Genetic dissection of high KRN trait and cob length has been undertaken by several researchers resulting in identification of loci controlling the traits. Further fine mapping of QTLs controlling KRN and cob length and TILLING strategies identified the underlying genes. All these studies are used temperate maize genotypes which are of limited use to researcher in sub-tropical region, and the sub-tropical maize production systems. Present investigation explores the availability of genetic regions responsible for KRN in sub-tropical maize germplasm. A total of 280 subtropical maize germplasm was analyzed for their KRN variation and selected 45 stable lines were subjected to molecular characterization using genes linked to KRN in maize. Diversity analysis was also performed to understand the possible association of character with the KRN gene in deciding its variation in the given population. It was revealed that four genes showed highest probability of influencing KRN traits in these tropical maize inbred lines. The remaining genes not establishing specific pattern of association with high and low KRN genotypes may need further study on its allelic variation.

Purpose: After apparently successful excision of breast cancer, risk of local recurrence remains high mainly in the area surrounding the original tumor, indicating that wound healing processes may be implicated. The proportional reduction of this risk by radiotherapy does not depend on the extent of surgery, suggesting that radiotherapy, in addition to killing tumor cells, may influence the tumor microenvironment.Experimental Design: We studied how normal and mammary carcinoma cell growth and motility are affected by surgical wound fluids (WF), collected over 24 h following breast-conserving surgery in 45 patients, 20 of whom had received additional TARGeted Intraoperative radioTherapy (TARGIT), immediately after the surgical excision. The proteomic profile of the WF and their effects on the activation of intracellular signal transduction pathways of breast cancer cells were also analyzed.Results: WF stimulated proliferation, migration, and invasion of breast cancer cell lines. The stimulatory effect was almost completely abrogated when fluids from TARGIT-treated patients were used. These fluids displayed altered expression of several cytokines and failed to properly stimulate the activation of some intracellular signal transduction pathways, when compared with fluids harvested from untreated patients.Conclusions: Delivery of TARGIT to the tumor bed alters the molecular composition and biological activity of surgical WF. This novel antitumoral effect could, at least partially, explain the very low recurrence rates found in a large pilot study using TARGIT. It also opens a novel avenue for identifying new molecular targets and testing novel therapeutic agents.

Supplementary Data from Targeted Intraoperative Radiotherapy Impairs the Stimulation of Breast Cancer Cell Proliferation and Invasion Caused by Surgical Wounding

The main target ofTthis study is to develop an antibacterial chitosan nanocomposite fabric among in situ crated silver nanoparticles utilizing Vitex leafTextract, which i s amedicinal herb. The CNCFs were evaluated using scanning electron microscopy (SEM), Fourier transform infrared (FT-IR) spectroscopy,Xray diffraction (XRD), and antimicrobial testing. Additionally, these CNCFs have excellent antibacterial characteristics. These CNCFs could be used in medical applic ations like surgical aprons, wound cleansing, wound dressing, and hospital bed mat erials, as they were manufactured utilising a simple and environmentally acceptable approach.

With the objective to investigate the inheritance pattern of fertility restoration of A1cytoplasm in multi-environments, six crosses were developed by crossing two A1 cytoplasm-based male sterile lines (A-lines) with three diverse restorers (R-lines). The parents, 6 F2s and 6 BC1s were evaluated in the rainy season at Delhi and summer season at Delhi and Dharwad. Four crosses viz.,576A1×PPMI 1012, 411A1× PPMI 1012, 576A1× ICMR 06111 and 411A1× ICMR 06111 showed a segregation ratio of fertile: semi-fertile: sterile as 9:3:4 and 1:1:2 in F2 and BC1 generations, respectively, based on pollen fertility and seed set percent data, indicating digenic supplementary gene action. In the other two crosses, i.e., 576A1× HTP 94/54 and 411A1× HTP 94/54, different digenic ratios of 9:7 and 1:3 of fertile and sterile plants were observed in F2 and BC1 generations, respectively, indicating complementary interaction of two genes. The deviation of expected ratios in the summer season at Delhi resulted due to excess sterile plants, as influenced by modifiers and environmental conditions that prevailed during the dry season. Thus, in the present investigation, fertility restoration of A1-based CGMS system was observed to be governed by two major genes. Still, with different types of epistatic interactions, digenic inheritance of A1 cytoplasm suggests that test of allelism and genetic mapping of fertility restorer genes can be taken up in the future.

