Abstract

At the Istituto Nazionale di Fisica Nucleare - Laboratori Nazionali del Sud (INFN-LNS) the beam commissioning of the high intensity Proton Source for the European Spallation Source (PS-ESS) was completed in November 2017. The ESS requires a high intensity proton beam (74 mA pulsed at 14 Hz of repetition rate), with fast Beam pulse rise/fall time (< 20 μs). In order to meet the project requirement, an electrostatic chopping system has been used in the Low Energy Beam Transport (LEBT). The design of the control system was done also to be the main element of the fast beam abort system and taking into account the radiation issue in the accelerator tunnel. This paper describes the performances of the chopper. The experimentally-achieved rise/fall times of the beam pulses measured by using an AC Current Transformer (ACCT) at the end of the LEBT collimator, are presented. An experimental investigation of the effects of different amounts and types of gas injected into the LEBT (for the sake of space charge compensation) has been carried out with respect to the beam and chopper parameters., Proceedings of the 9th Int. 162. Differential proteomic analysis highlights metabolic strategies associated with balhimycin production in Amycolatopsis balhimycina chemostat cultivations

Abstract

Abstract Background Proteomics was recently used to reveal enzymes whose expression is associated with the production of the glycopeptide antibiotic balhimycin in Amycolatopsis balhimycina batch cultivations. Combining chemostat fermentation technology, where cells proliferate with constant parameters in a highly reproducible steady-state, and differential proteomics, the relationships between physiological status and metabolic pathways during antibiotic producing and non-producing conditions could be highlighted. Results Two minimal defined media, one with low Pi (0.6 mM; LP) and proficient glucose (12 g/l) concentrations and the other one with high Pi (1.8 mM) and limiting (6 g/l; LG) glucose concentrations, were developed to promote and repress antibiotic production, respectively, in A. balhimycina chemostat cultivations. Applying the same dilution rate (0.03 h-1), both LG and LP chemostat cultivations showed a stable steady-state where biomass production yield coefficients, calculated on glucose consumption, were 0.38 ± 0.02 and 0.33 ± 0.02 g/g (biomass dry weight/glucose), respectively. Notably, balhimycin was detected only in LP, where quantitative RT-PCR revealed upregulation of selected bal genes, devoted to balhimycin biosynthesis, and of phoP, phoR, pstS and phoD, known to be associated to Pi limitation stress response. 2D-Differential Gel Electrophoresis (DIGE) and protein identification, performed by mass spectrometry and computer-assisted 2 D reference-map http://www.unipa.it/ampuglia/Abal-proteome-maps matching, demonstrated a differential expression for proteins involved in many metabolic pathways or cellular processes, including central carbon and phosphate metabolism. Interestingly, proteins playing a key role in generation of primary metabolism intermediates and cofactors required for balhimycin biosynthesis were upregulated in LP. Finally, a bioinformatic approach showed PHO box-like regulatory elements in the upstream regions of nine differentially expressed genes, among which two were tested by electrophoresis mobility shift assays (EMSA). Conclusion In the two chemostat conditions, used to generate biomass for proteomic analysis, mycelia grew with the same rate and with similar glucose-biomass conversion efficiencies. Global gene expression analysis revealed a differential metabolic adaptation, highlighting strategies for energetic supply and biosynthesis of metabolic intermediates required for biomass production and, in LP, for balhimycin biosynthesis. These data, confirming a relationship between primary metabolism and antibiotic production, could be used to increase antibiotic yield both by rational genetic engineering and fermentation processes improvement.

Abstract Background The alterations of the glutathione peroxidase enzyme complex system occur in physiological conditions such as aging and oxidative stress consequent to strenuous exercise. Methods Authors optimize the spectrophotometric method to measure glutathione peroxidase activity in rat red blood cell membranes. Results The optimization, when applied to age paired rats, both nulligravid and pregnant, shows that pregnancy induces, at seventeen d of pregnancy, an increase of both reactive oxygen substance concentration in red blood cells and membrane glutathione peroxidase activity. Conclusion The glutathione peroxidase increase in erythrocyte membranes is induced by systemic oxidative stress long lasting rat pregnancy.

