Berberine is an alkaloid from several medicinal plants originally used to treat diarrhea and dysentery as a traditional Chinese herbal medicine. In recent years, berberine has been discovered to exhibit a wide spectrum of biological activities in the treatment of diverse diseases ranging from cancer and neurological dysfunctions to metabolic disorders and heart diseases. This review article summarizes the clinical practice and laboratory exploration of berberine for the treatment of cardiometabolic and heart diseases, with a focus on the novel insights and recent advances of the underlying mechanisms recognized in the past decade. Berberine was found to display pleiotropic therapeutic effects against dyslipidemia, hyperglycemia, hypertension, arrhythmia, and heart failure. The mechanisms of berberine for the treatment of cardiometabolic disease involve combating inflammation and oxidative stress such as inhibiting proprotein convertase subtilisin/kexin 9 (PCSK9) activation, regulating electrical signals and ionic channels such as targeting human ether-a-go-go related gene (hERG) currents, promoting energy metabolism such as activating adenosine monophosphate-activated protein kinase (AMPK) signaling pathway, modifying gut microbiota to promote transforming of berberine into its intestine-absorbable form, and interacting with non-coding RNAs via targeting multiple signaling pathways such as AMPK, mechanistic target of rapamycin (mTOR), etc. Collectively, berberine appears to be safe and well-tolerated in clinical practice, especially for those who are intolerant to statins. Knowledge from this field may pave the way for future development of more effective pharmaceutical approaches for managing cardiometabolic risk factors and preventing heart diseases.

Extracellular vesicles derived from trophoblasts (T-EVs) play an important role in pregnancy, but the mechanism is not entirely clear. In this study, we found that HLA-E, which is mostly confined to the cytoplasm of trophoblast cells, was secreted by T-EVs. The level of HLA-E in T-EVs from unexplained recurrent spontaneous abortion (URSA) patients was lower than that in normal pregnancy (NP) and RSA patients who had an abnormal embryo karyotype (AK-RSA). T-EVs promoted secretion of IFN-γ and VEGFα by decidual NK (dNK) cells from URSA patients via HLA-E, VEGFα was necessary for angiogenesis and trophoblast growth, and IFN-γ inhibited Th17 induction. Glycolysis and oxidative phosphorylation (OxPhos) were involved in this process. Glycolysis but not OxPhos of dNK cells facilitated by T-EVs was dependent on mTORC1 activation. Inhibition of T-EV production in vivo increased the susceptibility of mice to embryo absorption, which was reversed by transferring exogenous T-EVs. T-EVs promoted secretion of IFN-γ and VEGFα by dNK cells to maintain pregnancy via Qa-1 in abortion-prone mouse models. This study reveals a new mechanism of pregnancy maintenance mediated by HLA-E via T-EVs.

Natural enemies that impact pest populations must be understood in order to build integrated pest control strategies and to understand the most important aspects affecting pest dynamics. Haloxylon ammodendron (C. A. Mey.) Bunge is an important perennial plant species extensively used in sand stabilization and wind prevention in arid areas. This study aimed to determine the main defoliators that damage H. ammodendron and the parasitoid complex associated with them. Twelve species of defoliators were found in Northern Xinjiang, and Teia dubia (Tauscher) (Lepidoptera: Lymantriidae), Scrobipalpa sp. (Lepidoptera: Gelechiidae), and Eucharia festiva Hüfnagel (Lepidoptera: Arctiidae) were the dominant pests. T. dubia is the predominant defoliator with three generations a year. Northwest China, Central Asia, and the Mediterranean region are potentially suitable habitats for T. dubia in the world, while Xinjiang is the primary distribution area in China. Parasitoids belonging to seven species and four families were reared from the larvae of T. dubia, they were all endoparasitoids and koinobiont. Cotesia sp. (Hymenoptera: Braconidae) is the dominant parasitoid and prefer to parasitic in the 3rd-5th instar larvae. The present study provides the basis for understanding the species composition and natural enemies of lepidopteran defoliators. It will be an effective tool for the integrated pest management programs of H. ammodendron forest.

