Abstract

Spinal muscular atrophy (SMA) is a neuromuscular disorder caused by loss of survival motor neuron (SMN) protein. While SMN restoration therapies are beneficial, they are not a cure. We aimed to identify potentially novel treatments to alleviate muscle pathology combining transcriptomics, proteomics, and perturbational data sets. This revealed potential drug candidates for repurposing in SMA. One of the candidates, harmine, was further investigated in cell and animal models, improving multiple disease phenotypes, including lifespan, weight, and key molecular networks in skeletal muscle. Our work highlights the potential of multiple and parallel data-driven approaches for the development of potentially novel treatments for use in combination with SMN restoration therapies.

Abstract

Due to constantly increasing traffic loads and ageing infrastructure, innovative materials for implementation in renovation and new bridge construction projects are gaining attention. One such innovation is hybrid structures consisting of Glass Fibre-Reinforced Polymer (GFRP) composite decks connected to a steel girder(s) superstructure. By combining the stiffness provided by steel members with the superior fatigue endurance and strength to weight ratio of FRP, the properties of both materials can be efficiently exploited. The main restriction to the implementation of hybrid steel-FRP structures lies in the lack of knowledge and efficient technical solutions for the connection between the two materials. Adhesively bonded and grouted connections have been identified as possible connection systems, whereas limited research has been performed on bolted connections. To address this research gap, this paper discusses an experimental investigation into the performance of two types of slip-resistant bolted connectors, namely conventional resin injected bolts and a novel connector system developed at TU Delft comprising of a bolted connector injected with steel reinforced resin (iSRR). Connectors’ performance is evaluated by means of long-term (sustained and cyclic) and short-term experiments with shear load. A single-lap shear joint configuration is used to compare performance of the two connector types. A vacuum infused GFRP multi-directional laminated plate, 20 mm thick, is connected to steel plates by means of M20 bolts. Results indicate comparable stiffness, resistance and ductility of the two connector types in short-term experiments. Superior fatigue performance of iSRR connector is found with 100 times more fully reversed load cycles that are needed to reach the same failure criterion as the conventional injected bolts., Steel & Composite Structures

Abstract

As traffic loads are ever increasing and bridge infrastructure is ageing, existing materials and connection methods used in renovation and new construction projects are reaching their limits. New materials, such as Fibre-Reinforced Polymer (FRP) in the form of hollow decks can be a competitive solution due to good resistance to fatigue and corrosion. By combining the stiffness provided by steel main and secondary girders with lightweight FRP decks, the properties of both materials can be efficiently utilised. However, the main restriction to the implementation of hybrid steel-FRP structures lies in a lack of knowledge regarding the level of interaction as well as efficient and durable connection solutions of FRP decks and steel girders. Adhesively bonded and grouted connections have been identified as possible connection systems, whereas limited research has been performed on bolted connections. This paper focuses on blind-bolted shear connectors. Connector performance is evaluated experimentally by means of short-term (static) experiments to determine shear resistance and long-term experiments with sustained (creep) and cyclic (fatigue) loading. A vacuum infused GFRP multi-directional laminated plate, 20 mm thick, is connected to steel plates by means of M20 blind-bolts. Static resistance, fatigue and creep behaviour of blind-bolted connectors, as a bearing type, are compared to results of a parallel experimental campaign conducted on slip-resistant injected connectors. A comparable static resistance and ductility is found. However, fatigue endurance is much lower compared to slip-resistant connectors. Therefore, due to low fatigue endurance, low initial stiffness and initial slip, the use of blind-bolted connectors is limited to applications where hybrid interaction and fatigue endurance is not required., Steel & Composite Structures

Abstract

Adenomatoid tumors are benign tumors of mesothelial origin that are usually encountered in the genital tract. Although they have been observed in other organs, the skin appears to be a very rare location, with only one case reported in the literature to our knowledge. We report a second case of an adenomatoid tumor, arising in the umbilicus of a 44-year-old woman. The patient presented with an 8-month-old erythematous and firm plaque under the umbilicus. A skin biopsy showed numerous microcystic spaces dissecting a fibrous stroma and lined by flattened to cuboidal cells with focal intraluminal papillary formation. This little-known diagnosis constitutes a diagnostic pitfall for dermatopathologists and dermatologists, and could be misdiagnosed as other benign or malignant entities. Abstract

With continuing improvements in spatial resolution of positron emission tomography (PET) scanners, small patient movements during PET imaging become a significant source of resolution degradation. This work develops and investigates a comprehensive formalism for accurate motion-compensated reconstruction which at the same time is very feasible in the context of high-resolution PET. In particular, this paper proposes an effective method to incorporate presence of scattered and random coincidences in the context of motion (which is similarly applicable to various other motion correction schemes). The overall reconstruction framework takes into consideration missing projection data which are not detected due to motion, and additionally, incorporates information from all detected events, including those which fall outside the field-of-view following motion correction. The proposed approach has been extensively validated using phantom experiments as well as realistic simulations of a new mathematical brain phantom developed in this work, and the results for a dynamic patient study are also presented. Abstract

