Aquaculture pond is a complex ecosystem where the microorganisms in the sediments, in the animal intestinal tract and in water interact with each other to influence the water quality and health of aquatic animals. In order to understand the spatial distribution and relationship of microbial communities in intensively farmed genetically improved farmed tilapia (GIFT, Oreochromis niloticus), 454 high-throughput pyrosequencing was applied to analyse the 16S rRNA gene of bacteria in intensive GIFT ponds in Wuxi City, Jiangsu Province, China. A total of 72 747 initial sequences were obtained from four depths of pond water, from tilapia large intestines and from pond sediment. The most common phylum in all samples was Proteobacteria, Actinobacteria and Cyanobacteria were the most abundant in water, Fusobacteria and Firmicutes in the large intestine and Chloroflexi in sediment samples. The sediment microbial community structure was comparatively similar to that of the tilapia large intestine. The microbial communities from different water depths were somewhat similar, especially the three most shallow samples, although the abundance of Actinobacteria gradually decreased with increased water depth. This data offer a preliminary exploration of the response mechanisms of the bacterial communities to aquafarming and contributes to the understanding of the status of bacterial communities of tilapia pond systems during the peak period of breeding from the aspect of their spatial distributions. The complete mitochondrial genome ofHyporhamphus intermediuswas determined to be 16,720 bp in length with (A + T) content of 56.3%, and it consists of 13 protein-coding genes, 22 tRNAs, two ribosomal RNAs, and a control region. The gene composition and the structural arrangement of theH. intermediuscomplete mtDNA were identical to most of the other vertebrates. Interestingly, two tandem repeat units were identified across tRNA-Pro and control region (2 41 bp), while in most of the fishes the tandem repeat units are located in the control region. Antibiotic resistance genes (ARGs) are emerging contaminants that pose a potential risk to human health worldwide. In this study, a metagenomic analysis was performed to investigate the distribution of ARGs in paddy field ecosystems, crayfish monoculture pond ecosystems, and rice-crayfish cultivation field ecosystems. The results showed that MacB and BcrA are two dominant ARGs, and macrolide is the dominant antibiotic not only in the water, but also in the sediment and gut of crayfish, in both the crayfish monoculture and the rice-crayfish cultivation ecosystems. Meanwhile, some lower-abundance ARGs in the water and sediment of crayfish cultivation ecosystems were significantly different in their abundance than those in rice paddy fields, and the ARGs in crayfish culture system and rice paddy fields showed even higher dissimilarities of diversity. Comprehensive analysis with redundancy analysis（RDA）and the distribution of dominant ARGs showed that the dissimilarity was related to the higher concentrations of total nitrogen (TN), total phosphorus (TP), chlorophyll a (Chla), permanganate index (COD Mn) , and nitrate in the water of rice paddy fields, and was related to the higher contents of N, P, K, and organic matter (OM) in the sediment of rice paddy fields. The source trackers of ARGs within the crayfish cultivation ponds and the rice-crayfish cultivation fields showed that the sediment in crayfish cultivation ponds mainly played the role of the 'sink' for ARGs, and the water and gut of crayfish mutually contributed to the high rates of ARGs. The ARG contribution rates of crayfish gut and sediment decreased and increased, respectively, in rice-crayfish cultivation fields and in crayfish monoculture ponds, which might be related to the lower crayfish biomass and the lower water depth in rice-crayfish cultivation fields. The widespread use of carbamate pesticides has led to numerous environmental and health concerns, including water contamination and perturbation of endocrine homeostasis among organisms. However, there remains a paucity of research elucidating the specific effects of methomyl on gut microbial composition and physiological functions. This study aimed to investigate the intricate relationship between changes in zebrafish bacterial communities and intestinal function after 56 days of sub-chronic methomyl exposure at environmentally relevant concentrations (0, 0.05, 0.10, and 0.20 mg/L). Our findings reveal significant methomyl-induced morphological changes in zebrafish intestines, characterized by villi shortening and breakage. Notably, methomyl exposure down-regulated nutrient and energy metabolism, and drug metabolism at 0.05−0.10 mg/L, while up-regulating cortisol, inflammation-related genes, and apoptotic markers at 0.20 mg/L. These manifestations indicate physiological stress imposition and disruption of gut microbiota equilibrium, impacting metabolic processes and instigating low-grade inflammatory responses and apoptotic cascades. Importantly, changes in intestinal function significantly correlated with shifts in specific bacterial taxa abundance, including Shewanella , Rubrobacter , Acinetobacter , Bacillus , Luteolibacter , Nocardia , Defluviimonas , and Bacteroides genus. Here we used RNA-Seq to explore the transcriptomic response and specific involvement of hepatic mRNA of juvenile Oreochromis niloticus (GIFT) as a result of dietary resveratrol supplementation (0.05 g/kg RES). More than 24,513,018 clean reads were reference genome guided assembly into 23,417 unigenes. 12,596 unigenes (29.64%) were annotated to GO database. There were 5, 179 and 1526 genes significantly differentially expressed genes at 15, 30 and 45 d respectively, and 8 KEGG pathways were enriched associated with this immune response. Hyperemia and compressed hepatic sinusoid, fibrosis of liver cell and abnormal hepatic epidermal cell revealed by H&E and SEM analysis respectively. Genes related with cytokine production (il12rb2 , scfr), immune system (ig8l , hlfl , cd226 , prf1l), autophagy regulation (atg4b), foxo signaling (ccnb2), steroid hormone biosynthesis (cyp3a40), fatty acid metabolism (scd1), metabolism (cacna1b) have been significantly decreased, while genes associated with such pathways above (leap-2 , prdx4 , mb , homer1 , mif , sat1 , cytbc1 _ 8) and the pathway of protein processing in endoplasmic reticulum (cne1 , tram1) have been significantly increased. These findings suggested RES activated some immune and biological process-related genes to enhance GIFT's innate immunity. It also suggested high concentration addition or long-time administration may bring negative effect in tilapia liver. Abstract Based on the RMB exchange rate regime reform launched by People's Bank of China (PBoC) on Aug 11, 2015, we investigate the effects of this reform on the cross-correlations among CNH, NDF and CNY markets using multifractal detrended cross-correlation analysis (MF-DCCA) method. The empirical results show that cross-correlations among CNH, NDF and CNY markets are multifractal. Moreover, we find that