Interference with TET enzymes by chelating ferrous ions can contribute to the altered methylation pattern. All tested cannabinoids displayed a strong affinity for Fe(II) ions. Cannabidiol and cannabinol exhibited potent inhibitory activities (IC 50 = 4.8 and 6.27 μM, respectively) towards the TET1 protein, whereas cannabigerol had no effect on the enzyme activity. An in silico molecular docking study revealed marked binding potential within the catalytic cavity for CBD/CBN, but some affinity was also found for CBG, thus the total lack of activity remains unexplained. These results imply that cannabinoids could affect the activity of the TET1 protein not only due to their affinity for Fe(II) but also due to other types of interactions (e.g., hydrophobic interactions and hydrogen bonding). A computational study was carried out to identify the structural determinant controlling the affinity, specificity and binding strength of several saturated and unsaturated fatty acids with Oryza sativa (Indica group) nonspecific lipid transfer protein (nsLTP2). Association between the number, position and conformation of hydrophobic patches and lipid binding properties of the protein was evidenced by docking analysis. Binding affinity is influenced by the number of carbon atoms, location of double bonds and hydroxyl group in the acyl chain. In the present study, the enzymatic and non-enzymatic antioxidants were evaluated in P. atriplicifolia , and P. abrotanoides in response to water deficit stress. The root harvesting was performed after 0, 3, 5, 7, and 9 days exposure to stress. In both species, total phenolics content, total flavonoids, enzymatic antioxidant activities, total tanshinones, tanshinone IIA, cryptotanshinone, hydroxy-cryptotanshinone, and proline substantially increased with the severity of stress. In the current study, the variations in tanshinone profile and the expression of genes (DXS1, HMGR, KSL) related to tanshinones biosynthesis were also investigated. The results revealed the negative trend between the concentration of malondialdehyde, and hydrogen peroxide as well as the harmonized augmentation in activities of enzymatic antioxidants including guaiacol peroxidase, and ascorbate peroxidase, exclusively on 7th day of stress. The expression ratios of all studied genes were markedly induced by the drought stress. The content of major oxidative stress-related indicators suggested that P. atriplicifolia should be better pre-adapted to water deficit physiological tolerance than P. abrotanoides. Finally, the experimental results can provide further insight into differential responses to imposed water-limited stress along with choice of the optimal eustress treatment to obtain the desired composition of pharmacologically useful diterpenoids. The accumulation of highly relevant bioactive phenylethanoid and iridoid glycosides produced in Rehmannia elata N.E. Brown ex Prein. in vitro shoot cultures was evaluated after culturing plant material under different light quality. Shoots of R. elata were cultured on Murashige and Skoog (MS) agar media without plant growth regulators (PGRs) or supplemented with 0.1 mg/L indole-3-acetic acid (IAA) and 0.8 mg/L N6-(2-isopentenyl)adenine (2iP) in the presence of white or photosynthetically active radiation (PAR) LED spectra. Ultrastructure of organogenic callus tissue, shoots and roots was observed using scanning electron microscopy (SEM). The highest number of regenerated shoots (4.35) were obtained on media supplemented with PGRs under PAR illumination. Phytochemical analysis of selected metabolites were determined using UHPLC technique. One of the iridoid glycoside, harpagide was detected at significantly higher level in shoots cultured under white light compared to PAR (0.66–1.08 mg/g DW and 0.27−0.29 mg/g DW, respectively), whereas larger contents of phenylethanoid glycoside - verbascoside (17.17–22.25 mg/g DW) were found in shoots cultured on PGRs supplemented media under both, white or PAR light conditions. Due to the presence of the biologically active R. elata metabolites, antimicrobial potential of methanolic extracts from shoot cultures against Gram-positive, Gram-negative bacteria and fungi as well as their cytotoxicity were evaluated. (sea knotgrass) as a promising source of ingredients for the cosmetic, food, pharmaceutical and veterinarian industries. To further explore this species, and to guarantee the supply of biomass for commercial purposes, it is necessary to ensure its sustainable production while assuring the preservation of its chemical and biological properties. In this context, this work aimed to cultivate this species in greenhouse conditions and to determine the influence of specific agronomic conditions, namely irrigation salinity and harvest, on the plant's growth performance and biological properties of obtained biomass. For that purpose, plants were grown in a greenhouse and irrigated with water with different salinities (0, 100, 200, 300 and 600 mM of sodium chloride - NaCl). After six weeks of irrigation, plants were cut 7 cm above soil level and submitted to an additional two harvests with a six-week interval. Plant growth performance was evaluated in terms of plant height, leaf number and surface area, moisture, and productivity. Acetone extracts were prepared from aboveground organs and evaluated for chemical composition (by spectrophotometric methods, and by ultra-high-resolution mass spectrometry - UHRMS), and for in vitro antioxidant properties [radical-scavenging activity (RSA) on DPPH and ABTS, ferric reducing antioxidant power (FRAP) and metal chelating activity on iron (ICA) and copper (CCA)]. Extracts were also