This study investigated the ability of immunity- and matrix- regulatory cells (IMRCs) to improve cognitive function in a rat model of vascular cognitive impairment.A chronic cerebral hypoperfusion (CCH) model was established in rats via permanent bilateral occlusion of the common carotid arteries (two-vessel occlusion, 2VO). The rats then received intravenous injections of IMRCs or saline. A single injection of different doses of IMRCs (1 × 10Intravenous injection of 4 × 10Intravenous injection of IMRCs protected against CCH-induced white matter injury and cognitive impairment inhibition of microglial activation and regulation of microglia polarization.

In response to COVID-19 epidemic and China-US trade frictions, based on the survey data of 413 companies, starting from the descriptive statistical analysis of whether the companies with different characteristics have migrated or not, with the help of the feature selection method in machine learning, The balanced characteristics of the enterprise migration survey data were analyzed by feature selection, and finally 13 important characteristic variables that affect whether the company migrated or not were selected from 102. On this basis, the effects of the dual effects of COVID-19 epidemic and China-US trade friction on the migration of enterprises were further investigated and analyzed through the Probit model. China is the world's largest rare earth producer and exporter, previous studies have shown that rare earth elements can cause oxidative damage in animal testis. However, the molecular mechanisms underlying these observations have yet to be elucidated. In this paper, male mice were fed with different doses (10, 20, and 40 mg/kg BW) of LaCl3 for 90 consecutive days, regulatory role of nuclear factor erythroid-2 related factor 2 (Nrf-2)/antioxidant response element (ARE) pathway in testicular oxidative stress induced by LaCl3 were investigated. Analysis showed that LaCl3 exposure could lead to severe testicular pathological changes and apoptosis in spermatogenic cells, it up-regulated the peroxidation of lipids, proteins and DNA, and induced the excessive levels of reactive oxygen species (ROS) production in mouse testis, reduced the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and glutathione S epoxide transferase (GST) as well as the glutathione (GSH) content. Furthermore, exposure to LaCl3 also downregulated the expression of Nrf2 and its target gene products, including heme oxygenase 1 (HO-1), glutamate-cysteine ligase catalytic subunit (GCLC), NAD(P)H dehydrogenase [quinine] 1(NQO1), protein kinase C (PKC), and phosphatidylinositol 3-kinase (PI3K), but upregulated the expression of Kelch-like ECH-related protein 1 (Keap1) in damaged mouse testes. Collectively, our data imply that the oxidative damage induced by LaCl3 in testis was related to inhibition of the Nrf-2/AREs pathway activation.

Color has strong relationship with food quality. In this paper, partial least square regression (PLSR) and least square-support vector machine (LS-SVM) models combined with six different color spaces (NRGB, CIELAB, CMY, HSI, I1I2I3, and YCbCr) were developed and compared to predict pH value and soluble solids content (SSC) in red bayberry. The results showed that PLSR and LS-SVM models coupled with color space could predict pH value in red bayberry (r = 0.93–0.96, RMSE = 0.09–0.12, MAE = 0.07–0.09, and MRE = 0.04–0.06). In addition, the minimum errors (RMSE = 0.09, MAE = 0.07, and MRE = 0.04) and maximum correlation coefficient value (r = 0.96) were found with the PLSR based on CMY, I1I2I3, and YCbCr color spaces. For predicting SSC, PLSR models based on CIELAB color space (r = 0.90, RMSE = 0.91, MAE = 0.69 and MRE = 0.12) and HSI color space (r = 0.89, RMSE = 0.95, MAE = 0.73 and MRE = 0.13) were recommended. The results indicated that color space combined with chemometric is suitable to non-destructively detect pH value and SSC of red bayberry.

Developing potent non-opioid pain medications is an integral part of the battle to conquer both chronic pain and the current opioid crisis. Although most screening approaches use in vitro surrogate targets, in vivo screening of analgesic candidates is a necessary preclinical step in drug discovery. Here, we report the design of a new automated behavioral testing apparatus based on the principle of a thermal place preference test (TPPT). This new design can detect, quantify, and differentiate behavioral responses to cold stimuli between sham and chronic constriction injury (CCI) rodents with up to 12 animals tested simultaneously. At an optimized temperature pair of 12.5 °C vs 30.0 °C (±0.5 °C), the TPPT design has captured the antinociceptive effects of morphine and pregabalin on CCI rats in individual 10 min tests. Moreover, it can differentiate analgesic effects by morphine or pregabalin from anxiolytic effects by diazepam. The results, along with the relatively low cost to construct the apparatus and moderately high throughput, make our TPPT design applicable for behavioral studies of chronic pain in rodents and for high-throughput in vivo screening of the next generation of pain medications.