this RMB exchange rate reform takes effects, as the persistence and the degree of multifractality of the cross-correlation between these series are weaker after the reform. Furthermore, we find that, after the reform, the persistent cross-correlation for CNH and CNY markets is stronger in the short term but weaker in the long term. While for NDF and CNY markets, the anti-persistent cross-correlation in the short term turn into persistent and persistent cross-correlation in the long term are stronger after the "8.11" RMB exchange rate regime reform. Finally, we find that the fat-tailed distribution is the main source of multifractality for NDF-CNY return series pair after the reform, while for CNH-CNY return series pair, the main source is still long-range correlation. The occurrence and human dietary exposure to 12 sulfonamides in fish samples along the lower reaches of the Yangtze River, China, were investigated in this study. The sulfonamides residue levels of 82 collected samples (21 caught and 61 aquacultured) were analyzed. Although 12 selected sulfonamides were detected in all fish samples along the lower reaches of the Yangtze River, the residual levels of caught and aquacultured samples were both below 5 μg kg −1 in the initial growth stage of fish. And sulfadiazine (SDZ) and sulfamethoxazole (SMX) were heavily used in the peak of fish growth. During this period, 17.4% of the samples contained concentrations of total sulfonamides above the maximum residue limit (100 μg kg −1 ); 13.0% were between 10 and 100 μg kg −1 , and the remaining (about 69.6%) were under 10 μg kg −1 . Through this study, we found residual levels of sulfonamides varied in different regions and in different fish species. Yixing and Changzhou were the main areas where the highest concentrations of sulfonamides were found. Huzhou and Suzhou areas were relatively free of pollution. The highest total sulfonamides concentrations were 1634.05 μg kg −1 and 472.97 μg kg− 1 in crab and bream, respectively. The sulfonamides concentrations in shrimp and perch were low. Based on the estimated daily intake (EDI) of Sulfonamides (SAs) in this study, SAs do not posed an immediate threat to human health; however, the potential risk to human safety could increase in the long run if the residual status of sulfonamides in aquatic products were left unattended. Much attention has been given to the safety and quality of aquatic products, including consuming Chinese mitten crab (Erio cheir sinensis), which offers both nutritional benefits and toxicological risks. Eighteen sulfonamides, 9 quinolones and 37 fatty acids were analyzed in 92 crab samples from primary aquaculture provinces in China. Enrofloxacin and ciprofloxacin have been mentioned as typical antimicrobials occurring at the greatest concentrations (>100 μg/kg, wm). By use of an in vitro method, the proportions of enrofloxacin, ciprofloxacin and essential fatty acids (EFAs, DHA and EPA) in ingested nutrients were determined to be 12 %, none and 95 %, respectively. The risk-benefit quotient (HQ) between the adverse effects of antimicrobials and nutritional benefits of EFAs in crabs found that HQs based on data after digestion were significantly less (HQ = 0.0086) than that of the control group where no digestion occurred (HQ = 0.055). This result suggested that (1) there was less risk posed by antimicrobials due to the consumption of crab, and (2) ignoring the bioaccessible fraction of antimicrobials in crabs might overestimate risks to the health of humans due to dietary exposure. Meaning bioaccessibility can improve the accuracy of the risk assessment process. Realistic risk evaluation should be recommended to achieve a quantified assessment of the dietary risks and benefits of aquatic products. In recent years, the food safety of aquatic products has become an increasing concern worldwide. Using conventional disinfectants and antibiotic drugs in the aquaculture industry may cause a decline in product quality. Compared to traditional water quality improvers, the green, emerging chemical potassium ferrate plays a significant role in improving aquaculture environments and the quality of aquatic products, yet the usage of potassium ferrate is blind, and the ecological risk is unclear due to the lack of knowledge of its scientific use. In this study, a probabilistic statistical model to assess the risk of potassium ferrate use is demonstrated for the first time. The results show that the annual output of potassium ferrate is about 900 t in China, with the usage dose ranging from 10 to 120 μg/L. The oxidation reduction potential (ORP) of potassium ferrate will return to a stable state within 72 h of the water quality treatment process, implying that there is no longer oxidative toxicity after 72 h. Acute toxicity tests have shown that potassium ferrate is of low toxicity to typical aquaculture organisms (cultured fish and crustaceans) with 72 h LC 50 values in the range of 95–609 mg/L, while it is highly toxic to the planktons tested with values of 1.0 and 4.3 mg/L. The joint probability curve analysis indicates that 30 μg/L (low daily usage dose), 60 μg/L (median daily usage dose), and 120 μg/L (high daily usage dose) of potassium ferrate can affect 1.06%, 2.15%, and 4.04% of aquatic species (aquaculture organisms and planktons), with a risk probability of 54.47%, 19.34%, 3.35%, while the risk to aquaculture organisms is 0. The research shows that the current concentrations of potassium ferrate used are of very low risk to aquaculture organisms, while also inhibiting planktons to improve water quality. This study plays an important role in guiding the safe use of potassium ferrate and improving water quality. Policymakers and scientists can draw support from these results and approaches when evaluating the use of potassium ferrate. • The total arsenic and its species within seventy-one Chinese mitten crab samples collected from China were investigated. • Organic and inorganic arsenic were all found (detectable rate were 100.0 %) in Chinese mitten crab. • Arsenobetaine is the most dominant form (52.0 % of total arsenic) of arsenic found in all Chinese mitten crab samples. • The low level of inorganic arsenic, estimating the hazards, measured in all samples indicate no health risks for human. Arsenic (As) is a major hazardous element in natural environments, and arsenic pollution is becoming an issue of concern worldwide. The more toxic inorganic arsenic, such as trivalent arsenite As(Ⅲ) and pentavalent arsenate As(Ⅴ), which normally can transform to organic arsenic compounds, such as arsenobetaine (AsB), arsenocholine (AsC), monomethyl arsenic acid (MMA), dimethyl arsenic acid (DMA), trimethyl arsenic acid (TMA), arsenosugars and arsenolipids. In this study, a total of 2130 individuals of Chinese mitten crabs were collected from seven locations and mixed to 71 samples. Total arsenic and six major species (AsC, AsB, DMA, MMA, As (Ⅲ), and As (Ⅴ)) were determined by inductively coupled plasma mass spectrometry (ICP-MS) and high-performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS), respectively. The