Abstract: Therapeutic Artificial Organs (TAO) are extracorporeal life-saving devices that are used to bridge patients from emergency to surgery and recovery. This paper details the research on TAOs in Korea along with their development as the clinical devices. In the heart and lung area, the artificial heart system (AnyHeart) and the pulsatile heart-lung machine (T-PLS) have been developed. The T-PLS has been used in 400 patients. In the renal replacement area, the portable hemofiltration machine (C-PAK) and its applications in home renal treatment are currently being tested through animal experiments. The multiple applications of the biomaterials, the electromechanical system designs and their control, the telecommunication technology and medical technology have been combined in the research and development of life-saving artificial organs for critically ill patients.

102. Sublingual application of liquid nitrendipine does not result in critical hypotension in healthy volunteers under phosphodiesterase-5 inhibition

Abstract: Introduction: The introduction of phosphodiesterase-5 inhibitors as sildenafil, tadalafil, or vardenafil, has tremendously improved the treatment of erectile dysfunction. Patients with the common comorbidity of cardiovascular disease and erectile dysfunction, however, are at risk for critical hypotension in case of self-treatment of cardiac angina with nitrates after the intake of a phosphodiesterase-5 inhibitor. Methods: We evaluated the safety of 5 mg sublingual nitrendipine after pre-treatment of 8 healthy male volunteers (42.1 ± 9.6 yrs) with 20 mg tadalafil. Randomly four different protocols were compared using six hours blood pressure recordings: (1) baseline, (2) 20 mg tadalafil, (3) 5 mg nitrendipine, and (4) 20 mg tadalafil + 5 mg nitrendipine. Results: The blood pressure was not significantly affected by tadalafil. Nitrendipine lowered the systolic blood pressure significantly by -1.91 mmHg (p = 0.0079). The co-medication of 20 mg tadalafil + 5 mg nitrendipine lowered the blood pressure significantly by -2.86 mmHg (p < 0.0001). Abstract: Glycogen storage disease type I (GSD-I) is a group of autosomal recessive disorders with an incidence of 1 in 100,000. The two major subtypes are GSD-Ia, caused by a deficiency of glucose-6-phosphatase (G6Pase), and GSD-Ib, caused by a deficiency of glucose-6-phosphate transporter (G6PT). We report that a substantial improvement was achieved following several infusions of hepatocytes in a patient with GSD-Ib. Hepatocytes were isolated from the unused cadaveric whole livers of two donors. At the first transplantation, approximately 2 × 109 cells (2% of the estimated recipient's total hepatocytes) were infused. Seven days later 1 × 109 (1% of liver mass) cryopreserved hepatocytes from the same donor were infused, and an additional 3 × 109 (3% of liver mass) cells from the second donor were infused 1 month after the second transplantation. After the hepatocyte transplantation, the patient showed no hypoglycemic symptoms despite the discontinuation of cornstarch meals. Liver biopsies on posttransplantation days 20 and 250 showed a normal level of glucose-6-phosphatase activity in presolubilization assay that was very low before transplantation. This was the first and successful clinical hepatocyte transplantation in Korea. In this study, hepatocyte transplantation allowed a normal diet in a patient with GSD-Ib, with substantial improvement in their quality of life. Hepatocyte transplantation might be an alternative to liver transplantation and dietary therapy in GSD-Ib.

Abstract: We present far ultraviolet (FUV: 912 - 1750AA) spectral imaging observations recorded with the SPEAR satellite of the interstellar OVI (1032AA), CIV (1550AA), SiIV (1394AA), SiII* (1533AA) and AlII (1671AA) emission lines originating in a 60 x 30 degree rectangular region lying close to the North Galactic Pole. These data represent the first large area, moderate spatial resolution maps of the distribution of UV spectral-line emission originating the both the highly ionized medium (HIM) and the warm ionized medium (WIM) recorded at high galactic latitudes., Comment: 5 figures

Abstract: Purpose: To examine how a one-time bolus injection of radiographic contrast media (RCM; iodixanol 320 and iomeprol 350) into the left coronary artery of six pigs affects tissue oxygen tension in the artery's supply area (pO2 LAD) compared to a 0.9% sodium-chloride (NaCl) bolus. Material and Methods: Each animal received a total of three boluses: one iodixanol, one iomeprol, and one NaCl (10 ml each). The radiographic contrast media and NaCl boluses were randomly assigned, and pO2 profiles were recorded. Results: 26.7±16.4 s after iodixanol injection, pO2 LAD had declined by 3.5% from 42.2±5.6 mmHg to 40.7±5.9 mmHg ( P = 0.0357). After 53±16.7 s, the initial value was restored. The pO2 LAD was 41.9±7.4 mmHg before iomeprol injection, and, 303.3±58.9 s after injection, pO2 LAD had declined by 13.1% to 36.4±7.5 mmHg ( P = 0.0001). After 577±22 s, the initial value was restored. The bolus application of an isotonic NaCl solution resulted in no effect on pO2 LAD. Immediately after injection, it increased by 3%. In the supply area of the right coronary artery and the peripheral skeletal muscle, no effect of the RCM or NaCl on tissue oxygen tension was observed. Furthermore, no effect on tissue temperature, heart rate, systolic and diastolic blood pressure, or cardiac output per minute occurred. Conclusion: The injection of RCM in a coronary artery can result in a significant local contrast-medium-induced microcirculation disorder. The high viscosity of iodixanol led to a very short-term insignificant effect on the microcirculation, while iomeprol induced a slight time-delayed pO2 decrease, which might be caused by the rigidification of erythrocytes. In comparison to a previous study with iopromide, the influence of iodixanol and iomeprol on myocardial oxygen tension was markedly less pronounced.

