The decline of insect pollinators is a global concern, and the use of pesticides has been identified as a potential cause for it. Glyphosate‐based herbicides (GBHs) are the world's most used pesticides, but until recent years they have been claimed to be safe for non‐target organisms, such as pollinators. Using controlled arena experiments, we investigated the effects on the learning and long‐term memory of buff‐tailed bumblebees, Bombus terrestris (L.) (Hymenoptera: Apidae), of a single field‐realistic dose of glyphosate, both in its pure form and as a commercial herbicide (Roundup Gold) containing the active ingredient (a.i.) glyphosate and other co‐formulants. We found that glyphosate in its pure form caused deterioration in the learning ability of bumblebees in a 10‐color discrimination experiment; the glyphosate‐treated bees discriminated colors over 10% worse than the untreated control bees. However, the commercially used GBH (Roundup Gold) had no observable effect on the learning ability of the bumblebees, despite the fact that this herbicide contained the same amount of glyphosate as its pure form. These findings shed light on the potential risks associated with agrochemicals previously considered safe for pollinators and emphasize the need for comprehensive risk assessments of pesticides, including evaluations of cognitive functions in pollinators. The transmission of pathogens from reservoir to recipient host species, termed pathogen spillover, can profoundly impact plant, animal, and public health. However, why some pathogens lead to disease emergence in a novel species while others fail to establish or do not elicit disease is often poorly understood. There is strong evidence that deformed wing virus (DWV), an (+)ssRNA virus, spills over from its reservoir host, the honeybee Apis mellifera, into the bumblebee Bombus terrestris. However, the low impact of DWV on B. terrestris in laboratory experiments suggests host barriers to virus spread in this recipient host. To investigate potential host barriers, we followed the spread of DWV genotype B (DWV-B) through a host's body using RT-PCR after experimental transmission to bumblebees in comparison to honeybees. Inoculation was per os, mimicking food-borne transmission, or by injection into the bee's haemocoel, mimicking vector-based transmission. In honeybees, DWV-B was present in both honeybee faeces and haemolymph within 3 days of inoculation per os or by injection. In contrast, DWV-B was not detected in B. terrestris haemolymph after inoculation per os, suggesting a gut barrier that hinders DWV-B's spread through the body of a B. terrestris. DWV-B was, however, detected in B. terrestris faeces after injection and feeding, albeit at a lower abundance than that observed for A. mellifera, suggesting that B. terrestris sheds less DWV-B than A. mellifera in faeces when infected. Barriers to viral spread in B. terrestris following oral infection may limit DWV's impact on this spillover host and reduce its contribution to the community epidemiology of DWV. Animals are often assumed to follow a strategy of energy maximization, and therefore should evaluate feeding options based on energy intake rates. However, at the proximal level, a direct estimate of energy intake rates, if that is possible at all, might require postabsorptive senses with relatively longer processing times, whereas an indirect estimate of energy intake through proxies like pre-absorptive senses of different sensory food properties might support rapid foraging decisions. Here, we show that nectar sugar concentration (sweetness) and nectar viscosity (resistance) drive preferences of bumble bees, Bombus terrestris , classical models for economic and foraging decision making. Using a tasteless/odourless biopolymer (Tylose), we created feeding options that differed in sweetness and resistance, properties that affect energy intake rate and can be immediately sensed. When energy intake rates were similar, bumble bees developed preferences based on sweetness and resistance. When energy intake rates were different, but sweetness and resistance were balanced against each other, bees developed no preferences. Decision dynamics during training indicated that bumble bees simultaneously evaluated sweetness and resistance to make decisions quickly (in seconds). These results suggest that bumble bees' food preferences are jointly affected by the immediate sensation of nectar sweetness and resistance as positively and negatively reinforcing properties, respectively, indicating a pre-absorptive proximate mechanism for rapid energy-sensitive decisions in bumble bees. • Bumble bees develop preferences based on nectar sugar concentration and viscosity. • Bumble bees quickly sample and compare the sensory properties of feeding options. • Pre-ingestive information contributes to bumble bees' floral preferences. Greenhouse tomato production is a growing business worldwide, and it is strongly promoted by bumblebee pollination. Although there are over 250 bumblebee species worldwide, very few species have been reared successfully for greenhouse tomato pollination. Those successfully managed species, especially Bombus terrestris, are shipped around the world for commercial use. However, managed bumblebees are known to escape greenhouse facilities, have established local populations, spread disease to local bumblebees, and are blamed for the declines of some indigenous bee species. An alternative to shipping exotic bumblebees around the world is to