151. 403 IDENTIFICATION AND CHARACTERIZATION OF A NOVEL MOUSE AND HUMAN MOPT GENE CONTAINING MORN MOTIF PROTEIN IN TESTIS

Abstract: By subtraction screening methods, we identified a novel mouse and its counterpart, human MOPT gene. The mouse and human MOT gene is localized on mouse chromosome 17E3 and human chromosome 2p22, and spans approximately 7 kb. Analysis of the mouse Mopt sequence revealed the existence of an ORF of 240 bp encoding a putative protein of 79aa amino acids. The predicted protein has a theoretical molecular mass of 8.89 kDa and a calculated isoelectric point of 5.82. The protein was unique; it did not show any similarities with other known protein except for Morn motif domain. Real-time reverse transcriptase polymerase chain reaction and Northern blot analysis revealed that mouse Mopt transcripts are highly and specifically expressed in adult testis and skeletal muscle. In situ hybridization and immunohistochemistry studies showed that mouse Mopt transcript and protein was confined mainly to round and elongated spermatids, except for a few individual dispersed spermatocytes, and increases in abundance in subsequent stages. To characterize Mopt functions in spermiogenesis, we examined Mopt protein distribution in late spermiogenesis by using immunogold electron microscopy: Mopt protein first appeared in the proacrosomic vesicles of the early Golgi phase spermatids. In the final step of spermiogenesis, Mopt expression was translocated from the head cap of an elongated spermatid to the nucleus of mature spermatozoa. However, no other testicular cell types, including somatic cells, spermatogenic cells, and residual cytoplasm that ultimately is engulfed as residual bodies into Sertoli cells, were found to bear Mopt-positive staining. This observation suggested that Mopt may play an important role in dynamic regulation of acrosome biogenesis during late spermiogenesis and an as yet uncharacterized role in oocyte activation during capacitation, after fertilization, or both.

155. Single-Stage Extracranial Carotid Artery Stenting and Intracranial Aneurysm Coiling: Technical Feasibility and Clinical Outcome.

Abstract: The coexistence of carotid artery stenosis and cerebral aneurysm in a patient presents challenges for treatment decision-making. The purpose of this study was to evaluate the technical feasibility and clinical outcome after singlestage extracranial carotid artery stenting (CAS) and ipsilateral intracranial aneurysm coiling in a single institution. From March 2005 to February 2011, 17 patients with 21 aneurysms underwent single-stage CAS and coiling for ipsilateral aneurysms. There were symptomatic atherosclerotic carotid stenoses with unruptured aneurysms in eight, ruptured or symptomatic aneurysms with simultaneous asymptomatic carotid stenoses in two and asymptomatic lesions in seven. CAS was followed by aneurysm coiling in all 17 patients. Clinical and radiological data were reviewed. There were two procedure-related complications: acute in-stent thrombosis in one and premature aneurysmal rupture in the other. After aneurysm coiling, complete occlusion was demonstrated in 17 aneurysms and near-total occlusion in four. No neurological deficit was found at discharge and follow-up outcomes were excellent in all the patients (mean, 32.9 months). Follow-up imaging studies were performed in all the patients, including neck CT angiography in 14 (mean, 26.1 months), brain MR angiography in 14 (mean, 31.2 months), and conventional angiography in three (mean, 14.7 months). They revealed two asymptomatic, mild carotid re-stenoses and one major aneurysmal recanalization requiring re-coiling. A single-stage CAS and coiling procedure appears to be feasible and the complication rate seems to be reasonable. We suggest that there is no need for separate therapeutic procedures when a patient has carotid artery stenosis and accompanying ipsilateral intracranial aneurysm. [ABSTRACT FROM AUTHOR]

