Tropospheric ozone (O3) pollution has been becoming prominent in North China Plain (NCP) in China since last decade. In order to clarify the source contribution and formation mechanism of O3, the critical precursors of volatile organic compounds (VOCs) were measured with both on-line and off-line methods in Luoyang City in summer of 2019. The concentrations of nitrogen oxides (NOx, sum of NO and NO2) and O3 were simultaneously monitored. Fifty-seven VOCs measured in U.S. Photochemical Assessment Monitoring Station (PAMS) showed daily concentrations in a range of 14.5 ± 5.33 to 29.2 ± 11.2 ppbv in Luoyang, which were comparable with those in other Chinese megacities. In China, owing to the methane concentration being below 0.75%, the coal ventilation air methane (CVAM) is usually emitted directly into the atmosphere, rather than utilized, which not only causes huge waste of energy but also exerts potential hazards to the greenhouse effect. It is important and practicable to save costs of development and investment by simulating enrichment and separation of CVAM with an aim to improve the efficiency and recovery of adsorption separation. Above all, it will have important practical significance to the development of adsorption separation. In this paper, the experiment of the pressure swing adsorption process was carried out on double towers built by our laboratory, and the Aspen Adsorption was used to simulate the process. The effect of the operation parameters on the desorbed methane concentration was studied by altering the feed concentration, the adsorbed pressure, and adsorbed and desorbed time. The results of simulation and experiment are basically consistent. The ratio of methane was decreased following the increasing concentration of the feed. The optimum adsorption pressure and time were found to be 210 kPa and 120 s, respectively. The optimum desorption times of experiment and simulation were 150 s and 120 s, respectively. Because there was a man-made 30 s time lag between the experiment and simulation to protect the vacuum pump, the results show that the simulation and experiment were matched well. Therefore, we can make use of Aspen Adsorption to design separation and enrichment of CVAM, providing theoretical and practical guidance for the gas separation and saving resources and energy.

Here we used a meta-analysis of several clinical trials to determine whether anti-Helicobacter pylori therapy has any positive effect on IBS patients. Here we compared the effective clinical remission rates between IBS patients treated with anti-H. pylori therapy and those who were not. This data would provide more clinical evidence regarding the efficacy of novel treatments and intervention points for IBS patients. Relevant studies were identified using keyword searches on various electronic databases, including PubMed, Embase, the Cochrane Central Register of Controlled Trials, CNKI, and CBM. Keywords included “helicobacter pylori” and “irritable bowel syndrome” among others. The literature was screened using relatively strict inclusion and exclusion criteria and RevMan 5.3.5 and Stata 15.1 software were used for meta-analysis and to assess publication bias and sensitivity. A total of ten studies met all of the inclusion criteria; these included 655 IBS patients with H. pylori infection, of these, 385 patients were in the experimental group and 270 patients were in the control group. Vibrio parahaemolyticus is the main pathogen of food-borne diarrheal in coastal areas. Through the study of pathogen characteristics of 1870 V. parahaemolyticus, the isolation rate of O4:KUT had increased significantly since 2013. In this study, we analyzed virulence, antimicrobial susceptibility, molecular, and epidemiological characteristics of a new serotype named O4:KUT2. O4:KUT2 strains had tlh+tdh+trh–toxRS/new– characteristics and were prevalent during 2013–2015. The 91.5% O4:KUT2 serotype strains were resistant to ampicillin. The growth curves of O4:KUT2 strains were different with O4:K9, O4:K8, and O3:K6 serotype strains. O4:KUT2 strains belonged to ST332 where four strains had a large fragment inserted at recA. Compared the whole genomes of O4:KUT2 strains with O4:K9 strain which also belonged to ST322 isolated from acute diarrhea patients in Zhejiang province in 2012, no different alleles at 2249 loci was found. This finding implied that O4:KUT2 strains might be derived from O4:K9 strains. Clinical presentation of patients positive for V. parahaemolyticus O4:KUT2 were no significant difference with patients positive for O3:K6, although their genetic characteristics were different. The appearance and the increase of proportion of the new serotype O4:KUT2 strains was aware that we should not relax the monitoring of the pathogen spectrum of acute diarrheal patients.

We present ADIOS 2, the latest version of the Adaptable Input Output (I/O) System. ADIOS 2 addresses scientific data management needs ranging from scalable I/O in supercomputers, to data analysis in personal computer and cloud systems. Version 2 introduces a unified application programming interface (API) that enables seamless data movement through files, wide-area-networks, and direct memory access, as well as high-level APIs for data analysis. The internal architecture provides a set of reusable and extendable components for managing data presentation and transport mechanisms for new applications. ADIOS 2 bindings are available in C++11, C, Fortran, Python, and Matlab and are currently used across different scientific communities. ADIOS 2 provides a communal framework to tackle data management challenges as we approach the exascale era of supercomputing.