Dendrobium officinale (D. officinale) is commonly used as a functional food or herbal medicine worldwide, but often adulterated with low-priced materials. In this study, we attempted to develop an integrated method of least-squares support vector machine and nuclear magnetic resonance spectroscopy (LS-SVM-NMR) to identify and quantify the adulteration of D. officinale powder. We found that LS-SVM-NMR can identify the adulterated D. officinale powder with an overall accuracy of 100% and quantify its purity with an R2 of 0.999 and RMSE of 1.410 at model validation phase. In addition, our results from the double-blinded test revealed that LS-SVM-NMR can yield a classification accuracy of 98% for distinguishing the adulterated D. officinale powder, and predict its purity with an R2 of 0.998 and RMSE of 2.660. Moreover, adulteration evaluation reports can be obtained in 10 min. Therefore, LS-SVM-NMR proposed herein could be used as a promising and time-saving tool for the quality evaluation of D. officinale or even other food products.

An extensive study collected in situ data along the Yellow Sea (YS) and East China Sea (ECS) to assess the radiometric properties and the concentration of the water constituents derived from Moderate Resolution Imaging Spectroradiometer (MODIS). Thirteen high quality match-ups were obtained for evaluating the MODIS estimates of Rrs(λ), chlorophyll a (Chi a) and concentrations of suspended particulate sediment matter (SPM). For MODIS Rrs(λ), the mean absolute percentage difference (APD) was in the range of 20%-36%, and the highest uncertainty appeared at 412 nm, whereas the band ratio of Rrs(λ) at 488 nm compared with that at 547 nm was highly consistent, with an APD of 7%. A combination of near-infrared bands and shortwave infrared wavelengths atmosphere correction algorithm (NIR-SWIR algorithm) was applied to the MODIS data, and the estimation accuracy of RIS were improved at most of the visible spectral bands except 645 nm, 667 nm and 678 nm. Two ocean-colour empirical algorithms for Chi a estimation were applied to the processed data, the results indicated that the accuracy of the derived Chi a values was obviously improved, the four-band algorithms outperformed the other algorithm for measured and simulated datasets, and the minimum APD was 35%. The SPM was also quantified. Two regional and two coastal SPM algorithms were modified according to the in situ data. By comparison, the modified Tassan model had a higher accuracy for the application along the YS and ECS with an APD of 21%. However, given the limited match-up dataset and the potential influence of the aerosol properties on atmosphere correction, further research is required to develop additional algorithms especially for the low Chi a coastal water.

Cervical cancer is a common gynecological malignancy, and miR-133b is an abnormally expressed cervical cancer gene, which suggests that miR-133b may be involved in the occurrence and development of cervical cancer. However, the underlying mechanism is still unclear. miR-133b was overexpressed or silenced in the cervical cancer cell line C33A. Brefeldin A-inhibited guanine nucleotide-exchange protein 1 (ARFGEF1) was combined with overexpression of miR-133b in C33A cells. Cell Counting Kit-8, clone formation, and Transwell assays were performed to determine the influence of miR-133b and ARFGEF1 on clone formation, proliferation, migration, and invasion of C33A cells. The interaction between miR-133b and ARFGEF1 was verified using a luciferase reporter assay. Finally, the mRNA and protein expression of miR-133b and ARFGEF1 in the tumor and adjacent normal tissues of cervical cancer patients was detected by real-time quantitative PCR, Western blotting, and immunohistochemistry. Objective. GIT1 is identified as a novel tumor oncogene in breast cancer. In this article, we aimed to explore the role of GIT1 in the progression of head and neck squamous cell carcinoma (HNSCC). Methods. GIT1 expression in HNSCC was detected by RT-qPCR, immunohistochemistry assay, and Western blot. HNSCC cell proliferation, migration, and invasion were examined by CCK-8 assay, Wound healing assay, and Transwell assay, respectively. Cell apoptosis was detected by flow cytometric analysis. Results. In our study, GIT1 was notably upregulated in HNSCC tissues and cells. Moreover, GIT1 expression level had positive corelation with pathological grade and nodal status of HNSCC. Functional experiments showed that knockdown of GIT1 restrained HNSCC proliferation, invasion, migration, and EMT and facilitated cell apoptosis. Furthermore, GIT1 knockdown was found to restrain HNSCC tumor growth and lung metastasis. Additionally, PI3K/AKT/mTOR signal pathway inhibitors suppressed the effect of GIT1 on HNSCC cell progression. Conclusion. GIT1 was upregulated in HNSCC and facilitated HNSCC cell progression by inducing PI3K/AKT/mTOR signal pathway. Therefore, we suggested that GIT1 might be a potential target for HNSCC treatment.