target hazard quotient (THQ) was utilized to evaluate the human health risk. The total arsenic concentration and totals for the six arsenic forms are 0.25–1.66 mg/kg and 0.05–1.19 mg/kg wet mass, respectively. Except for HH and LH, the six arsenic species concentrations accounted for more than 50.0 % of the total arsenic. The less toxic AsB is the most predominant in crabs, comprising 50.0 %–90.0 % of the sum of six types of arsenic. The more toxic inorganic arsenic only range from 0.01 to 0.21 mg/kg wet mass, much less than the limit content of 0.50 mg/kg inorganic As in crustacea. The THQ values of inorganic arsenic through the consumption of Chinese mitten crabs are estimated, and the values are all less than 1, indicating that intaking Chinese mitten crabs collected from China will not cause an appreciable hazard risk to human health. Osteoarthritis (OA) is marked by cartilage deterioration, subchondral bone changes, and an inflammatory microenvironment. The study introduces the Microneedle-Delivered Polydopamine-Exosome (PDA@Exo MN), a therapeutic that not only preserves cartilage and promotes bone regeneration but also improves localized drug delivery through enhanced penetration capabilities. PDA@Exo MN shows strong reactive oxygen species (ROS) scavenging abilities and high biocompatibility, fostering osteogenesis and balancing anabolic and catabolic processes in cartilage. It directs macrophage polarization from M0 to the anti-inflammatory M2 phenotype. RNA sequencing of treated chondrocytes demonstrates restored cellular function and activated antioxidant responses, with modulated inflammatory pathways. The PI3K-AKT-mTOR pathway's activation, essential for PDA@Exo's effects, is confirmed via bioinformatics and Western blot. In vivo assessments robustly validate that PDA@Exo MN prevents cartilage degradation and OA progression, supported by histological assessments and micro-CT analysis, highlighting its disease-modifying impact. The excellent biocompatibility of PDA@Exo MN, verified through histological (H&E) and blood tests showing no organ damage, underscores its safety and efficacy for OA therapy, making it a novel and multifunctional nanomedical approach in orthopedics, characterized by organ-friendliness and biosecurity., (© 2024 The Author(s). Background: Diabetic atherosclerosis is one of the main causes of morbidity and mortality worldwide, but its therapeutic options are limited. Liraglutide (LIR), a synthetic analog of GLP-1 approved as an anti-obesity drug by the FDA, has been reported as a promising drug for diabetic atherosclerosis. However, the main problem with LIR is its use that requires regular parenteral injections, which necessitates the improvement of drug delivery for increased efficiency and minimization of injection numbers., Results: The objective of our present study was to prepare and characterize nanoparticles (BSA@LIR-PMF) for targeted drug delivery using LIR-encapsulated platelet membrane fragments (PMF) coated bovine serum albumin (BSA). We used various methods to characterize the prepared nanoparticles and evaluated their efficiency on diabetes-induced atherosclerosis in vitro and in vivo. The results showed that the nanoparticles were spherical and had good stability and uniform size with intact membrane protein structure. The loading and encapsulation rates (LR and ER) of BSA@LIR-PMF were respectively 7.96% and 85.56%, while the cumulative release rate was around 77.06% after 24 h. Besides, we also examined the impact of BSA@LIR-PMF on the proliferation, migration, phagocytosis, reactive oxygen species (ROS) levels, oxidative phosphorylation, glycolysis, lactate and ATP levels, and lipid deposition in the aortas. The results indicated that BSA@LIR-PMF could effectively inhibit ox-LDL-stimulated abnormal cell proliferation and migration, reduce the level of ROS and lactate concentration, and enhance the level of ATP, thereby improving oxidative phosphorylation in ox-LDL-treated cells., Conclusion: BSA@LIR-PMF significantly inhibited diabetes-induced atherosclerosis. Macrophages play a pivotal role in the healing of diabetic ulcers. The sustained elevation of glucose levels damages the insulin signaling pathway in macrophages, leading to dysfunctional macrophages that struggle to transition from pro-inflammatory (M1) to reparative (M2) states. Therefore, modulating macrophage inflammatory responses via the insulin pathway holds promise for diabetic ulcer treatment. Additionally, the presence of biofilm impedes drug penetration, and the resulting immunosuppressive microenvironment exacerbates the persistent infiltration of pro-inflammatory M1 macrophages. Therefore, we designed an array of dissolvable microneedle (denoted as NPF@MN) loaded with self-assembled nanoparticles that could deliver NPF nanoparticles, acid-sensitive NPF-releasing Protocatechualdehyde (PA) with hypoglycemic and insulin-like effects, regulating macrophage polarization to an anti-inflammatory M2 phenotype. Additionally, this study extensively examined the mechanism by which NPF@MN accelerates the healing of diabetic ulcers through the activation of the insulin signaling pathway. Through RNA-seq and GSEA analysis, we identified a reduction in the expression of pathway-related factors such as IR, IRS-1, IRS-2, and SHC. The widespread use of carbamate pesticides has raised significant environmental and health concerns, particularly regarding water contamination and the disruption of defense systems in organisms. Despite these concerns, research on the differential impacts of pesticides on male and female organisms remains limited. This study focused on methomyl, investigating sex-specific differences in liver antioxidant defenses and inflammatory response indices in male and female zebrafish after 56 days of exposure to environmentally relevant concentrations (0, 0.05, 0.10, and 0.20 mg/L). Our findings indicate that methomyl exposure significantly increased ROS content in zebrafish livers, inducing oxidative stress and activating enzymatic antioxidant defenses such as SOD, CAT, and GSH-Px activities. Sub-chronic exposure altered the expression of apoptosis-related genes ( Bax/Bcl2a and Caspases3a ), resulting in liver cell apoptosis in a concentration-dependent manner, with the 0.20 mg/L concentration causing the most severe damage. Additionally, methomyl exposure at environmentally relevant concentrations triggered persistent inflammatory responses in liver tissues, evidenced by increased transcription levels of inflammatory factor genes and the activation of toll-like receptors, heightening susceptibility to exogenous allergens. It is noteworthy that oxidative damage indicators (AST, ROS, MDA) and inflammatory gene expressions ( IL-1β , TNF-α ) were significantly higher in female livers compared to male livers at 0.10-0.20 mg/L methomyl exposure. Consequently, our study underscores the potential adverse effects of environmental methomyl exposure on aquatic organisms and highlights the need for heightened consideration of the risks posed by environmental endocrine disruptors to female health and safety.