Low-density lipoprotein (LDL)-mimetic lipid nanoparticles (LNPs), decorated with MRI contrast agents and fluorescent dyes, were prepared by the covalent attachment of apolipoprotein-mimetic peptide (P), Gd(iii)-chelate (Gd), and sulforhodamine B (R) moieties on the LNP surface. The functionalized LNPs were prepared using the amide-forming potassium acyltrifluoroborate (KAT) ligation reaction. The KAT groups on the surface of LNPs were allowed to react with the corresponding hydroxylamine (HA) derivatives of P and Gd to provide bi-functionalized LNPs (PGd-LNP). The reaction proceeded with excellent yields, as observed by ICP-MS (for B and Gd amounts) and MALDI-TOF-MS data, and did not alter the morphology of the LNPs (mean diameter: ca. 50 nm), as shown by DLS and cryoTEM analyses. With the help of the efficient KAT ligation, a high payload of Gd(iii)-chelate on the PGd-LNP surface (ca. 2800 Gd atoms per LNP) was successfully achieved and provided a high r1 relaxivity (r1 = 22.0 s−1 mM−1 at 1.4 T/60 MHz and 25 °C; r1 = 8.2 s−1 mM−1 at 9.4 T/400 MHz and 37 °C). This bi-functionalized PGd-LNP was administered to three atherosclerotic apoE−/− mice to reveal the clear enhancement of atherosclerotic plaques in the brachiocephalic artery (BA) by MRI, in good agreement with the high accumulation of Gd in the aortic arch as shown by ICP-MS. The parallel in vivo MRI and ex vivo studies of whole mouse cryo-imaging were performed using triply functionalized LNPs with P, Gd, and R (PGdR-LNP). The clear presence of atherosclerotic plaques in BA was observed by ex vivo bright field cryo-imaging, and they were also observed by high emission fluorescent imaging. These directly corresponded to the enhanced tissue in the in vivo MRI of the identical mouse., LDL-mimetic lipid nanoparticles, decorated with MRI contrast agents and fluorescent dyes, were prepared by the covalent attachments of an apoB100-mimetic peptide, Gd(iii)-chelate, and rhodamine to enhance atherosclerosis in the in vivo imaging.

Abstract

The Oak Ridge National Laboratory (ORNL), in collaboration with Eaton Corporation, has performed computational research and development to design an innovative, direct-contact heat exchanger (DCHE) that is optimized for a low-temperature organic Rankine cycle. A computational fluid dynamics (CFD) model of DCHE was developed in STAR-CCM+ which was later calibrated and validated against the experimental data from literature. The validated CFD model was used to develop an industry-relevant liquid-liquid direct-contact heat exchanger system with water and pentane working fluids. This work heavily relied on high-performance computing (HPC) resources to investigate multiple designs and to identify a baseline design. The innovative design consists of two chambers connected by a converging-diverging nozzle. Phase change for pentane, from liquid to vapor, occurs in the first chamber, whereas the second chamber serves as a separator. Outlets in the second chamber are staggered to prevent entrainment of the liquid water by the gaseous pentane. CFD results confirm that the design behaves as expected and the addition of baffles enhances mixing and heat transfer for higher flow rates while preventing entrainment of gaseous pentane by the liquid water.

Abstract

Extracellular vesicles (EVs) form an endogenous transport system for intercellular transfer of biological cargo, including RNA, that plays a pivotal role in physiological and pathological processes. Unfortunately, whereas biological effects of EV-mediated RNA transfer are abundantly studied, regulatory pathways and mechanisms remain poorly defined due to a lack of suitable readout systems. Here, we describe a highly-sensitive CRISPR-Cas9-based reporter system that allows direct functional study of EV-mediated transfer of small non-coding RNA molecules at single-cell resolution. Using this CRISPR operated stoplight system for functional intercellular RNA exchange (CROSS-FIRE) we uncover various genes involved in EV subtype biogenesis that play a regulatory role in RNA transfer. Moreover we identify multiple genes involved in endocytosis and intracellular membrane trafficking that strongly regulate EV-mediated functional RNA delivery. Altogether, this approach allows the elucidation of regulatory mechanisms in EV-mediated RNA transfer at the level of EV biogenesis, endocytosis, intracellular trafficking, and RNA delivery.

Abstract

Spinal muscular atrophy (SMA) is a neuromuscular disorder caused by loss of survival motor neuron (SMN) protein. While SMN restoration therapies are beneficial, they are not a cure. We aimed to identify novel treatments to alleviate muscle pathology combining transcriptomics, proteomics and perturbational datasets. This revealed potential drug candidates for repurposing in SMA. One of the lead candidates, harmine, was further investigated in cell and animal models, improving multiple disease phenotypes, including SMN expression and lifespan. Our work highlights the potential of multiple, parallel data driven approaches for development of novel treatments for use in combination with SMN restoration therapies.

Abstract

Methanol represents an attractive substrate for biotechnological applications. Utilization of reduced one-carbon compounds for growth is currently limited to methylotrophic organisms, and engineering synthetic methylotrophy remains a major challenge. Here we apply an in silico-guided multiple knockout approach to engineer a methanol-essential Escherichia coli strain, which contains the ribulose monophosphate cycle for methanol assimilation. Methanol conversion to biomass was stoichiometrically coupled to the metabolization of gluconate and the designed strain was subjected to laboratory evolution experiments. Evolved strains incorporate up to 24% methanol into core metabolites under a co-consumption regime and utilize methanol at rates comparable to natural methylotrophs. Genome sequencing reveals mutations in genes coding for glutathione-dependent formaldehyde oxidation (frmA), NAD(H) homeostasis/biosynthesis (nadR), phosphopentomutase (deoB), and gluconate metabolism (gntR).
