Mit Dichlorketen entstand als Hauptprodukt N-Dichloracetyl-2-phenyl-Δ2-piperidein. Die Cycloaddition von Ketenen an 5,6-Dihydro-1,3-oxazine ergab die sauerstoffhaltigen Cepham-Analogen 7 und 8, die Cycloaddition an 5,6-Dihydro-1,3-thiazine fuhrte zu den schwefelhaltigen Cepham-Analogen 9,10 und 11. The carbocyclic cepham analog 3 and N-diphenylacetyl-2-phenyl-Δ2-piperideine 4 as a side product were obtained by [2 + 2] cycloaddition of diphenyl ketene and 2-phenyl-Δ1-piperideine. Der pH-abhangige Einflus von Lysin auf die Hydrolyse von Acetylsalicylsaure zu Salicylsaure und auf die Bildung von Lysinderivaten der Acetylsalicylsaure wurde durch HPLC-Untersuchungen geklart. Die Bildung von e-N-Acetyl-lysin, α-N-Acetyl-lysin und wenig Diacetyl-lysin erfolgt rasch bei pH-Werten der Losung von 6 und hoher, sehr langsam jedoch bei dem fur gebrauchliche Injektionslosungen eingestellten pH-Wert 5. Das Auftreten des postulierten Salicyloyllysins in Acetylsalicylsaure-Lysin-Losungen wurde durch Vergleich mit der unabhangig synthetisierten Verbindung ausgeschlossen. Interactions of Acetylsalicyclic Acid and Lysine in Solution The influence of lysine on the hydrolysis, and on the formation of lysine derivatives, of acetylsalicylic acid is pH dependent and was elucidated by HPLC. There is rapid formation of e-N-acetyllysine, α-N-acetyllysine and of traces of diacetyllysine at pH values of 6 and higher, but very slow formation at pH 5 which is used for injectable solutions. Highly oriented samples of lipid and gramicidin A' (8:1 molar ratio) have been prepared with the samples extensively hydrated (approximately 70% water v/w). These preparations have been shown to be completely in a bilayer phase with a transition temperature of 28/sup 0/C, and evidence is presented indicating that the gramicidin is in the channel conformation. An estimate of the disorder in the alignment of the bilayers parallel with the glass plates used to align the bilayers can be made from the asymmetry of the nuclear magnetic resonances (NMR). Such an analysis indicates a maximal range of disorder of +-3/sup 0/. Uniformly /sup 15/N-labeled gramicidin has been biosynthesized by Bacillus brevis grown in a media containing /sup 15/N-labeled Escherichia coli cells as the only nitrogen source. When prepared with labeled gramicidin, the oriented samples result in high-resolution /sup 15/N NMR spectra showing 12 resonances for the 20 nitrogen sites of the polypeptide. The frequency of the three major multiple resonance peaks has been interpreted to yield the approximate orientation of the N-H bonds in the peptide linkages with respect to the magnetic field. The bond orientations are only partially consistent with the extant structural models of gramicidin.