appraised for in vitro anti-inflammatory activity on lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Increased salinity and multiple harvests reduced plant growth and yields; the highest productivity was obtained in plants irrigated with freshwater at 2nd harvest (346 g/m2 of dried biomass). The extracts maintained the in vitro biological properties and interesting chemical profile, however, these depended on the irrigation salinity and harvest regime. The highest antioxidant activities were obtained in extracts from plants irrigated with freshwater at the 3rd harvest (DPPH: 96.2%; ABTS: 89.1%; CCA: 61.6%; FRAP: 136%). The extracts from biomass obtained in the 1st harvest of freshwater-irrigated plants, and from those treated with 100 and 200 mM of NaCl, had significant anti-inflammatory properties. The main compounds detected were mostly flavonols (myricetin and quercetin glycosides), which varied according to both irrigation salinity and harvest. Our results indicate that sea knotgrass can be cultivated in greenhouse conditions aiming industrial commercial applications, irrigated with freshwater or with irrigation solutions with moderate salinity. This study aims to explore the effect of pulsed electric field (PEF) treatment as a method very likely to result in reversible electroporation of Scutellaria baicalensis Georgi underground organs, resulting in increased mass transfer and secondary metabolites leakage. PEF treatment with previously established empirically tailored parameters [E = 0.3 kV/cm (U = 3 kV, d = 10 cm), t = 50 µs, N = 33 f = 1 Hz] was applied 1-3 times to S. baicalensis roots submerged in four different Natural Deep Eutectic Solvents (NADES) media (1-choline chloride/xylose (1:2) + 30% water, 2-choline chloride/glucose (1:2) + 30% water, 3-choline chloride/ethylene glycol (1:2), and 4-tap water (EC = 0.7 mS/cm). Confocal microscopy was utilized to visualize the impact of PEF treatment on the root cells in situ. As a result of plant cell membrane permeabilization, an extract containing major active metabolites was successfully acquired in most media, achieving the best results using medium 1 and repeating the PEF treatment twice (baicalein wogonin = 2.15 µg/mL). After successful extraction, PEF treatment allowed the plants to continue growing, with the lowest survival rate across treated groups being 60%. Additionally, an enhancement in plant growth parameters (length and fresh mass of the roots) and significant changes in the S. baicalensis root phytochemical profile were also observed.

Background/Objectives: Basella alba L. (Malabar spinach, Basellaceae), widely consumed as a leafy vegetable, produces dark-colored fruits rich in betacyanins, including rare 6-glycosylated derivatives called gomphrenins. Comprehensive studies on the anti-inflammatory potential of its gomphrenin fraction (A) and crude extract (B) employed various analytical and biological methods. Methods: Cytotoxicity and anti-inflammatory effects were tested on human and animal cell models using SRB, DCF-DA, Griess, MDA, and ELISA assays. Results: Both the crude extract and enriched gomphrenin fraction exhibited significant anti-inflammatory and antioxidant effects in vitro. They inhibited pro-inflammatory cytokines IL-1β and IL-6, reduced oxidative stress markers (ROS, NO), and decreased lipid peroxidation. The enriched gomphrenin fraction (A) showed stronger antioxidant and anti-inflammatory effects, particularly in reducing ROS and NO levels, though not always concentration-dependent. Both A and B inhibited NF-κB and COX activity. Conclusions: These findings highlight the therapeutic potential of B. alba f. rubra fruit extract and betalain fraction for inflammation-related conditions, encouraging further exploration of their mechanisms and application.

The aim of this study was to develop and optimize polymeric films based on cellulose derivatives-hydroxypropylmethylcellulose (HPMC), methylcellulose (MC), and sodium carboxymethylcellulose (NaCMC)-as well as pullulan, polyvinyl alcohol (PVA), polyvinylpyrrolidone (PVP), and glycerol (GLY) as plasticizer incorporating Reynoutria japonica extract for potential use in periodontal and gum disease treatment. Over 80 formulations were fabricated using the solvent-casting method, 6 of which were selected for further investigation based on their mechanical properties, mucoadhesion, and disintegration profiles, including three placebo films (OP1 (PVA/PVP/MC400CP/NaCMC/GLY), OP2 (PVA/PVP/MCA15C/NaCMC/GLY), and OP3 (PVA/PVP/HPMC/NaCMC/GLY)) and three films containing R. japonica extract (OW1, OW2, and OW3). The films demonstrated uniform structural characteristics, with the formulations containing PVA with a high hydrolysis degree (98-99%) and methylcellulose derivatives showing prolonged dissolution times due to physical cross-linking, while the inclusion of NaCMC reduced dissolution time without compromising mucoadhesiveness. The study also described the release kinetics of resveratrol and piceid from the OW2 films using three semi-empirical models: the Korsmeyer-Peppas model, a first-order kinetic model, and a multidimensional approach. The multidimensional model demonstrated a strong fit, with a correlation coefficient (R 2 ) of 0.909 for resveratrol, compared to 0.894 and 0.908 for the Korsmeyer-Peppas and first-order models, respectively. For piceid, the multidimensional model showed a correlation coefficient (R 2 ) of 0.958, outperforming the Korsmeyer-Peppas (0.823) and first-order models (0.932). The active compounds released in sustained-release tests, including resveratrol and piceid, suggest that these films could provide an extended therapeutic effect.