156. GNSS-RO residual ionospheric error (RIE): a new method and assessment

Abstract: Global Navigation Satellite System (GNSS) radio occultation (RO) observations play an increasingly important role in monitoring climate changes and numerical weather forecasts in the upper troposphere and stratosphere. Because the magnitudes of the RO bending angle are small at these altitudes, quantifying and removing residual ionospheric error (RIE) are critical to accurately retrieve atmospheric temperature and refractivity. Yet, RIEs remain poorly characterized in terms of the global geographical distribution and its variations with the local time and altitude influenced by the solar cycle and solar geomagnetic disturbances. In this study we developed a new method to determine RIE from the RO excess phase measurement on a profile-by-profile basis. The method, called the ϕex-gradient (dϕex/dht) method, is self-sufficient and based on the vertical derivative of the RO excess phase (ϕex) with respect to tangent height (ht), which can be applied to individual RO bending angle observations for RIE correction. In addition to the RIE in bending angle measurements, RIEs can also be found in the RO ϕex measurements in the upper atmosphere where an exponential dependence is expected. RIEs are likely to impact the RO temperature retrieval by inducing a small-scale variance that is solar-cycle-dependent. We found that the RIE values derived from the dϕex/dht method can be both positive and negative, which is fundamentally different from the κ method that produces only positive RIE values. The new algorithm reveals a latitude-dependent diurnal variation with a larger daytime negative RIE (up to ∼ 3 µrad) in the tropics and subtropics. Based on the observed RIE climatology, a local-time-dependent RIE representation is used to evaluate its impacts on reanalysis data. We examined these impacts by comparing the data from the Goddard Earth Observing System (GEOS) data assimilation (DA) system with and without the RIE. The RIE impact on GEOS DA temperature is mainly confined to the polar regions of the stratosphere. Between 10 and 1 hPa the temperature differences are ∼ 1 K and exceed ∼ 3–4 K in some cases. These results further highlight the need for RO RIE correction in modern DA systems.

157. Diurnal variations of NO2 tropospheric vertical column density over the Seoul metropolitan area from the Geostationary Environment Monitoring Spectrometer (GEMS): seasonal differences and the influence of the a priori NO2 profile

Abstract: The Geostationary Environment Monitoring Spectrometer (GEMS), launched in 2020, provides both temporally and spatially continuous air quality data from geostationary Earth orbit (GEO). This study first investigates the seasonal variations and diurnal behavior of nitrogen dioxide (NO2) tropospheric vertical column densities (TropVCDs) over the Seoul metropolitan area (SMA) using GEMS data, retrieved by the IUP-UB algorithm. We find that the magnitude of the NO2 TropVCDs and their diurnal behavior have significant seasonal dependences. In January, the highest NO2 TropVCD values in the range 27.5–28.9×1015 molec.cm-2 during the four seasons were observed at 15:00 local time (LT) and NO2 TropVCD increases from the first retrieved values at 10:00 LT. On the other hand, we find the lowest values (7.4–8.8×1015 molec.cm-2) are at ∼14:00 LT in July. The VCD values in July increased up to 10:00 LT and then decreased until 14:00 LT but then began to increase again. These different diurnal behaviors of the TropVCDs in the different seasons reflect the differences in photochemical and meteorological conditions as well as the emissions of NOx. Photochemical transformations are typically more rapid in July and slower in January. The absolute values and diurnal behavior of NO2 TropVCDs are significantly influenced by the wind speed, except in July. Moderate (wind speed ≥3 m s−1) or strong wind (wind speed >5 m s−1) reduced the magnitude of the diurnal behavior in January, implying that the NO2 plumes were transported downwind. Finally, we compared the retrieved NO2 TropVCDs by using different a priori NO2 data simulated by TM5 and WRF-Chem, calculated using the most recent emission inventories. Although simulated VCDs from WRF-Chem and TM5 show differences of up to a factor 2.75, retrieved NO2 TropVCDs using each a priori data have almost identical values and diurnal behaviors, except in July. Notably, the diurnal behavior of the retrieved NO2 TropVCDs is independent of that from the two chemical transport models, indicating that observations of slant column densities are the dominant factor in determining the diurnal behavior of NO2 TropVCDs. Changes in the model horizontal resolution and volatile organic compound (VOC) emission inventory do not significantly affect the retrieved NO2 TropVCDs in this study. However, when the a priori NO2 vertical profile was fixed as the values at 13:45 LT, the diurnal patterns of NO2 TropVCDs showed significant changes, with differences of up to −18.3 %.

