Trigonella foenum-graecum (fenugreek), a legume has been used as spice throughout the world to enhance the sensory quality of foods, is considered to have a high nutraceutical value. The antidiabetic actions of Trigonella seeds have been speculated due to the presence of steroid saponins, alkaloids, and fiber content. Five bioactive compounds, namely, trigonelline, isoorientin, orientin, vitexin, and isovitexin, were detected and quantified by HPLC system. Further rapid detection of bioactive compounds was achieved using high-performance liquid chromatography hybrid electrospray quadrupole time-of-flight mass spectrometric (HPLC-ESI-QTOF-MS/MS) method. Twenty-five compounds were tentatively identified based on their retention time and measured accurate mass. The fragmentation patterns of known compounds were utilized to elucidate their structures. Six of these identified compounds, that is, 4-hydroxyisoleucine, trigonelline, isoorientin, isovitexin, pinitol, and sarsasapogenin, were simultaneously quantified by UHPLC-ESI-MS/MS method under the multiple reactions monitoring (MRM) mode using ultrahigh-performance liquid chromatography hybrid electrospray triple quadrupole linear ion trap mass spectrometer. The analytical method was validated and successfully applied for simultaneous determination of compounds in Fenugreek seeds, which indicated the suitability of the validated method for its quality control.

Introduction: Atrophic vaginitis is a common problem in postmenopausal women and results from decreased levels of blood estrogen. It is associated with symptoms of itching, burning, dyspareunia, and postmenopausal bleeding. The present study evaluated the effects of fenugreek extract on atrophic vaginitis., Materials and Methods: This randomized controlled clinical trial was performed on 60 postmenopausal women in Ardabil, Iran, in 2018. The participants were selected using block randomization with the allocation ratio 1:1. Those in the intervention group received 0.5g (the applicator filled to the half-full mark) fenugreek vaginal cream 5% twice a week for 12 weeks. The control group received conjugated estrogens vaginal cream at the dose of 0.625 mg (the applicator filled to the half-full mark) containing 0.3 mg of conjugated estrogens. Atrophic vaginitis was evaluated before and after the treatment through clinical examination, clinical signs, and measurement of Vaginal Maturation Index (VMI)., Findings: After the 12-week intervention and modification of the baseline score, the mean (standard error) score for atrophic vaginitis signs was 3.100 (1.43-4.75). This difference was statistically significant in intragroup comparison and in favor of the control group in intergroup comparison (p=0.001). VMI was less than 49% in 86.7% and 46.7% of the participants in the intervention and control groups, respectively. This was a significant difference in favor of the control group (p=0.001)., Conclusion: The results of this study showed that total fenugreek extract could be effective in treating signs of atrophic vaginitis, but it was not as effective as ultra-low-dose estrogen., (Copyright© Bentham Science Publishers; For any queries, please email at epub@benthamscience.net.)

The objective of this study was to hydrophobically modify fenugreek gum (FG) and to further evaluate the potential application of the obtained derivative in liver-targeted drug delivery system. Stearic acid (C 18 ) was conjugated with FG (FG-C 18 ) by a simple esterification reaction. The obtained FG-C 18 was then characterized on its chemical structure by Fourier transform infrared spectroscopy and 1 H-nuclear magnetic resonance. The self-assembled nanomicelles (NMs) of FG-C 18 in water were prepared by an ultrasonication method. The average diameter and zeta potential of FG-C 18 NMs were 196.70 ± 6.12 nm and -31.79 ± 1.58 mV, respectively. FG-C 18 NMs appeared as spherical particles under transmission electron microscopy and possessed a critical micellar concentration of 0.042 mg/ml by pyrene fluorescence probe method. A low toxicity of FG-C 18 was revealed on both HepG2 and MCF-7 cells at 0.1-100 mg/ml. Haemolysis of FG-C 18 was less than 5%. Cellular uptake of coumarin-6 into HepG2 cells was enhanced by treating with C6-loaded FG-C 18 NMs compared to free coumarin-6. These results suggest that FG-C 18 have a potential application for a liver targeted drug delivery.

The purpose of this in vitro study was to investigate the effect of Trigonella foenum graecum seed extract (TFGSE) as a root canal irrigation agent on the surface of instrumented root canals and on changes in the mineral contents of the root dentin. A total of 90 extracted human mature maxillary central incisor teeth with a single root and without root resorption were selected. The specimens were subdivided into five groups according to irrigation protocol groups G0: distilled water (n = 15): G1: Sodium hypochlorite (5.25% NaOCl); G2: 1% TFGSE G3: NaOCl +17% EDTA (3 min); G4: NaOCl+1% TFGSE; G5: 1% TFGSE +17% EDTA. All the roots were sectioned longitudinally into two halves and examined under scanning electron microscope/energy dispersive X-ray analysis (SEM/EDX) to assess changes in the mineral content of the tooth. Smear layer removal from root canals was evaluated with scanning electron microscopy (SEM). A statistical analysis was performed using the Kruskal-Wallis test. The most effective irrigation protocols in removing smear layer occurred with TFGSE and NaOCl, both with rinsing with 17% EDTA. TFGSE was more effective than NaOCl for removing the smear layer. The results showed the ability of 1% TFGSE to remove the smear layer from the root canals. TFGSE is a promising alternative irrigation agent for root canals., (© 2019 Wiley Periodicals, Inc.)

This work is about the utilization of fenugreek seed as an antibacterial and anticancer agents. The antibacterial activity of fenugreek seed extract on six pathological bacteria strains were specified through conventional biochemical tests using the Vitek2 automated system and diffusion agar method. The anticancer activities of fenugreek seed extract, on MCF-7 breast cancer cells, liver cancer HCAM cells and the non-cancerous Vero cell lines, were investigated using colorimetric MTT assay. Results showed that the highest activity of the extract of the seed was found on Staphylococcus aureus and Pseudomonas aeruginosa (22 mm and 17 mm diameter of inhibition zones respectively). The seed extract showed proliferative inhibition on MCF-7 cell line at a concentration of 400 µg/ml and 72 h of the incubation period. This was accompanied by insignificant apoptosis or necrosis. The seed extract showed no anticancer effect on liver and Vero cell lines. This work emphasizes that fenugreek seed extract is a potential source of antibacterial and anticancer agents., .

Background: Polycystic ovary syndrome (PCOS) is a frequent medical condition characterized by both metabolic and reproductive disorders. Different pharmaceutical treatments have been proposed for PCOS. However, side effects of long-term treatments and their probable low efficacy have made complementary and alternative treatments a valuable option. Recent reports have indicated the increased use of complementary treatments. Herbal medicine, as part of complementary medicine, was find introduced in traditional Persian and Chinese medicine. Medicinal herbs have used for a long time in the treatment of gynecological and infertility problems of PCOS patients. In this study, we aimed to review herbal medicines used for PCOS worldwide., Methods: PubMed, Embase, Cochrane, and Scopus databases were searched for clinical trials and Randomized Controlled Trials based on related keywords. Data were collected from 1990 to 2019., Results: According to a multitude of studies, a wide spectrum of herbs can be used to improve various aspects of PCOS. Herbs such as Cinnamomum verum, Trigonella foenum-graecum L., and Vitex agnus-castus can impact on menstrual and ovulatory dysfunctions, obesity, insulin resistance, lipid-metabolism dysfunction, and androgen excess-related conditions., Conclusion: Some plants as natural remedies may have beneficial effects on improving different aspects of PCOS; but further studies are needed to investigate their mechanisms and safety.