IntroductionTo explore the role of ferritin in placenta, serum and umbilical cord blood of pregnant women and the changes of oxidative stress injury as well as cell apoptosis in placenta in the pathogenesis of preeclampsia (PE).Material and methodsSixty pregnant women with severe PE were assigned into early-onset and late-onset PE group. Another 60 cases of normal late pregnant women with similar gestational weeks were divided into early-onset and late-onset control group. Maternal serum and fetal umbilical cord blood ferritin content was determined by automatic biochemical immunoassay system; mRNA expression levels of ferritin and ferritin heavy chain (FTH) were detected by reverse transcription real-time fluorescence quantitative polymerase chain reaction (RT-qPCR). Western Blot was used to detect the relative expression level of ferritin and apoptosis; the contents of total superoxide dismutase (T-SOD) and malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) were detected by colorimetry.ResultsSerum uric acid (UA) and creatinine (Cr) levels of PE groups were significantly higher when compared to the controls. The serum ferritin levels in blood sample and umbilical cord blood sample were significantly higher relative to the controls. However, the mRNA and protein levels of ferritin levels in placenta samples were significantly lower compared with the controls. The placental cleaved caspase-3, Bcl-2 levels were significantly lower than the early onset PE group. The levels of GSH-Px and MDA in placenta were significantly higher.ConclusionsThese results may assist understanding the pathogenesis of PE and provide potential biomarkers for diagnosis of PE.

Abstract: Diffuse large B-cell lymphoma (DLBCL), an aggressive cancer of the B cells, is the most common pathological type of non-Hodgkin’s lymphoma (NHL), and the typical heterogeneity of the disease is due to metastasis, which indicates a poor prognosis. Currently, the key mechanism of metastasis remains largely unknown, and research is focusing on the same in DLBCL. Recent studies have focused on the role of mesenchymal-epithelial transition (MET) and epithelial-mesenchymal transition (EMT), the extracellular matrix (ECM), chemokines, cancer stem cells, and non-coding RNAs in DLBCL. Here, we have summarised the advances and challenges in the investigation of metastasis in DLBCL and attempted to reveal the potential targets that can improve patient survival.

In China, the opening of high-speed rails (HSR) brings significant changes to the source-destination spatial distance, the accessibility of destinations, and the spatial structure of tourist flows in each region, exerting varied HSR effects on different types of cities. Against this backdrop, it is meaningful to deeply explore tourists’ preference for city destinations in the light of HSR effects. The exploration could contribute greatly to the planning, marketing, management, and sustainable development of urban tourism. This paper takes Xiangtan and Yueyang as typical cases of the diffusion effect and the corridor effect of HSR. Firstly, the factors affecting destination choice were identified, and the attribute levels were configured, forming multiple virtual alternatives. Next, questionnaire surveys were carried out to collect tourists’ selections between each pair of alternatives. Further, a discrete choice model was constructed to assign a weight to each factor, reflecting its importance to tourists’ decision-making regarding their destination selection and to disclose the law of tourists’ preferences for destinations. The results showed that (1) Under the HSR diffusion effect, the top three factors affecting tourists’ preference for destinations in Xiangtan are convenience, connection time, and popularity, under HSR corridor effect, the top three factors affecting the tourists’ preference for destinations in Yueyang are reputation, convenience, and leisure and reception facilities (LRFs). (2) The destination preference is closely associated with personal features like gender, income, occupation, and fellow travelers. Tourists with different personal features give different attention to the various influencing factors. The research findings provide a reference for the sustainable development of urban tourism.