develop local species for greenhouse pollination. Such an approach has the dual benefits of creating a new industry of insect rearing while reducing threats to local bee communities. In this study, we successfully reared Bombus haemorrhoidalis, which is the most common bumblebee species in Northern Pakistan, in a laboratory and compared its effectiveness as a tomato pollinator with that of commercial B. terrestris in a greenhouse. We found that the effectiveness of B. haemorrhoidalis in tomato pollination in a greenhouse is very similar to that of B. terrestris when it comes to the fruit size, number of seeds, and fruit weight. Our study provides an example of how to rear a native bumblebee species to pollinate local crops, which is a method that could potentially substitute the importation of non-ingenious bumblebees. Question: The large earth bumble bee (Bombus terrestris) maintains a social core gut-microbiota, similar as known from the honey bee, which plays an important role for host health and resistance. Experiments under laboratory conditions with commercial hives are limited to vertically transmitted microbes and neglect influences of environmental factors or external acquisition of microbes. Various environmental and landscape-level factors may have an impact on the gutmicrobiota of pollinating insects, with consequences for pollinator health and fitness in agroecosystems. Still, it is not fully clear whether access to different flower diversities will have a significant influence on the bumble bee microbiota. Here, we tested in a semi-field experiment if the bumble bee microbiota changes over time when exposed to different flower diversities within outdoor flight cages. We used commercial hives to distinguish between vertically and horizontally transmitted bacteria, respectively from the nest environment or the exposed outside environment. Result: The sequential sampling of foraging workers over a period of 35 days indicated a temporal progression of the bumble bee microbiota when placed outside. The microbiota increased in diversity and changed in composition and variability over time. We observed a major increase in relative abundance of the families Lactobacillaceae, Bifidobacteriaceae and Weeksellaceae. In contrast, major core-taxa like Snodgrassella and Gilliamella declined in their relative abundance over time. The genus Lactobacillus showed a high diversity and strain specific turnover, so that only specific ASVs showed an increase over time, while others had a more erratic occurrence pattern. Exposure to different flower diversities had no significant influence on the progression of the bumble bee microbiota. Conclusion: The bumble bee microbiota showed a dynamic temporal succession with distinct compositional changes and diversification over time when placed outdoor. The exposure of bumble bees to environmental conditions, or environmental microbes, increases dissimilarity and changes the gut-community composition. Insect pollination is an important process in the natural ecosystem and plays a vital role in crop reproduction. The bumblebee, Bombus terrestris (L.), is a widely used pollinator for crops, especially tomato. The efficiency of bee pollination is related to many factors, including biotic factors such as bee species, nutrition plants, and herbivores. Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) is one of the most common and serious pests of tomato plants. However, whether B. tabaci affects the pollination behaviour of B. terrestris on tomato, and how plant–herbivore–pollinator interactions are unclear. In this study, we found that B. terrestris preferred B. tabaci-damaged plants. Furthermore, plant volatiles of tomato plants damaged by B. tabaci were detected and analysed. Electroantennogram (EAG) and the Y-tube bioassay were used for olfactory responses of B. terrestris to plant volatiles. Interestingly, methyl hexanoate and eugenol induced by B. tabaci in tomato plants were found to significantly attract B. terrestris. Therefore, Bemisia tabaci influenced B. terrestris preference through volatiles. The plant volatile-mediated interaction between B. tabaci and B. terrestris could enrich our understanding of the interaction between herbivores and pollinators. Purpose: Bumblebees are an important group of insects in the pollination of various vegetables, fruits, oilseeds, legumes, and the fodder crops. Compared to honeybees, they have a wider choice of hosts and a longer flight period. These bees are used especially for the pollination of plants in greenhouses and are commercially produced for this purpose. Recently, serious decreases have been occurring in bumblebee populations due to various reasons such as pathogens, and some of species are even threatened with extinction. Due to the worldwide decline in pollinator insects, determining the distribution and prevalence of bumblebee pathogens is of great importance. Therefore, this study was conducted to determine the incidence and prevalence of pathogens in Turkish bumblebee populations and how much of each pathogen was in bumblebee samples. Methods: A total of 172 Bombus terrestris (Linnaeus,1758) samples (21 samples from commercial enterprises, 79 samples from greenhouses and 72 samples from nature) were randomly collected from 3 provinces (Antalya, Mersin and İzmir) where greenhouse cultivation is intensively carried out in Turkey. Eighty-nine of these samples were collected in the spring and eighty-three in the autumn. The presence of four pathogens (Nosema