Context: Alopecia areata (AA) is a common form of localized, non-scarring hair loss. The pathogenesis of the disease is unknown. Previous evidence suggested the involvement of Th2 cytokines in disease pathogenesis. Aim: To determine serum level of total IgE, this is mainly influenced by Th2 cytokines, in Egyptian patients with AA. Materials and Methods: Fifty subjects with AA (28 males and 22 females) were selected from Dermatology Outpatient Clinic, Menoufiya University Hospital from February 2012 to December 2012. Subjects with other conditions that might elevate serum IgE were excluded from the study. Fifty age- and sex-matched healthy subjects were selected as a control group. Venous blood samples were taken from cases and controls for measurement of total serum IgE by enzyme-linked immunosorbent assay. Skin biopsy was taken from every case from an active area of hair loss. Results: Total serum IgE was elevated in 27 (54%) cases. Its values among patients ranged from 13.5 IU/ml to 780 IU/ml. There was a statistically significant difference between cases and controls with regard to mean value of serum IgE (P < 0.05). Mean value of IgE did not vary significantly with disease severity, patients' age, patients' gender, disease duration, site of patients' gender, and positive family history of AA. No correlation was found between serum IgE levels and histopathological changes detected in examined cases. Conclusions: Total serum IgE is elevated in AA. Background: Lipoid proteinosis (Urbach-Wiethe disease) is a rare progressive autosomal recessive disorder, characterized histologically by deposition of periodic acid Schiff-positive, diastase resistant, hyaline-like material into the skin, upper aerodigestive tract, and internal organs. Main observation: We report two cases of lipoid proteinosis. A 2-year-old girl presented with vesiculobullous skin lesions on her face, trunk, extremities and scalp, inability to protrude the tongue and hoarseness of voice that appeared few months after birth. The other case is a 4-year-old girl, who presented with waxy papules on face and trunk, hoarseness of voice and enlarged lips and tongue. The lesions healed leaving pitted scars in both cases. Based on clinical, histopathological and laryngoscopy findings, lipoid proteinosis was diagnosed in both cases. Acitretin was started in a dose of 0.5 mg/kg/day in every child. Complete remission of cutaneous lesions and improvement of the hoarseness was observed after one year. Conclusion: Acitretin may be benificial for treatment of mucosal and cutaneous lesions in lipoid proteinosis. [ABSTRACT FROM AUTHOR]

Keloids are slow growing neoplasms characterized by benign proliferation of fibroblasts that is due, at least in part, to altered cytokine profiles. Stem cells were claimed to play a role in skin tumor development. However, their role in keloid formation is unclear. The current study investigated the immunoreactivity of CD34 and c-KIT antibodies in 30 cases with keloid lesions together with normal skin biopsies of 30, sex and age-matched subjects representing the control group. Examined keloid sections showed positive dermal stromal immunoreactivity for CD34 in 76.7% of cases. CD34 expression intensity and H score were upregulated in keloid tissue relative to normal skin ( p < 0.0001, p = 0.0002, respectively) and in perilesional relative to lesional tissue ( p = 0.03, p < 0.001, respectively). c-KIT showed positive dermal stromal expression in all cases. Dermal c-KIT expression intensity and H score were upregulated in keloid tissue relative to normal skin ( p < 0.008, p < 0.001, respectively) and in perilesional relative to lesional tissue ( p < 0.0001, p < 0.001, respectively). Lesional skin showed more staining of basal keratinocytes when compared to perilesional tissue ( p < 0.0001). Hematopoietic stem cells may share in keloid pathogenesis. Further studies are warranted to gain firmer conclusion about the exact role played by these cells and the significance of their perilesional accumulation. The future therapy of keloid scars may have to target this stem cell population in order to deprive these tumors of their regenerative cell pools. [ABSTRACT FROM AUTHOR]

The aim of the present investigation was to evaluate the efficacy of Withania somnifera on tricarboxylic acid (TCA) cycle enzymes and electron transport chain in azoxymethane-induced experimental colon cancer in mice. Azoxymethane at the dose of 15 mg/kg body weight was induced intraperitoneally once in a week for 28 days. The progression in colon tumor development was correlated with the appearance of the histological biomarker and aberrant crypt foci (ACF). Azoxymethane-induced colon cancer animals were treated with 400 mg/kg body weight of W. somnifera once in a week orally for 28 days. After the experimental period, the animals were killed and analyzed for TCA cycle key enzymes, such as isocitrate dehydrogenase (ICDH), succinate dehydrogenase (SDH), malate dehydrogenase (MDH), and α-keto glutarate dehydrogenase (α-KGDH). The modulating effect of W. somnifera on TCA cycle key enzymes and electron transport chain complexes were investigated against colon cancer induced by azoxymethane in Swiss albino mice. Decreased activities of TCA cycle key enzymes such as ICDH, SDH, MDH, and α-KGDH in colon cancer bearing animals were observed. W. somnifera administration normalized these enzyme levels in azoxymethane-induced experimental mice. These results suggested that W. somnifera is the promising chemotherapeutic agent for the treatment of colon cancer. [ABSTRACT FROM AUTHOR]