158. Regulation of Fc gamma receptor expression and phagocytosis of a human monoblast cell line U937. Participation of cAMP and protein kinase C in the effects of IFN-gamma and phorbol ester

Abstract: We investigated the positive and negative effects of IFN-gamma, PMA, dibutyryl cAMP (Bt2cAMP), dexamethasone and transforming growth factor-beta (TGF-beta) on Fc gamma R subtype expression and phagocytosis of a human monoblast cell line, U937. IFN-gamma increased and Bt2cAMP decreased Fc gamma RI expression determined by a mAb 32.2, whereas PMA and Bt2cAMP increased Fc gamma RII expression determined by a mAb IV-3. Phagocytosis was measured microscopically by counting ingested aggregated human IgG- or BSA-treated ox E (Eo'-IgG or Eo'-BSA). IFN-gamma increased the phagocytosis of Eo'-IgG but not that of Eo'-BSA, and PMA increased the phagocytosis of both Eo'-IgG and Eo'-BSA. Bt2cAMP decreased both basal and IFN-gamma- and PMA-augmented phagocytosis of U937 cells. Dexamethasone also inhibited both basal and IFN-gamma-augmented Fc gamma RI expression and PMA-augmented Fc gamma RII expression and phagocytosis, but did not affect IFN-gamma-augmented phagocytosis of Eo'-IgG. The augmentation of phagocytosis of Eo'-IgG by IFN-gamma thus seems to be due mainly to the increased internalizing process rather than to increased Fc gamma RI expression. TGF-beta slightly decreased Fc gamma R expression. In a study of the participation of protein kinase C (PK-C), it was found that H-7, a PK-C inhibitor, did not inhibit either IFN-gamma- or PMA-enhanced Fc gamma RI and Fc gamma RII expression, respectively, and 1-oleoyl-2-acetylglycerol and N-(6-phenylhexyl)-5-chloro-1-naphthalenesulfonamide, both PK-C activators, did not show any apparent increase in Fc gamma R expression and phagocytosis. These results show that Fc gamma RI and Fc gamma RII expression on U937 cells is regulated by different mechanisms and that IFN-gamma and PMA play their roles in Fc gamma R expression and phagocytosis by different pathways. It is possible that cAMP but not PK-C plays an important role in the regulation of Fc gamma R expression and phagocytosis.

Abstract: Inhibition of fluid flow instabilities in the melt by the axial magnetic field in Czochralski silicon crystal growth (AMCZ) is investigated precisely by a high‐sensitivity striation etch in conjunction with temperature measurements. The magnetic field strength (B) was varied up to 4.0 kG, incremented mostly in 0.5‐kG/2.5‐cm crystal length. The convection flow was substantially suppressed at B≥1.0 kG. A low oxygen level of 2–3 ppm and a high resistivity of 400 Ω cm is achieved in the AMCZ silicon crystals at B≥1.0 kG. Random striations at B=0, characteristic of turbulent convection, assumed progressively a periodicity, indicative of oscillatory convection at 0.35

Abstract: The microsegregation behaviour of antimony in the faceted and non-faceted Czochralski silicon crystal growth was analyzed quantitatively. Using small melt heights and no rotation, dopant striations of various small spacings were eliminated. Interface demarcation and spreading resistance measurements were used for the segregation analysis. The dopant concentration and its fluctuation during the faceted growth were both higher than during non-faceted growth. On the other hand, fluctuations of the microscopic growth rate were about the same in magnitude and periodicity in the two growth regions.

