Abstract

MicroRNAs (miRs) were involved in numerous cardiovascular diseases, especially ischemic heart diseases, but the miR changes during cardiac ischemia–reperfusion (I/R) injury following sevoflurane (SEV) preconditioning are still unknown. This study aims to investigate the effect of miR‐874 on cardiac I/R injury in mouse models pretreated with SEV. Following establishment of mouse models with myocardial I/R injury, mice were pretreated with SEV. The functional mechanism of miR‐874 in I/R injury was explored when miR‐874 and the Janus kinase 2 (JAK2)/signal transducer and activator of transcription 3 (STAT3) signaling pathway were inhibited. Terminal deoxynucleotidyl transferase (TdT)‐mediated dUTP biotin nick‐end labeling (TUNEL) staining was used to detect cardiomyocyte apoptosis and dual luciferase reporter gene assay to identify the targeting relationship between miR‐874 and STAT3. Expression of the JAK2/STAT3 signaling pathway and apoptosis‐related genes was determined. Initially, upregulated miR‐874 was observed in I/R mice. Then, miR‐874 inhibition improved cardiac function of I/R mice, inhibited cardiomyocyte apoptosis (also shown as decreased Bcl‐2 associated X protein B [Bax] and increased B‐cell lymphoma‐2 [Bcl‐2]), and activated the JAK2/STAT3 signaling pathway. STAT3, a target gene of miR‐874, was upregulated following miR‐874 inhibition. Finally, we also observed that the effect of miR‐874 was lost when the JAK2/STAT3 signaling pathway was blocked. The findings indicate miR‐874 as a contributory role in cardiac I/R injury, with miR‐874 inhibition alleviating cardiac I/R injury in mice following SEV pretreatment by targeting STAT3 through the JAK2/STAT3 signaling pathway. Abstract

Background Patients with developmental dysplasia of the hip (DDH) may have decreased blood supply to the femoral heads. Finding a non-invasive method to evaluate whether the femoral heads in patients with DDH are ischemic is paramount for orthopedic surgeons. Purpose To identify whether parameters reflecting perfusion and diffusion in intravoxel incoherent motion (IVIM) sequences can be used to assess ischemia in femoral heads in patients with DDH after closed reduction. Material and Methods Twenty-eight patients with DDH who had undergone closed reduction were enrolled. IVIM data were acquired using a 3-T magnetic resonance scanner, regions of interest were placed on the epiphyses; ADCslow, ADCfast, f, and ADCfast×f were measured. A Mann-Whitney U test was performed to compare ADCslow, ADCfast, f, and ADCfast×f between the lesion and control sides. Receiver operating characteristic curves were generated with respective cut-off values. The lesion sides were classified based on the International Hip Dysplasia Institute (IHDI) classification. ADCslow, ADCfast, f, and ADCfast×f were compared among the groups. Results ADCslow was higher and ADCfast, f, and ADCfast×f were lower on the lesion sides ( P = 0.000-0.002). The optimal cut-off value for ADCfast×f, ADCfast, ADCslow, and f were 0.030, 0.626, 0.000251, and 0.636, respectively. Higher IHDI classification scores on the lesion side were associated with lower ADCfast, f, and ADCfast×f, and higher ADCslow values. Abstract

In this paper, the effect of the ramie fiber properties on yarn quality was analyzed. Due to the complex nonlinear relationship between the ramie fiber properties and yarn quality, three methods, the grey analysis combined with BP neural network, principle component analysis combined with BP neural network and pure BP neural network were applied to predict yarn quality on the basis of ramie fiber properties, respectively. The grey analysis and principle component analysis were expected to reduce the input layer node numbers of BP neural network, then the network structure can be simplified, therefore the prediction accuracy and stability can be improved. Compared with that of pure BP neural network, the results got from the other two methods are both better, the mean relative error between the forecast results and measured values of ramie yarn quality, such as the strength, strength irregularity, unevenness and neps, were all reduced greatly.

Abstract

The technology of reducing yarn hairiness using air-jet nozzle on the winder (JetWind) is a combination of the ring spinning and air-jet (vortex) spinning technologies. A dynamic model for the coupling between the ramie yarn hair and the airflow field inside the hairiness-reducing nozzle on the winder is developed in this research. Based on the model, computer simulation of the dynamics of the hair in the nozzle is performed and the hairiness-reducing principle is theoretically investigated. The high-speed photography technology is used to capture experimentally the dynamics of the hair in the nozzle to verify the simulation result. The dynamics of the hair in the nozzle is obtained through the computer simulation which indicates the principle of reducing ramie yarn hairiness using air-jet nozzle on the winder is to make the hair wrap onto the yarn body with the action of the swirling airflow. The high-speed photography validates the simulation result.

Abstract

Background: This study demonstrates a critical role in CNS innate immunity of the microglial Toll-like receptor 4 (TLR4) in the induction and maintenance of behavioral hypersensitivity in a rat model of bone cancer pain with the technique of RNA interference (RNAi). We hypothesized that after intramedullary injection of Walker 256 cells (a breast cancer cell line) into the tibia, CNS neuroimmune activation and subsequent cytokine expression are triggered by the stimulation of microglial membrane-bound TLR4. Results: We assessed tactile allodynia and spontaneous pain in female Sprague-Dawley (SD) rats after intramedullary injection of Walker 256 cells into the tibia. In a complementary study, TLR4 small interfering RNA (siRNA) was administered intrathecally to bone cancer pain rats to reduce the expression of spinal TLR4. The bone cancer pain rats treated with TLR4 siRNA displayed significantly attenuated behavioral hypersensitivity and decreased expression of spinal microglial markers and proinflammatory cytokines compared with controls. Only intrathecal injection of TRL4 siRNA at post-inoculation day 4 could prevent initial development of bone cancer pain; intrathecal injection of TRL4 siRNA at post-inoculation day 9 could attenuate, but not completely block, well-established bone cancer pain. Conclusions: TLR4 might be the main mediator in the induction of bone cancer pain. Further study of this early, specific, and innate CNS/microglial response, and how it leads to sustained glial/neuronal hypersensitivity, might lead to new therapies for the prevention and treatment of bone cancer pain syndromes.