Abstract: The decays $J/\psi\to p\bar{p}$ and $J/\psi\to n\bar{n}$ have been investigated with a sample of 225.2 million $J/\psi$ events collected with the BESIII detector at the BEPCII $e^+e^-$ collider. The branching fractions are determined to be $\mathcal{B}(J/\psi\to p\bar{p})=(2.112\pm0.004\pm0.031)\times10^{-3}$ and $\mathcal{B}(J/\psi\to n\bar{n})=(2.07\pm0.01\pm0.17)\times10^{-3}$. Distributions of the angle $\theta$ between the proton or anti-neutron and the beam direction are well described by the form $1+\alpha\cos^2\theta$, and we find $\alpha=0.595\pm0.012\pm0.015$ for $J/\psi\to p\bar{p}$ and $\alpha=0.50\pm0.04\pm0.21$ for $J/\psi\to n\bar{n}$. Our branching-fraction results suggest a large phase angle between the strong and electromagnetic amplitudes describing the $J/\psi\to N\bar{N}$ decay.

Abstract: Based on a data sample of 106 M $\psi^{\prime}$ events collected with the BESIII detector, the decays $\psi^{\prime}\ar\gamma\chi_{c0, 2}$,$\chi_{c0, 2}\ar\gamma\gamma$ are studied to determine the two-photon widths of the $\chi_{c0, 2}$ states. The two-photon decay branching fractions are determined to be ${\cal B}(\chi_{c0}\ar\gamma\gamma) = (2.24\pm 0.19\pm 0.12\pm 0.08)\times 10^{-4}$ and ${\cal B}(\chi_{c2}\ar\gamma\gamma) = (3.21\pm 0.18\pm 0.17\pm 0.13)\times 10^{-4}$. From these, the two-photon widths are determined to be $\Gamma_{\gamma \gamma}(\chi_{c0}) = (2.33\pm0.20\pm0.13\pm0.17)$ keV, $\Gamma_{\gamma \gamma}(\chi_{c2}) = (0.63\pm0.04\pm0.04\pm0.04)$ keV, and $\cal R$ $=\Gamma_{\gamma \gamma}(\chi_{c2})/\Gamma_{\gamma \gamma}(\chi_{c0})=0.271\pm 0.029\pm 0.013\pm 0.027$, where the uncertainties are statistical, systematic, and those from the PDG ${\cal B}(\psi^{\prime}\ar\gamma\chi_{c0,2})$ and $\Gamma(\chi_{c0,2})$ errors, respectively. The ratio of the two-photon widths for helicity $\lambda=0$ and helicity $\lambda=2$ components in the decay $\chi_{c2}\ar\gamma\gamma$ is measured for the first time to be $f_{0/2} =\Gamma^{\lambda=0}_{\gamma\gamma}(\chi_{c2})/\Gamma^{\lambda=2}_{\gamma\gamma}(\chi_{c2}) = 0.00\pm0.02\pm0.02$.

108. The potential effects of fresh water content on the primary production in the Chukchi Sea

Abstract: The in situ primary production rates and various environmental variables were investigated in the Chukchi Sea during the 3rd RUSALCA expedition, which was conducted in 2012, to identify the current status of primary production. A 13C-15N dual tracer technique was used to measure the daily primary production rates, which ranged from 0.02 to 1.61 g C m−2 d−1 (mean ± SD = 0.42 ± 0.52 g C m−2 d−1). The primary production rates showed large regional differences, with the southern region (0.66 ± 0.62 g C m−2 d−1) producing approximately five times as much as the northern region (0.14 ± 0.10 g C m−2 d−1), which was primarily due to the differences in phytoplankton biomasses induced by regional nutrient conditions. The primary production rates in the Chukchi Sea were averaged using data acquired during the three different RUSALCA expeditions (2004, 2009, and 2012) as 0.33 g C m−2 d−1 (SD = 0.40 g C m−2 d−1), which was significantly lower than previously reported rates. In addition to strong seasonal and interannual variations in primary production, recent decreases in the concentrations of major inorganic nutrients and chlorophyll a could be among the reasons for the recent low primary production in the Chukchi Sea because the primary production is mainly affected by nutrient concentration and phytoplankton biomass. The nutrient inventory and primary production appear to be largely influenced by the freshwater content (FWC) variability in the region due to the significant relationships between FWC, nitrate concentrations (r = 0.54, p < 0.05) and primary production rates (r = 0.56, p < 0.05). Moreover, we found highly significant relationships between the nutrient levels and the primary production rates (r = 0.75, p < 0.001). In conclusion, the primary production in the Chukchi Sea is primarily controlled by nutrient availability which is strongly related to the FWC variability. Our results imply that the predicted increase in freshwater accumulation might cause a decrease in primary production by lowering the nutrient inventory in the euphotic zone of the Chukchi Sea.

109. Mechanical Behavior of Potato and Sweet Potato under Impact and Compression Loading

Abstract: Mechanical properties of potato and sweet potato were measured under impact and compression loading. The test apparatus consisted of disgital storage oscilloscope and simple mechanisms which can apply compression and impact forces to potatoes and sweet potatoes. The mechanical properties could be measured while the tissues were ruptured in a very short period time less than 10 ms by impact loading. Rupture force, energy, and deformation were measured as mechanical properties of potatoes and sweet potatoes under impact and compression loading. Rupture forces under impact and compression loading were in the range of 84.1 to 93.7N and 128.9 to 132.2N for external tissues and 60.1 to 64.8N and 158.9 to 171.1N for internal tissues of potato and sweet potato, respectively. Compression speeds and drop heights for each test were in the range of 1.25 to 62.5mm/min and 8 to 24cm.

110. Steroid withdrawal in living donor renal transplant recipients using tacrolimus and cyclosporine: a randomized prospective study*

Abstract: Steroids have been a mainstay of immunosuppressive regimens in renal transplantation despite their adverse effects. The introduction of new immunosuppressant has improved the survival rates and prompted trials of steroid withdrawal. We conducted a randomized prospective study to compare steroid withdrawal at 6 months post-transplant between tacrolimus + mycophenolate mofetil (MMF) (FK group) versus cyclosporine A + MMF (CsA group). Steroid was withdrawn at 6 months post-transplant under the condition of no rejection episode proven by biopsy and maintenance of serum creatinine level2.0 mg/dl. Fourteen recipients were excluded because of acute rejection within 6 months or protocol violation. Steroid could be tapered off in 62 in FK group and 55 in CsA. Three cases in FK group and five in CsA had acute rejection within another 6 months after steroid withdrawal (P0.05). At 12 months, the incidence of post-transplant diabetes was 18.6% vs. 8.0% in FK and CsA group. And hypercholesterolemia was presented in 8.5% vs. 2.0%, hypertension in 47.5% vs. 56.0%, and serum creatinine level 1.18 +/- 0.24 mg/dl vs. 1.18 +/- 0.20 mg/dl, respectively (P0.05). Steroid withdrawal may be carried out successfully using both FK and CsA with MMF, but long-term follow-up is necessary.

