123. Creation, Development, and Delivery of a New Interactive First-Year Introduction to Engineering Course.

Abstract

This complete evidence-based practice paper is focused on the initiation, development, and execution of the second component of a two-course sequence for first-year engineering students at a large, public university in the southeastern U.S. This sequence was developed with input from an engineering school-wide committee and represents a thorough restructuring of the school's first-year introduction to engineering program. The restructuring was designed to support the J.B. Speed School of Engineering's effort to have a common first year throughout all engineering majors. The new course sequence provides students with a more applicable and realistic exposure to the engineering experience. The second component of this sequence (Engineering Methods, Tools, & Practice II) exclusively takes place in the Engineering Garage (EG), a 15,000 ft2 makerspace that provides the course with individual classrooms in addition to laboratory-analogous workstations that further augments course delivery. Key logistical challenges, such as student safety and space accommodations, that needed to be overcome for full course realization are discussed. Each academic year, more than 500 first-year engineering students are exposed to this interactive course, which introduces students to fundamental engineering skills – including teamwork, design, project management, technical writing, critical thinking, programming, communication (including written, oral, and graphical), and an introduction to engineering research. The course includes extensive introductory design pedagogy coupled with project management; including two individual design challenges during the semester, and culminating in a team-based Cornerstone project that all students present at the end of the semester. For conveying key instructional topics to the students, a few select classes are held in the EG classroom(s), while additional instruction is delivered online via supplementary, instructor-created videos. The majority of the course meetings days occur in the EG makerspace and these meetings are activity-based. Course instruction and activities are designed to methodically expose students to the aforementioned skills, as well as other topics that pertain to engineering fundamentals. Many of the course deliverables have been designed to be dual-purpose, in that they build student understanding of essential engineering skills while also assisting progression towards Cornerstone project completion. The vast majority of in-class activities are team-based, with teams created very early in the semester to be diverse in both background and discipline. Key features of the course include various student-utilized hand tools, various software, Arduinobased circuitry, 3D printers, and hardware-based Programmable Logic Controllers (PLCs). This course has quickly become high-profile amongst students, faculty, and metropolitan industry alike. End-of-semester surveys suggest strong, positive student feedback pertaining to teamwork development. Other institutional entities such as the entrepreneurship school have partnered with course administrators to educate students on topics synergistic with engineering. New course features have been added with the help of local industries. Future work includes adding new Cornerstone projects with the help and cooperation of local industry partners. 124. Computational Instruction through PLCs in a Multi-Disciplinary Introduction to Engineering Course.

Abstract

This paper is focused on the inclusion of hardware-based Programmable Logic Controllers (PLCs) in a first-year engineering introduction course, Engineering Methods, Tools, & Practice II, at the University of Louisville's JB Speed School of Engineering. Each academic year, more than 500 first-year engineering students are exposed to this interactive course, which introduces students to fundamental engineering skills - including teamwork, design, project management, technical writing, critical thinking, programming, communication (including written, oral, and graphical), 3D printing, and an introduction to engineering research. Previously, this course culminated in a final project that combined construction and mechanical design of a windmill system utilizing data acquisition driven solely by Arduino programming and circuitry. The most recent iteration of the course added the instruction of PLCs through the collaboration with an industry partner prior to beginning the final project. The PLCs provide the students with a secondary form of computational methodology instruction. A key benefit of the PLC addition is the variation in logic and programming approach that is exposed to students. The inclusion of hardware based PLCs allows for the instruction of ladder logic, which is an alternative form of programming that gives students a different perspective. PLC control logic provides a sequential, real-time, and loop-based perspective versus the typical procedural and object-oriented approaches to which most students are limited in engineering education. The PLC addition is also beneficial in that it provides student exposure to systems and programming principles that are applicable within numerous industrial settings, which can also be useful when interviewing for their first co-operative internship during their sophomore year as many engineering disciplines rely on controlling systems and processes. Since the course continues to employ Arduino programming in addition to the PLC ladder logic programing, students are provided with a greater breadth of programming exposure and experience in integrating multiple hardware systems (PLC and Arduino) within their project. Abstract

