Synthetic astaxanthin is an effective nutritional strategy for improving shrimp body color and promoting growth. However, the optimal amount of astaxanthin in feed also varies with the synthetic technology and purity. In the present study, five diets containing different doses of synthetic astaxanthin (0% (CON), 0.02% (AX0.02), 0.04% (AX0.04), 0.08% (AX0.08), and 0.16% (AX0.16)) were administered to Penaeus monodon (initial body weight: 0.3 ± 0.03 g) for 8 weeks. With an increase in astaxanthin content in feed, weight gain and specific growth rate increased initially and subsequently decreased, with the highest value appearing at AX0.08. Dietary astaxanthin supplementation obviously improved the carapace and muscle color by enhancing astaxanthin pigmentation. Meanwhile, the fatty acid profile was altered by dietary astaxanthin, as evidenced by a decline in palmitic acid proportion, along with an increase in n-3 polyunsaturated fatty acids (n-3 PUFA) contents in muscle. In addition, dietary astaxanthin supplementation regulated prawn's antioxidant capacity. In the hemolymph, the activities of glutamic pyruvic transaminase (GPT) showed a significantly decrease trend with linear effect. The activities of glutamic oxaloacetic transaminase (GOT) and the contents of malondialdehyde (MDA) were first downregulated and then upregulated with significantly quadratic pattern. In the hepatopancreas, the activities of superoxide dismutase (SOD) and the contents of MDA were significantly downregulated with the increase of dietary astaxanthin levels. Reduced glutathione (GSH) contents and catalase (CAT) activities were also significantly decreased in group AX0.08. Correspondingly, astaxanthin decreased GSH and MDA contents under transportation stress. Moreover, the mRNA expression of immune genes (traf6, relish, and myd88) were inhibited by dietary astaxanthin supplementation. Based on the results of polynomial contrasts analysis and Duncan's test, dietary synthetic astaxanthin is a suitable feed additive to improve the growth, body color, antioxidant capacity, and nonspecific immunity of P. monodon. According to the second-order polynomial regression analysis based on the weight gain, the optimal supplementation level of dietary astaxanthin was 90 mg kg−1 in P. monodon. [ABSTRACT FROM AUTHOR]

Bioluminescent bacteria (BLB) are the most widely distributed light-emitting microorganisms, most of which are found as symbionts of free-living marine organisms, including the crustacean species. In this study, 4 out of 7 marine crustaceans in the northeastern section of Manila Bay were found to contain bioluminescent bacteria. Marine crustacean species namely Thenus orientalis (Oriental Slipper Lobster), Oratosquilla oratoria (Mantis Shrimp), Penaeus monodon (Giant Tiger Prawn), Litopenaeus vannamei (Whiteleg shrimp), Scylla serrata (Mud Crab), Portunus pelagicus (Blue Swimming Crab), and Charybdis feriata (Crucifix crab), were swabbed after collection. Bioluminescent bacteria were isolated from the inner surface of the carapace, eyes, abdomen, and abdominal segments of the crustacean samples. All glowing colonies were isolated and upscaled. Only bioluminescent bacteria from T. orientalis, O. oratoria, and P. monodon completed the isolation process and were identified using 16S rRNA gene sequencing for phylogenetic analysis. Furthermore, results from molecular identification through Nucleotide BLAST identified that it was Vibrio alginolyticus and Mucus bacterium. At the same time, the presence of Vibrio rumoiensis was also identified which was proved to be understudied and needs exploration. This study was aimed to identify the present bioluminescent bacteria in selected marine crustaceans in Manila Bay, Philippines through microbiological isolation, molecular identification, and phylogenetic reconstruction. The inability to precisely identify biological species has direct impacts on conservation efforts, which is especially true among taxa of great commercial interest such as representatives of the Penaeini tribe. Those shrimp supply more than half of the global demand for crustaceans and have important functional roles in ocean ecosystems. Recent systematic studies suggest that the diversity of taxa within Penaeini is underestimated, and there are controversial taxonomic classifications. In this work, we used phylogenetic and phylogeographic approaches to assess the genetic diversity and evolutionary of representatives of the Penaeini tribe, focusing on the mitochondrial cytochrome oxidase subunit I gene. A total of 1680 COI sequences from 24 species were accessed on the Bold Systems platform. We detected several identification errors and synonyms among the 15 taxa of the tribe, principally within the genus Penaeus. Phylogeographic analyses allowed the identification of highly structured populations within several ecoregions of the Atlantic (19), Indian (16), and Pacific (10) oceans. The delimitation methods (GMYC, bPTB, and BINs) suggest are cryptic species such as Fenneropenaeus indicus (2 MOTUs), Litopenaeus vannamei (2 MOTUs), Penaeus monodon (3 MOTUs), and P. semisulcatus (4 MOTUs). The highest cryptic diversity concentration was found in the Indo-West Pacific region, suggesting it as the center of origin for the tribe. Nowadays, bacterial diseases caused by Vibrio spp. pose a significant obstacle to aquaculture growth, affecting socioeconomic development. The presence of pathogenic Vibrio spp. are gaining attention due to widespread mortality in fish and shellfish worldwide. This study investigates the distribution of virulence genes and their pathogenicity by experimentally infecting a healthy group of cultured shrimp, specifically Penaeus monodon. A total of 225 Vibrio spp. isolated from 210 cultured shrimp from various geographical regions in Peninsular Malaysia were examined, resulting in 13 Vibrio spp. being identified and characterized. According to virulence gene assessment, only eight of the 11 virulence genes were detected in the isolates, including chiA (89%), flaC (74%), tlh (72%), toxR (70%), luxR (40%), pirA (11%), pirB (11%), and hlyA (0.4%). Other virulence genes, such as tdh, trh, and ctxA, were not found in any of the isolates (0%). Then, three Vibrio isolates, including V. parahaemolyticus S12-3, V. campbellii S10-4, and V. rotiferianus S24-4, were selected for pathogenicity test based on highest number of virulence genes in each species. These species were subjected to immersion challenge test against postlarvae (PL) P. monodon approximately PL-20 with three different concentrations of 1 × 106, 1 × 107, and 1 × 108 CFU/mL. The V. parahaemolyticus S12-3 was determined to have lethal concentration 50 (LC50) 1 × 105 CFU/mL after 168 hr of exposure. The immersion challenge test resulted in a pale hepatopancreas and empty guts in P. monodon. Histopathological examination showed sloughing of epithelial cells in the hepatopancreatic tubule, hemocytic infiltration, massive vacuolation, and loss of hepatopancreatic tubule structure. Overall, the study demonstrated the virulence and pathogenicity of V. parahaemolyticus S12-3 toward P. monodon, which could be associated with its virulence properties. This study aims to reduce the cost of allergen testing for Guangzhou, China by limiting the number of allergens for which patients are tested, and provide a testing panel to improve diagnostic and therapeutic efficiency. This retrospective study of real-world data from 2012 to 2019 included 39,570 patients with suspected allergies in Guangzhou, southern China. All the patients were tested for one or more of the following allergens serum specific immunoglobulin E (sIgE): Dermatophagoides pteronyssinus, Dermatophagoides farinae, cat dander, dog dander, Artemisia vulgaris, Aspergillus fumigatus, Alternaria alternata, Blattella germanica, egg whites, milk, wheat, peanuts, soybeans, Cancer pagurus, and Penaeus monodon by PhadiaCAP 1000. Totally, only the positive rates of allergens sIgE in D. farinae, D. pteronyssinus, milk, egg whites, B. germanica, C. pagurus, A. alternata, and P. monodon were > 10%, the other allergens were between 4–7%. Moreover, among the allergic diseases, dust mites exhibited the overall highest positive rate, followed by milk and B. germanica. In children, milk was the main allergen, whereas in adults, mites, cockroaches, shrimp, and crab allergens had higher positive rates. The optimal scale analysis shows that the multiple sensitization classification of patients can be divided into three categories: I D. farinae and D. pteronyssinus; II. C. pagurus, P. monodon, and B. germanica; III. Milk and egg whites. Generally, a panel including 4 allergens can detect > 90% of the potential allergy in this local population. White Spot Syndrome Virus (WSSV) is a major virus found in shrimp that causes huge economic loss in shrimp farms. A selective diagnostic approach for WSSV is required for the early diagnosis and protection of farms. This work proposes a novel recognition method based on improved Convolutional Neural Network (CNN) namely Dense Inception Convolutional Neural Network (DICNN) for diagnoses of WSSV disease. Initially, the process of data acquisition and data augmentation is carried out. The Inception structure is then used to improve the performance of multi-dimensional feature extraction. As a result, the proposed work has the highest accuracy of 97.22% when compared to other traditional models. The proposed work is targeted to Litopenaeus Vannamei (LV), and Penaeus Monodon (PM) diversities for major threats detection of White Spot Syndrome (WSS). Performance metrics related to accuracy have been compared with other traditional models, which demonstrate that our model will efficiently recognize shrimp WSSV disease. [ABSTRACT FROM AUTHOR]