Scutellaria baicalensis Georgi is a valuable medicinal plant of the Lamiaceae family. Its roots have been used in Traditional Chinese Medicine (under the name Huang-qin) since antiquity and are nowadays included in Chinese and European Pharmacopoeias. It is abundant in bioactive compounds which constitute up to 20% of dried root mass. These substances are lipophilic flavones with unsubstituted B-ring, baicalein, and wogonin and their respective glucuronides-baicalin and wogonoside being the most abundant. The content of these compounds is variable and the environmental factors causing this remain partially unknown. The role of these compounds in stress response is still being investigated and in our efforts to measure the effect of NaCl treatment on S. baicalensis growth and metabolic profile, we hope to contribute to this research. Short-term exposure to salt stress (50, 100, and 150 mM NaCl) resulted in a marked increase of baicalein from 1.55 mg to 2.55 mg/g DM (1.6-fold), baicalin from 8.2 mg to 14.7 mg (1.8-fold), wogonin from 4.9 to 6.8 (1.4-fold), and wogonoside from 3.3 to 6.8 mg/g DM (2-fold) in the roots. Conversely, in the aerial parts, the content of individual major flavonoids: carthamidine-7-O-glucuronide and scutellarein-7-O-glucuronide decreased the most by 10-50% from 18.6 mg to 11.3 mg/g (1.6-fold less) and from 6.5 mg to 3.4 mg/g DM (0.52-fold less), respectively. The amino acid profile was also altered with an increase in root concentrations of the following amino acids: arginine from 0.19 to 0.33 mg/g (1.7-fold), glutamate from 0.09 to 0.16 mg/g DM (1.6-fold), alanine from 0.009 to 0.06 mg/g (6.8-fold), proline from 0.011 to 0.029 (2.4-fold) and lysine from 0.016 to 0.063 mg/g (3.9-fold). Aspartate concentration decreased from 0.01 to 0.002 mg/g (4.8-fold less) at 150 mM NaCl. In the aerial parts, the concentration and variation in levels of specific amino acids differed among groups. For instance, the glutamate content exhibited a significant increase exclusively in the treatment group, rising from 0.031 to 0.034 mg/g, representing a 1.2-fold increase. Proline concentration showed a marked increase across all treated groups with the highest from 0.011 to 0.11 mg/g (10-fold). The family Caryophyllaceae comprises more than 2600 species spread widely across all the continents. Their economic importance is mainly as ornamentals (carnation) and as weeds in agriculture. Some species have been used traditionally (and some are still) in herbal medicine or as emulsifiers in food processing. These applications are based on the high content of triterpenoid saponins. Typical for this family are also ribosome-inactivating proteins (RIPs), which are potentially highly toxic. Agrostemma githago L. (common corncockle) was historically considered a serious toxicological hazard owing to cereal grain contamination by its seeds. Notwithstanding, it was also recommended as a drug by various herbalists. In this review, the literature was searched in the PubMed, Google Scholar, and Scopus databases for papers focused on the chemical composition and bioactivity of the two accepted species of the Agrostemma genus. This systematic review adhered to the Preferred Reporting Items for Systematic Reviews and MetaAnalysis (PRISMA) guidelines. Current research reports the cytotoxicity against neoplastic cells; the protection against oxidative stress; the suppression of Leishmania major culture growth; the inhibition of protein synthesis; and the antiviral, anti-angiogenic, and antihypercholesterolemic activities of common corncockle. The future prospects of using A. githago saponins as adjuvants in drug formulations and enhancing the cytotoxicity of RIPs are also discussed.

A comprehensive study of the interactions of human serum albumin (HSA) and α-1-acid glycoprotein (AAG) with two isoquinoline alkaloids, i.e., allocryptopine (ACP) and protopine (PP), was performed. The UV-Vis spectroscopy, molecular docking, competitive binding assays, and circular dichroism (CD) spectroscopy were used for the investigations. The results showed that ACP and PP form spontaneous and stable complexes with HSA and AAG, with ACP displaying a stronger affinity towards both proteins. Molecular docking studies revealed the preferential binding of ACP and PP to specific sites within HSA, with site 2 (IIIA) being identified as the favored location for both alkaloids. This was supported by competitive binding assays using markers specific to HSA's drug binding sites. Similarly, for AAG, a decrease in fluorescence intensity upon addition of the alkaloids to AAG/quinaldine red (QR) complexes indicated the replacement of the marker by the alkaloids, with ACP showing a greater extent of replacement than PP. CD spectroscopy showed that the proteins' structures remained largely unchanged, suggesting that the formation of complexes did not significantly perturb the overall spatial configuration of these macromolecules. These findings are crucial for advancing the knowledge on the natural product-protein interactions and the future design of isoquinoline alkaloid-based therapeutics.

