196. Psychotropic and Neuroreceptor Effects of Cyclopropylglycine Upon Intranasal Administration.

Abstract

The effects of chronic intranasal administration of cycloprolylglycine (CPG, 1 and 2 mg/kg) on the behavior of BALB/c and C57BL/6 mice and the brain neuroreceptor characteristics in BALB/c mice were studied. Intranasal administration of CPG (1 and 2 mg/kg, 2 weeks) to BALB/c mice in the Porsolt test resulted in a significant antidepressant-like effect more potent than that from intraperitoneal administration of CPG. At the same time, intranasal administration of CPG to C57BL/6 mice in the tail-suspension test was the same as that upon intraperitoneal administration and led to a moderate antidepressant-like effect (1 mg/kg, 4 weeks). Intranasal CPG did not produce any effect on anxiety or exploratory activity in the closed cross-maze test. Radioligand analyses showed an increased density of GABAA receptors in the PFC (1 mg/kg) and reduced density of NMDAand 5HT2A receptors in the hippocampus and striatum, respectively, after intranasal administration of CPG (1 and 2 mg/kg) for two weeks while not affecting mGluII receptor binding in the hippocampus. The significant difference in hippocampal NMDA binding between the two routes may indicate that this receptor is involved in the mechanism of the antidepressant-like effect of CPG. Abstract

The study examined the effect of GTS-201, a low-molecular weight mimetic of brain-derived neurotrophic factor (BDNF) loop 2, on persistent alcohol craving in outbred male and female albino rats with ethanol preference score ~50% developed in the free choice paradigm between 10% ethanol and water over 24 weeks. Both single and subchronic (5 days) injections of GTS-201 in a daily dose of 5 μg/kg reduced alcohol deprivation effect in female, but not in male rats. The possibility of in vivo sex-dependent regulation of modeled alcohol craving with a low-molecular-weight dipeptide mimetic of BDNF loop 2 was demonstrated and sex-related differences in this effect were revealed [ABSTRACT FROM AUTHOR]

Abstract

Experimental results for the new nerve growth factor (NGF) mimetic GK-2 [bis-(N-monosuccinyl-L-glutamyl-L-lysine)hexamethylenediamide] administered perorally toWistar rats in a streptozotocin-induced type 2 diabetes mellitus model are reported. The antihyperglycemic activity of GK-2 was determined in a dose range from 1 to 10 mg/kg. The most pronounced effect was observed at doses of 2.5 and 5 mg/kg. GK-2 was also found to reduce the severity of polydipsia and polyphagia and to produce a positive effect on the body-weight dynamics of diabetic animals. The dose dependence of GK-2 effects was described by a bell-shaped curve typical of regulatory peptides with various chemical structures. The ten-fold greater width of the dose–activity curve of the drug observed in this work combined with its high safety suggests that a satisfactory therapeutic index for GK-2 as a potential antidiabetic drug for peroral administration can be expected. Abstract

A new series of bis-(2,3,4-trimethoxybenzyl)alkanediamines were obtained. Their anti-ischemic and antiarrhythmic activities were studied. Replacement of the central N atom of the triazaalkane linker in the previously studied compound ALM-802 by a carbon atom and shortening of the linker length led to preservation of the cardiotropic activity spectrum of the corresponding compounds. The most active compounds among the new series were ALM-844 [N1,N5-bis(2,3,4-trimethoxybenzyl)pentane-1,5-diamine dihydrochloride] and ALM-851 [N1,N3-bis(2,3,4-trimethoxybenzyl)propane-1,4-diamine dihydrochloride].