bombi, Crithidia bombi, Apicystis bombi, and Locustacarus buchneri) was investigated by PCR using universal primers. Results: The overall prevalence of Nosema bombi, Crithidia bombi, Apicystis bombi, and Locustacarus buchneri was determined as 7.55%, 9.3%, 11.62%, and 4.65%, respectively. Co-infections (5.81%) were only detected in wild-caught (nature) samples. C. bombi and A. bombi infections were detected at higher rates in the spring samples than in the autumn samples (p < 0.05). There was no significant difference between the spring and autumn samples with respect to the presence of N. bombi and L. buchneri (p > 0.05). Conclusion: The results obtained could be important in determining the prevalence and spread rates of the bumblebee diseases in Turkey and to determine appropriate protection measures. The information gathered should increase our knowledge about the presence of these pathogens in Turkey and could contribute to improve apiarist's practice. More studies are needed to determine the transmission pathways of these pathogens between the populations. Climate change is currently considered one of the major threats to biodiversity and is associated with an increase in the frequency and intensity of extreme weather events, such as heatwaves. Heatwaves create acutely stressful conditions that may lead to disruption in the performance and survival of ecologically and economically important organisms, such as insect pollinators. In this study, we investigated the impact of simulated heatwaves on the performance of queenless microcolonies of Bombus terrestris audax under laboratory conditions. Our results indicate that heatwaves can have significant impacts on bumblebee performance. However, contrary to our expectations, exposure to heatwaves did not affect survival. Exposure to a mild 5-day heatwave (30-32 ℃) resulted in increased offspring production compared to those exposed to an extreme heatwave (34-36 ℃) and to the control group (24 ℃). We also found that brood-care behaviours were impacted by the magnitude of the heatwave. Wing fanning occurred occasionally at temperatures of 30-32 ℃, whereas at 34-36 ℃ the proportion of workers engaged in this thermoregulatory behaviour increased significantly. Our results provide insights into the effects of heatwaves on bumblebee colony performance and underscore the use of microcolonies as a valuable tool for studying the effects of extreme weather events. Future research, especially field-based studies replicating natural foraging conditions, is crucial to complement laboratory-based studies to comprehend how heatwaves compromise the performance of pollinators. In recent years, a significant reduction in the number of insect pollinators has been reported. During the period 2021-2022, a study was carried out of pollinator types in tobacco cenosis at the experimental field of the Institute of Tobacco and Tobacco Products – Markovo. The study was carried out on oriental tobacco ecotype Krumovgrad (variety Krumovgrad 90) in area of 0.2 ha. The plants were grown under irrigation conditions and compliance with all agrotechnical measures. The survey was carried out during the period of mass flowering of tobacco by visual observations and the use of an entomological bag. The bee species Apis mellifera L., Bombus terrestris L., as well as flies from family Syrphidae-Episyrphus balteatus De Geer, Scaeva pyrastri L., Sphaerophoria spp. and Syrphus ribesii L. were established. In 2021, out of 679 reported pollinators, the most were representatives of honey bee A. mellifera- 618, followed by B. terrestris- 34 and the representatives of family Syrphidae –27. Food security is threatened by climate change and associated abiotic stresses that affect the flowering stage and the biochemistry of flowers and fruits. In tomato, managed insect pollination and grafting elite tomato varieties onto robust rootstocks are widely practiced commercially to enhance tomato crop profitability, particularly under suboptimal conditions. However, little is known about rootstock–pollinator interactions and their impact on the chemical composition of fruit. In this study, a commercial tomato F1 hybrid (Solanum lycopersicum L.) was self-grafted and grafted onto a set of experimental rootstocks and cultivated under optimal and saline (75 mM NaCl) conditions in the presence of managed bumblebee pollinators (Bombus terrestris). The number of visits (VN) and total visiting time (TVT) by pollinators to different grafted plants were monitored through an RFID (radio-frequency identification) tracking system, while targeted metabolites (hormones, sugars, and organic and amino acids) and mineral composition were analyzed in the fruit juice by UHPLC-MS and ICP-OES, respectively. Pollinator foraging decisions were influenced by the rootstocks genotype and salinity treatment. Experimental rootstocks predominantly increased pollinator attraction compared to the self-grafted variety. Interestingly, the pollinator parameters were positively associated with the concentration of abscisic acid, salicylic acid, malate and fumarate, and tyrosine in salinized fruits. Moreover, a high accumulation of sodium was detected in the fruits of the plants most visited by pollinators, while rootstock genotype-specific responses were found for nitrogen and potassium concentrations. In addition to the known effect on yield, these findings underscore the synergic interactions between rootstocks, pollinators, and environmental stressors on tomato fruit composition.

