Abstract

The purpose of this study was to evaluate the antioxidant and anticholinesterase activities of yellow and white bitter yams from South Western Nigeria using methanolic extraction and simulated gastrointestinal digestion models. The phenolic compounds in the bitter yam varieties were evaluated by high performance liquid chromatography with a diode array detector (HPLC-DAD). The total phenolic content of the bitter yams was measured by the Folin-Ciocalteu method, reductive potential by assessing the ability of the bitter yam to reduce FeCl3 solution, and the antioxidant activities were determined by the 2,2-diphenyl-1-picrylhydrazyl radical (DPPH·) scavenging activity, 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical cation (ABTS·+) scavenging activity, nitric oxide radical (NO·) scavenging ability, hydroxyl radical scavenging ability, and ability to inhibit Fe2+-induced lipid oxidation. The HPLC-DAD analysis revealed the presence of some phenolic compounds in the studied bitter yam varieties, with varying degree of quantitative changes after cooking. The antioxidant indices (total phenolic content, total flavonoid content, reducing power, DPPH· scavenging activity, ABTS·+ scavenging activity, and NO· scavenging activity) were higher in the simulated gastrointestinal digestion model compared to the methanolic extract, with the in vitro digested cooked white bitter yam ranking higher. Similarly, the in vitro digested yams had a higher inhibitory action against lipid oxidation compared to the methanolic extracts, with the cooked white bitter yam ranking high. The methanolic extracts and in vitro enzyme digests showed no acetylcholinesterase inhibitory abilities, while methanolic extracts and the in vitro enzyme digest displayed some level of butyrylcholinesterase inhibitory activities. Therefore the studied bitter yams could be considered as possible health supplements.

Abstract

The aim of this study was to investigate the toxicity of Syzygium jambos and Solanum guaraniticum leaf extracts, through Artemia salina lethality testing and cytotoxicity evaluations in rat spleen lymphocytes, using methyl tetrazolium (MTT), neutral red uptake (NRU), trypan blue dye exclusion, and lactate dehydrogenase (LDH) leakage assays, besides by their in vitro effects on acetylcholinesterase (AChE) activity. The LC50 calculated in the A. salina bioassay demonstrated that both extracts might well be toxic. Solanum guaraniticum presented cytotoxic effects against lymphocytes, as demonstrated by viable cells count reductions, NR uptake, and increased LDH leakage. Syzygium jambos appears to present immunomodulatory properties, increasing lymphocyte mitochondrial activity and inhibiting AChE activity. The results demonstrated the possible harmful effects of these vegetal preparations and may be helpful in therapeutic decisions and future studies with respect to the toxicology of these extracts, when used as phytotherapeutic medicines.

Abstract

The effect of drying on the phytoconstituents, antioxidative, and anti‐inflammatory properties of Bryophyllum pinnatum leaves was investigated. The phenolic constituents were characterized using HPLC‐DAD. The aqueous extraction was done and various assays (Inhibition of membrane stabilization, albumin Denaturation and heat‐induced hemolysis, malondialdehyde (MDA), and reduced glutathione (GSH) contents, as well as superoxide dismutase (SOD) activity), were carried out on human erythrocytes. The fresh portion (89.12 µg/ml) exhibited the highest potential to inhibit heat‐induced hemolysis compared to the standard drug—Diclofenac (91.51 µg/ml). Freeze‐dried sample showed the highest inhibitory potential on albumin denaturation ([Freeze‐dried‐330.72 µg/ml], [Diclofenac‐318.63 µg/ml]) and membrane destabilization ([Freeze‐dried‐331.93 µg/ml], [Diclofenac‐289.57 µg/ml]) when compared with Diclofenac. Similarly, the freeze‐dried sample showed the highest GSH and SOD level and lowest MDA level when human erythrocytes challenged with tertiary butyl hydroperoxide (tBHP) were treated with the extract. This study confirms the retention of a considerable quantity of bioactive constituents of plants when freeze‐dried. Practical applications: The ideal method of drying Bryophyllum pinnatum and possible anti‐inflammatory potential was investigated. Abstract