For decades, changes in fish catch composition and the marine environment have been monitored worldwide and recorded in databases like FAO FishStatJ and the European Union Copernicus Marine Service. However, the complexity and high variability in the dataset makes it challenging to find meaningful information through conventional data analytical methods. Therefore, in this pilot data mining study, we employed association rule mining algorithms (Apriori, ECLAT, and FP-Growth) to find frequently occurring itemsets in the fish-catch composition and marine environment data of the west and east coasts of India during the past decade (2011-2020). Firstly, the inherent spatial and temporal variations in fish-catch composition and marine environment (sea surface temperature and chlorophyll) on the west and east coasts of India were statistically analyzed and described. Then, the data were preprocessed, selected, and transformed into categorical attributes. By applying the association rule mining algorithms written in the Python language in the Google Colab workspace, we obtained frequent itemsets of fish catch and marine environment with different levels of minimum support and confidence. The preliminary results showed linear and inverse associations between changes in the sea surface temperature, chlorophyll concentration, and major catch groups, such as anchovies, Indian oil sardine, Indian mackerel, hairtails, butterfish-pomfrets, Bombay duck, flatfish, tunas, giant tiger prawn, crabs, lobsters, and cephalopods. Among the tested data mining algorithms, FP-Growth was found to be more efficient and reliable in finding associations between the spatiotemporal dynamics of the marine environment and fish distribution and abundance. Despite the success in identifying new crustacean allergens other than tropomyosin, many potential allergens remain unidentified. At present, there are no diagnostic and treatment crustacean allergens on the Russian market, including tropomyosin of invertebrates. The aim of the research was to explore allergenic properties of tropomyosin and other proteins in water-salt extract from tiger shrimp (Penaeus monodon). Raw and boiled meat of giant tiger shrimp was used to prepare allergenic extracts. To identify the specific activity of experimental allergens, sera from patients allergic to crustaceans with an established degree of activity (class 3) were used. The levels of specific IgE were from 3.5 to 17.5 IU/mL. ELISA was performed with the allergen samples. Polyacrylamide gel electrophoresis of raw and boiled shrimp allergen preparations was followed by western blotting with the most allergen-specific serum. It has been shown that the extracts from raw and boiled shrimp have allergenic activity (bind IgE). The heat treatment of shrimp had no effect on allergenic activity except for one sample, where a strong reaction with the antigens was found. It is proved that the selected extraction conditions make it possible to reveal the multifractional nature of the protein profile of raw and boiled shrimp extracts. In western blotting experiments, the raw shrimp extract showed a weak reaction; tropomyosin was not detected. At the same time, five protein fractions were identified in the boiled shrimp samples which reacted with IgE antibodies of a patient allergic to crustaceans. Laranjeira et al.'s study evaluated the effects of extracts of the aromatic plant I Pterospartum tridendatum i , an important source of active compounds, in treating acute and chronic inflammation in adult rats using an experimental model of osteoarthritis and ear edema and correlated it with its phytochemical composition. Bioactive components are substances that positively influence the organism, resulting in a physiological benefit and/or reduction in the risk of developing certain pathologies. Ahmed et al. evaluated the effects of various plant matrices and their phytochemical compounds in terms of modulating the immune system and signaling pathways involved in anti-tumor immunity. The results show that this effect depends on the chemical structure of the antioxidant substances used, as some of them, working in combination with the field, show a strong anti-tumor effect, while others, such as resveratrol, lose their anti-tumor properties under the influence of a moderate-intensity static magnetic field [[2]]. Abstract The current study profiled the dual isotopes of carbon and nitrogen in feeds and muscles of farmed Penaeus monodon for evaluation of feed efficiency in making up the prawn muscle. Signatures of both feed and resulted muscles of P. monodon were subjected to two‐ and three‐source linear mixing models to elucidate the contribution of each feed item in the building of the muscle. The results revealed that carbon and nitrogen in different feed substances have different influences on the growth and nutrient uptake by the prawn. Different growth stages showed isotopic switching within prawn muscles in the course of their building up. Generally, marine sources were the most enriched in both δ13C and δ15N. Likewise, δ13C of wild‐caught prawns (−16.30 ± 0.72‰) were superior over farmed prawns (−18.00 ± 0.59‰) (p 0.01). The isotopic composition of P. monodon mirrored those of the ingredients contained in its feed and varied across treatment levels. Comparing to other ingredients, marine macroalgae exhibited significantly higher (p 0.01). Moreover, a clear distinction was observed between wild and farmed P. monodon, and such a distinction is clearly explained by δ13C composition. In conclusion, multiple sources of δ13C and δ15N in feeds are incorporated more efficiently in muscles than single sources.

Acute hepatopancreatic necrosis disease (AHPND) has inflicted massive economic losses and posed a considerable threat to the development of penaeid shrimp aquaculture in Malaysia. The restriction on antibiotic uses have necessitated exploring alternatives preventive measures against AHPND outbreak, such as the use beneficial bacteria and nutritional additives. In this study, diets which were fed to Penaeus monodon postlarvae (PL15) were fed with diets containing probiotic (L. plantarum at 108 CFU/mL), prebiotic (S. polycystum at 2%) and synbiotic diets (combination of probiotic and prebiotic) formulations. These diets were administered in triplicate for 35 days, followed by an immersion challenge with 6 × 105 CFU/mL AHPND-causing V. parahaemolyticus S2–4. Sampling of shrimp’s cephalothorax and abdomen from the feeding trial and the immersion challenge were conducted for gene expression and histopathology analyses. Results indicated that the synbiotic-fed group displayed the most significant weight gain, specific growth rate, and protein conversion ratio among the tested groups. In addition, shrimp cephalothorax from this group displayed a significant immune response, with expression of LGBP, peroxinectin, prophenoloxidase during post-feeding trial, and expression of prophenoloxidase, toll-like receptor, penaeidin, during post-challenge trial. Furthermore, the highest L. plantarum concentration in abdomen, lowest V. parahaemolyticus S2–4 concentration in cephalothorax and highest percent survival of shrimp during post-challenge were observed in this group. Therefore, this study highlighted the positive effect of synbiotic-supplemented diet on growth, immune response, and disease resistance of shrimp, offering a promising and sustainable solution to alleviate substantial production losses in shrimp farming.

This research sought to assess the effects of dietary supplements with Gracilaria lichenoides and Bacillus amyloliquefaciens, either individually or combined, on the growth performance, antioxidant capacity, and intestinal function of Penaeus monodon. A total of 840 shrimps were randomly assigned to 28 tanks with an average initial weight of (1.04 ± 0.03) g (30 shrimp per tank) with 7 different treatment groups and 4 replicates per treatment. The control treatment (C) consisted of a basal diet; in contrast, the experimental groups were complement with varying levels of G. lichenoides (3% or 8%), either alone (S3 and S8) or in combination with B.amyloliquefaciens at different concentrations (3% G. lichenoides and 109 CFU/g—S3B9; 8% G. lichenoides and 1011 CFU/g B. amyloliquefaciens—S8B11; 109 CFU/g B. amyloliquefaciens—S9; 1011 CFU/g B. amyloliquefaciens—B11). The results indicated that the maximum values of final body weight (FBW) (10.49 ± 0.90) g, weight gain rate (WGR) (908.94 ± 33.58) g, and specific growth rate (SGR) (4.20 ± 0.06) g were perceived in the 3% G. lichenoide diet treatment, and compared with the control group, the difference was significant (p < 0.05). The whole-body lipid content of shrimp in the B9 group was significantly higher than that in the B11 group (p < 0.05), but no significant difference was observed when compared with shrimp fed other diets (p > 0.05). The ash content of shrimp in the B9 group was found to be significantly higher than that in the S3B9 group (p < 0.05). Furthermore, the lipase activity in the stomach and intestines of the experimental groups exhibited a statistically significantly increase compared to the control (p < 0.05). In comparison to the control group, the hepatopancreas of the S3 group exhibited a significant increase in the activities of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and antioxidant genes [SOD, catalase (CAT), GSH-Px, thioredoxin (Trx), Hippo, and NF-E2-related factor 2 (Nrf2)] expression levels (p < 0.05). Additionally, the activities of total antioxidant capacity (T-AOC), SOD, peroxidase (POD), and antioxidant genes (CAT, GSH-Px, Trx, and Hippo) in the S3B9 treatment of hepatopancreas showed significant improvement (p < 0.05). The inclusion of dietary G. lichenoides and B. amyloliquefaciens resulted in enhanced relative expression of intestinal lipid metabolism genes (fatty acid synthetase (FAS), lipophorin receptor (LR), fatty acid transport protein 1 (FATP1)) and suppressed the expression of the long-chain fatty acid-CoA ligase 4 (LCL4) gene. Analysis of microbiota sequencing indicated improvements in composition and structure, with notable increases in Firmicutes at the phylum level and Vibrio at the genus level in the S3 group, as well as an increase in Tenericutes at the genus level in the S8B11 group. Overall, the inclusion of dietary G. lichenoides and B. amyloliquefaciens positively impacted the growth, antioxidant capacity, and microbial composition of shrimp, with particular enhancement observed in shrimp fed a supplementary 3% G. lichenoides diet.