Abstract: Finite difference computations were used to follow boron migration in the melt and segregation in CZ Si crystal growth. With no applied magnetic field, transport near the growth interface was dominated by advection, resulting in a nearly uniform radial distribution of boron. Application of a 1 kG axial field calmed the melt motion enough that advection and diffusion appeared to play equal roles in transport near the crystal: the boron profile under the interface showed some characteristics of a diffusion boundary layer, but was quite shallow. Increasing to 2 kG further suppressed the melt motion until transport near the growth interface appeared to be strongly dominated by diffusion. Abstract: Computer simulation leading to controlled large diameter Czochralski crystal growth is discussed. A simple mathematical model, which describes the different crystal growth phases including neck‐in, fast flat top, roll‐over to constant diameter bulk growth, and tail‐off is presented. This model, in conjunction with a computer‐implemented simulator, is used to simulate silicon crystal growth. Good agreement between simulation results and experimental crystal growth is obtained.

Abstract: The formation of microdefects in pedestal‐grown silicon crystals was investigated by using x‐ray topography. Small‐diameter (4–5 mm) crystals were grown without crystal rotation, whereby all potential fluid flow instabilities in the melt were significantly reduced. Microdefects (most likely B‐type defects) of a uniform distribution with a density of 3×106 cm−3 were still formed in the crystal. No microdefects were present, however, in regions close to the crystal rim, apparently due to the out‐diffusion of point defects responsible for the formation of the microdefects; the estimated diffusion coefficient is about 1×10−5 cm2/sec in the temperature range ∼1000–1400 °C.

Abstract: In a series of electron microscopic studies of human urinary stones, a stone composed of sodium orthophosphate hydrate was identified. The stone was recovered from a patient who succumbed to advanced renal failure. A massive failure of the sodium pump, which cotransports phosphate across the brush border membrane of the proximal tubules is thought to be responsible for such an exceptional stone. This appears to be the first description of sodium phosphate crystal in a human urinary stone. Electron microscopy is a useful tool for stone analysis.

Abstract: Calcification is common in human placentas and is widely recognized as a normal part of maturation and aging of this organ. Eleven human placentas were studied by light and electron microscopy to elucidate the mechanism of placental calcification. Earliest mineral deposits were seen along the trophoblastic basement membrane of the chorionic villi undergoing fibrinoid degeneration. Transmission electron microscopic examination revealed crystalline deposits within small membrane-bound vesicles; the latter appear to be derived from degenerating cells and were particularly numerous within the basement membrane. X-ray microanalysis of these deposits revealed calcium and phosphorus peaks and the pattern of calcium hydroxyapatite was noted by electron diffraction. This pattern of calcification, i.e., precipitation of calcium hydroxyapatite in association with extracellular membrane bound vesicles, is similar to that seen in physiologic and pathologic calcifications of other tissues.

Abstract: Various stones were examined by a combined electron microscopy. Morphological parameters of calcium oxalates and phosphates, struvite, urates, cystine, gypsum, siliceous deposits in urinary, prostatic and salivary calculi, and calcium carbonate in gallstones were defined. Study also disclosed that stones frequently contain organic crystals in addition to the organic matrix. Membranous cell debris was observed in the nuclei of small stones and were believed to be common nidus of various stones. In view of a very limited number of crystals occurring in human stones, which differ obviously in their habits and growth patterns, an expeditious identification of stone components is possible by scanning electron microscopy. SEM with x-ray analysis is a powerful tool for research and diagnosis of various stones. It is most suitable for the study of surface phenomena, such as crystal growth, detection of phenomena, such as crystal growth, detection of organic matrix and minor components of the stones, and is a wide open field for a further study.