Objective. Due to the increased parasitic resistance, there is a need to explore alternative insecticides, including natural products, such essential oils. In this sense, the aim of this study was to evaluate, for the first time, the insecticidal and repellent effect of Cinnamomun zeylanicum (free and nanoemulsion forms) in vitro and in vivo. Materials and methods. For that, the insecticidal effect of the free form was tested at 1.0, 5.0 and 10%, while the nanoemulsion form was tested at 0.5, 1.0 and 5.0% against adult house flies, Musca domestica. For validation of the test, the flies were sprayed with the diluent (water and triton 10%) and with blank nanoemulsion (without C. zeylanicum essential oil). Moreover, in vivo test was performed using cows naturally infested with Haemotobia irritans. Results. The results demonstrated that cinnamon oil (10%) and the nanoemulsion (5%) were 100% effective against M. domestica after 90 minutes of exposure. The repellent effect was tested in vivo using 5% of cinnamon oil on Holstein cows naturally infested by Haemotobia irritans. The flies were counted at 0, 1, 2, 3, 9 and 24 h after cinnamon oil treatment. It was verified significant decrease (p<0.05) in the number of flies on cows sprayed with cinnamon in all times. Conclusions. Based on these results, C. zeylanicum showed insecticidal effect (in vitro) against domestic flies and repellent effect (in vivo) against the horn fly., Resumen Objetivo. Debido a la mayor resistencia parásita, hay una necesidad de explorar insecticidas alternativos, incluidos los productos naturales, tales como aceites esenciales. En este sentido, el objetivo de este estudio fue evaluar, para el primer equipo, el efecto insecticida y repelente de Cinnamomum zeylanicum (formas libres y nanoemulsión) in vitro e in vivo. Materiales y métodos. Para ello, el efecto insecticida de la forma libre se ensayó a 1.0, 5.0 y 10%, mientras que la forma nanoemulsión se ensayó a 0.5, 1.0 y 5.0% frente a las moscas domésticas adultas, Musca domestica. Para la validación de la prueba, las moscas fueron rociados con el diluyente (agua y triton 10%) y con nanoemulsión en blanco (sin aceite esencial de C. zeylanicum). También, fueron realizados pruebas in vivo usando vacas naturalmente infestadas con Haemotobia irritans. Resultados. Los resultados demostraron que el aceite de canela (10%) y la nanoemulsión (5%) fueron 100% eficaz contra M. domestica después de 90 minutos de exposición. El efecto repelente se ensayó in vivo usando 5% de aceite de canela en vacas Holstein, naturalmente infestados por Haemotobia irritans. Se contaron las moscas a las 0, 1, 2, 3, 9 y 24 h después del tratamiento el aceite de canela. Se verificó disminución significativa (p<0.05) en el número de moscas vivas de vacas canela pulverizada en todo momento. Conclusiones. En base a estos resultados, C. zeylanicum mostró un efecto insecticida (in vitro) contra las moscas domésticas y efecto repelente (in vivo) contra la mosca de los cuernos.

Abstract

Lantana camara, the widely studied species, and L. montevidensis, the less studied species of the genus Lantana are both used in traditional medicine for the same purpose (anti-asthma, anti-ulcer, anti-tumor, etc). However, little is known about the toxicity of L. montevidensis and there is limited information on its chemical constituents. Here, we investigated for the first time the genotoxicity and cytotoxicity of the ethanolic (EtOH) and aqueous extracts from the leaves of Lantana montevidensis in human leukocytes, as well as their ossible interaction with human erythrocyte membranes in vitro. The antioxidant activities of both extracts were also investigated in chemical and biological models. Treatment of leukocytes with EtOH or aqueous extracts (1-480 µg/mL) did not affect DNA damage index, but promoted cytotoxicity at higher concentrations (240-480 µg/mL). Both extracts did not modify the osmotic fragility of human erythrocytes. The extracts scavenged DPPH radical and prevented Fe2+-induced lipid peroxidation in rat’s brain and liver homogenates, and this was likely not attributed to Fe (II) chelation. The HPLC analysis of the extracts showed different amounts of polyphenolic compounds (isoquercitrin, gallic acid, catechin, ellagic acid, apigenin, kaempferol, caffeic acid, rutin, quercitrin, quercetin, chlorogenic acid, luteolin) that may have contributed to these effects. Abstract