Tanshinones, are a group of diterpenoid red pigments present in Danshen - an important herbal drug of Traditional Chinese Medicine which is a dried root of Salvia miltiorrhiza Bunge. Some of the tanshinones are sought after as pharmacologically active natural products. To date, the biosynthetic pathway of tanshinones has been only partially elucidated. These compounds are also present in some of the other Salvia species, i.a. from subgenus Perovskia, such as S. abrotanoides (Kar.) Sytsma and S. yangii B.T. Drew. Despite of the close genetic relationship between these species, significant qualitative differences in their diterpenoid profile have been discovered. In this work, we have used the Liquid Chromatography-Mass Spectrometry analysis to follow the content of diterpenoids during the vegetation season, which confirmed our previous observations of a diverse diterpenoid profile. As metabolic differences are reflected in different transcript profile of a species or tissues, we used metabolomics-guided transcriptomic approach to select candidate genes, which expression possibly led to observed chemical differences. Using an RNA-sequencing technology we have sequenced and de novo assembled transcriptomes of leaves and roots of S. abrotanoides and S. yangii. As a result, 134,443 transcripts were annotated by UniProt and 56,693 of them were assigned as Viridiplantae. In order to seek for differences, the differential expression analysis was performed, which revealed that 463, 362, 922 and 835 genes indicated changes in expression in four comparisons. GO enrichment analysis and KEGG functional analysis of selected DEGs were performed. The homology and expression of two gene families, associated with downstream steps of tanshinone and carnosic acid biosynthesis were studied, namely: cytochromes P-450 and 2-oxoglutarate-dependend dioxygenases. Additionally, BLAST analysis revealed existence of 39 different transcripts related to abietane diterpenoid biosynthesis in transcriptomes of S. abrotanoides and S. yangii. We have used quantitative real-time RT-PCR analysis of selected candidate genes, to follow their expression levels over the vegetative season. A hypothesis of an existence of a multifunctional CYP76AH89 in transcriptomes of S. abrotanoides and S. yangii is discussed and potential roles of other CYP450 homologs are speculated. By using the comparative transcriptomic approach, we have generated a dataset of candidate genes which provides a valuable resource for further elucidation of tanshinone biosynthesis. In a long run, our investigation may lead to optimization of diterpenoid profile in S. abrotanoides and S. yangii, which may become an alternative source of tanshinones for further research on their bioactivity and pharmacological therapy., (© 2024. This study delved into the influence of ecological and seasonal dynamics on the synthesis of secondary metabolites in the medicinal halophyte Limonium algarvense Erben, commonly known as sea lavender, and examined their antioxidant and anti-inflammatory properties. Aerial parts of sea lavender were systematically collected across winter, spring, summer, and autumn seasons from distinct geographic locations in southern Portugal, specifically "Ria de Alvor" in Portimão and "Ria Formosa" in Tavira. The investigation involved determining the total polyphenolic profile through spectrophotometric methods, establishing the chemical profile via liquid chromatography electrospray ionization quadrupole time-of-flight mass spectrometry (LC-ESI-QTOF-MS/MS), and evaluating in vitro antioxidant properties using radical and metal-based methods, along with assessing anti-inflammatory capacity through a cell model. Results unveiled varying polyphenol levels and profiles across seasons, with spring and autumn samples exhibiting the highest content, accompanied by the most notable antioxidant and anti-inflammatory capacities. Geographic location emerged as an influential factor, particularly distinguishing plants from "Ria de Alvor". Seasonal fluctuations were associated with environmental factors, including temperature, which, when excessively high, can impair plant metabolism, but also with the presence of flowers and seeds in spring and autumn samples, which also seems to contribute to elevated polyphenol levels and enhanced bioproperties of these samples. Additionally, genetic factors may be related to differences observed between ecotypes (geographical location). This study underscores sea lavender's potential as a natural source of antioxidant and anti-inflammatory agents, emphasizing the significance of considering both geographic location and seasonal dynamics in the assessment of phenolic composition and bioactive properties in medicinal plant species.

Vanicosides A and B isolated from Reynoutria sachalinensis rhizomes are disaccharide phenylpropanoid esters with proven antioxidant activity. Our earlier study showed the cytotoxic activity of vanicosides against melanoma cells, but the mechanism of cell death has not been elucidated. Based on the chemical structure of vanicosides, we proposed that they may induce cell death by generating reactive oxygen species (ROS) into melanoma cells. Moreover, the glucose molecule in their structure can affect the glucose transporters (GLUTs), upregulated in cancer cells. The A375 (melanotic) and C32 (amelanotic) melanoma cell lines were applied. Cell viability assay and ROS-Glo™ assay were performed before and after blocking of Glucose Transporter Type 1 (GLUT1) by WZB117. Fibroblasts and the SKOV-3 line were included in the study to test selectivity in the action of vanicosides and help to elucidate the mechanism of action. Upon incubation with vanicosides, high production of ROS occured, especially inside C32 cells, which was significantly reduced after GLUT-1 blocking. The A375 cells produced less ROS. Melanoma cells were simillary sensitive to the cytotoxic effects of vanicosides, which was clearly enhanced when vanicosides were used together with the WZB117 (GLUT1 inhibitor). The SKOV-3 line and the fibroblasts showed much less sensitivity to the cytotoxicity of vanicosides, also used together with WZB117. Moreover, no significant ROS formation was observed in these lines. The study proved that vanicosides generate ROS inside melanoma cells. In this research, a HPLC analysis, along with transcriptomics tools, was applied to evaluate chitosan and water stress for the prediction of phenolic flavonoids patterns and terpenoid components accumulation in Salvia abrotanoides Karel and S. yangii . The results indicated that the tanshinone contents under drought stress conditions increased 4.2-fold with increasing drought stress intensity in both species. The rosmarinic acid content in the leaves varied from 0.038 to 11.43 mg/g DW. In addition, the flavonoid content was increased (1.8 and 1.4-fold) under mild water deficit conditions with a moderate concentration of chitosan (100 mg L -1 ). The application of foliar chitosan at 100 and 200 mg L -1 under well-watered and mild stress conditions led to increases in hydroxyl cryptotanshinone (OH-CT) and cryptotanshinone (CT) contents as the major terpenoid components in both species. The expressions of the studied genes (DXS2, HMGR, KSL, 4CL, and TAT) were also noticeably induced by water deficit and variably modulated by the treatment with chitosan. According to our findings, both the drought stress and the application of foliar chitosan altered the expression levels of certain genes. Specifically, we observed changes in the expression levels of DXS and HMGR, which are upstream genes in the MEP and MVA pathways, respectively. Additionally, the expression level of KSL, a downstream gene involved in diterpenoid synthesis, was also affected. Finally, the present investigation confirmed that chitosan treatments and water stress were affected in both the methylerythritol phosphate pathway (MEP) and mevalonate (MVA) pathways, but their commitment to the production of other isoprenoids has to be considered and discussed.