Abstract: On the basis of an extensive electron microscopic study of human urinary stones, a step by step method of stone analysis by scanning electron microscopy (SEM) morphology was developed. A preliminary blind study of 100 consecutive stones demonstrated that the method requires minimum amounts of training and is highly accurate. With several hours of training, accuracy of the stone analysis by SEM morphology alone exceeded 95%. The data indicate that, with further refinements, SEM stone analysis can be accomplished expeditiously and exceed other methods known for stone analysis with economy and accuracy.

Abstract: Calcium oxalate stones are very common and increasing. Crystal growth is no less important than the crystal nucleation in the pathogenesis of stone formation. The crystal growth was studied in human calcium oxalate stones by a combined electron microscopy and x-ray diffraction. The main mode of weddellite growth was interpenetration twinning of tetrahedral bipyramids. Bipyramids may form as initial crystal seeds, develop from anhedral crystals (crystals which lack flat symmetric faces) of spherular or mulberry shape, develop on the surface of preformed bipyramids by spiral dislocation mechanisms, or develop on whewellite crystal by heterogeneous nucleation and epitaxy. Heterogeneous nucleations of whewellite on weddellite, and calcium apatite on whewellite were also observed. Whewellite grew mainly by parallel twinning. Interpenetration twinning was exceptional. Transformation of anhedral to euhedral (completely bounded by flat faces that are set ar fixed angles to one another) whewellite occurred by parallel fissurations followed by brick wall like stacking of the crystals, while euhedral transformation of weddellite occurred by protrusion of bipyramids frm anhedral crystal surface. Occasionally, an evidence of crystal dissolution was noted. Although an aggregation of crystals is believed to play a pivotal role in stone nidus formation, growth in size of the formed crystals, and twinning and epitactic crystal intergrowth apparently play a significant role in the obstructive urinary stone formation.

Abstract: A previous study of urinary stones by a combined electron microscopy demonstrated the potential of scanning electron microscopy and X-ray analysis as an analytic tool for urinary stones. Electron diffraction was chosen for the final confirmation of crystals in the study. Although electron diffraction is highly accurate for this purpose, it is desirable to establish the sensitivity of X-ray analysis for the identification of stone components relative to the more commonly used methods. Eighty six consecutive urinary stones were analyzed by X-ray analysis and the findings were compared with those of X-ray diffraction, infrared spectrometry and chemical analysis. The results indicate that X-ray analysis exceeds X-ray diffraction and infrared spectroscopy in its sensitivity for the identification of stone components several fold. This was largely due to the inability of the latter methods to detect apatite in more than half of the apatite containing stones. The findings in X-ray analysis had the best correlation with chemical analysis, which was applied mainly to the detection of apatite. X-ray analysis is particularly suited for the detection of rare and minor inorganic components of urinary stones such as silica and gypsum, and is obviously one of the most powerful tools for the analysis of urinary stones. Further application of X-ray analysis to urinary stone is likely to discern rare inorganic components of urinary stones overlooked by other methods.

Abstract: The oligodendrocytes in culture serve as an important model for the study of demyelination diseases. We have previously originated a method of isolating human oligodendrocytes. In order to establish their three dimensional morphology, scanning electron microscopy of the cultured oligodendrocytes was performed. The oligodendrocytes bulk isolated from adult human brain were maintained in culture for more than 2 months. At the various periods in culture, the cells were studied by scanning electron microscopy and immunofluorescence staining using marker antibodies for the identification of oligodendrocytes. The three dimensional organization and the surface morphology of the cultured oligodendrocytes were investigated. They displayed an extensive network of the cell processes and characteristic surface morphology.