Infectious and parasitic agents (IPAs) and their associated diseases are major environmental stressors that jeopardize bee health, both alone and in interaction with other stressors. Their impact on pollinator communities can be assessed by studying multiple sentinel bee species. Here, we analysed the field exposure of three sentinel managed bee species (Apis mellifera, Bombus terrestris and Osmia bicornis) to 11 IPAs (six RNA viruses, two bacteria, three microsporidia). The sentinel bees were deployed at 128 sites in eight European countries adjacent to either oilseed rape fields or apple orchards during crop bloom. Adult bees of each species were sampled before their placement and after crop bloom. The IPAs were detected and quantified using a harmonised, high-throughput and semi-automatized qPCR workflow. We describe differences among bee species in IPA profiles (richness, diversity, detection frequencies, loads and their change upon field exposure, and exposure risk), with no clear patterns related to the country or focal crop. Our results suggest that the most frequent IPAs in adult bees are more appropriate for assessing the bees' IPA exposure risk. We also report positive correlations of IPA loads supporting the potential IPA transmission among sentinels, suggesting careful consideration should be taken when introducing managed pollinators in ecologically sensitive environments. Bumblebees are important pollinators in both natural and agricultural ecosystems, but their survival and pollination service are threatened by extensive pesticide use. Due to regulation changes, acetamiprid has become the only neonicotinoid substance that can be used without restrictions and in open‐field cultivations in the European Union. Yet, we know little about how this active ingredient affects bumblebees' foraging behaviour and if such effects are similarly detrimental to those of other neonicotinoids. Here we investigated how a 14‐day‐long (chronic) exposure to low (5 ppb) and high (2500 ppb) concentrations of acetamiprid in syrup affected different aspects of foraging behaviour in buff‐tailed bumblebees (Bombus terrestris, Linnaeus). We recorded individual foraging at artificial food patches during one‐hour‐long trials and then compared several foraging‐related measures between differently dosed individuals. We found that 75.12% of the total syrup consumption occurred at the first‐exploited patch, but individuals did not exhibit any bias toward pesticide‐treated food patches. Chronic exposure to acetamiprid had little effect on individual foraging decisions at the first‐exploited food patch but significantly affected the time to the first feeding and the number of feedings at the second‐exploited patch in interaction with body mass. The duration of the first feeding was affected only by foragers' body mass. Our finding indicates that chronic exposure to a high but field‐realistic concentration of acetamiprid may alter some aspects of bumblebees' foraging behaviour. If such behavioural changes accumulate during consecutive foraging bouts, chronic exposure to this pesticide might lead to a reduction of daily resource collection, ultimately jeopardising colony fitness or plant (crop) pollination. Invasive species are predicted to adjust their morphological, physiological and life‐history traits to adapt to their non‐native environments. Although a loss of genetic variation during invasion may restrict local adaptation, introduced species often thrive in novel environments. Despite being founded by just a few individuals, Bombus terrestris (Hymenoptera: Apidae) has in less than 30 years successfully spread across the island of Tasmania (Australia), becoming abundant and competitive with native pollinators. We use RADseq to investigate what neutral and adaptive genetic processes associated with environmental and morphological variation allow B. terrestris to thrive as an invasive species in Tasmania. Given the widespread abundance of B. terrestris, we expected little genetic structure across Tasmania and weak signatures of environmental and morphological selection. We found high gene flow with low genetic diversity, although with significant isolation‐by‐distance and spatial variation in effective migration rates. Restricted migration was evident across the mid‐central region of Tasmania, corresponding to higher elevations, pastural land, low wind speeds and low precipitation seasonality. Tajima's D indicated a recent population expansion extending from the south to the north of the island. Selection signatures were found for loci in relation to precipitation, wind speed and wing loading. Candidate loci were annotated to genes with functions related to cuticle water retention and insect flight muscle stability. Understanding how a genetically impoverished invasive bumblebee has rapidly adapted to a novel island environment provides further understanding about the evolutionary processes that determine successful insect invasions, and the potential for invasive hymenopteran pollinators to spread globally. This note documents the first record of the invasive buff‐tailed bumblebee Bombus terrestris nesting in an above‐ground nest box deployed for birds in Australia. The box in question had been installed as nesting habitat for the endangered forty‐spotted pardalote Pardalotus quadragintus on Bruny Island, Tasmania. The bumblebee nest appeared to be constructed on a partially built pardalote nest inside the box and so may have caused the abandonment of a nesting attempt by pardalotes. However, the remains of this bumblebee nest were not large enough to prevent forty‐spotted pardalotes constructing a nest in the box during the subsequent breeding season. Further research will be needed to ascertain the degree to which bumblebees disrupt breeding of forty‐spotted pardalotes. [ABSTRACT FROM AUTHOR]