Purpose: We found in previous study that metformin could treat sepsis myocarditis in a mouse model. We employed the zebrafish model organism to investigate the effect of metformin on sepsis myocarditis. Methods and Results: Wild-type zebrafish was used to establish a sepsis myocarditis model and combined with image software analysis and cytokine detection, the protective dose of metformin was determined. The results showed that immersion with Escherichia coli could cause 75% mortality in zebrafish and make larvae appear as characteristics of severe sepsis myocarditis. Pretreatment with 10 mM metformin for 3 hours could effectively reduce heart congestion and swelling in zebrafish with sepsis myocarditis and increased the heart rate. It could reduce the mortality and prolong the survival time of zebrafish with sepsis myocarditis; Tg(mpx: EGFP) transgenic zebrafish were adopted to explore the number of neutrophils in zebrafish heart before and after metformin protection, and metformin could maintain the number of neutrophils in zebrafish heart; quantitative real-time reverse transcription–polymerase chain reaction showed that metformin could reduce the expression of pro-inflammatory factors, tumor necrosis factor-α and interleukin (IL)-6, and could promote the anti-inflammatory factor, transforming growth factor-β and IL-10 expression. Conclusion: We established a zebrafish sepsis myocarditis model and applied metformin in advance to provide a protective effect on the zebrafish heart.

Purpose: Interferon regulatory factor 4 (IRF4) is identified as a transcriptional factor and plays an important role in the immune response in mammals; however, there are few reports about the function of zebrafish IRF4. Methods: We first amplified the coding sequence of irf4a from the testis of zebrafish. Besides, the fragments of irf4a , P2A , EGFP , and Tol2 vector were added for homologous recombination. By sequencing, we can get the Tol2-ef1α-irf4a-EGFP recombinant plasmid and it was microinjected into zebrafish embryos. Fluorescence observation was proceeded at days 3 post fertilization; F0 generations expressing green fluorescence in multiple tissues throughout the body were screened as the founder and raised them to sexual maturity. After mating with WT zebrafish to generate F1 offspring, polymerase chain reaction was used to identify whether irf4a was integrated into the zebrafish genome. Conclusion: We obtained the systematic overexpressed irf4a transgenic zebrafish with green fluorescence labeled in spine, eyes, heart, brain, and other tissues. The transgenic zebrafish will be used as a tool for the role of IRF4a in the immune response to the inflammation preconditioning in the future study.

Abstract Background Lipotoxicity contributes to diabetic myocardial disease. In this study, we investigated the lipid species contributing to lipotoxicity and the relationship with peroxisomal β-oxidation in the heart of diabetic mice. Methods Male C57BL/6 mice were randomly divided into a Diabetic group (intraperitoneal injection of STZ) and a Control group (saline). Cardiac function indexes [ejection fraction (EF%) and fractional shortening (FS%)] were evaluated by echocardiography. Morphological changes in the myocardial tissues and mitochondria were assessed by electron microscopy following hematoxylin and eosin staining. Blood myocardial injury indexes and lipids were measured using an automatic biochemical analyzer. Cardiac ATP levels were analyzed using a commercially available kit. mRNA levels of glucose transporter 4 (GLUT4), fatty acid binding protein 3 (FABP3), palmitoyl transferase 1α (CPT-1α), acyl-CoA oxidase 1 (AOX1), D-bifunctional protein (DBP), 3-ketoacyl-CoA thiolase A (THLA), uncoupling protein (UCP) 2 and UCP3 were investigated by quantitative reverse-transcription polymerase chain reaction. FABP3 protein expression was analyzed by Western blotting. Non-targeted metabolomics by LC-MS/MS was applied to evaluate profile of lipid metabolism in heart. Results Compared with controls, EF% and FS% were significantly reduced in diabetic mice. Furthermore, blood myocardial injury indexes and lipids, as well as myocardial mitochondrial cristae fusion were significantly increased. In the diabetic heart, GLUT4 expression was decreased, while expression of FABP3, CPT-1α, AOX1, DBP, THLA, UCP2 and UCP3 was increased, and ATP levels were reduced. Background: The population structure of human diarrheagenic Escherichia coli (DEC) isolates derived from worldwide collections remains undefined. Methods: A total of 1196 clinical isolates were obtained from a multilocus sequence typing (MLST) database. Genetic diversity analysis, MLST analysis, and phylogenetic analysis combined with different pathotypes were performed through a variety of calculation software applications. Results: All isolates were categorized as one of 579 different sequence types (STs). The eBURST algorithm resolved these 579 STs into 27 clonal complexes (CCs), 37 concatemers, and 210 singletons, revealing a high level of genetic diversity in the population structure of DEC. CC10 was the most prevalent CC, comprising 276 (23.08%, 276/1196) isolates with 85 (14.68%, 85/579) STs widely distributed in 20 countries. The population structure of five common pathotypes was highly diversified, and isolates with the same ST or CC were heterogeneous for different pathotypes. Sequence variations were more abundant in fumC and gyrB than in the other five genes, and these exhibited the highest degree of nucleotide diversity (0.03886 and 0.03075, respectively) and the greatest number of polymorphic nucleotide sites (137 and 139, respectively). The dN/dS ratios of seven analyzed loci varied from 0.0083 (recA) to 0.0434 (purA), and the ratio for the concatenated sequence was 0.2518, revealing the effects of purifying selection on housekeeping genes during the evolutionary process. Alsophila latebrosa is a common and widespread tree fern of Cyatheaceae. Its complete chloroplast genome is first assembled and reported with 155,724 bp in length, including a large single copy (LSC) region of 85,800 bp, a small single copy (SSC) region of 21,620 bp, and a pair of inverted repeats (IRs) of 24,152 bp. The genome has 133 genes, including 89 protein-coding genes, 33 tRNA genes, eight rRNA genes and three pseudogenes. Maximum likelihood approach was employed to construct the phylogenetic relationship among ten ferns including A. latebrosa. The result showed that A. latebrosa was most related to A. costularis as a sister group with 100% bootstrap support. The complete chloroplast genome sequences of A. latebrosa will provide valuable genomic information to further illuminate phylogenetic classification of Cyatheacea.

