uma técnica eficaz, permitindo a formação de quantidades superiores de calo embriogénico. Os ensaios de indução de organogénese in vitro realizados, recorrendo a material vegetal estabelecido in vitro (linha TJ), proveniente de uma árvore adulta, demonstraram, à semelhança do que se observou na maturação de embriões somáticos, uma baixa capacidade organogénica desta linha, visto que das gemas isoladas apenas 13,3% geraram rebentos caulinares Somatic embryogenesis is a process that allows the fast propagation of economical important species and a powerful tool in breeding and genetic transformation. One of the most critical phases in this process is the embryo maturation, because during this stage it is usual to find a significative number of anomalous embryos, what may have implications in their conversion in emblings. Somatic embryogenesis induction and somatic embryo development of the Solanaceous tamarillo tree (Cyphomandra betacea) have been studied at the Biotechnology Laboratoty of FCTUC, and successfully used on the regeneration of these plants from several types of explants. It is a two-step system in which somatic embryogenesis is induced in MS medium containing an auxin and high sucrose concentrations, and the embryogenic tissues obtained must be transferred to an auxin-free medium, with reduced sucrose levels, in order to obtain somatic embryos. Previous work has demonstrated that, by optimizing maturation conditions, a higher yield and a better quality of the formed embryos can be achieved. In this work, it was intended to optimize the maturation of somatic embryos of tamarilho by testing different types of embryogenic callus in different maturation conditions. Callus at different ages, embryogenic origin and quality were assessed by varying the composition of culture medium, in particular by addition of activated charcoal, various concentrations of sucrose and other osmotic agents other than sucrose. The expression of different capacities by the embryogenic lines studied before the treatments applied, thus confirming the importance of the origin and age of the explants to obtain mature somatic embryos. The callus induced from micropropagated material of a mature tree (TJ-CE3) revealed an absence of embryogenic competence, which can be explained by the endogenous low levels of hydrogen peroxide in this callus line (about 6 μmol / l), where compared to those detected in embryogenic callus (about 12 μmol / l) induced at the same time but from seminal material source (line TV-CE2). In general, it was line TV-CE1 that showed a better response in terms of the number of embryos formed being ABA (2 mg / l, TMA2) the most effective compound in this type of callus, promoting the development of 204 embryos/ 100 mg callus. In the case of line TV-CE2, a callus more recently established, contrary to expectations, it showed lower responses to TV-CE1, the best results being obtained for the middle TM2 (50.3 somatic embryos /100 mg callus). Control of the maturation and conversion processes by histological analysis allowed observe the effects of culture conditions on embryo xiii development, in particular in the apical-basal pattern and in the apical meristem organization. It was observed that, even in the abnormal embryos apical meristem is organized according to normal, suggesting that this type of embryos would have the potential to develop normally and achieve conversion to plantlets. In addition to the maturation tests performed were evaluated factors that influence the induction of somatic embryogenesis as the genotype, the size of the explant and the environment conditions, more specifically to their chemical composition. In these tests it was observed differences in the rate of induction observed for the 3 genotypes and the amount of callus observed. Red tamarilho genotype showed higher induction rates, hovering around 68% of explants with embryogenic tissue. Fragmentation of leaf explants into sections of 3-10 mm proved to be an effective technique, allowing the formation of higher amounts of embryogenic callus. The induction of organogenesis assays performed in vitro, using established plant material in vitro (TJ line) from a mature tree, shown, similar to that observed in the maturation of somatic embryos, low organogenic capacity of this line, since that of gems isolated only 13.3% generated stem shoots

Bambara groundnut (Vigna subterranea [L.] Verdc.) is mainly grown for human consumption. However, several factors limit a wider adoption of the crop including the presence of antinutritional factors in the seeds that lower product quality and protein availability but also the plant susceptibility to pests and diseases. Tissue culture techniques are very scanty in Bambara groundnut and should be developed before carrying out genetic transformation for the crop improvement. Therefore, here, an efficient system for in vitro shoot induction from cotyledons derived from mature seeds has been established. Different types and concentrations of plant growth regulators were used to induce buds in embryo-free cotyledon explants. Cotyledons were cut transversally or longitudinally into three segments: proximal, middle and distal part. The influence of explant orientation on the medium, the type of segment and landrace has then been studied. Benzylaminopurine (3 mg.l(-1)) alone or combined with a-naphthaleneacetic acid (0.05 mg.l(-1)) induced multiple shoot formations. The organogenic potential was restricted to the proximal segment of cotyledons. Frequency of bud induction (30%) and average number of buds per explant (12) were higher when the adaxial side of the proximal segment was in contact with the medium. Shoot regeneration from cotyledon explants of ten Bambara landraces revealed that the response is genotype-dependent with varieties Ci6, Ci2, Ci4 and Ci15 exhibiting 20 to 30% shoot regeneration and six to ten buds per explant. Regenerated shoot buds excised from explants were elongated and rooted on MS basal medium devoid of plant growth regulators. All rooted plantlets survived to the transfer on a sand soil mixture, and morphologically normal plants were hardened and transferred to greenhouse for further growth to maturity and seed set.

The physic nut (Jatropha curcas L.) is considered a potential source for the biofuel production, because of its high productivity and oil quality extracted from the seeds. However, its use is unfeasible for human and animal food due to antinutritional factors like the curcin protein and specially the secondary metabolites – phorbol esters. The application of biotechnological tools, like tissue culture, can be used to overcome these limitations. In this context, the aim of this work was to propagate whole plants in vitro using shoot cultures obtained from physic nut seeds germinated in vitro and in vivo; to determine the reproducibility of a regeneration protocol available in the literature, and to define through histological studies, the origin of the regenerated plant in vitro. For the in vitro propagation, shoots were used as explants sources, which were placed on MS medium supplemented with the cytokinins BAP (6- benzylaminopurine), KIN (kinetin) e 2-iP (2- isopentenyl adenine), 2,0 mg.L-1. BAP showed the best results in the development of the shoots. This regulator was used alone or in association with gyberelins for the elongation of the shoots. The indole-3-butyric-acid was used for the rooting. The supplementation of the medium culture with BAP was efficient in the development of the shoots and the elongation was superior with the use of GA3 alone. The rooting was achieved with AIB. To determine the reproducibility of an organogenesis protocol, cotyledons from physic nut seeds germinated in vitro were placed on MS medium supplemented with BAP and AIB for the promotion of aerial which were elongated with BAP and rooting with AIB. The cotyledonary explants were placed with the abaxial and adaxial face in contact with the medium to establish the best position of it in the in vitro regeneration. Whole plants of J. curcas L. were obtained using the protocol tested. The best position of the cotyledonar segment was the adaxial face in contact with the medium, where 56% of the explants formed shoots. The histological analysis was made with cotyledonary explants collected in sequential intervals of 0, 5, 10, 15 e 25 days of permanency in the regeneration medium. The anatomical study allowed the accompaniment of the organogenesis in physic nut, where parenchymal cells next to the adaxial region of the cotyledonar explants were divided and formed the shoots. The data obtained from this work showed that both protocols used, from shoots tips and cotyledonar explants, were efficient in the regeneration process; the histological analysis suggests that regeneration occurs via direct organogenesis and possibly presents multicellular origin. O pinhão manso (Jatropha curcas L.) é considerado uma fonte potencial para a produção de biocombustíveis, tendo por base a alta produtividade e qualidade do óleo extraído de suas sementes. Entretanto, seu uso é inviabilizado para alimentação humana e animal devido à presença de fatores antinuticionais como a curcina e, em especial, os ésteres de forbol. A utilização de ferramentas biotecnológicas, como a cultura de tecidos, pode vir a minimizar ou solucionar essas limitações. Nesse contexto, o objetivo desse trabalho foi propagar in vitro plantas inteiras via cultivo de ápices caulinares obtidos a partir de sementes de pinhão manso germinadas in vivo e in vitro; determinar a reprodutibilidade de um protocolo de regeneração disponível na literatura e definir, através de estudos histológicos, a origem da planta regenerada in vitro. Para a propagação in vitro utilizou-se ápices caulinares, como explantes, que foram colocados em meio MS suplementado com as citocininas BAP (6-benzilaminoprina), KIN (cinetina) e 2-iP (2-isopenteniladenina) na concentração de 2,0 mg.L-1. O BAP apresentou os melhores resultados no desenvolvimento dos ápices caulinares. Este e a giberelina foram utilizados juntos no mesmo meio ou separados, para a indução do alongamento dos mesmos. O ácido indol-3-butiríco (AIB) foi usado para o enraizamento. A suplementação do meio de cultura com BAP foi eficiente no desenvolvimento dos ápices caulinares e o alongamento apresentou êxito com o uso do GA3 individualmente. O enraizamento com o AIB foi conseguido. A fim de determinar a reprodutibilidade do protocolo de organogênese, cotilédones de sementes de pinhão manso germinadas in vitro foram colocados em meio MS suplementado com BAP e AIB para a indução da formação de partes aéreas, estas foram alongadas com BAP e enraizadas com AIB. Além disso, os explantes cotiledonares foram colocados com a face abaxial e adaxial em contato com o meio de cultivo a fim de estabelecer a melhor posição do mesmo na regeneração in vitro. Plantas inteiras de J. curcas L. foram obtidas a partir da utilização do protocolo testado. A melhor posição do segmento cotiledonar foi a face adaxial em contato com o meio, onde 56% dos explantes formaram partes aéreas. A análise histológica foi feita com a coleta de explantes cotiledonares em intervalos sequenciais de 0, 5, 10, 15 e 25 dias de permanência do meio de regeneração. O estudo anatômico possibilitou o acompanhamento da organogênese do pinhão manso, no qual células parenquimáticas próximas à região adaxial do explante cotiledonar se dividiram e formaram as partes aéreas. Os dados obtidos nesse trabalho mostraram que ambos os protocolos utilizados, a partir de ápices caulinares e de explantes cotiledonares, foram eficientes no processo de regeneração; as análises histológicas sugerem que a regeneração ocorre via organogênese direta e possivelmente apresenta origem multicelular.

International audience; Psychosomatic disorders are classically related to the body outside the brain. Yet, as the Freudian concept of actual neuroses lets us imply, there is no reason why the circuits of the brain cannot be affected by a psychosomatic process. For Lacan the psychosomatic phenomena are related to the creation of a conditioned reaction of the organism through signifiers, which having frozen and therefore lost their signifying function are transformed into signs, which coming from the Other, obtain an imperative quality for the organism, thus disturbing its function. Peirce's phaneroscopy helps us conceptualise this procedure as a semiotic reduction which proceeds from the thirdness of the signifier to the secondness of the signal (which can bring about conditioned reflexes or reactions of a different kind) or even go further to the firstness of complete automation. Yet, beyond the typical psychosomatic phenomena this process of semiotic reduction could also apply to mood disorders (depression, bipolar disorder, delirious mood, athymhormia, panic disorder and so on) and other clinical conditions likewise. According to Post this clinical phenomenon of the tendency towards automation is a result of the kindling neurophysiological mechanism which could explain passing from mood disorders of reactional type to more automatic mood states. Yield also presents large inter-month variability as well as variability among trees in the field. To simulate this complex system we developed a plant growth model using i) plant source-sink relationships as drivers of plant phenotypic plasticity and ii) stochastic rules relating inflorescence characteristics to plant nutritional status.

