Supramolecular chirality usually plays an important role in biological processes and living systems. A novel achiral liquid crystalline polymer where the molecular chirality can be transferred is described, which provides a simple method to realize the chiral selfassembly in achiral polymer systems. However, the supramolecular chiral structures are often easily destroyed by external stimulus due to the weak non-covalent interactions between building blocks, which limits the application of the chiral supramolecular materials to a large extent. In the most systems composed of achiral precursors, the destroyed chiral ordered structures cannot be recovered in the absence of the pre-chiral resource. In order to improve the stability of supramolecular chiral structure and overcome its strong dependence on the chiral resource, a series of achiral liquid crystalline polymers containing phenyl benzoate groups as the mesogens and cinnamic acid groups as the cross-linkable center in the polymer side-chains are designed and synthesized. Combining with the induction of chiral limonene and photo-crosslinking, the supramolecular chirality constructed from achiral polymers can be permanently memorized in a simple way. Even when the helical structure is destroyed by polar solvent or high temperature, the memorized chiral information enables the complete self-recovery behavior without any pre-chiral resource. OBJECTIVE: To probe into the mechanism of Shenfu injection in the treatment of hemorrhagic shock based on network pharmacology and molecular docking technology. METHODS: The main components and action targets of Shenfu injection were obtained by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform. Hemorrhagic shock related targets were obtained by searching the databases of GeneCards, Online Mendelian Inheritance in Man, therapeutic target database and Drugbank. The drug targets and disease targets were intersected and Venn diagram was drawn. The intersection targets were uploaded to STRING for visualization to obtain the proteinprotein interaction ( PPI) network diagram. Cytoscape 3. 9. 1 was used to construct the “ drug-component-targetdisease” network diagram. Gene ontology (GO) functional enrichment analysis and Kyoto encyclopedia of genes and genomes (KEGG) pathway enrichment analysis were performed by using R 4. 2. 1. Pymol 2. 4. 0 and AutoDock Tools 1. 5. 7 were used for molecular docking. RESULTS: The two components of Shenfu injection were ginseng and aconite extract, 25 active components were screened, and 30 common targets were obtained after intersected with drug targets and disease targets. The PPI diagram showed that the degree of TNF, CASP3, IL-1β, PTGS2, ESR1 and CASP8 were higher. The network diagram of Shenfu injection-active component-gene target-disease showed that Beta-sitosterol, Ginsenoside rh2, Deltoin, Karanjin and PTGS2, NCOA2, PTGS1, ADRB2 were of high biological importance. GO functional enrichment analysis mainly focused on the regulation of apoptosis and inflammatory factors. KEGG pathway enrichment analysis showed that influenza A and other pathways were associated with hemorrhagic shock. In molecular docking, the change of intermolecular binding energy of molecular docking confirmed that the effective components of Shenfu injection could bind stably to the target related to hemorrhagic shock. CONCLUSIONS: Shenfu injection has the characteristics of multi-components and multi-targets in the treatment of hemorrhagic shock, which may be related to its reduction of cell apoptosis and inhibition of inflammation. Objective: To explore and analysisi the effects of recombinant human epidermal growth factor combined with photorejuvenation M22 on skin barrier function in patients with acne pitted scars. Methods: From September 2020 to February 2022, 120 cases of patients with acne pitting scars who were diagnosed and treated in our hospital were selected as the research subjects. All the cases were divided into the combination group and the control group with 60 cases in each group, accorded to the simple allocation principle of1:1. The control group were treated with recombinant human epidermal growth factor, and the combination group were treated with photorejuvenation M22 based on the treatment of the control group. Both groups were treated and observed for 4 weeks. Results: After treatment, the total effective rates of the combination group were 