The use of by-products from the agri-food industry is a promising approach for production of value-added, polyphenol-rich dietary supplements or natural pharmaceutical preparations. During pistachio nut processing, a great amount of husk is removed, leaving large biomass for potential re-use. The present study compares antiglycative, antioxidant, and antifungal activities as well as nutritional values of 12 genotypes belonging to four pistachio cultivars. Antioxidant activity was measured using DPPH and ABTS assays. Antiglycative activity was evaluated as inhibition of advanced glycation end product (AGE) formation in the bovine serum albumin/methylglyoxal model. HPLC analysis was performed to determine the major phenolic compounds. Cyanidin-3- O -galactoside (120.81-181.94 mg/100 g DW), gallic acid (27.89-45.25), catechin (7.2-11.01), and eriodictyol-7- O -glucoside (7.23-16.02) were the major components. Among genotypes, the highest total flavonol content (14.8 mg quercetin equivalents/g DW) and total phenolic content (262 mg tannic acid equivalent/g DW) were in KAL1 (Kaleghouchi) and FAN2 (Fandoghi), respectively. The highest antioxidant (EC 50 = 375 μg/mL) and anti-glycative activities were obtained for Fan1. Furthermore, potent inhibitory activity against Candida species was recorded with MIC values of 3.12-12.5 µg/mL. The oil content ranged from 5.4% in Fan2 to 7.6% in Akb1. The nutritional parameters of the tested cultivars were highly variable: crude protein (9.8-15.8%), ADF (acid detergent fiber 11.9-18.2%), NDF (neutral detergent fiber, 14.8-25.6%), and condensed tannins (1.74-2.86%). Finally, cyanidin-3- O -galactoside was considered an effective compound responsible for antioxidant and anti-glycative activities.

Ducrosia anethifolia (DC.) Boiss. is an aromatic medicinal plant that has been traditionally used as an analgesic to treat headaches, backaches, colic, and cold. This study evaluated the yield, physiological, and phytochemical traits of 24 populations for 2 consecutive years under the water stress condition. The seed yield and physiological traits demonstrated the highest values in the first and second year, respectively. Hydrogen peroxide (H 2 O 2 ), proline, malondialdehyde (MDA), and antioxidant activity enzymes were elevated, while chlorophyll, carotenoids, relative water content (RWC), and yield decreased under drought stress. High-performance liquid chromatography (HPLC) was also applied to assess the changes in some polyphenolic compounds in response to water stress. The increase in some phenolic compounds, such as p-coumaric acid, was recorded due to drought stress, while there was a decrease in flavonoids, that is luteolin and quercetin. Among the populations, Abarkuh2 indicated the highest increase in p-coumaric acid (96%) in response to drought stress. In general, high diversity among the studied populations provides new insights into choosing the beneficial populations for medicinal and food purposes. Oregano and marjoram are important aromatic spices in the food industry, as well as medicinal plants with remarkable antioxidant properties. Despite their popularity, little is known about treatments that would influence the antioxidant capacity of essential oils. In this study, different spectra of LED light, namely blue, red, white, blue-red, and natural ambient light as a control, were applied to assess the essential oil content, composition, flavonoid, phenolic, and antioxidant capacity of oregano and marjoram. GC-MS analysis revealed thymol, terpinen-4-ol, sabinene, linalool, p-cymene, and γ-terpinene as the main compounds. In oregano, the thymol content ranged from 11.91% to 48.26%, while in marjoram it varied from 17.47% to 35.06% in different samples. In oregano and marjoram, the highest phenolic contents were in blue (61.26 mg of tannic acid E/g of DW) and in white (65.18 mg of TAE/g of DW) light, respectively, while blue-red illumination caused the highest increase in total flavonoids. The antioxidant activity of oregano and marjoram extract was evaluated using two food model systems, including DPPH and β-carotene bleaching. The highest antioxidant capacity was obtained in control light in oregano and blue-red light in marjoram. The results provide information on how to improve the desired essential oil profile and antioxidant capacity of extracts for industrial producers.