Some species of bumblebees can be discriminated from each other based on distinct morphological features, while for some species, morphological characteristics are not sufficient for identification. The use of wing vein characteristics is one of the important methods used in the identification of bee species or subspecies. In Bombus terrestris colonies, individuals belonging to four different caste groups are grown throughout the life cycle, namely (i) worker bees, (ii) young queen bees, (iii) male bees produced from haploid eggs laid by the founding queen, and (iv) male bees produced from haploid eggs laid by worker bees. In this study, wing vein characteristics of individuals belonging to these 4 different caste groups produced in B. terrestris colonies were examined, and values for these characteristics were determined. For this purpose, 160 individuals (40 for each caste group) were used in the study. The difference between the groups for each characteristic determined in the study was found statistically significant (P<0.05). The results of the study are of a nature to contribute to future research on wing characteristics of bumblebees. [ABSTRACT FROM AUTHOR]

Diapause, an adaptative strategy for survival under harsh conditions, is a dynamic multi-stage process. Bombus terrestris, an important agricultural pollinator, is declining in the wild, but artificial breeding is possible by imitating natural conditions. Mated queen bees enter reproductive diapause in winter and recover in spring, but the regulatory mechanisms remain unclear. Herein, we conducted a comparative 4D label-free proteomic analysis of queen bees during artificial breeding at seven timepoints, including pre-diapause, diapause, and post-diapause stages. Through bioinformatics analysis of proteomic and detection of substance content changes, our results found that, during pre-diapause stages, queen bees had active mitochondria with high levels of oxidative phosphorylation, high body weight, and glycogen and TAG content, all of which support energy consumption during subsequent diapause. During diapause stages, body weight and water content were decreased but glycerol increased, contributing to cold resistance. Dopamine content, immune defense, and protein phosphorylation were elevated, while fat metabolism, protein export, cell communication, signal transduction, and hydrolase activity decreased. Following diapause termination, JH titer, water, fatty acid, and pyruvate levels increased, catabolism, synaptic transmission, and insulin signaling were stimulated, ribosome and cell cycle proteins were upregulated, and cell proliferation was accelerated. Meanwhile, TAG and glycogen content decreased, and ovaries gradually developed. These findings illuminate changes occurring in queen bees at different diapause stages during commercial production. Many pollinators, such as bees, hummingbirds and bats, use multi-destination routes (traplines) to exploit familiar plant resources. However, it is not clear to what extent the mechanisms underpinning trapline development and optimisation are comparable across species. Here we compared route formation, repeatability and efficiency by foragers of two social bee species, the solo foraging bumblebee Bombus terrestris and the mass foraging honey bee Apis mellifera, in the same laboratory conditions. In a simple routing task (with four artificial flowers), all bumblebees and honey bees developed a route, although honey bees were slower to do so. In a more complex routing task (with six flowers), however, only bumblebees developed a route between the 6 flowers. Honey bees took a longer time to discover all flowers and developed routes between fewer flowers. Comparing bumblebees and honey bees using the same experimental paradigm thus revealed key behavioural differences likely resulting from their contrasting collective foraging strategies. Significance statement: Pollinators face some of the most complex foraging challenge among animals: collecting small amounts of nectar from large numbers of flowers in a minimum of time. Previous studies report bees, birds and bats rely on spatial memory to develop routes optimising foraging efficiency. Here we compared this routing behaviour in two major pollinators, the buff-tailed bumblebee and the Western honey bee, in the same experimental conditions. We found that bumblebees were much faster and more efficient at visiting flowers following repeatable sequences than honey bees. In uncertain environments, animals often face the challenge of deciding whether to stay with their current foraging option or leave to pursue the next opportunity. The voluntary decision to persist at a location or with