88.3 %, which were higher than 66.7 % of the control group (P<0.05). The combined group had fewer clinical symptoms such as erythema regression time than the control group (P<0.05). After treatment, the proportion of skin oil in the two groups were lower than that before treatment, and the water content in the skin were higher than that before treatment (P<0.05). The depth of scar base after treatment in the two groups were significantly lower than that before treatment, and the combination group were significantly lower than that in the control group (P<0.05). The subjective and objective aesthetic scores of the two groups after treatment were higher than those before treatment, and the combination group were higher than that of the control group (P<0.05). Conclusion: Recombinant human epidermal growth factor combined with photorejuvenation M22 can improve skin barrier function in patients with acne pitting scar, can reduce the depth of scar base and the ratio of skin oil, increase skin water content, can promote the improvement of clinical symptoms, and improve skin aesthetics score, thereby improving the overall treatment effect. Objective To investigate the relationship between lymphocyte number reduction, cell proliferation and apoptosis induced by hypoxia exposure. Methods Lymphocytes were isolated from spleen of C57BL/6 mice and cultured in hypoxia (1%O2) and normoxia (21%O2) for 12, 24 and 48 h, respectively. T and B lymphocytes were assessed by flow cytometry. Proliferation and apoptosis of lymphocytes were detected by CFSE and AnnexinⅤ-FITC/PI method, respectively. The morphology of lymphocytes was observed by scanning electron microscope. The mRNA and protein expression levels of apoptosis-related factor bcl-2 homologous antagonist killer protein (Bak), murine B-cell lymphoma/leukemia-2 (bcl-2) and cysteine aspartate protease-3 (caspase-3) were detected by real-time quantitative reverse transcription PCR (qPCR) and Western blot assay. Results Hypoxic exposure for 12, 24 and 48 h can reduce the number of T and B lymphocytes, inhibit the proliferation of lymphocytes and promote cell apoptosis. Scanning electron microscopy showed that at the same time, the characteristic morphological changes of apoptosis appeared earlier in the hypoxic group. After 12, 24 and 48 h hypoxia exposure, the mRNA and protein expression levels of Bak and caspase-3 in lymphocytes were up-regulated, while the mRNA and protein expression levels of bcl-2 were down-regulated. Conclusion Hypoxic exposure mediates the decline of lymphocyte numbers by inhibiting lymphocyte proliferation and promoting apoptosis. [ABSTRACT FROM AUTHOR]

Objective: To study the therapeutic effect of IL-6 antibody on psoriatic mice, and to explore the effect of IL-6 antibody treatment on peripheral blood helper T cells and dendritic cells of psoriatic mice. Methods:Thirty 6-8 week old SPF grade BALB/c mice were divided into Normal group, Model group and IL-6 group according to the random number table method. The Model group and IL-6 group were used to establish psoriasis mouse models by applying imiquimod. Mice in the IL-6 group were treated by subcutaneous injection of IL-6 antibody in the dorsal alopecia. The psoriasis area and severity index (PASI), epidermal thickness and number of blood vessels in the skin lesions of the three groups of mice were compared, serum TNF-α, IL-17 and IL-23 levels, peripheral blood dendritic cells, Th17 cells and Th22 cells ratios and IL-6, IL-21 and STAT3 protein expressions in skin lesions. Results: After IL-6 antibody treatment, PASI, epidermal thickness and blood vessel number, serum TNF-α, IL-17 and IL-23 contents, peripheral blood dendritic cells and Th17 cells of mice in Model group and IL-6 group were compared. The proportions of IL-6, IL-21 and STAT3 in skin lesions were significantly higher than those in the Normal group (P<0.05); compared with the Model group, the PASI in the skin lesions of the IL-6 group mice, epidermal thickness and blood vessel number, serum TNF-α, IL-17 and IL-23 levels, peripheral blood dendritic cells, Th17 cells and Th22 cell ratios, and IL-6, IL-21 and STAT3 protein expressions in skin lesions were significantly decreased (P<0.05). Conclusion: IL-6 antibody has a good therapeutic effect on the psoriasis mouse model, and its mechanism may be related to the influence of Th17/Th22 and dendritic cell levels by inhibiting the IL-6/STAT3 pathway.
