Abstract

The detection of tumours in an early phase of tumour development in combination with the knowledge of expression of tumour markers such as epidermal growth factor receptor (EGFR) is an important prerequisite for clinical decisions. In this study we applied the anti-EGFR nanobody (99m)Tc-D10 for visualizing small tumour lesions with volumes below 100 mm(3) by targeting EGFR in orthotopic human mammary MDA-MB-468 and MDA-MB-231 and subcutaneous human epidermoid A431 carcinoma mouse models. Use of nanobody (99m)Tc-D10 of a size as small as 15.5 kDa enables detection of tumours by single photon emission computed tomography (SPECT) imaging already 45 min post intravenous administration with high tumour uptake (>3% ID/g) in small MDA-MB-468 and A431 tumours, with tumour volumes of 52.5 mm(3) ± 21.2 and 26.6 mm(3) ± 16.7, respectively. Fast blood clearance with a serum half-life of 4.9 min resulted in high in vivo contrast and ex vivo tumour to blood and tissue ratios. In contrast, no accumulation of (99m)Tc-D10 in MDA-MB-231 tumours characterized by a very low expression of EGFR was observed. Here we present specific and high contrast in vivo visualization of small human tumours overexpressing EGFR by preclinical multi-pinhole SPECT shortly after administration of anti-EGFR nanobody (99m)Tc-D10. The article titled "The Moral Customer - A Paradox?" deals with the phenomenon of paradox in consumer behavior. Examples are given of customers who want to behave morally and sustainably, but still have unresolved problems and therefore choose pragmatic solutions that do not always align with their own moral values. The article emphasizes the importance of creativity and willingness to implement in order to offer sustainable products that are affordable for everyone. It is also pointed out that companies should go beyond "common sense" to combine sustainability with other values such as ethics. Abstract

The gene transfer routes for integration of single copy transgenes into tissues by infection or electroporation are evaluated. Fluorescence in situ hybridization technique indicates that a consistent, expression level-independent association potential is imparted to nuclear matrix by the retroviral infectants, whereas electroporated proviral inserts mostly localize to the loop fraction.

179. Probiotic yogurt in the elderly with intestinal bacterial overgrowth: endotoxaemia and innate immune functions

Abstract

A study was conducted in healthy elderly living independently in senior housing to assess the impact of a probiotic yoghurt supplement on small intestinal bacterial overgrowth. Twenty-three participants with positive and thirteen participants with negative hydrogen breath test were studied before and after a period of 4 weeks of probiotic yoghurt administration. Intestinal permeability, plasma endotoxin levels, phagocytic activity of leucocytes, cytokine production by monocytes and free radical response of neutrophils were determined. Intestinal permeability was similar for the two groups and was unaffected by probiotic treatment. Both plasma endotoxin levels and the basal phagocytic activity of leucocytes decreased after yoghurt intake in the two groups. Exposure of monocytes and neutrophils ex vivo led to an increased cytokine response and free radical response, respectively. The normalisation of the various cytokine responses was more apparent in the group with positive breath test. In addition, the plasma levels of lipoplysaccharide binding protein and soluble CD14, lipoplysaccharide pattern recognition receptors and surrogate markers of lipoplysaccharide permeability were diminished by the end of the study. In conclusion, probiotic administration in the elderly normalises the response to endotoxin, and modulates activation markers in blood phagocytes, and therefore may help reduce low-grade chronic inflammation

Abstract

Throughout world history, copper has been a significant metal for a vast number of cultures, from the oldest civilizations on record to the Bronze Age and Greek and Roman antiquity. Though replaced by iron as the primary metal for tools and weapons in ancient civilizations, copper found new resurgence in the nineteenth century when it was discovered to have particularly high thermal and electrical conductivity. Copper mining quickly escalated into a large-scale industry, and because of its vast reserves and innovative mining techniques, the United States seized the reins of global production with the opening of significant copper mines in Tennessee and Michigan in the 1840s and Montana in the 1870s. Copper-mining prosperity and America's dominance of the industry came with a heavy environmental price, however. As rich copper deposits declined with increased mining efforts, large deposits of leaner ores—oftentimes less than one percent pure—had to be mined to keep pace with America's technological thirst for copper. Processing such ore left an inordinate amount of industrial waste, such as tailings and slag deposits from the refining process and toxic materials from the ores themselves, and copper mining regions around the United States began to see firsthand the landscape degradation wrought by the industry. In The Legacy of American Copper Smelting, Bode J. Morin examines America's three premier copper sites: Michigan's Keweenaw Peninsula, Tennessee's Copper Basin, and Butte- Anaconda, Montana. Morin focuses on what the copper industry meant to the townspeople working in and around these three major sites while also exploring the smelters'environmental effects. Each site dealt with pollution management differently, and each site had to balance an EPA-mandated cleanup effort alongside the preservation of a once-proud industry. Morin's work sheds new light on the EPA's efforts to utilize Superfund dollars and/or protocols to erase the environmental consequences of copper-smelting while locals and preservationists tried to keep memories of the copper industry alive in what were dying or declining post-industrial towns. This book will appeal to anyone interested in the American history of copper or heritage preservation studies, as well as historians of modern America, industrial technology, and the environment.