Scutellaria baicalensis Georgi is a valuable medicinal plant of the Lamiaceae family. The roots, Scutellariae baicalensis radix , are valued in the traditional medicine of East Asia and are also listed in several pharmacopeias, such as the Chinese and European versions. The roots contain a high amount of flavones, such as baicalein, wogonin and their glucuronides, baicalin and wogonoside, respectively, with rare structures of unsubstituted B-ring. These major constituents are responsible for its pharmacological activity, mainly anti-inflammatory, antiviral, and antitumor, as well as BDZ-receptor modulating. There is a fast-growing demand for both the crude drug and the individual flavonoids obtained from it. However, the variability of content and composition of flavonoids in the roots is significant and affects pharmaceutical use, and little is known about the influence of various factors on root quality. In our experiments, we use aeroponics to determine the effect of electroporation as an abiotic stressor on plant growth, development, and root mass, as well as on its metabolic profile., Results: Electroporation significantly impacted plant growth and the content of flavonoids, especially baicalein and wogonin, depending on the treatment parameters. Concentrations of aglycones were increased in at least half of the treatment conditions. The greatest amounts (a 2.5-fold increase compared to controls) were recorded after applying an electrical field characterized by the following parameters: E = 3 kV/cm, t = 100 μs, and N = 10. In conclusion, electrostimulation is an innovative and efficient way to increase plant growth and yield in an aeroponic system, as well as modulate the profile and content of bioactive flavones in the roots. However, the fine-tuning of these parameters, such as the electrical field strength (E), length (t), and number (N) of impulses delivered, is of great importance. Polygoni Cuspidati Rhizoma et Radix ( syn. rhizomes of Reynoutria japonica Houtt.) is a pharmacopoeial raw material in Europe and China. In traditional medicine, one of the applications for Reynoutria japonica rhizomes is wound healing. In a recent in vitro study, we demonstrated that ethanol and acetone extracts from this herbal drug have the potential to heal oral gum wounds. However, considering that a majority of herbal medicines have been traditionally administered as water decoctions, in the present study, a decoction of Reynoutria japonica rhizomes was prepared and detailed tests to determine its in vitro gingival wound healing activity were conducted. We used the primary human gingival fibroblasts (HGF) incubated with a decoction to determine cell viability (MTT assay), cell proliferation (the confocal laser scanning microscope-CLSM), and cell migration (wound healing assay). Moreover, the collagen type III expression was examined using immunocytochemical staining. The studied decoction was qualitatively and quantitatively characterized using the validated HPLC/DAD/ESI-HR-QTOF-MS method. The Folin-Ciocalteu assay was used to determine the total phenols and tannins content. Additionally, HPLC-RI analysis of decoction and the previously obtained ethanol and acetone extracts was used to determine the composition of saccharides. Low concentration (from 50 to 1000 µg/mL) of decoction after 24 h incubation caused a significant increase in HGF cell viability. No cytotoxic effect was observed at any tested concentration (up to 2000 µg/mL). The lowest active concentration of decoction (50 µg/mL) was selected for further experiments. It significantly stimulated human gingival fibroblasts to proliferate, migrate, and increase the synthesis of collagen III. Phytochemical analysis showed significantly fewer polyphenols in the decoction than in the ethanol and acetone extracts tested earlier. In contrast, high levels of polysaccharides were observed. In our opinion, they may have a significant effect on the oral wound healing parameters analyzed in vitro. The results obtained encourage the use of this raw material in its traditional, safe form-decoction.

Ducrosia anethifolia Boiss is a medicinal and aromatic plant that is used as a food additive and drug in the areas of natural distribution. In this study, eight populations from different geographical regions of Iran were evaluated for their essential oil composition, total phenolics and flavonoids as well as for free radical scavenging activity during two consecutive years under water deficit. Analysis of variance was performed using a randomized block design at three levels of irrigation and 2 years, using the GLM procedure of SAS software and cluster analysis was used according to Ward's method using squared Euclidean dissimilarity. The highest essential oil and total phenolics/flavonoids content were obtained in the second year. GC-MS analysis was used to analyze the essential oil components. In normal conditions, cis-chrysanthenyl acetate (39.1-66.4%) and α-pinene (1.02-4.7%) were recognized as major compounds. The essential oil components were elevated under water stress. The highest increase in cis-chrysanthenyl acetate (21.32%) in response to water stress was observed in Abarkuh1. Elevation in total phenolic, flavonoid and antioxidant activity was also observed in plants subjected to water deficit. The highest content of phenolic acids was measured in Semirom (136.74 mg TAE/g DW), while the highest flavonoid content was in Kerman (6.05 mg QUE/g DW) in severe water stress condition. Among traditional Iranian herbs, Perovskia species (a subgenus of Salvia ), while being valued ornamentals, are also studied for numerous potential pharmacological and therapeutic aspects. The current study was conducted to assess the effectiveness of two species of arbuscular mycorrhizal fungi (AMF), Funneliformis mosseae and Rhizophagus intraradices, separately and in combination, in terms of the essential oil content and compositions along with the enzymatic and non-enzymatic antioxidants in Salvia abrotanoides and S. yangii in response to three levels of irrigation, including 100% FC as well-watered, 75% FC (moderate irrigation deficit), and 50% FC (severe irrigation deficit). In both species, essential oil content, enzyme antioxidant activities, total phenolics, and flavonoids were increased significantly with the severity of stress; this increase was more pronounced in mycorrhizal inoculated herbs. Furthermore, leaf phosphorus concentration, relative water content, chlorophylls a and b , and total carotenoids decreased in parallel with reducing soil moisture; albeit, AMF inoculation improved the stress symptoms under increasing severity of water restriction compared with their control conditions. In addition, the percentage of root colonization was positively correlated with the relative water content (RWC) and leaf phosphorus concentration. Taking into account the essential oil groups, AMF colonization elevated some essential oil components, such as oxygenated monoterpenes, 1,8-cineol, camphor, and borneol, whereas the main sesquiterpenes, including E-β-caryophyllene and α-humulene, remarkably decreased. Taken together, these findings highlighted the role of symbiosis with AMFs in increasing the tolerance of water deficit stress in S. abrotanoides and S. yangii and improving their essential oil composition.