Citizen scientist-based environmental monitoring and public education are becoming increasingly popular. However, current technologies for antibiotic-based novel contaminant identification are still restricted to laboratory sample collection and analysis due to detection methodologies and apparatus limitations. This study developed a time-resolved immunofluorescence-based simultaneous field-based assay for ciprofloxacin (CIP) and enrofloxacin (ENR) that matches test results to geographic locations. The assay helps the public understand the potential levels of antibiotic exposures in their environments and helps them take appropriate action to reduce risk. The assay was developed using smartphones and social software in addition to rapid testing. The method uses a portable, low-cost analytical kit with a smartphone app to build a field-based detection platform for the detection and analysis of ENR and CIP in water and aquatic products. The methodological evaluation was good, with detection limits of 0.4 ng/mL and 0.5 ng/g for ENR in water and fish, and quantification limits of 1.2 ng/mL and 1.4 ng/g, with recoveries of 89.0 %-101.0 % and 78.0 %-97.0 %. For CIP in water and fish, the limits of detection were 0.3 ng/mL and 0.4 ng/g, the limits of quantification were 0.9 ng/mL and 1.2 ng/g, and the recoveries were 75.0 %-91.0 % and 72.0 %-89.0 %, both with coefficients of variation <15 %. These limits were sufficient to prevent the two antibiotics from crossing over during simultaneous detection. The assay was validated using real samples to assess the effectiveness of the assay platform in field deployments, and the results were consistent with those obtained through liquid chromatography-tandem mass spectrometry (LC-MS) and enzyme-linked immunoassay (ELISA) techniques. In addition, the TRFIA assay process requires less time, uses more portable instruments, and is less complex than traditional methods. To investigate the activities of intestinal digestive enzymes, liver antioxidant enzymes, immunological enzymes, and glucometabolic enzymes in largemouth bass ( Micropterus salmoides ) under the biofloc model, an experiment was conducted in 300-liter glass tanks. The experiment comprised a control group, which was fed a basal diet, and a biofloc group, where glucose was added to maintain a C/N ratio of 15. Each group had three parallel setups, with a stocking density of 20 fish per tank. The experiment ran for 60 days, employing a zero-water exchange aquaculture model. The results showed that at the end of the culture period, there were no significant differences between the initial weight, final weight, WGR, SGR, and SR of the biofloc group and the control group of largemouth bass ( p > 0.05), whereas the lower FCR and the higher PER in the biofloc group were significant ( p < 0.05); intestinal α-amylase, trypsin, and lipase activities of largemouth bass in the biofloc group were significantly increased by 37.20%, 64.11%, and 51.69%, respectively, compared with the control group ( p < 0.05); liver superoxide dismutase and catalase activities, and total antioxidant capacity of largemouth bass in the biofloc group were significantly increased by 49.26%, 46.87%, and 98.94% ( p < 0.05), while the malondialdehyde content was significantly reduced by 19.91% ( p < 0.05); liver lysozyme, alkaline phosphatase, and acid phosphatase activities of largemouth bass in the biofloc group were significantly increased by 62.66%, 41.22%, and 29.66%, respectively ( p < 0.05); liver glucokinase, pyruvate kinase, glucose-6-phosphate kinase, pyruvate kinase, glucose-6-phosphatase, and glycogen synthase activities were significantly increased by 46.29%, 99.33%, 32.54%, and 26.89%, respectively ( p < 0.05). The study showed that the biofloc model of culturing largemouth bass can not only enhance digestive enzyme activities, antioxidant capacity, and immune response but can also promote the process of glucose metabolism and reduce feeding costs. This study provides data support for healthy culturing of largemouth bass in future production, provides a theoretical reference for optimizing the biofloc technology culture model, and is crucial for promoting the healthy and green development of aquaculture.

Advancements in monitoring and operation of aquaculture environments has minimized the concentrations of some residual pollutants in cultured aquatic products. However, currently most aquatic products are "farmed", and relationships among residual pollutants in tissues of crabs were still unclear. In this study, 64 typical pollutants, including 25 antibiotics, 15 metal, 23 organochlorine pesticides, and one dioxin-like compound inducing hydrocarbon-receptor (AHR) activity were measured in Chinese mitten crab ( Eriocheir Sinensis ) risks of consumption assessed and ranked. The superposition of properties including severity and relative potency of effects and parameters describing persistence and exposure along with rates of usage and identification of groups most likely to be exposed were assessed in combination to rank likelihood of dietary exposure and probabilities of adverse effects for each contaminant. The results indicated that the total scores per pollutants found that Cadmium (Cd), Heptachlor epoxide (HEPE), dioxin TEQ exhibited the greatest scores and explained the severity of dietary risk, while source analysis found that the three main pollutants resulted from the ambient environment and are not due to specific aquaculture processes. Bacterial infected wounds, which is characterized by easy infection, multiple inflammation and slow healing, is a complex symptom, resulting from metabolic disorder of the wound microenvironment. In this study, a series of self-healing double-network hydrogels based on KGRT peptide (Lys-Gly-Arg-Thr) with antibacterial, anti-inflammatory and optimizing cellular functions were designed to promote the healing of infected wounds with full-thickness skin defects. Moreover, the dextran hydrogelintroduces a large number of side chains, which are entangled with each other in the Schiff base network to form an interpenetrating structure. The hydrogel might regulate cell metabolism, differentiation and vascular endothelial growth factor (VEGF) function. Importantly, both in vitro and in vivo data showed that hydrogel not only has good antibacterial properties (99.8 %), but also can eradicate bacterial biofilm, effectively reduce inflammation (down-regulated IL-1β, TNF-α and ROS) and accelerate chronic wound healing process by speeding-up wound closure, increasing granulation tissue thickness, collagen deposition, angiogenesis (up-regulated CD31). The hydrogel could up-regulate mRNA expression of PI3K, AKT, ERK, eNOS, HIF-1α and VEGF, which were correlated with wound healing. Consistently, the hydrogel could promote infected wounds healing and inhibit inflammation through ERK/eNOS signaling pathway. Oxidative stress and reactive oxygen species drive ischemic stroke and its related complications. New antioxidant medications are therefore crucial for treating ischemic stroke. We developed Ti 2 C@BSA-ISO nanocomposites loaded with the hydrophobic drug isoquercetin (ISO) encapsulated in BSA on Ti 2 C nano-enzymes as a novel therapeutic nanomedicine for the treatment of ischemic stroke targeting reactive oxygen species (ROS). TEM visually proved the successful preparation of Ti 2 C@BSA-ISO, and the FTIR, XPS, zeta potential and DLS together demonstrated the acquisition of Ti 2 C@BSA-ISO. In addition, the enzyme-mimicking activity of Ti 2 C was evaluated and the antioxidant capacity of Ti 2 C@BSA-ISO was verified. Ti 2 C@BSA-ISO was able to reverse the decrease in cellular activity caused by ROS. Experiments in vivo showed that Ti 2 C@BSA-ISO could promote neuroprotection and scavenging of ROS in the hippocampal CA1 area and cerebral cortex of rats, thereby inhibiting cellular death and alleviating ischaemic stroke. Specifically, Ti 2 C@BSA-ISO alleviated ischemic stroke by inhibiting NLRP3/caspase-1/GSDMD pathway-mediated pyroptosis. Our study demonstrates the effectiveness of nanomedicines that can be directly used as drugs for the treatment of ischemic stroke in synergy with other drugs, which greatly expands the application of nanomaterials in the treatment of ischemic stroke.