Mutationen in Genen, die für Kernhüllproteine codieren sind mit einer stetig zunehmenden Anzahl menschlicher Erkrankungen verbunden, die als Envelopathien bezeichnet werden. Erstaunlicherweise betrifft die Pathologie dieser Krankheiten spezifische Gewebe und Organe, obwohl entsprechende Proteine meist ubiquitär exprimiert werden. So führen beispielsweise Defekte in Emerin, einem Protein der inneren Kernhülle, zur X-chromosomalen Emery- Dreifuss Muskeldystrophie (EDMD). Diese Krankheit ist durch Muskelschwäche oder – schwund gekennzeichnet. Defekte im Kernhüllprotein MAN1 sind dagegen mit Krankheiten verbunden, die Knochen- und Hautgewebe betreffen. Interessanterweise besitzen beide Proteine eine evolutionär hoch konservierte Domäne, die sog. LEM-Domäne. LEM-Domänen Proteine können mit der Kernlamina interagieren, ebenso mit dem sog. Barrier-to- Autointegration Factor (BAF) sowie mit zahlreichen Transkriptionsfaktoren. Dennoch ist die funktionelle Rolle der LEM-Domänen Proteine bis dato nicht vollständig aufgeklärt. In der vorliegenden Studie sollten daher die Funktionen von MAN1 und Emerin während der Frühentwicklung von Xenopus laevis untersucht werden. Vorangehende Untersuchungen zeigten, dass Mikroinjektionen von XMAN1- Antikörpern in Zwei-Zell-Stadien befruchteter Eizellen zu einem Arrest der Zellteilung in der injizierten Blastomere führten. Da dabei eine Störung der Kernhüllbildung spekuliert wurde, sollte durch Antikörper-vermittelter Inhibition von XMAN1 die Bildung von in vitro Kernen im Xenopus Eiextrakt untersucht werden. Dabei wurden Kerne beobachtet, die dekondensiertes Chromatin zeigten, bei denen jedoch eine Fusion von Membranvesikeln zu einer durchgehenden Kernhülle nicht stattgefunden hatte. Frühere Charakterisierungen von MAN1 und Emerin zeigten unterschiedliche Expressionsmuster während der Entwicklung von X. laevis. Da XMAN1 ubiquitär exprimiert und Xemerin jedoch erstmals ab Stadium 41 nachweisbar ist, war es mittels Mikroinjektion von Xemerin möglich zu zeigen, dass es in der Lage ist den Arrest der Zellteilung zu verhindern. Es wurde daher die These aufgestellt, dass MAN1 und Emerin während der Frühentwicklung von Xenopus überlappende Funktionen besitzen. Um diese These zu prüfen, wurde zunächst unter Verwendung des Proximity Ligation Assays untersucht, ob beide Proteine miteinander interagieren können. Mit Hilfe dieser Methode konnte gezeigt werden, dass Interaktionen beider Proteine innerhalb der Kernhülle lokalisieren. The objectives of this study were to establish appropriate conditions for obtaining plant regeneration and acclimatization of the 'RB92579' and 'RB93509' sugarcane cultivars and to elucidate the shoots origin through histological analysis. For both cultivars, obtaining shoots showed better results with the culture of explants on a callus induction medium containing 2.0mg L-1 2,4-dichlorophenoxyacetic acid, followed by cultivation on a shoot induction medium containing 0.1mg L-1 kinetin and 0.2mg L-1 benzilaminopurine. The MS medium without growth regulators proved to be appropriate for elongation and rooting of shoots and the use of the composed substrate of vermiculite + MS salts was effective for acclimatization. Histological analysis revealed that the origin of the shoots in both cultivars occurred through indirect organogenesis. Os objetivos deste estudo foram estabelecer condições apropriadas para a obtenção de regeneração de plantas e aclimatização das cultivares 'RB92579' e 'RB93509' de cana-de-açúcar e elucidar a origem das brotações através da análise histológica. Em ambas as cultivares, as brotações obtidas apresentaram os melhores resultados com a cultura dos explantes em um meio de indução de calo contendo 2,0mg L-1 de ácido 2,4-diclorofenoxiacético, seguido do cultivo em meio indutor de brotações, contendo 0,1mg L-1 de cinetina e 0,2mg L-1 benzilaminopurina. O meio MS sem reguladores de crescimento foi adequado para o alongamento e enraizamento das brotações e da utilização do substrato composto de vermiculite + sais do meio MS, foi eficaz para a aclimatização. A análise histológica revelou que a origem das brotações em ambas as cultivares ocorreu via organogênese indireta.

Recent years have seen a growth in the Brazilian production of strawberries, however, the participation of Brazil in world production is still low.Aiming to increase production and improve the quality of fruits produced alternatives have been sought, such as better adapted cultivars to the production regions and best seedlings.Considering that the plant tissue culture can be a technique for improvements in this production system, the work was developed aiming to assess the conditions of seed germination of strawberry cv. Camiño Real and to establish protocols for regeneration through organogenesis, to the genetic lineages derived from cultivars Camiño Real and Festival.To assess the conditions of germination, it was used fresh seeds and stored for 2 and 12 months. These were submitted to different treatments of break dormancy and the presence or absence of light during germination. Based on the evaluations of germination rate and fresh weight of the plants obtained, it was concluded that the seed storage significantly reduces the viability thereof, for fresh seeds the treatments of scarification with sandpaper commercial and with sulfuric acid and commercial presented the best results.It was also concluded that the strawberry seeds showed positive photoblastic behavior. For regeneration through organogenesis of genetic lineages obtained from seeds it was used leaf and petiole explants conditioned in culture medium supplemented with different concentrations of growth regulators.The variables evaluated were: percentage of explants with calluses, with buds and shoots, number of buds and shoots per explants, percentage of rooted plants, fresh weight of plants and percentage of plant survival after acclimatization. Through these assessments it was possible to conclude that the leaf explants showed better results for the genotypes tested. The growth regulators recommended are TDZ and AIB, at concentrations of 2,0 mg L-1 and 0,1 mg L-1 respectively, for the lineage obtained from cultivating Camiño Real, and 1,5 mg L-1 and 0,1 mg L-1 respectively, for the lineage derived from the cultivar Festival. The results of seed germination and plant regeneration have shown that it is possible to apply the technique of plant tissue culture for strawberry (FragariaxananassaDuch). Juntamente com a amora e a framboesa, o morango (Fragaria x ananassa) faz parte do grupo dos pequenos frutos vermelhos. Pertencente a família Rosaceae, o morango destaca-se pelo valor nutricional e pela importância econômica. Nos últimos anos tem se observado um crescimento na produção brasileira de morangos, no entanto, a participação do Brasil na produção mundial ainda é reduzida. Visando o aumento da produção e a melhoraria da qualidade dos frutos produzidos têm se buscado alternativas, tais como cultivares melhor adaptadas as regiões de produção e mudas de melhor qualidade. Considerando que a cultura de tecidos vegetais pode ser uma técnica para introdução de melhorias neste sistema de produção, o trabalho foi desenvolvido com os objetivos de avaliar as condições de germinação de sementes de morangueiro cv. Camiño Real e estabelecer os protocolos de regeneração por meio de organogênese, para as linhagens genéticas obtidas a partir das cultivares Camiño Real e Festival. Para avaliação das condições de germinação, foram utilizados aquênios frescos e armazenados por 2 e 12 meses. Estes foram submetidos à diferentes tratamentos para germinação in vitro e fotoperíodo de 16 horas e de 0 horas. A partir das avaliações da porcentagem de germinação e da massa fresca das plântulas obtidas, concluiu-se que o armazenamento dos aquênios reduz significativamente a viabilidade dos mesmos, para aquênios frescos os tratamentos de escarificação com lixa comercial e com ácido sulfúrico foram os que apresentaram melhores resultados. Concluiu-se ainda que as sementes de morango apresentam comportamento fotoblástico positivo. Para a regeneração, por meio de organogênese indireta, das linhagens genéticas obtidas a partir das sementes foram utilizados explantes foliares e peciolares acondicionados em meio de cultura suplementado com diferentes concentrações de reguladores de crescimento. As variáveis avaliadas foram: porcentagem de explantes com calos, com gemas e com brotos, número de gemas e de brotos por explante, porcentagem de plântulas enraizadas, massa fresca das plântulas e porcentagem de sobrevivência das plantas após a aclimatação. Por meio destas avaliações foi possível concluir que os explantes foliares apresentaram melhores resultados para os genótipos testados. Os reguladores de crescimento recomendados foram TDZ e AIB, nas concentrações de 2,0 mg L-1 e 0,1 mg L-1 respectivamente, para a linhagem obtida a partir da cultivar Camiño Real, e 1,5 mg L-1 e 0,1 mg L-1 respectivamente, para a linhagem obtida a partir da cultivar Festival. Os resultados obtidos de germinação de sementes e de regeneração de plantas, mostraram que é possível aplicar a técnica de cultura de tecidos vegetais para o morangueiro (Fragaria x ananassaDuch).

Paleobotanical studies suggest that roots evolved at least twice independently during land plant diversification, once in lycophytes and once in euphyllophytes. Auxin promotes postembryonic root initiation in both groups but from different cell types. In several euphyllophytes, such as Arabidopsis, rice, and maize, AS2/LOB-domain (ASL/LBD) proteins act directly downstream of auxin and are conserved elements necessary for root initiation. It is currently unknown whether similar or different genetic mechanisms act downstream of auxin for root initiation in lycophytes and euphyllophytes. We searched for ASL/LBD proteins in genome sequences spanning the tree of life to retrace their evolutionary history. We performed a phylogenetic analysis of ASL/LBD proteins and mapped the functions of all characterized ASL/LBD onto the phylogenetic trees. We identified a clade specifically associated with root development, which includes no lycophyte sequence. This points toward the existence of distinct genetic mechanisms downstream of auxin for root initiation in lycophytes and euphyllophytes.

International audience; The effects of endocrine disrupting chemicals (EDCs) on reproduction are well known, whereas their developmental effects are much less characterized. However, exposure to endocrine disruptors during organogenesis may lead to deleterious and permanent problems later in life. Zebrafish (Danio rerio) transgenic lines expressing the green fluorescent protein (GFP) in specific organs and tissues are powerful tools to uncover developmental defects elicited by EDCs. Here, we used seven transgenic lines to visualize in vivo whether a series of EDCs and other pharmaceutical compounds can alter organogenesis in zebra-fish. We used transgenic lines expressing GFP in pancreas, liver, blood vessels, inner ear, nervous system, pharyngeal tooth and pectoral fins. This screen revealed that four of the tested chemicals have detectable effects on different organs, which shows that the range of effects elicited by EDCs is wider than anticipated. The endocrine disruptor tetrabromobi-sphenol-A (TBBPA), as well as the three drugs diclofenac, trichostatin A (TSA) and valproic acid (VPA) induced abnormalities in the embryonic vascular system of zebrafish. Moreover, TSA and VPA induced specific alterations during the development of pancreas, an observation that was confirmed by in situ hybridization with specific markers. Developmental delays were also induced by TSA and VPA in the liver and in pharyngeal teeth, resulting in smaller organ size. Our results show that EDCs can induce a large range of developmental alterations during embryogenesis of zebrafish and establish GFP transgenic lines as powerful tools to screen for EDCs effects in vivo.

