In this work, Bunocephalus doriae was cytogenetically analyzed. A karyotype with a diploid number of 2n= 50 comprising 6m, 10sm, 6st, and 28a (FN= 72) chromosomes was observed. The occurrence of an asymmetric karyotype with a large number of acrocentric chromosomes distinguishes this species from others the Order Siluriformes. An exclusive character observed is the first pair of subtelocentric as the largest chromosome pair of the complement. NORs detected using AgNO3 were located in the terminal regions, on the short arm of a subtelocentric chromosome pair (pair 11), in a secondary constriction. C-banding revealed heterochromatic centromeric regions on several chromosomes of the complement after C-banding. This is the first cytogenetic description of this species and the first cytogenetic report on a member of the family Aspredinidae.No presente trabalho, Bunocephalus doriae. foi analisado citogeneticamente. O número diploide encontrado foi 2n= 50 compreendendo 6m, 10sm, 6st, and 28a (NF= 72) cromossomos. A ocorrência de um cariótipo assimétrico com um grande número de cromossomos acrocêntricos distingue esta espécie das demais pertencentes à Ordem Siluriformes e foi observada como característica exclusiva a presença do primeiro par subtelocêntrico, sendo o maior do complemento. As NORs detectadas pelo AgNO3 foram observadas na região terminal do braço curto de um par cromossômico subtelocêntrico (par 11), em uma constrição secundária. A heterocromatina, após o bandamento C, foi visualizada em regiões centroméricas de vários cromossomos do complemento. Esta é a primeira descrição citogenética desta espécie e a primeira descrição de um membro da família Aspredinidae.

This study presents chromosomal data of Megalonema platanum from rio Tibagi, Paraná, Brazil and from rio Paraná, Argentina. The diploid number was equal 54 with karyotype composition of 24m+16sm+2st+12a in both populations. The AgNOR sites were detected in the terminal position of a submetacentric pair of the two analyzed populations, coinciding with secondary constrictions on the short arm of pair 15. CMA3 and FISH with 18S rDNA probe displayed fluorescent signals that correspond to the AgNOR sites and secondary constriction. The presence of a small acrocentric supernumerary chromosome can be observed in M. platanum from rio Tibagi, with centromeric heterochromatin. Others heterochromatic blocks were evidenced in the terminal position of some chromosome and one metacentric large chromosome pair, probably the first pair, showed an interstitial heterochromatin. In the population of the rio Paraná were still observed heterochromatic blocks in both ends in some chromosomes. This work brings for the first time cytogenetic date of M. platanum, which is a very rare species in the rio Paraná basin and may be endangered.Este estudo apresenta dados cromossômicos de Megalonema platanum do rio Tibagi, Paraná, Brasil e do rio Paraná, Argentina. O número diploide foi igual 54 com composição cariotípica de 24m+16sm+2st+12a em ambas populações. Os sítios AgNORs foram detectados na posição terminal de um par submetacêntrico das duas populações analisadas, coincidindo com constrição secundária no braço curto do par 15. CMA3 e FISH com sonda de DNAr 18S exibiram sinais fluorescentes que correspondem aos sítios AgNORs e à constrição secundária. A presença de um pequeno cromossomo supranumerário acrocêntrico foi observado em M. platanum do rio Tibagi, com heterocromatina centromérica. Outros blocos heterocromáticos foram evidenciados na posição terminal de alguns cromossomos e um par cromossômico submetacêntrico grande, provavelmente o primeiro par, mostrou heterocromatina intersticial. Na população do rio Paraná foram observados ainda blocos heterocromáticos em ambas regiões terminais em alguns cromossomos. Este trabalho mostra pela primeira vez dados citogenéticos de M. platanum, que é uma espécie muito rara na bacia do rio Paraná e pode estar ameaçada de extinção.

This study reports the description of the karyotype of Mugil incilis from Venezuela. The chromosome complement is composed of 48 acrocentric chromosomes, which uniformly decrease in size. Therefore, the homologues can not be clearly identified, with the exception of one of the largest chromosome pairs, classified as number 1, whose homologues may show a subcentromeric secondary constriction, and of chromosome pair number 24, which is considerably smaller than the others. C-banding showed heterochromatic blocks at the centromeric/pericentromeric regions of all chromosomes, which were more conspicuous on chromosomes 1, given the C-positive signals include the secondary constrictions. AgNO3 and fluorescent in situ hybridization (FISH) with 45S rDNA demonstrated that the nucleolus organizer regions are indeed located on the secondary constrictions of chromosome pair number 1. FISH with 5S rDNA revealed that the minor ribosomal genes are located on this same chromosome pair, near the NORs, though signals are closer to the centromeres and of smaller size, compared to those of the major ribosomal gene clusters. This is the first description of co-localization of major and minor ribosomal genes in the family. Data are discussed from a cytotaxonomic and phylogenetic perspective.Se presenta la primera descripción del cariotipo de Mugil incilis de Venezuela. El complemento cromosómico está compuesto por 48 cromosomas acrocéntricos uniformemente decrecientes en tamaño. Por lo tanto, los homólogos no pueden ser claramente identificados, con excepción de uno de los pares de mayor tamaño, clasificado como número 1, cuyos homólogos poseen una constricción secundaria subcentromérica, y el par de cromosomas número 24, considerablemente más pequeño que los otros. El bandeo-C reveló bloques heterocromáticos en las regiones centroméricas/pericentroméricas de todos los cromosomas, más conspicuas en el cromosoma 1 en el que las señales C-positivas se encuentra localizada precisamente en la constricción secundaria. La tinción con AgNO3 y la Hibridación Fluorescente in situ (FISH) con sonda 45S rDNA revelaron que las regiones organizadoras del nucléolo están ciertamente localizadas sobre la constricción secundaria del cromosoma número 1. FISH con 5S rDNA reveló que los genes ribosomales menores están ubicados en este mismo par cromosómico, en posición proximal a las NORs, aunque cercanas al centrómero y de menor tamaño en comparación con los clúster de genes ribosomales mayores. Ésta es la primera descripción de co-localización de genes ribosomales mayores y menores en la familia Mugilidae. Los datos se discuten bajo perspectivas citotaxonómicas y filogenéticas.