111. Role of protein kinase C-η in cigarette smoke extract-induced apoptosis in MRC-5-cells

Abstract: Cigarette smoke (CS) is a major risk factor for emphysema, which causes cell death in structural cells of the lung by mechanisms that are still not completely understood. We demonstrated previously that CS extract (CSE) induces caspase activation in MRC-5 human lung fibroblasts, activated protein kinase C-η (PKC-η), and translocated PKC-η from the cytosol to the membrane. The objective of this study was to investigate the involvement of PKC-η activation in a CSE-induced extrinsic apoptotic pathway. We determined that CSE increases expression of caspase 3 and 8 cleavage in MRC-5 cells and overexpression of PKC-η significantly increased expression of caspase 3 and 8 cleavage compared with control LacZ-infected cells. In contrast, dominant negative (dn) PKC-η inhibited apoptosis in MRC-5 cells exposed to CSE and decreased expression of caspase 3 and 8 compared with control cells. Exposure to 10% CSE for >8 h significantly increased lactate dehydrogenase release in PKC-η-infected cells compared with LacZ-infected cells. Additionally, PKC-η-infected cells had an increased number of Hoechst 33342 stained nuclei compared with LacZ-infected cells, while dn PKC-η-infected cells exhibited fewer morphological changes than LacZ-infected cells under phase-contrast microscopy. In conclusion, PKC-η activation plays a pro-apoptotic role in CSE-induced extrinsic apoptotic pathway in MRC-5 cells. These results suggest that modulation of PKC-η may be a useful tool for regulating the extrinsic apoptosis of MRC-5 cells by CSE and may have therapeutic potential in the treatment of CS-induced lung injury.

112. Experimental Studies of Wide Flange Beam to Square Concrete-Filled Tube Column Joints with Stiffening Plates Around the Column

Abstract: The strength and stiffness of wide flange beams to rectangular tube column joints are reduced significantly if the connections are not reinforced. However, the reinforcement for the connections can be complex and increase the fabrication costs significantly because the tube columns are closed sections. This paper describes the force transfer mechanism and the cyclic performance of wide flange beams to square concrete-filled tube column joints reinforced with stiffening plates around the column. The first phase of this research program was to assess the force transfer mechanism at the joint using an analytical yield line method. An experimental program was conducted to verify the proposed model. The test results showed that the derived nominal strength equation provided a reasonable prediction. As the second phase of the research program, full-scale joint subassemblage tests were conducted to assess the cyclic performance of the joints. A total of seven specimens were tested under cyclic loading. Test results showed that total rotation of 0.04 rad which is required for special moment resisting frames could be obtained.

Abstract: Front the production on the farm to the consumer, agricultural products are subject to various physical treatments involving mechanical techniques and devices. It is essential to understand the physical laws governing the response of these biological materials so that the machines, processes, and handling operations can be designed fur maximum efficiency and the highest quality of the end products. A compression test system was developed to test the physical properties of fruits including apple, pear, and peach which may lead to a better understanding of the physical laws. The test system consisted of a digital storage oscilloscope and simple mechanism which can apply quasi-static compression to fresh fruits. Rupture force, energy, and deformation were measured at the five levels of compression speed from 1.25 to 62.5 mm/min for each internal and external tissues. Rupture forces for apple and pear were in the range of 42.2 to 46.2 N and 38.8 to 41.2 N for external and infernal tissues, respectively. Rupture forces fir peach external tissues were in the range of 48.2 to 54.0 N.

Abstract: Impact is one of the major cause of damage to fruits druing varios processes from the production on the farm to the consumer. The tissue of fruits are ruptured in a very short period time less than 10ms by impact loading. Mechanical behavior of fruits under impact loading can be analyzed better with high speed sampling data acquisition system and one of them is a digital storage oscilloscope. A impact test system was developed to test the physical properties of fruits including apple, pear, and peach which may lead to a better understanding of the physical laws. The test system consisted of a digital storage oscilloscope and simple mechanism which can apply impact force to fresh produce. Rupture force, energy, and deffrmation were measured at the five levels of drop heights from 4 to 24cm fur each internal and external tissues. Rupture forces for apple and pear were in the range of 72.9 to 87.7 N and 70.8 to 84.1 N for external and internal tissues, respectively. Rupture forces far peach external tissues were in the range of 43.4 to 65.0 N.

Abstract: We have investigated the Sb-induced reconstruction of the Si(112) surface using low energy electron diffraction (LEED) and scanning tunneling microscopy (STM). Upon Sb adsorption on the clean reconstructed Si(112) surface at 300°C, the Si(112)‐(111)1×1‐Sb surface was obtained. The present STM study gathered the following findings: The Sb-adsorbed Si(112) surface is composed of saw toothlike nanofacets, which are composed of the (557) plane and the (111) plane. The (557) plane consists of about five (111) planes with five times the width of bulk-terminated (111)1×1 unit cell and (001) plane. It is also about 8.5A in height and tilted at a 9.9° angle with respect to the basal plane (112). Based on observation, it appears that the Sb atoms on the (111) planes substitute for the topmost Si atoms on the ideal Si(111)1×1 surface. Based on the STM results, we suggest a structural model and discuss the reconstructing mechanism of nanofacets induced by Sb adsorption.

Abstract: Vapour pressures of butyl acetate + benzene or toluene or o- or m- or p-xylene were measured by static method at 298.15 ± 0.01 K over the entire composition range. The activity coefficients and excess molar Gibb's free energies of mixing (G E) for these binary mixtures were calculated by fitting vapour pressure data to the Redlich–Kister equation using Barker's method of minimizing the residual pressure. The G E values for the binary mixtures containing benzene are positive; while these are negative for toluene, ortho, meta and para xylene system over the whole composition range. The G E values of an equimolar mixture for these systems vary in the order: benzene > m-xylene > o-xylene > p-xylene > toluene

And it was known by the experiment with plasticine or aluminum material that an symmetric product with `c` channel section and the product with flanged `h` section could be bended because of asymmetric shape of billet.