Whether aspirin platelet reactivity affects platelet function and clinical outcomes with different antiplatelet therapies in patients with mild stroke or transient ischemic attack (TIA) remains unclear. We conducted a subgroup analysis of the PRINCE trial.Patients with mild stroke or TIA were randomized into aspirin+ticagrelor, or aspirin+clopidogrel groups; aspirin reaction units (ARU) were measured at the baseline and after 7 ± 2 days to assess response to treatment. High on-treatment platelet reactivity (HPR) was defined as ≥550 ARU (poor response to aspirin). The platelet functions of ticagrelor and clopidogrel were measured using the VerifyNow P2YAmong 628 enrolled patients, 69 (11%) were poor aspirin responders. After 7 ± 2 days, the proportion of poor P2YIn patients with minor stroke or TIA, clopidogrel, and particularly ticagrelor, decreased platelet function in poor versus good aspirin responders. The poor platelet reactivity of aspirin could not sufficiently reduce the risk of recurrent stroke with ticagrelor or clopidogrel; however, HPR (poor aspirin response) may have a protective effect on clinically relevant major bleeding.

Supplying adequate nitrogen (N) to meet crop demand is critical for enhancing agricultural sustainability. Not only fertilizer N, but also N from other available sources should be considered in N supply capacity. We conducted a 10-year farming experiment using a split-plot design with two different main fertilizer management approaches and three N application strategies as add-on sub-treatments. Based on the experiment, we estimated the total N supply (TNsupply) for the summer maize cropping system, through considering environmental, soil, crop residue, and fertilizer N sources. An appropriate TNsupply was established by correlating TNsupply with the relative yield (RY), N input and output, and N use efficiency (NUE). The results revealed a wide variation in TNsupply (from 88 to 755 kg ha−1). The RY, N input, and N output fitted well to TNsupply using linear-plateau, linear, and linear-plateau models, respectively. The lower limits of TNsupply for achieving the maximum RY and N output were 361 and 358 kg ha−1, respectively. The relationship between N input and N output was described as linear-plateau. We determined the slope of the linear curve (55.4%) as the lower limit of NUE, beyond which the upper limit of TNsupply was determined to be less than 497 kg ha−1. Thus, appropriate TNsupply values ranged from 325 to 497 kg ha−1 for summer maize production, which could ensure enough N supply for higher yields and avoid excessive N input for higher NUE and lower environmental N loss. Our findings highlight that TNsupply can be an alternative indicator for evaluating N management.

Feature selection is an important research field in machine learning since high-dimensionality is a common characteristic of real-world data. It has two main objectives, which are to maximize the classification accuracy while minimizing the number of selected features. As the two objectives are usually in conflict with each other, it makes feature selection a multi-objective problem. However, the large search space and discrete Pareto front makes it not easy for existing evolutionary multi-objective algorithms. In order to deal with the above mentioned difficulties in feature selection, an entropy driven multiobjective particle swarm optimization algorithm is proposed to remove redundant feature and decrease computational complexity. First, its basic idea is to model feature selection as a multiobjective optimization problem by optimizing the number of features and the classification accuracy in supervised condition simultaneously. Second, a particle initialization strategy based on information entropy is designed to improve the quality of initial solutions, and an adaptive velocity update rule is used to swap between local search and global search. Besides, a specified discrete nondominated sorting is designed. These strategies enable the proposed algorithm to gain better performance on both the quality and size of feature subset. The experimental results show that the proposed algorithm can maintain or improve the quality of Pareto fronts evolved by the state-of-the-art algorithms for feature selection.

Bovine tuberculosis, caused by both MTB(Mycobacterium tuberculosis) and MB(Mycobacterium bovis), is one of the most serious zoonotic diseases in the world. Both pathogenic MTB and MB interact closely with the host alveolar macrophages. However, the mechanisms of defense response of macrophages to MTB or MB still need further researches. Presently, in-depth comparative proteome analysis was performed on the MB- and MTB- infected macrophage. We identified that macrophages was more sensitive to the MB infection. Both pathogens could induce strong energy metabolism of macrophages. To eliminate MB and protect the host, a bunch of proteins and signaling which involved in autophagy- and inflammatory-related progresses were highly activated in macrophages following MB infection. In contrast, only proteins relevant to energy metabolisms were highly expressed in macrophages following MTB infection. In summary, we proposed that macrophages were more sensitive to MB attacks and could deployed proteins functioned in autophagy- and inflammatory-related progresses to protect the host. Our research provide a novel insight into the different mechanisms of defense responses of macrophages to both MTB and MB infection.