Contamination by odor substances such as geosmin (GSM) and 2-methylisoborneol (2-MIB) was examined in the cultured water from aquaculture farming in the region of the Hongze Lake in 2022, and some factors influencing residual levels of them in the water were analyzed. Geographically, high concentrations of GSM were located mainly in the north and northeast culture areas of the lake, while those of 2-MIB were found in the northeast and southwest. Analysis of the water in the enclosure culture revealed significant differences in the concentrations of GSM and 2-MIB among the cultured species. The mean concentrations of GSM in culture water were ranked in the order: crab > the four major Chinese carps > silver and bighead carp, and silver and bighead carp > crab > the four major Chinese carps for 2-MIB. The concentration of GSM was significantly higher at 38.99 ± 18.93 ng/L in crab culture water compared to other fish culture water. Significant differences were observed in GSM concentrations between crab enclosure culture and pond culture, while 2-MIB levels were comparable. These findings suggest that cultural management practices significantly affect the generation of odor substances. The taste and odor (T&O) assessment revealed that the residual levels of GSM and 2-MIB in most samples were below the odor threshold concentrations (OTCs), although high levels of GSM and 2-MIB in all water bodies were at 30.9% and 27.5%, respectively. Inspired by the experience of relieving inflammation in infants with milk, antioxidant-engineered milk-derived extracellular vesicles (MEVs) are developed to evaluate their potential for accelerating wound healing. In this work, MEVs with polydopamines (PDA) are engineered using the co-extrusion method. Subsequently, the authors incorporated them into a Schiff-based crosslink hydrogel, forming a skin dosage form that could facilitate the wound healing process. The antioxidant properties of PDA assist in the anti-inflammatory function of engineered MEVs, while the gel provides better skin residency. The PDA@MEVs+GEL formulation exhibits excellent biocompatibility, pro-angiogenic capacity, and antioxidant ability in vitro. Furthermore, in vivo experiments demonstrate its efficacy in wound repair and inflammation inhibition. Mechanistically, PDA@MEVs+GEL simultaneously promotes the growth, migration, and anti-inflammation of 3T3 cells by activating PI3K-AKT pathway. Moreover, PDA@MEVs+GEL exhibits enhanced functionality in promoting wound healing in vivo, attributed to its ability to inhibit inflammation, stimulate angiogenesis, and promote collagen synthesis. In conclusion, this study delves into the mechanism of MEVs and underscores the improved efficacy of engineered extracellular vesicles. E-commerce platforms have become a significant sales channel for processed tilapia products such as frozen tilapia fillets, pickled tilapia, and canned tilapia in China. As food safety issues are worldwide concerned, the imbalance between the nutritional benefits of fish and the risk of contamination has become a major constraint on fish consumption. Therefore, it is necessary to assess the safety of tilapia products sold on e-commerce platforms. We conducted a quantitative benefit-risk assessment of arsenic and nutrient levels for tilapia products sold on Chinese e-commerce platforms using the hazard quotient (HQ). A total of 147 tilapia products were collected from the central Chinese e-commerce platforms Tmall, Taobao, and Jingdong for arsenic determination. Arsenic concentrations in tilapia products ranged from 0.004 to 0.820 mg·kg -1 . The inorganic arsenic content of tilapia products was lower than the national limit (0.1 mg·kg -1 ). One-way analysis of variance revealed that there was no significant difference in arsenic levels in tilapia products among different regions (p > 0.05), while there was a significant difference in product form, with canned tilapia containing significantly higher arsenic levels than frozen tilapia fillets and pickled tilapia fillets (p < 0.05). We conducted an aquaculture experiment to analyze the nutrient levels of tilapia. The mean value of EPA + DHA content of tilapia was 20.634 mg·100 g -1 . The HQ values of tilapia products ranged from 0.004 to 0.736. In a word, the nutritional benefits of consuming tilapia products exceed the risk of arsenic exposure. Microalgae has been widely used in aquaculture to improve both the water environment and fish growth; however, the current understanding of the effects of microalgae addition on the key players involved in regulating the water environment and fish health, such as microorganisms, remains limited. Here, a 50-day mesocosm experiment was set up to simulate the culture of Genetic Improvement of Farmed Tilapia (GIFT, Oreochromis niloticus ) with an average weight of 14.18 ± 0.93 g and an average length of 82.77 ± 2.80 mm. Different amounts of Chlorella pyrenoidosa were added into these artificial systems to investigate dynamics of bacterial communities in aquaculture water, fish gill, and gut using amplicon-based high-throughput sequencing technology. Our results showed that Chlorella pyrenoidosa addition increased diversity and network complexity of gill-associated bacterial communities rather than those of the water and gut. Furthermore, more biomarkers in the gill-associated bacterial communities were detected in response to Chlorella pyrenoidosa addition than the water and fish gut samples. These findings highlighted the high sensitivity of gill-associated bacterial communities in response to the Chlorella pyrenoidosa addition, implying Chlorella pyrenoidosa addition could play important roles in regulating the fish mucosal immunity by altering the gill-associated microbiota.

Objective: Through the comparison of different prediction models, we hope to find a promising statistical method to evaluate the prognosis of patients with acute ischemic stroke (AIS) after thrombolytic therapy., Methods: Data of 518 patients who received thrombolytic therapy were retrospectively collected in this study. Among them, 362 patients met the eligibility criteria, so their data such as age, sex, smoking history, previous medical history, clinical and laboratory indicators were analyzed. According to the 3 month follow-up results, 266 patients were included in a good prognosis group (modified Rankin Scale (mRS) score ≤2) and 96 in a poor prognosis group (3≤mRS≤6). All variables with P<0.05 in univariant and multivariant analyses were assigned in logistic regression model and artificial neural network (ANN) model to predict neurological prognosis, and the performance of the two models were compared., Results: Age, NIHSS scores, the serum concentration of immediate glucose, APTT and MBP at admission were found to be the predictive factors through ANN and logistic regression analysis. The binary logistic regression model revealed that the percentage correction, the precision, recall and F1 score of the regression model were 79%, 69.23%, 37.50% and 48.65%, respectively. While those of ANN were 79.98%, 69.70%, 37.25%, and 49.66%, correspondingly., Conclusions: ANN model is as effective as a logistic regression model in predicting the prognosis of AIS after thrombolytic therapy with rt-PA. Polyculture operations in freshwater aquaculture ponds can disrupt microbial communities. High-throughput sequencing was used to assess the impact of polyculture operations on bacterial and three sub-microeukaryote communities (fungi, zooplankton, and eukaryotic phytoplankton) in Penaeus vannamei aquaculture ponds containing oriental river prawns and giant freshwater prawns, respectively. The results showed that the bacterial community was