The diagnostic power offirst‐order reversal curve (FORC) diagrams has recently beenenhanced by an extended measurement protocol that yields three additional FORC‐like diagrams: theremanent (remFORC), induced (iFORC), and transient (tFORC) diagrams. Here, we present micromagneticsimulations using this extended protocol, including numerical predictions of remFORC, iFORC, and tFORCsignatures for particle ensembles relevant to rock magnetism. Simulations are presented for randomlypacked single‐domain (SD) particles with uniaxial, cubic, and hexagonal anisotropy, and for chains ofuniaxial SD particles. Noninteracting particles have zero tFORC, but distinct remFORC and iFORC signals,that provide enhanced discrimination between uniaxial, cubic, and hexagonal anisotropy types. Increasinginteractions lessen the ability to discriminate between uniaxial and cubic anisotropy but reproduces achange in the pattern of positive and negative iFORC signals observed for SD‐dominated versus vortex‐dominated samples. Interactions in SD particles lead to the emergence of a bi‐lobate tFORC distribution,which is related to formation offlux‐closure in super‐vortex states. A predicted iFORC signal associated withcollapsed chains is observed in experimental data and may aid magnetofossil identification in sediments.Asymmetric FORC and FORC‐like distributions for hexagonal anisotropy are explained by the availability ofmultiple easy axes within the basal plane. A transition to uniaxial switching occurs below a critical value ofthe out‐of‐plane/in‐plane anisotropy ratio, which may allow FORC diagrams to provide insight into thestress state of hexagonal minerals, such as hematite.

The Day diagram is used extensively in rock magnetism for domain state diagnosis. It has beenshown recently to be fundamentally ambiguous for 10 sets of reasons. This ambiguity highlights theurgency for adopting suitable alternative approaches to identify the domain state of magnetic mineralcomponents in rock magnetic studies. We evaluate 10 potential alternative approaches here and concludethat four have value for identifying data trends, but, like the Day diagram, they are affected by use of bulkparameters that compromise domain state diagnosis in complex samples. Three approaches based onremanence curve and hysteresis loop unmixing, whensupervisedby independent data to avoidnonuniqueness of solutions, provide valuable component‐specific information that can be linked byinference to domain state. Three further approaches based onfirst‐order reversal curve diagrams providedirect domain state diagnosis with varying effectiveness. Environmentally important high‐coercivityhematite and goethite are represented with variable effectiveness in the evaluated candidate approaches.These minerals occur predominantly in noninteracting single‐domain particle assemblages inpaleomagnetic contexts, so domain state diagnosis is more critical for ferrimagnetic minerals. Treating thehigh‐coercivity component separately following normal rock magnetic procedures allows focus on the morevexing problem of diagnosing domain state in ferrimagnetic mineral assemblages. We suggest a moveaway from nondiagnostic methods based on bulk parameters and adoption of approaches that provideunambiguous component‐specific domain state identification, among which variousfirst‐order reversalcurve‐based approaches provide diagnostic information.

Fusarium oxysporum is a globally distributed soilborne fungal pathogen causing root rots, bulb rots, crown rots and vascular wilts on a range of horticultural plants. Pathogenic F. oxysporum isolates are highly host specific and are classified as formae speciales. Narcissus is an important ornamental crop and both the quality and yield of flowers and bulbs can be severely affected by a basal rot caused by F. oxysporum f. sp. narcissi (FON); 154 Fusarium isolates were obtained from different locations and Narcissus cultivars in the United Kingdom, representing a valuable resource. A subset of 30 F. oxysporum isolates were all found to be pathogenic and were therefore identified as FON. Molecular characterisation of isolates through sequencing of three housekeeping genes, suggested a monophyletic origin with little divergence. PCR detection of 14 Secreted in Xylem (SIX) genes, previously shown to be associated with pathogenicity in other F. oxysporum f. spp., revealed different complements of SIX7, SIX9, SIX10, SIX12 and SIX13 within FON isolates which may suggest a race structure. SIX gene sequences were unique to FON and SIX10 was present in all isolates, allowing for molecular identification of FON for the first time. The genome of a highly pathogenic isolate was sequenced and lineage specific (LS) regions identified which harboured putative effectors including the SIX genes. Real-time RT-PCR, showed that SIX genes and selected putative effectors were expressed in planta with many significantly upregulated during infection. This is the first study to characterise molecular variation in FON and provide an analysis of the FON genome. Identification of expressed genes potentially associated with virulence provides the basis for future functional studies and new targets for molecular diagnostics.