Abstract: Thirty retail samples of heat-processed corn foods, i.e. corn flakes, corn-based breakfast cereals, tortilla chips and corn chips, were analysed for fumonisins--fumonisin B1 (FB1), fumonisin B2 (FB2) and hydrolysed FB1 (HFB1)--as well as for protein- and total-bound FB1. Bound (hidden) fumonisins cannot be detected by conventional analysis. Improved methods for the determination of bound FB1 were developed. The protein-bound FB1 was extracted with 1% sodium dodecylsulfate (SDS) solution. The SDS, which interfered with high-performance liquid chromatography (HPLC) analysis, was then separated from protein-bound FB1 by complexing with methylene blue followed by solvent extraction and hydrolysis with 2 N KOH. To measure total-bound FB1, the sample itself was hydrolysed with KOH. In both cases, clean-up was accomplished on an OASIS polymeric solid-phase extraction column and the bound fumonisins were determined by HPLC measurement of HFB1. Fourteen of 15 samples of corn flakes and other corn-based breakfast cereals analysed contained detectable levels of FB1 with a mean in positive samples of 67ng g(-1) (13-237 ng g(-1)). Two samples also had detectable levels of FB2 (21-23ng g(-1)). Bound FB1 was found in all samples; the mean protein-bound FB1 measured was 58 ng g(-1) (22-176 ng g(-1)) and the mean total-bound FB1 measured was 106 ng g(-1) (28-418 ng g(-1)), reported as FB1 equivalents after correction for recoveries of HFB1. There was an average of about 1.3 times more FB1 in the bound form compared with extractable FB1, and this was about twice as much as protein-bound FB1. Seven of the 15 samples of alkali-processed corn-based foods, such as tortilla chips and corn chips, contained FB1 and three contained HFB1 with means in measurable positive samples of 78 (48-134) and 29 (13-47) ng g(-1), respectively. Five of these alkali-processed corn foods contained bound FB1; the mean measurable protein-bound FB1 was 42 ng g(-1) (39-46 ng g(-1)) and the mean measurable total-bound FB1 was 100 ng g(-1) (54-209 ng g(-1)). HFB1 derived from bound FB1 in selected samples was confirmed by HPLC with mass spectrometry (MS).

Abstract: The short heterodimer partner (SHP) (NR0B2) is an orphan nuclear receptor whose function in pancreatic β-cells is unclear. Mitochondrial uncoupling protein (UCP2) in β-cells is upregulated in obesity-related diabetes, causing impaired glucose-stimulated insulin secretion (GSIS). We investigated whether SHP plays a role in UCP2-induced GSIS impairment. We overexpressed SHP in normal islet cells and in islet cells overexpressing UCP2 by an adenovirus-mediated infection technique. We found that SHP overexpression enhanced GSIS in normal islets, and restored GSIS in UCP2-overexpressing islets. SHP overexpression increased the glucose sensitivity of ATP-sensitive K+ (KATP) channels and enhanced theATP/ADP ratio. A peroxisome proliferator-activated receptor gamma (PPARγ) antagonist, GW9662, did not block the SHP effect on GSIS. SHP overexpression also corrected the impaired sensitivity of UCP2-overexpressing β-cells to methylpyruvate, another energy fuel that bypasses glycolysis and directly enters the Krebs cycle. KATP channel inhibition mediated by dihydroxyacetone, which gives reducing equivalents directly to complex II of the electron transport system, was similar in Ad-Null-, Ad-UCP2- and Ad-UCP2+Ad-SHP-infected cells. The mitochondrial metabolic inhibitor sodium azide totally blocked the effect of SHP overexpression on GSIS. These results suggest that SHP positively regulates GSIS in β-cells and restores glucose sensitivity in UCP2-overexpressing β-cells by enhancing mitochondrial glucose metabolism, independent of PPARγ activation.

Abstract: This study was carried out to estimate the effects of breed and environment such as sex, test station, test year, test season, parity, initial and final weight on average daily gain, age at 90kg, backfat thickness, feed efficiency, lean percent and selection index on the basis of the performance data collected from 25,790 pigs of Duroc, Yorkshire and Landrace breeds which were performance-tested at the Korea Swine Testing Station from 1991 to 2002. The results obtained in the study are summarized as follows; 1. The means of the major economic traits were estimated as 959.95±0.699g for average daily gain, 138.36± 0.072days for age at 90kg, 1.41±0.001cm for backfat thickness, 2.33±0.001 for feed efficiency, 56.71±0.018% for lean percent and 221.65±0.113 for selection index. 2. The effect of breed was statistically significant for all studied traits. Briefly, Duroc showed the best per- formance for the average daily gain and age at 90kg. Landrace had the best performances for the backfat thickness and lean meat percent. In feed efficiency and selection index, Yorkshire had a better score than other breeds. 3. The least-squares means of female and male for the traits studied were 923.05±1.289g and 974.53±0.856g for average daily gain, 139.74±0.145days and 137.21±0.097days for age at 90kg, 1.49±0.002cm and 1.39± 0.002cm for backfat thickness, 2.43±0.002 and 2.28±0.002 for feed efficiency, 56.43±0.034% and 56.81± 0.023% for lean percent and 211.37±0.194 and 224.61±0.129 for selection index. Therefore, males were superior to females for all traits examined. 4. The effect of test station was statistically significant for all traits except for selection index. Performances for age at 90kg, backfat thickness, feed efficiency and lean meat percent collected from Test station 2 were higher than those from Test station 1. However, Test station 1 showed better average daily gain. 5. The initial weight and final weight included as a covariate in this study had a significant influence on average daily gain, age at 90kg, backfat thickness, feed efficiency and selection index. From the absolute values of the estimated regression coefficients, it was inferred that the final weight had greater effect for the investigated traits than the initial weight.

This paper describes the two monitoring programs, how they are implemented, items of being measured, analytical techniques, quality control programs and R&D activities associated with the monitoring. Also, some of the latest data obtained by the monitoring programs are introduced.