less sensitive than the microeukaryote communities to both the polyculture activity and environmental variations. The polyculture of giant freshwater prawns rather than oriental river prawns was the primary factor affecting the beta diversity of the three sub-microeukaryote communities. This may be due to the larger biomass of the polyculture varieties of giant freshwater prawns compared with oriental river prawns. The polyculture activity of giant freshwater prawns with a higher density and that of oriental river prawns with a lower density increased the stochasticity of the community assembly of the three sub-microeukaryote communities. It also affected the topological properties of the microbial communities, including greater correlations between ecosystem elements, and reducing the correlations among zooplanktons. The eukaryotic phytoplankton was the only microbial community that could also be explained by nutrient variation (mainly the total nitrogen). Background: Spinal astrocytes contribute to chronic itch via sensitization of itch-specific neurons expressing gastrin-releasing peptide receptor (GRPR). However, whether microglia-neuron interactions contribute to itch remains unclear. In this study, we aimed to explore how microglia interact with GRPR + neurons and promote chronic itch., Methods: RNA sequencing, quantitative real-time PCR, western blot, immunohistochemistry, RNAscope ISH, pharmacologic and genetic approaches were performed to examine the roles of spinal NLRP3 (The NOD-like receptor family, pyrin-containing domain 3) inflammasome activation and IL-1β-IL1R1 signaling in chronic itch. Grpr-eGFP and Grpr KO mice were used to investigate microglia-GRPR + neuron interactions., Results: We observed NLRP3 inflammasome activation and IL-1β production in spinal microglia under chronic itch conditions. Blockade of microglial activation and the NLRP3/caspase-1/IL-1β axis attenuated chronic itch and neuronal activation. Type 1 IL-1 receptor (IL-1R1) was expressed in GRPR + neurons, which are essential for the development of chronic itch. Our studies also find that IL-1β + microglia are localized in close proximity to GRPR + neurons. Consistently, intrathecal injection of IL1R1 antagonist or exogenous IL-1β indicate that the IL-1β-IL-1R1 signaling pathway enhanced the activation of GRPR + neurons. Furthermore, our results demonstrate that the microglial NLRP3/caspase-1/IL-1β axis contributes to several different chronic itches triggered by small molecules and protein allergens from the environment and drugs., Conclusion: Our findings reveal a previously unknown mechanism in which microglia enhances the activation of GRPR + neurons through the NLRP3/caspase-1/IL-1β/IL1R1 axis. Objective: To investigate the mechanism of Neferine in treating endometriosis fibrosis by TGF-β/ERK signaling pathway through a combination of network pharmacological analysis of Lotus embryos, in vivo animal experiments, and in vitro cell experiments., Methods: The active ingredients of the drug lotus embryos, the drug targets and the targets of endometriosis were determined from the TCMSP database, the Swiss Target Prediction database and GeneCard and Online Mendelian Inheritance in Man. The String database and Cytoscape 3.6.3 software were used to construct the network of common target protein interactions between drug and disease, as well as the target network. GO and KEGG enrichment analysis of the common targets was performed. We designed endometriosis mouse models with Neferine to investigate the therapeutic effect of Neferine on the fibrosis model of endometriosis and its mechanism of action. Different methods were used to evaluate the treated endometriotic lesion tissue and the untreated ectopic lesion tissue. The 12Z cells (human endometriosis immortalized cells) were cultured in vitro and treated with Neferine to detect cell viability and the effects of invasion and metastasis., Results: The results of GO function and KEGG enrichment analysis showed that the role pathways of lotus germ were TGF-β signaling pathway, ERK1/2 signaling pathway, IL-17 signaling pathway, TNF signaling pathway, AGE-RAGE signaling pathway, and PI3K-Akt signaling pathway. Neferine which is one of the effective active ingredients of lotus germ, significantly inhibited the expression of fibronectin, collagen I, connective tissue growth factor, and smooth muscle actin by activating the TGF-β/ERK pathway in vivo , which is required for the fibrosis process of endometriosis. Neferine also significantly inhibited the proliferation, invasion and metastasis ability of 12Z cells., Conclusion: Neferine inhibits the progression of endometriosis both in vitro and in vivo . Purpose: To observe the crystalline lens rise (CLR) in horizontal and vertical orientations using anterior segment optical coherence tomography (AS-OCT)., Methods: Non-invasive swept-source AS-OCT was used to measure the lens thickness, CLR, and angle-to-angle distance (ATA) in both the horizontal and vertical orientations. Anterior chamber depth (ACD) and horizontal white-to-white corneal diameter were obtained using the Pentacam HR (Oculus Optikgeräte GmbH). Axial length was obtained using the IOLMaster 700 (Carl Zeiss Meditec AG). The paired t test was used to analyze the difference in CLR between the two orientations. Pearson correlation analysis was performed to investigate the correlations between CLR and other ocular variables., Results: This prospective observational study comprised 99 eyes (99 patients) that underwent Visian Implantable Collamer Lens (STAAR Surgical) implantation for myopic correction. The mean CLR was 64.29 ± 168.04 and 208.09 ± 173.12 µm in the horizontal and vertical orientations, respectively. The vertical CLR (VCLR) was significantly greater than the horizontal CLR (HCLR) ( P < .05). Both the HCLR and VCLR were positively correlated with lens thickness and negatively correlated with ACD (all P < .05). The difference in CLR (VCLR-HCLR) was positively correlated with the axial length and the difference in ATA between the two orientations ( P < .05)., Conclusions: VCLR was greater than HCLR in most patients with myopia, especially in the longer eyes. This nonuniform distribution in CLR implied different placements of the iridocorneal angles in the horizontal and vertical orientations and should be considered for the selection of ICL size and placement position. Exosomes are extracellular vesicles which carry specific molecular information from donor cells and act as an intercellular communication vehicle, which have emerged as a novel cell-free strategy for the treatment of many diseases including inflammatory disease. Recently, rising studies have developed exosome-based strategies for novel inflammation therapy due to their biocompatibility and bioactivity. Researchers not only use native exosomes as therapeutic agents for inflammation, but also strive to make up for the natural defects of exosomes through engineering methods to improve and update the property of exosomes for enhanced therapeutic effects. The engineered exosomes can improve cargo-loading efficiency, targeting ability, stability, etc., to achieve combined and diverse treatment strategies in inflammation diseases. Herein, a comprehensive overview of the recent advances in application studies of native and engineered exosomes as well as the engineered methods is provided. Meanwhile, potential application prospects, possible challenges, and the development of clinical researches of exosome treatment strategy are concluded from plentiful examples, which may be able to provide guidance and suggestions for the future research and application of exosomes., (© Tsinghua University Press 2022.)