Abstract: The concept that phospholipids, phosphatidyl serine (PS) in particular, play a role in tissue calcification has emerged recently and progressed rapidly. Membranous cellular degradation products upon various tissue injuries apparently serve as nidi of calcium apatite nucleation. In view of the strong affinity of calcium for PS, and the frequent occurrence of calcification in apposition to the inner surface of membrane where PS resides, it is concluded that exposure of PS to the extracellular fluid with a high calcium concentration upon cell injury is largely responsible for the dystrophic calcification. An abundance of membranous vesicles and other unidentified organic matrices were visualized in decalcified urinary stones. The findings also indicate the prospect of the role of degenerate cell membrane in stone formation as well. Furthermore, red cells adherent to bipyramid calcium oxalate dihydrate crystals apparently enhance crystal growth by mechanism analogous to the kink site formation. Lipid matrix in the form of membrane plays an important role in dystrophic calcification as well as calcium containing stone formation.

Abstract: Deposition of calcium-containing apatite mineral occurs widely in association with cardiovascular and noncardiovascular medical devices and biomaterials, is the leading cause of failure of contemporary bioprosthetic heart valves, and limits the functional lifetime of experimental (and potentially clinical) mechanical blood pumps and polymeric heart valves. Calcification of bioprosthetic tissue is primarily intrinsic, related to cuspal connective tissue cells and fragments, and collagen. In contrast, the predominant site of calcific crystals on flexing polymeric surfaces in blood pumps or valve prostheses is extrinsic, associated with adherent cells, thrombus, or pseudointima. Pathologic calcification shares key features with physiologic skeletal mineralization, including crystal initiation through the mediation of cell membranes, usually in the form of extracellular vesicles. This suggests a unified hypothesis for normal and abnormal mineralization. Several approaches are being studied experimentally for the inhibition of bioprosthetic heart valve calcification. Controlled-release diphosphonate therapy, perhaps in conjunction with an anticalcification cuspal pretreatment, appears most effective. Research objectives in biomaterial-associated calcification include (1) development of animal models, (2) determination of initial crystal nucleation events and sites, (3) elucidation of the relative roles of host, implant, and mechanical determinants, and (4) development of approaches for the inhibition of mineralization.

Abstract: Calcification of human aortic valve and aortic media occurs regularly, increases with age, and is distinctively associated with a zone of lipid accumulation. Ultrastructurally, the accumulated lipids are seen as cellular degradation products derived from senescent and degenerate fibrocytes and smooth muscle cells. The products when deposited in the matrix are morphologically similar to the matrix vesicles described in other calcifying tissues, and serve as the initial site of calcification rather than collagen or elastic fibers. Scattered among the smaller and more typical matrix vesicles, there are seen frequently giant vesicle-like structures measuring several microns in diameter. Many of these large calcified bodies contain needle-shaped, radially arranged apatite crystal deposits. Some of the large calcifying bodies are bounded by folded structures suggesting a membrane component, at times obscured by a more dense floccular osmiophilic deposition. Alcianophilic apparent proteoglycan particles are also adherent to these large calcified bodies. The substance forming the large calcified bodies might be a complex of phospholipids derived from cell membrane and proteoglycan derived from ground substance, this combination possible serving as a nidus for calcification.

Abstract: Electron-probe microanalysis (EPM) is an ideal technique with which to study biological calcification. It is particularly effective in identification of crystalline or non-crystalline deposits of minute size in tissues, and in detecting artifacts which may occur during tissue processing. Human aorta, aortic valves, tumours, joint fluid, and calculus specimens were analyzed via scanning electron microscopy (SEM), EPM, transmission electron microscopy (TEM), and selected-area electron diffraction (ED). Crystals found in the specimens were definitively identified by combined SEM-EPM. It is apparent that EPM is an invaluable tool that will potentially improve the acuity of 'in house' laboratory diagnosis of many pathological calcifications.

Abstract: Aging changes of aortic valves are thought to underlie the mechanism of calcification, which leads to calcific aortic stenosis in humans. The study of calcification in the aging valvular connective tissue has been hindered by the lack of a suitable animal model. In search of the model, canine aortic valves demonstrated age changes including calcification remarkably similar to those in humans. The mechanism of calcification was studied in the aortic valves of aged Beagles by electron microscopy. Fibroblasts in the canine aortic valves showed the most prominent age changes. The cells accumulated numerous residual bodies and appeared to disintegrate. The resultant membranous cellular degradation products which sequestered in the extracellular space were the nidi of calcification. It appeared that the membrane of cell debris played an important role in calcification. Canine aortic valve is an ideal model for the study of calcification in relation to aging of the valvular connective tissue.

