Abstract

The effect of mercury on the room temperature, high cycle fatigue properties of three alloys: Cu-5.5 pct Al, Cu-7.3 pct Al, and Cu-6.3 pct Al-2.5 pct Fe has been determined. Severe embrittlement under cyclic loading in mercury is associated with rapid crack propagation in the presence of the liquid metal. A pronounced grain size effect is noted under mercury, while fatigue properties in air are insensitive to grain size. The fatigue results are discussed in relation to theories of adsorption-induced liquid metal embrittlement.

Abstract

Abstract 3049 Poster Board II-1025 Colocalization and assembly of blood coagulation factors in the presence of negatively charged phospholipids leads to a 1,000-fold increase in the rate of thrombin generation compared to the solution reaction. We have established prothrombin fragment 1, the region of prothrombin containing the γ-carboxy-glutamic acid domain, as a probe for anionic phospholipids including phosphatidylserine. Prothrombin fragment 1 binds with high affinity to phosphatidylserine-containing membranes in vitro and identifies phosphatidylserine exposure relevant for the site of assembly of coagulation complexes in vivo. In order to determine the effect of phosphatidylserine exposure on thrombus formation during the laser injury model in vivo, we treated mice orally with the Cu2+ chelator tetrathiomolybdate for one week prior to study. This treatment has been shown to suppress phosphatidylserine exposure in rats (PNAS, 100: 6700-05, 2003). After copper chelator treatment, normal partial thromboplastin times (39 sec vs 42 sec, p=0.5) and whole blood counts in treated versus untreated mice were similar, suggesting that copper chelation did not affect the function of coagulation factors or total blood cell counts. Annexin V and Prothrombin fragment 1 were also used to measure phosphatidylserine exposure after thrombin (1 U/ml) stimulation of washed platelets using flow cytometric analysis. Platelets from untreated mice exhibited 2-fold increase in binding of both Annexin V and Prothrombin fragment 1 after thrombin stimulation; these values are similar to those previously reported. In contrast, the platelets of treated mice did not expose phosphatidylserine upon thrombin stimulation. Treatment with copper chelator did not affect platelet degranulation, as determined by surface exposure of P-selectin in flow cytometry. In addition, total phospholipid content and the ratio of outer to inner membrane phospholipids was not affected by treatment with copper chelator, suggesting that any reduction in detection of phosphatidylserine was due to reduction in exposure on the cell surface in response to an appropriate stimulus rather than reduced biosynthesis. Fluorescently conjugated Prothrombin fragment 1 or fluorescently conjugated antibody directed against phosphatidylserine were used as probes to follow the kinetics of phosphatidylserine exposure after the laser injury of cremaster muscle arterioles of a living mouse using high speed fluorescence intravital microscopy. Endogenous platelets were labeled with a fluorescently conjugated Fab fragment of an anti-CD41 antibody and fibrin deposition was measured using a fluorescently conjugated antibody that recognizes fibrin but not fibrinogen. We observed a 42% reduction (median of 18 thrombi, p=0.02) in Prothrombin fragment 1 binding and a 60% reduction (median of 27 thrombi, p=0.0002) in anti-phosphatidylserine binding after laser injury compared to untreated animals (n=58 thrombi). The accumulation of platelets during thrombus formation was not affected by the treatment when compared to untreated mice (p=0.4). On the other hand, fibrin deposition was reduced by 64% in treated mice (median of 38 thrombi, p=0.001) when compared to untreated animals (39 of thrombi). These data suggest that suppression of phosphatidylserine exposure reduces assembly of coagulation complexes resulting in a suboptimal concentration of thrombin for full fibrin generation but sufficient thrombin to activate platelets to yield a normal platelet thrombus. This emphasizes the importance of the exposure of anionic phospholipids as the surface for the colocalization of the coagulation complexes in vivo. Abstract

The article presents a study which examined the effects of metformin on collagen glycation and diastolic dysfunction in diabetic myocardium. The methods conducted include the process of comparing untreated and metformin-treated diabetes in the basal state and during volume infusion. It is concluded that untreated diabetics exhibited increased diastolic chamber stiffness and that the chronic metformin used prevented the abnormalities of function and composition.