Introduction: Targeted intraoperative radiotherapy (TARGIT - IORT) as a tumor bed boost during breast conserving surgery is an established option for women with early breast cancer. In a previous study our group could demonstrate a beneficial effect of TARGIT-IORT on overall survival after neoadjuvant chemotherapy driven by non-breast cancer mortality, especially from cardiac or pulmonary diseases. This was in line with results of TARGIT-A trial that demonstrated an improved overall survival after TARGIT-IORT that was driven by other causes of death than breast cancer but in our analysis we could not exclude a selection bias because we had no data regarding pulmonary and cardiac co-morbidities. In this study we present an analysis of breast cancer associated and non-breast cancer associated mortality and morbidity in a lower risk population treated with TARGIT-IORT boost or external boost (EBRT) and with a complete dataset of pulmonary and cardiac co-morbidities at baseline. Material/Methods: In this non-randomized cohort study involving 125 patients with early breast cancer we compared outcomes of 59 patients who received a tumor bed boost with IORT (TARGIT-IORT) during lumpectomy versus 66 patients with external (EBRT) boost. All patients received whole breast radiotherapy. Local recurrence rates, distant recurrence rates, mortality, causes of death and incidence of new pulmonary and cardiac morbidity were compared. The small event rate did not allow a formal comparison, so the results are reported descriptively. Results: There were no statistically significant differences between the two groups regarding age, menopausal status, nodal status, ER status, PR status, HER2 status, tumor size, grading, cardiac or pulmonary diseases and hypertension. Median follow up was 60 months. Local recurrence occurred in 3 cases in the EBRT group (4.5%) and 4 cases in the TARGIT-IORT group (6.8%). Distant recurrences were observed in 5 cases in the EBR group (7.6%) and 3 cases in the TARGIT-IORT group (5.1%). 9 patients in the EBRT group (13.6%) died in the follow-up of 60 months and 6 patients (10.2%) in the TARGIT-IORT group. No death from a pulmonary disease occurred and the incidence of new pulmonary and new cardiac diseases showed no difference between both groups with 3 cases in the EBRT group (4.5%) and 2 cases in the TARGIT-IORT group (3.4%). Death from cardiac disease occurred with an incidence of 7 cases in the EBRT group (10.6%) and only 2 cases in the TARGIT-IORT group (3.4%). 2 patients in the EBRT group died of breast cancer (3.0%) whereas this did not happen at all in the TARGIT-IORT group. Conclusion: This analysis of an average risk group of breast cancer patients well balanced for pulmonary and cardiac co-morbidities demonstrates that TARGIT-IORT as an anticipated boost is not inferior to external beam radiotherapy boost. Trends for an improved overall survival after TARGIT-IORT have been reported before. The trend for a lower incidence of death from cardiac disease is in line with earlier considerations that TARGIT-IORT may induce miR-223 in the peritumoral breast tissue resulting in a downregulation of the local expression of epidermal growth factor (EGF) and a decreased activation of epidermal growth factor receptor (EGFR). This mechanism could explain an effect of TARGIT-IORT on the vascular system. In the currently recruiting TARGIT-B study this aspect will be prospectively investigated. USG guided FNAC of intra-abdominal masses is a good diagnostic modality to facilitate treatment planning. Histopathological correlation aids in sensitivity and specificity of Ultra sound guided FNAC thereby making it first investigation of choice for evaluation of intra-abdominal masses. This study attempts to assess the utility of Ultra sound guided fine needle aspiration cytology of intra-abdominal masses and their histopathological correlation. Methodology: This Prospective study will be conducted in Cyto-histopathology division of Department of Pathology, JNMC, Wardha. Total 30 patients with abdominal masses will be enrolled in the study. Fine needle aspiration of the abdominal lump will be done in the Department of Radiology, using real time USG. The data will be analysed using appropriate statistical programmes. Expected Results: A significant correlation is expected between histopathology and ultrasound guided fine needle aspiration cytology of intra-abdominal masses. Conclusion: Conclusion will be drawn from on the basis of results obtained from this study.

The unprecedented clinical success of Chimeric Antigen Receptor (CAR) T cell therapy in hematological malignancies has led researchers to study its role in solid tumors. Although, its utility in solid tumors especially in neuroblastoma has begun to emerge, preclinical studies of its efficacy in other solid tumors like osteosarcomas or gliomas has caught the attention of oncologist to be tried in clinical trials. Malignant high-grade brain tumors like glioblastomas or midline gliomas in children represent some of the most difficult malignancies to be managed with conventionally available therapeutics, while relapsed gliomas continue to have the most dismal prognosis due to limited therapeutic options. Innovative therapies such as CAR T cells could give an additional leverage to the treating oncologists by potentially improving outcomes and ameliorating the toxicity of the currently available therapies. Moreover, CAR T cell therapy has the potential to be integrated into the therapeutic paradigm for aggressive gliomas in the near future. In this review we discuss the challenges in using CAR T cell therapy in brain tumors, enumerate the completed and ongoing clinical trials of different types of CAR T cell therapy for different brain tumors with special emphasis on glioblastoma and also discuss the future role of CAR T cells in Brain tumors.
