Abstract

Wave is a common environmental load that often causes serious damages to offshore structures. In addition, the stability for the submarine artificial slope is also affected by the wave loading. Although the landslide of submarine slopes induced by the waves received wide attention, the research on the influence of solitary wave is rare. In this study, a 2-D integrated numerical model was developed to investigate the stability of the foundation trench under the solitary wave loading. The Reynolds-averaged Stokes (RANS) equations were used to simulate the propagation of a solitary wave, while the current was realized by setting boundary inlet/outlet velocity. The pore pressure induced by the solitary wave was calculated by Darcy’s law, and the seabed was characterized by Mohr–Coulomb constitutive model. Firstly, the wave model was validated through the comparison between analytical solution and experimental data. The initial consolidation state of slope under hydrostatic pressure was achieved as the initial state. Then, the factor of stability (FOS) for the slope corresponding to different distances between wave crest and slope top was calculated with the strength reduction method. The minimum of FOS was defined as the stability index for the slope with specific slope ratio during the process of dynamic wave loading. The parametric study was conducted to examine the effects of soil strength parameters, slope ratio, and current direction. At last, the influence of upper slope ratio in a two-stage slope was also discussed.

156. The relationship between neutrophil-lymphocyte ratio and early renal fibrosis and renal prognosis in patients with lupus nephritis

Abstract

Objective: To explore the relationship between neutrophil-lymphocyte ratio (NLR) and early renal fibrosis and renal prognosis in patients with lupus nephritis. Methods: A total of 186 patients with lupus nephritis admitted to our hospital were enrolled and grouped according to the standard of “NLR=3.175”. There were 90 patients in the higher NLR group and 96 patients in the lower NLR group. The correlation and independent relationship between NLR/estimated glomerular filtration rate (eGFR) and pathological indicators were explored by comparing the differences of physiological indicators between the two groups. The correlation between NLR/eGFR and renal insufficiency and renal prognosis were explored. Results: The higher NLR group showed increased levels of hs-CRP, white blood cells, neutrophils, platelets, PLR values, eGFR, D-dimer, crescent compared with the lower NLR group, while lymphocyte count in the higher NLR group was lower than that in the lower NLR group. Platelets, neutrophils, white blood cells, serum anhydride, serum C4 and vascular cellulose had positive correlations with NLR. eGFR, age, systolic blood pressure, diastolic blood pressure, neutrophils, leukocytes, NLR, blood uric acid, triglycerides, crescent ratio, endothelial hyperplasia, spherical sclerosis ratio, intraglomerular leukocyte infiltration, platinum loop, microthrombus, nuclear fragmentation, interstitial inflammatory cell infiltration, renal tubular atrophy, interstitial fibrosis, and mesangial cell showed negative correlation with matrix hyperplasia, arteriolar wall thickening and pathological activity scores, but showed positive correlation with hemoglobin and low-density lipoprotein. Conclusion: NLR affects some important renal functions to a certain extent and is not the only risk factor for renal prognosis in patients with LN.

Abstract

The material innovation is prerequisite to accelerating sluggish oxygen evolution reaction (OER) kinetics, thus promoting the realization of hydrogen energy community. Herein, we develop an oxygen-vacancy-rich TiO2 supported RuO2 catalyst (RuO2@r-TiO2) towards improved OER activity and stability. The oxygen vacancy on TiO2 not only supplies electrons to produce lower valence Ru, but also provides sufficient anchoring site for the deposition of RuO2 nanocrystal. Beyond that, it can generate strong electronic interaction between TiO2 and supported RuO2, and thereby tailors the intermediates' adsorption energy on the RuO2 surface. As a result, the derived RuO2@r-TiO2 catalyst exhibits superior OER activity and stability with the overpotential of 211 mV at a current density of 10 mA cm−2 and negligible activity degradation after 6 h operation, outperforming the non-oxygen-vacancy counterpart (223.3 mV, 12.75% activity loss) and RuO2 catalyst (234.6 mV, 42.86% activity loss). Abstract