The aim of this study was to evaluate the net effects of dietary consumption on the potential neurodevelopmental effects of largemouth bass (Micropterus Salmoides, simplify as bass) cultured in China, especially when the bait of largemouth bass is changed from iced trash fish to formulated feed. A total of three bait group bass samples were collected from main producing areas during harvest period, including formulated feed group, iced trash fish group, and both group (converting from iced trash fish to formulated feed). Net neurodevelopment value (expressed as intelligence quotient or IQ) was calculated using FAO/WHO deterministic methods and probabilistic assessment model. The results showed net IQ of largemouth bass fed by the three baits groups all showed positive or beneficial neurodevelopmental effects, with the values of 1.55 ± 0.97 (formulated feed group), 3.17 ± 1.76 (iced trash fish group), and 2.65 ± 1.29 (both group) respectively. Bass samples fed with ice trash fish showed the highest neurodevelopmental effect. In this work, β-cyclodextrin porous polymers (P-CDPs) functionalized novel covalent organic frameworks (P-CDPs/COFs) were synthesized by a simple and facile method. After combined with Pd 2+ via electrostatic interaction, the Pd 2+ @P-CDPs/COFs nanocomposites were prepared and utilized as novel electrode materials to fabricate the non-enzyme electrochemical sensors for high-sensitivity detection of Norfloxacin (NF). Due to the host-guest recognition, excellent adsorption performance and catalytic performance of Pd 2+ @P-CDPs/COFs, the prepared sensor exhibited excellent electrochemical performance for detecting NF under the optimum conditions, which showed two linear ranges of 0.08-7.0 μM and 7.0-100.0 μM with a low detection limit of 0.031 μM (S/N = 3). Additionally, the obtained sensor has also been successfully applied to measure NF with satisfactory results in the real medicinal samples of NF eye-drops. Objective: To explore the mechanism of Thunberg Fritillaria in treating endometriosis (EMs) based on network pharmacology and the effect of Peiminine on the MEK/ERK pathway., Methods: We applied Chinese medicine system pharmacology analysis platform (TCMSP) database and literature search to screen the main chemical components of Fritillaria thunbergii Miq and created a Vanny map from the databases of TCMSP, GENECARDS, Online Mendelian Inheritance in Man (OMIM), and some others. The STRING database was used to construct the protein interaction network of Fritillaria thunbergii Miq and EMs. The overlapping targets and enriched pathways were discovered using the cells of the innate immune annotation database (DAVID) and the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses. To test the mechanism of Peiminine, the active ingredients of Fritillaria thunbergii, in the therapy of EMs, we designed cell assays and animal research. EMs mouse models were treated with several therapies, including fibrosis inhibitor in Peiminine by utilizing Hematoxylin-eosin staining (HE staining), MASSON staining, Immunohistochemistry, Immunofluorescence, quantitative real-time PCR (qRT-PCR) experiment, and Western blotting test. We evaluated the anti-endometriotic effects of Peiminine using 12Z human endometriotic cells. Cell Counting Kit 8 was used to assess the vitality of 12z cells (CCK8). We evaluated the migration ability of 12z cells by cell scratch test., Results: The effective active ingredients of Fritillaria thunbergii Miq in the treatment of EMs are Pelargonidin, Beta-sitosterol syringaresinol, Peimisine Pelargonidin-3, 5-diglucoside Ziebeimine Zhebeiresinol Verticine Solatubin OSI-2040 Chaksine Peiminine Peiminoside Peiminoside&#95;qt, and 6-Methoxyl-2-acetyl-3-methyl-1, 4-naphthoquinone-8-O-beta-D-glucopyranoside. The critical targets for Fritillaria thunbergii Miq treating EMs are NOS2/PTGS1/AR/PPARG/PTGS2/NCOA2/RXRA/PGR/NR3C1/NCOA1/SLC6A4/OPRM1/BCL2 and ESR1. The results of GO function and KEGG enrichment analysis showed that the role pathway was estrogen-related signaling and thyroid hormone-related signaling. The expression of E-cadherin was decreased in EMs while MEK1/2, P-ERK, N-cadherin and vimentin were all increased in MASSON, immunofluorescence, Real-time PCR and Western blotting. To comprehensively investigate the effects of exposure to legal doses of sulfamethoxazole (SMZ) in Nile tilapia (Oreochromis niloticus), fishes were exposed to diets supplemented with different doses of SMZ (NS, normal feed; LS, 20 mg/kg·day; MS, 200 mg/kg·day; and HS, 1000 mg/kg·day) for 4 weeks and then fed with normal feed for 4 weeks. General SMZ accumulation, growth performance, intestinal short-chain fatty acids, intestinal flora diversity, composition, and function were systemically evaluated. Results indicated that the SMZ accumulation in O. niloticus muscles, intestinal contents, and aquaculture environment positively correlated to the exposure dose. The growth performance, measured by weight increase, was MS>LS>NS, while HS antibiotics retarded the growth. SMZ-exposed O. niloticus had an increased number of fat particles in the liver and a change in the content of intestinal SCFAs. Moreover, SMZ exposure changed the biological diversity of the intestinal flora and subsequently induced microbiota dysbiosis, primarily inhibiting the growth of Fusobacteria, especially in HS group. Overall, exposure to higher SMZ doses than the recommended ones impair general intestinal functions and provokes health risk in fish. The occurrence of 27 antibiotics (18 sulfonamides and 9 quinolones) in 92 samples of cultured Chinese mitten crab (Eriocheir sinensis) from three provinces (Anhui, Jiangsu and Liaoning) was investigated. These 21 antibiotics were detected at least once in crabs from these provinces with detection frequencies of 3.70-90.91%. Sulfonamides were detected in 53.7% of the samples at concentrations of 0.1-10 μg/kg in Jiangsu, while quinolones were detected with 90.9% of samples containing concentrations of 1-100 μg/kg in Liaoning province. Enroflxacin, ciprofloxacin, sulfaquinoxaline, sulfameter, sulfadoxine, and sulfamethoxazole were the mainly used antibiotics and enroflxacin were present at a high concentration (>100 μg/kg). Dietary assessments showed that residual antibiotics in crabs from China were far below the maximum residue limit (MRL) of total sulfonamides and quinolones, and there was almost no risk associated with crab consumption. To investigate the effect of chiral pesticide fenvalerate (FV) on the micro-ecological environment of aquaculture pond sediment, we used an indoor static experiment to observe the effects of FV added at different concentrations with different chiral isomers on the changes in the sediment bacterial community. The 16S rDNA high-throughput sequencing technique was used to conduct sequencing and analysis of the bacterial community structure as well as changes in aquaculture pond sediments after 4 weeks of cultivation. The results showed that the microbial alpha diversity indices (Sobs and Shannon indices) of the treated groups were significantly lower than those of the control group after 4 weeks (P < 0.05), and the values in the high-concentration group were significantly lower than those of the low-concentration group (P < 0.05). In terms of bacterial group composition, the proportion of abundance of Proteobacteria and Acidobacteria in the treated groups were greater than in the control group after 4 weeks, while the proportion of abundance of Bacteroidetes and Verrucomicrobia were lower. In the high-concentration FV treatment group, the proportion of abundance of Bacteroidetes, Acidobacteria, Chloroflexi, Nitrospinae, unclassified&#95;k&#95;norank, Ignavibacteriae, and Nitrospirae were significantly different from those of the other groups (P < 0.05). Principal coordinate analysis (PCoA) and ANONISIM/Adonis analysis showed that the cis-enantiomer had a stronger effect on the bacterial community as the concentration of FV increased. In addition, the linear discriminant analysis effect size (LEfSe) and linear discriminant analysis (LDA) results revealed differences in the level of enrichment of bacterial groups caused by FV at different concentrations and isomer levels. Collectively, this study showed that FV residue has a pronounced effect on bacterial communities in sediment, which becomes more significant with increasing exposure concentration. The effects of the cis- and trans-enantiomers of FV on the sediment environment are different; the cis-enantiomer has a stronger effect on the bacterial community.

