Abstract

Background: The significant changes in a series of biochemical activities during the process of tumorigenesis and development, including those in glucose, lipid, and amino acid metabolism can contribute to the ability of the cancer cells to proliferate indefinitely. WZ35 is a new small molecule YAP inhibitor, which can mediate YAP activity to inhibit the growth of gastric cancer, breast cancer and hepatocellular carcinoma. In this article, we explored how WZ35 can modulate YAP activity to influence the metabolism of hepatocellular carcinoma (HCC) cells to inhibit their proliferation activity. Methods: The Gene Expression Omnibus (GEO) data, the Cancer Genome Atlas (TCGA) data, immunohistochemistry (IHC) and preclinical mouse model was utilized to detect the differential expression and vital role of YAP in HCC cells. A series of in vitro and in vivo experiments were performed to explore the antitumor activity of WZ35 and how it can target YAP activity to inhibit the tumor progression. UCSC combined JASPAR public databases were used to predicted possible binding sites of TEAD on GLUT1 promoter. Additionally, seahorse energy metabolism experiments and metabolomics analysis were utilized to explore the possible metabolic changes induced by WZ35. The clinical use of YAP and GLUT1 was verified by bioformatics and tissue microarrays of immunocytochemistry. Results: WZ35 can significantly attenuate the proliferation and growth of HCC. In terms of mechanism(s), it was demonstrated that the suppressive effects of WZ35 on YAP were achieved by promoting ROS production and the drug can exert its significant YAP inhibitory capacity to decrease the level of GLUT1, a glucose transporter located on the surface of cytomembrane, thereby resulting in a significant decrease in the ability of cells to take up glucose and thus perturb the metabolism. Moreover, through bioinformatics mining, it was proposed that YAP/GLUT1 has the potential to serve as promising target for HCC therapy. Conclusions: Our findings indicate that potential inhibitory effects of WZ35 were achieved by affecting the ROS-YAP-GLUT1 signal axis to induce the metabolic reprogramming of hepatocellular carcinoma cells. YAP/GLUT1 could serve as an important molecular target for both the diagnosis and treatment of HCC.

Abstract

The advent of graphene opens up the research into two-dimensional (2D) materials, which are considered revolutionary materials. Due to its unique geometric structure, graphene exhibits a series of exotic physical and chemical properties. In addition, single-element-based 2D materials (Xenes) have garnered tremendous interest. At present, 16 kinds of Xenes (silicene, borophene, germanene, phosphorene, tellurene, etc.) have been explored, mainly distributed in the third, fourth, fifth, and sixth main groups. The current methods to prepare monolayers or few-layer 2D materials include epitaxy growth, mechanical exfoliation, and liquid phase exfoliation. Although two Xenes (aluminene and indiene) have not been synthesized due to the limitations of synthetic methods and the stability of Xenes, other Xenes have been successfully created via elaborate artificial design and synthesis. Focusing on elemental 2D materials, this review mainly summarizes the recently reported work about tuning the electronic, optical, mechanical, and chemical properties of Xenes via surface modifications, achieved using controllable approaches (doping, adsorption, strain, intercalation, phase transition, etc.) to broaden their applications in various fields, including spintronics, electronics, optoelectronics, superconducting, photovoltaics, sensors, catalysis, and biomedicines. These advances in the surface modification of Xenes have laid a theoretical and experimental foundation for the development of 2D materials and their practical applications in diverse fields. We herein introduce the principle of proximity assay into tetramolecular G-quadruplexes guided by various biomolecules for the construction of a sensing strategy. Our strategy is based on the co-localization of intermolecular G-rich strands guided by a recognition event of a specific biomolecule to its corresponding affinity ligand. In such case, the local concentration among intermolecular strands is significantly increased to trigger the following self-assembly that served as the peroxidase-mimicking activity. Abstract

The catalytic activity of photozymes can be regulated through light irradiation time and intensity, but they still suffer from low activity in physiologically neutral pH (typically, pH < 5). Herein, through simple post-synthetic incorporation of Pt, the neutral pH activity of the g-C3N4 nanosheet (CNNS) was activated. Abstract