Crimean-Congo hemorrhagic fever virus (CCHFV) is a tick-borne arbovirus that can cause bleeding and death in humans. The mortality rate in humans is between 5 to 30%. The pathogen is prevalent in more than 30 countries in the world. In China, reports of strains of CCHFV have been concentrated in Xinjiang province. However, the CCHFV strain has never been reported in Inner Mongolia, China. This study reports new CCHFV strains, HANM-18, from Hyalomma asiaticum and Hyalomma dromedarii collected in Alxa Left Banner and Alxa Right Banner in Inner Mongolia. Complete sequences of CCHFV were obtained by the nested PCR technique and used for analyzing the identity and evolutionary relationship with other CCHFV strains. Interestingly, our results showed that the S and L fragments of the HANM-18 strain had high degrees of identity with Xingjiang isolate strains, and the M fragment had significant identity with South African isolates. These analyses also indicate that the HANM-18 strain may have been prevalent in northwest Inner Mongolia for many years. This discovery will be helpful in CCHF prevention and control in Inner Mongolia, and it also adds new evidence to the epidemiology of CCHF in China.

Postmenopausal women taking estrogen supplements are at a lower risk of advanced colorectal cancer, but the underlying mechanism remains unclear. Thus, this study examined the role of estrogen in colorectal cancer. Estrogen receptor expression levels in in situ colorectal cancer tissue from female patients increased significantly, indicating their estrogen sensitivity. Compared with the sham-operated group, the growth of MC38 tumors was enhanced in ovariectomized mice, which was reversed in ovariectomized mice with E2 supplementation. The PD-L1+ M2-like macrophage, regulatory T (Treg) cell, and myeloid-derived suppressor cell (MDSC) populations significantly increased, and the population of cytotoxic CD8+ T cells declined in MC38 tumors in ovariectomized mice, which were all reversed in ovariectomized mice with E2 supplementation. MC38 cell-derived extracellular vesicles (MC38-EVs), but not EVs derived from MC38 cells treated with E2 (E2-MC38-EVs), were involved in the establishment of immunosuppressive tumor microenvironment. E2-MC38-EVs contained lower TGF-β1 levels and were less capable of inducing Treg cells than MC38-EVs in vitro. Overall, these results show that estrogen treatment prevents MC38 tumor growth via regulating the tumor immune microenvironment through MC38-EVs.

There is a strong capacity for hair cell regeneration after damage in the inner ear of non-mammals. However, mammalian hair cells are substantially unable to regenerate. To obtain insights into the mechanism of this difference, we analyzed the transcriptomic changes in the mouse cochleae suffered from gentamicin damage and compared them with those in the chick cochleae suffered from the same damage. The results indicated that 2,230 genes had significantly differential expression between the gentamicin- and saline-treated mouse cochleae. Some of the differentially expressed genes were grouped into 265 signaling pathways, including the Notch, Wnt (Wingless and INT-1), Bmp (bone morphogenetic protein), FGF (fibroblast growth factor) and Shh (sonic hedgehog) pathways. Using pharmacological inhibitors or agonists of these pathways, the effects of these pathways on hair cell regeneration were further studied. The results indicated that Bmp alone and its coregulation with the Notch or Wnt signaling pathways increased the numbers of generated cells from transdifferentiation or proliferation in the mouse cochlea after damage, in addition to the reported coregulation of Notch and Wnt. Thus, this work indicates a new signaling pathway (Bmp) and its synergetic coregulation in mammalian hair cell regeneration, providing potential therapeutic targets to increase mammalian hair cell regeneration.