Fenugreek gum is modified using two different techniques: microwave-assisted and thermal synthesis technique of graft copolymerization. The synthesis was done by varying the concentration of acrylamide monomer and CAN initiator to get the best-grafted product. The physicochemical characteristics were determined by elemental analysis, FTIR spectroscopy, SEM, XRD, DSC, TGA, number-average molecular weight and intrinsic viscosity. All the grafted materials were tested by flocculation of different synthetic suspension media as well as river water. The process of deflocculation and refloccculation was also carried out and it was observed that refloccculation process has a good potential as the flocculation efficacy increased to a considerable extent. The flocculation efficacy increased from 92.05% to 94.32% upto a maximum of 95.98% in refloccculation process. The flocculation efficacy was found out to be 95.98%, 40.52% and 68.94% respectively kaolin, coal fine and iron ore suspensions. Flocculation was done in kaolin suspension of pH 2, 4, 7 and 10 and at different temperature range of 10 0 , 25 0 , 40 0 and 55 0 C. Atherosclerosis is one of the leading causes of death in patients with cardiovascular diseases worldwide. Although some progress has been made in the treatment of cardiovascular diseases, the morbidity and mortality of cardiovascular diseases still continue to rise. At present, it is an important topic for researchers to develop safe and effective drugs from natural products to prevent and treat cardiovascular diseases. Diosgenin (DSG) is a plant sterol saponin mainly found in natural medicinal plants such as fenugreek seeds and wild yam tubers. More and more studies have reported that DSG has significant pharmacological activities such as anticancer, cardiovascular protection, hypolipidemic, anti-inflammatory, and neuroprotection. Furthermore, diosgenin is also an important basic raw material for the preparation of steroids and contraceptives in the pharmaceutical industry. Numerous preclinical studies have shown that DSG has great potential in the treatment of various cardiovascular diseases in vivo and in vitro, especially in atherosclerosis. This review mainly discusses the effects of DSG on endothelial dysfunction, lipid profile, and macrophage foam cell formation, VSMC viability, thrombosis and inflammation during the formation of atherosclerosis. Also, the mechanism of DSG on atherosclerosis was elaborated in detail. It is noteworthy that newly synthesized DSG derivatives and DSG delivery systems have good antithrombotic activity and pharmacokinetic characteristics.

Dietary administration of some plant-derived substances have been proved of great economic value in aquaculture. In order to investigate the effects of dietary fenugreek seed extracts (FSE) on juvenile blunt snout bream (Megalobrama amblycephala), a feeding trial was conducted for 8 weeks. The results showed that final weight (FW), weight gain (WG), feed conversion ratio (FCR) and specific growth rate (SGR) were not significantly affected by dietary FSE levels. The whole body lipid contents of fish fed with 0.04%, 0.08% and 0.16% FSE diets were significantly lowered compared to the control group. Dietary FSE diets significantly affected plasma complement component 3 (C3), immunoglobulin M (IgM), albumin (ALB) and total protein (TP). The relative expressions of acetyl CoA carboxylase (ACC), fatty acid synthase (FAS) and sterol regulatory element binding protein-1 (SREBP1) mRNA in the liver of fish decreased significantly with increasing dietary FSE levels from 0% up to 0.04%. FSE supplementation diets lowered the liver pro-inflammatory genes expressions by regulating tumor necrosis factor-α (TNF-α) and interleukin 8 (IL-8) mRNA levels and increased anti-inflammatory genes expression by regulating transforming growth factor (TGF-β) and interleukin 10 (IL-10). FSE diets increased growth factor-1 (IGF-1) and target of rapamycin (TOR) mRNA levels from 0% up to 0.04%, 0.04% FSE diets significantly increased growth factor-1 (IGF-1) mRNA levels and S6 kinase-polypeptide 1 (S6K1) mRNA levels compared to the control group. 0.04% FSE diets significantly increased superoxide dismutase (SOD) activities and 0.08% FSE diets significantly increased catalase (CAT) and glutathione peroxidase (GPx) activities, 0.16% FSE diets significantly increased total antioxidant capacity (T-AOC) activities compared to the control group. Additionally, compared to the control group, 0.04% dietary FSE significantly up-regulated nuclear factor erythroid 2-related factor 2 (Nrf2) mRNA levels and glutathione peroxidase-1 (GPx1) mRNA levels, at the same time, 0.02%, 0.04%, 0.08%, 0.16% FSE diets significantly down-regulated kelch-like ECH-associated protein 1 (Keap1) mRNA levels. However, no significant effects were observed on copper zinc superoxide dismutase (Cu/Zn-SOD) and manganese superoxide dismutase (Mn-SOD). Fenugreek is a leguminous plant belongs to the family Fabaceae, which is extensively cultivated as a semiarid crop in Northern Africa, the Mediterranean, India, and Canada. In the present review paper, first we summarized the extraction, purification, chemical composition, molecular structure, and rheological behavior of the mucilages isolated from Fenugreek seeds (FSG), and then their functional properties presented to elucidate the potential application of this traditional source of hydrocolloids in food, pharmaceutical, and other industries. To date, there is no technique that can successfully remove the attached protein from FSG. From a structural point of view, galactose and mannose are the most abundant polysaccharide in FSG composition, suggesting a galactomannan-like structure. FSG is the most soluble of the seed gums. FSG solutions at various temperatures and concentrations showed a time-dependent shear thinning behavior. Furthermore, these hydrocolloids can be employed for the fabrication of eco-friendly packaging systems. Antioxidant capacity and anti-fungi activity of FSG has been proved in different studies. In conclusion, industrial applications of FSG are possible due to its strong thickening properties. The present study was accomplished to examine and compare the effect of specific antioxidant-rich oils on hyperglycemia, dyslipidemia, renal function markers and oxidative renal damage in diabetic rats for four weeks. Papaya (P), olive (O), fenugreek (Fe), bitter gourd (B) and fish (Fi) oils were used for this purpose. Streptozotocin (STZ) was injected intraperitoneally in a single dose to induce diabetes. All oils were given orally at a dose of 3g/kg for four weeks in respective group after induction of diabetes. After treatment with oils, blood was collected, and their kidneys were stored. The level of fasting blood glucose (FBG), glycated hemoglobin (HbA1c), total cholesterol (TC), triglycerides (TG), low-density lipoprotein-cholesterol (LDL-C) and very low-density lipoprotein-cholesterol (VLDL-C) increased while amylase and high-density lipoprotein cholesterol (HDL-C) level decreased in the diabetic rats. These changes were augmented by fenugreek, bitter gourd and olive oils treatment. Diabetic rats showed elevated renal function markers in serum, including, serum creatinine (Scr), blood urea nitrogen (BUN) and alkaline phosphatase (ALP), which were restrained significantly by fenugreek and bitter gourd oil treatment. Moreover, fenugreek and bitter gourd oils treatment significantly modulated the level of thiobarbituric reactive substances (TBARS), malonaldehyde (MDA) and catalase (CAT) in the kidney of diabetic rats. The histopathological examination also showed the protective effect of these oils. The study suggests that vegetable oils are effective in reducing hyperglycemia, dyslipidemia and renal damage related to the side effects of diabetes. Thus they may have therapeutic value for preventing diabetes side effects and may be included in oil diet treatment synergically. Thus, our data suggest that oils as potent antidiabetic agent and beneficial in the control of diabetes-related abnormalities such as hyperglycemia, dyslipidemia and renal damage of STZ induced rat model of type 2 diabetes. Our study also supports the suggestion that synergistic possibilities exist concerning the use of these oils in the treatment of diabetes mellitus.