Abstract: A survey for aflatoxin B1 (AFB1), fumonisin B1 and ochratoxin A (OTA) was conducted on 127 samples that included 30 food-grade barley, 32 barley foods, 18 food-grade corn and 47 corn foods, randomly collected during 1998-99 in Seoul, Korea. The presence of mycotoxins was analysed by direct competitive enzyme-linked immunosorbent assay (ELISA), and most of the positive samples from ELISA were confirmed using high-performance liquid chromatography (HPLC). Recoveries of AFB1 and OTA spiked at 10 ng g(-1) and FB1 spiked at 50 ng g(-1) were 106, 87 and 105% by ELISA, whereas those by HPLC were 80, 79 and 84%, respectively. Detection limits by ELISA for AFB1, FB1 and OTA were 1, 5 and 5 ng g(-1), and those by HPLC were 0.6, 35 and 1 ng g(-1). Naturally occurring AFB1, FB1 and OTA were found in 4/32 (12%), 2/32(6%) and 4/32 (12%) samples of barley foods with an average of 26, 16 and 9 ng g(-1), respectively. AFB1 and FB1 in corn foods were detected in 4/47 (8%) and 9/47 (19%) samples with the average being 20 and 74 ng g(-1), while no OTA was found in any corn foods samples. No AFB1, FB1 or OTA was detected in any of food-grade barley and corn samples. This is the first report on the natural co-occurrence of AFB1 and FB1 in barley and corn foods as well as on surveillance of OTA in Korea.

Abstract: A specially-built EMM (Electrochemical Micro Machining)/PECM (Pulse Electrochemical Machining) cell, a electrode tool filled with non-conducting material, a electrolyte flow control system and a small & stable gap control unit are developed to achieve accurate dimensions of spindle recesses. Two electrolytes, aqueous sodium nitrate and aqueous sodium chloride are investigated in this study. The former electrolyte with few pits on the surface of workpiece has better machine-ability than the latter one with many pits on the surface of workpiece. It is easier to control the machining depth precisely with pulse electrical current than direct electrical current. This paper also presents an identification method for the machining depth by in-process analysis of applied electrical current and interelectrode gap size. The interelectrode gap characteristics, including pulse electrical current, effective volumetric electrochemical equivalent and electrolyte conductivity variations, are analyzed using the model and experimental results.

Abstract: Previously, we prepared two different monoclonal antibodies (mAbs) against human 4-1BB (CD137): an agonistic mAb BBK-1 and an antagonistic mAb BBK-2. In this paper, we describe the molecular cloning of these two mAbs and present comparisons of their amino acid sequences. cDNAs encoding the heavy (H) and light (L) chains of the two mAbs were cloned by screening of cDNA libraries constructed from hybridomas secreting these mAbs. Comparisons of amino acid sequences of the two mAbs showed that, while the constant regions of the H and L chains were identical between the two mAbs, the variable region showed 45% identity in H chains and 48% identity in L chains. This suggests that these two mAbs recognize different epitopes of 4-1BB and may have different effects on the activity of 4-1BB.

Abstract: Background Heart transplantation is an established tool for the treatment of terminal heart failure. Hyperlipidemia is a common problem following heart transplantation and has been implicated as an additional risk factor in the development of transplant coronary artery disease (TxCAD). Therefore, heart recipients are commonly treated with inhibitors of cholesterol synthesis (HMG-CoA reductase inhibitors). However, these patients have an increased risk of developing rhabdomyolysis due to elevated concentrations of HMG-CoA reductase inhibitors under co-administration with the immunosuppressive cyclosporin A (CsA). Aim of the study Aim of our study was to obtain pharmacokinetic data on pravastatin whilst monitoring the safety and efficiency of the lipid lowering therapy in heart-transplant recipients under immunosuppression with CsA and to compare these data to those of a healthy control group. Subjects, materials and methods Eleven patients (30.2 +/- 12.3 months after transplantation) receiving immunosuppressive therapy consisting of cyclosporin A, prednisone and azathioprine with LDL cholesterol (LDL-C) concentrations exceeding 3.9 mmol/l and 8 control subjects were included into the study. In addition to the immunosuppressive therapy, the patients received a daily dose of 40 mg/day pravastatin for the first 8 days which was then reduced to 10 mg/day administered until Day 29. Blood was sampled for pharmacokinetic profiling (maximum concentration of the drug (Cmax), time to reach Cmax (tmax), area under the concentration vs. time curve (AUC(0-24h)), elimination half-life time (tcl)) and measurement of the parameters of clinical chemistry on Days 1, 8 and 29. The control group received a single dose of 60 mg pravastatin and the values of Cmax and AUC(0-24h) were normalized for a dose of 10 mg. Results Pravastatin 40 mg/day for 1 week in the patient group caused a significant reduction in total cholesterol (C) and LDL-C from 8.11 +/- 1.20 mmol/l and 5.88 +/- 1.15 mmol/l to 6.91 +/- 1.01 mmol/l and 4.72 +/- 1.05 mmol/l, respectively (p = 0.005 and p = 0.003). Triglycerides and HDL cholesterol (HDL-C) concentrations did not change significantly. Mean values for Cmax of pravastatin were 384.2 ng/ml, 392.0 ng/ml and 115.1 ng/ml in patients on Days 1, 8 and 29, respectively. After normalization for a dose of 10 mg, the corresponding values of C(max-DN10mg) and Cmax were 96.0 ng/ml, 98.0 ng/ml and 115.1 ng/ml on study Days 1, 8 and 29. These values were 7-8 times higher than the normalized value of C(max-DN10mg) for the control group (13.7 ng/ml). The corresponding values of AUC(0-24h) were 1228.2 ng/ml x h, 1214.1 ng/ml x h and 345.9 ng/ml x h in the patient group on study Days 1, 8 and 29 as well as 157.5 ng/ml x h in the control group prior to normalization. After normalization for a dose of 10 mg, the values of AUC(0-24h-DN10mg) in the patient group were approximately 12 times higher than those of the control group. However, no significant differences between the 2 groups were observed in tmax and tcl. Within the patient group, no significant increase in Cmax or AUC was found on Day 1 to Day 8. The results of creatine kinase (CK), lactate dehydrogenase (LDH), aspartate aminotransferase (ASAT) and alanine aminotransferase (ALAT) showed also no significant increase during the observation period. Conclusion Heart-transplant recipients treated with the HMG-CoA reductase inhibitor pravastatin generally show higher plasma concentrations of this drug than control subjects. However, our data suggest that the HMG-CoA reductase inhibitor pravastatin can be used effectively in these patients receiving the immunosuppressive cyclosporin A. The pharmacokinetic data obtained indicate that there is no significant cumulation of the drug following multiple dosages in spite of increased drug concentrations after a single oral dosage.