one option is a critical cognitive ability in animal temporal decision-making. Little is known about whether foraging insects form temporal expectations of reward and how these expectations affect their learning and rapid, short-term foraging decisions. Here, we trained bumblebees on a simple colour discrimination task whereby they entered different opaque tunnels surrounded by coloured discs (artificial flowers) and received reinforcement (appetitive sugar water or aversive quinine solution depending on flower colour). One group received reinforcement immediately and the other after a variable delay (0–3 s). We then recorded how long bees were willing to wait/persist when reinforcement was delayed indefinitely. Bumblebees trained with delays voluntarily stayed in tunnels longer than bees trained without delays. Delay-trained bees also waited/persisted longer after choosing the reward-associated flower compared to the punishment-associated flower, suggesting stimulus-specific temporal associations. Strikingly, while training with delayed reinforcement did not affect colour discrimination, it appeared to facilitate the generalisation of temporal associations to ambiguous stimuli in bumblebees. Our findings suggest that bumblebees can be trained to form temporal expectations, and that these expectations can be incorporated into their decision-making processes, highlighting bumblebees' cognitive flexibility in temporal information usage. Significance statement: The willingness to voluntarily wait or persist for potential reward is a critical aspect of decision-making during foraging. Investigating the willingness to persist across various species can shed light on the evolutionary development of temporal decision-making and related processes. This study revealed that bumblebees trained with delays to reinforcement from individual flowers were able to form temporal expectations, which, in turn, generalised to ambiguous stimuli. Honeybees are well-established bioindicators for different types of pollutants. This study aims to establish another species of the Apidae family as a bioindicator, with a distinct behaviour and life cycle. The bumblebee Bombus terrestris was used as a bioindicator for 12 metals. Bumblebee hives were placed at sampling sites in and around the city of Aachen, Germany, and metal concentrations were assessed using ICP-MS. Metal concentrations were compared to those found in honeybees described in the literature. Spatial differences in metal patterns were investigated by comparing two land-use types: urban and agrarian. Seasonal differences were compared by taking samples in spring and summer. All analysed metals were detected above the detection limit and within or even above the concentration range found in honeybees. Significant spatial differences were found for the metalloid B and the metal Cd with higher concentrations at the agrarian sites than the urban sites. Significant seasonal differences were found for 8 metals: Fe, Zn, Cu, Ni, Cd, and As concentrations were higher in summer than in spring, while B and V concentrations were higher in spring. To categorise the results, we applied the honeybee contamination index (HCI) and adapted it to bumblebee purposes. According to the HCI, only one agrarian site showed a high contamination level. This study shows that bumblebees are suitable bioindicators for metals. The obtained data can serve as a first baseline in the establishment of additional monitoring studies or risk assessments. Aims Diets and parasites influence the gut bacterial symbionts of bumble bees, but potential interactive effects remain overlooked. The main objective of this study was to assess the isolated and interactive effects of sunflower pollen, its phenolamides, and the widespread trypanosomatid Crithidia sp. on the gut bacterial symbionts of Bombus terrestris males. Methods and results Bumble bee males emerged in microcolonies fed on either (i) willow pollen (control), (ii) sunflower pollen, or (iii) willow pollen spiked with phenolamide extracts from sunflower pollen. These microcolonies were infected by Crithidia sp. or were pathogen-free. Using 16S rRNA amplicon sequencing (V3–V4 region), we observed a significant alteration of the beta diversity but not of the alpha diversity in the gut microbial communities of males fed on sunflower pollen compared to males fed on control pollen. Similarly, infection by the gut parasite Crithidia sp. altered the beta diversity but not the alpha diversity in the gut microbial communities of males, irrespective of the diet. By contrast, we did not observe any significant alteration of the beta or alpha diversity in the gut microbial communities of males fed on phenolamide-enriched pollen compared to males fed on control pollen. Changes in the beta diversity indicate significant dissimilarities of the bacterial taxa between the treatment groups, while the lack of difference in alpha diversity demonstrates no significant changes within each treatment group. Conclusions Bumble bees harbour consistent gut microbiota worldwide, but our results suggest that the gut bacterial communities of bumble bees are somewhat shaped by their diets and gut parasites as well as by the interaction of these two factors. Recent developments allowed