A new karyotype of Wiedomys pyrrhorhinus (Wied, 1821) is described, including G- and C-banding and Ag-NOR sites from specimens collected in the municipality of Morro do Chapéu, situated in the northern region of the Chapada Diamantina, state of Bahia. Karyological studies of W. pyrrhorhinus have shown a constant diploid number (2n) of 62 with two different numbers of autosomal arms (FNa): 86 and 90, respectively. The new karyotype revealed 2n = 62 and FNa = 104, being the higher autosomal number found so far for this genus. The X chromosome is a large acrocentric and the Y chromosome is a small acrocentric. The analysis of the karyotype morphology suggests that this new karyotype is more closely related to the previous karyotype with 2n = 62 and FNa = 90, described from Caetité, Bahia State. Further studies, comparing different populations of W. pyrrhorhinus, including molecular approaches, may help to better understand the geographical limits of each population and their phylogenetic relationships in the Caatinga biome

Karyotypes and testis structure of 14 species representing 9 genera (Latissus, Bubastia, Falcidius, Kervillea, Mulsantereum, Mycterodus, Scorlupaster, Scorlupella and Zopherisca) of the planthopper tribe Issini (Issidae) are presented. All the karyotypes are illustrated by meiotic and occasionally mitotic figures. The male karyotypes of most of the species analyzed are 2n = 26 + X, the exception being Falcidius limbatus, which has a karyotype of 2n = 24 + neo-XY. The latter is the first report of the neo-XY system in the family Issidae. The species studied are found to be similar in having NORs on the largest pair of autosomes, but differ significantly in the amount and distribution of C-heterochromatin along the chromosomes. In contrast to the conserved chromosome numbers, the highly variable follicle number in the testes suggests rapid evolution of the tribe Issini. On the basis of its specific follicle number, it is proposed that Zopherisca tendinosa skaloula Gnezdilov & Drosopoulos, 2006 be upgrade to a species: Z. skaloula stat. n. The cytological and taxonomic significance of results presented are discussed.

A recent substantial rearrangement of the 882 described eleutherodactyline frog species has considerably improved the understanding of their systematics. Nevertheless, many taxonomic aspects of the South American eleutherodactyline species remain unknown and require further investigation using morphological, cytogenetic and molecular approaches. In this work, the karyotypes of the Brazilian species Ischnocnema juipoca (Atibaia and Campos do Jordão, SP), Barycholos cf. ternetzi (Uberlândia, MG, and Porto Nacional, TO), and Pristimantis crepitans (Chapada dos Guimarães and São Vicente, MT) were analyzed using Giemsa staining, Ag-NOR labeling, and C-banding techniques. All individuals had a diploid number of 22 chromosomes, but the Fundamental Numbers were different among species. The herein described low chromosome number of Pristimantis crepitans is unique within this genus, suggesting that cytogenetically this species is not closely related either to its congeneric species or to Ischnocnema. In addition, karyotype differences, mainly in the NOR position, clearly distinguished the two Barycholos populations, besides indicating the existence of a so far undescribed species in this genus. A taxonomic review could clarify the systematic position of P. crepitans and verify the hypothetic new Barycholos species.

The subgenus Ceratochloa of the genus Bromus includes a number of closely related allopolyploid forms or species that present a difficult taxonomic problem. The present work combines data concerning chromosome length, heterochromatin distribution and nuclear genome size of different 6x, 8x and 12x accessions in this subgenus. Special attention is paid to the karyotype structure and genomic constitution of duodecaploid plants recently found in South America. Hexaploid lineages possess six almost indistinguishable genomes and a nuclear DNA content between 12.72 pg and 15.10 pg (mean 1Cx value = 2.32 pg), whereas octoploid lineages contain the same six genomes (AABBCC) plus two that are characterized by longer chromosomes and a greater DNA content (1Cx = 4.47 pg). Two duodecaploid accessions found in South America resemble each other and apparently differ from the North American duodecaploid B. arizonicus as regards chromosome size and nuclear DNA content (40.00 and 40.50 pg vs. 27.59 pg). These observations suggest that the South American duodecaploids represent a separate evolutionary lineage of the B. subgenus Ceratochloa, unrecognized heretofore.

Pseudacteon flies, parasitoids of worker ants, are being intensively studied as potentially effective agents in the biological control of the invasive pest fire ant genus Solenopsis (Hymenoptera: Formicidae). This is the first attempt to describe the karyotype of P. curvatus Borgmeier, P. nocens Borgmeier and P. tricuspis Borgmeier. The three species possess 2n = 6; chromosomes I and II were metacentric in the three species, but chromosome pair III was subtelocentric in P. curvatus and P. tricuspis, and telocentric in P. nocens. All three species possess a C positive band in chromosome II, lack C positive heterochromatin on chromosome I, and are mostly differentiated with respect to chromosome III. P. curvatus and P. tricuspis possess a C positive band, but at different locations, whereas this band is absent in P. nocens. Heterochromatic bands are neither AT nor GC rich as revealed by fluorescent banding. In situ hybridization with an 18S rDNA probe revealed a signal on chromosome II in a similar location to the C positive band in the three species. The apparent lack of morphologically distinct sex chromosomes is consistent with proposals of environmental sex determination in the genus. Small differences detected in chromosome length and morphology suggests that chromosomes have been highly conserved during the evolutionary radiation of Pseudacteon. Possible mechanisms of karyotype evolution in the three species are suggested.