Abstract: A new method is proposed for effective analysis of laminated plates incorporating accurate through-the-thickness distribution of displacements, strains and stresses in the finite element formulation. It is a two-step analysis procedure. In the first step, displacements are obtained using a post-processing procedure based on the three-dimensional stress equilibrium equations and the thermoelasticity equations, from the results of FSDT finite element analysis. In the second step, the higher-order through-the-thickness distribution of displacements are reflected on the subsequent finite element analysis. The effectiveness of the present approach for the analysis of laminated plates is shown by numerical examples.

Abstract: We have previously synthesized a new cationic liposome that displays high efficiency and low toxicity, 3 beta[l-ornithinamide-carbamoyl] cholesterol (O-Chol), using solid-phase synthesis. In this study, O-Chol was applied to in vitro and in vivo models of ovarian cancer. Intraperitoneal gene delivery for peritoneal disseminated ovarian cancer in nude mice was achieved using a stable chloramphenicol acetyl transferase (CAT)-expressing ovarian cancer cell line (OV-CA-2774/CAT), which allowed us to quantify the exact tumor burden of organs. When luciferase and beta-galactosidase genes were used as reporter genes, O-Chol showed better efficiency than other commercial transfection reagents such as lipofectin, lipofectAMINE, DC-Chol, and FuGENE 6, both in vitro and in vivo. Moreover, the transfection efficiency of this new cationic lipid reagent remained high in serum-containing medium and under serum-free conditions. Furthermore, in vivo transfection with O-Chol showed high levels of gene expression specific to peritoneal tumor cells. Consequently, the O-Chol:DNA lipoplex appears to offer potential advantages over other commercial transfection reagents because of (1) its higher level of gene expression in vitro and in vivo; (2) its reduced susceptibility to serum inhibition; and (3) its highly selective transfection into tumor cells. These results suggest that the O-Chol:DNA lipoplex is a promising tool in gene therapy for patients with peritoneal disseminated ovarian cancer.

Abstract: The presence of zearalenone (ZEN) in Korean foods such as barley, barley-based foods, corn, corn-based foods, fruits and vegetables was investigated by direct competitive enzyme-linked immunosorbent assay (dcELISA) and the liquid chromatography (LC) method. A total of 164 samples was collected from Seoul, Korea, in 1998 and 1999 including 30 barley, 32 barley-based foods, 18 corn, 47 corn-based foods, 19 dried red pepper, nine dried jujube, and nine dried persimmon. Average recoveries of ZEN from barley and corn by dcELISA were 138 and 145%, and by LC were 91 and 83% respectively at spiking levels of 5-500 ngg(-1). The detection limit for ZEN was 3 ng g(-1) by dcELISA and 4 ngg(-1) by LC. From 164 samples analysed, ZEN was found by dcELISA in 35 samples (21%) with a mean level of 30 ng g(-1). Barley-based foods had the highest incidence of ZEN (38%) followed by barley (33%), corn-based foods (26%) and corn (22%). No ZEN was detected in any red pepper, jujube or persimmon samples. A high degree of correlation (r2 = 0.86) was observed between the 35 positive samples detected by dcELISA and 31 samples by LC.

Abstract: In this study, we identified a new tobamovirus from diseased Aporcactus flagelliformis cactus plants, named it rattail cactus necrosis-associated virus (RCNaV), and determined its complete genome sequence. The full RCNaV genome consisted of 6,506 nucleotides and contained four open reading frames coding for proteins of M(r) 128 kDa (3,441 nt), 185 kDa (4,929 nt), 55 kDa (1452 nt), 36 kDa (1,005 nt) and 19 kDa (513 nt) from the 5' to 3' end, respectively. The overall similarities for the four ORFs of RCNaV were from 32.5% to 64.1% and from 17.0% to 67.3% to those of the other tobamoviruses, at the nucleotide and amino acid level, respectively. Comparison of the coding and non-coding regions of the virus with those of other tobamoviruses showed that RCNaV is the most closely related to cactus mild mottle virus.

Abstract: We report on a study of the process e(+)e(-) -> pi(+/-) (D (D) over bar*)(-/+) at root s = 4.26 GeV using a 525 pb(-1) data sample collected with the BESIII detector at the BEPCII storage ring. A distinct charged structure is observed in the (D (D) over bar*)(-/+) invariant mass distribution. When fitted to a mass- dependent- width Breit- Wigner line shape, the pole mass and width are determined to be M-pole (3883: 9 +/- 1.5 (stat) +/- 4.2 dsyst__ MeV= c(2) and Gamma(pole) = (24: 8 +/- 3.3 (stat) +/- 11: 0 (syst)) MeV. The mass and width of the structure, which we refer to as Z(c)(3885), are 2 sigma and 1 sigma, respectively, below those of the Z(c)(3900) -> pi(+/-) J/psi peak observed by BESIII and Belle in pi(+)pi(-) J/psi final states produced at the same center- of- mass energy. The angular distribution of the pi Z(c)(3885) system favors a J(P) = J(P) = 1(+) quantum number assignment for the structure and disfavors 1(-) or 0(-). The Born cross section times the (D (D) over bar*) branching fraction of the Z(c)(3885) is measured to be sigma(e(+)e(-) -> pi(+/-)Z(c)(3885)(-/+)) x B(Z(c)(3885)-/+ -> (D (D) over bar*)(-/+) = (83.5 +/- 6.6 (stat) +/- 22.0 (syst)) pb. Assuming the Z(c)(3885) -> (D (D) over bar*)(-/+) signal reported here and the Z(c)(3900) -> pi J/psi signal are from the same source, the partial width ratio (Gamma(Z(c)(3885) -> D (D) over bar*)/Gamma(Z(c)(3900) -> pi J/psi)) = 6.2 +/- 1.1 (stat) +/- 2.7 (syst) is determined.