It is of great significance for air pollution control and personnel safety guarantee to master the release characteristics of harmful gases in the process of Limnoperna fortunei corruption. In view of the lack of research on the environmental pollution caused by the corruption of Limnoperna fortunei, a model experiment was designed to study the three harmful gases of NH3, H2S, and CH4 in the putrid process of Limnoperna fortunei by considering the density of Limnoperna fortunei and the time of leaving water. The results show that: (1) The recognition and processing of outliers based on wavelet decomposition and K-means algorithm can effectively reduce the standard deviation and coefficient of variation of the data set and improve the accuracy of the data set. (2) The variation of NH3 and H2S gas concentrations with the time of water separation satisfies polynomial linear regression (R2 > 99%). (3) At a density of 0.5–7.0 × 104 mussels/m2, the highest concentration of NH3 reached 47.9777–307.9454 mg/m3 with the increase in the density of Limnoperna fortunei and the extension of the time away from water, far exceeding the occupational exposure limit of NH3 of 30 mg/m3, potentially threatening human health and safety. The highest detection value of H2S concentration is 0.1909–5.0946 mg/m3, and the highest detection concentration of CH4 is 0.02%, both of which can be ignored.

Abstract Background Surfactants have attracted increasing interest for their capability to improve the enzymatic hydrolysis of lignocellulosic biomass. Compared to chemical surfactants, biosurfactants have a broader prospect for industrial applications because they are more environmentally friendly and more effective in some researches. Commercial cellulase preparations are mainly composed of endoglucanases (EGs) and cellobiohydrolases (CBHs) that possess carbohydrate-binding modules (CBMs). However, the effects of lipopeptide-type biosurfactants on enzymatic saccharification of lignocellulose and adsorption behaviors of cellulases with CBMs remain unclear. Results In this study, we found that Bacillus sp. W112 could produce a lipopeptide-type biosurfactant from untreated biomass, such as wheat bran and Jerusalem artichoke tuber. The lipopeptide could enhance the enzymatic hydrolysis of dilute acid pretreated Giant Juncao grass (DA-GJG) by fungal and bacterial enzymes. The enhancement increased over a range of temperatures from 30 to 50 °C. Lipopeptide was shown to be more effective in promoting DA-GJG saccharification than chemical surfactants at low dosages, with a best stimulatory degree of 20.8% at 2% loading of the substrates (w/w). Lipopeptide increased the thermostability of EG and CBH in commercial cellulase cocktails. Moreover, the dual effects of lipopeptide on the adsorption behaviors of cellulases were found. It specifically lowered the non-productive binding of cellulases to lignin and increased the binding of cellulases to cellulose. In addition, we investigated the influence of lipopeptide on the adsorption behaviors of CBHs with CBMs for the first time. Our results showed that lipopeptide reduced the adsorption of CBM-deleted CBH to DA-GJG to a greater extent than that of intact CBH while the non-productive binding of intact CBH to lignin was reduced more, indicating that lipopeptide decreased the binding of CBMs onto lignin but not their combination with cellulose. Conclusions In this study, we found that lipopeptide from Bacillus sp. W112 promoted the enzymatic hydrolysis of DA-GJG at relative low loadings. The stimulatory effect could be attributed to increasing the cellulase thermostability, reducing non-productive adsorption of cellulases with CBMs caused by lignin and enhancing the binding of cellulases to cellulose.