Galactomannan extracted from Trigonella persica endosperm like to other galactomannans is useful in the medicine and pharmaceutical industry such as drug delivery due to its unique physicochemical properties. As soon as germination takes place, the hydrolysis of galactomannan begins and its structure changes. Evaluation of galactomannan content extracted from the germinating seeds (GEGS) and the non-germinated seeds (GENGS) showed that the amount of galactomannan was highest at 24 h after imbibition and was completely consumed approximately 48 h after imbibition. It seems that galactomannan is used for storage reserve synthesis in the cotyledon during germination. Based on the quantitative assessment, FT-IR, macroscopic and microscopic observations, galactomannan of T. persica seeds have high an M/G ratio (approximately 5:1) and this ratio does not alter during germination. Therefore, changes in galactomannan structure (functional group and band) caused that GEGS have different physicochemical properties than GENGS. A novel electropolymerized molecularly imprinted polymer (MIP) film, based on diosgenin on a glassy carbon electrode (GCE), has been synthesized. Based on density functional theory (DFT), para-aminobenzoic acid (pABA), which is a green substance, was selected from five functional monomers by Gaussian software for MIP production, to be the suitable monomer. The MIP's synthesis conditions were optimized, and the imprinting effect was confirmed by comparing the electrochemical reaction of MIP, with that of non-imprinted polymer (NIP). The calibration curve of diosgenin on MIP/GCE was obtained with a linear range of 0.003 to 0.13 mM. The limit of detection (LOD) and limit of quantification (LOQ) were determined to be 8.95 × 10 -4  mM and 2.98 × 10 -3  mM, respectively. This method exhibited good stability, high sensitivity, and high selectivity for diosgenin. The developed method is the first method reported to be used for the electroanalysis of diosgenin, and it has been successfully applied to the analysis of diosgenin in Trigonella foenum graecum seed extract., (Copyright © 2019. Plants are extensively used in treating inflammatory disorders. The current study focused on evaluating anti-inflammatory potential of Trigonella gharuensis. The ethanolic and n-hexane extracts of T. gharuensis were given orally at the dose of 400mg/kg/day. Various in vivo inflammatory models such as carrageenan-, histamine-, dextran- and serotonin-induced paw edemas; xylene-induced ear edema, and castor oil-induced diarrhea models were used for validation of different mechanisms of autacoid inhibition. Gas chromatography-mass spectrometry analyses were performed to find out compounds responsible for anti-inflammatory properties. Both extracts significantly inhibited (P<0.05) carrageenan-, histamine-, dextran- and serotonin-induced paw edema in early and late acute inflammation phases. Suppression of xylene-induced ear edema supported suggested inhibition of autacoids. Attenuation of castor oil-induced diarrhea suggested prostaglandin inhibition by both extracts and supported inhibition of carrageenan-induced inflammation in the late phase. GC-MS analysis indicated constituents with considerable biological activities such as, saturated and unsaturated fatty acid esters, coumarins, terpenes, and aromatic and aliphatic compounds in the extracts. In conclusion, extracts of T. gharuensis possess significant anti-inflammatory activity which might be ascribed to the inhibition of autacoids.

The study aimed to determine the impact of treated domestic wastewater on seed germination, seedling growth and reserve mobilization from Fenugreek (Trigonellafoenum graecum L.). Seeds were germinated by soaking in distilled water (H 2 O) or wastewater treated with various methods: activated sludge processes (T1), facultative lagoons treatment (T2) and dilution. Results show high levels of organic matter (OM), suspended solids (TSS) and nutrients in TWW (T2) than that of TWW (T1). The embryo length and biomass of fenugreek imbibed by TWW were higher in TWW (T2) compared with TWW (T1). There was more reduction in free amino acids and soluble sugar contents in fenugreek treated with TWW (T1) than treated by TWW (T2). Important solutes leakage is recorded by measuring electric conductivity during seed imbibition with TWW. Improving the quality of wastewater by dilution (50%) stimulated germination of seeds and the growth of the tested plant. Moreover, it significantly reduces the solutes leakage and enhanced seed metabolites accumulation.

The aim of the present research was to determine the trace metal accumulations in Trigonella foenum-graecum irrigated with three different water regimes (ground water, canal water and sugar mill water). Also, transfer factors, pollution load indices, and health risk indices were assessed to evaluate metal transport and accumulation through the food chain. The analysis was conducted by Atomic Absorption Spectrophotometer (Shimadzu model AA-6300) to evaluate the concentration of metals in water, soil and vegetables. Trace metal concentrations in water samples ranged from 0.84 to 1.67, 0.42 to 0.72, 0.45 to 0.85, 2.51 to 9.99, 1.21 to 1.92, 1.82 to 9.98 and 0.64 to 0.91 mg/kg for Cd, Cr, Cu, Fe, Ni, Zn and Mn, respectively. The mean metal concentrations in soil samples were determined as 0.25, 0.86, 0.96, 3.37, 0.4, 0.44 and 2.31 mg/kg for Cd, Cr, Cu, Fe, Ni, Zn and Mn, respectively. Trace metal accumulations in T. foenum-graecum samples gathered from where soil samples were taken are as follows: The contents of Cd, Cr, Cu, Fe, Ni, Zn and Mn ranged from 0.48 to 1.06, 0.11 to 0.35, 0.15 to 0.29, 1.43 to 8.39, 0.39 to 0.54, 2.1 to 3.05 and 0.42 to 0.47 mg/kg, respectively. Statistical analyses showed that the treatments have non-significant effect (p > 0.05) on concentrations of metals in T. foenum-graecum samples collected from three sites for Ni, Cr, Cu, Zn and Mn and significant effect on Fe and Cd.

The effects of soaking and cooking on soluble sugars, alpha-galacto-oligosaccharides (α-GOS) and soluble dietary fibres (SDF) and insoluble dietary fibres (IDF) were assessed in five legumes (lentil, chickpea, fenugreek, faba bean and Egyptian faba bean). In raw seeds, total α-GOS content ranged from 2500 mg/100 g (chickpea) to over 4000 mg/100 g (fenugreek). Stachyose was predominant in fenugreek, lentil and chickpea, whereas verbascose was the main α-GOS in faba bean and Egyptian faba bean. IDF represented 69-87% of the total dietary fibres in all studied legumes, while SDF content varied noticeably. During soaking, total α-GOS content decreased between 10% (lentil and faba bean) and 40% (chickpea). In fenugreek, soaking reduced IDF and increased SDF concentration, possibly due to partial IDF solubilisation from the cell wall. Cooking further decreased α-GOS and increased total dietary fibre content. The different behaviours of these five legumes during processing illustrate the high biodiversity within legume species.

Background Natural medicinal plants have been the focus of current research for developing neuroprotective agents to be used in the diabetes-linked cognitive dysfunction. Trigonella foenum-graecum seeds (known as fenugreek, methi in Hindi), is a well-known traditional medicinal herb and possesses anti-diabetic, anti-oxidant, and anti-inflammatory properties. Purpose This study was undertaken to explore the ameliorative effects of T. foenum-graecum seed extract on diabetes-induced cognitive dysfunction. Methods Experimental diabetes was induced by administering a single dose of streptozotocin (60 mg/kg) through intraperitoneal dose. Cognitive function was assessed using a T-maze and the Morris water maze. Lipid peroxidation levels and oxidative stress in the hippocampus was measured. Quantification of hippocampal CA1 and CA3 regions was done using cresyl violet stain. Results Diabetic rats demonstrated learning and memory impairment, which was evident from poor performance in behavioral tasks, i.e. T-maze and Morris water maze tasks. Learning and memory impairment in diabetic animals is associated with increased blood glucose levels, increased oxidative stress in the hippocampus and decreased number of neurons in the CA1 and CA3 regions of the hippocampus. The diabetic rats administered with T. foenum-graecum showed improved performance in behavioral tasks, and these changes were associated with decreased blood glucose levels, decreased oxidative stress in the hippocampus, and decreased neuronal loss from the CA1 and CA3 regions of the hippocampus. Conclusion In conclusion, administration of T. foenum-graecum seed extract ameliorates diabetes-linked cognitive dysfunction in rats by decreasing blood glucose levels, reducing lipid peroxidation and oxidative stress in the hippocampus, and preventing neuronal loss from the hippocampus.