We aimed to evaluate the antiproliferative and genotoxic potential of aqueous extracts prepared with fresh and dry leaves of Rubus sp. on meristematic divisions of Allium cepa root tips, as well as the phenolic compounds in the extracts. Aqueous extracts were prepared by infusion of blackberry fresh leaves collected at Santa Maria municipality, Rio Grande do Sul state (RS), southern Brazil (at concentrations 20 and 60 g.L-1), and dry leaves collected at Nova Palma municipality, RS (at concentrations 10 and 30 g.L-1). Distillated water was used as negative control while 1% glyphosate solution was used as positive control. Phenolic compounds were determined by High Performance Liquid Chromatography (HPLC). We analyzed a total 4000 cells per treatment. Statistical analysis was performed using Tukey and Scott-Knott tests (p < 0.05). Both fresh- and dry-leaf aqueous extracts at both concentrations inhibited cell proliferation, showing no significant difference from one another yet showing significant difference from the control treatments. HPLC analysis revealed that dry-leaf aqueous extracts show higher total amount of phenolic compounds. Quercetin was the compound found in highest amounts in dry-leaf aqueous extracts while caffeic acid was the predominant compound in fresh-leaf aqueous extracts. Thus, we conclude that both fresh- and dry-leaf aqueous extracts of blackberry have intense antimitotic activity but no genotoxic activity in the in vivo plant test system., O presente trabalho teve o propósito de avaliar o potencial antiproliferativo e genotóxico de extratos aquosos de folhas frescas e secas de Rubus sp. sobre a divisão meristemática de pontas de raízes de Allium cepa, bem como os compostos fenólicos presentes nos referidos extratos. Os extratos aquosos foram preparados por infusão de folhas frescas de amoreira coletadas no município de Santa Maria – RS e de folhas secas em Nova Palma – RS, em duas concentrações. Água destilada foi usada como controle negativo e glifosatos 1% como controle positivo. Os compostos fenólicos foram determinados por cromatografia líquida de alta eficiência (CLAE). Foram analisadas 4000 células por tratamento. A análise estatística foi realizada pelos testes Tukey e Scott-Knott (p < 0,05). Os extratos aquosos, tanto de folhas frescas como de folhas secas, nas duas concentrações inibiram a proliferação celular, não diferindo significativamente entre si, mas diferindo dos controles. Por CLAE, o extrato aquoso de folhas secas apresentou maior quantidade total de compostos fenólicos. A quercetina foi o elemento encontrado em maior quantidade no extrato de folhas secas e o ácido cafeico foi predominante no extrato de folhas frescas. Assim, conclui-se que os extratos aquosos de folhas frescas e secas de amoreira-preta tem intensa ação antimitótica, mas não genotóxica pelo sistema teste in vivo vegetal.

Abstract

Lantana camara, the widely studied species, and L. montevidensis, the less studied species of the genus Lantana are both used in traditional medicine for the same purpose (anti-asthma, anti-ulcer, anti-tumor, etc). However, little is known about the toxicity of L. montevidensis and there is limited information on its chemical constituents. Here, we investigated for the first time the genotoxicity and cytotoxicity of the ethanolic (EtOH) and aqueous extracts from the leaves of Lantana montevidensis in human leukocytes, as well as their possible interaction with human erythrocyte membranes in vitro. The antioxidant activities of both extracts were also investigated in chemical and biological models. Treatment of leukocytes with EtOH or aqueous extracts (1-480 µg/mL) did not affect DNA damage index, but promoted cytotoxicity at higher concentrations (240-480 µg/mL). Both extracts did not modify the osmotic fragility of human erythrocytes. The extracts scavenged DPPH radical and prevented Fe2+-induced lipid peroxidation in rat's brain and liver homogenates, and this was likely not attributed to Fe (II) chelation. The HPLC analysis of the extracts showed different amounts of polyphenolic compounds (isoquercitrin, gallic acid, catechin, ellagic acid, apigenin, kaempferol, caffeic acid, rutin, quercitrin, quercetin, chlorogenic acid, luteolin) that may have contributed to these effects. Abstract

Several species of the genus Ceiba (Malvaceae) are ethnopharmacologically used. Thus, this study aimed to investigate the in vitro beneficial properties of the aqueous stem bark extract of Ceiba speciosa. The extract presented a great amount of phenolic compounds (117.4 ± 6.2 mg GAE/g). The antioxidant activity was assessed by DPPH (IC50 = 42.87 μg/mL), ORAC (2351.17 μmol TE/g) and FRAP (235.94 μM FeSO4/g) methods. In addition, the extract reduced MCF-7 cell viability as assessed by MTT. However, it prevented mitochondrial membrane depolarization and reduced caspase-9 activity induced by hydrogen peroxide. In conclusion, these findings indicate the extract is an excellent source of natural antioxidants and is able to protect ROS-induced cell death. Therefore, C. speciosa extract may possess beneficial properties for application in pharmaceutical industry as an antioxidant.