Specific antibodies that humans acquire as a result of disease or after vaccination are needed to effectively suppress infection with a specific variant of SARS CoV-2 virus. The S protein of the D614G variant of coronavirus is used as an antigen in known vaccines to date. It is known that COVID-19 disease resulting from infection with this coronavirus can often be very dangerous to the health and lives of patients. In contrast, vaccines produce antibodies against an older version of the protein S-D614G (January 2020) and therefore have difficulty recognizing new variants of the virus. In our project we propose to obtain specific and precise antibodies by means of so-called controlled infection against specific infectious variants of the SARS-CoV-2 virus "here and now". Currently, several variants of this pathogen have already emerged that threaten the health and lives of patients. We propose to reduce this threat by partially, but not completely, blocking the fusion mechanism of the SARS-CoV-2 virus into human respiratory cells. According to our plan, this can be achieved by inhibiting cathepsin L activity in respiratory cells, after introducing natural and non-toxic cysteine protease inhibitors into this area. We obtain these inhibitors by our own method from natural, "human body friendly" natural resources. We hypothesize that blocking cathepsin L will reduce the number of infecting viruses in cells to such an extent that COVID-19 developing in infected individuals will not threaten their health and life. Ajowan ( Trachyspermum ammi L.) is considered a valuable spice plant with a high thymol content. Seed yield, essential oil constituents, polyphenolic composition, and antioxidant capacity of ajowan ( Trachyspermum ammi L.) populations were evaluated in three (normal, moderate, and severe) water irrigation regimes. The highest essential oil content (5.55%) was obtained under normal condition in the Yazd population. However, both essential oil and seed yield showed significant reductions as a result of water stress. According to gas chromatography-mass spectrometry (GC-MS) analysis, thymol (61.44%), γ-terpinene (26.96%), and p -cymene (20.32%) were identified as the major components of the oil. The highest (89.01%) and the lowest (37.54%) thymol contents were in Farsmar and Hamadan populations in severe stress condition, respectively. Based on HPLC analysis, chlorogenic (3.75-47.35 mg/100 g), caffeic (13.2-40.10 mg/100 g), and ferulic acid (11.25-40.10 mg/100 g) were identified as the major phenolic acids, while rutin was determined as the major flavonoid (11.741-20.123 mg/100 g). Moreover, total phenolic and flavonoid contents were elevated under drought stress treatment, while antioxidants responded inconsistently to stress based on two model systems. Overall, the Yazd population exhibited a superior response to water stress, as evidenced by its less reduced thymol and oil yield content, while Arak and Khormo had the highest accumulation of polyphenolic compounds.

Polygonum maritimum L. (sea knotgrass) belongs to a genus commonly used in folk medicine to treat inflammation-related disorders. In vitro pharmacological studies have confirmed these properties that were ascribed to bioactive flavonoids, such as myricetin and quercetin glycosides. Therefore, this study aimed at establishing a micropropagation procedure for sea knotgrass for obtaining standardized materials for its potential commercial cultivation. For that, a complete plant regeneration protocol was developed by improving shoot multiplication from nodal explants, rooting and acclimatization procedures, followed by the assessment of the phenolic profile of the in vitro -produced plants. The combination of 3 mg/L 6-benzylaminopurine (BA) + 0.1 mg/L indole-3-acetic acid (IAA) induced the maximum shoot formation (10.3), which was significantly increased from the first to the second cycle (18.3). The best rooting capacity was observed on shoots derived from the control medium (100%), followed by 2 mg/L kinetin (KIN) (97%) and 3 mg/L BA + 0.1 mg/L IAA (90%); however, the shoot number at the end of the rooting phase was higher on shoots derived from 3 mg/L BA + 0.1 mg/L IAA (6.16). The plant growth regulators used in the multiplication phase influenced survival in the acclimatization process, and plants derived from the control medium had the highest survival percentage (63.1%). Acetone extracts made from aerial organs of micropropagated sea knotgrass showed a predominance of the flavonoid myricetin-3- O -rhamnoside (8.135 mg/g). The search for mitochondria-targeted drugs has dramatically risen over the last decade. Mitochondria are essential organelles serving not only as a powerhouse of the cell but also as a key player in cell proliferation and cell death. Their central role in the energetic metabolism, calcium homeostasis and apoptosis makes them an intriguing field of interest for cancer pharmacology. In cancer cells, many mitochondrial signaling and metabolic pathways are altered. These changes contribute to cancer development and progression. Due to changes in mitochondrial metabolism and changes in membrane potential, cancer cells are more susceptible to mitochondria-targeted therapy. The loss of functional mitochondria leads to the arrest of cancer progression and/or a cancer cell death. Identification of mitochondrial changes specific for tumor growth and progression, rational development of new mitochondria-targeted drugs and research on delivery agents led to the advance of this promising area. This study investigated the effect of drought stress on the amount of phenolic and flavonoid compounds as well as H 2 O 2 and malondialdehyde (MDA) in Achillea pachycephala. The expression patterns