Background: The seed of Trigonella foenum-graecum L. (Methika in Sanskrit) is a well known kaphahara (balancing kapha) herb in Ayurveda indicated in Prameha or early diabetes mellitus. It is also useful in obesity and reduces lipid level of blood., Purpose: We aimed to explore the metabolites present in the plant extract and to establish the combination synergy and the network pharmacology along with the underlying the mechanism of action involved., Study Design: LC-MS/MS based metabolite screening followed by ADME screening and finally network pharmacology exploration of the mechanism of action involved against hyperlipidemia and hypolipidemia with neighbourhood based combination synergy approach., Methods: Ethanolic extract of Trigonella foenum-graecum L. (TFHE) was subjected to LC-MS/MS analysis to identify the active constituents. Oral bioavailability and drug likeness was screened for all the compounds. Databases- Binding DB, DAVID, KEGG and STRING were used to gather information to develop the networks. The networks were constructed using Cytoscape 3.2.1. Combination synergy analysis was performed with the help of Cytoscape network analyzer tool with neighbourhood approach., Results: The LC-MS/MS analysis identified 13 compounds which were found to be bio-available and drug like following the QED and Veber drug likeness parameters. The pathway analysis showed enrichment for different pathways like MAPK pathway (p-4.69E-07), JAK-STAT pathway (p-6.30E-05), Adipocytokine (p-0.00179), Type 2 Diabetes mellitus (0.00441), Insulin signalling pathway (p-0.0121), mTOR signalling pathway (p-0.000378), which are all connected to hyperlipidemia and hyperglycemia. The combination synergy network identified 23 targets interacting with 13 compounds based on a network neighbourhood approach., Conclusion: The network pharmacology analysis strongly suggested the multimode evidences that TFHE largely works on the insulin signalling pathway and mainly based on its antioxidant potential due to its interaction with carbonic anhydrase. Various compounds were found to be interacting with key proteins that activates EGFR/AKT/mTOR signalling cascade which has therapeutic implication in hyperglycemia and hyperlipidemia. Background The antidiabetic and antioxidant effects of Trigonella foenum-graceum have been suggested. The effects of hydroalcoholic extract of the plant seeds and metformin against the diabetes-induced memory impairment were investigated. Materials and methods The rats were treated: (1) control, (2) diabetic (3-6) and diabetic rats treated by 50, 100 and 200 mg/kg of the plant extract or metformin. The rats were diabetic by streptozotocin (STZ, 55 mg/kg). After the passive avoidance test, malondialdehyde (MDA), nitric oxide (NO) metabolites, total thiol (SH), catalase (CAT) and superoxide dismutase (SOD) were determined in the brain. Results In the diabetic group, at 3, 24 and 48 h after receiving a shock, the latency to enter the dark room was lower than for the controls (p < 0.001). All doses of the extract and metformin increased the latencies to enter the dark at 3 and 24 h after the shock treatment (p < 0.05-p < 0.001). Additionally, the two higher doses of the extract and metformin increased the latency at 48 h after the shock (p < 0.05-p < 0.001). Diabetes also elevated MDA and NO metabolites, while it reduced thiol, SOD and CAT in the hippocampal and cortical tissues (p < 0.001). Treatment of the diabetic animals by the highest dose of the extract and also metformin reduced the MDA and NO metabolites, while it improved thiols, SOD and CAT (p < 0.01-p < 0.001). Conclusions Based on our findings, metformin and the hydro-alcoholic extract from the T. foenum-graceum seed prevented memory deficits resulting from diabetes. Preventing oxidative damage in the brain may at least, in part, be responsible for the positive effects of the extract and metformin.

This study aimed to evaluate the protective effect of a polysaccharide extracted from fenugreek seeds (Trigonella foenum-graecum) (FWEP) against insecticide-thiamethoxam (TMX)-induced hepatotoxicity. Obtained data exhibited potent antioxidant and antibacterial potentialities. On other trend, in vivo, adult female rats were divided into four groups: controls; TMX (100 mg/kg of body weight); TMX + FWEP at two graded doses, respectively (100 and 200 mg/kg of body weight) for 30 d. Up to TMX treatment, our data showed a significant increase in plasma markers of hepatotoxicity including alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, and in lactate dehydrogenase (LDH) activities, which is coordinated with decline in total protein and albumin levels. Remarkably, a clear sign of genotoxicity was delivered by total disruption in hematological parameters and micronucleus (MN) test shown by severe chromatin degradation. These data were also associated with oxidative stress set up, histological and DNA injuries. However, co-administration with FWEP succeeded significantly in a dose-dependent manner in reducing and healing liver's hematological and genotoxic induced by TMX injuries.

This study reports a facile and ecofriendly approach for the ultrasound assisted synthesis of silver and iron oxide nanoparticles and their enhanced antibacterial and antioxidant activities. The fenugreek seed extract was used as reducing, capping, and stabilizing agent in the synthesis of nanoparticles. The transmission electron microscopy results showed that nanoparticles synthesized by ultrasonication have a smaller size (~20 nm) as compared to the nanoparticles fabricated by magnetic stirring (~40 nm). The color change of the solution from milky white to brown suggested the formation of silver nanoparticles which was confirmed by the presence of an absorbance peak at 396 nm. The results of powder X-ray diffraction and energy dispersive X-ray spectroscopy confirmed the crystallinity and elements present in nanoparticles synthesized using fenugreek seed extract. Fourier transform infrared analysis showed that the fenugreek seed phytochemicals were coated on the nanoparticle surface. Thermal gravimetric analysis showed the thermal degradation and stability of nanoparticles. Magnetization study of iron oxide nanoparticles confirmed the superparamagnetic nature. The silver nanoparticles showed antibacterial activities against both gram-negative ( Escherichia coli ) and gram-positive ( Staphylococcus aureus ) bacteria, while no antibacterial activities were observed for iron oxide nanoparticles. The ultrasound assisted nanoparticles showed higher stability and antibacterial and antioxidant activity compared with the nanoparticles fabricated by magnetic stirring.

Fenugreek is known since ancient times as a traditional herbal medicine of its multiple beneficial effects. Fenugreek's most studied and employed effect is its hypoglycemic property, but it can also be useful for the treatment of certain thyroid disorders or for the treatment of anorexia. The regulation of glucose homeostasis is a complex mechanism, dependent on the interaction of different types of hormones and neurotransmitters or other compounds. For the study of how diosgenin and fenugreek seeds modify insulin sensitivity, we used a rat insulin resistance model induced by high-fat diet. Diosgenin in three different doses (1mg/bwkg, 10mg/bwkg, and 50 mg/bwkg, respectively) and fenugreek seed (0.2 g/bwkg) were administered orally for 6 weeks. Insulin sensitivity was determined by hyperinsulinemic euglycemic glucose clamp method. Our research group found that although glucose infusion rate was not significantly modified in either group, the increased insulin sensitivity index and high metabolic clearance rate of insulin found in the 1 mg/kg diosgenin and the fenugreek seed treated group suggested an improved peripheral insulin sensitivity. Results from the 10 mg/kg diosgenin group, however, suggest a marked insulin resistance. Fenugreek seed therapy results on the investigated anabolic hormones support the theory that, besides insulin and gastrointestinal peptides, the hypothalamic-hypopituitary axis regulated hormones synchronized action with IGF-1 also play an important role in the maintaining of normal glucose levels. Both diosgenin and fenugreek seeds are capable of interacting with substrates of the above-mentioned regulatory mechanisms, inducing serious hormonal disorders. Moreover, fenugreek seeds showed the ability to reduce the thyroid hormone levels at the periphery and to modify the T4/T3 ratio. It means that in healthy people this effect could be considered a severe side effect; however, in hypothyroidism this effect represents a possibility of alternative natural therapy.