Abstract

At the Istituto Nazionale di Fisica Nucleare Laboratori Nazionali del Sud (INFN-LNS) the beam commissioning of the high intensity Proton Source for the European Spallation Source (PS-ESS) started in November 2016. Beam stability at high current intensity is one of the most important parameter for the first steps of the ongoing commissioning. Promising results were obtained since the first source start with a 6 mm diameter extraction hole. The increase of the extraction hole to 8 mm allowed improving PS-ESS performances and obtaining the values required by the ESS accelerator. In this work, extracted beam current characteristics together with Doppler shift and emittance measurements are presented, as well as the description of the next phases before the installation at ESS in Lund., Proceedings of the 8th Int. Abstract

The Superconducting Cyclotron (CS) is a three sectors compact accelerator with a wide operating diagram, capable of accelerating heavy ions with q/A from 0.1 to 0.5 up to energies from 2 to 100 MeV/u. The proposed upgrade to increase the light ion beam intensity by means of extraction by stripping implies many modifications of the median plane. The main activities of the mechanical upgrade are: the actuation of the new magnetic channels for the extraction by stripping and the realization of the two extraction modes, by stripping and by electrostatic deflection. For the magnetic channels and compensating iron bars, we are studying the problems of mechanical handling. To obtain the two extraction modes, we are trying to design a new set that allows for the exhange of two devices: electrostatic deflectors and and stripper with its magnetic channels for stripping extraction., Proceedings of the 21st Int. Abstract

DESI (Dark Energy Spectroscopic Instrument) is a Stage IV ground-based dark energy experiment that will study baryon acoustic oscillations (BAO) and the growth of structure through redshift-space distortions with a wide-area galaxy and quasar redshift survey. To trace the underlying dark matter distribution, spectroscopic targets will be selected in four classes from imaging data. We will measure luminous red galaxies up to $z=1.0$. To probe the Universe out to even higher redshift, DESI will target bright [O II] emission line galaxies up to $z=1.7$. Quasars will be targeted both as direct tracers of the underlying dark matter distribution and, at higher redshifts ($ 2.1 < z < 3.5$), for the Ly-$\alpha$ forest absorption features in their spectra, which will be used to trace the distribution of neutral hydrogen. When moonlight prevents efficient observations of the faint targets of the baseline survey, DESI will conduct a magnitude-limited Bright Galaxy Survey comprising approximately 10 million galaxies with a median $z\approx 0.2$. In total, more than 30 million galaxy and quasar redshifts will be obtained to measure the BAO feature and determine the matter power spectrum, including redshift space distortions.

Abstract

The NUMEN (NUclear Matrix Elements for Neutrinoless double beta decay) project was recently proposed with the aim to investigate the nuclear response to Double Charge Exchange reactions for all the isotopes explored by present and future studies of 0νββ decay. The expected level of radiation in the NUMEN experiment imposes severe limitations on the average lifetime of the electronic devices. During the experiments, it is expected that the electronic devices will be exposed to about 105 neutrons/cm2/s according to FLUKA simulations. This paper investigates the reliability of a System On Module (SOM) under neutron radiation. The tests were performed using thermal, epithermal, and fast neutrons produced by the Instituto de Pesquisas Energéticas e Nucleares 4.5 MW Nuclear Research Reactor. The results show that the National Instruments SOM is robust to neutron radiation for the proposed applications in the NUMEN project. [ABSTRACT FROM AUTHOR]

Abstract

The biological treatment with bio-slurry reactors has been effectively considered as a remediation technology for the removal of organic pollutants from soils or sediments, characterized mainly by sandy and clayey fractions. In this context, the treatment of marine sediments contaminated by hydrocarbons, through a remediation technique, represent a topic of particular interest for the scientific community. In this work, bioslurry technology has been studied for the treatment of marine contaminated sediments with a Total Petroleum Hydrocarbons (TPH) content of 888.57 mg kg-1. The experimental campaign was divided into three phases, for a period of about 75 days. TPH removal efficiency was evaluated in two reactors, operating in parallel and in batch mode. In the first phase, reactors had the same characteristics and operating conditions while, in the second phase, the erythromycin antibiotic was added into one of the two reactors. Surprisingly, the addition of erythromycin improved the TPH removal efficiencies and reduced treatment times. Finally, the third phase was characterized by the substitution of the liquid phase, present in the reactors, with saline water, in order to deepen the study on the removal and transfer mechanisms typical of the bio-slurry systems. We report the draft genome sequence of Microbispora sp. strain ATCC-PTA-5024, a soil isolate that produces NAI-107, a new lantibiotic with the potential to treat life-threatening infections caused by multidrug-resistant Gram-positive pathogens. The draft genome of strain Microbispora sp. ATCC-PTA-5024 consists of 8,543,819 bp, with a 71.2% G+C content and 7,860 proteincoding genes.