As one of the few animals with variation in the number of rib pairs (RIB), the pig is a good model to study the mechanism of RIB regulation. Quantitative trait loci (QTL) for porcine RIB are present on Sus scrofa chromosome 7 (SSC7). Although several candidate genes exist in this QTL region on SSC7, the causal gene has yet to be verified. Beijing Black pig with 14–17 RIB is a good population for candidate gene mining and 1104 individuals were genotyped using the Illumina Porcine 50K BeadChip. A total of 14 SNPs from 95.49 to 97.78 Mb on SSC7 showed genome‐wide significant association with RIB. On SSC7, a locuszoom plot using pairwise linkage disequilibrium displayed the narrowest linkage region encompassing only two genes, ABCD4 and VRTN. In mice, a transcriptome expression profile was obtained using three embryos at E9.5 (the critical period for rib formation). ABCD4 was highly expressed, but no expression of VRTN was detected. On SSC6, there were four genome‐wide significant SNPs from 106.42 to 106.92 Mb associated with RIB. GREB1L and MIB1, in this region, were regarded as novel candidate genes. These results revealed a crucial candidate causal gene on SSC7 and novel genes on SSC6 for rib number and provided interesting new insights into its genetic basis. [ABSTRACT FROM AUTHOR]

Abstract

Background The critical role of antibody Fc-mediated effector functions in immune defense has been widely reported in various viral infections. These effector functions confer cellular responses through engagement with innate immune cells. The precise mechanism(s) by which immunoglobulin G (IgG) Fc domain and cognate receptors may afford protection are poorly understood, however, in the context of HIV/SHIV infections. Many different in vitro assays have been developed and utilized to measure effector functions, but the extent to which these assays capture distinct antibody activities has not been fully elucidated. Results In this study, six Fc-mediated effector function assays and two biophysical antibody profiling assays were performed on a common set of samples from HIV-1 infected and vaccinated subjects. Biophysical antibody profiles supported robust prediction of diverse IgG effector functions across distinct Fc-mediated effector function assays. While a number of assays showed correlated activities, supervised machine learning models indicated unique antibody features as primary contributing factors to the associated effector functions. Additional experiments established the mechanistic relevance of relationships discovered using this unbiased approach. Conclusions In sum, this study provides better resolution on the diversity and complexity of effector function assays, offering a clearer perspective into this family of antibody mechanisms of action to inform future HIV-1 treatment and vaccination strategies.

Diverse Ab effector functions mediated by the Fc domain have been commonly associated with reduced risk of infection in a growing number of nonhuman primate and human clinical studies. This study evaluated the anti-HIVAb effector activities in polyclonal serum samples from HIV-infected donors, VAX004 vaccine recipients, and healthy HIV-negative subjects using a variety of primary and cell line-based assays, including Ab-dependent cellular cytotoxicity (ADCC), Ab-dependent cell-mediated viral inhibition, and Abdependent cellular phagocytosis. Additional assay characterization was performed with a panel of Fc-engineered variants of mAb b12. The goal of this study was to characterize different effector functions in the study samples and identify assays that might most comprehensively and dependably capture Fc-mediated Ab functions mediated by different effector cell types and against different viral targets. Deployment of such assays may facilitate assessment of functionally unique humoral responses and contribute to identification of correlates of protection with potential mechanistic significance in future HIV vaccine studies. Multivariate and correlative comparisons identified a set of Ab-dependent cell-mediated viral inhibition and phagocytosis assays that captured different Ab activities and were distinct from a group of ADCC assays that showed a more similar response profile across polyclonal serum samples. The activities of a panel of b12 monoclonal Fc variants further identified distinctions among the ADCC assays. These results reveal the natural diversity of Fc-mediated Ab effector responses among vaccine recipients in the VAX004 trial and in HIV-infected subjects, and they point to the potential importance of polyfunctional Ab responses.