Das Molekulargewicht verschiedener Gelatinepraparate (Knochen- und Hautgelatine; pI = 4,9 bzw. 8,1) wurde durch Sedimentation in der analytischen Ultrazentrifuge nach der Methode vonArchibald und der Sedimentationsgleichgewichtsmethode in verschiedenen Pufferlosungen und bei unterschiedlicher Temperatur bestimmt. Fur die Gelatine wurde jeweils in verdunnten Salzlosungen bei 40°C, in konzentrierten KSCN-Losungen bei 25°C und in 6 m Guanidinhydrochloridlosung bei 40°C das gleiche Molekulargewicht gefunden. Die Gelatine lag bei diesen Versuchsbedingungen molekulardispers vor. Die Gelatine wurde durch Ausfallen und durch Losungsmittel-Gradienten-Chromatographie fraktioniert. Von den Fraktionen wurden die Viskositatszahlen [η]40, die SedimentationskonstantenS 40, 0 und die MolekulargewichteM w mit Hilfe der Ultrazentrifuge bestimmt. Fur die Molekulargewichtsabhangigkeit der Viskositatszahlen und der Sedimentationskonstanten der untersuchten Fraktionen gelten die Beziehungen: $$\begin{gathered} [\eta ]_{40} = 7,75 \cdot 10^{ - 2} ml/g M_w^{0,51} \hfill \\ s_{40,w}^{0,w} = 0,61 Svedberg M_w^{0,17} . \hfill \\ \end{gathered} $$ Die fur Gelatine in der Literatur publizierten unterschiedlichen [η]-M w -Beziehungen werden auf den unterschiedlichen Verzweigungs- und Vernetzungsgrad der Praparate zuruckgefuhrt. Der kleine Exponent ders-M-Beziehung wird u. E. von der Durchspulbarkeit der untersuchten Gelatinemolekule verursacht. Bei der Gelchromatographie last sich die Molekulargewichtsabhangigkeit des Elutionsvolumens der untersuchten Gelatinefraktionen und von verschiedenen Proteinen durch die gleiche Eichkurve darstellen. Zur Erklarung dieses Befundes wird gezeigt, das die gelchromatographische Trennung beider Molekularten nicht nach dem hydrodynamischen Volumen [η] ·M, sondern mit groser Wahrscheinlichkeit bei den knauelformigen Gelatinemolekulen nach dem Tragheitsradius $$(\overline {r^2 } )^{1/2} $$ und bei den ellipsoidformigen Proteinmolekulen nach der grosen Halbachse erfolgt. Die Molekulargewichtsverteilung der Gelatine wurde durch Losungsmittel-Gradienten-Chromatographie und durch Gelchromatographie bestimmt. Die erhaltenen Verteilungskurven stimmen gut uberein. Die Losung der untersuchten Knochengelatine enthalt bei 40°C keine Aggregate, sondern Einzelmolekule im Molekulargewichtsbereich 4 · 103 bis 10 · 106.

Kinetische Untersuchungen der Reaktivitat von Aminopenicillinen belegen bei Ciclacillin eine sterische Hinderung der α-Aminogruppe der Phenylacetamido-Seitenkette. Vergleichende Haltbarkeitsuntersuchungen an Arzneizubereitungen von Ciclacillin und Ampicillin ergaben, das die aus der sterischen Hinderung resultierende hohere Stabilitat von Ciclacillin auch bei Injektions-Losungen, Suspensionen sowie eingeschrankt bei Pulvern und Tabletten nachweisbar ist. Influence of Steric Hindrance on the Degradation of Ciclacillin and Ampicillin Kinetic measurements of the reactivity of aminopenicillins demonstrated steric hindrance of the side chain amino group of ciclacillin. Comparative stability measurements of formulations of ciclacillin and ampicillin, showed that increased stability resulting from steric hindrance is also present in injectable solutions, suspensions and to a certain degree in powders and tablets.

Die Reaktionsmechanismen der strahlungslosen Desaktivierung von Molekeln in angeregten Elektronenzustanden und die zugrundeliegenden Prinzipien werden am Beispiel neuerer fluoreszenzspektroskopischer Ergebnisse an Losungen aromatischer Verbindungen (Merocyanine, Stickstoff-Heterocyclen, Saureamide usw.) dargestellt und erlautert.

Cytisin reagiert mit Brom unter Bildung eines Perbromids und eines Dibromids. NMR-, IR- und massenspektroskopische Untersuchungen ergeben fur das Dibromid die Struktur 1 eines 3,5-Dibromcytisins. Das NMR-Spektrum des Cytisins wird diskutiert. Fur das Cytisinperbromid wird auf Grund vergleichender Untersuchungen an den Perbromiden von Pyridin, α-Pyridon, Piperidin und Morpholin die Struktur 2 vorgeschlagen. The NMR spectrum of cytisine is discussed. Investigations of the perbromides of α-pyridone, pyridine, piperidine and morpholine gave structure 2 for cytinsine perbromide.