Polysaccharide monooxygenases (PMOs) and Glycoside Hydrolases (GH) play a crucial role in breaking down chitin and cellulose. Research on PMOs has mainly focused on their role in the production of bioethanol from biomass. Still, the potential of PMOs as virulence factors in microorganisms is not been fully studied. Some pathogenic bacteria secrete PMOs, which have been identified as virulence factors. The ability of Vibrio species to metabolize chitin is well-known, but the question of which Vibrio species contains the PMO gene remains unanswered. Materials and methods: Black tiger shrimp Penaeus monodon was collected from a culture pond in Long An, Vietnam. Thiosulfate-citrate-bile salts-sucrose and CHROMagar Vibrio agar were used to isolate, Vibrio bacteria from the shrimp samples. Mixed bases primers were designed to detect PMOs gene from Vibrio strains. Results: Total of 35 different strains of bacteria were isolated from the guts and pancreas of the shrimp sample. Based on biochemical tests, there were a total of 20 different Vibrio strains identified among 35 isolated strains. The PCR results showed that six isolates could have the PMOs gene. The partial 16S rDNA fragments of six isolate Vibrio strains were sequenced. Comparing 16S rDNA sequences with sequences available in NCBI databases revealed that all the sequences are Vibrio spp. 16S rDNA sequences.

Astaxanthin is an efficient nutrition strategy to relieve oxidative stress and enhance crustacean body color. Because of technological and economic benefits, synthetic astaxanthin is favored by the aquatic feed industry. However, whether synthetic astaxanthin can achieve the effects of natural astaxanthin needs further investigation. Here, we evaluated the effects of synthetic astaxanthin and natural astaxanthin (dietary supplementation with Haematococcus pluvialis and Phaffia rhodozyma) on the growth performance, body color, fatty acid composition and antioxidant capacity of black tiger prawn (Penaeus monodon). Compared with the control group (no astaxanthin supplementation), only synthetic astaxanthin significantly increased the weight gain rate (WG) and specific growth rate (SGR), together with the elevation of feed efficiency ratio (FER). All three types of astaxanthin enhanced body color by increasing astaxanthin deposition. The highest value of (a*) in the carapace and muscle occurred in the synthetic astaxanthin group. Synthetic astaxanthin significantly increased the n-3 polyunsaturated fatty acid (n-3 PUFA) contents in muscle. As expected, different sources of astaxanthin inhibited glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) activities in the hemolymph and decreased the contents of reduced glutathione (GSH) and malondialdehyde (MDA) in the hepatopancreas. Correspondingly, the structure of hepatopancreas was enhanced by astaxanthin. In addition, three types of astaxanthin almost indistinguishably enhanced the ability of shrimps to resist stress caused by transportation simulation. In conclusion, these results indicate that dietary synthetic astaxanthin has a better effect than natural astaxanthin on growth promotion, body color enhancement and n-3 PUFA deposition in muscle of black tiger prawn. Hence, synthetic astaxanthin is a suitable feed additive to promote the healthy and sustainable development of the shrimp industry.

Vibrio parahaemolyticus, the causative agent of Acute hepatopancreatic necrosis disease (AHPND), was discovered in 2013 as a unique isolate that produces toxins and kills penaeid shrimps in devasting nature in Bangladesh and causes severe economic losses. This research aimed to understand the expressions of immune genes in different stages of the host species, Penaeus monodon, against virulence and toxin genes upon being challenged with V. parahaemolyticus. Healthy post-larvae (PL) samples were collected from southwestern of Bangladesh from July 2021 to August 2022. The tryptic soy agar with 1.5% sodium chloride (NaCl) was used to inoculate the cells of V. parahaemolyticus, and the tryptic soy broth (TSB) with 1.5% NaCl was used to transfer the colonies. The spectrophotometry measured bacteria density. PCR, qPCR, SDS-PAGE, and Western blot measured gene expression and survivability after the immersion challenge. The 1 × 105CFU/mL of V. parahaemolyticus was used for 144 h.p.i (hours post-infection) challenge to six stages of post-larvae (PL) of P. monodon (PL20, PL25, PL30, PL35, PL40, and PL45), PL30 and PL35 showed 100% mortality by day 72 (h.p.i.) after exposure that indicated most vulnerable to V. parahaemolyticus. The expression of immune and toxic genes was confirmed by qPCR. The immune genes toll-like receptors (TLR), prophenoloxidase (ProPO), lysozyme (lyso), and penaeidin (PEN) of PL20 and PL25 of P. monodon were expressed robustly up-trends. PL30 and PL35 showed the lowest gene expression at the end of 72 (h.p.i.). At the end of the 144 (h.p.i.) exposure, the immune genes TLR, ProPO, lyso, and PEN expressed highest in PL45 than other post-larvae stages of P. monodon. The toxic genes (pirA, ToxR, ToxA, ToxB, tlh, tdh, and trh) in PL30 and PL35 of P. monodon after exposure of V. parahaemolyticus were expressed highest at the end of the 72 (h.p.i.). The lowest toxic genes expressions were revealed in PL20 and PL45 at the end of the 144 (h.p.i.). The SDS-PAGE analysis of proteins from the bacterium revealed identical protein profiles with toxic genes, and those toxins were further confirmed by Western blot. The 20 kDa, 78 kDa (ToxR), 20 kDa, 25 kDa (ToxA), 25 kDa (ToxB), 20 kDa, 27 kDa, 75 kDa (tdh), and 20 kDa, 27 kDa, 75 kDa, and 78 kDa (trh) proteins were strong responses in Western blot, indicating the crucial involvement of these immune-related genes in the defense and recovery of the first-line defense mechanisms during V. parahaemolyticus infection to shrimp. The all-toxic genes showed a unique homology and those derived from the common ancestor compared with V. parahaemolyticus (NCBI accession no. AP014859.1). All clades were derived with different traits with very low genetic distance, where the overall mean distance was 3.18 and showed a very uniform and homogenous pattern among the lineages. The V. parahaemolyticus infection process in different PL stages in P. monodon revealed novel insights into the immune responses. The responses may lead to the subsequent production of a DNA vaccine, enhancing shrimp health management to minimize the economic losses due to AHPND experiencing an outbreak of early mortality syndrome (EMS) toward sustainable production P. monodon (shrimp).