Abstract: We report on a study of the process e+ e- → π± (DD*)∓ at sqrt[s] = 4.26 GeV using a 525 pb(-1) data sample collected with the BESIII detector at the BEPCII storage ring. A distinct charged structure is observed in the (DD*)∓ invariant mass distribution. When fitted to a mass-dependent-width Breit-Wigner line shape, the pole mass and width are determined to be Mpole = (3883.9±1.5(stat)±4.2(syst)) MeV/c2 and Γpole = (24.8±3.3(stat)±11.0(syst)) MeV. The mass and width of the structure, which we refer to as Zc(3885), are 2σ and 1σ, respectively, below those of the Zc(3900) → π± J/ψ peak observed by BESIII and Belle in π+ π- J/ψ final states produced at the same center-of-mass energy. The angular distribution of the πZc(3885) system favors a JP = 1+ quantum number assignment for the structure and disfavors 1- or 0-. The Born cross section times the DD* branching fraction of the Zc(3885) is measured to be σ(e+ e- → π± Zc(3885)∓)×B(Zc(3885)∓ → (DD*)∓) = (83.5±6.6(stat)±22.0(syst)) pb. Assuming the Zc(3885) → DD* signal reported here and the Zc(3900) → πJ/ψ signal are from the same source, the partial width ratio (Γ(Zc(3885) → DD*)/Γ(Zc(3900) → πJ/ψ)) = 6.2±1.1(stat)±2.7(syst) is determined.

Abstract: Experiments were carried out to verify the effect of the solubility change of the Ca(OH)2 sorbent on the removal of SO2 in a lab-scale spray dryer system. First, to identify the solubility effect, HNO3 was added into Ca(OH)2 slurry to increase the solubility of Ca(OH)2 by neutralization. The improvement of SO2 removal by 5-10% occurred under the condition of both 50% relative solubility and constant pH of slurry. But SO2 removal efficiency decreased steeply when the relative solubility approached 100% and the pH of the slurry decreased rapidly. For the practical application of the solubility effect, NH4Cl and NH4OH were selected as additives. Ammonium chloride (NH4Cl) which could improved Ca(OH)2 solubility removed more SO2 by 5-10% than NH4OH which showed no change in solubility. The optimal result was obtained when the slurry was fed at the ratio of 0.25mol NH4Cl/mol Ca(OH)2. The Ca(OH),/NH4Cl sorbent could remove more SO2 and improve Ca(OH)2 utilization than Ca(OH)2 sorbent in the Ca/S stoichiometric range of 1.0-1.4. It was concluded that the increase of sorbent solubility made the improvement in SO2 removal efficiency and Ca(OH)2 utilization.

Abstract: In this study we investigated whether a disturbance in microcirculation is detectable in heart recipients with cardiac allograft vasculopathy (CAV) and severe hypercholesterolemia (n = 11) and in 7 heart recipients without CAV in comparison to patients with severe coronary artery disease (n = 49) and age-matched apparently healthy subjects (n = 100). For this purpose, the flow velocity of erythrocytes through cutaneous capillaries at the nail fold of the finger was measured under resting conditions. In addition, reactive hyperemia in the same capillaries after a 3-min ischemia was determined. Patients with CAV and severe lipid disorder showed a pathological reduction in mean capillary erythrocyte velocity under resting conditions with v(RBC) = 0.10 +/- 0.07 mm/s. The latter was significantly and relevantly lower than in patients with coronary three-vessel disease (v(RBC) = 0.46 +/- 0.35 mm/s). It was notable that under resting conditions temporary cessation of flow occurred in 8 of the 11 patients which did not occur in healthy subjects and rarely in patients with three-vessel disease (1 of 49 patients). In comparison to age-matched healthy subjects (v(max) = 1.46 +/- 0.52 mm/s), the patients with three-vessel disease showed a significant reduction in postischemic maximum erythrocyte velocity (v(max) = 0.85 +/- 0.55 mm/s), with a considerable shortening of the duration of reactive hyperemia. Patients with CAV demonstrated a total loss of postischemic reactive hyperemia (only 1 of the 11 patients presented a weak reactive hyperemia). Since no macroangiopathy was detectable in the upstream arm arteries, primary cutaneous microangiopathy can be assumed in patients with cardiac allograft vasculopathy and severe hypercholesterolemia.

139. The X-ray Diffraction Patterns of Bimetallic FexM100-x (M = Mo and W) Nanoclusters

Abstract: We produced the bimetallic FexMo100-x and FexW100-x nanoclusters with cluster size of 1.5−4.5 nm by thermally decomposing a mixture of two metal carbonyl vapor species and recorded the X-ray diffraction (XRD) patterns of them for the mole percent (x) range of 0 ≤ x ≤ 100. The XRD patterns changed from that of pure M (M = Mo and W) nanoclusters to that of pure Fe nanoclusters as x increased. The solubilities of bimetallic nanoclusters were 2−3 times higher than those of the corresponding bulk alloys, respectively. We observed that the cluster size decreased whereas the atomic level strain increased as the degree of alloying increased. We also observed that the cluster size effect on the broadening of the full width at half-maximum (fwhm) of the XRD peaks was stronger than the strain effect. Both cluster size decrease and atomic level strain increase of FexW100-x nanoclusters were larger than those of FexMo100-x nanoclusters.

Abstract: New blend hydrogels based on poly(vinyl alcohol) (PVA) and methylcellulose (MC) were prepared by crosslinking in an aqueous solution with glutaraldehyde (GA) in the presence of HCl. The state of the miscibility of the blend hydrogel films was examined over the entire composition range by differential scanning calorimetry (DSC) and dynamic mechanical analysis (DMA). Depressions of the melting and crystallization temperatures of PVA were observed with increasing MC content and crosslinking density via the DSC. The determination of the glass transition temperature of blend hydrogels by DMA indicated that they exhibit a higher miscibility than the non-crosslinked blends.