The Japanese mantis shrimp Oratosquilla oratoria (Stomatopoda; Crustacea) is one of the most economically important aquatic species of Pacific shrimp and it is distributed from Japan to the coast of China, the Philippines, the Malay Peninsula, and the Hawaiian Islands. Early studies described certain characteristics of spermatogenesis and the sperm ultrastructure in Stomatopoda, but the composition of sperm basic nuclear proteins (SBNPs) remains completely unknown. We studied the sperm ultrastructure of O. oratoria using transmission electron microscopy and the histone composition using immunofluorescence and immunoelectron microscopy. We found that the spherical nucleus is adjacent to the electron translucent external coat, which occurs in early spermatids. The acrosomal structure begins to form at the junction of the nucleus and the external coat. At the mid‐spermatid stage, part of the chromatin appears to be more electron‐dense than the external coat side. The aflagellate sperm of O. oratoria, are rounded or slightly ovoid in shape and have a consistent granular nucleus, an acrosome structure of pushpin shape and a spherical vesicular body in which faintly granular material is scattered. The acrosome consists of an acrosomal vesicle, perforatorium, and subacrosomal material. The sperm contains histones H2A, H2B, H3, H4, H3.3, H2AX, and H2AZ as well as some histone modifications, that is, H3K9me3, H3K4me2, H3S10ph, H4Kac, and H2A + H4S1ph. Histones are localized not only in the nucleus of the sperm but also in other structures outside the nucleus. The results may provide new perspectives for systematic studies of crustaceans and their sperm chromatin components. These findings extend the study of the sperm structure of Stomatopoda and provide basic data to elucidate the epigenetic mechanism of fertilization.

Ultrafine Fe2O3 nanorods anchored on graphene oxide (GO) are synthesized by a one-pot route under mild conditions in presence of KMnO4 and Ni foam. Ni foam is beneficial for the formation of Fe2O3 rather than FeOOH under otherwise the same conditions, while KMnO4 accelerates the hydrolysis of FeCl3, which promotes the nucleation of Fe2O3 and the achievement of uniform ultra-small morphology thereby. As an anode material for lithium-ion batteries, the Fe2O3/GO manifests a reversible capacity of 1004 mA h g−1after 500 cycles at 200 mA g−1, and even presents a high reversible capacity of 722 mA h g−1 at 1600 mA g−1. The excellent electrochemical properties of Fe2O3/GO are attributed to the ultrafine nanorods of Fe2O3 and the strong interaction between Fe2O3 and GO, which can reduce the diffusion distance of Li+ ions, enlarge the electrode/electrolyte contact area and improve the cyclic stability of iron oxide.

Objectives: This study Aims to explore the role of placental Cripto-1 in the incidence of an adherent placenta. Material and methods: Ten pregnant women with placenta increta, 20 pregnant women with placenta previa and 30 women with normal pregnant were enrolled in this study. Reverse transcription-polymerase chain reaction (RT-PCR) was used to measure the expression of Cripto-1 in the placenta while as the analysis of placental Cripto-1 was performed by Western blotting Results: The placenta increta group showed higher levels of Cripto-1 in the center of the increta as compared to the non-implantation area. The level of placental Cripto-1 in the placenta increta was higher than that of the placenta accrete. The expression of placental Cripto-1 in the placenta increta and placenta previa groups was higher than that of control. Conclusions: Placental Cripto-1 is involved in the regulation of placental tissue invasion. Additionally, excessive placental growth or penetration into the myometrium are likely to be involved in the development of placenta increta.

Activin A, a multifunctional cytokine of transforming growth factor-β (TGF-β) superfamily, can be produced by the diverse immune cells. NK cells in peripheral blood are one of the major immune cells applied to cancer therapy in recent years. However, whether activin A can be produced by natural killer (NK) cells and be involved in regulation of peripheral blood NK cells activities of mouse are not well characterized. Here, we found that activin type IIA and IIB receptors and signaling molecules Smad2, 3 were expressed in peripheral blood NK cells of mouse by flow cytometry and RT-PCR. The cultured blood NK cells of mouse not only produced activin βA chain protein by intracellular cytokine staining, but also secreted mature activin A protein by enzyme-linked immunosorbent assay (ELISA), and the production was promoted by IL-2. In addition, IL-2 as a positive control obviously promoted IFNγ production of mouse blood NK cells in vitro. However, activin A suppressed IFNγ production, but enhanced IL-2 synthesis and did not alter IL-10 production. Moreover, we found that activin A significantly suppressed the ability of NK cells to lyse target cells. These data revealed that blood NK cells of mouse were not only the target cells in response to activin A, but also the source of activin A, suggesting that activin A may play an important role in regulation of NK cells activities of mouse in an autocrine / paracrine manner.