Genome-wide analyses of the effector- and toxin-encoding genes were used to examine the phylogenetics and evolution of pathogenicity amongst diverse strains of Pseudomonas syringae causing bacterial canker of cherry (Prunus avium), including pathovars P. syringae pv morsprunorum (Psm) races 1 and 2, P. syringae pv syringae (Pss) and P. syringae pv avii. Phylogenetic analyses revealed Psm races and P. syringae pv avii clades were distinct and were each monophyletic, whereas cherry-pathogenic strains of Pss were interspersed amongst strains from other host species. A maximum likelihood approach was used to predict effectors associated with pathogenicity on cherry. Pss possesses a smaller repertoire of type III effectors but has more toxin biosynthesis clusters than Psm and P. syringae pv avii. Evolution of cherry pathogenicity was correlated with gain of genes such as hopAR1 and hopBB1 through putative phage transfer and horizontal transfer respectively. By contrast, loss of the avrPto/hopAB redundant effector group was observed in cherry-pathogenic clades. Ectopic expression of hopAB and hopC1 triggered the hypersensitive reaction in cherry leaves, confirming computational predictions. Cherry canker provides a fascinating example of convergent evolution of pathogenicity that is explained by the mix of effector and toxin repertoires acting on a common host.

The oomycete pathogen Phytophthora cactorum causes crown rot, a major disease of cultivated strawberry. We report the draft genome of P. cactorum isolate 10300, isolated from symptomatic Fragaria x ananassa tissue. Our analysis revealed that there are a large number of genes encoding putative secreted effectors in the genome, including nearly 200 RxLR domain containing effectors, 77 Crinklers (CRN) grouped into 38 families, and numerous apoplastic effectors, such as phytotoxins (PcF proteins) and necrosis inducing proteins. As in other Phytophthora species, the genomic environment of many RxLR and CRN genes differed from core eukaryotic genes, a hallmark of the two-speed genome. We found genes homologous to known Phytophthora infestans avirulence genes including Avr1, Avr3b, Avr4, Avrblb1 and AvrSmira2 indicating effector sequence conservation between Phytophthora species of clade 1a and clade 1c. The reported P. cactorum genome sequence and associated annotations represent a comprehensive resource for avirulence gene discovery in other Phytophthora species from clade 1 and, will facilitate effector informed breeding strategies in other crops.

Verticillium dahliae infection of strawberry (Fragaria x ananassa) is a major cause of disease-induced wilting in soil-grown strawberries across the world. To understand what components of the pathogen are affecting disease expression, the presence of the known effector VdAve1 was screened in a sample of Verticillium dahliae isolates. Isolates from strawberry were found to contain VdAve1 and were divided into two major clades, based upon their vegetative compatibility groups (VCG); no UK strawberry isolates contained VdAve1. VC clade was strongly related to their virulence levels. VdAve1-containing isolates pathogenic on strawberry were found in both clades, in contrast to some recently published findings. On strawberry, VdAve1-containing isolates had significantly higher virulence during early infection, which diminished in significance as the infection progressed. Transformation of a virulent non-VdAve1 containing isolate, with VdAve1 was found neither to increase nor decrease virulence when inoculated on a susceptible strawberry cultivar. There are therefore virulence factors that are epistatic to VdAve1 and potentially multiple independent routes to high virulence on strawberry in V. dahliae lineages. Genome sequencing a subset of isolates across the two VCGs revealed that isolates were differentiated at the whole genome level and contained multiple changes in putative effector content, indicating that different clonal VCGs may have evolved different strategies for infecting strawberry, leading to different virulence levels in pathogenicity tests. It is therefore important to consider both clonal lineage and effector complement as the adaptive potential of each lineage will differ, even if they contain the same race determining effector.