Gelling properties of three galactomannan dispersions (fenugreek (FG), guar (GG) and locust bean gum (LBG)) were compared by strain and frequency sweep viscoelasticity, and temperature cycle at a constant strain and frequency in the concentration range 1 to 2 wt%. For FG and GG dispersions, tan δ remain <1 in the linear viscoelastic region during strain sweep analysis. FG and GG dispersions also showed a sol to gel transition at higher frequencies compared to LBG, indicating gelation by inter-polymer interactions and network formation. Conversely, strain and frequency sweep analysis of LBG dispersions revealed poor gelling ability. The fractal dimensions of the gels were 1.82, 2.38 for FG and 1.64, 2.10 for GG, using Shih (1990) and Wu-Morbidelli model (2001), respectively. According to the Shih model, FG and GG fell into the strong gel regime, but the Wu-Morbidelli model revealed that both dispersions were in the transition gel regime, more inclined towards weak gels, which is a more accurate description of the gels. FG and GG could form thermo-reversible gels, while for LBG, an improvement in gel strength was observed after the temperature cycle. Currently, metallic nanoparticles possessing versatile heterogeneous catalytic functionality such as in hydrogenation, water splitting, hydrogen production and CO 2 reduction for global pollution remediation have been paid great attentions due to their high chemical stability, superior activity and unique electrical and optical properties. However, the gradual degradation of their catalytic activity on multiple usage limits the monometallic nanoparticles to industrial applications. Herein, we fabricated the highly porous fenugreek polysaccharide assisted green synthesis of AuPd nanostructures for heterogeneous catalytic hydrogenation of the industrial usable highly toxic 4-nitrophenol to the medicinally useful 4-aminophenol. The aqueous method developed in the present work is environmentally friendly, simple and low-cost procedure. The fabricated bimetallic porous AuPd nanostructures characterized using SEM, TEM, UV-Vis, XRD, XPS and FTIR analysis. Background: Typical hydrolysis times of saponins generally do not take into consideration the effect of time on the degradation of the target compounds, namely sapogenins. When producing natural extracts, it should be borne in mind that conducting hydrolysis to yield a target compound might also affect the final composition of the extracts in terms of other bioactive compounds. In our study, saponin-rich extracts from fenugreek, quinoa, lentil, and soybean were produced and their acid hydrolysis to give sapogenin-rich extracts was conducted over different periods (0-6 h). The disappearance of saponins and appearance of sapogenins was analyzed using high-performance liquid chromatography-diode array detection-mass spectrometry (HPLC-DAD-MS) and gas chromatography-mass spectrometry (GC-MS), respectively. The impact of hydrolysis on the phytosterols and tocopherol in the extracts was also evaluated., Results: Fenugreek showed the highest saponin content (169 g kg -1 ), followed by lentil (20 g kg -1 ), quinoa (15 g kg -1 ), and soybean (13 g kg -1 ). Hydrolysis for 1 h caused the complete disappearance of saponins and the greatest release of sapogenins. Hydrolyzed fenugreek and quinoa extracts contained the highest amounts of sapogenins and minor fractions of phytosterols and tocopherol. Hydrolyzed extracts of lentil and soybean contained a major fraction of phytosterols and a low fraction of sapogenins. In all cases, sapogenins decreased after 1 h of hydrolysis, phytosterols slightly decreased, and tocopherol was unaffected. Standards of diosgenin and oleanolic acid also showed this decreasing pattern under acid hydrolysis conditions., Conclusion: Hydrolysis times of 1 h for saponin-rich extracts from the assayed seeds guarantee the maximum transformation to sapogenin-rich extracts, along with phytosterols and tocopherol. In this study, 1% and 2% of macerated fenugreek oil was added to the feeds of rainbow trout with an average weight of 25.79 ± 1.5 g. At the end of the study, growth rate, blood parameters and NBT (Nitroblue Tetrazolium) level of rainbow trout were determined. The best feed ratio (FCR) was observed in the control group (0.77). Statistically significant differences were found only in MID values (P&lt;0.05), although there was a numerical increase in all blood parameters. There was no statistically significant difference between NBT levels (P&gt; 0.05). Although the best weight gain was in the control group as in the FCR values, the maximum elongation was measured at D1 and then at D2 (P &lt;0.05). The best survival rate was obtained with 96.66% in D1 while the worst was observed in D2 with 60% (p&lt;0,05).

The application of plant extracts for therapeutic purposes has been used in traditional medicine since the plants are a source of a great variety of chemical compounds that possess biological activity. Actually, the effect of these extracts on diseases such as cancer is being widely studied. Colorectal adenocarcinoma is one of the main causes of cancer related to death and the second most prevalent carcinoma in Western countries. The aim of this work is to study the possible effect of two fenugreek (Trigonella foenum graecum) protein hydrolysates on treatment and progression of colorectal cancer. Fenugreek proteins from seeds were hydrolysed by using two enzymes separately, which are named Purafect and Esperase, and were then tested on differentiated and undifferentiated human colonic adenocarcinoma Caco2/TC7 cells. Both hydrolysates did not affect the growth of differentiated cells, while they caused a decrease in undifferentiated cell proliferation by early apoptosis and cell cycle arrest in phase G1. This was triggered by a mitochondrial membrane permeabilization, cytochrome C release to cytoplasm, and caspase-3 activation. In addition, the hydrolysates of fenugreek proteins displayed antioxidant activity since they reduce the intracellular levels of ROS. These findings suggest that fenugreek protein hydrolysates could be used as nutraceutical molecules in colorectal cancer treatment.

Fenugreek ( Trigonella Foenum-Graecum ) seeds flavonoids (FSF) have diverse biological activities, while the antidepressant-like effect of FSF has been seldom explored. The aim of this study was to evaluate the antidepressant-like effect of FSF and to identify the potential molecular mechanisms. LC-MS/MS was used for the determination of FSF. Chronic restraint stress (CRS) was used to establish the animal model of depression. Observation of exploratory behavior in the forced swimming test (FST), tail suspension test (TST) and sucrose preference test (SPT) indicated the stress level. The serum corticosterone (CORT) level was measured. The monoamine neurotransmitters (5-HT, NE and DA) and their metabolites, as well as monoamine oxidase A (MAO-A) enzyme activity in the prefrontal cortex, hippocampus and striatum, were evaluated. The protein expression levels of KLF11, SIRT1, MAO-A were also determined by western blot analysis. The results showed that FSF treatment significantly reversed the CRS-induced behavioral abnormalities, including reduced sucrose preference and increased immobility time. FSF administration markedly restored CRS induced changes in concentrations of serum corticosterone, prefrontal cortex neurotransmitters (NE, 5-HT and DA), hippocampus neurotransmitters (NE, 5-HT and DA) and striatum neurotransmitters (NE). FSF treatment exhibited significant inhibition of MAO-A activity in the prefrontal cortex and hippocampus. FSF also significantly down-regulated the KLF11, SIRT1 and MAO-A protein expression levels in the prefrontal cortex and hippocampus. These findings indicate that FSF could exhibit an antidepressant-like effect by down-regulating the KLF11/SIRT1-MAO-A pathways, inhibiting MAO-A expression and activity, as well as up-regulating monoamine neurotransmitters levels.