The phylogenetic relationships of 15 taxa from Hystrix and the related genera Leymus (NsXm), Elymus (StH), Pseudoroegneria (St), Hordeum (H), Psathyrostachys (Ns), and Thinopyrum (E) were examined by using the Giemsa C-banded karyotype. The Hy. patula C-banding pattern was similar to those of Elymus species, whereas C-banding patterns of the other Hystrix species were similar to those of Leymus species. The results suggest high genetic diversity within Hystrix, and support treating Hy. patula as E. hystrix L., and transferring Hy. coreana, Hy. duthiei ssp. duthiei and Hy. duthiei ssp. longearistata to the genus Leymus. On comparing C-banding patterns of Elymus species with their diploid ancestors (Pseudoroegneria and Hordeum), there are indications that certain chromosomal re-arrangements had previously occurred in the St and H genomes. Furthermore, a comparison of the C-banding patterns of the Hystrix and Leymus species with the potential diploid progenitors (Psathyrostachys and Thinopyrum) suggests that Hy. coreana and some Leymus species are closely related to the Ns genome of Psathyrostachys, whereas Hy. duthiei ssp. duthiei, Hy. duthiei ssp. longearistata and some of the Leymus species have a close relationship with the E genome. The results suggest a multiple origin of the polyploid genera Hystrix and Leymus.

A cytogenetic study of bisexual species belonging to the genera Cirrorhynchus, Dodecastichus and Otiorhynchus is presented in order to confirm their taxonomic position. The karyotype characterization was accomplished by an analysis of mitotic and meiotic chromosomes after differential staining, namely by C-banding, silver impregnation, DAPI and CMA3. A review of the cytogenetic data for the tribe Otiorhynchini contributed to knowledge of chromosomal evolution in this group. An investigation of five of the species studied showed some similarities such as a sex chromosome system of "parachute type" (Xyp), the presence of 10 autosomal bivalents (2n = 22) and heterochromatin localized around centromeres. These observations are similar to those already described for Otiorhynchini species, and confirm the karyological conservatism of this weevil group. In contrast, another species Cirrorhynchus kelecsenyi has an additional four autosomal bivalents (n% = 14 + Xyp, 2n = 30), which differs considerably from the chromosomal homogeneity of the other genera. Karyotypic evolution in this species was achieved most probably by increasing the number of chromosomes by centric fissions, resulting in variation in the number of acrocentric chromosomes. DAPI-positive and CMA3-negative reactions of heterochromatic DNA in all the species studied suggest that it has an AT-rich composition. Impregnating chromosomes with silver nitrate reveals NORs on one pair of autosomes, and probably argentophilic material in the interspace between the X and y sex chromosomes. The karyological findings support the taxonomical revision of Otiorhynchini based on morphological characters.

Heterochromatin is one of the most dynamic components in the genome of species. Previous studies on the heterochromatin content and distribution in Heteroptera (insects with holokinetic chromosomes) have shown that the species belonging to the family Coreidae are interesting model organisms since they show very diverse C bands patterns. In the present work, we analyzed the C-band pattern in individuals of Holhymenia rubiginosa from different populations collected in different years. This species has the diploid karyotype 2n = 27/28 = 24 + 2m + X0/XX (male/female). C-bands are terminally, subterminally or interstitially located on 10-17 chromosomes and a remarkable heterochromatin heteromorphism is observed in the meiotic bivalents: in the presence/absence of bands, in the size of bands and number of bands. A heteromorphism is also inferred in the number of ribosomal genes from the difference in the fluorescent in situ hybridization signals between NOR-homologues. Chiasmata are generally located opposite to conspicuous C-bands, but in some bivalents chiasmata are also observed in close proximity to C-bands. Considering the striking variation in heterochromatin content between individuals and populations it is suggested that heterochromatin should be selectively neutral in H. rubiginosa.

The stingless bees Melipona rufiventris and M. mondury were analyzed cytogenetically by conventional staining with Giemsa, C-banding and sequential staining with the fluorochromes CMA3/DA/DAPI. Both species presented 2n = 18 and n = 9, except for one colony of M. rufiventris, in which some individuals had 2n = 19 due to the presence of a B chromosome. After Giemsa staining and C-banding the chromosomes appeared very condensed and presented a high heterochromatic content, making it difficult to localize the centromere and therefore to visualize the chromosomes morphology. The constitutive heterochromatin was located in interstitial chromosome regions covering most of the chromosomes extension and consisted mainly of AT, as shown by DAPI staining. The euchromatin was restricted to the chromosome extremities and was GC-rich, as evidenced by CMA3 staining. The B chromosome was CMA3-negative and DAPI-positive, a heterochromatic constitution similar to that of the A genome chromosomes.

Cytogenetic analyses performed in Cephalosilurus apurensis, Microglanis aff. cottoides and Pseudopimelodus bufonius revealed that the three species have 2n = 54 chromosomes: C. apurensis presented six metacentric (M), 28 submetacentric (SM), 14 subtelolocentric (ST), and six acrocentric (A) chromosomes, while M. aff. cottoides showed 10M, 32SM, 10ST and 2A, and P. bufonius had 12M, 30SM and 12ST. The nucleolus organizer regions (NORs) were present on the short arm of a middle-sized ST pair, identified as pair 19, in C. apurensis NORs were found on the short arm of a middle-sized ST (pair 23) and on the long arm of a middle-sized ST (pair 22) in M. aff. cotttoides and on the short arm of three middle-sized ST pairs, identified as pairs 9, 10 and 11, in P. bufonius. C-banding revealed a very small amount of constitutive heterochromatin in the chromosomes of all species, including the NORs. The occurrence of 2n = 54 in all species of the family Pseudopimelodidae and its absence among species of the closely related Pimelodidae and Heptapteridae may be important in identifying Pseudopimelodidae species.

Cytogenetic studies of the family Lycosidae (Arachnida: Araneae) are scarce. Less than 4% of the described species have been analyzed and the male haploid chromosome numbers ranged from 8+X 1 X 2 to 13+X 1 X 2 . Species formerly classified as Lycosa were the most studied ones. Our aim in this work was to perform a comparative analysis of the meiosis in " Lycosa " erythrognatha Lucas, " Lycosa " pampeana Holmberg and Schizocosa malitiosa (Tullgren). We also compared male and female karyotypes and characterized the heterochromatin of " L. " erythrognatha . The males of the three species had 2n = 22, n = 10+X 1 X 2 , all the chromosomes were telocentric and there was generally a single chiasma per bivalent. In " Lycosa " pampeana , which is described cytogenetically for the first time herein, the bivalents and sex chromosomes showed a clustered arrangement at prometaphase I. The comparison of the male/female karyotypes (2n = 22/24) of " Lycosa " erythrognatha revealed that the sex chromosomes were the largest of the complement and that the autosomes decreased gradually in size. The analysis of the amount, composition and distribution of heterochromatin with C-banding and staining with DAPI- and CMA 3 - showed that " Lycosa " erythrognatha had little GC-rich heterochromatin in the pericentromeric region of all chromosomes. In addition, the actual occurrence of the genus Lycosa in the Southern Hemisphere is discussed.