Die Trennung und die Eigenschaften des cis- und des trans-Isomeres werden beschrieben. Das trans-Isomer entspricht einem bicyclischen Azetidinon-(2), das durch Photolyse eines in einer Bamford-Stevens-Reaktion gewonnenen Diazoketons dargestellt wurde. 1. 1. For many years hysterectomy has been the safest of all major abdominal procedures and has been extraordinarily successful from the point of view of end results. 2. 2. For these reasons the indications for the operation have generally been extended, and it is now regarded as the preferable procedure whenever pelvic surgery is required in women in whom childbearing is no longer possible or in whom it would be impossible after the necessary pelvic surgery had been performed. 3. 3. This point of view has for many years been accepted in the Department of Gynecology of the Tulane University School of Medicine, which also, since 1950, has followed the policy of performing total hysterectomy routinely when the abdominal operation is indicated. This rule has not been violated since 1951. As the subtotal operation decreased in popularity, even before this policy went into effect, the vaginal technique was employed more frequently, particularly in Negro women. 4. 4. The policy of routine abdominal hysterectomy is justified by the end results, which include the elimination of symptoms referable to the retained cervix and the elimination of the possible development of cancer of the cervical stump. The policy is also completely justified by the safety of routine total abdominal hysterectomy. 5. 5. The good results achieved by this policy are attributable to a variety of factors. Chief among them are the practice of medical consultation in all cases of associated organic disease and the careful supervision of residents in training, who do the bulk of all gynecologic surgery. 6. 6. This contribution is based upon an analysis of the 6,280 hysterectomies performed on the Tulane Gynecological Unit of Charity Hospital of Louisiana at New Orleans between 1939 and 1954, inclusive, and in particular upon the 1,246 operations performed in the 1952–1954 period, when the policy was in effect of performing routine total abdominal hysterectomy whenever abdominal hysterectomy was indicated. The mortality rate for the whole period was 0.8 per cent and for the 1952–1954 period 0.4 per cent. A series of 639 hysterectomies performed in private practice over a sixteen and one-half year period is also briefly discussed from the standpoint of the safety of routine total abdominal hysterectomy.

Bei der Umsetzung von Pyrrylmagnesiumbromid mit Athylenoxid entstehen 2- und 3-Hydroxyathylpyrrol als Hauptprodukte, 1-Hydroxyathylpyrrol in Spuren. Das bisher unbekannte 3-Hydroxyathylpyrrol zeichnet sich ebenso wie die 1-Verbindung durch eine intramolekulare OH … π-Wasserstoffbindung aus und unterscheidet sich dadurch in charakteristischer Weise von 2-Hydroxyathylpyrrol, bei dem eine intramolekulare NH … O-Wasserstoffbindung nachgewiesen werden konnte. Pyrrolylmagnesium bromide reacts with ethylene oxide to give a mixture of nearly equal amounts of 2-and 3-hydroxyethylpyrrole as main product, to gether with a small amount of 1-hydroxyethylpyrrole. 3-Hydroxyethylpyrrole, a new compound, has intramolecular OH … π-hydrogen bonding as well as the 1-isomer. A characteristic difference is thus given between the 3-isomer and 2-hydroxyethylpyrrole, for which intramolecular NH … O-hydrogen bonding could be indicated.

In solutions of polar conjugated compounds in polar solvents a displacement of the absorption band towards the red has been observed with decreasing temperature. The initial displacement to the red of the fluorescent band becomes a displacement to the blue. The wave-number position of the fluorescence maximum occurs at a temperature for which the first time of relaxation of the dipole orientation of the solvent roughly agrees with the lifetime of the excited state. At lower temperatures the orientation equilibrium of the excited system does not fall completely into line with the emission of the fluorescence.

Cheap and easily constructed safety cells are described which can be used for absorption and fluorescence measurements over a wide range of temperature. Solvents are listed which can be used in these cells for studying the effect of solvents upon electronic spectra between 100 and 400°K.

Entscheidend sind die IR-Spektren: Fur die CO-Valenzschwingung und die CH-Valenzschwingungen des Azetidinon-(2)-Ringes und fur die CH2-Valenzschwingungen des kondensierten Piperidins konnen durch naherungsweise Berechnung und Vergleich mit Amiden Erwartungsbereiche angegeben werden. Eine auf den Azetidinon-(2)-Ring zuruckgehende UV-Absorption ist weder bei dem unsubstituierten noch bei den phenylsubstituierten Azabicyclooctanonen feststellbar. Die NMR-Spektren zeigten charakteristische chemische Verschiebungen fur die gespannten Protonen des Azetidinon-(2)-Ringes. There is conclusive evidence from the infrared spectra: Small frequency ranges of the CO and CH valency vibrations of the azetidin-2-one ring and of the CH2 valency vibrations of the fused piperidine result from calculation by approximation and from comparison with amides of higher and lower strain. A characteristic ultraviolet absorption having its origin in the azetidin-2-one rig could not be observed. In the strained protons of the azetidin-2-one ring are in accordance with the postulated structure.