Abstract: We study e(+)e(-) -> pi(+) pi(-)h(c) at center-of-mass energies from 3.90 to 4.42 GeV by using data samples collected with the BESIII detector operating at the Beijing Electron Positron Collider. The Born cross sections are measured at 13 energies and are found to be of the same order of magnitude as those of e(+)e(-) -> pi(+) pi(-) J/Psi but with a different line shape. In the pi(+/-)h(c) mass spectrum, a distinct structure, referred to as Z(c)(4020) is observed at 4. 02 GeV/c(2). The Z(c)(4020) carries an electric charge and couples to charmonium. A fit to the pi(+/-)h(c) invariant mass spectrum, neglecting possible interferences, results in a mass of (4022.9 +/- 0.8 +/- 2.7) MeV/c(2) and a width of (7.9 +/- 2.7 +/- 2.6) MeV for the Z(c)(4020), where the first errors are statistical and the second systematic. The difference between the parameters of this structure and the Z(c) (4025) observed in the D*(D) over bar* final state is within 1.5 sigma, but whether they are the same state needs further investigation. No significant Z(c)(3900) signal is observed, and upper limits on the Z(c)(3900) production cross sections in pi +/- h(c) at center-of-mass energies of 4.23 and 4.26 GeVare set.

Abstract: The temperature dependence of the optical activity in the amorphous and therefore nonbirefringent solid state is used here to study the conformational properties of polyisocyanates and isotactic polyketones. Optically active polyisocyanates, which are not entirely amorphous in the solid state, were rendered amorphous by blending, while the glass transition temperature of the inherently amorphous polyketones was varied from 145 to 1 °C by terpolymerization of alternating styrenic/CO units with alternating 1-alkene/CO units. In both polymer systems, the optical activity changed significantly at temperatures only above Tg. This result, which was controlled for the contribution of degradation and epimerization of stereocenters in the polyketones, requires that the chiral optical properties of these polymers arise from long-range conformational properties of the chain that are impeded from changing with temperature only in the glassy state. Abstract: The occurrence of aflatoxin M1 (AFM1) in pasteurized milk and dairy products was investigated by using direct competitive enzyme-linked immunosorbent assay (ELISA) and high-performance liquid chromatography (HPLC). The recoveries of AFM1 from the samples spiked at levels between 5 and 500 pg/ml were 88.0-106.5% for pasteurized milk and 84.0-94.0% for yoghurt by ELISA. By HPLC, the recoveries were 103-120% for pasteurized milk and 87.0-93.0% for yoghurt. The limits of detection were found to be 2 pg/ml by ELISA and 10 pg/ml by HPLC. Among a total of 180 samples collected in Seoul, Korea, the incidence of AFM1 in pasteurized milk, infant formula, powdered milk and yoghurt was 76, 85, 75, and 83%, respectively, with a mean concentration of 18, 46, 200, and 29 pg/g, respectively, when determined by ELISA. These results obtained by ELISA were closely related to those by HPLC for AFM1 (r2 = 0.9783).

147. The analysis of void wave propagation in adiabatic monodispersed bubbly two-phase flows using an ensemble-averaged two-fluid model

Abstract: A three-dimensional two-fluid model has been developed using ensemble-averaging techniques. The two-fluid model was closed for adiabatic two-phase bubbly flows using cell averaging which accounted for the dispersed phase distribution in the region of the averaging volume. The phasic interfacial momentum exchange includes the surface stress developed on the interface which is induced by the relative motion of the phases. The surface stress has been obtained by treating the interface as an elastic spherical shell. A characteristic analysis revealed that the one-dimensional system of two-fluid conservation equations which were derived is well-posed over a range of void fractions with increased value of the interfacial pressure. The propagation of void fraction disturbances (i.e. the void wave) has also been analyzed by performing a dispersion analysis. The speed, stability and damping of the linear void waves have been obtained. To study finite amplitude void waves, the system of equations has been transformed into a moving coordinate system, and asymptotic solutions of the transformed nonlinear void wave equation have been obtained. The speed and the stability of different types of nonlinear void waves have been found to be sensitive to the closure relations of the two-fluid model. Among the different constitutive parameters, the interfacial pressure difference in the continuous phase and the void fraction gradient in the non-drag force are found to be the most significant in determining behavior of void waves in bubbly flows. The derived void wave speed agrees well with the void wave data of bubbly air–water flow.

148. Effects of oxygen on invertase expression in continuous culture of recombinant Saccharomyces cerevisiae containing the SUC2 gene

Abstract: The yeast SUC2 gene, cloned on a multicopy plasmid pRB58, was used to study the effect of oxygen on the invertase expression of the recombinant Saccharomyces cerevisiae. Glucose repression was not the only factor affecting the invertase expression. The results obtained from the single-stage continuous cultures under microaerobic conditions showed that invertase expression was also strongly dependent on oxygen availability, and moving from anaerobic to aerobic conditions led to a five-fold increase in specific invertase activity. However, the cell yields under anaerobic conditions were quite low compared to those under aerobic conditions. These opposite effects of oxygen on cell growth and gene expression offer a strategy for maximizing invertase productivity by a two-stage continuous culture. The first stage was operated at a low level of glucose, around 100 mg/l, under aerobic conditions in order to obtain a high yield of yeast biomass, and the second stage maintained anaerobic conditions with residual glucose levels of 50 mg/l to derepress and fully induce invertase expression. The two-stage continuous culture resulted in a 2.5-fold increase in invertase productivity over that of a single-stage continuous culture.

Abstract: A new device structure for a single-electron-tunnelling transistor with a dual-gate geometry has been fabricated based on the silicon-on-insulator structure prepared by SIMOX wafers. The split gate of the transistor is the lower-level gate and located above the inversion layer 2DEG active channel, which yields strong carrier confinement with a fully controllable tunnelling potential barrier. The transistor operates at low temperatures and exhibits single-electron tunnelling behaviour through a nano-size quantum dot. The Coulomb blockade oscillation is demonstrated at 15 mK and its periodicity is 16.4 mV in the upper gate voltage. For the nonlinear transport regime, Coulomb staircases are clearly observed up to four current steps in the range of 100 mV drain-source bias. The I-V characteristics near zero bias display a typical Coulomb gap due to the one-electron charging effect. From the width of the blockade regime the dot capacitance is estimated to be .