Background The number of research articles on health-related quality of life (HRQoL) has been strikingly increasing. This study aimed to explore the general trends and hotspots of HRQoL. Methods Based on the Web of Science database, research on HRQoL published between 2000 and 2019 were identified. A bibliometric analysis was performed based on the number of articles, citations, published journals, authors' addresses, and keywords. Descriptive analysis, visualization of geographic distribution and keyword clustering analysis were applied to the collected data. Results The annual number of articles showed growth over the past twenty years, but the annual total citations and annual citations per article were both in decreasing trends. Articles about HRQoL were more likely to be published in journals of multi-subject categories. The HRQoL research was mainly distributed across North America and Europe throughout the twenty years and ushered in a vigorous development worldwide after 2015. Cooperation strength between domestic institutions was much greater than that of international institutions. HRQoL research had six concentrated clusters: HRQoL, Depression, Obesity, Disability, Oncology, Fatigue. Conclusion This study provided an overall perspective of global research trends and hotspots in HRQoL, and a potential insight for future research. HRQoL research had experienced significant increasing development during 2000–2019, especially the HRQoL measurement instruments, however, there were significant regional disparities in scientific output in HRQoL.

Metformin has been demonstrated to be beneficial for the treatment of an impaired myocardium as a result of ischemia/reperfusion (I/R) injury, and miR-34a may be involved in this process. The aim of the present study was to determine the mechanisms by which metformin attenuated myocardial I/R injury–induced apoptosis. In the in vivo I/R model using Sprague–Dawley rats, metformin reduced the area of damaged myocardium and serum creatine MB isoform (CKMB) activity resulting in protection of the myocardium. Metformin also reduced apoptosis and the expression of apoptosis associated proteins, including caspase 3 and cleaved caspase, and decreased the expression of miR-34a, which is upregulated during I/R injury, which in turn resulted in corresponding changes in expression of Bcl-2, a direct target of miR-34a both in vitro and in vivo. To further examine the role of miR-34a in this process, H9C2 cells were transfected by a miR-34a mimic and inhibitor. Overexpression of miR-34a increased apoptosis in H9C2 cells induced by oxygen–glucose deprivation/recovery and knockdown of miR-34a expression–reduced apoptosis under the same conditions. Therefore, the effect of metformin on miR-34a in vitro were assessed. Metformin decreased the deacetylation activity of silent information regulator 1 resulting in reduced Ac-p53 levels, which reduced the levels of pri-miR-34a, and thus in turn reduced miR-34a levels. To confirm these results clinically, 90 patients with ST-segment elevation myocardial infarction following percutaneous coronary intervention were recruited. Patients who took metformin regularly before infarction had lower miR-34a levels and lower serum CKMB activity. Metformin also improved the sum ST-segment recovery following I/R injury. In conclusion, metformin may be helpful in the treatment of myocardial I/R.

Summary Activin A levels are elevated during multiple severe infections and associated with an increased risk of death. However, the role of activin A in bacterial infection is still unclear. Here, we found that activin A levels were increased during S. aureus skin infection in mice. Administration of activin A increased the bacterial burden and promoted the spread of bacteria in vivo. Moreover, activin A inhibited neutrophil chemotaxis to N-formylmethionine-leucyl-phenylalanine via the type IIA activin receptor (ActRIIA) in vitro and impaired ActRIIA+ neutrophil recruitment to infection foci in vivo. Additionally, we identified a novel subpopulation of neutrophils, ActRIIA+ neutrophils, which exhibit superior phagocytic capacity compared to ActRIIA− neutrophils and possess an N2-like immunoregulatory activity via secreting IL-10 and TGF-β. Taken together, these findings indicate that activin A inhibits the recruitment of ActRIIA+ neutrophils to infected foci, leading to the impairment of bacterial clearance, and thus may hamper early infection control.