This experiment was conducted to assess the effects of replacing fish meal with fermented corn gluten meal (FCGM) in diets on the growth performance, immune response and intestinal digestive function in Penaeus monodon. In this experiment, grade levels of FCGM (4.4%, 8.8%, 13.2%, 17.6%, 22.0%) were added in the basic diet to instead of 14.3%, 28.6%, 42.9%, 57.1% and 71.4% fish meal and six kinds of isonitrogenous and isoenergetic experimental diets were prepared to feed [initial weight (12.00±0.25) g] Penaeus monodon for 8 weeks. The experimental results showed as follows: ① compared with the control group, when fermented corn protein meal replaced 42.8% and less of fish meal, it did not significantly affect the growth performance of Penaeus monodon (P>0.05); when fermented corn protein meal replaced 57.1% of fish meal, it significantly reduced the weight gain rate and specific growth rate of Penaeus monodon (P<0.05), and the feed coefficient was significantly higher when the replacement level reached 71.4% (P<0.05). ② Compared with the control group, fermented corn protein meal did not significantly (P>0.05) affect serum immunity indexes of Penaeus monodon when 42.8% and less of fish meal was replaced by fermented corn protein meal; 57.1% of fish meal was replaced by fermented corn protein meal significantly (P< 0.05) reduced serum peroxidase (POD) and acid phosphatase (ACP) activities of Penaeus monodon, while when the replacement ratio reached 71.4%, ACP, POD, superoxide dismutase (SOD) and lysozyme (LZM) activities were significantly lower (P<0.05) than those of the control group; alkaline phosphatase (AKP) activity did not differ significantly (P>0.05) among the treatment groups. ③ Substitution of 42.8% and less of fishmeal by fermented corn protein powder did not affect the intestinal digestive enzyme activities of Spotted Penaeus monodon compared to the control group (P>0.05), whereas substitution levels of 42.8% and more significantly decreased intestinal protease activity, amylase activity and lipase activity (P<0.05). In conclusion, replacing fish meal with FCGM controlled 42.9% in the diet does not have negative effects on growth performance, immune function and intestinal digestive capacity of Penaeus monodon, but higher substitute levels showed serious adverse effects on Penaeus monodon. Objectives: This study was to identify and characterize Decapod iridescent virus 1 (DIV1) in the outbreaks reported in two whiteleg shrimp farms and one black tiger shrimp farm located in northern Taiwan in 2020. Methods: The histopathology, electron microscopy and polymerase chain reaction (PCR) specific for the DIV1 were used to identify the virus, and the phylogenetic analysis was performed by comparing the major capsid protein gene fragment of DIV1s from Taiwan with reference sequences of the family Iridoviridae. Results: DIV1 was identified by diagnostic PCR and caused mild mortality (20%) in cultured Penaeus monodon and high mortality (100%) in cultured whiteleg shrimp. Cultured P. monodon was first found to be infected with DIV1 through natural route of infection. Histopathological examination showed dark‐eosinophilic cytoplasmic inclusions in the degenerative cells of targeted hematopoietic tissues. For electron microscopy, a non‐enveloped virus particle was observed from homogenates of mixed target organs through negative staining with a diameter of 112±2 nm. Nucleotide sequences of DIV1 isolates from the Taiwanese outbreak are 100% identical to those from the PRC. Conclusions: Based on the clinical evidence, mortality rates, histopathology, electron microscopy examinations and phylogenetic analysis, it is believed that DIV1 is the causative agent of the outbreak. This review summarises the current knowledge of mineral nutrition for penaeid shrimp. It investigates how the aquatic environment and the lifecycle of shrimp affect requirements and the role that minerals play in shrimp health. Methods of supplying minerals via either water or to feed, and novel ways of supplementing minerals within feed, are discussed. The requirements for individual minerals are summarised with recommendations for minimum levels of dietary inclusion for semi‐intensive and intensive commercial shrimp culture presented where data permits. Estimates of dietary requirement remain broad for most minerals for the main shrimp production species (Penaeus vannamei, Penaeus monodon and Penaeus japonicus), with some essential minerals remaining unstudied (Table 2 in Section 5.10). Penaeus monodon (tiger shrimp), Metapenaeus monoceros (brown shrimp), and Fenneropenaeus indicus (white shrimp) are the most economically important shrimp species in the waters of the Bay of Bengal, Bangladesh. This is the first analytical study to assess three major shrimp stocks using both length-based and catch-based methods, such as length-based Bayesian biomass estimation (LBB), length-based indicator (LBI), and a catch-based method entitled JABBA (Just Another Bayesian Biomass Assessment), to explore and process the data; estimate the growth parameters, with length at first capture; present relative biomasses; and approximate the reference points. The parameters of the von Bertalanffy Growth Function (VBGF) analysis for the tiger, brown, and white shrimps were L∞ = 113.0 mm, 85.4 mm, and 76.4 mm, respectively, for carapace length. Our results showed that the relative biomass level (B/BMSY) of the tiger shrimp was 0.43, suggesting an overfished status, and brown and white shrimps were 0.84 and 0.96, indicating that they were fully exploited but not overfished. This study, therefore, advised an optimum carapace length limit to catch from 57.0–70.0 mm for tiger shrimp, 44.0–53.0 mm for brown shrimp, and 40.0–48.0 mm for white shrimp. The estimated maximum sustainable yield (MSY) reference points were as follows: optimal biomass BMSY = 3116 mt, 15,885 mt, and 2649 mt for tiger, brown, and white shrimp, respectively, and optimal harvest rate uMSY = 12%, 33%, and 8% for tiger, brown, and white shrimp, respectively. The average annual catch values for the last ten years were 265 mt, 2396 mt, and 115 mt below the estimated MSY values of 389 mt, 4899 mt, and 209 mt for tiger, brown, and white shrimp, respectively. But, brown shrimp had the estimated highest carrying capacity (31,770 mt) and intrinsic growth rate (0.66) than the tiger and white shrimp, which was replicated distinctly in the graphical representation of the Kobe plot and the surplus production plot. Microbial community inhabiting the intestine of the shrimp (Penaeus monodon) and their surrounding environments (e.g., water and sediment) is considered as a key contributing factor for the sustainable farming of shrimp. Indiscriminate application of antibiotics in aquaculture is a growing concern due to the emergence of antibiotic resistant bacteria (ARB), more specifically the antibiotic resistance genes (ARGs). The present study investigates the microbiome composition and 19 ARGs from four different shrimp farming systems; (i) cluster, (ii) extensive, (iii) semi-intensive, and (iv) improved extensive in the southwest coastal region of Bangladesh. In doing so, the study applied advanced 16S rRNA-based metagenomic sequencing to study the bacterial composition. Moreover, gene specific polymerase chain reaction (PCR) was employed to detect the ARGs in shrimp, water, and sediments of different farming systems. In the current study, bacteria from the phylum Proteobacteria and Firmicutes were predominant among the samples (n = 12) collected from the different farming systems followed by Actinobacteria, Bacteroidetes, and Cyanobacteria. Firmicutes was the predominant phylum in the gut of shrimp cultured in the cluster (relative abundance 53.33%) and semi-intensive (relative abundance 59.2%) culture systems. Results indicated that the bacterial community structure was significantly (p < 0.05) distinct among gut, sediment, and water samples as well as the farming systems. The shared operational taxonomic unit (OTU) in the sediment sample (16,495) was nearly double than the gut (7,931) and water (8,513) bacterial communities. The improved extensive farming system showed 1,289 (11.05%) shared OTUs among gut, sediment, and water followed by semi-intensive (6.87%), cluster (6.27%), and extensive (5.46%) farming system. Among the tested ARGs, sul1, cat, gyrA(C), tetA, tetC, tetX, ere(A), vanR, and dfrA1 were predominant in water and sediment samples. Semi-intensive farming system had the highest prevalence of ARGs (21.05%) while the lowest prevalence was found in extensive (5.26%) farming system. Overall, the study provides a comprehensive scenario of bacterial composition and growing emergence of ARGs in shrimp farming of Bangladesh. Seafood elemental profiling (SEP) is the quantification of a range of elements in seafood products and may serve in addressing questions of seafood provenance and quality. Traditional methods for analyzing soft tissue present several limitations for the industry-level use of SEP. Portable handheld X-ray fluorescence (pXRF) analysis is a promising alternative to conventional methods; however, its application for biological analysis has not been fully established. Intact giant tiger prawn (Penaeus monodon) abdomens were analyzed with a Vanta