establishing virtual‐reality (VR) setups to study multiple aspects of visual learning in honey bees under controlled experimental conditions. Here, we adopted a VR environment to investigate the visual learning in the buff‐tailed bumble bee Bombus terrestris. Based on responses to appetitive and aversive reinforcements used for conditioning, we show that bumble bees had the proper appetitive motivation to engage in the VR experiments and that they learned efficiently elemental color discriminations. In doing so, they reduced the latency to make a choice, increased the proportion of direct paths toward the virtual stimuli and walked faster toward them. Performance in a short‐term retention test showed that bumble bees chose and fixated longer on the correct stimulus in the absence of reinforcement. Body size and weight, although variable across individuals, did not affect cognitive performances and had a mild impact on motor performances. Overall, we show that bumble bees are suitable experimental subjects for experiments on visual learning under VR conditions, which opens important perspectives for invasive studies on the neural and molecular bases of such learning given the robustness of these insects and the accessibility of their brain. This study explored the flower visiting behaviors and pollination abilities of mason bees (Osmia excavata Alfken (Hymenoptera: Megachilidae)), bumble bees (Bombus terrestris (Linnaeus, 1758) (Hymenoptera: Apidae)), and Italian honey bees (Apis mellifera ligustica Spinola (Hymenoptera: Apidae)) in apple orchards in early spring in Jinan (located in the central region of Shandong) and Yantai (located in the Peninsula of Shandong). We compared the pollen collection patterns, flower visiting behavior, flying speed, and effects on apple pollination of the 3 types of bees. The frequencies of flower visits were significantly higher for mason bees (12.89/min in Jinan and 10.63/min in Yantai) than bumble bees and Italian honey bees in the 2 regions. The single flower residence times were significantly higher for Italian honey bees (8.22 s in Jinan and 9.43 s in Yantai), but Italian honey bees were most affected by the climate. The 3 bees differed significantly in terms of the amount of apple pollen collected and their effects on the fruit setting rate in apples (mason bees > bumble bees > Italian honey bees). The results showed that the mason bee was the most suitable pollinating species for spring apple orchards; Bumble bees were more suitable as alternative pollinators during cloudy and low temperatures; Italian honey bees were able to take advantage of their large number of worker bees in sunny and warm weather. Abstract Background The standard evolutionary theory of ageing proposes that ageing occurs because of a trade-off between reproduction and longevity. Eusocial insect queens exhibit positive fecundity-longevity associations and so have been suggested to be counter-examples through not expressing costs of reproduction and through remodelling conserved genetic and endocrine networks regulating ageing and reproduction. If so, eusocial evolution from solitary ancestors with negative fecundity-longevity associations must have involved a stage at which costs of reproduction were suppressed and fecundity and longevity became positively associated. Using the bumblebee (Bombus terrestris), we experimentally tested whether queens in annual eusocial insects at an intermediate level of eusocial complexity experience costs of reproduction, and, using mRNA-seq, the extent to which they exhibit a remodelling of relevant genetic and endocrine networks. Specifically, we tested whether costs of reproduction are present but latent, or whether a remodelling of relevant genetic and endocrine networks has already occurred allowing queens to reproduce without costs. Results We experimentally increased queens’ costs of reproduction by removing their eggs, which caused queens to increase their egg-laying rate. Treatment queens had significantly reduced longevity relative to control queens whose egg-laying rate was not increased. Reduced longevity in treatment queens was not caused by increased worker-to-queen aggression or by increased overall activity in queens. In addition, treatment and control queens differed in age-related gene expression based on mRNA-seq in both their overall expression profiles and the expression of ageing-related genes. Remarkably, these differences appeared to occur principally with respect to relative age, not chronological age. Conclusions This study represents the first simultaneously phenotypic and transcriptomic experimental test for a longevity cost of reproduction in eusocial insect queens. The results support the occurrence of costs of reproduction in annual eusocial insects of intermediate social complexity and suggest that reproductive costs are present but latent in queens of such species, i.e. that these queens exhibit condition-dependent positive fecundity-longevity associations. They also raise the possibility that a partial remodelling of genetic and endocrine networks underpinning ageing may have occurred in intermediately eusocial species such that, in unmanipulated conditions, age-related gene expression depends more on chronological than relative age.