It is possible to determine the optimum time for permanence of vegetative propagules (mini-cuttings) inside a greenhouse for rooting, and this value can be used to optimize the structure of the nursery. The aim of this study was to determine the dynamics of adventitious rooting in mini-cuttings of three clones of Eucalyptus benthamii x Eucalyptus dunnii. Sprouts of H12, H19 and H20 clones were collected from mini-stumps that were planted in gutters containing sand and grown in a semi-hydroponic system. The basal region of the mini-cuttings was immersed in 2,000 mg L-1 indole-3-butyric acid (IBA) solution for 10 seconds. The rooting percentage of the mini-cuttings, the total length of the root system and the rooting rate per mini-cutting were also evaluated at 0 (time of planting), 7, 14, 21, 28, 35, 42, 49 and 56 days. We used logistic and exponential regression to mathematically model the speed of rhizogenesis. The rooting percentage was best represented as a logistic model, and the total length of the root system was best represented as an exponential model. The clones had different speeds of adventitious rooting. The optimum time for permanence of the mini-cuttings inside the greenhouse for rooting was between 35 and 42 days, and varied depending on the genetic material. O tempo ideal de permanência de propágulos vegetativos (miniestacas) no interior da casa de vegetação para a rizogênese é possível de ser determinado matematicamente, o que pode otimizar as instalações do viveiro. O objetivo deste estudo foi determinar a dinâmica de enraizamento de miniestacas de três clones de Eucalyptus benthamii x Eucalyptus dunnii. Brotações dos clones H19, H12 e H20 foram coletadas de minicepas plantadas em canaletão com areia e cultivadas sob sistema semi-hidropônico. A região basal da miniestaca foi imersa em solução de 2.000 mg L-1 de ácido indolbutírico (AIB) por 10 segundos. A porcentagem de enraizamento de miniestacas, o comprimento total de raízes e a taxa de enraizamento por miniestaca foram avaliados a cada sete dias (0 - instante da estaquia, 14, 21, 28, 35, 42, 49 e 56 dias). Foram utilizadas as regressões logística e exponencial para a modelagem matemática da velocidade dos processos rizogênicos. O modelo logístico teve o melhor ajuste para o percentual de enraizamento, e o modelo exponencial para o comprimento total do sistema radicular. Os clones apresentaram diferentes velocidades de enraizamento. O tempo ideal de permanência das miniestacas dentro da casa de vegetação para o enraizamento está entre 35 a 42 dias, podendo variar em função do material genético.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior The present study was conducted to evaluate the influence of ethylene, polyamines and gas exchange in in vitro morphogenesis of ornamental pepper. Seeds of Capsicum annuum L. were surface-sterilized and inoculated in MS medium supplemented with B5 vitamins, myo-inositol, sucrose and agar. Benzyladenine and 3-indoleacetic acid were added to the medium to elicit organogenesis from cotyledon and hypocotyl segments. The effect of gas exchange was evaluated comparing three flask sealings: rigid polypropylene lid (PRV) without vents; PRV with one or two vents covered with a membrane (MilliSeal® AVS-045 Air Vent). Photosynthetic pigments, morphologic characters and histological analyses from each treatment were performed. The effects of ethylene inhibitor, promoter and scavenger (aminoethoxyvinilglycine AVG, silver tiosulfate STS, 1-aminocyclopropane-1-carboxylic acid ACC, and mercury perchlorate- MP, respectively), polyamines (spermine, spermidine and putrescine) and its inhibitor (methylglyoxal-bis-guanylhydrazone - MGBG) were also analysed by the addition of these compounds into the regeneration medium. Plants maintained in glass flasks capped with vented lids showed more vigorous growth and differentiated anatomical structures as compared to the control. These treatments resulted in higher plants, higher number and more expanded leaves, greater fresh and dry weights, and photosynthetic pigments. However, for adventitious shoot bud and calli induction, the explant source was more effective than flask sealing. AVG, STS and MP caused a greater regeneration frequency. ACC and MGBG induced a lower regeneration frequency, high callus formation and showed the higher levels of ethylene. On the other hand, when the polyamines were added to the medium, ethylene levels were lower than control and regeneration frequency was higher. These results indicate that higher air exchange is beneficial for in vitro culture of Capsicum annuum L. and the endogenous levels of ethylene and polyamines interfere directly in morphogenic responses during in vitro regeneration of this species. O presente trabalho teve por objetivo avaliar a influência do etileno, das poliaminas e das trocas gasosas na morfogênese in vitro de pimenteira ornamental. Sementes de Capsicum annuum L. foram desinfestadas e inoculadas em meio MS suplementado com vitaminas B5, mio-inositol, sacarose e ágar. Para a organogênese, explantes cotiledonares e hipocotiledonares foram cultivados no meio descrito, porém adicionado de 6-benziladenina (BA) e ácido indolacético (AIA). O efeito das trocas gasosas foi avaliado utilizando três tipos de vedações dos frascos: tampas de polipropileno rígido sem membrana, com uma e com duas membranas (MilliSeal® AVS-045 Air Vent). Análises histológicas, de pigmentos fotossintéticos e de características morfológicas foram realizadas. Também foram realizados tratamentos com inibidores (aminoetoxivinilglicina AVG, tiossulfato de prata - STS), sequestrador (perclorato de mercúrio PM) e promotor da produção de etileno (ácido 1-carboxílico 1-aminociclopropano - ACC), além da adição de poliaminas (putrescina, espermidina e espermina) e de seu inibidor (metilglioxal-bis-guanil hidrazona MGBG) ao meio de regeneração. Plântulas cultivadas em frascos com tampas com membranas apresentaram estruturas anatômicas diferenciadas e crescimento mais vigorosos comparados ao controle. Estes tratamentos resultaram em plântulas maiores, com folhas mais expandidas, maior número de folhas, maior massa fresca, maior teor de massa seca e maior quantidade de pigmentos fotossintéticos. Todavia, na indução de organogênese os tipos de vedação não foram significativos, tendo o tipo de explante sido mais determinante na formação de gemas e calos. AVG, STS e o PM induziram alta frequência de regeneração. O ACC e o MGBG induziram menor taxa de regeneração, elevada calogênese e apresentaram níveis mais altos de etileno. Por outro lado, quando se adicionou as poliaminas ao meio de cultura, os níveis de etileno foram mais baixos que o controle e a frequência de regeneração foi mais alta. Os resultados deste estudo mostram que maiores trocas gasosas são benéficas no cultivo in vitro de Capsicum annuum L. e que os níveis endógenos de etileno e poliaminas interferem diretamente nas respostas morfogênicas durante a regeneração in vitro desta espécie.

Resumo: O objetivo deste trabalho foi estabelecer um protocolo de micropropagacao para Pinus taeda, pelas tecnicas de organogenese direta e multiplicacao de gemas axilares. Sementes de familias selecionadas (F27 e B05) e de pomar comercial, fornecidas pela empresa Battistella Florestal (Rio Negrinho ¡V SC) foram submetidas a tratamento de estratificacao (5¢XC „b 1¢XC durante 25 dias) para a superacao da dormencia. Brotacoes apicais obtidas de plantulas com cinco ou dez dias de germinacao foram as fontes de explantes. Para a inducao da formacao de brotacoes adventicias e/ou axilares, foram utilizados meio de cultura WV5 com 0, 11, 22 ou 44 ƒÝM de 6-benzilaminopurina (BAP) combinada com 0,05 ƒÝM de acido naftaleno acetico (ANA) e meio de cultura WPM com 11 a 44,0 ƒÝM de BAP e 0; 0,5 ou 0,75 ƒÝM de ANA durante 14 dias, e subcultivo para meio sem reguladores e WPM. Para a tecnica de multiplicacao de brotacoes axilares, utilizaram-se brotacoes apicais (0,5 a 2,0 cm de comprimento) e segmentos nodais (1,0 a 4,0 cm de comprimento), em meios de cultura WV5 e GDm, acrescidos de concentracoes de 0,1 a 4,0 ƒÝM de BAP, e cultivo durante quatro e oito semanas. Brotacoes de 1,5 cm de comprimento foram submetidas a inducao ao enraizamento, por nove dias em meios GDm/2, WV5/2, WV3/2 ou em agar-agua, com 2,68 ƒÝM de ANA e 0,44 ƒÝM de BAP seguida de subcultivo em meio de cultura sem reguladores. A aclimatizacao de mudas foi feita diretamente em casa de vegetacao ou com periodo previo em sala de crescimento. Os melhores resultados para a inducao de brotacoes multiplas foram obtidos com explantes de cinco dias de idade, cultivados em meio de cultura WV5, 44,0 ƒÝM de BAP e 0,05 ƒÝM de ANA, obtendo-se 50% de explantes com brotacoes e numero medio de 4,8 brotacoes por explante. Para a multiplicacao de gemas axilares, segmentos nodais de 1,0 cm de comprimento, inoculados em meio WV5 contendo 2,5 ƒÝM de BAP foram os mais eficientes, com 90% de explantes com brotacoes e 4,0 brotacoes por explante em media. Com brotacoes apicais, observou-se maior porcentagem de alongamento (286,5%) em meio WV5 sem reguladores, com explantes de 0,5 cm de comprimento. A melhor duracao do subcultivo foi de oito semanas. O genotipo influenciou os resultados de germinacao, inducao e multiplicacao de brotacoes, sendo que as melhores respostas ocorreram com a familia F27, seguido da B05 e genotipos do pomar comercial. O meio de cultura WV5 permitiu a manutencao prolongada das culturas in vitro, as quais apresentaram vigor durante dois anos. Para inducao de raizes, recomenda-se a formulacao de agar-agua, acrescida de 2,68 ƒÝM de ANA e 0,44 ƒÝM de BAP durante nove dias, seguida de transferencia para meio GDm/2, sem reguladores para o desenvolvimento de raizes (55,6% de enraizamento). A sobrevivencia das mudas aclimatizadas foi de 85% apos 40 dias, sendo os 20 primeiros dias em sala de crescimento e os dias posteriores em casa de vegetacao. Concluiu-se que foi estabelecido um protocolo de micropropagacao para P. taeda.