Twenty five patients were thus premedicated, 25 others received no premedication. All were put to sleep with Alfatesine. The results were judged on the clinical symptoms and the variations of histamine in whole blood. This premedication seems very efficacious, in particular in subjects with a high risk of histamine liberation.

Abstract: Silica calculi in man are extremely rare; only 12 cases have been reported. In an electron microscopic study of human urinary stones, siliceous deposits were encountered in three out of 180 stones as a discrete component of stones. There were no histories of magnesium trisilicate administration in these cases. Morphology of the deposits was distinctive from any other components of the stones and bore some resemblance to animal silica calculi. Siliceous deposits in human urinary stones appear to be more common than generally recognized.

Abstract: In order to study the mechanism of dystrophic calcification, an anoxic incubation of rat renal cortex in a tissue culture medium was performed in vitro. Calcium and phosphate in the medium were adjusted to 1.6 and 1.2 mM/1 respectively. Calcification occurred in apposition to the inner surface of membranous cellular degradation products and associated with the flocculent densities within the degenerate organelles. The chemical nature of the flocculent density was not determined. In view of the known affinity of calcium for acidic phospholipids, particularly phosphatidyl serine (PS), which lines the inner surface of the plasma membrane, calcification along the inner surface of membrane was thought to be related to the presence of PS. Accumulation of calcium in mitochondria, which is presumably dependent upon residual substrate for energy production, appeared to cause calcification as well. Amorphous calcium phosphate in the form of spheroids, and possibly fine fibrils and granules, also appeared to play a role in calcification by their transformation into apatite. The seemingly simple phenomenon of tissue calcification is complex. Nephrocalcinosis in vitro is remarkably similar to the calcification in acute tubular necrosis in vivo, and is a convenient model with which to study the mechanism of calcification. It is concluded that the cellular degradation products are the initial loci of calcification and have a likely role in urolithiasis.

Abstract: Fc epsilon receptor (Fc epsilon R) expression on several human cell lines (U937, RPMI 8866, HL 60, THP-1, and Molt 4) and its regulation were examined by immunofluorescent analysis using a monoclonal anti-human Fc epsilon R antibody, H107. Phorbol ester (PMA), recombinant gamma interferon (IFN-gamma) and H107 itself enhanced Fc epsilon R expression on a FC epsilon R positive cell line U937, whereas these reagents did not induce FC epsilon R expression on the Fc epsilon R negative cell lines, Molt 4, HL 60 and THP-1. Dexamethasone not only suppressed by 50% the spontaneous Fc epsilon R expression on U937 cells but also completely inhibited the enhancement of their Fc epsilon R expression on U937 cells induced by PMA, IFN-gamma or H107. Dexamethasone caused a little suppression of Fc epsilon R expression by RPMI 8866 cells. The results showed that Fc epsilon R expression on a human monoblast cell line U937 was up- or down-regulated by a variety of physiological or pharmacological agents. These experimental systems provide a good model for the investigation of the regulatory mechanisms of Fc epsilon R expression.

190. Low burst-promoting activity (BPA) production by cord mononuclear cells is due to functional immaturity of monocytes

Abstract: Cord blood mononuclear cells produced lower burst-promoting activity (BPA) than adult peripheral blood mononuclear cells when stimulated with phytohemagglutinin (PHA). In order to examine the cellular basis of the low production of BPA by PHA-stimulated cord blood mononuclear cells in the context of the functional immaturity of T cells or monocytes, we studied BPA production by T cells or monocytes from cord blood and adult peripheral blood. Cord T cells produced as much BPA as adult T cells. Monocytes themselves did not produce significant BPA at the concentration used in this experiment (1 x 10(5)/ml). BPA production by adult T cells was significantly enhanced by the presence of autologous monocytes. BPA production by cord T cells was also enhanced by the presence of adult monocytes but not by that of cord monocytes. Cord monocytes did not enhance BPA production by adult T cells either. These results indicate that cord monocytes are primarily responsible for the low BPA production by PHA-stimulated cord mononuclear cells.