of the key genes and their molecular mechanisms in the phenylpropanoid pathway (PAL, CHS, CHI, F3H, F3'H, F3'5'H, FLS) were also assessed during drought stress using quantitative real-time polymerase chain reaction (qRT-PCR). The samples were harvested at 0, 7, 14, 21 and 28 days after exposure to drought stress. High-performance liquid chromatography (HPLC) analysis was performed to determine the changes of phenolic and flavonoid compounds - chlorogenic acid, caffeic acid, rutin, luteolin-7-O-glycoside, 1,3-dicaffeoylquinic acid, apigenin-7-O-glycoside, luteolin, apigenin and kaempferol - during stress conditions. Concentrations of most of the compounds increased with increasing drought stress duration. Most of the phenolic acids continued to accumulate with increasing duration of stress, while flavonoids dramatically decreased at day 28 of stress. Chlorogenic acid was the most abundant phenolic acid (4.97 mg/100 g dry weight [DW]) at the beginning of the experiment, while it decreased at day 7 and increased again at day 21. However, different trends were observed for some flavonoids, such as luteolin and apigenin. At the beginning of stress treatment, high accumulation of free radicals (H 2 O 2 ) and lipid peroxidation (MDA) led to elevated expression of most of the flavonoid genes. MDA increased from 22.66 to 43.28 μmol g -1 DW at day 28. CHS gene expression was elevated at day 7, while chi gene expression remained unchanged. At the end of the stress period, most of the flavonoid concentrations and expression of the relevant genes also increased. The results can facilitate selection of appropriate drought conditions to obtain the highest levels of flavonoids such as luteolin and apigenin and phenolic compounds such as chlorogenic acid for improved health benefits., (Copyright © 2019 Elsevier Ltd. Cholinesterase inhibition is one of the most treatment strategies against Alzheimer's disease (AD) where metal accumulation is also strongly associated with pathology of the disease. In the current study, we assessed inhibitory effect against acetyl- (AChE) and butyrylcholinesterase (BChE) and metal-chelating capacity of twelve diterpenes: arucadiol, miltirone, tanshinone IIa, 1-oxomiltirone, cryptotanshinone, 1,2-didehydromiltirone, 1,2-didehydrotanshinone IIa, 1β-hydroxycryptotanshinone, 15,16-dihydrotanshinone, tanshinone I, isotanshinone II, 1(S)-hydroxytanshinone IIa, and rosmarinic acid, isolated from Perovskia atriplicifolia and Salvia glutinosa. The compounds were tested at 10 μg/mL using ELISA microtiter assays against AChE and BChE. QSAR and molecular docking studies have been also performed on the active compounds. All of the compounds showed higher [e.g., IC 50  = 1.12 ± 0.07 μg/mL for 1,2-didehydromiltirone, IC 50  = 1.15 ± 0.07 μg/mL for cryptotanshinone, IC 50  = 1.20 ± 0.03 μg/mL for arucadiol, etc.)] or closer [1,2-didehydrotanshinone IIa (IC 50  = 5.98 ± 0.49 μg/mL) and 1(S)-hydroxytanshinone IIa (IC 50  = 5.71 ± 0.27 μg/mL)] inhibition against BChE as compared to that of galanthamine (IC 50  = 12.56 ± 0.37 μg/mL), whereas only 15,16-dihydrotanshinone moderately inhibited AChE (65.17 ± 1.39%). 1,2-Didehydrotanshinone IIa (48.94 ± 0.26%) and 1(S)-hydroxytanshinone IIa (47.18 ± 5.10%) possessed the highest metal-chelation capacity. We evaluated the antioxidant properties of four main flavones from Scutellaria baicalensis: baicalein, wogonin and their glucuronides - baicalin and wogonoside. We used three in vitro assays: free radical scavenging with 2,2'-diphenylpicrylhydrazyl radical, transition metal ions reducing power by phosphomolybdenum assay and inhibition of the hydroxyl radical-induced peroxidation of linoleic acid assay. All flavones have antioxidant capacity, which differs depending on the structure and mechanisms of activity. In all tests, only baicalein - the aglycone with three adjacent hydroxyl groups - exhibited consistent antioxidant effect. Wogonin protected linoleic acid against oxidation. Baicalin displayed less potent antioxidant properties whereas wogonoside did not have significant antioxidant activity.

The antioxidant and antimutagenic properties of the extract from the oriental natural drug - Belamcanda chinensis (L.)DC. (Iridaceae) rhizomes have been examined. Three different antioxidant assays and Salmonella antimutagenic test have been used. The extract has strong free radical scavenging activity as shown by the DPPH assay with EC50 of 63.4 microg/mL. Similarly, the high ability to reduce the transition metal ions was shown by phosphomolybdenum assay as well as effective prevention of hydroxyl radical induced linoleic acid peroxidation. The latter averaged 77.9% with an EC50 of 45 microg/mL. The mutations induced in two Salmonella typhimurium strains (TA98 and TA100) by both directly (NQNO) and indirectly acting (2AF) chemical mutagens were inhibited efficiently in dose dependent manner for concentrations of 50-500 microg/mL. The inhibition percentage in TA98 was 62.8% for direct mutagen and 94.4% for enzymatically activated promutagen. In TA100 the inhibition was 82.7% and 73.5%, respectively.

The four flavonoids baicalin, baicalein, wogonoside, and wogonin have been isolated from Scutellaria baicalensis (Lamiaceae) root extract as main bioactive components. We evaluated their antimutagenic activity as potential chemopreventive natural substances using the Ames test. Free radical scavenging properties have also been measured with the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. Both crude extract and isolated flavone glycosides and aglycones have antimutagenic and free radical scavenging properties. The lipophilic flavone baicalein has been confirmed to be the most powerful antimutagen among the compounds tested.