Tilapia were exposed to 0, 0.2, 2, 20, 200 µg/L methomyl for 30 days, and then transferred to methomyl-free water for 18 days. Caspase-8 in serum, apoptosis rate, microstructure and ultra-microstructure of testis were checked after methomyl exposure and at 18 days after transferring to methomyl-free water. There were no significant changes in Caspase-8 activity, apoptosis rate, and tissue structure in testis exposed to 0.2 and 2 µg/L compared with control. However, when tilapia exposed to 20 and 200 µg/L, the Caspase-8 activity and apoptosis rate were induced significantly, and tissue damage happened compared with the control. Thus it would appear 2 µg/L methomyl might be considered as the no observed adverse effect level. Recovery data showed that the effects produced by lower concentration of 20 µg/L were reversible but not at the higher 200 µg/L concentration.

Fenvalerate (FV) is widely used in aquaculture because of their broad spectrums and high efficiency. However, little is known regarding the elimination of FV influenced by environment factors in aquaculture water, especially its cis-trans isomers (cis-FV and trans-FV). In the present study, factors influencing the aquaculture environment (open type, temperature, pH and light) were selected, and the elimination dynamics of FV and its cis-trans isomers in aquaculture water using orthogonal experiments were investigated. The results showed that the half-life and elimination rate range of FV were 4.75-11.95 days and 65-93%, respectively, while those of trans-FV were 4.60-11.82 days and 67-93% and those of cis-FV were 4.94-12.04 days and 64-92%, respectively. The elimination rate of trans-FV was better than that of cis-FV. Additionally, analysis of variance (ANOVA) of the orthogonal experimental data indicated that the environmental factors of open type, temperature, and pH significantly influenced the elimination rate of cis- and trans-FV (P < 0.05), that is, in the aquaculture season, high temperature and pH facilitate to eliminate FV. This study would improve our understanding of natural degradation associated with FV and guide safe to use associated with pesticide in aquaculture.

Objectives: In 2014, the International Association for the Study of Lung Cancer (IASLC)/International Thymic Malignancies Interest Group (ITMIG) launched a worldwide Tumor Node Metastasis (TNM) staging proposal for the next edition of thymic tumours. The objective of the current study was to evaluate the proposed new staging system specific to the thymic well-differentiated neuroendocrine carcinoma (TWDNC)., Methods: From November 2003 to July 2014, 61 consecutive patients were enrolled in this study with pathologically confirmed TWDNC in Shanghai Chest Hospital. Clinical and pathological data were retrospectively reviewed. Survival analysis was performed using the Kaplan-Meier and log-rank tests. Validity evaluation was addressed by Cox proportional hazards regression model, after adjusting for potential confounders and visually assessing the distinction of curves generated based on the staging system of Masaoka-Koga and the proposed TNM ones., Results: Thymic carcinoids made up 4% of total thymic tumours in our institution. The 5-year overall survival (OS) rate and the disease-free survival (DFS) rate were 72 and 41%, respectively. Neither Masaoka-Koga staging system nor the proposed TNM system showed ordered appropriateness visually in survival curves and the prognostic demarcation between stages was poor on both OS and DFS., Conclusions: The IASLC/ITMIG suggested that the TNM and Masaoka-Koga staging systems fail to predict the clinical course of TWDNC patients. Objectives: Chest computed tomography (CT) scanning has been widely utilized in thymoma identification and staging as well as in follow-up monitoring for recurrence. However, the relationship between some CT imaging features and pathological types, clinical stage, completeness of resection, or prognosis in thymoma has not been well explored., Methods: We retrospectively reviewed preoperative CT imaging for 238 thymoma patients, who had undergone thymectomy from October 2007 to December 2011. All CT parameters were assessed in each case based on clinical and pathological data. Survival analysis was performed by using the Kaplan-Meier and log-rank tests., Results: Tumour contours (P = 0.008), homogeneity (P = 0.009), degree of enhancement (P = 0.013), fat plane obliteration with adjacent structures (P < 0.001), the presence of mediastinal lymphadenopathy (P = 0.010), irregular infiltration into the lung (P = 0.012) and tumour shape (P = 0.007) were associated with the World Health Organization (WHO) histological classification. Lobulated or irregular tumour contours (P < 0.001), presence of calcifications (P = 0.002), infiltration of surrounding fat (P < 0.001), irregular infiltration into the lung (P < 0.001), irregular infiltration into vascular (P < 0.001), more abutment of vessels (P < 0.001) and pulmonary changes adjacent to the tumour (P < 0.001) were associated with the more advanced Masaoka-Koga clinical stage. Tumour contours (P < 0.001), infiltration of surrounding fat (P = 0.008), irregular infiltration into the lung (P < 0.001) and degree of abutment of vessel circumference (P = 0.001) were associated with completeness of resection. With multivariate analysis, no CT image features could reliably predict on the overall or disease-free survival rate., Conclusions: CT imaging does have some features, which are significantly correlated with the WHO classification, the Masaoka-Koga clinical staging and the completeness of resection, although it has no definite role to evaluate preoperatively the survival rate of thymoma patients., (© The Author 2014. Lung cancer is a leading cause of cancer-related death worldwide, and non-small cell lung cancer (NSCLC) constitutes ~85% of lung cancers. However, the mechanisms underlying the progression of NSCLC remain unclear. In this study, we found the mRNA and protein expression levels of integrin αv are both increased in NSCLC tissues compared to healthy ones, which indicates that integrin αv may play an important role in NSCLC progression. To further investigate the roles of integrin αv in NSCLC, we overexpressed the integrin αv gene in the NSCLC cell line A549, and found that the cell proliferative ability increased. The apoptosis of A549 cells was inhibited with overexpression of integrin αv. To elucidate the molecular mechanism underlying the role of integrin αv in promoting NSCLC progression, we studied the expression of proteins from a number of important pathways associated with tumorigenesis, and found that the extracellular signal regulated protein kinase (ERK)1/2 signaling pathway may be involved in the mediation of the observed integrin αv effects. component of an important pathway for tumorigenesis, the ERK 1/2. Following inhibition of ERK 1/2 signaling, the proliferation of A549 cells induced by integrin αv was reduced, while the inhibition of apoptosis was attenuated. Our findings demonstrate that integrin αv promotes the proliferation of the human lung cancer cell line A549 by activating the ERK 1/2 signaling pathway, which suggests that this pathway may be a promising target for the treatment of human lung cancer.