191. Dense-core matrical mitochondrial bodies in oncocytic adenoma of the thyroid

Abstract: Single and/or multiple, round and homogeneous electron-dense bodies occurred within the mitochondrial matrix of a mitochondrion-rich follicular adenoma of the thyroid. These non-membrane-bound bodies varied greatly in size and often occupied a large portion of the mitochondrion. They were present in normal-sized or large mitochondria that exhibited cristolysis and/or dilatation of the intracristal space of the residual cristae and formation of intramatrical myelin figures. The globule-containing mitochondria frequently exhibited bundles of parallel intramatrical filamentous inclusions that were 6 to 8 nm in width and were randomly arranged in clusters. Megamitochondria up to 4.4 micrometers in widest diameter occurred in a frequency of one to two per cell and they displayed similar bundles of filamentous inclusions. This evidence of electron-dense bodies in non-steroid-hormone-secreting cells suggests that the bodies result from nonspecific mitochondrial reaction to injury rather than being inclusions specific for steroid-synthesizing cells.

Abstract: Zinc has long been recognized as essential to normal growth and development in animals and in man. Its role in regulating metabolic events at the molecular level is becoming evident, but it is not yet possible to correlate biochemical function to the pathology of deficiency. Studies by a number of investigators have indicated that feeding animals with a diet low in zinc causes an abnormal condition on skin in addition to growth retardation. These include the rats (Todd et al., 1934), mice (Day and Skidmore, 1947), swine (Kernkam and Ferrin, 1953), cattle (Miller and Miller, 1962), lambs (Ott et al., 1964) goats (Miller et al., 1964), dogs (Robertson and Burns, 1963), rabbits (Graham and Telle, 1967), squirrel monkeys (Macapinlac et al., 1967), hamsters (Boquist and Lernmark, 1969), guinea pigs (McBean et al., 1972). In man (Prasad et al., 1963) the clinical aspects of zinc deficiency are remarkably similar to those described above for animals. They consisted of growth retardation and rough skin with hyperpigmentation. These observations indicate the importance of zinc in skin metabolism.

Abstract: Since the last IAEA conference, the scientific programme of JET has focused on the qualification of the integrated operating scenarios for ITER and on physics issues essential for the consolidation of design choices and the efficient exploitation of ITER. Particular attention has been given to the characterization of the edge plasma, pedestal energy and edge localized modes (ELMs), and their impact on plasma facing components (PFCs). Various ELM mitigation techniques have been assessed for all ITER operating scenarios using active methods such as resonant magnetic field perturbation, rapid variation of the radial field and pellet pacing. 200. Electron microscopic pathology of urinary stones

Abstract: Urolithiasis occurs commonly. With modern technology, stones can be easily removed. However, the major threat of urolithiasis is in its high recurrence rate. Many recurrences can be medically prevented. For medical prevention, proper knowledge of stone components is essential.Urinary stones have been analyzed mainly by x-ray diffraction, infrared spectroscopy and chemical analysis. Each method suffers from limitations in sensitivity and reliability. Primarily because of their inability to detect minor components of the stones, two or more methods are frequently combined. Recently it has become apparent that scanning electron microscopy (SEM), especially when combined with x-ray analysis (XA), has a sensivity in stone analysis exceeding other conventional methods.SEM of urinary stones did not gain popularity mainly due to the perceived high cost, the difficulty of sectioning stones, and the variability of crystal morphology.