The IABs represent one of only two groups of iron meteorites that did not form by fractional crystallization of liquid Fe-Ni in the core of a differentiated planetesimal. Instead, they are believed to originate from a partially differentiated body that was severely disrupted by one or more impacts during its early history. We present a detailed microstructural and paleomagnetic study of the Odessa and Toluca IAB meteorites, with a view to further constraining the complex history of the IAB parent body. X-ray photoemission electron microscopy and energy dispersive spectroscopy were used to generate high-resolution Ni/Fe maps. The crystallographic architecture of Odessa was analysed using electron backscatter diffraction. Paleomagnetic signals and the magnetic properties of several microstructures were also assessed using X-ray magnetic circular dichroism. Odessa exhibits a complex series of microstructures, requiring an unusual evolution during slow cooling. A conventional Widmanstätten microstructure, consisting of multiple generations of kamacite lamellae surrounded by M-shaped diffusion profiles, developed via continuous precipitation to temperatures below ∼400 °C. Multiple generations of pearlitic plessite nucleated from kamacite/taenite (T > 400 °C) and tetrataenite rim/taenite interfaces (T < 400 °C), via a process of discontinuous precipitation. Rounded rafts of Ni-rich taenite, observed within some regions of pearlitic plessite, are shown to have the same crystallographic orientation as the parental taenite, and a non-standard orientation relationship with the enclosing kamacite. Contrary to current theories, these rafts cannot have formed by coarsening of pre-existing pearlitic plessite. A new bowing mechanism is proposed, whereby rafts of Ni-enriched taenite form between advancing lobes of an irregular reaction front during discontinuous precipitation. Diagenetic alteration of magnetic minerals occurs in all sedimentary environments and tends to be severe in reducing environments. Magnetic minerals provide useful information about sedimentary diagenetic processes, which makes it valuable to use magnetic properties to identify the diagenetic environment in which the magnetic minerals occur and to inform interpretations of paleomagnetic recording or environmental processes. We use a newly developed first-order reversal curve unmixing method on well-studied samples to illustrate how magnetic properties can be used to assess diagenetic processes in reducing sedimentary environments. From our analysis of multiple data sets, consistent magnetic components are identified for each stage of reductive diagenesis. Relatively unaltered detrital and biogenic magnetic mineral assemblages in surficial oxic to manganous diagenetic environments undergo progressive dissolution with burial into ferruginous and sulfidic environments and largely disappear at the sulfate-methane transition. Below the sulfate-methane transition, a weak superparamagnetic to largely noninteracting stable single domain (SD) greigite component is observed in all studied data sets. Moderately interacting stable SD authigenic pyrrhotite and strongly interacting stable SD greigite are observed commonly in methanic environments. Recognition of these characteristic magnetic components enables identification of diagenetic processes and should help to constrain interpretation of magnetic mineral assemblages in future studies. A key question for future studies concerns whether stable SD greigite forms in the sulfidic or methanic zones, where formation in deeper methanic sediments will cause greater delays in paleomagnetic signal recording. Ensuring health of food plants is critical to maintaining the quality and productivity of crops and for sustenance of the rapidly growing human population. There is a close linkage between food security and societal stability; however, global food security is threatened by the vulnerability of our agricultural systems to numerous pests, pathogens, weeds, and environmental stresses. These threats are aggravated by climate change, the globalization of agriculture, and an over-reliance on nonsustainable inputs. New analytical and computational technologies are providing unprecedented resolution at a variety of molecular, cellular, organismal, and population scales for crop plants as well as pathogens, pests, beneficial microbes, and weeds. It is now possible to both characterize useful or deleterious variation as well as precisely manipulate it. Data-driven, informed decisions based on knowledge of the variation of biotic challenges and of natural and synthetic variation in crop plants will enable deployment of durable interventions throughout the world. These should be integral, dynamic components of agricultural strategies for sustainable agriculture.

Background: Accurate and quantitative phenotypic data in plant breeding programmes is vital in breeding to assess the performance of genotypes and to make selections. Traditional strawberry phenotyping relies on the human eye to assess most external fruit quality attributes, which is time-consuming and subjective. 3D imaging is a promising high-throughput technique that allows multiple external fruit quality attributes to be measured simultaneously. Results: A low cost multi-view stereo (MVS) imaging system was developed, which captured data from 360° around a target strawberry fruit. A 3D point cloud of the sample was derived and analysed with custom-developed software to estimate berry height, length, width, volume, calyx size, colour and achene number. Analysis of these traits in 100 fruits showed good concordance with manual assessment methods. Conclusion: This study demonstrates the feasibility of an MVS based 3D imaging system for the rapid and quantitative phenotyping of seven agronomically important external strawberry traits. With further improvement, this method could be applied in strawberry breeding programmes as a cost effective phenotyping technique.