Mucinous colorectal adenocarcinoma (CRC) is conventionally defined by extracellular mucin comprising >50% of the tumour area, while tumours with ≤50% mucin are designated as having a mucinous component. However, these definitions are largely arbitrary and comparisons of clinico-molecular features and outcomes by proportion of mucinous component are limited. A cohort of 1643 patients with stage II/III cancer was examined for tumour mucinous component, DNA mismatch repair (MMR) status, BRAFmutation and tumour infiltrating lymphocytes (TILs). Tumours with ≤50% mucinous component exhibited similar characteristics as mucinous tumours, including association with female gender, proximal location, high grade, TIL-high, defective MMR (dMMR) and BRAFmutation. Proportion of mucinous component did not stratify disease-free survival (DFS). In univariate analysis dMMR status, but not histological grade, stratified survival for mucinous and mucinous component tumours; however, in multivariate analysis dMMR status was not an independent predictor. BRAFmutation prognostic value depended on mucinous differentiation and MMR status, with poor prognosis limited to non-mucinous pMMR tumours (HR 2.61, 95% CI 1.69–4.03; p< 0.001). TIL status was a strong independent predictor of DFS in mucinous/mucinous component tumours (HR 0.40, 95% CI 0.23–0.67; p<0.001), and a superior predictor of prognosis compared with histological grade, MMR and BRAFmutation. Mucinous component and mucinous stage II/III CRCs exhibit clinico-molecular resemblances, with histological grade and BRAFmutation lacking prognostic value. Prognosis for these tumours was instead strongly associated with TIL status, with the most favourable outcomes in TIL-high dMMR tumours, whilst TIL-low tumours had poor outcomes irrespective of MMR status.

Abstract

Aberrant transcriptional silencing of genes by promoter hypermethylation is an important mechanism of tumour suppressor gene (TSG) inactivation in cancer. DNA hypomethylating agents such as 5-Aza-2’deoxycytidine (5-Aza-dC) are approved for the treatment of myelodysplastic syndrome and show promise in the treatment of solid tumours. These therapies are thought to exert their anti-tumour effects by reactivating hypermethylated genes. However, resistance to these drugs inevitably develops and this is associated with the silencing of genes initially re-expressed by therapy, termed gene re-silencing. The molecular basis of this is unknown, however understanding how it occurs may aid in the development of strategies to prevent or overcome resistance. We hypothesised that other chromatin modifications may silence gene expression in the absence of methylation. To study gene re-silencing following 5-Aza-dC exposure, a model gene (MLH1) and cell line (the colorectal carcinoma cell line RKO) were identified and validated. Gene reactivation was dependent on DNMT1 depletion and hypomethylation, nucleosome eviction and the acquisition of active histone marks at the MLH1 promoter. However, close inspection of the early stages of gene re-silencing showed that loss of expression occurred despite persistent hypomethylation. Instead, gene re-silencing coincided with the reassembly of nucleosomes at the MLH1 transcription start site, which preceded DNA re-methylation. Unexpectedly, changes in non-CpG methylation (methylation at CpT, CpA and CpC dinucleotides) were also observed, which we find may be attributable to alterations in the expression of the DNMT3A, DNMT3B or DNMT3L genes. Finally, despite the persistence of the repressive histone mark H3K9me3 following 5-Aza-dC treatment we find that, in this model, inhibitors of H3K9 methyltransferases do not delay gene re-silencing. Abstract

Cancer is caused by genetic and epigenetic changes resulting in altered patterns of gene expression. This study explored how somatic epigenetic events may predispose to the development of cancer and how genetic variation can underlie these events.Firstly, the role of constitutional methylation of the BRCA1 and BRCA2 genes in breast cancer development was investigated in BRCA-mutation negative cases of familial and early-onset breast cancer. No BRCA2 methylation was detected. Low level BRCA1 methylation was seen in 5/266 (2%) familial cases, 4/20 (20%) early-onset and 5/252 (2%) controls. No underlying sequence variants were found in regulatory regions of BRCA1 in methylation-positive cases. One familial case had two affected cousins who showed methylation (1-13%) – they all shared the same BRCA1 haplotype, suggesting a genetic basis for the epimutation. In another familial case, methylation detected in buccal cells (5%) was elevated in a cerebral metastasis (17%), suggesting a causative role in tumourigenesis in that case. However, methylation was found at similar low levels soma-wide and in the primary breast carcinoma in an early-onset case.Secondly, the role of the minor (T) allele of the MGMT enhancer rs16906252 C>T SNP, which correlates with MGMT promoter methylation, was studied in colorectal cancer (CRC). In a case-control study, the T allele was associated with increased risk of CRC showing MGMT-methylation, but not of developing CRC. Monoallelic methylation, mostly of the T allele, was observed in MGMT-methylated tumours retaining protein expression, producing discordant methylation and protein expression profiles. In peripheral blood and normal colorectal mucosa (NCM) of CRC cases and controls, low-level MGMT methylation was linked to the T allele.