Sulfated derivatives of the galactomannan family have multiple biological activities. In the present study, sulfated galactomannan from fenugreek gum (SFG) were prepared by chlorosulfuric acid/pyridine (CSA/Py) method. In order to acquire the derivatives with the highest degree of substitution (DS), the experiment optimum conditions of the sulfation were determined by Box-Behnken design (BBD). The quadratic regression model analysis confirmed that reaction time was the most significant parameter for the impact of DS. Under the selected conditions, maximum DS was obtained as 0.490. Results of FT-IR and X-ray photoelectron spectroscopy (XPS) indicated the present of SO 3 - group. In 13 C NMR spectroscopy, the original C-6 peaks did not disappear completely and the newly peaks appeared at δ 63.2 and 64.0 illustrating the incomplete substitution were predominantly on C-6 position. After sulfation, size exclusion chromatography combined with multi-angle laser light scattering (SEC-MALLS) detected all the weight average molecular mass (M W ) of sulfated derivatives decrease rapidly. The risk of pesticides on the human health and environment has drawn increasing attention. Today, new tools are developed to reduce pesticide adverse effects. This study aimed to evaluate the toxicity induced by, thiamethoxam (TMX), and the cytoprotective effect of a novel polysaccharide, named fenugreek seed water polysaccharide (FWEP) in vitro using H9c2 cardiomyoblastes and in vivo using Wistar rat model. Animals were assigned into four groups per eight rats each: group 1 served as a control group, group 2 received TMX, group 3, and group 4 received both FWEP and TMX tested at two doses (100 and 200 mg/kg, respectively). Regarding the in vitro study, our results demonstrated that TMX induced a decrease in H9c2 cell viability up to 70% with the highest concentration. In vivo, TMX injection induced marked heart damage noted by a significant increase in plasma lactate dehydrogenase, creatine phosphokinase, troponin-T, aspartate amino transferase activities, cholesterol, and triglyceride levels. Concomitant alterations in cardiac antioxidant defense system revealed depletion in the levels of glutathione and non-protein thiol and an increase in the activity of superoxide dismutase, catalase, and glutathione peroxidase. Similarly, a significant increase in heart lipid, malondialdehyde, advanced oxidation protein product and in protein carbonyls levels was also noted. In addition, heart tissues histo-architecture displayed major presence of apoptosis and necrosis as confirmed by DNA degradation. However, supplementation with FWEP alleviated heart oxidative damage and genotoxicity. In this manner, ABTS radical-scavenging activity, linoleic acid oxidation tests and heart genomic and DNA nicking assay had proved FWEP strong antioxidant potential. Aerogels are obtained by laccase/2,2,6,6-tetramethylpiperidin-1-yl)oxyl (TEMPO)-oxidation of galactomannans (GMs) from the leguminous plants fenugreek (Trigonella foenum-graecum), sesbania (Sesbania bispinosa) and guar (Cyamopsis tetragonolobus). GM oxidation in aqueous solutions causes a viscosity increase, resulting in structured and stable hydrogels. Upon lyophilization, water-insoluble aerogels are obtained, capable of water uptake up to several times their own weight. The materials derived from the three gums have been analyzed and compared. Chemical modifications have been studied by electrospray-ionization mass spectrometry (ESI-MS) and Fourier-transform infrared spectroscopy (FTIR). Polymer structure has been determined by liquid-state and semi-quantitative solid-state nuclear magnetic resonance (NMR) spectroscopy and chemical stability by incubation under variable conditions of pH, ionic strength and solvent nature. The results show that hydrogel formation is due to oxidation of primary hydroxyl groups to carbonyl and carboxyl groups and subsequent formation of hemiacetal and ester bonds. Fenugreek displays the highest stability, compared to guar and sesbania rehydrated aerogels. Progressive effects of inhalation of gasoline fumes on the liver and the protective potential with fenugreek seed supplementation were evaluated in adult male rats. Twenty-four rats were divided into four groups, unexposed control and fenugreek groups, as well as exposed groups to gasoline fumes for 6 h/6 days/week for 10 weeks, with and without supplementation of fenugreek seed powder in food (5%w/w). Exposure to gasoline fumes resulted in a significant elevation in serum alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, gamma-glutamyl transferase, total bilirubin, total cholesterol, total triglycerides, and low-density lipoprotein, along with a significant decrease in high-density lipoprotein, total protein, and albumin contents compared to the control. Meanwhile, liver oxidative stress markers, malondialdehyde, hydrogen peroxide, and nitric oxide, were highly detected with decreased antioxidants, superoxide dismutase and catalase. Also, levels of inflammatory markers, interleukin(IL)-1β and IL-6, were significantly elevated with fibrotic markers, transforming growth factor (TGFβ1), fibroblast growth factor (FGF1), and collagen I. Histopathological studies illustrated that rats exposed to gasoline fumes have degenerated hepatocytes with cellular infiltration and necrotic areas, along with remarkable deposition of collagen fibers, suggesting an incidence of liver fibrosis. Administration of fenugreek seeds, rich with antioxidant and anti-inflammatory components, during gasoline exposure showed significant amelioration through suppressing oxidative stress and inflammation.

Biomimetic nanocomposites containing hydroxyapatite and natural polymers are promising candidates for bone grafting. In this work, a tricomponent bioactive nanocomposite (nHA-CH-TFSP) assembling nano-hydroxyapatite (nHA), Chitosan (CH) and Trigonella foenum graecum seed polysaccharide (TFSP) was developed using co-precipitation method and investigated against bicomponent nHA-CH nanocomposite for bone tissue engineering. In contrast to nHA-CH nanocomposite, nHA-CH-TFSP nanocomposite exhibited rough and interconnected porous structure as revealed by SEM with the porosity (60.3 ± 0.17) lying in the range of cancellous bone. The incorporation of TFSP in nHA-CH substantially enhanced the in vitro water absorption capacity and protein adsorption ability along with appropriate biodegradation rate. Additionally, the nHA-CH-TFSP nanocomposite exhibited superior antibacterial activity. The nHA-CH-TFSP evinced a compressive strength of 6.7 ± 0.24 MPa and a compressive modulus of 100 ± 1.4 MPa, which fulfill the strength requisite of cancellous bone and could provide strong support for the growth of osteoblasts cells. Furthermore, the in vitro bioactivity study demonstrated its excellent biomineralization capacity in comparison to nHA-CH. The synthesized nHA-CH-TFSP nanocomposite exhibited better cytocompatibility towards the MG-63 cells along with its haemocompatible nature. Pyrexia occurs due to infection, malignancy and other diseases. Majority of the antipyretic drugs are synthetic in nature which exerts side effects such as gastric ulcer, hepatic necrosis and renal damage. The antipyretic potential of the hydro-alcoholic extracts of Achillea millefolium, Taraxacum officinale, Salix alba and Trigonella foenum were investigated on the yeast-induced pyrexia in albino rats. Paracetamol was used as a positive control. Rectal temperature of albino rats was verified immediately before the administration of the extracts or vehicle or paracetamol and yet again at 1-hour gap for 6 hours using a digital thermometer. The animals having pyrexia were divided into four groups Group1: Paracetamol was given to positive control. Group 2: Distilled water was given to negative control. Group 3: (250mg/kg) extract of the plant was given to rats (treatment group 1). Group 4: (500mg/kg) extracts of the plant was given to albino rats (Treatment group 2). The extracts were also phytochemically screened for alkaloids, tannins, saponins, flavonoids, cardiac glycosides and phenols. The hydro-alcoholic extracts of plants with the dose of 500mg/kg showed significant (p<0.0001) decrease in yeast-induced pyrexia, as compared with that of set drug paracetamol (150mg/kg) where the extract dose 250mg/kg was less effective than that of standard drug (p<0.05). Phytochemical screening showed the presence of alkaloids, tannins, flavonoids, saponins and phenols. This study showed that hydro-alcoholic extracts of all plants under study at a dose of 500mg/kg have significant antipyretic potential in yeast-induced elevated temperature.

Seeds and sprouts are of considerable interest due to their numerous pro-health benefits. The aim of this study was to investigate the effect of germination on the mineral composition (performed by flame absorption atomic spectroscopy), total phenolic content, antioxidant activity, as well as phenolic profiles (before and after alkaline hydrolysis by high-performance liquid chromatography) of chia, golden flax, evening primrose, phacelia and fenugreek seeds. Generally, significant (p < 0.05) changes in the individual minerals composition of the seeds, improvement of their antioxidant properties, as well as increase in levels of individual phenolic compounds was found after seeds germination. Alkaline hydrolysis allowed to release free forms of phenolics and to confirm (chromatographically) their significantly higher amounts when compared to the nonhydrolyzed fraction. Gallic, protocatechuic, caffeic, p-coumaric, ferulic and sinapic acids, as well as quercetin and kaempferol were identified in analyzed seeds and sprouts. Background: Herbal medicines long have been used in the management of diabetes mellitus (DM)., Objective: This study was conducted to ascertain if fenugreek compared with glibenclamide had any impacts on controlling blood glucose in patients with uncontrolled type II DM on conventional therapy., Methods: A total of 12 patients with uncontrolled DM and on metformin were recruited and divided into two groups. Patients in group 1 received 2 g fenugreek per day, whereas those in group 2 received glibenclamide 5 mg once daily. The impacts of fenugreek on the glycemic control and lipid profile were measured before initiation of the regimen and then after 12 weeks., Results: Only 9 of the 12 study participants completed the study. Fenugreek at 2 g/day caused an insignificant drop in fasting blood glucose (P = 0.63), but the fasting insulin level increased significantly (P = 0.04). The ratio of high- to low-density lipoprotein was significantly decreased from before to after treatment (P = 0.006). Fenugreek did not cause any notable adverse impacts on hepatic and renal functions throughout the study., Conclusion: Fenugreek could be used as adjuvant therapy to anti-diabetic drugs to control blood glucose, and further studies are needed.

Background Fenugreek seeds have shown antinociceptive effect in animal studies. This double-blind placebo controlled trial was designed to study the effect of fenugreek transdermal patch 10% (FDP) for management of inguinal hernia (IH) post-operative pain. Methods Standardized extract of fenugreek seeds was used for FDP formulation. Ninety patients treated with 10% FDP, diclofenac dermal patch 1% (DP) and placebo twice daily after IH surgery. The pain intensity score was evaluated using a visual analogue score (VAS) up to 48 h after operation. Morphine consumption and diclofenac suppository demand were evaluated too. Results The pain score was significantly reduced in FDP group in comparison with the placebo group. This effect was also significantly different from DP up to 6 h after surgery (p<0.05). Morphine consumption and diclofenc suppository demand were significantly decreased in FDP group (p<0.05). Conclusion In all, results of the present study indicated that FDP decreases pain score and demand for morphine in post-surgery patients in comparison to diclofenac patch, and this preparation could be a suit option as a natural antinociceptive agent for pain management.