[POR] Foram comparados os efeitos de diferentes dietas e da transição alimentar sobre o desempenho zootécnico e a morfologia do trato digestório de larvas de pacu alimentadas com duas dietas comerciais e uma dieta experimental microencapsulada produzida por gelificação interna. Larvas com quatro dias de vida receberam os seguintes protocolos alimentares: somente náuplios de artêmia em quantidades crescentes (controle positivo); larvas mantidas em jejum (controle negativo); três tipos de alimento formulado durante todo o experimento (dieta experimental microencapsulada, dieta comercial NRD1.2/2.0, Inve, USA, e dieta Poli-Peixe 450F, PoliNutri, Brasil); e três protocolos de transição alimentar com náuplios de artêmia durante os seis primeiros dias, seis dias de coalimentação, e somente as respectivas dietas formuladas após esse período. O experimento foi conduzido por 23 dias em delineamento inteiramente casualizado, com avaliações periódicas do crescimento e do sistema digestório. A taxa de sobrevivência foi determinada ao final do experimento. Os melhores desempenhos de crescimento e sobrevivência, assim como mais acelerada organogênese do sistema digestório, foram verificados entre os animais que receberam alimento vivo. As larvas que passaram pela transição alimentar apresentaram médias intermediárias de crescimento e sobrevivência e não diferiram entre si. Entre os grupos que receberam apenas dieta inerte, somente naquele alimentado com a dieta nacional houve sobrevivência até o final do experimento. Nenhuma das dietas formuladas é adequada para uso como alimento exclusivo para larvas de pacu. Four-day old pacu larvae received the following treatments: only artemia nauplii in increasing amount during the experiment (positive control); larvae maintained at fasting (negative control); three types of formulated diets throughout the experiment (experimental microencapsulated diet, commercial diet NRD1.2/2.0, Inve, USA, and diet Poli-Peixe 450F, PoliNutri, Brazil); and three weaning protocols in which artemia nauplii were given from the 1 st to 6 th days, six days of co-feeding (artemia nauplii + the respective formulated diet), and only respective diets formulated after this period. The experiment was conducted for 23 days in a completely randomized design, with periodical evaluations of growth and digestive system. Survival rate was determined at the end of the experiment. The best performances of growth and survival, as well as the organogenesis of the digestive tract, were verified for the animals which received live feed. The larvae at weaning treatments induced intermediate growth and survival, which did not differ statistically between them. Considering the treatments that received only formulated diets as first feed, the PoliNutri diet was the unique that showed alive larvae at the end of experiment. None of the formulated diets are adequate as initial exogenous food for pacu larvae. Dissertação de mestrado em Biodiversidade e Biotecnologia Vegetal apresentada ao Departamento de Ciências da Vida. A espécie Arbutus unedo (Ericaceae) possui porte arbustivo ou arbóreo. O medronheiro é capaz de suportar temperaturas baixas (até -12ºC) e é tolerante à seca. Encontra-se largamente distribuído ao longo da bacia do Mediterrâneo, desde Espanha até à Turquia, algumas zonas do Norte de África e ilhas Mediterrânicas, e ainda na costa Atlântica, incluindo Irlanda e Portugal. O medronheiro tem várias aplicações: estabilização de solos, programas de reflorestação, floricultura e na produção de compota, geleia, aguardente mel e biomassa. Apesar de ser tradicionalmente uma espécie NUC (www.underutilized-species.org), o medronheiro tem também sido a aposta de vários agricultores, sendo actualmente reconhecida como uma espécie com elevado potencial. O presente trabalho insere-se num projecto mais alargado de melhoramento do medronheiro e é mais uma contribuição para um melhor conhecimento dos processos de propagação desta espécie. Foram estudadas diferentes técnicas de propagação: germinação de sementes, embriogénese somática, organogénese e multiplicação em meio líquido. Relativamente à germinação de sementes, a baixa percentagem de germinação pode ser contornada com pelo menos 14 dias de estratificação a 4ºC, obtendo-se assim percentagens de germinação próximas de 100%. Apesar de não assegurar a manutenção do genótipo, é uma técnica simples de aplicar. Em ralação à embriogénese somática, vários genótipos foram avaliados, não se tendo verificado diferenças na percentagem de indução. Os embriões somáticos passavam pelos estados de desenvolvimento típicos: globular, cordiforme, torpedo e cotiledonar, embora com algumas variações relativamente a este padrão geral. Uma percentagem considerável dos embriões formados apresentava algum tipo de anomalia, o que pode comprometer o uso desta técnica para fins de propagação. Nos estudos relacionados com a organogénese, verificou-se que a multiplicação do medronheiro por esta técnica apresenta grande potencial, devido ao considerável número de plantas obtidas. No entanto, esta técnica deve ser alvo de estudos mais pormenorizados, de forma a perceber os mecanismos envolvidos e assim explorar todo o seu potencial. O crescimento em meio líquido de plantas de medronheiro revelou-se bastante eficaz na sua multiplicação, no entanto, o enraizamento das plantas obtidas foi muito reduzido. Assim, para que a técnica possa ser aplicada, esta limitação deve ser contornada. A estabilidade genética das plantas propagadas foi testada através de vi marcadores moleculares. Não se verificaram diferenças entre perfis, pelo que provavelmente não existem diferenças entre as plantas-mãe e as plantas in vitro testadas. No entanto, uma análise mais pormenorizada deve ser realizada. Nos ensaios de melhoramento, foram abordadas três técnicas distintas: polinização artificial, embriogénese polínica e poliploidização. No que diz respeito à polinização artificial, a germinação do pólen no estigma foi observada. Os tubos polínicos percorreram todo o comprimento do estilete em 20-24 h. Este resultado foi obtido nos ensaio realizados in vitro e in vivo, o que abre boas perspectivas para possíveis cruzamentos entre diferentes genótipos. Relativamente à embriogénese polínica, quando as anteras foram colocadas em cultura não se obtiveram embriões, mas sim calo que não revelou capacidade embriogénica. Foi também estabelecida uma relação entre a morfologia dos botões florais e o estado de desenvolvimento dos micrósporos, o que pode ser útil em futuras tentativas de obtenção de haplóides. Nos ensaios de poliploidização usou-se colquicina e orizalina. Os tratamentos com orizalina e em meio líquido foram mais agressivos para as plantas. Apesar de a ploidia das plantas obtidas não ter sido testada, verificou-se um crescimento reduzido, o que pode indicar diferenças na sua ploidia. Apesar de preliminares, os resultados obtidos abrem boas perspectivas no que diz respeito a diferentes técnicas de propagação do medronheiro. No entanto, para que possam ser utilizadas de forma efectiva na propagação em massa, alguns estudos mais detalhados terão de ser levados a cabo, no sentido de optimizar algumas das técnicas. No melhoramento do medronheiro, este trabalho representa um pequeno avanço. Os resultados da polinização artificial são especialmente interessantes, e permitirão a realização de um programa de cruzamentos de genótipos, no sentido de obter variedades com características vantajosas Arbutus unedo (Ericaceae) is a small shrub or tree that can stand low temperatures (until -12ºC) and is tolerant to water stress. This species is spread around the Mediterranean basin, from Spain to Turkey, some areas in the North Africa and Mediterranean islands, and also in the Atlantic coast, including Ireland and Portugal. The strawberry tree has innumerous applications such as soil stabilization, reforestation programs, floriculture, and production of jams, spirits, honey and biomass. Although being traditionally considered a NUC species (www.underutilized-species.org), the great interest recently shown by farmers and other stakeholders has increased the potential of this tree. The present work is part of a more broad project which the main goal is the improvement of strawberry tree. Different propagation techniques were studied: seed germination, somatic embryogenesis, organogenesis and multiplication in liquid medium. The results concerning seed germination showed that the rates were low. However, stratification at 4ºC (for at least 14 days) increased germination to values close to 100%. Although this technique does not assure propagation of selected trees it is easy to apply and can be used to increase variability and to better germinate hybrid seeds between genotypes. Somatic embryogenesis induction was tested in different genotypes, and no significant differences were observed. In spite of some variations, somatic embryo morphology was similar to the general pattern, going through globular, hearth, torpedo and cotyledon stages. A considerable rate of abnormal somatic embryos was observed which can compromise the use of this technique for propagation purposes. The propagation of the strawberry tree through organogenesis has a great potential since a large number of plants can be obtained. However, more work must be carried out before it can be used for propagation purposes. The results also showed that strawberry tree plants can be propagated in liquid medium opening the way for the use of bioreactors. However, these plants were difficult to root. Therefore, this limitation must be overcome, in order to apply this technique for propagation purposes. The genetic stability of propagated plants was tested, and no differences were found between the mother plants and the in vitro propagated plants. Nevertheless, a more detailed analysis must be carried out with more primers and more plants. viii Regarding plant breeding, three different techniques were used: artificial pollination, pollen embryogenesis and polyploidization. Concerning the artificial pollination, pollen germination in stigma was observed and the growth of the pollen tubes through the style occurs in 20-24 h. This result was obtained not only in vitro but also in in vivo pollinations, which offers good perspectives to control artificial hybridization. With regard to pollen embryogenesis, callus were obtained but without embryogenic or organogenic potential. A relation between flower buds and microspore development stage was established which may be useful in further attempts to obtain haploids through microspore culture. In polyploidization assays, colchicine and oryzalin were used. The treatments with oryzalin and liquid medium were showed to be more aggressive to the plants. Although the ploidy level was not assessed, a low growth rate was observed, which can indicate different ploidy levels among the plants. These preliminary results opened new ways of research for the breeding of Arbutus unedo and are one more contribution to the study of this species. However, in order to use these techniques for mass propagation, mores studies must be carried out. The pollination results are especially interesting, for future crossbreeding assays and for the obtention of plants with new characteristics.

The capacity of preformation and neoformation and the structure of winter buds are vegetative attributes that may vary between plant species and according to ontogenetic stages of the same species. The present study describes and evaluates these features for the four tree species of Proteaceae occurring in Patagonia. In particular, it analyzes the structure and development of the distal buds of the trunk and the preformed or neoformed nature of the organs involved. Two of the species, Embothrium coccineum and Lomatia hirsuta, have scaly buds, in which primordia of green leaves are covered by cataphylls. The shoots of both species may include neoformed organs, more frequently so in juvenile trees. Lomatia ferruginea and Gevuina avellana have naked buds with a low number of primordia; in juvenile and adult trees of both species trunk shoots are entirely preformed. The structure of buds and shoots suggests two different growth modalities of the axes, which would be related to ecological breadth (narrower in the species with naked buds than in those with scaly buds) and distribution area of these species. The considerable morphological differences between the two Lomatia species studied raise the question whether they ought to be included in the same genus. Fil: Magnin, Noel Amaru. A fundamental and unresolved question of life is how organs are formed. The shape and form of organs emerges by spatio temporally controlled division and motility of cells. As both processes are tightly coordinated, interactions amongst cells are required to ensure stability and integrity. Many genetic networks controlling the polarised cell motility or promoting cell division have been identified. Action between cells results in an increased complexity. Yet cells give rise to regular patterned organs. The form of objects and their motion is subject to physical laws. Cells are of an active character, divide and move, interesting properties for a material, with potentially new knowledge emerging from their study. Here, we perform a quantitative characterisation of two experimental models for tissue morphogenesis. Using cultured epithelial sheets we address the mechanical properties of growth control and identify regulatory mechanisms. Based on this study, we propose a phenomenological description of tissue dynamics, reproducing the observed data. Using the methods developed to understand the cultured sheet, we approach the role of mechanics in the migration of an embryonic tissue. We measure the directed motion of the tissue and show that the findings can be reproduced by coupling the biophysical model of motile cells to a dynamically regulated polarisation mechanism. Formgebung von Organen ist ein grundlegendes, ungelöstes Problem des Lebens. Ihre Gestalt resultiert aus raumzeitlich kontrollierten Zellteilungen sowie Bewegungen von Zellen. Um mechanische Stabilität sowie Integrität des Gewebes zu gewährleisten, werden Zell-Zell Wechselwirkungen benötigt. Viele genetische Netzwerke kontrollieren die polarisierte Zellbeweglichkeit oder fördern die Zellteilung. Interaktion zwischen Zellen führt zu einer erhöhten Komplexität. Dennoch bilden sich reguläre Muster. Die Form von Objekten und deren Bewegung unterliegt physikalischen Gesetzen. Zellen sind von aktiver Art, teilen und bewegen sich, interessante Eigenschaften für ein Material, welche in neuen Erkenntnissen münden könnten. In dieser Arbeit führen wir eine quantitative Charakterisierung von zwei Modellsystemen der Morphogenese von Geweben durch. Anhand von kultivierten Epithelien behandeln wir die mechanischen Eigenschaften der Wachstumskontrolle und identifizieren Regulationsmechanismen. Darauf aufbauend, schlagen wir eine phänomenologische Modellbeschreibung für Gewebedynamik vor, welche die Beobachtungen reproduziert. Wir machen Gebrauch von diesen Methoden um die Mechanik der Migration eines embryonalen Epithels zu verstehen. Dabei messen wir die gerichtete Bewegung des Gewebes und zeigen, dass die resultierenden Daten durch Kopplung der biophysikalischen Motilitätsbeschreibung an einen dynamisch regulierten Polarisationsmechanismus reproduziert werden.