Euchroma gigantea was karyotypically studied using conventional staining, C-banding, silver nitrate staining and ribosomal fluorescent in situ hybridization (rDNA FISH). Broad wide autosomal polymorphism and a complex sex determination system were found in this beetle. Karyotype complements ranging from 2n = 32, X1X2X3Y1Y2Y3 to 2n = 36,X1X2X3Y1Y2Y3 were detected in the sample analyzed. Punctiform supernumerary chromosomes were present in the different karyotypes. The karyotypic evolution of Brazilian E. gigantea may have taken two directions, reduction in the diploid number of 2n = 36 to 24 through centric fusions or 2n = 24 to 36 due to chromosomal fissions. In addition, pericentric inversions were also involved. The complex multiple sex mechanism of this species seems to be old and well established since it is found in specimens from different populations. Small pericentromeric blocks of constitutive heterochromatin were located on the autosomes and terminal blocks were also found on some small pairs. The sex chromosomes showed larger constitutive heterochromatin blocks. Silver nitrate staining during prophase I of meiosis showed labeling of the sex chromosome chain. However, the rDNA sites could only be precisely determined by FISH, which permitted the identification of these ribosomal sites on chromosomes X1 and X2 of this species.

Cytogenetic analyses were performed in Cyphocharax modestus, collected at Paranapanema River and Tietê River (São Paulo State, Brazil). A karyotype with 2n = 54 chromosomes was observed in the animals from both Brazilian freshwater river systems. One to four B chromosomes were also detected in individuals from the Paranapanema River, which represents the probable first report of more than a single supernumerary element in a species of the Curimatidae group. C-banding revealed centromeric and telomeric heterochromatin blocks in several chromosomes of the normal karyotype complement of C. modestus. Moreover, while some B chromosomes were characterized by the complete absence of C-bands, others were totally heterochromatic. Although there was a prevalence of B chromosomes in males of C. modestus, at least one supernumerary element was found in males and/or females of several other populations of the species, which suggests that the presence of these chromosomes seems to represent a general trait of C. modestus. A possible origin of the described B chromosomes may be related to the occurrence of a chromosome non-disjunction followed by the loss of euchromatic segments, an event that should have occurred in chromosomes that present conspicuous centromeric heterochromatic blocks and even in chromosomes that lack C-bands in this region, resulting in small supernumerary elements.

O número cromossômico diplóide de Schlumbergera truncata e Schlumbergera x buckleyi, de indivíduos com diferentes tipos de coloração de pétalas, foi determinado usando-se pontas de raízes. A utilização de 8-hidroxiquinoleína 0,003 M à 36 °C por 3 horas possibilitou melhor separação cromossômica. Técnica de bandeamento C e de coloração Giemsa permitiram o estudo cariológico dessas espécies. O híbrido Schlumbergera × buckleyi (rósea) apresenta 2n = 22 cromossomos com fórmula cariotípica 16 M + 6 SM. Schlumbergera truncata, apresentando pétalas nas cores vermelha, branca e pink, possui 2n = 22 cromossomos, formulação cariotípica idêntica à de Schlumbergera × buckleyi, enquanto a planta com flores de coloração amarelada mostrou 2n = 34 cromossomos. A classificação cromossômica foi baseada no índice centromérico. Nas plantas que apresentam coloração vermelha, branca, pink e rósea nas pétalas, o melhor período de obtenção de metáfases corresponde ao período de florescimento. Schlumbergera truncata com flores amareladas apresenta dois picos anuais de divisão mitótica. Esses resultados dão suporte à um melhor entendimento da biologia no gênero Schlumbergera e auxiliam na classificação taxonômica nos casos onde apenas as características fenotípicas não são suficientemente confiáveis para a classificação das plantas no mesmo táxon.The diploid chromosome number of Schlumbergera truncata and Schlumbergera × buckleyi, in individuals with different types of petal color, was determined using root tips. The use of 8-hydroxyquinolein 0.003 M at 36 ºC provided better chromosome separation. C-banding technique and Giemsa coloration allowed the karyological study of these species. Schlumbergera × buckleyi hybrid (light pink) species has 2n = 22 chromosomes with karyotype formula 16M + 6SM. Schlumbergera truncata with red, white, and pink petals and 2n = 22 chromosomes has karyotype formula identical to Schlumbergera × buckleyi, while the plant with yellowish flowers has 2n = 34 chromosomes. Chromosome classification was based on the centromeric index. In plants with white, red, pink and light pink petal color, the best time to obtain metaphases is during flowering. Schlumbergera truncata with yellowish flowers has two annual peaks of mitotic division. These results give us a better understanding of the biology of the genus Schlumbergera and aid in taxonomic classification where phenotypic characteristics alone are not reliable enough to classify plants of the same taxon.

Differences in the karyology of two species, Centricnemus leucogrammus and Peritelus familiaris (Coleoptera: Curculionidae), were investigated in order to elucidate their taxonomic position of the taxa. Previously both species were placed in one genus whereas the latest taxonomic revision puts them in separate genera. Cytogenetic analysis of P. familiaris and C. leucogrammus showed significant differences in karyotype structure and confirmed their present taxonomic status. The diploid set of C. leucogrammus consists of 22 chromosomes with a fundamental number of arms (FN) of 45 and little variation in morphology and length. Peritelus familiaris has 24 chromosomes with FN of 47 and a more diverse karyotype. The karyotype evolution might have occured by centric fissions of autosomes. At pachytene and diplotene in spermatocytes, each chromosome bivalent showed a small band of pericentric heterochromatin. The bands were hardly visible or undetectable in other stages of spermatogenesis, namely mitotic metaphase, diakinesis, metaphase I and II. The nucleolar organizer regions (NORs) were active at premeiotic stages and early meiosis, but invisible at meiotic metaphase I, metaphase II, and mitotic metaphase. These results indicate the usefulness of cytogenetic methods in taxonomic evaluations.