M series XRF portable analyzer following a novel soft tissue protocol. Exploratory statistics (principal component analysis, nonmetric multidimensional scaling, and canonical discriminant analysis), as well as random forest models, have been implemented with pXRF profiles, yielding 81% accuracy when assigning the geographical origin of P. monodon. The results of this study highlight that SEP via pXRF is a viable industry-level analysis, and its application will depend on improved instrument calibration to account for fluctuating wetness factors that are influenced by cooking, storage, and other pre- and post-harvest treatments. As the demand for seafood increases, so does the incidence of seafood fraud. Confirming provenance of seafood is important to combat fraudulent labelling but requires a database that contains the isotopic and elemental "fingerprints" of authentic seafood samples. Local isotopic and elemental databases can be scaled up or combined with other databases to increase the spatial and species coverage to create a larger database. This study showcases the use of isotopic and elemental fingerprints of the black tiger prawn (Penaeus monodon) to develop a database that can be used to securely store the data necessary for determining provenance. The utility of this database was tested through querying and building seven different datasets that were used to develop models to determine the provenance of P. monodon. The models built using the data retrieved from the database demonstrated that the provenance of P. monodon could be determined with >80% accuracy. As the database was developed using MySQL, it can be scaled up to include additional regions, species, or methodologies depending on the needs of the users. Combining the database with methods of determining provenance will provide regulatory bodies and the seafood industry with another provenance tool to combat fraudulent seafood labelling. The aim of this work was to evaluate the spatial and temporal distribution of shrimp species in the Lake Nokoué-Cotonou Channel complex for an efficient management of the fishery. The results of one year sampling from five stations (North, South, East, Central and West) in the complex revealed the following proportions: Penaeus notialis 73.68%, Macrobrachium macrobrachion 13.81%, Macrobrachium vollenhovenii 5.36%, Penaeus monodon 2.68%, Macrobrachium felicinum 2.20%, Penaeus kerathurus 1.72%, Macrobrachium cf. zariquieyi 0.27%, and Macrobrachium equidens 0.27%. The species M. cf. zariquieyi and M. equidens were never reported from Benin, but are now found in Lake Nokoué. Spatially, Penaeus (Penaeus notialis) are more abundant in the west (Acadja concentration zone), while Macrobrachium (Macrobrachium macrobrachion) are more dominant in the east (near the Ouémé entrance) and north (near the Sô entrance) of the lake. Penaeus are strongly present in the lagoon complex from January to August, while Macrobrachium are more abundant in Lake Nokoué from September to December. Spearman's correlation analysis revealed that increasing salinity, pH, transparency and total dissolved solids (TDS) positively influenced the abundance of P. notialis , while increasing salinity, pH, transparency, and TDS negatively influenced the abundance of M. macrobrachion at the 5% significance level. For an efficient management of the shrimp fishery, it is desirable that the shrimp fishery in Lake Nokoué, mainly in the Cotonou Channel, which is a very sensitive area for their seasonal migrations, benefits from at least three months of respite per year, from November to January. This period corresponds to the strong migration of juvenile Penaeus towards the lake. Sixteen novel circular rep-encoding DNA sequences with high sequence homologies to previously described SPHINX and BMMF sequences were isolated for the first time from non-bovine foods (pork, wild boar, chicken meat, Alaska pollock, pangasius, black tiger shrimp, apple, carrot, and sprouts from alfalfa, radish, and broccoli). The phylogenetic analysis of the full-length circular genomes grouped these together with previously described representatives of SPHINX/BMMF group 1 and 2 sequences (eight in each group). The characterization of genome lengths, genes present, and conserved structures confirmed their relationship to the known SPHINX/BMMF sequences. Further analysis of iteron-like tandem repeats of SPHINX/BMMF group 1-related genomes revealed a correlation with both full-length sequence tree branches as well as Rep protein sequence tree branches and was able to differentiate subtypes of SPHINX/BMMF group 1 members. For the SPHINX/BMMF group 2 members, a distinct grouping of sequences into two clades (A and B) with subgroups could be detected. A deeper investigation of potential functional regions upstream of the rep gene of the new SPHINX/BMMF group 2 sequences revealed homologies to the dso and sso regions of known plasmid groups that replicate via the rolling circle mechanism. Phylogenetic analyses were accomplished by a Rep protein sequence analysis of different ssDNA viruses, pCRESS, and plasmids with the known replication mechanism, as this yielded deeper insights into the relationship of SPHINX/BMMF group 1 and 2 Rep proteins. A clear relation of these proteins to the Rep proteins of plasmids could be confirmed. Interestingly, for SPHINX/BMMF group 2 members, the relationship to rolling circle replication plasmids could also be verified. The effects of three feed additives in a low fish meal diet on growth, antioxidant capacity and intestinal microbiota of Penaeus monodon were studied to enhance the understanding of these effects and to provide basic data and a reference for the formulation and optimization of feed for P. monodon. A total of 630 healthy, homogenous shrimp (4.58 ± 0.05 g) were randomly divided into seven groups with three replicates per group and 30 shrimp per replicate in a breeding barrel (500 L). The additive names and amounts were 0 (CT); vitamin B6, 100 mg/kg (V1); vitamin B6, 200 mg/kg (V2); phytase, 1000 U/kg (P1); phytase, 2000 U/kg (P2); 0.2% yucca extract (Y1); and 0.4% yucca extract (Y2). The experiment lasted 8 weeks. The results showed that compared to CT, V1 and Y1 had a significant increasing tendency toward weight gain (WG) (p < 0.05) and had a significant decreasing tendency in the food conversion ratio (FCR) (p < 0.05). P2 had a significant increasing tendency in α-amylase activity (p < 0.05), and P1 had a significant increasing tendency in total superoxide dismutase (T-SOD) (p < 0.05). The next-generation sequencing of intestinal microbiota showed that Proteobacteria was the most abundant phylum in the seven groups, accounting for 29.33%, 56.67%, 55%, 45.33%, 73%, 39.33% and 64.33% of the total. Compared to CT, the Proteobacteria was significantly high (p < 0.05) in P2 and Y2, and the Bacteroidota in all other groups decreased significantly (p < 0.05). The functional prediction of FAPROTAX indicated that there was no significant difference (p > 0.05) in functional components among all groups. According to growth performance, antioxidant capacity and effects on intestinal microbiota, vitamin B6 (100 mg/kg), phytase (2000 U/kg) and yucca extract (0.2%) can be recommended as additives for the diet of P. monodon.

The aim of this experiment was to investigate the effects of the partial substitution of fish meal with soybean products and chicken meal on the growth performance, antioxidant capacity and intestinal microbiota of Penaeus monodon. A total of 450 healthy, consistent shrimp were randomly divided into five groups, with three replicates per group and 30 shrimp per replicate. The proportion of fish meal substituted with soybean products and chicken meal in the five feed groups was 0% (FM), 40% (40SC), 60% (60SC), 80% (80SC) and 100% (100SC). The experiment lasted for 8 weeks. The results showed that, compared to the FM group, the 40SC and 60SC groups had a decrease in WG and SR, but there was no significant difference (p > 0.05). In contrast, compared to the FM group, the FCR in the 100SC group was significantly increased (p < 0.05), while there was no significant difference among the FM and 40SC, 60SC and 80SC groups (p > 0.05). Compared to the FM group, the ACP in the 80SC and 100SC groups significantly increased (p < 0.05), while the 40SC and 60SC groups had no significant difference (p > 0.05). The AKP in the 100SC group was significantly higher than that in the FM group (p < 0.05), while there were no significant differences among the other four groups (p > 0.05). There were no significant differences in T-AOC and T-SOD among all the treatment groups (p > 0.05). The next-generation sequencing of the intestinal microbiota showed that Proteobacteria was the most abundant phylum in the five groups, accounting for 37.67%, 66%, 40%, 40% and 43.33%, respectively. Compared to the FM group, the Fusobacteriota in the other four groups decreased significantly (p < 0.05). The functional prediction of FAPROTAX indicated that no functional components were observed which are harmful to the body. Considering the effects on growth performance, antioxidant capacity and intestinal microbiota, it is feasible to use soybean products and chicken meal to replace 60% of fish meal in the feed of P. monodon.