Congenital Hypothyroidism (CH) is the most common endocrine disorder, affecting one out of 2000-4000 newborns. Most CH are due to a defect in thyroid embryonic development and they can lead to severe phenotypes if not treated correctly. Multiple observations argue in favor of a genetic cause in a minority of thyroid dysgenesis, but to date, only few cases could be explained by a mutation in one of the genes coding for the factors known to be important in thyroid development and/or function (NKX2-1, PAX8, FOXE1, TSHR). This is the reason why it was important to develop new models allowing the discovery of new genes/mechanisms potentially implicated in the gland organogenesis. To that purpose, we set up in the laboratory a structure enabling the use of zebrafish as an animal model. The latter is indeed more and more used by developmental biologists, including by scientists interested in thyroid development.The first step of our project consisted in a deeper characterization of the model, notably by the study of the expression patterns of the thyroid functional differentiation markers. Furthermore, the exact role of the Tsh/Tshr signaling – main regulator of thyroid growth and function in mammals – was dissected. In a second part of the project, we generated a stable transgenic line (tg(tg:mCherry)) allowing the visualization of thyroid development in living embryos and in a dynamic manner, thanks to real-time imaging techniques. On the one hand, this tool enabled us to better understand the morphological aspect of the different stages of thyroid development, such as the budding, evagination, relocalization or folliculogenesis. On the other hand, the use of double transgenic fishes obtained by crossing tg(tg:mCherry) with other lines expressing GFP in surrounding structures of interest, allowed us to highlight the contacts between the cardiovascular system and thyroid, and this along the whole gland development. The introduction of this model within the laboratory paves the way for the discovery and the study of thyroid intrinsic and extrinsic genes/mechanisms which might play a role on its development.L’hypothyroïdie congénitale (HC) est une maladie relativement fréquente, touchant un nouveau-né sur 2000-4000. La majorité des HC sont dues à un défaut dans le développement embryonnaire de la glande, et peuvent mener à des phénotypes sévères si elles ne sont pas correctement traitées. Il existe plusieurs arguments en faveur d’une cause génétique dans une minorité de ces dysgénésies thyroïdiennes mais, à ce jour, seuls quelques cas ont pu être reliés à une mutation dans un des gènes codant pour des facteurs connus pour être importants dans le développement/la fonction de la glande (NKX2-1, PAX8, FOXE1, TSHR). C’est pour cette raison qu’il est important de développer de nouveaux modèles pouvant permettre la découverte de nouveaux gènes/mécanismes potentiellement impliqués dans l’organogénèse de la glande. A cette fin, nous avons mis en place au sein du laboratoire une structure permettant l’utilisation du poisson-zèbre comme modèle animal. Ce dernier est en effet de plus en plus utilisé par les biologistes du développement, y compris par les scientifiques qui s’intéressent au développement thyroïdien.La première étape de notre travail a consisté en une caractérisation approfondie du modèle, notamment par l’étude du réseau d’expression des marqueurs de différenciation fonctionnelle de la glande. Syngonanthus mucugensis Giul. subsp. mucugensis is an herbaceous plant with significant economic value in the ornamental dry flower business. The restricted occurrence of the municipality Mucugê-BA, Brazil, exclusively associated with extractive exploitation, has considered this species as endangered. The objective of this work was to evaluate the organogenic potential of three different types of S. mucugensis subsp. mucugensis explants to promote the development of an alternative method to the propagation of the genetic resources of this important plant. The morphogenetic capacities of the leaf, stem and root this species was tested using Murashige and Skoog culture medium at half salt concentration and different concentrations of growth of regulators benzylaminopurine - BAP (0.00; 2.22 and 4.44 µM), and naphthalene acetic acid - NAA (0.00; 1.34 and 2.68 µM). The morphoanatomic events that lead to formation of shoots were described. Stems proved to be the best source of explants, showing 58.75% regeneration of shoot by direct organogenesis in the absence of growth regulators, and 32.18 and 47.55% of shoot regeneration by indirect organogenesis in the presence of 2.22 and 4.44 µM BAP, respectively. As for leaves, there was callus formation, but without regenerating shoots. Morphogenesis was not observed when roots were used as explants. The histological analyses showed that shoot regeneration in S. mucugensis subsp. mucugensis occurred both indirectly, by unorganized tissue differentiation, and directly through returning to merismatic activity in differentiated mature cells and preexisting bud proliferation.Syngonanthus mucugensis Giul. subsp. mucugensis é uma herbácea com grande potencial de utilização no comércio de flores secas ornamentais. A ocorrência restrita ao município de Mucugê-BA, Brasil, associado à exploração extrativista tem levado essa espécie ao risco de extinção. Neste estudo, objetivou-se avaliar o potencial organogênico de três diferentes tipos de explantes de S. mucugensis subsp. mucugensis visando ao desenvolvimento de um método alternativo para a sua propagação. A capacidade morfogênica de caule, folha e raiz foi testada utilizando o meio de cultura Murashige e Skoog com metade da concentração salina e diferentes concentrações dos reguladores de crescimento benzilaminopurina - BAP (0,00; 2,22 e 4,44 µM) e ácido naftaleno acético - ANA (0,00; 1,34 e 2,68 µM). Os eventos morfoanatômicos que levaram a formação dos brotos foram descritos. O caule demonstrou ser a melhor fonte de explante, apresentando 58,75% de regeneração de brotos via organogênese direta, em meio livre de regulador de crescimento; e 32,18 e 47,55% de regeneração de brotos por organogênese indireta na presença de 2,22 e 4,44 µM de BAP, respectivamente. Para folha, foi obtida a formação de calos sem regeneração de brotos, não sendo observada morfogênese, quando se utilizou raiz como explante. As análises histológicas mostraram que a regeneração de brotos em S. mucugensis subsp. mucugensis ocorreu tanto por via indireta, pela diferenciação do tecido calogênico desorganizado, quanto por via direta pela retomada da atividade meristemática em células maduras diferenciadas e proliferação de gemas pré existentes.

Shapes emerge at all scales and in all physical systems, living or not, as the result of interactions between the elements of the system according to a certain set of rules. The crucial issue of morphogenesis study is to identify which elements and which rules are responsible for the emergence of the shape's striking features that one seeks to explain. In developmental biology, the molecular context and the central role of genes often mask that, as in any material system, geometrical shapes emerge following interactions well de ned in time and space, interactions in which genes and proteins are de nitely crucial actors. In this manuscript, we will study the morphogenesis of two distinct systems. The rst one refers to XIXth century's osmosis experiments by Dr. St ephane Leduc. It consists in the spontaneous formation of silica tubes growing evenly on both sides of a central fracture, and occurs when injecting a solution in another, a precipitate forming at the interface. The rest of the manuscript deals with lung morphogenesis in mammals. We will rst build the theoretical framework of a very general growth model based upon the observations of molecular biology as well as the geometry of the organ. Then, we will see how elementary considerations of geometry and di usion account for the patterning of involved genes, and also for the emergence of the striking features of embryonic lung morphology : the arborescence, the fact that bronchi never penetrate another, and the apparition of a characteristic distance between distal epithelium and mesothelium. We will also introduce theoretical tools that allow a deeper understanding of the underlying mechanisms. The first step in plant transformation using somatic embryogenesis or organogenesis for plant regeneration in vitro has been the identification of genotypes with the best totipotency. Thus, the objective of this experiment was to evaluate the totipotency of ten cultivars and one soybean line from the Coodetec Gene Bank. The somatic embryos developed from immature cotyledons measuring from 4 to 6 mm in length, and the organogenesis was evaluated from mature soybean seeds. The cultivars CD 201, CD 216 and CD 220 were the most promising genotypes to produce somatic embryos, whereas the cultivar CD 217 had the highest quantity of differentiated explants, which represents the best regeneration efficiency for participating in organogenic systems. In the experiment with somatic embryogenesis, time to plant regeneration was ten months, but the plants were regenerated after only four months in the experiment with organogenesis. A identificação de genótipos que possuam maior totipotência quando submetidos à embriogênese somática e organogênese é a primeira etapa para a condução de um programa de transformação genética. Este trabalho teve como objetivo avaliar o potencial de embriogênese somática e organogênese em nove cultivares de soja e uma linhagem, do banco de germoplasma da Coodetec. Cotilédones imaturos de soja com 4 a 6 mm de comprimento foram utilizados para produção de embriões somáticos, induzidos com 2,4-D, e sementes maduras de soja foram usadas para avaliação do potencial organogênico. As cultivares CD 201, CD 216 e CD 220 mostram-se promissoras para produção de embriões somáticos, enquanto que para o sistema organogênico a cultivar CD 217 apresentou a maior quantidade de explantes diferenciados e manteve a maior eficiência de regeneração. Para obtenção de uma planta via embriogênese somática foram necessários 10 meses, em virtude do período necessário para obtenção dos explantes. O sistema organogênico se mostrou mais rápido, e plantas completas de soja foram obtidas em quatro meses de cultivo in vitro.

Nitric oxide (NO) is a gaseous molecule that participates in numerous plant signalling pathways. It is involved in plant responses to pathogens and development processes such as seed germination, flowering and stomatal closure. Using a permeable NO-specific fluorescent probe and a bacterial reporter strain expressing the lacZ gene under the control of a NO-responsive promoter, we detected NO production in the first steps, during infection threads growth, of the Medicago truncatula–Sinorhizobium meliloti symbiotic interaction. Nitric oxide was also detected, by confocal microscopy, in nodule primordia. Depletion of NO caused by cPTIO (2-(4-carboxyphenyl)-4,4,5,5-tetramethyl imidazoline-1-oxyl-3-oxide), an NO scavenger, resulted in a significant delay in nodule appearance. The overexpression of a bacterial hmp gene, encoding a flavohaemoglobin able to scavenge NO, under the control of a nodule-specific promoter (pENOD20) in transgenic roots, led to the same phenotype. The NO scavenging resulting from these approaches provoked the downregulation of plant genes involved in nodule development, such as MtCRE1 and MtCCS52A. Furthermore, an Hmp-overexpressing S. meliloti mutant strain was found to be less competitive than the wild type in the nodulation process. Taken together, these results indicate that NO is required for an optimal establishment of the M. truncatula–S. meliloti symbiotic interaction

Conselho Nacional de Desenvolvimento Científico e Tecnológico Different methodologies for in vitro regeneration have already been developed for several soybean cultivars. However, this species has proven to be recalcitrant to tissue culture and its in vitro morphogenetic response has been considered genotype dependent. Taking into account the need to develop reliable protocols for regeneration and acclimatization in soybean, the objectives of this study were to induce somatic embryogenesis and organogenesis in order to standardize protocols. Somatic embryos were obtained from immature cotyledons of four commercial soybean cultivars: CAC-1, CD 219 RR, CS 303 TNKCA and FMT Tucunaré. The methodologies for somatic embryogenesis adopted were effective. It was found that the embryogenic potential was genotype-specific, and the cultivars CAC-1 and CD 219 RR showed the higher responses, with an average of 10.83 and 13.36 somatic embryos per responding cotyledon respectively. Only somatic embryos derived from CAC-1 were able to regenerate plants, leading to eight acclimatized plants. Direct organogenesis was achieved from cotiledonary nodes of CAC-1 soybean and 156 regenerated plants were recovered. Clonal fidelity was evaluated through the use of the ISSR marker assay and eight recovered plants showed variations in the amplification profile. In addition, the same plants were analyzed for oil and protein content using the NIRS methodology. Plants with higher oil and protein contents in comparison with CAC-1 control plants were identified, indicating the presence of somaclonal variation. Taken together, the results of this study indicate specific protocols that can be used for soybean genetic transformation in the Soybean Quality Breeding Program that is under development at BIOAGRO/UFV. Diferentes metodologias de regeneração in vitro já foram desenvolvidas para diversas cultivares de soja. No entanto, essa espécie tem se mostrado recalcitrante, e suas respostas morfogênicas in vitro tem sido consideradas genótipo-específicas. Considerando-se a necessidade de desenvolvimento de protocolos confiáveis de regeneração e de aclimatização em soja, os objetivos do presente trabalho foram induzir a embriogênese somática e a organogênese visando à padronização de metodologias. Para os experimentos de embriogênese somática foram utilizados cotilédones imaturos de quatro cultivares comerciais de soja: CAC-1, CD 219 RR, CS 303 TNKCA e FMT Tucunaré. As metodologias adotadas para a obtenção de embriões somáticos a partir de explantes cotiledonares mostraram-se eficientes. Verificou-se que o potencial embriogênico foi genótipo-específico, sendo que as cultivares CAC-1 e CD 219 RR apresentaram as melhores respostas, com médias de produções de embriões por cotilédones de 10,83 e 13,36, respectivamente. Apenas embriões derivados da cultivar CAC-1 germinaram, sendo possível aclimatizar oito plantas dessa cultivar. Para os experimentos de organogênese, foram utilizadas sementes de soja das variedades CAC-1, sendo possível recuperar 156 plantas regeneradas. Essas foram submetidas à análise de fidelidade clonal por meio da utilização de marcadores ISSR, tendo sido identificadas oito plantas com variações no padrão de amplificação em relação a cultivar CAC-1. Além disso, as mesmas plantas foram submetidas a análises da porcentagem de óleo e de proteína por meio da metodologia NIRS. Plantas com elevados conteúdos de óleo e de proteína, quando comparadas com as testemunhas, foram identificadas, indicando presença de possíveis variantes somaclonais. Em conjunto, os resultados do presente trabalho indicam protocolos que podem ser utilizados, posteriormente, em experimentos de transformação genética de soja, aplicáveis no Programa de Melhoramento Genético da Qualidade do Óleo e Proteína da Soja em desenvolvimento no BIOAGRO/UFV.