We report several chromosomal traits in 11 species from 8 genera of the planthopper family Issidae, the tribes Issini, Parahiraciini and Hemisphaeriini. All species present a 2n = 27, X(0) chromosome complement known to be ancestral for the family. The karyotype is conserved in structure and consists of a pair of very large autosomes; the remaining chromosomes gradually decrease in size and the X chromosome is one of the smallest in the complement. For selected species, analyses based on C-, AgNOR- and CMA3- banding techniques were also carried out. By fluorescence in situ hybridization, the (TTAGG)n probe identified telomeres in all species, and the major rDNA loci were detected on the largest pair of autosomes. In most species, ribosomal loci were found in an interstitial position while in two species they were located in telomeric regions suggesting that chromosomal rearrangements involving the rDNA segments occurred in the evolution of the family Issidae. We conducted a cytogenetic study of four hyline frog species ( Dendropsophus elegans, D. microps, D. minutus and D. werneri) from southern Brazil. All species had 2 n = 30 chromosomes, with interspecific and intraspecific variation in the numbers of metacentric, submetacentric, subtelocentric and telocentric chromosomes. C-banding and fluorochrome staining revealed conservative GC-rich heterochromatin localized in the pericentromeric regions of all species. The location of the nucleolus organizer regions, as confirmed by fluorescent in situ hybridization, differed between species. Telomeric probes detected sites that were restricted to the terminal regions of all chromosomes and no interstitial or centromeric signals were observed. Our study corroborates the generic synapomorphy of 2 n = 30 chromosomes for Dendropsophus and adds data that may become useful for future taxonomic revisions and a broader understanding of chromosomal evolution among hylids. The karyotypes of Dryomys laniger and D. nitedula from Turkey were studied using C-banding and AgNOR staining. The standard karyotypes found in both species were fairly similar to previously published data (2n = 46, NF = 92 in D. laniger; 2n = 48, NF = 96 in D. nitedula). The C-banding pattern revealed a relatively small amount of heterochromatin in both karyotypes and C-heterochromatin was concentrated at centromeric areas of most autosomes and the X chromosome. Heterochromatin changes have apparently not been responsible for karyotypic divergences between the studied species. The AgNORs were recorded in the pericentromeric region of two autosome pairs in the complement of D. laniger, and at a single autosome pair of D. nitedula. The complement of D. laniger could be derived from that of D. nitedula after a tandem fusion of two autosomal pairs, and the assumed rearrangement also included the NOR region. Karyotypes are presented for 10 species of Gyrinus Geoffroy, 1762: G. minutus Fabricius, 1798, G. caspius Ménétriés, 1832, G. paykulli Ochs, 1927, G. distinctus Aubé , 1836 var. fairmairei Régimbart, 1883, G. marinus Gyllenhal, 1808, G. natator (Linnaeus, 1758), G. opacus Sahlberg, 1819, G. substriatus Stephens, 1869, G. suffriani Scriba, 1855, G. urinator Illiger, 1807 and for Orectochilus villosus (Müller, 1776) (Coleoptera: Gyrinidae). The 10 Gyrinus species have karyotypes comprising 13 pairs of autosomes plus sex chromosomes which are X0 (♂), XX (♀), with the X chromosomes the longest in the nucleus. O. villosus has 16 pairs of autosomes plus X0, XX sex chromosomes. The data obtained by Saxod and Tetart (1967) and Tetart and Saxod (1968) for five of the Gyrinus species are compared with our results. Saxod and Tetart considered the X chromosome to be the smallest in the nucleus in all cases, and this is considered to result from confusion arising from uneven condensation of some of the chromosomes. Small differences between the chromosomes of different Gyrinus species have been detected, but not between Greenland and Swedish populations of G. opacus, nor between typical G. distinctus from France and G. distinctus var. fairmairei from Kuwait. Different cytogenetic techniques including C-banding, base-specific fluorochromes and silver nitrate staining were used to compare the karyotypes of three species of bugs, representatives of the Neotropical genus Antiteuchus, namely A. mixtus, A. sepulcralis and A. macraspis. The species have holokinetic chromosomes which is typical of the Hemiptera, and their diploid chromosome number is 2n = 14 and they have an XX/XY (female/male) sex chromosome system. C-banding revealed telomeric blocks of constitutive heterochromatin in most chromosomes of these three species. In each species, CMA3-positive blocks were observed in the telomeric heterochromatin region of chromosome pair 1. This chromosome pair carried the nucleolar organizer region (NOR), which silver nitrate staining revealed is located in the telomeric heterochromatin. In contrast to the result obtained with CMA3, entire chromosome complement stained homogenously with DAPI, except for the telomeric region of pair 1, which remained unstained.

The Coreidae (Heteroptera) have holokinetic chromosomes and during male meiosis the autosomal bivalents segregate reductionally at anaphase I while the sex chromosomes do so equationally. The modal diploid chromosome number of the family is 2n = 21, with a pair of m-chromosomes and an X0/XX sex chromosome system. A 2n = 24/26 (male/female) and an X1X20/X1X1X2X2 sex chromosome system were found in Spartocera batatas (Fabricius). C-banding and fluorescent-banding revealed the presence of AT-rich heterochromatic bands medially located on all the autosomes, and one telomeric band on both the X1 and X2 chromosomes. This banding pattern differed from the telomeric heterochromatin distribution found in most other heteropteran species. The X1 and X2 chromosomes were intimately associated during male meiosis and difficult to recognize as two separate entities. Based on a comparison with the behaviour of sex chromosomes in other coreids and other heteopterans with multiple sex chromosomes it is suggested that the particular behaviour of X1 and X2 chromosomes in coreid species with multiple sex chromosome systems evolved as an alternative mechanism for ensuring the proper segregation of the sex chromosomes during meiosis.