Abstract

Enzymatic degradation of beta-lactam antibiotics can be detected by cutting a wall of inoculum into the agar 3 mm from an antibiotic disc. Inactivation of the drug as it diffuses through the wall of inoculum results in distortion of the resulting inhibition zone. This procedure can be applied to the Stokes, Bauer-Kirby, International Collaborative Study (ICS) and Calibrated Dichotomous Sensitivity Test (CDS) methods so that they provide information about any detrimental effect of a pathogen on beta-lactam antibiotics, in addition to the effect the antibiotics have on the pathogen. 143. Selective depression of conduction of premature action potentials in canine Purkinje fibres by class lb antiarrhythmic drugs: comparison with la and Ic drugs.

Abstract

Microelectrodes were used to record action potentials and to estimate their conduction velocity in canine Purkinje fibres 8-15 mm long mounted in a tissue bath. The effects of varying stimulation rates and protocols were studied in the presence of nine different class I antiarrhythmic drugs at each of two concentrations (high and low therapeutic range). In all cases, as stimulation rate increased (range of cycle lengths 1000 ms to 200 ms), conduction velocity in the presence of a drug fell progressively below that in control solution at the same rate. No major differences in rate dependent behaviour at steady state were observed between the subclasses la, lb, and Ic. Differences were apparent, however, in the rate at which conduction velocity fell after a sudden decrease in cycle length. This was studied using two protocols. In the first of these, the conduction velocity was recorded of each action potential of a 20 beat train induced after a long rest period. In the presence of class lb drugs (lignocaine, tocainide, and mexiletine) there was a rapid decline within 2-3 beats to a new equilibrium level of conduction velocity. Class Ia drugs (quinidine, disopyramide, and procainamide) required 12-16 beats to achieve equilibrium, and class Ic agents (flecainide, encainide, and lorcainide) produced slow falls in conduction velocity that did not reach equilibrium within the 20 beat trains. The second protocol involved interpolation of increasingly premature extrastimuli. Class Ib drugs progressively slowed conduction of premature beats as the diastolic interval was reduced below 300-400 ms. Class Ia and Ic agents had no significant effect except when the diastolic interval was less than zero (that is, a premature beat arising during repolarisation of normal drive action potential). A combination of lignocaine (Ib) with quinidine (Ia) and flecainide (Ic) behaved similarly to class Ib drugs alone. It is concluded that class Ib antiarrhythmic drugs, with fast kinetics of interaction with sodium channels, may selectively depress early extrasystoles with little effect on normal beats. Abstract

The mass spectra of 18 substituted 1-(p-R2-phyeyl)-3-R3-4-(p-R1-phenylimino)-2-pyrazolin-5-ones have been obtained. The major cleavages under electron bombardment take place in the pyrazoline nucleus, producing four major fragment ions whose abundance is related primarily to the electronic properties of R1. These ions appear to be produced from the molecular ion via an intermediate formed by the loss of CO. Abstract

In dogs the effects of acute myocardial infarction (AMI) on right and left heart pressure relationships were studied. Right atrial pressure was a poor predictor of left heart pressures. Pulmonary artery end-diastolic, left atrial, and left ventricular end-diastolic pressures tended to equalize in normals (except with tachycardia) and after AMI. In a previously scarred left ventricle, however, after superimposed AMI and marked elevations in left ventricular end-diastolic pressure, prominent atrial impact waves resulted in this being appreciably higher than the other two pressure measurements. Abstract

1 The cellular electrophysiological effects of amiodarone and its metabolite desethylamiodarone (DEA) were studied in guinea-pig ventricular myocardium by use of standard microelectrode techniques. 2 Both compounds produced significant increases in action potential duration (Class III antiarrhythmic effect) and decreases in maximum rate of depolarization (Class I effect), at clinically relevant concentrations. 3 The Class I effects were rate-dependent, with small (0-16%) falls in maximum depolarization rate in the absence of stimulation ('resting block') and progressively larger effects at decreasing interstimulus intervals (range 1200-300 ms). 4 The kinetics of onset and offset of the Class I effect in response to a step change in driving rate were quite fast for both drugs (comparable to those reported for Class Ib agents). 5 It is concluded that this unique combination of Class III action plus Class I effects with fast onset and offset kinetics may help explain the great efficacy of amiodarone in antiarrhythmic therapy.