O objetivo deste estudo foi avaliar o efeito de combinações de 6-benzilaminopurina (BAP) e ácido α-naftalenoacético (NAA), meios de cultura e antibióticos beta-lactâmicos sobre a regeneração in vitro de segmentos internodais de laranja doce ('Pêra', 'Valência' e 'Bahia') e limão 'Cravo'. Para a indução de brotações, os explantes internodais foram cultivados em meio Murashige e Skoog (MS) contendo as seguintes combinações de BAP/ANA: 0,0/0,0; 0,25/0,0; 0,25/0,25; 0,5/0,0; 0,5/0,5; 1,0/0,0; 2,0/0,0; 2,0/0,25; 2,0/0,5; e 1,0/2,0 mg L-1. A fim de testar a influência dos meios de cultura na organogênese, as formulações (MS), Murashige e Tucker (MT) e "woody plant medium" (WPM) foram avaliadas, associadas à melhor combinação de reguladores de crescimento obtida no experimento anterior. Foram utilizados quatro antibióticos beta-lactâmicos (timentin, cefotaxima, trihidrato de meropenem e augmentin), em diferentes concentrações, para avaliar sua influência na regeneração in vitro. As melhores respostas foram obtidas quando os explantes foram cultivados em meio MS com 500 mg L-1 de cefotaxima com as seguintes combinações BAP/ANA: 0,5 + 0,25 mg L-1; para 'Cravo'; 1,0 + 0,25 mg L-1; para 'Valência' e 'Bahia'; e 1,0 + 0,5 mg L-1; para 'Pêra'. Genótipos, meios de cultura e antibióticos beta-lactâmicos afetam a resposta morfogenética de tecidos maduros de citros. The objective of this study was to evaluate the effects of 6-benzylaminopurine (BAP) and α-naphthaleneacetic acid (NAA) combinations, basal media and beta-lactam antibiotics on in vitro organogenesis from mature stem segments of 'Pêra', 'Valência' and 'Bahia' sweet oranges and 'Cravo' rangpur lime. For induction of shoot regeneration, the segments of the four cultivars were placed on Murashige and Skoog (MS) medium containing the following BAP/NAA concentrations: 0.0/0.0; 0.25/0.0; 0.25/0.25; 0.5/0.0; 0.5/0.5; 1.0/0.0; 2.0/0.0; 2.0/0.25; 2.0/0.5; and 2.0/1.0 mg L-1. In order to test the influence of the culture media on shoot-bud induction, (MS), Murashige and Tucker (MT), and woody plant medium (WPM) formulations were evaluated, associated with the best combination of plant growth regulators obtained in the previous experiment. The influence of four beta-lactam antibiotics (timentin, cefotaxime sodium salt, meropenem trihydrate and augmentin) on shoot regeneration was determined. Better regeneration responses were achieved when internodal segments were cultured onto MS-based medium with 500 mg L-1 cefotaxime with the following BAP/NAA concentrations: 0.5 + 0.25 mg L-1 for 'Cravo', 1.0 + 0.25 mg L-1 for 'Valência' and 'Bahia', and 1.0 + 0.5 mg L-1 for 'Pêra'. Their propagation through the tissue culture allows the plant production in a large scale, with high sanitary in a short period of time. Another application of this technique is the regeneration or in vitro morphogenesis that consists in the organs induction by somatic embryogenesis or organogenesis, which is a pre-requirement for genetic transformation. The current work aimed at optimizing the multiplication and in vitro regeneration of Cydonia oblonga Mill. rootstocks, cultivars Adams and MC. For the multiplication experiments were used apicals shoots with excised tip (1,5cm), and inoculated in MS½ and MS¾ or MS medium modified (¾ of the normal concentration of NH4NO3 and KNO3 and EDTA-Ferric), containing different BAP concentrations or interaction among BAP and AG3 concentrations. The explants were maintained in growth chamber with 16h photoperiod, 48 μmol m-2 s-1 density photon flux and 25±2ºC temperature, for 40 days. The variables analyzed were shoots number per explants, shoots length, node number per shoot and percentage of hyperhydric shoots. In the regeneration experiments it was used as initial explants entire leaves or third basal leaves, young or adult leaves, with or without petiole, inoculated in MS medium, supplemented with TDZ (0, 1, 2, 3 and/or 4mg dm-3) and ANA (0,1mg dm-3), maintained in darkness for 40 days. Later, they were transferred to MS medium containing 1mg dm-3 TDZ and maintained in the light for 30 days more. After that period, the percentage of regenerate explants, the shoots number for regenerate explant and type of organogenesis formed was evaluated. Both cultivars present potential for in vitro propagation, however 'Adams' is more responsive, because it was obtained 6,3 shoots per explant, with 2,8mg dm-3 BAP, while ' MC' presents larger sensibility to this plant growth regulator and to develop the hyperhydric explants, however it demonstrated to be more efficient to the in vitro regeneration, presenting 26% of regenerating explants (with 1mg dm-3 TDZ). It was verified for both cultivars, the entire explant leaves were more responsive for the regeneration than basal thirds and that this happens mainly in the basal area of the explant, through direct and 9 indirect organogenesis. Based on the multiplication and the regeneration results, it was concluded that is possible the in vitro multiplication of these cultivars in large scale, as well as to improve the regeneration rates for future plant breeding works for characteristics of interest, through genetic transformation. Os marmeleiros (Cydonia oblonga Mill.) são excelente alternativa de diversificação de porta-enxertos para pereiras, devido ao interesse por plantas com porte reduzido, rápida frutificação, uniformidade nos pomares e boa qualidade das frutas. A sua propagação por meio da cultura de tecidos permite a produção de plantas homogêneas, em larga escala, com alta qualidade sanitária e num curto espaço de tempo. Outra aplicação desta técnica é a regeneração ou morfogênese in vitro, que consiste na formação de órgãos, seja por meio de embriogênese somática ou organogênese, sendo esta um pré-requisito para a transformação genética. O presente trabalho teve como objetivo otimizar a multiplicação e regeneração in vitro dos porta-enxertos de Cydonia oblonga Mill., cultivares Adams e MC. Para os experimentos de multiplicação foram utilizadas brotações apicais com ápice excisado (1,5cm), inoculadas em meio MS½ e MS¾ ou MS modificado (¾ da concentração normal de NH4NO3 e KNO3 e Ferro na forma de EDTA-Férrico), contendo diferentes concentrações de BAP ou interação entre BAP e AG3, mantidas em câmara de crescimento com fotoperíodo de 16 horas, 48 μmol m-2 s-1 e 25±2ºC de temperatura por 40 dias. Após, analisaram-se as variáveis: número de brotações por explante, comprimento das brotações, número de nós por brotação e percentagem de explantes com hiperidricidade. Nos experimentos de regeneração utilizou-se como explante inicial folhas inteiras ou terços basais, jovens ou adultas, com ou sem pecíolo, inoculados em meio MS, suplementados com TDZ (0, 1, 2, 3 e/ou 4mg dm-3) e ANA (0,1mg dm-3), permanecendo no escuro por 40 dias. Posteriormente, foram transferidos para meio MS contendo 1mg dm-3 de TDZ e mantidos na luz por mais 30 dias. Após esse período, avaliou-se a percentagem de explantes regenerados, o número de brotações por explante regenerante e tipo de organogênese formada. As duas cultivares estudadas apresentam potencial para a propagação in vitro, porém Adams foi mais responsivo, pois obteve-se 6,3 brotações por explante, com 2,8mg dm-3 de BAP, enquanto MC apresenta maior sensibilidade a altas concentrações deste regulador de crescimento e à vitrificação, porém demonstrou ser mais eficiente durante a regeneração in vitro, apresentando 7 valores aproximados de 26% de explantes regenerantes (utilizando 1,0mg dm-3 de TDZ). Para ambas cultivares, o explante folha inteira é mais responsivo à regeneração do que terços basais e que esta ocorre principalmente na região basal do explante, via organogênese direta ou indireta. Baseado nos resultados obtidos tanto na fase de multiplicação quanto na de regeneração, verifica-se que é possível multiplicar in vitro estas cultivares em larga escala, bem como melhorar as taxas de regeneração para futuros trabalhos de melhoramento para características de interesse, via transformação genética.

In order to evaluate the formation of adventitious buds and in vitro regeneration of sour orange plants (Citrus aurantium L.) two organogenesis-inducing experiments were conducted. In the first experiment, the induction and in vitro regeneration of adventitious buds were tested on epicotyl and internodal segments under the influence of BAP or KIN associated with NAA. The second experiment evaluated the in vitro regeneration of sour orange plants related to different explant types (epicotyls segments, internodal segments of in vitro germinated plantlets and internodal segments of greenhouse cultivated plants). Data collected on both experiments included the percentage of responsive explants (explants that formed buds), and the number of buds per explant. The addition of BAP showed the best organogenic response. In vitro germinated epicotyl segments and internodal segments are recommended as explants for sour orange in vitro organogenesis. Rooting of regenerated shoots was achieved without the need of auxin in the medium. Com o objetivo de avaliar a formação de gemas adventícias e regeneração in vitro de plantas de laranja-azeda (Citrus aurantium L.), foram realizados dois experimentos de indução à organogênese. No primeiro experimento, a indução e a regeneração in vitro de gemas adventícias foram investigadas a partir de segmentos internodais e segmentos de epicótilo sob o efeito de BAP ou CIN associados com ANA. O segundo experimento avaliou a regeneração in vitro de plantas de laranja-azeda em função do tipo de explante (segmentos de epicótilo, segmentos internodais de plantas germinadas in vitro e segmentos internodais de plantas cultivadas em casa de vegetação). Os dados coletados em ambos os experimentos incluíram a porcentagem de explantes responsivos (explantes que formaram gemas) e número de gemas por explante. A adição de BAP revelou a melhor resposta organogenética. Segmentos de epicótilo e segmentos internodais são explantes recomendados para a indução de organogênese in vitro de laranja-azeda. Enraizamento das brotações foi alcançado sem a adição de auxinas ao meio de cultura.

Hydrangea macrophylla is a ligneous plant that has attracted the attention of many plant breeders and agronomists for the purpose of enhancing its phenotypic plasticity. However, this plasticity was always exploited empirically. Can this plasticity be assessed by a more scientific approach? In this work, the phenotypic variation is analysed via a description of the different development sequences of the plant and by exposing the plant to different contrasted environments. The architectural unit consists of two morphogenetic units: the Vegetative Unit (VU) and the Vegetative and Floral Unit (VFU). They result in four successive development sequences: an organogenetic phase accompanied by continuous growth (sequence A), floral transformation (sequence B), dormancy (sequence C) and flower bloom (sequence D). Under the effect of environmental factors, the formation of the mixed terminal bud (sequence B) provides a considerable source of spatial variability, whereas the absence or presence of dormancy (sequence C) is responsible for a source of temporal variation. The in-depth description of the architectural unit with its morphological components and the characterisation of the four development sequences provide a necessary scientific basis to identify environmental effects on plant development and for the integrated use of its plasticity.; Hydrangea macrophylla est une plante ligneuse présentant une grande plasticité phénotypique qui a suscité l'intérêt de nombreux sélectionneurs et agronomes. Discos foliares de 0,5 cm de diâmetro, com a face adaxial em contato com o meio de cultura, foram introduzidos em placa de petri. Organogênese foi obtida utilizando-se meio MS acrescido de 0,5 ou 1,0 mg L-1 de BAP (6-benzilaminopurina). Após 4 a 8 semanas, observou-se formação de calos nas bordas dos discos foliares. Os calos formados foram transferidos para meio MSM, acrescido de 1,0 mg L-1 de GA3 (ácido giberélico), sob fotoperíodo de 16 horas. Desenvolvimento de gemas adventícias foi obtido a partir dos calos, sendo estas alongadas e enraizadas no mesmo meio de cultura, após periódicas repicagens, e aclimatadas em casa de vegetação.