Background: Coronavirus disease 2019 (COVID-19) is a new viral respiratory disease and whether pregnant women are at increased risk of infection is unknown. Viral pneumonia is an important indirect cause of maternal death. Little is known about the effects of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) during pregnancy. Objective: To describe the clinical characteristics of COVID-19 in pregnancy and their newborn infant, and we sought to explored whether the SARS-CoV-2 can be intrauterine vertically transmitted. Study Design: The study was a case series study conducted in the obstetric ward of Tongji Hospital affiliated to Huazhong University of science and technology, Wuhan, China. Demographic, clinical, laboratory and radiological profiles of the SARS-CoV-2 infection case series. A systematic testing procedure for SARS-CoV-2 infection using oropharyngeal swab, placenta tissue, vaginal mucus, and breast milk of mothers. and oropharyngeal swab, umbilical cord blood, and serum of newborns was conducted. Results: We have conducted the most thorough virological assessment to date, and we include a longer clinical observation in mother-infant dyads during hospitalization. The clinical course and outcomes of three pregnant women who acquired SARS-CoV-2 infection late pregnancy are described in mother-infant dyads. Two had caesarean delivery in their third trimester. All patients showed an uneventful perinatal course, and a successful outcome. No infants became infected by vertical transmission or during delivery. Conclusion: No evidence to suggest the potential risk of intrauterine vertical transmission in the case series and further in-depth study is needed. Both the pregnancy woman and infant showed fewer adverse maternal and neonatal outcomes.

Abstract

Electrospun polymer/metal composite nanofibers have received much attention in the field of bone tissue engineering and regenerative medicine (BTERM) owing to their extracellular matrix- (ECM-) like structure, sufficient mechanical strength, favorable biological properties, and bone induction. In particular, electrospun nanofibers containing strontium (Sr) can significantly promote bone repair and regeneration by mediating osteolysis and osteogenesis, which offers a promising bioactive material for BTERM. In this review, we summarized the effects of electrospun nanofibers containing Sr on stem cells, osteoblasts, and osteoclasts in BTERM. Also, current challenges and future perspectives for electrospun nanofibers containing Sr in BTERM are briefly outlined. It is hoped that the systematic overview will inspire the readers to further study Sr-containing nanofibers for BTERM and accelerate their translation from the bench to the clinic.

185. Blood biomarkers for post-stroke cognitive impairment: A systematic review and meta-analysis.

Abstract

Post-stroke cognitive impairment (PSCI) is a frequent consequence of stroke, which affects the quality of life and prognosis of stroke survivors. Numerous studies have indicated that blood biomarkers may be the key determinants for predicting and diagnosing cognitive impairment, but the results remain varied. Therefore, this meta-analysis aims to summarize potential biomarkers associated with PSCI. PubMed, Web of Science, Embase, and Cochrane Library were comprehensively searched for studies exploring blood biomarkers associated with PSCI from inception to 15 April 2022. 63 studies were selected from 4,047 references, which involves 95 blood biomarkers associated with the PSCI. We meta-analyzed 20 potential blood biomarker candidates, the results shown that the homocysteine (Hcy) (SMD = 0.35; 95 %CI: 0.20–0.49; P < 0.00001), c-reactive protein (CRP) (SMD = 0.49; 95 %CI: 0.20–0.78; P = 0.0008), uric acid (UA) (SMD = 0.41; 95 %CI: 0.06–0.76; P = 0.02), interleukin 6 (IL-6) (SMD = 0.92; 95 % CI: 0.27–1.57; P = 0.005), cystatin C (Cys-C) (SMD = 0.58; 95 %CI: 0.28–0.87; P = 0.0001), creatinine (SMD = 0.39; 95 %CI: 0.23–0.55; P < 0.00001) and tumor necrosis factor alpha (TNF-α) (SMD = 0.45; 95 %CI: 0.08–0.82; P = 0.02) levels were significantly higher in patients with PSCI than in the non-PSCI group. Based on our findings, we recommend that paramedics focus on the blood biomarkers levels of Hcy, CRP, UA, IL-6, Cys-C, creatinine and TNF-α in conjunction with neuroimaging and neuropsychological assessment to assess the risk of PSCI, which may help with early detection and timely preventive measures.