Janus kinase/signal transducers and activators of transcription (JAK/STAT) signaling pathway plays an important role in antiviral immunity. This research reports the full-length DOME receptor gene in Penaeus monodon (PmDOME) and examines the effects of PmDOME and PmSTAT silencing on immune-related gene expressions in shrimp hemocytes during white spot syndrome virus (WSSV) infection. PmDOME and PmSTAT were up-regulated in shrimp hemocytes upon WSSV infection. Suppression of PmDOME and PmSTAT showed significant impacts on the expression levels of ProPO2 (melanization), Vago5 (interferon-like protein) and several antimicrobial peptides, including ALFPm3, Penaeidin3, CrustinPm1 and CrustinPm7. Silencing of PmDOME and PmSTAT reduced WSSV copy numbers and delayed the cumulative mortality caused by WSSV. We postulated that suppression of the JAK/STAT signaling pathway may activate the proPO, IFN-like antiviral cytokine and AMP production, resulting in a delay of WSSV-related mortality. This study investigated the occurrence of microplastics (MPs) in the gastrointestinal tracts (GT) and tissues of four common shrimps (including two wild-caught shrimps and two farmed shrimps) collected from a high-diversity lagoon in central Vietnam. The numbers of MP items in greasy-back shrimp (Metapenaeus ensis), green tiger shrimp (Penaeus semisulcatus), white-leg shrimp (Litopenaeus vannamei), and giant tiger shrimp (Penaeus monodon), determined per weight and individual, were 0.7 ± 0.3, 0.6 ± 0.2, 1.1 ± 0.4, and 0.5 ± 0.3 (items/g-ww), and 2.5 ± 0.5, 2.3 ± 0.7, 8.6 ± 3.5, 7.7 ± 3.5 (items/individual), respectively. The concentration of microplastics in the GT samples was significantly higher than that in the tissue samples (p < 0.05). The number of microplastics in the farmed shrimp (white-leg shrimp and black tiger shrimp) was statistically significantly higher than the number of microplastics in the wild-caught shrimp (greasy-back and green tiger shrimps) (p <0.05). Fibers and fragments were the dominant shapes of the MPs, followed by pellets, and these accounted for 42–69%, 22–57%, and 0–27% of the total microplastics, respectively. The chemical compositions determined using FTIR confirmed six polymers, in which rayon was the most abundant polymer, accounting for 61.9% of the MPs found, followed by polyamide (10.5%), PET (6.7%), polyethylene (5.7%), polyacrylic (5.8%), and polystyrene (3.8%). As the first investigation on the MPs in shrimps from Cau Hai Lagoon, central Vietnam, this study provides useful information on the occurrences and characteristics of the microplastics in the gastrointestinal tracts and tissues of four shrimp species that live in different living conditions. Simple Summary: To enhance farming efficiency, most farmers prefer to purchase Penaeus larvae as opposed to hatching them themselves. However, counting small and highly congested larvae during transactions is a challenging task to accomplish manually. We intend to improve counting precision and decrease human labor costs. In this work, an equal keypoint regression method is proposed to address these challenges. We employed five different types of smartphones to capture thousands of high-resolution images under various challenging environmental conditions. Then, we selected 1420 images to build a high-resolution dataset. In addition, this high-resolution dataset included general point annotations for use. Following training with this dataset, we obtained a model that we tested with a real Penaeus monodon larvae dataset. The results showed that the average model accuracy for the 720 images with seven density groups in the test dataset was 93.79%. Ultimately, our trained model demonstrated greater efficiency than the classical density map algorithm. Today, large-scale Penaeus monodon farms no longer incubate eggs but instead purchase larvae from large-scale hatcheries for rearing. The accurate counting of tens of thousands of larvae in these transactions is a challenging task due to the small size of the larvae and the highly congested scenes. To address this issue, we present the Penaeus Larvae Counting Strategy (PLCS), a simple and efficient method for counting Penaeus monodon larvae that only requires a smartphone to capture images without the need for any additional equipment. Our approach treats two different types of keypoints as equip keypoints based on keypoint regression to determine the number of shrimp larvae in the image. We constructed a high-resolution image dataset named Penaeus_1k using images captured by five smartphones. This dataset contains 1420 images of Penaeus monodon larvae and includes general annotations for three keypoints, making it suitable for density map counting, keypoint regression, and other methods. The effectiveness of the proposed method was evaluated on a real Penaeus monodon larvae dataset. The average accuracy of 720 images with seven different density groups in the test dataset was 93.79%, outperforming the classical density map algorithm and demonstrating the efficacy of the PLCS. Shellfish are a leading cause of allergies worldwide, affecting about one-tenth of the general population. The sarcoplasmic calcium-binding protein, also known as allergen Pen m 4, is an important factor in shrimp allergies. Our objective was to assess the most effective techniques for producing a recombinant Pen m 4 protein as a potential tool for diagnosing shrimp allergies. In this study, for the first time, we produced a functional recombinant Pen m 4 protein in a eukaryotic system, Pichia pastoris , and analyzed it against Escherichia coli -produced equivalents in enzyme-linked immunosorbent and reverse-phase protein microarray assays. A dual tag system based on the maltose-binding protein was successfully used to increase the yield of Pen m 4 by 1.3–2.3-fold in both bacteria and yeast, respectively. Immunological characterization showed that N-glycosylation is neither crucial for the folding of Pen m 4 nor its recognition by specific IgE. However, the Ca2+-depletion assay indicated a dependence on calcium ion presence in blood samples. Results demonstrate how a comparative analysis can elucidate essential allergen manufacturing points. Commercial cultivation of Litopenaeus vannamei, popularly known as white leg shrimp or vannamei shrimp, has significantly boosted the global shrimp industry over the last 10 years. Higher yield potential, increasing market demand, shorter cultivation cycles and less vulnerability to diseases prompted the major shrimp-producing countries to introduce Litopenaeus vannamei. However, as of today, no academic sources are available supporting the commercial farming of Litopenaeus vannamei in Bangladesh—an important actor in the global shrimp industry. In recent years, Bangladesh's shrimp industry has already started to lose global competitiveness due to the deficient production (0.0347 tonne/ha) of traditionally cultivated black tiger shrimp (Penaeus monodon), which is highly susceptible to white sport syndrome diseases. This opinion article aims to spark a policy stimulation justifying the immediate policy measures to introduce high-yielding Litopenaeus vannamei species to revitalize the declining shrimp industry in Bangladesh. In this study, the expression of four reference genes (β-actin, EF1-α, GAPDH, and SubF0), two prophenoloxidase genes (proPO1 and proPO2), and the distribution of phenoloxidase (PO) activity in eight tissues (carapace, cuticle, hemolymph, hepatopancreas, muscle, pereiopods, pleopods, and uropods) of black tiger shrimp (Penaeus monodon) were evaluated. According to the geNorm and NormFinder algorithms, the most stable reference gene or the best combinations of reference genes for the evaluation of proPO1 and proPO2 expression are SubF0 for cuticle, muscle, and uropods, EF1-α for hemolymph and pereiopods, EF1-α + SubF0 for carapace, β-actin + SubF0 for hepatopancreas, and β-actin + GAPDH for pleopods. In the hemolymph and uropods, proPO1 expression was significantly stronger than that of proPO2 (433 and 5 times higher, respectively). However, in hepatopancreas, proPO2 expression was significantly stronger than proPO1 (36 times higher). Trypsin cleaves the proPO zymogen into active PO in most of the studied tissues, except hepatopancreas. PO activity (units/mg protein) following trypsin activation varied among the tissues in the following order: uropods > hemolymph > pleopods > pereiopods > hepatopancreas > carapace > cuticle > muscle. The results indicate that proPO1 is dominant in hemolymph, whereas proPO2 is dominant in hepatopancreas. Shrimp are economically important candidates for aquaculture in Tanzania; however, their farming is constrained by reliance on costly feeds, attributed to the use of fishmeal as the main ingredient. This study determined the efficacy