Conselho Nacional de Desenvolvimento Científico e Tecnológico The present study was carried out to evaluate the effects of the growth regulators BAP and TDZ on the induction of adventicious shoots of the pigeon pea (cultivars "Anão" and "Fava Larga"). So, zygotic embryos and epicotyledonary segments from in vitro-germinated plantlets were used. In the shoot induction from embryonic axes, the following variables were evaluated at the end of 30 days under culture: the percent callus, the number of roots and shoots formed at both ends (proximal and distal), as well as the presence of contamination and explants without response. The formation of roots were restricted to the explants cultured in the MSØ medium and at the proximal end. In the explants cultured in the MS medium added with cytokinins, an intensive callus formation was generally observed at the proximal end. In both cultivars, the culture medium supplemented with combination of BAP and TDZ at concentrations 4.44μM and 2.27μM respectively, made possible the formation of higher shoot numbers by explant at distal end, but they were reducedsized shoots. Although the lower number of shoots in the explants cultured in mediums added with only BAP (2.22μM) or TDZ (2.27μM), they showed more expanded leaves and vigorous aspect. In shoot induction from the epicotyl segments aseptically obtained from plantlets aged 7 days, the number of roots and shoots formed at both ends were evaluated on the 32o day. The intensity, coloration and texture of the callus were visually evaluated and generally described. Again, the root formations were observed at the basal end of the explants cultured in MSØ medium, whereas a higher number of root formation by explant were observed, compared to the control medium without AgNO3. In both cultivars and both ends of the explants, the medium added with combination of two cytokinins (2.22μM BAP and 2.27μM TDZ) made possible the formation of callus that were visually higher, but at the presence of AgNO3 occurred lower callus formation. The shoot were restricted to basal end of the explants cultured in the medium added with cytokinins and AgNO3, and this combination made possible the formation of higher shoot number. Histological sections revealed the adventicious origin of the shoots via direct organogenesis in the embryonic explants, as well as indirect via in the epicotyl explants. According to the results, the following conclusions were drawn: the use of the embryonic axis, epicotyl segments and both BAP and TDZ cytokinins in inducing the adventicious shoots of the pigeon pea (cultivars "Anão" and "Fava Larga") is promising; the use of AgNO3 is necessary to induce the shoots from the epicotyl segments; and although the addition of precursors and the measurement of the ethylene levels have not been accomplished, the responses obtained in this study may be related to the effect of this hormone. Those data will be useful as base for the accomplishment of complementary studies that are targeted to the improvement of the efficiency in the in vitro regeneration of the pigeon pea, as well as the ex vitro elongation and acclimatization phases. O presente estudo objetivou avaliar os efeitos dos reguladores de crescimento BAP e TDZ na indução de brotações adventícias de feijão guandu (cultivares "Anão" e "Fava Larga") a partir de embriões zigóticos e segmentos de epicótilo de plântulas germinadas in vitro. Na indução de brotações a partir de eixos embrionários, ao final de 30 dias de cultivo foram avaliados o percentual de calejamento, o número de raízes e brotações formadas em ambas as extremidades (proximal e distal) bem como a presença de contaminação e de explantes sem reposta. A formação de raízes se manteve restrita aos explantes cultivados em meio MSØ e na extremidade proximal. Nos explantes cultivados em meio MS acrescido de citocininas, de maneira geral, independente da concentração de BAP e, ou, TDZ testadas, observou-se intensa formação de calo na extremidade proximal. Em ambos os cultivares o meio de cultura suplementado com a combinação de BAP e TDZ nas concentrações de 4,44 μM e 2,27 μM respectivamente, possibilitou a formação de maior número de brotações por explante na extremidade distal, mas essas apresentaram tamanho reduzido. Apesar de haver formação de menor número de brotações nos explantes cultivados em meios acrescidos de apenas BAP (2,22 μM) ou TDZ (2,27 μM), essas apresentaram folhas mais expandidas e aspecto vigoroso. Na indução de brotações a partir de segmentos de epicótilo obtidos assepticamente de plântulas com 7 dias de idade, avaliou-se ao final de 32 dias de cultivo o número de raízes e de brotações formadas em ambas as extremidades. A intensidade, coloração e textura dos calos foram avaliadas visualmente e descritas de forma geral. Novamente, a formação de raízes foi observada no extremo basal dos explantes cultivados em meio MSØ sendo que, na presença de AgNO3, observou-se a formação de maior número de raízes por explante comparado ao meio controle sem AgNO3. Em ambos os cultivares e em ambas as extremidades dos explantes, o meio acrescido da combinação das duas citocininas (2,22 μM de BAP e 2,27 μM de TDZ) possibilitou a formação de calos visualmente maiores, sendo que na presença de AgNO3 a formação de calo foi menor. As brotações se restringiram ao extremo basal dos explantes cultivados em meio acrescido de citocininas e AgNO3, sendo que esta combinação possibilitou a formação de maior número de brotações. Seções histológicas revelaram a origem adventícia das brotações via organogênese direta nos explantes embrionários e via indireta nos explantes do epicótilo. Conclui-se que a utilização do eixo embrionário, segmentos de epicótilo e das citocininas BAP e TDZ na indução de brotações adventícias de feijão guandu cvs. "Anão" e "Fava Larga" é promissora; a utilização de AgNO3 se faz necessário para a indução de brotações a partir de segmentos de epicótilo; e embora a adição de precursores bem como a mensuração dos níveis de etileno não tenham sido realizados, as respostas obtidas no presente estudo podem estar relacionadas ao efeito deste hormônio. Os resultados obtidos neste trabalho fornecem subsídeos a estudos complementares, necessários para o aperfeiçoamento da eficiência de regeneração in vitro de feijão guandu, bem como das fases de alongamento e aclimatização ex vitro.

Azadirachta indica A. Juss, popularmente conhecido como nim, é uma espécie arbórea que se destaca por possuir substâncias de ação inseticida, fungicida, bactericida e nematicida. Sementes de nim foram inoculadas em meio de cultura WPM (Wood Plant Medium) contendo diferentes concentrações de ácido giberélico (GA3) (0; 3,0; 6,0; 9,0; e 12,0 mg L-1). Após 30 dias, cotilédones obtidos a partir de plântulas germinadas in vitro foram inoculados em meio WPM suplementado com ácido 2,4-diclorofenoxiacético (2,4-D) (0,0; 1,0; 2,0; e 3,0 mg L-1) e, ou, 6-benzilaminopurina (BAP) (0,0; 1,0; 2,0; e 3,0 mg L-1). As culturas foram incubadas no escuro, a 28 ºC. Os calos formados foram avaliados com base na sua coloração e textura, e três subcultivos foram realizados mensalmente em meio WPM contendo 2,0 mg L-1 BAP, na presença de luz. A cada 30 dias, avaliou-se o número de brotos formados a partir dos calos subcultivados. Entre os meios testados, o mais apropriado para germinação in vitro de nim foi o WPM na ausência de GA3. Explantes cotiledonares cultivados em WPM suplementado com 2,0 mg L-1 de BAP promoveram a maior formação de calos com potencial morfogênico. Quando esses calos foram transferidos para meio de composição similar, obteve-se alta formação de brotações até o terceiro subcultivo. Azadirachta indica A. Seeds of neem were inoculated in WPM (Wood Plant Medium) containing different concentrations of gibberelic acid (GA3) (0, 3, 6, 9, and 12 mg L-1). After 30 days, cotyledons were inoculated in WPM supplemented with 0.0; 1.0; 2.0 and 3.0 mg L-1 acid 2,4-phenoxyacetic acid (2,4-D) and/or 0.0; 1.0; 2.0 and 3.0 mg L-1 6-benzylaminopurine (BAP). The cultures were kept in the dark at 28 ºC. Differentiated calli were evaluated based on their coloration and texture, and three subcultures were carried out on a monthly basis in WPM medium with 2.0 mg L-1 BAP, in the presence of light. Every 30 days, the number of differentiated shoot-buds was evaluated. WPM medium lacking GA3 promoted the highest in vitro seed germination indexes. Cotyledonary-derived explants cultured in WPM medium with 2.0 mg L-1 BAP yielded calli with high morphogenic potential. When those calli were transferred to medium with similar composition, a high shoot formation was achieved up to the third subculture. HERRERA, R.C. Dynamic models of grapevine development appear as promising tools to evaluate the relevance of training systems in order to improve both quality and quantity of production. Grapevine development is driven by different factors such as environment, genotype, and agronomic practices. The phenotypic plasticity is mainly controlled by an endogenous trait, the trophic competition between organs, which is determined by the other different factors. The development of the organs can be decomposed in three processes, the iniation (organogenesis), the expansion (morphogenesis) and the biomass growth. For grapevine, axis organogenesis was observed appear as the most plastic process. This study is a first step towards a predictive model, allowing to simulate the effects of different training systems and of contrasted environmental conditions on grapevine development. This work combines approaches based on ecophysiology and on mathematical modelling. The originality of this work was to use these two approaches together to take advantage of tools and concepts of both, and to fill in their respective deficiencies in order to model plant-environment interactions. GreenLab model and its associated optimization tools allowed to quantify precisely the source-sink relationships between organs and the trophic competition at the plant scale. The ecophysiological approach aims to input in GreenLab model new formalisms of grapevine organogenesis response to environmental constraints. In this way, a stochastic simulator of grapevine development in contrasted environments (temperature, water deficit, irradiance level) was built., La modélisation dynamique du développement de la vigne offre de nombreuses possibilités pour évaluer la pertinence des pratiques culturales pour répondre à des objectifs de production. Le développement de la vigne résulte de l’effet de différents facteurs, environnementaux, génotypiques et techniques lorsqu’elle est cultivée. Une des principales variables endogènes, résultant des effets combinés de ces trois facteurs et qui détermine en grande partie la plasticité phénotypique est la compétition trophique entre les organes. Le développement des organes peut-être décomposé en trois processus, l’initiation (organogenèse), l’expansion (morphogenèse), et la croissance en biomasse. Pour la vigne, l’organogenèse des axes est le processus de développement présentant la plus grande plasticité. Ce travail établit les bases pour la construction d’un modèle prédictif, permettant de simuler le développement de plantes de vignes soumises à une gamme de pratiques culturales et dans des environnements contrastés. Notre travail combine deux approches de modélisation, écophysiologique et mathématique. L’originalité de ce travail a été d’utiliser les deux approches de façon conjointe pour tirer parti des outils et des concepts de chacune et pour combler leurs lacunes respectives dans un objectif de modélisation des interactions plante-environnement. Le modèle GreenLab et les outils d’optimisation qui lui sont associés ont permis de quantifier précisément les relations sources-puits entre les organes et le niveau de compétition trophique qui en résulte à l’échelle de la plante. L’approche écophysiologique a permis d’établir de nouveaux formalismes de la réponse des processus d’organogénèse aux contraintes environnementales, et de les implémenter dans le modèle GreenLab. Un simulateur stochastique du développement de la vigne dans des environnements variés (thermiques, hydriques, lumineux) a ainsi pu être développé.