Abstract Background Xylan, the major constituent of hemicellulose, is composed of β-(1,4)-linked xylopyranosyl units that for the backbone, with side chains formed by other chemical moieties such as arabinose, galactose, mannose, ferulic acid and acetyl groups. Acetyl xylan esterases and α-l-arabinofuranosidases are two important accessory enzymes that remove side chain residues from xylan backbones and may act in synergy with other xylanolytic enzymes. Compared with enzymes possessing a single catalytic activity, multifunctional enzymes can achieve lignocellulosic biomass hydrolysis using a less complex mixture of enzymes. Results Here, we cloned an acetyl xylan esterase (PcAxe) from Penicillium chrysogenum P33 and expressed it in Pichia pastoris GS115. The optimal pH and temperature of the recombinant PcAxe (rPcAxe) for 4-nitrophenyl acetate were 7.0 and 40 °C, respectively. rPcAxe is stable across a broad pH range, retaining 100% enzyme activity om pH 6–9 after a 1 h incubation. The enzyme tolerates the presence of a wide range of metal ions. Sequence alignment revealed a GH62 domain exhibiting α-l-arabinofuranosidase activity with pH and temperature optima of pH 7.0 and 50 °C, in addition to the expected esterase domain. rPcAxe displayed significant synergy with a recombinant xylanase, with a degree of synergy of 1.35 for the hydrolysis of delignified corn stover. Release of glucose was increased by 51% from delignified corn stover when 2 mg of a commercial cellulase was replaced by an equivalent amount of rPcAxe, indicating superior hydrolytic efficiency. Conclusions The novel bifunctional enzyme PcAxe was identified in P. chrysogenum P33. rPcAxe includes a carbohydrate esterase domain and a glycosyl hydrolase family 62 domain. This is the first detailed report on a novel bifunctional enzyme possessing acetyl xylan esterase and α-l-arabinofuranosidase activities. These findings expand our current knowledge of glycoside hydrolases and pave the way for the discovery of similar novel enzymes.

In typical median and small aeroengines, the air used to realize the functions such as cooling of turbine blades and disks, sealing of turbine cavities and bearing chambers, adjusting of rotating assembly axial load is normally drawn through the rear cavity of centrifugal impeller, so the thorough understanding of flow characteristics and pressure distribution and the proposal of the corresponding control methods in the cavity are the key to design the rational secondary air system. With an impeller rear cavity in a small turbofan engine as an object, the current study was dedicated to the investigation of flow control methods in the cavity. Two methods, namely, baffle and swirl-controlled orifice, were proposed to regulate the pressure loss and distribution in the cavity. Furthermore, the influence of geometry parameters of the two methods such as the length of baffle, the space between the baffle and rotating disk wall, the orientation, and radial position of swirl-controlled orifice was investigated. The CFD results show that the swirl-controlled orifice which could deswirl the flow is more effective in regulating the pressure loss and its distribution in cavity than baffle. The variation of the radial position of the swirl-controlled orifice had little influence on pressure loss but obvious influence on pressure distribution; therefore, decreasing the radial position could reduce the axial load on the rotating disk without changing the outlet pressure.

In-depth analysis of the influence of mixed working fluids on the thermodynamic performance of a throttling refrigeration cycle through experimental research. After optimizing the calculation of two ternary mixed working fluids with different compositions, the charging test is carried out, and the throttle characteristic curve of the mixed working fluid the law of temperature and pressure changes in the experiment was analyzed. The results show that: a mixed working fluid with a mixing ratio of 30.39mol% R14, 10.73mol% R170 and 58.87mol% R600A, at a pressure ratio of 2.5/0.1, a low temperature of -67.7°C is obtained, and the cooling time is 113.42min. The refrigeration system COP is 0.076; the mixing ratio is 30.39mol%R14, 15.73mol%R23 and 53.87mol%R600A mixed working fluid, in the case of a pressure ratio of 2.14/0.1, a low temperature of -76.4°C is obtained, and the cooling time is 160min, the refrigeration system COP is 0.106.