The total steroidal saponins, particularly its major steroidal sapogenin (diosgenin), are the main active principles of fenugreek seed extract. In this study, an ethanol-salt aqueous two-phase system (ATPS) was explored for the purification of the total steroidal saponins, and the process conditions were optimized by response surface methodology (RSM). Under the optimized conditions, the RSM predicted recovery of the total steroidal saponins in the top phase of ATPS was 97.9%, which agreed with the average experimental recovery (98.3 ± 4.2% ( n = 6)). Moreover, a rapid micellar electrokinetic chromatography (MEKC) method was developed for the determination of diosgenin from extracts. The diosgenin content in the ATPS top phase extract was 3-fold higher than that in crude extract, suggesting this ATPS having a great potential for purification pharmacological active ingredients from fenugreek seeds.

Trigonella foenum graecum L. is a dietary herb used in traditional medicine system. In this study, we investigated the cytotoxicity, antitumor, antimetastatic and antiangiogenic effect of the steroidal compound, ethyl iso-allocholate isolated from T. foenum graecum L. seeds against A549 lung cancer cells in vitro and in vivo. Among all the isolated compounds, the ethyl iso-allocholate rendered the highest cytotoxicity potential. It showed least percentage cell viability in trypan blue assay and lowest nuclei count in hoechst staining. The caspase glo assay and western blot analysis showed the significant caspase 3 cleavage, indicating caspase dependent apoptosis. Consistent with the in vitro data, ethyl iso-allocholate showed highest percentage tumor growth inhibition i.e. 80 ± 5% in zebrafish, equivalent to doxorubicin. It significantly reduced angiogenesis to 5 ± 0.8% (**P < 0.01), compared to negative control group which was 60 ± 2%. The ethyl iso-allocholate showed 55 ± 3% inhibition in liver metastasis. To investigate the safety of the compounds on normal tissues, the percentage mortality was examined. The ethyl iso-allocholate showed zero percent mortality of zebrafish. These results indicate that the steroidal derivative isolated from T. foenum-graecum seeds induces caspase dependent apoptosis in cancer cells and reduces tumor growth, metastasis and angiogenesis in vivo, as well as it is safe on the normal tissues. The in vitro and in vivo anticancer studies suggest that the cytotoxic compound ethyl iso-allocholate has potential application in pharmaceutical industry., (Copyright © 2018. Ethanol extracts (EE) from fenugreek and quinoa seeds with different total content of inhibitory compounds (TIC, total saponin plus phenolic) were prepared with and without concentration of TIC (CEE -concentrated EE-, and EE, respectively). Their inhibitory activity on pancreatic lipase and α-amylase was assessed by traditional in vitro methods (with or without orbital shaking), and by simulating intestinal digestion. CEE contained higher contents of TIC than EE, being fenugreek superior to quinoa (p < 0.001). The extracts inhibited enzymes in a dose-dependent manner, CEE extracts being stronger (fenugreek for lipase -p = 0.009-, and quinoa for α-amylase -p < 0.001-). Shaking did not impact the activity. Intestinal conditions worsened the inhibition of lipase, but slightly catalyzed the α-amylase. Longer times of reaction worsened activities. The importance of assessing the inhibitory activity of extracts under simulated intestinal conditions is concluded, being fenugreek more interesting than quinoa, especially against pancreatic lipase., (Copyright © 2018 Elsevier Ltd. Ajowan, thyme and fenugreek are spice and aromatic crops with a number of medicinal properties which are known as important sources of essential phytochemicals. The purpose of this study was to investigate the antioxidant capacity, total phenolic content and antifungal activities of these plant extracts on growth of Fusarium solani, an important plant pathogen, soil saprophyte and one of the causal agents of fusariosis in human and animals. Their total antioxidant activity was measured using DPPH radical scavenging assay and their antimicrobial activity was determined through poison food assay at two concentrations (1000 and 1500 ppm) and spore germination assay in vitro. All methanolic extracts showed high antioxidant activity which among them methanolic extract of thyme demonstrated higher antioxidant potential with a low IC50 (16.50 mg ascorbic acid/g). Also, the highest phenolic content (70.55 mg GAE g-1) was observed in methanolic extract of thyme. The highest and lowest amount of thymol was determined in methanolic extract of thyme and aqueous extract of ajowan. Methanolic extracts of thyme leaves and ajowan seeds at concentration of 1500 ppm were potentially effective against F. solani over the control treatments by 90.33% and 85.73%, respectively. Followed by hydro-ethanolic and aqueous extracts exhibited a lesser percentage of inhibition. The MIC value for methanolic extract of thyme and ajowan was 3.75 mg/ml followed by hydro-ethanolic and aqueous extracts, respectively. The amount of calculated MFC was ranging from 7.5 to 30 mg/ml for thyme methanolic and fenugreek aqueous extracts, respectively.

Fenugreek gum (FG), guar gum (GG), and locust bean gum (LBG) dispersions were studied for their flow behaviour, intrinsic viscosity, viscoelasticity and the effect of environmental stress on their physicochemical properties. Shear thinning behaviour was observed in all three gum dispersions prepared at concentrations > 0.25 wt%. The impact of the change in concentration (0.125 to 2 wt%) on dispersion flow behaviour and consistency coefficient were determined using power law model. Both GG and LBG dispersions have a higher intrinsic viscosities (16.93 ± 0.02 dL/g and 15.20 ± 0.02 dL/g) compared to FG (13.46 ± 0.02 dL/g), but FG has higher average viscosity molecular weight (3.10 ± 0.01) compared to GG and LBG. This could impart higher apparent viscosity to FG dispersions compared to GG and LBG. All gum dispersions above 0.1 wt% concentration showed entanglement among the polymer chains. Variation in pH (3-7) did not significantly affect viscosity of the dispersions while it was significantly decreased with the addition of salt. The FG had superior gelling properties compared to GG, and LBG at 1 wt% concentration which could have potential application in certain food and non-food products., (Copyright © 2018 Elsevier B.V. Objectives: The growing rate of neurological disorders is a major concern in today's scenario. Today's research is focusing on therapeutic interventions providing benefits in these disorders. Presently, drugs of natural origin have gained more interest for the treatment of central nervous system disorders for their efficacy and less/ no side effects. This review is emphasizing the cited roles of Trigonella foenum graecum (fenugreek) and its constituents in different neurological manifestations., Method: A review of the literature, relevant to the role of fenugreek and its major constituents including saponins and alkaloids in different neurological aspects and in delineating the health benefits, was conducted., Results: The cited research acknowledged that fenugreek and its constituents exert positive influence on neurological health. Few studies have reported the beneficial role of fenugreek and its constituents like trigonelline in pathological symptoms of Alzheimer's disease. Similarly, other studies evidenced the neuroprotective, antidepressant, antianxiety as well as modulatory effect on cognitive functions and Parkinson's disease., Discussion: Large populations are the sufferers of the neurological disorders, pointing the need for investigation of such therapeutic interventions which target and delay the underlying pathological hallmarks and exert positive influence on different neurological health problems. Hypolipidemic, hypoglycemic, antioxidant, and immunomodulatory effects of fenugreek and its constituents with their potential role in various neurological disorders were already reported. In future, it would be of even greater interest to further develop more effective dosage, supplementation period, and to evaluate the therapeutic potentials of fenugreek and its constituents in neurological disorders by exploring underlying cellular and molecular mechanisms.