Cytogenetic studies were performed on two sympatric species of Characidium, C. gomesi and C. cf. zebra, from the Grande River basin, Minas Gerais State, Brazil. Although both species had a chromosome number of 50 with a karyotype exclusively consisting of meta- and submetacentric chromosomes, interspecific diversity was detected concerning the size of the two first chromosome pairs of the karyotypes. Active nucleolus organizer regions (NORs) were located at the terminal position on the long arm of the 17th pair of C. gomesi and at subterminal position on the long arm of the 23rd pair of C. cf. zebra. For both species the fluorochrome CMA3 stained only the NOR-bearing pair of chromosomes. The heterochromatin pattern also showed some differentiation between these species restricted to the centromeric or pericentromeric region of C. cf. zebra and practically absent in C. gomesi. These data are discussed concerning chromosome diversification in this fish group.

Chromosomal studies were carried out on a population of the fish Hoplerythrinus unitaeniatus (jeju) from Prata river in the São Francisco river basin, Minas Gerais, Brazil using conventional Giemsa staining, C-banding, silver nitrate nucleolar organizer region (Ag-NOR) staining and fluorescent in situ hybridization (FISH) with 18S and 5S rDNA probes. We found a high degree of inter- and intra-individual variability with the identification of 2n = 50, 2n = 51 and 2n = 52 karyotypes at nearly the same frequency within the population. Intra-individual variation in chromosomal morphology and, consequently, karyotype formulae was also observed, chiefly in the specimens with 2n = 50 and 2n = 52 chromosomes. Ag-NORs and 18S rDNA sites also showed numerical and chromosomal variation similar to that found for the 5S rDNA sites. Some putative hypotheses are considered in order to explain these results.

The chromosomal localization of the As51 satellite DNA was identified by fluorescent in situ hybridization (FISH) in specimens of the characid fish Astyanax scabripinnis and Astyanax fasciatus, which are considered species complexes because of their extensive karyotypical and morphological variability. A conserved chromosomal distribution of the As51 satellite, coincident with distal C-banded segments was demonstrated. The alternative interstitial localization of this satellite DNA and possible alterations of its structure suggest that this sequence underwent quantitative, positional and structural variations, as the A. scabripinnis and A. fasciatus complexes diverged.

Cytogenetic studies were conducted on Mugil trichodon from Margarita Island, Venezuela. The species showed a karyotype 2n=48 with entirely acrocentric chromosomes (Arm number, NF=48). Chromosomes gradually decreased in size and did not allow a clear distinction of homologues, except for one marker pair, which showed a conspicuous secondary constriction. C-banding showed heterochromatic blocks restricted at the centromeric regions of all the chromosomes. Some were more obvious than others, with the exception of the chromosome pair that had a secondary constriction with entirely heterochromatic short arms. Sequential staining with Giemsa and AgNO3 demonstrated the conspicuous secondary constrictions corresponding to the NORs, and these had significant intraindividual size variations between both homologous chromosomes. The data obtained here support the contention that Mugilidae have a conservative chromosome macrostructure and reinforce the hypothesis that small structural chromosome rearrangements involving active NOR sites are the main cause of the karyotypic diversification seen in this group.

In the present study, five callichthyid species belonging to the subfamily Corydoradinae were karyotyped: three species of Aspidoras and two of Corydoras. The three species of Aspidoras had the same diploid number, 2n = 46 chromosomes, similar karyotypic formulae, with most chromosomes metacentric or submetacentric, single interstitial Ag-NORs and C-band positive segments mainly found in the centromeric position. The comparative analysis of cytogenetic data available for the genus Aspidoras and other species of Corydoradinae suggest that several events of centric fusion occurred in the origin of the species of Aspidoras. The two analyzed species of Corydoras showed high diploid numbers, 2n = 74 in C. sodalis and 2n = 90 in C. britskii. While C. sodalis exhibited single Ag-NORs and terminal, interstitial and centromeric C-band positive segments in almost all chromosomes, C. britskii showed multiple Ag-NORs and a small number of C-band positive segments found in the terminal position in one acrocentric (A) pair and in the interstitial position in one subtelocentric (ST) pair. The occurrence of high diploid numbers and many ST and A chromosomes are uncommon among the Corydoradinae, suggesting the occurrence of a high number of chromosome rearrangements, mainly centric fissions, in the origin of the Corydoras species studied.

Cytogenetical studies based on conventional coloration by Giemsa, C-banding and Ag-NOR were performed on 2 species of bats from the vespertilionid family: Lasiurus cinereus (Beauvois, 1796) and Lasiurus ega (Gervais, 1856). The 2n was 28 and FN was 48 in both species. The constitutive heterochromatin is located in centromeric regions in the two species and in the short arm of the subtelocentric X chromosome in L. ega. NORs were observed in the secondary constriction of the smaller autosome in both species.

The aim of this study was to describe mitotic and meiotic chromosomes of Cycloneda sanguinea using C-banding, fluorescent in situ hybridization (FISH) rDNA probes, and sequential FISH/Ag-NOR staining. The chromosome number was 2n = 18 + XX for females and 2n = 18 + Xy for males. The X chromosome was metacentric and the Y chromosome was very small. During meiosis, the karyotypic meioformula was n = 9 + Xy p, and sex chromosomes configured a parachute at metaphase I. At the beginning of pachytene, bivalents were still individualized, and sex chromosomes were associated end-to-end through the heteropycnotic region of the X chromosome. Later in pachytene, further condensation led to the formation of a pseudo-ring by the sex bivalent. All chromosomes showed pericentromeric heterochromatin. FISH and sequential FISH/Ag-NOR staining evidenced the location of the nucleolar organizer region in one pair of autosomes (at spermatogonial metaphase). During meiosis, these genes were mapped to a region outside the sex vesicle by FISH, although Xy p was deeply stained with silver at metaphase I. These results suggest that these argyrophilic substances are of a nucleolar protein nature, and seem to be synthesized by a pair of autosomes and imported during meiosis (prophase I) to the sex pair, during the association of the sex chromosomes.