Abstract

A 2.45 GHz ' 0.1 T microwave discharge Proton Source has been designed and assembled at INFN-LNS for the European Spallation Source (PS-ESS) in order to produce pulsed beams of protons up to 74 mA nominal current, at 75 keV of energy, with a transverse emittance containing 99 % of the nominal proton current below 2.25 π mm mrad and a beam stability of ± 2 %. The challenging performances of the machine have triggered specific studies on the maximization of the proton fraction inside the plasma and of the overall plasma density, including dedicated modelling of the wave-to-plasma interaction and ionization processes. The plasma conditioning phase started in July and excellent RF to plasma coupling, more than 99.5% is evident since the beginning. Reflected power fluctuation less than 0.05 % was measured providing a great starting point to reach the beam stability requested by the ESS accelerator., Proceedings of the 22nd Int. Abstract

Charged particle acceleration using ultra-intense and ultra-short laser pulses has gathered a strong interest in the scientific community and it is now one of the most attractive topics in the relativistic laser-plasma interaction research. Indeed, it could represent the future of particle acceleration and open new scenarios in multidisciplinary fields, in particular, medical applications. One of the biggest challenges consists of using, in a future perspective, high intensity laser-target interaction to generate high-energy ions for therapeutic purposes, eventually replacing the old paradigm of acceleration, characterized by huge and complex machines. Abstract

The chemical synthesis of novel chemotherapeutical leads is evolving thanks to possibility to design molecules with desired physical-chemical and, thus, biological properties. The imidazolium salts, recently proven effective to inhibit bacterial and/or cancer cell growth, posses an amphiphilic nature that is conferred by the imidazolium cation having a polar head generally coupled with aliphatic side chains. Thus, biological properties of imidazolium salts can be tuned through modifications involving the cation structure and/or the anion nature. By covalently linking two imidazolium rings, di-imidazolium salts were obtainedobtain differing in: i) kind of anions; ii) geometric isomerization of di-imidazolium anions; iii) length of the imidazolium alkyl side chains. Preliminarly, eleven di-imidazolium salts, differing for their anionic counterparts, were assayed for: i) antibacterial property, quantified as the minimal concentration inhibiting at least the 90% of bacterial growth (MIC90) using Escherichia coli and Micrococcus luteus as Gram-negative and Gram-positive tester strains; ii) antitumoral activity measured as the concentration inhibiting the 50% of cell growth (IC50) using SKBR-3 breast cancer cell line. All the assayed salts possesses biological activity showing i) 0.1-0.5 and 25-50 µg/ml as MIC90 values against of M. luteus, and E. coli; respectively, and ii) 30-55 µg/ml as IC50 values. Among the tested di-imidazolium salts, three were chosen to further investigate the relationship between biological efficacy and either length of alkyl side chains or imidazolium ring configurations. Geometric isomerization revealed few or no effect while a positive correlation between alkyl chain length and cell-growth inhibitory efficacy was shown. Although further studies have to be performed to elucidate the molecular mechanisms leading to cell growth arrest, this study provide insights on the attractive possibility of di-imidazolium salt exploitation as chemotherapeutical compounds whose activity can be tuned by modifying structural characteristics.

Abstract

In this work, we prepared poly(caprolactone) (PCL) monofilaments incorporating different amounts of the antiseptic chlorhexidine diacetate (CHX) and evaluated the effect of "online" drawing during the melt spinning or of an "offline" cold drawing on the antimicrobial and physical properties of PCL/CHX monofilaments. The results showed that the drawing conditions strongly affected the properties of the threads. In particular, the filaments exhibited antimicrobial activity for all the draw ratios (DR) adopted, but on increasing DR the antibacterial efficiency reduced. These results were correlated with the CHX surface concentration and with the release kinetics.

Abstract

Biocompatible Ag nanocomposites were prepared by photoreduction of ammoniacal silver acetate in the presence of a polyaminocyclodextrin, namely the poly-{6-[3-(2-(3-aminopropylamino)-ethylamino)-propylamino]}-(6-deoxy)-b-CD (amCD, figure 1). The obtained Ag-amCD systems, which possess an oniontype structure [1] with a metal core surrounded by several layers of the capping agent, were characterized by means of various complementary techniques. In particular, FT-IR spectroscopy confirmed the presence of the amCD scaffold in the composite, and evidenced a partial oxidative degradation of the polyamine branches,due to the fact that these groups function as sacrificial reducing agents in the photoinduced process of formation of the Ag metal core. TEM and SAED micrographs evidenced that the Ag cores possess a relatively low polydispersity and a significantly crystalline character.Then, in consideration of the well-known antimicrobial activityof nanosized silver, our Ag-amCD systems were assayed for antibacterial activity, quantified as the minimal concentration inhibiting at least the 90% of bacterial growth (MIC90), using Escherichia coli and Kocuria rhizophila as Gram-negative and Gram-positive tester strains. This analysis revealed 5 and 1 μg/ml as MIC90 values against E. coli and K. rhizophila; respectively. In addition, thanks to their peculiar features, the systems function as potential supramolecular drug carriers, effectively able to bind the b-lactam antibiotic Ampicillin (amp, figure 2) as demonstrated by polarimetric measurements. Antimicrobial assays reveals a five-fold improved activity of Ag-amCD-amp probablydue to synergistic action of Agnanoparticles and amp.