of green algae Chaetomorpha sp. on survival, feed utilization, biochemical composition, and fatty acids profile of the cultured Penaeus monodon as a potential candidate for replacing fishmeal. Four experimental feeds, F0, F1, F2, and F3, denoting fishmeal replacement levels of 0, 10, 20, and 30%, respectively, were made. Post-larvae weighing 0.49 ± 0.06 g were stocked at 15 individuals/m3 in 2 m3 hapa nets in three replicates and reared for 45 days. The F2 treatment showed a significantly higher (p < 0.05) survival rate than the other treatments. F2 and F3 improved growth significantly with F3 exhibiting the highest weight gain (8.02 ± 0.26 g) compared to the other treatments. The biochemical composition varied significantly (p<0.05) across treatment levels. The F2 treatment demonstrated the highest muscle somatic index (MSI) of 71.77 ± 2.32%. The F3 treatment exhibited significantly higher (p<0.05) crude protein (CP) than the other treatments. Crude fat (CF) in F0 and F2 treatments were significantly higher (p<0.05) than the other replacement levels. The F0 and F1 treatments exhibited higher proportions of saturated fatty acids (SFA) than the other treatments. In contrast, F2 and F3 treatments had high levels of polyunsaturated fatty acids (PUFA). The replacement of Chaetomorpha algae at 20 to 30% levels in feeds improved feed utilization, biochemical composition, and fatty acids profile of the farmed Penaeus monodon. However, the replacement of Chaetomorpha algae at 30% resulted in high saturated fatty acids but lowest survival rates. Anti-lipopolysaccharide factor is a class of antimicrobial peptides with lipopolysaccharide-binding structural domains, which has a broad antimicrobial spectrum, high antimicrobial activities, and broad application prospects in terms of the aquaculture industry. However, the low yield of natural antimicrobial peptides and their poor expression activity in bacteria and yeast have hindered their exploration and utilization. Therefore, in this study, the extracellular expression system of Chlamydomonas reinhardtii, by fusing the target gene with the signal peptide, was used to express anti-lipopolysaccharide factor 3 (ALFPm3) from Penaeus monodon in order to obtain highly active ALFPm3. Transgenic C. reinhardtii T-JiA2, T-JiA3, T-JiA5, and T-JiA6, were verified using DNA-PCR, RT-PCR, and immunoblot. Additionally, the IBP1-ALFPm3 fusion protein could be detected not only within the cells but also in the culture supernatant. Moreover, the extracellular secretion containing ALFPm3 was collected from algal cultures, and then its bacterial inhibitory activity was analyzed. The results showed that the extracts from T-JiA3 had an inhibition rate of 97% against four common aquaculture pathogenic bacteria, including Vibrio harveyi, Vibrio anguillarum, Vibrio alginolyticus, and Vibrio parahaemolyticus. The highest inhibition rate of 116.18% was observed in the test against V. anguillarum. Finally, the minimum inhibition concentration (MIC) of the extracts from T-JiA3 to V. harveyi, V. anguillarum, V. alginolyticus, and V. parahaemolyticus were 0.11 μg/μL, 0.088 μg/μL, 0.11 μg/μL, and 0.011 μg/μL, respectively. This study supports the foundation of the expression of highly active anti-lipopolysaccharide factors using the extracellular expression system in C. reinhardtii, providing new ideas for the expression of highly active antimicrobial peptides. The black tiger shrimp, Penaeus monodon, is the most economically important penaeid shrimp in Bangladesh for both culture and capture fishery. The culture of these penaeid shrimps is always risky due to different diseases and environmental stressors. This study aimed to assess the relative expression of the Penaeus monodon Rab7 gene in the post larvae (PL) of the P. monodon cultured under different salinity conditions. We challenged the PL of the P. monodon in aquariums with two different salinity and dissolved oxygen conditions during a 7-day test. The relative expression of Rab7 was upregulated during the experimental period of 36 hours. Average differences in the relative expression of Rab7 were 0.3-folds and 0.5-folds, and 0.4-folds and 0.6-folds after 18 and 36 hours of rearing in 2 and 54 ppt salinity treated shrimp PL, respectively, when ß-actin was used as an internal control. On the other hand, when elongation factor 1 alpha (EF1a) was used for normalization, the average relative expression of Rab7's fold differences were 0.2-folds and 0.5-folds, and 0.3-folds and 0.6-folds after 18 and 36 hours of rearing in 2 and 54 ppt salinity treated shrimp PL, respectively. Therefore, our results suggested that the relative expression of the Rab7 was positively correlated with an increased level of salinity and challenge duration. The significant increase in the expression level of Rab7 in the PL of P. monodon exposed to high salinity stress (54 ppt) conditions indicated its functional role in osmotic stress in shrimps. However, further studies are necessary to elucidate the mechanisms of how Rab7 is upregulated in shrimp cells after applying different levels of salinity using a larger sample group. The Mekong Delta produces about 70 % of Vietnam's national aquaculture output, and water quality management is a key factor in maintaining high levels of production and profitability. Effective water management strategies depend on understanding the spatial and temporal variations in water quality along major waterways. Therefore, this study aimed to assess the spatial and temporal fluctuation of water quality in the waterways of five provinces with intensive aquaculture operations in the Mekong Delta. The study sites included two inland provinces (An Giang and Can Tho) where striped catfish, Pangasianodon hypophthalmus (Sauvage, 1878), is farmed, and three coastal provinces (Soc Trang, Bac Lieu, and Ca Mau) where white leg shrimp, Litopenaeus vannamei (Boone, 1931), and black tiger shrimp Penaeus monodon Fabricius, 1798, are cultured. At each sampling site, physicochemical parameters were monitored for 12 months on the main rivers which supply water to aquaculture systems. Water quality data were analysed and compared based on the coefficient of variation (CV), Pearson correlation coefficients to identify relationships among variables and principal component analysis (PCA) to identify 2-3 key parameters having the most significant influence on spatial and temporal variation in water quality in each province. BOD, COD, PO43-, S2-, NO2- and NO3- were highly variable (CV 40-120 %). Significant interrelationships ranged from -0.5 to 0.5 between many water quality parameters. Overall, water in most provinces was considered polluted. However, the water quality parameters, except for S2- and NO2-, were within acceptable levels specified by National Technical Regulation on Surface Water Quality for Protection of Aquatic Life. Abstract Severe shrimp disease outbreaks have a destructive impact on shrimp aquaculture and its associated downstream food processing industries. Thus, it is essential to develop proper methods for shrimp disease control, which emphasizes the importance of food safety. In this study, we performed biochemical tests and gut microbiome analysis using uninfected control and VpAHPND‐infected Penaeus monodon samples. Biochemical tests were performed to assess the phenoloxidase (PO) activity, respiratory Burst (RB) activity, nitrite concentration, superoxide dismutase (SOD) activity, total hemocyte count (THC), and total protein concentrations. Overall, upregulations were detected in these biochemical tests, which showed the activation of the immune response in P. monodon during acute hepatopancreatic necrosis disease (AHPND) infection, especially at 6 hpi and 12 hpi. Besides that, shrimp gut samples were collected and pooled (n = 3), followed by DNA extraction, PCR amplification targeting the V3/V4 16S ribosomal RNA (rRNA) region, next‐generation sequencing (NGS), and bioinformatics analysis. Proteobacteria was the most abundant phylum in both samples. The Rhodobacteraceae family and Maritimibacter genus were proposed to be vital forshrimp health maintenance. VpAHPND bacterial colonization and secondary Vibrio infections were postulated to have occurred based on the higher abundances of Vibrionaceae family and Vibrio genus in the VpAHPND‐infected sample. Firmicutes phylum together with Photobacterium and Aliiroseovarius genera were inferred to be pathogenic or related factors of AHPND infections. In conclusion, physiology (immune response activation) and gut microbiome changes of disease tolerant P. monodon during AHPND infection were identified. Both biochemical tests and 16S rRNA analysis are proposed as a combined strategy for shrimp health diagnosis for ensuring shrimp health maintenance, disease control, and food safety.