172. Achieving Ultrasensitive Chromogenic Probes for Rapid, Direct Detection of Carbapenemase-Producing Bacteria in Sputum

Abstract

Carbapenemase-producing bacteria (CPB) stand as the most dangerous “superbugs” in the clinic. Rapid point-of-care (POC) detection of CPB in clinical samples is key to timely and effective infection management. We herein report the first ultrasensitive chromogenic probe that allows direct POC detection of CPB in clinical sputum samples at a sample-to-result time of less than 15 min. This chromogenic probe is modularly designed by conjugating the carbapenem core with a benzene derivative bearing an electronegativity-tunable substituent. Unexpectedly high sensitivity was achieved simply by choosing strong electron-withdrawing substituents, such as −N+(CH3)3, without resorting to complex molecular design. Through integrating the probes with a portable paper chip, 24 out of 80 clinical sputum samples from sepsis patients with lung infections were quickly diagnosed as CPB-positive, exhibiting 100% clinical sensitivity and specificity. This low-cost paper chip assay can be readily performed on-site, breaking through the dilemma of rapid CPB detection, especially in resource-limited settings.

178. Acquiring Offshore Project Sanction during Low Oil Price Environment: Rejuvenating Reservoir with Depleted Reservoir Energy via Innovative Low Capex Gas Injection Project

Abstract

Maturing a secondary recovery project in an offshore multi-fault complex reservoir is challenging because considerable investment is required, especially in a low oil price environment and/or when the asset can no longer produce. However, an 85% reduction in cost with no reduction in reserves was achieved for such a reservoir through de-risking the opportunity and revising the development concept, allowing the project to be sanctioned during a low oil price environment. A water injection project with an estimated cost of 165 MMUSD was initially envisaged during the feasibility stage to increase the oil recovery of a depleted reservoir, and contractual expectations were agreed accordingly. This is due to the availability of seawater offshore and water injection being the standard improved recovery approach in the region. Without adhering to the familiarity, a reservoir simulation study was performed comparing multiple options of secondary recovery. The reservoir simulation indicated that gas injection would provide higher incremental recovery while the investment can be significantly reduced by utilizing the existing gas lift system as a gas injection system. Several improved oil recovery schemes were evaluated with the history matched dynamic model to address the depleted energy. This includes water dumpflood, water injection, and gas injection with recovery factors of 32%, 56%, and 58%, respectively, in comparison to the depletion drive recovery factor of 30%. The gas injection scheme provides the highest recovery factor and expected fastest response and thus was prioritized as the main focus over water injection. The existing gas compressor with ullage, gas source from current associated gas production, and existing idle wells are key enablers for the project. The existing assets also enable the acceleration and further de-risking by a pilot gas injection before drilling infills. The paper highlights the numerous examples of cost-saving initiatives while maximizing the oil recovery from the reservoir. This value-focused approach enabled the project to be sanctioned during a period where most offshore improved recovery projects were abandoned due to the low oil price.

188. Disturbance Observer-based Nonsingular Fast Terminal Sliding Mode Fault Tolerant Control of a Quadrotor UAV with External Disturbances and Actuator Faults.

Abstract

This paper addresses a nonsingular fast terminal sliding mode fault tolerant control (NFTSMFTC) scheme based on disturbance observer (DO) for a quadrotor UAV in the presence of external disturbances and actuator additive faults. Firstly, by analyzing the dynamic characteristics of a quadrotor UAV, the model is decoupled into outer-loop (position subsystem) and inner-loop (attitude subsystem). Secondly, the DO is utilized to deal with external disturbances and actuator additive faults effectively. Furthermore, nonsingular fast terminal sliding mode control (NFTSMC) can avoid the chattering problems of traditional sliding mode control (SMC) and singularity problems of terminal SMC. Moreover, compared with nonsingular terminal SMC, it behaves the faster convergence rate. So the NFTSMC method is applied to design controller. Finally, in the framework of Lyapunov theory, the stability of the closed-loop system is analyzed, and the tracking error is asymptotically stable. This study focuses on the evolution of the fraction of rattling particles in deviator deformation until the critical state. Numerical simulations using the discrete element method reveal the presence of rattling particles (with zero or only one contact with neighbouring particles) even in a very dense packing system. The results show that the initial fraction of rattling particles depends on sample density and particle size distribution. With the increase of deviator strain, the number and volume fractions of rattling particles gradually approach a steady critical state from either a loose or a dense starting point. An effective void ratio, which is calculated by treating rattling particles as voids, can be viewed as new state parameter describing the effective packing density of sands.