Background: Diosgenin is a very important plant secondary metabolite and raw material for the drug indus- try. Plant sources rich in diosgenin include yam (Dioscorea spp.) and fenugreek (Trigonella foenum-graecum L.). A method for diosgenin extraction from yam extracts has previously been validated, but its extraction from fenugreek plants still requires validation. In addition, all available methods require time-consuming additional purification steps. The present study was aimed at developing a low cost, less time-consuming single-step method for diosgenin extraction from fenugreek., Methods: This study represents a method developed for diosgenin extraction from fenugreek plants without any additional/supportive purification methods such as chromatography or thin-layer chroma- tography. Diosgenin yield estimation and purity analysis by HPLC method, along with accuracy and preci- sion analysis, is presented., Results: Five different fenugreek varieties were subjected to a newly developed diosgenin extraction method, and an HPLC chromatogram showed a single peak corresponding to diosgenin. Yield was determined by the standard curve method. Limit of detection (LOD) and limit of quantification (LOQ) for the assay were found to be 0.0312 and 0.102 &mu;g, respectively; tcalculated for slope and other statistical parameters were found to be significant (P value < 0.001) for this method., Conclusions: We have developed a fast, accurate and low cost method for diosgenin extraction from fenu- greek. Although the authors have studied this method only in fenugreek plants, it could be applied to the extraction of a few other plant secondary metabolites, which will help researchers to save time and effort.

Rheumatoid arthritis is a chronic inflammatory joint disease characterized by synovial proliferation and tissue destruction. Pro-inflammatory cytokines like interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α) play a key role in the disease process and elevate energy expenditure, which further increases the joint pain and stiffness. The present study was undertaken to explore the anti-arthritic potential of fenugreek mucilage in adjuvant induced arthritic rats. In the present study, paw volume was measured on the 7 th , 14 th and 21 st day. Finally, animals were anaesthetized; blood samples and tissues were collected for the assay of inflammatory enzymes like cyclooxygenase, lipoxygenase; evaluated the level of cytokines like IL-6, TNF-α, arthritic index and rheumatoid factor. Fenugreek mucilage exhibited maximum percentage of edema inhibition at a dose of 75 mg/kg on 21st day of adjuvant arthritis. The effect was higher than that of standard drug, indomethacin. The activities of inflammatory enzymes and concentration of mediators were decreased on treatment with fenugreek mucilage. Cytology of synovial fluid showed mild inflammation with normal synoviocytes (mesothelial cells) tried to bring back to normal characteristics on supplementation with fenugreek mucilage. Based on the observations, it can be suggested that fenugreek mucilage possesses promising anti-arthritic property and it can be used as a therapeutic agent for arthritis., (Copyright © 2018 Elsevier Inc. Cadmium (Cd), an economically valuable metal, is widely used in various industrial processes. Although it is of economic value, it is hazardous to human health. Cd accumulates in vital organs where it causes various diseases. Natural compounds with chelating or antioxidant properties have been tested to reduce the toxic effect of Cd. The anti‑oxidant, anti‑diabetic and hypocholesterolemic properties of fenugreek (Trigonella foenum-graecum) leaves make it a candidate for investigation as protective agent against Cd‑induced toxicity. In the present study, the protective effects of fenugreek leaf extract (FLE) on cell viability, morphology, and whole genomic transcription in cadmium chloride (CdCl2)‑treated rat liver cells were analyzed. The cells were treated with 25 µM CdCl2 alone, or co‑treated with 5 µg/ml FLE for 48 h. The co‑treated cells were pretreated with FLE for 2 or 4 h, followed by CdCl2 treatment. Genomic transcription analysis was performed in the CdCl2‑treated cells following treatment for 6 h. The CdCl2 caused a significant decrease in viability (35.8±4.1%) and morphological distortion of the cells, compared with the untreated control cells; whereas 4 h pretreatment with FLE (5 µg/ml) reversed the Cd‑induced morphology alteration and increased the cell viability to 102±3.8%. Genomic transcription analysis of the CdCl2 only‑treated cells showed 61 upregulated and 124 downregulated genes, compared with 180 upregulated and 162 downregulated genes in the FLE pretreated cells. Furthermore, 37 and 26% of the affected total genomic genes in the CdCl2 only‑treated cells were involved in binding and catalytic activities, respectively, whereas 50 and 20% of the genes in the FLE pretreated cells were involved in binding and catalytic activities, respectively. In conclusion, these results suggested that genome transcriptome modulation may be important in the protective effect of FLE against Cd‑induced toxicity in normal rat liver cells.

The rheology of carboxymethyl fenugreek gum (CFG) is characterized and applied it as a binder for the silicon anode of lithium-ion batteries (LIBs). First, a series of carboxymethyl fenugreek gum (namely CFG-1, CFG-2 and CFG-3) was prepared and characterized by Fourier transform infrared spectrophotometer and hydrogen nuclear magnetic resonance spectrometry studies. In rheological measurements, the CFG solutions show higher flowability, weaker viscoelastic behavior and better thixotropy than that of fenugreek gum (FG) solution. Furthermore, the higher degrees of substitution (DS) of CFG solutions lead to lower apparent viscosity and higher flow index, weaker elastic effect and smaller relaxation time, and higher viscosity recoverability than those obtained for lower DS of CFG solutions. In electrochemical measurements, the polar hydroxyl and carboxyl groups in the CFG skeleton chain provide bonding points between the CFG binders and the silicon electrode to produce binder-silicon bonds, which cause strong hydrogen bonding, and resulted in excellent electrochemical stability and specific capacity. The electrochemical performances were measured via the silicon electrodes with FG and CFG at 5 wt% content. As the DS is increased, the initial Coulombic efficiency can increase from 88.74% to 91.3%. When the electrode undergoes a volume change, the CFG binder with silicon electrodes can hold high capacity of above 1500 mAh/g after 200 charge-discharge cycles. Therefore, the carboxymethyl fenugreek gum as a binder may also be a novel extended binder-design, with applications for silicon anodes of LIBs., (Copyright © 2018 Elsevier B.V. Ethanolic extracts of green tea leaves (GTE) and fenugreek seeds (FSE) were screened for their antibacterial activity against four food pathogenic strains using disc diffusion method. The two extracts revealed antimicrobial activity against selected bacterial strains. GTE showed the highest antibacterial activity to Escherichia coli and Staphylococcus aureus at a concentration of 1% with inhibition zone equal to 29.45 ± 0.64 mm and 25.68 ± 1.2 mm, respectively. In addition, the effect of GTE and FSE combined with chitosan coating on the shelf life of Pacific white shrimp (PWS) (Litopenaeus vannamei) during refrigerated storage have been studied. Our results indicated that using GTE or FSE during the refrigerated storage of PWS led to significantly decreased Total Volatile Bases Nitrogen, Thiobarbituric acid reacting substances, total bacterial count, and pH. The sensory properties of PWS have improved considerably in the samples treated GTE or FSE. These findings suggested that the application of chitosan coating combined with GTE or FSE to PWS is advisable to achieve better quality during refrigerated storage.

The efficient production of saponin-rich extracts is of increasing interest due to the bioactive properties that have being demonstrated for these compounds. However, saponins have a poor bioavailability. In this respect, the knowledge about the bioaccessibility of saponins as a first step before bioavailability has been scarcely explored. In this study, the production of ultrasound-assisted extracts of saponins from edible seeds (quinoa, soybean, red lentil, fenugreek and lupin) was carried out with ethanol, ethanol:water or water. Extraction yield, total saponin (TSC), fat and total phenolics content (TPC) were determined. Then, the bioaccessibility of saponins after the in vitro gastrointestinal digestion of the extracts was determined and the effect of TPC and fat in the extracts on bioaccessibility was evaluated. The highest saponin-rich extracts were obtained by ethanol, being fenugreek and red lentil the richest extracts (12% and 10%, respectively). Saponins from ethanol:water extracts displayed variable bioaccessibility (from 13% for fenugreek to 83% for lentil), but a bioaccessibility closer to 100% was reached for all ethanol extracts. Correlation studies showed that TPC of the extracts negatively affected the bioaccessibility of saponins, whereas fat of the extracts enhanced this parameter. As summary, ultrasound-assisted extraction is shown as an efficient method for obtaining saponin-rich extracts from edible seeds, being ethanol the most advantageous solvent due to the richness of saponins and the successful bioaccessibility from these extracts, likely caused by the co-extracted fat with ethanol. Regardless of the extracts, phenolic compounds or fat may hinder or enhance the bioaccessibility of saponins, respectively.