Antibiotics have been used to control the aquatic environment in both therapeutic and prophylactic ways. Antibiotics are particularly difficult to extract due to their strong interactions with biological matrices. In this study, UPLC-MS/MS method was developed and validated for quantitative confirmatory analysis of multi-class antibiotics residues in fish and shrimp. Fourteen antibiotics belonging to sulphonamides, β-lactams, quinolones, sulfones and macrolides were determined within one chromatographic run. The samples were suspended in 0.1 M HCl, and the analytes were extracted into ethyl acetate. The extracts were defatted with cyclohexane. The limits of quantification (LOQ) ranged from 0.24 to 1.32 µg kg−1 for fish and 0.42–1.62 µg kg−1 for shrimp samples. The recoveries ranged from 75 to 105%. The method was applied to the analysis of farmed freshwater Tilapia fish (Oreochromis niloticus) and shrimp (Penaeus monodon) collected in Sri Lanka. Sulfacetamide (4.31 ± 0.70 µg kg−1) and sulfamethoxypyridazine (0.75 ± 0.15 µg kg−1) were detected in the fish, and sulfapyridine (0.21–0.56 µg kg−1) and sulfadoxine (0.35–1.44 µg kg−1) were detected in the shrimp samples. The taxonomic diversity of penaeid shrimps from the northwest coast of Peninsular Malaysia was investigated based on morphometric and molecular approaches. For geometric morphometrics (GM) approach, eighteen homologous landmarks were analyzed with principal component analysis (PCA) and canonical variate analysis (CVA) in Morpho J software. The morphological variations among species were attributed to body shape, rostrum, carapace head, and telson. The first four components accounted for 87.27% in the PCA, while for CVA, the first three components contributed to 78.47%. Although not absolute, there is a tendency for closely related species to cluster together. The CVA analyses clearly differentiated Metapenaeopsis stridulans, Megokris sedili, Metapenaeus brevicornis and Mierspenaeopsis sculptilis into discrete clusters, and highlighted the closeness of the groups; Mierspenaeopsis sculptilis - M. hardwickii, Penaeus merguiensis - P. semisulcatus, Metapenaeus affinis - M. dobsoni - M. ensis, and Penaeus monodon - P. pulchricaudatus. Molecular phylogeny among these species of penaeid shrimp were examined using mitochondrial cytochrome c oxidase subunit I (COI) gene sequences. Geometric morphometric analyses revealed shape overlap among the 12 shrimp species, yet significant differences were also detected. The morphometric and molecular multispecies analyses were largely in agreement. The phylogenetic signal was assessed by mapping the morphometric data onto three phylogenetic trees (Neighbour Joining-NJ, Maximum Likelihood-ML, and Bayesian Inference-BI) generated from the partial mitochondrial COI on the same 12 species. Results revealed non-significance (no phylogenetic signal) for NJ but significant phylogenetic signals (evolutionary significance) for ML and BI, suggesting that the shape difference among the penaeid shrimp species investigated was related to their evolutionary history. The NJ tree is prone to errors when dealing with deeper divergence times, whereas ML and BI trees are ideal for phylogeny tree reconstruction, which applies a model of sequence evolution on the data. The objective of this study was to evaluate the impact of varying concentrations of longkong pericarp extract (LPE) on the physicochemical properties of alginate-based edible nanoparticle coatings (NP-ALG) on shrimp. For developing the nanoparticles, the alginate coating emulsion with different LPE concentrations (0.5, 1.0, and 1.5%) was ultrasonicated at 210 W with a frequency of 20 kHz for 10 min and a pulse duration of 1s on and 4 off. After that, the coating emulsion was separated into four treatments (T): T1: Coating solution containing basic ALG composition and without the addition of LPE or ultrasonication treatment; T2: ALG coating solution converted into nano-sized particles with ultrasonication and containing 0.5% LPE; T3: ALG coating solution converted into nano-sized particles with ultrasonication and containing 1.0% LPE; T4: ALG coating solution converted into nano-sized particles with ultrasonication and containing 1.5% LPE. A control (C) was also used, where distilled water was used instead of ALG coating. Before coating the shrimp, all the coating materials were tested for pH, viscosity, turbidity, whiteness index, particle size, and polydispersity index. The control samples had the highest pH and whiteness index and was followed by the lowest viscosity and turbidity (p < 0.05). Among the T1–T4 coating materials, T4 coating had higher turbidity, particle size, polydispersity index, but lower pH, viscosity, and whiteness index (p < 0.05). To study the quality and shelf-life of the shrimp, all coated shrimp samples were refrigerated at 4 °C for a period of 14 days. At 2-day intervals, physiochemical and microbial analyses were performed. The coated shrimp also had a lower increase in pH and weight loss over the storage period (p < 0.05). Coatings containing 1.5% LPE significantly reduced the polyphenol oxidase activity in the shrimp (p > 0.05). The addition of LPE to NP-ALG coatings demonstrated dose-dependent antioxidant activity against protein and lipid oxidation. The highest LPE concentration (1.5%) led to increased total and reactive sulfhydryl content, along with a significant decrease in carbonyl content, peroxide value, thiobarbituric acid reactive substances, p-anisidine, and totox values at the end of the storage period (p < 0.05). Additionally, NP-ALG-LPE coated shrimp samples exhibited an excellent antimicrobial property and significantly inhibited the growth of total viable count, lactic acid bacteria, Enterobacteriaceae, and psychotropic bacteria during storage. These results suggested that NP-ALG-LPE 1.5% coatings effectively maintained the quality as well as extended the shelf-life of shrimp during 14 days of refrigerated storage. Background: Salinity is one of the main influencing factors in the culture environment and is extremely important for the survival, growth, development and reproduction of aquatic animals. Methods: In this study, a comparative transcriptome analysis (maintained for 45 days in three different salinities, 30 psu (HC group), 18 psu (MC group) and 3 psu (LC group)) was performed by high-throughput sequencing of economically cultured Penaeus monodon. P. monodon gill tissues from each treatment were collected for RNA-seq analysis to identify potential genes and pathways in response to low salinity stress. Results: A total of 64,475 unigenes were annotated in this study. There were 1,140 upregulated genes and 1,531 downregulated genes observed in the LC vs. HC group and 1,000 upregulated genes and 1,062 downregulated genes observed in the MC vs. HC group. In the LC vs. HC group, 583 DEGs significantly mapped to 37 signaling pathways, such as the NOD-like receptor signaling pathway, Toll-like receptor signaling pathway, and PI3K-Akt signaling pathway; in the MC vs. HC group, 444 DEGs significantly mapped to 28 signaling pathways, such as the MAPK signaling pathway, Hippo signaling pathway and calcium signaling pathway. These pathways were significantly associated mainly with signal transduction, immunity and metabolism. Conclusions: These results suggest that low salinity stress may affect regulatory mechanisms such as metabolism, immunity, and signal transduction in addition to osmolarity in P. monodon. The greater the difference in salinity, the more significant the difference in genes. This study provides some guidance for understanding the low-salt domestication culture of P. monodon. At present, food security is a matter of debate of global magnitude and fulfilling the feeding requirement of > 8 billion human populations by 2030 is one of the major concerns of the globe. Aquaculture plays a significant role to meet the global food requirement. Shrimp species such as Litopenaeus vannamei, Penaeus monodon, and Macrobrachium rosenbergii are among the most popular food commodities worldwide. As per Global Outlook for Aquaculture Leadership survey, disease outbreaks have been a matter of concern from the past many decades regarding the shrimp aquaculture production. Among the past disease outbreaks, white spot disease caused by the white spot syndrome virus is considered to be one of the most devastating ones that caused colossal losses to the shrimp industry. Since the virus is highly contagious, it spreads gregariously among the shrimp population; hence, practicing proper sanitization practices is crucial in order to have disease-free shrimps. Additionally, in order to control the disease, antibiotics were used that further leads to bioaccumulation and biomagnification of antibiotics in several food webs. The bioaccumulation of the toxic residues in the food webs further adversely affected human too. Recently, immunostimulants/antivirals were used as an alternative to antibiotics. They were found to enhance the immune system of shrimps in eco-friendly manner. In context to this, the present paper presents a critical review on the immunostimulants available from plants, animals, and chemicals against WSSV in shrimps. Looking into this scenario, maintaining proper sanitation procedures in conjunction with the employment of immunostimulants may be a viable approach for preserving shrimp aquaculture across the globe. Some Vibrio species can cause food-borne diseases in humans, including cholera, vomiting, septicemia, and gastroenteritis, which are associated with the consumption of contaminated seafood products. The study was conducted to detect antimicrobial-resistant Vibrio species in shrimp and shrimp environments in Bangladesh. Samples of shrimp (n = 50), water (n = 50), and mud (n = 50) were collected aseptically from 50 different shrimp culture ponds in the Khulna region of Bangladesh. Identification of Vibrio species was based on cultural and staining characteristics, biochemical tests, and polymerase chain reaction (PCR). Antimicrobial resistance profiles were determined using a disk diffusion assay. By PCR, Vibrio isolates were found in 34% (95% CI: 26.9%–41.9%) of the samples, of which the detection rate was significantly higher in shrimp (54%), compared to mud (26%) and water (22%). Moreover, V. cholerae, V. parahaemolyticus, and V. alginolyticus were detected in 24.7%, 15.3%, and 4% of the samples, respectively. Among them, the detection rate of V. cholerae and V. alginolyticus was significantly higher in shrimp samples than in other samples. V. parahaemolyticus was also higher in the shrimp samples, but the difference was not statistically significant. Vibrio isolates showed high to moderate resistance (92.2%–15.7%) to ampicillin, amikacin, cefotaxime, tetracycline, ceftazidime, gentamicin, nalidixic acid, levofloxacin, and ciprofloxacin, and low resistance (3.9%) to imipenem, meropenem, chloramphenicol, and trimethoprim-sulfamethoxazole. Interestingly, 52.9% of the isolates were multidrug resistant, and the multiple antibiotic resistance index was up to 1.0. To our knowledge, this is the first study in Bangladesh detecting these three Vibrio species (V. parahaemolyticus, V. alginolyticus, and V. cholerae) from shrimp and shrimp environments by molecular approach in the same study. These findings reveal the alarmingly high occurrence of antimicrobial-resistant Vibrio species in shrimp and shrimp environments, which should be of concern to both the shrimp industry and public health management.