(Microanatomia larval do peixe-rei marinho Odontesthes argentinensis do Rio Grande do Sul – Brasil) Larvas de Odontesthes argentinensis foram cultivadas sob condições controladas em tanques de 15 litros (temperatura: 21-22,5ºC; salinidade: 30-35‰; fotoperíodo 13h claro:11h escuro; aeração e renovação da água constantes; densidade: sete larvas/litro e alimentação: nauplii de Artemia spp.) na Estação Marinha de Aquicultura (EMA) da Universidade Federal do Rio Grande (FURG) para descrição microanatômica e funcional do sistema digestório entre a eclosão e o 30º dia de vida. Amostragens histológicas e biometrias (Regressão Linear: comprimento-idade e peso-idade) respeitaram as idades: recém-eclodidos (zero dias=0D), 1D, 3D, 6D, 12D, 16D, 20D e 30D pós-eclosão com anestesia com MS-222. Para histologia foram fixadas por 12h (0D≥idade≤20D) e por 24h (idade=30D) em líquido de Bouin, submetidas ao protocolo de inclusão em parafina para montagem permanente em Bálsamo do Canadá, coloração com Hematoxilina-Eosina e microtomia de 7μm em orientação sagital. Ao 0D a espécie possui: sistema nervoso e tecido muscular estriado esquelético (miomérico) bem diferenciados; boca funcional; tubo digestório com luz contínua e em diferenciação acelerada (concomitantemente com a rápida formação hepática e pancreática) o que permite uma nutrição mista (endógena e exógena); não há orifício anal ainda formado; cavidade faringiana portadora de brânquias funcionais; vitelo grande e ocupando a maior parte da cavidade visceral. Larvas de 12D apresentam todos os sistemas de órgãos não-gametogênicos caracterizados. Até o 30D O. argentinensis não apresenta estômago e cecos pilóricos. A salinidade parece ser um fator importante para a sobrevivência larval mesmo com o tegumento, o sistema respiratório e o sistema urinário funcionais, o que implica em mortalidade multifatorial. To histology were fixed by 12h (0D≥age≤20D) and 24h (age=30D) in Bouin’s fluid to embedding in paraffin and permanent mounting in balsam, staining with Hematoxylin-Eosin under 7μm thickness microtomia in sagittal cut plane. At 0D life the species show: nervous system and striated skeletal muscle tissue (myomera) well differentiated; functional mouth; digestory tube in fast differentiation with a continuous lumen (together with fast hepatic and pancreatic formation) what permits a mixed nutrition (exogenous and endogenous); there is no anus formed; the pharyngeal cavity possess functional gills; the yolk sac is great and inhabit the greatest portion of the visceral cavity. 12D larvae show all internal no-gametal organ systems characterized. Until 30D O. argentinensis no develops stomach and pyloric ceca. The salinity can be an important factor to larval surviving although the tegument is present, respiratory and urinary systems were functional, what can denote a multifactorial mortality.

The Tapeinochilos ananassae (Hassk) K. Schum is a tropical species that has a great acceptance in the world of floriculture market and presents, in the Brazilian Northeast, fitossanitary problems in which the Rhabdovirus is highlighted. In the present work, aiming to assess the establishment and development of various explants in the in vitro culture of T. ananassae, including the study of biochemical and anatomical variables. For this, lateral buds and mature zygotic embryos were inoculated in MS medium, full or reduced to its half ionic strength, using three sources of antioxidants: ascorbic acid, activated charcoal and PVP, in the concentrations 0,25g.L-1; 3,0g.L-1 and 0,5g. L-1. The ½MS culture medium supplemented with 3,0g. L-1 activated charcoal promoted a higher rate of success in the establishment of zygotic embryos. The plants formed from the culture of embryos were propagated by nodal segments in different concentrations of BAP and GA3: ½MS; ½MS + 0,5mg.L-1 GA3; ½MS + 1,0mg.L-1 GA3; ½MS + 0,5mg.L-1 BAP; ½MS + 0,5mg.L-1 BAP + 0,5mg.L-1 GA3; ½MS + 0,5mg.L-1 BAP + 1,0mg.L-1 GA3; and showed better development when cultivated in ½MS medium free from growth regulators. The lateral buds had not shown morfogenetic development under the conditions of study, besides a large phenolic oxidation had been seen, regardless of the culture medium that had been used, and a high level of microbial contamination, especially bacterial, when buds that had come from basal branches are used. In the enzymatic analysis of lateral buds that were cultivated in vitro in a process of oxidation, had been realized that the activity of peroxidase had declined through the time, suggesting the loss of organogenetic potential, but the activity of polyphenoloxidase had tended to increase during the oxidation process of explants. O cultivo in vitro é uma ferramenta de grande importância na produção de mudas com qualidade fitossanitária. O Tapeinochilos ananassae (Hassk) K. Schum é uma espécie tropical com grande aceitação no mercado floricultor mundial. No Nordeste brasileiro, vem enfrentando sérios problemas fitossanitários causados principalmente pelo Rhabdovirus. O presente trabalho teve como objetivo avaliar o estabelecimento e desenvolvimento de diferentes explantes no cultivo in vitro de T. ananassae, incluindo o estudo de variáveis bioquímicas e anatômicas. Para isso, gemas laterais e embriões zigóticos maduros foram inoculados em meio MS completo ou reduzido à metade da força iônica, utilizando três fontes de antioxidantes: ácido ascórbico, carvão ativado e PVP, nas concentrações 0,25g L-1; 3,0g L-1 e 0,5g L-1. O meio de cultivo ½MS suplementado com 3,0g. L-1 de carvão ativado promoveu um maior índice de sucesso no estabelecimento de embriões zigóticos. As plantas formadas a partir da cultura de embriões foram propagadas por segmentos nodais em diferentes concentrações de BAP e GA3: ½MS; ½MS+0,5mg.L-1 GA3; ½MS+1,0mg. L-1 GA3; ½MS+0,5mg.L-1 BAP; ½MS+0,5mg.L-1 BAP+0,5mg.L-1 GA3; ½MS+0,5mg.L-1 BAP+1,0mg.L-1 GA3. O meio de cultivo constituído de ½ dos sais MS e isento de reguladores de crescimento mostrou-se mais adequado para o cultivo de segmentos nodais de T. ananassae. As gemas laterais não apresentaram desenvolvimento morfogênico nas condições de estudo deste trabalho. Além disso, foi observado grande oxidação fenólica, independentemente do meio de cultivo utilizado, e elevado índice de contaminação microbiana, sobretudo bacteriana, quando do uso de gemas advindas de ramos basais. Nas análises enzimáticas das gemas laterais cultivadas in vitro em processo de oxidação, percebeu-se que a atividade da peroxidase decaiu ao longo do tempo, sugerindo a perda do potencial organogenético; já a atividade da polifenoloxidase tendeu a aumentar ao longo do processo de oxidação dos explantes.

Primary lymphoid follicles are structures which are important for adaptive immune responses in mammals. Within the follicles follicular dendritic cells (FDC) are maintained by constant stimuli provided by B cells. It is thought that the FDC are important for immune response. It is of interest to know how lymphoid follicles are regulated in order to understand their role in various autoimmune diseases in which these follicles are created ectopically. With the help of a tissue simulation relying on an agent-based cell model on top of a regular triangulation various scenarios suggested by the available experimental data have been investigated. In order to cope with the complexity in the simulation of immune tissue the regular triangulation has been implemented for the use on parallel computers. The algorithms for kinetic and dynamic regular triangulation have been created newly. Also the cell model underlying the simulation has been designed newly in many aspects. The simulations allowed to identify common factors that regulate the formation of lymphoid follicles normally during organogenesis in development and lymphneogenesis in the course of diseases. The generation of FDC from local stromal populations under the influence of B cell aggregates is shown to be possible with the given experimental parameters. The sequence of the organogenesis and lymphneogenesis can be described with regard to the morphology of the B and T zone. Tests for the stability of the primary lymphoid follicle system constraints the regulation of the B cell efflux. The required lymphatic vessels around the lymphoid follicle are shown to be negatively correlated with the FDC network. Moreover it is shown that the adjacent T zone consisting of its own stromal population and T cells has similar regulation principles. This easily explains the intermediate ring of B cells found around the T zone during development and certain signaling molecule deficiencies. A major result of this thesis is that the generation of FDC needs negative regulation while a number of other possible mechanisms is incompatible with the available experimental data. Moreover the observed microanatomy was brought into a functional relationship with data on the cellular level finally culminating in the proposal of new experiments that shed light on the dynamics of the primary lymphoid follicle. One conclusion is that the FDC directly or indirectly influence the angiogenesis and lymphangiogenesis processes in secondary lymphoid tissues. The work presented here may help to guide experiments with the help of computers in order to reduce the amount of experiments and design them in a way to maximize the amount of information about biological systems. In Säugetieren spielen primäre lymphoide Follikel eine wichtige Rolle bei der adaptiven Immunantwort. Innerhalb der Follikel werden follikuläre dendritische Zellen (FDZ) durch eine konstante Stimulation durch B-Zellen generiert und erhalten. Diese FDZ-Population is wichtig für bestimmte Immunreaktionen. Die Erzeugung und Regulierung der Follikel is interessant insbesondere im Hinblick auf ihre ektopische Erzeugung im Laufe von Autoimmunreaktionen. Mit Hilfe von Gewebesimulationen ausgehend von einem Individual-basierten Zellmodells, das auf einer Delaunay-Triangulation aufbaut, wurden verschiedene aus Experimenten generierter Szenarien untersucht. Zur Lösung des Komplexitäts-Problems der vielen zu simulierende Zellen des Gewebes wurde größtenteils neue Algorithmen zur parallelen Verarbeitung der Kinetik und Dynamik der Delaunay-Triangulation erstellt. Das darauf aufbauende Zellmodell ist ebenfalls der parallelen Verarbeitung angepaßt. Die Simulationen führten zur Identifizierung gemeinsamer Zusammenhänge der Regulation lymphoider Follikel währen der Organogenese und der krankheitsbedingten Lymphneogenese. Die Erzeugung von FDZ aus lokalen Populationen stromaler Zellen durch Aggregate von B-Lypmozyten ist im Bereich der experimentellen Parameter möglich. Die Sequenz der Musterbildung von B- und T-Zone des Follikels während Organogenese und Lymphneogenese kann simuliert werden. Stabilitätskriterien für das primäre lymphoide Follikel schränken die möglichen Mechanismen der Regulation des B-Lymphozyten-Efflux ein. Die dafür notwendigen lymphatischen Gefäße um das Follikel sind negativ mit den FDZ korreliert, um eine Position um das Follikel herum einzunehmen. Ebenso wird gezeigt, das die spezifischen stromalen Populationen der T-Zone zusammen mit den T-Lymphozyten ähnlichen Organisationsprinzipien gehorchen, wie das benachbarte Follikel. Dies erklärt auf einfache Weise, das Auftreten einer charakteristischen intermediären Morphologie des Follikels als B-Lymphozyten-Ring um die T-Zone sowohl während der normalen Entwicklung als auch als Resultat bestimmter Signalmolekül-Defizite. Ein zentrales Ergebnis dieser Arbeit ist, dass die Erzeugung der FDZ einer negativen Regulation unterliegen muss, während eine Reihe anderer denkbarer Mechanismen nicht mit den verfügbaren experimentellen Daten in Einklang zu bringen ist. Darüber hinaus konnten Morphologie und Funktion mit Daten der zellulären Ebene in Beziehung gesetzt werden. Daraus sind neue Experimente zur Dynamik primärer lymphoider Follikel abgeleitet worden. Ein Beispiel hierfü ist der Einfluss der FDZ auf die Angiogenese und Lymphangiogenese in sekundären lymphoiden Geweben. Die hier aufgeführte Arbeit stellt so einen Beitrag dar, Experimente mit Hilfe von Computer Simulationen zu entwerfen, um sowohl die Zahl der notwendigen Experimente zu reduzieren als auch deren Informationsgewinn über ein biologisches System zu maximieren.