Cytogenetic analysis based on the distribution of C-bands showed two groups of karyotypes in a Trypoxylon nitidum population from the Rio Doce Park, State of Minas Gerais, Brazil. One of these groups, that was identical to a previously described karyotype (n = 15; 2n = 30), had a stable chromosome number and was rich in acrocentric chromosomes, whereas the other had a variable chromosome number (n = 12 to 14; 2n = 25 to 28) and was rich in pseudo-acrocentric chromosomes. We propose a hypothesis explaining the dynamics of the modifications which occurred in the karyotype of this species, based on the minimum interaction theory of Imai et al. (1986, 1988, 1994) and on the chromosome rearrangements and heteromorphisms observed by us.

The stingless bees of the Partamona genus have been studied taxonomically, ecologically and behaviourally, but cytogenetic studies are still rare. The objective of this study was to obtain cytogenetic data to contribute to Partamona peckolti species characterization. Heterochromatin was localized in all chromosome pericentromeric regions but some blocks could be visualized on some large chromosomes arms. A large heterozygous DA-CMA3-positive band was observed on one large chromosome arm, but was completely absent when C banding was applied before fluorochrome staining, with only one small positive band being visualized. Sequential DA-CMA3-NOR staining of interphase nuclei provided coincident positive responses. This suggests that DA-CMA3-positive bands of P. peckolti correspond to nucleolar organizer regions, as previously confirmed for another Partamona species by FISH.

A cytogenetic analysis of thirteen specimens of Bryconamericus aff. Iheringii revealed a diploid number of 52 chromosomes and a karyotype of 8M+22SM+10ST+12A with a fundamental number (FN) of 92. The nucleolus organizer regions (NORs) were studied by means of AgNO3 and CMA3 staining as well as by FISH using an 18S rDNA probe. NORs were found to be located at the terminal position on the short arm of the submetacentric chromosome pair. C-banding showed strong telomeric and centromeric staining in the majority of the chromosomes, and a similar pattern was observed after treatment with AluI restriction enzyme

Using conventional staining with acetic orcein and C-banding techniques it was investigated constitutive heterochromatin chromosomal polymorphisms and the mitotic and the meiotic behavior of male and female chromosomes of Boophilus microplus (Canestrini, 1887). Some differences were detected in the population of southern Brazil as compared to the data of other authors for populations in other latitudes. The differences being mainly concerned with the distribution of constitutive centromeric heterochromatin and variation in the length of heterochromatic blocks in the pericentromeric regions of some chromosome pairs.

The cytogenetic analysis of Frieseomelitta dispar and F. francoi revealed the chromosome numbers 2n = 30 and n = 15 and a karyotypic formula 2K = 4M+2Mt+4A+20A M. The number of chromosomes observed was consistent with those reported for other Frieseomelitta species. The occurrence of the Mt chromosome and other features of the karyotype formulae suggest a close relationship between F. dispar, F. francoi and F. varia. Nevertheless, it was possible to differentiate the karyotypes of the species by DAPI/CMA3 staining, which revealed GC-rich regions on two chromosome pairs of F. dispar: one acrocentric and one pseudoacrocentric. In F. francoi, the same kinds of regions were observed on a pair of metacentrics and on a pair of acrocentrics. Our analysis also confirmed the chromosome number conservation in Frieseomelitta and suggests that infrequent pericentric inversion could constitute a synapomorphy for the group including F. dispar, F. francoi, and F. varia.

Saltator is a genus within family Thraupidae, the second largest family of Passeriformes, with more than 370 species found exclusively in the New World. Despite this, only a few species have had their karyotypes analyzed, most of them only with conventional staining. The diploid number is close to 80, and chromosome morphology is similar to the usual avian karyotype. Recent studies using cross-species chromosome painting have shown that, although the chromosomal morphology and number are similar to many species of birds, Passeriformes exhibit a complex pattern of paracentric and pericentric inversions in the chromosome homologous to GGA1q in two different suborders, Oscines and Suboscines. Hence, considering the importance and species richness of Thraupidae, this study aims to analyze two species of genus Saltator, the golden-billed saltator ( S. aurantiirostris) and the green-winged saltator ( S. similis) by means of classical cytogenetics and cross-species chromosome painting using Gallus gallus and Leucopternis albicollis probes, and also 5S and 18S rDNA and telomeric sequences. The results show that the karyotypes of these species are similar to other species of Passeriformes. Interestingly, the Z chromosome appears heteromorphic in S. similis, varying in morphology from acrocentric to metacentric. 5S and 18S probes hybridize to one pair of microchromosomes each, and telomeric sequences produce signals only in the terminal regions of chromosomes. FISH results are very similar to the Passeriformes already analyzed by means of molecular cytogenetics ( Turdus species and Elaenia spectabilis). However, the paracentric and pericentric inversions observed in Saltator are different from those detected in these species, an observation that helps to explain the probable sequence of rearrangements. As these rearrangements are found in both suborders of Passeriformes (Oscines and Suboscines), we propose that the fission of GGA1 and inversions in GGA1q have occurred very early after the radiation of this order. This work presents novel data that has allowed us to propose an evolutionary model for the karyotype of the European ground squirrel ( Spermophilus citellus , L. 1766) based on sex chromosome variants. Routine C- and G-banding, specific fluorochrome staining (CMA 3 , DAPI) and FISH experiments were used to characterize the pattern of chromosomal variation in populations of S. citellus in the Southeastern part of the Balkan Peninsula, the ancestral area of the species with the highest genetic diversity. The established diploid chromosome number of S. citellus is 2 n = 40. All autosomes are biarmed: 12 pairs of large to small submetacentrics, 5 pairs of large to small subtelocentrics, one medium-sized and one small pair of metacentrics. Three different morphologies are observed for both X and Y chromosomes. The X chromosomes are classified as acrocentric, subtelocentric, or submetacentric. In most of the individuals studied, the Y chromosome was dot-like, with the exception of a few presenting small or medium-sized acrocentrics and metacentrics. Blocks of heterochromatin of different sizes were found in the centromeric regions of all autosomes and X chromosomes, while all Y chromosome variants appeared to be entirely heterochromatic. CMA 3 fluorescence was bright in the majority of the C-positive regions and in the acrocentric X chromosome. These results were supported by DAPI staining, which showed that the CMA 3- positive sites were DAPI-negative and approximately of the same distinct sizes. In situ hybridization with the (TTAGGG)n probe produced a staining typical of telomeric repeats at the edges of all chromosomes. Notably, the Y chromosome showed stronger accumulation of telomeric repeats than that observed on other chromosomes. The nature of the C-, G- and fluorochrome staining of the sex chromosomes of S. citellus suggested that the amplification of constitutive heterochromatin and the pericentric inversions were likely sufficient to explain the differences between the morphology of sex chromosome. The observed sex chromosome variability reinforces the differences among the populations of the S. citellus in the Southeastern part of the Balkan Peninsula and correlate with specific geographical areas. The triatomines are insects vectors of the protozoan Trypanosoma cruzi, etiologic agent of Chagas disease. There are six species of greatest importance vector, out more, Triatoma infestans, Panstrongylus megistus, T. brasiliensis, T. sordida, Rhodnius neglectus and T. pseudomaculata. P. megistus presents great geographic distribution and has been collected in 22 Brazilian states. Intraspecific chromosomal variations have been detected in Triatominae. Thus, the present study aimed to analyze the chromosomal characteristics of P. megistus from seven different Brazilian states (Rio Grande do Norte, Rio Grande do Sul, Paraná, São Paulo, Minas Gerais, Bahia and Santa Catarina) by means of classical cytogenetic techniques. All insects analyzed showed the same chromosomal characteristics: 21 chromosomes, sex determination system the type X1X2Y, relative size of autosomes with little variation, relative size of the sex chromosomes (Y>X1>X2), chromocenter in meiotic prophase formed only by sex chromosomes and the presence of heterochromatin only in sex chromosome Y. Thus, this paper presents evolutionary data on the populations of P. megistus of Brazil demonstrating that populations from different Brazilian states have chromosomal homogeneity. The genus Pseudophoxinus Bleeker, 1860 is found in a wide range of habitats in central Anatolia, but it is not well known from a cytogenetic aspect. In this study the first karyotypic description of the spring minnows Pseudophoxinus crassus (Ladiges, 1960) and P. hittitorum Freyhof & Özulug, 2010 by means of conventional methods (Giemsa staining, C-banding, silver nitrate impregnation (Ag-NORs)) was performed. Both species are endemic and have restricted distributions in Central Anatolia. P. crassus and P. hittitorum have the same diploid chromosome number, 2n = 50, patterns of distribution of constitutive heterochromatin (CH), and localization of nucleolus organizer regions (NORs), but differ in their karyotypic formulae (KFs). The C-banding technique revealed clear pericentromeric blocks of CH in many chromosomes; Ag-NORs treatment revealed consistent positive signals at the end of the short arms of a submetacentric chromosome pair, likely homologous in both species. The article reports on the discovery of a new cytotype among a population of mole rats Nannopalax xanthodon inhabiting southeastern Anatolia, Turkey. Topics discussed include C-banding and NOR distribution pattern among N. xanthodon's population in the area and the characteristics of the new cytotype that has 2n=46 chromosomes. Also mentioned is the possibility of chromosomal variation between mole rat populations in the region.

Spermatogonial and primary spermatocyte chromosomes of Paragomphus lineatus (Selys) and Nepogomphus modestus (Selys) of family Gomphidae have been described on the basis of C-banding and NOR staining. Both the species possess 2n (♂) =23, n (♂) =12 without m chromosomes with XO(♂)/XX(♀) type sex determining mechanism. The sex chromosome is the largest element in the whole complement in Paragomphus lineatus but it is medium sized in Nepogomphus modestus. In Paragomphus lineatus, all the autosomal bivalents possess terminal C-bands while X chromosome is C-positive throughout the length but lightly stained in the centre. Similarly, terminal NOR bands are present on all the autosomal bivalents and X chromosome shows terminal NOR bands with large interstitial band. In Nepogomphus modestus, all the autosomal bivalents except one bivalent possess terminal C-bands while X chromosome represents large C-positive region on one end and small C-negative region on the other end. NOR staining shows terminal / non terminal NOR bands on some autosomal bivalents while X chromosome possesses dark NOR band only on one end. [ABSTRACT FROM AUTHOR]

Using C-banding and FISH methods, the karyotype of MC1611 induced mutant of bread wheat, which develop additional spikelets at a rachis node (trait 'supernumerary spikelets') was characterized. It was determined that the mutant phenotype is not associated with aneuploidy and major chromosomal rearrangements. The results of genetic analysis showed that supernumerary spikelets of the line are caused by a mutation of the single Bh-D.1 gene, influenced by the genetic background. The mutation causes abnormalities of inflorescence morphogenesis associated with the development of ectopic spikelet meristems in place of floral meristems in the basal part of the spikelets, causing the appearance of additional spikes at a rachis node. The mutant phenotype suggests that the Bh-D gene determines the fate of the lateral meristems in ear, which develops as floral meristem and gives rise to floral organs in wild-type inflorescences. In the bh-D.1 mutant, the floral meristem identity is impaired. The characterized mutant can be used in further studies on molecular genetic basis of development of wheat inflorescence. Scincoidea, the reptilian clade that includes girdled lizards, night lizards, plated lizards and skinks, are considered as a lineage with diversity in sex-determining systems. Recently, the hypothesis on the variability in sex determination in skinks and even more the absence of sex chromosomes in some of them has been rivalling. Homologous, evolutionary stable XX/XY sex chromosomes were documented to be widespread across skinks. However, sex determination in the other scincoidean families is highly understudied. ZZ/ZW sex chromosomes have been identified only in night lizards and a single species of plated lizards. It seems that although there are different sex chromosome systems among scincoidean lineages, they share one common trait: they are mostly poorly differentiated and often undetectable by cytogenetic methods. Here, we report one of the exceptions, demonstrating for the first time ZZ/ZW sex chromosomes in the plated lizard Zonosaurus madagascariensis . Its sex chromosomes are morphologically similar, but the W is clearly detectable by comparative genomic hybridization (CGH), suggesting that the Z and W chromosomes highly differ in sequence content. Our findings confirm the presence of female heterogamety in plated lizards and provides novel insights to expand our understanding of sex chromosome evolution in scincoidean lizards.