Bumblebees are crucial for both natural ecosystems and agriculture, but their decline in distribution and abundance over the past decade is alarming. The global importance of bumblebees in natural ecosystems and agricultural food production cannot be overstated. However, the reported decline over the past decade has led to a surge of interest in understanding and addressing bumblebee population decline. Hence, we aimed to detect disruptions in the gut microbiome of male and worker bumblebees reared indoor and outdoor to assess potential resilience to environmental stress. Using the Illumina MiSeq platform for 16s rRNA amplicon sequencing, we analyzed the gut microbiome of male and worker bees that were raised indoors (designated as the IM and IW group) and those that were raised outdoors (also designated as the OM and OW group). Our results show presence of core bacteria Neisseriaceae, Orbaceae, Lactobacillaceae and Bifidobacteriaceae from indoor reared worker bees. However, a higher abundance of Bifidobacterium and absence of Fructobacillus from indoor reared worker bees was also observed. Indoor-reared male bees had lower diversity and fewer observed OTUs compared to outdoor-reared male bees. Additionally, the relative abundance of Actinobacteriota, Bacteroidota, and Firmicutes was significantly lower in indoor-reared males, while Proteobacteria was significantly increased. Despite this, we did not observe any dysbiosis in the gut microbiota of indoor-reared bumblebees when comparing the role of the gut symbionts among the groups. Background: Global warming is expected to impact the chemical communication between flowering plants and their pollinators. Surprisingly, it is unknown whether and how temperature-induced changes in scent emission affect pollinator behavior. Strawberry (Fragaria x ananassa) is a plant primarily pollinated by bees and hoverflies, with the former group being particularly attracted to the floral scent they emit. Results: Using chemical analytical, electrophysiological, and behavioral approaches we tested whether temperature-induced shifts in floral scent of strawberry affect chemical communication with its main bee pollinators (Apis mellifera, Bombus terrestris, Osmia bicornis). While strawberry flowers in the optimum scenario released 10.4 ng/flower/hour, mainly p-anisaldehyde (81%) and seven other scent compounds, in the warmer scenario, the flowers did not emit any detectable scent. In the behavioral experiments, the pollinators were attracted by the scents of the optimum scenario. Conclusions: We predict that the absence of detectable scent emissions from strawberry plants grown under heat stress will reduce the attractiveness of the flowers to the bee pollinators. Our study raises important ecological and agricultural questions, as decreased attractiveness of flowers to pollinators might potentially lead to insufficient bee pollination, with potential negative consequences for ecosystem functioning and crop yields, particularly in regions reliant on bees as primary pollinators. Social organization is commonly dynamic, with extreme examples in annual social insects, but little is known about the underlying signals and mechanisms. Bumble bee larvae with close contact to a queen do not differentiate into gynes, pupate at an earlier age, and are commonly smaller than siblings that do not contact a queen. We combined detailed observations, proteomics, microRNA transcriptomics, and gland removal surgery to study the regulation of brood development and division of labor in the annual social bumble bee Bombus terrestris. We found that regurgitates fed to larvae by queens and workers differ in their protein and microRNA composition. The proteome of the regurgitate overlaps significantly with that of the mandibular (MG) and hypopharyngeal glands (HPG), suggesting that these exocrine glands are sources of regurgitate proteins. The proteome of the MG and HPG, but not the salivary glands, differs between queens and workers, with caste-specificity preserved for the MG and regurgitate proteomes. Queens subjected to surgical removal of the MG showed normal behavior, brood care, and weight gain, but failed to shorten larval development. These findings suggest that substances in the queen MG are fed to larvae and influence their developmental program. We suggest that when workers emerge and contribute to larval feeding, they dilute the effects of the queen substances, until she can no longer manipulate the development of all larvae. Longer developmental duration may allow female larvae to differentiate into gynes rather than to workers, mediating the colony transition from the ergonomic to the reproductive phase. Entomopathogenic fungi (EPF) are insecticide alternatives for pest control. Their ability to easily adhere and quickly penetrate the insect cuticle is a key factor for their selection, which has received too little consideration so far. Here, we evaluated the impact of five EPF on the survival of Drosophila suzukii, a worldwide invasive pest of soft-skinned fruits. The most efficient EPF was then selected, and a second efficacy assay was performed by exposing D. suzukii adults to the EPF for different durations: 10 seconds, 1 minute, 10 minutes, 1 hour and 3 hours. Finally, EPF safety was assessed on two non-target beneficial insects frequently encountered in the same crops affected by D. suzukii, namely Orius laevigatus and Bombus terrestris. We found Beauvaria bassiana to be the most efficient EPF, killing over 95% of the flies within 10 days. The exposure time impacted the mortality rates: 50% of the flies died within 4 days after a 3-hours exposure to B. bassiana, whereas 6 days were needed to reach the same result with 10 seconds of exposure. Whatever the exposure time, this EPF always needed ten days to be lethal for more than 95% of individuals. Beauvaria bassiana was not lethal for the non-target species. Thus, B. bassiana is an option to control D. suzukii without harming beneficial insects. Further studies are now needed under real cultivation conditions to assess whether B. bassiana can be included in biocontrol strategies against D. suzukii. [ABSTRACT FROM AUTHOR]

Adaptations that attract pollinators to flowers are central to the reproductive success of insect‐pollinated plants, including crops. Understanding the influence of these non‐rewarding traits on pollinator preference is important for our future food security by maintaining sufficient crop pollination. We have identified substantial variation in flower shape, petal size, corolla‐tube length, petal spot size and floral volatile compounds among a panel of 30 genetically distinct lines of Vicia faba. Using this variation, we found that Bombus terrestris was able to distinguish between natural variation in petal spot size, floral volatile emissions and corolla‐tube length. Foragers showed some innate preference for spotted flowers over non‐spotted flowers and preferred shorter corolla‐tube lengths over longer tubes. Our results suggest that some floral traits may have significant potential to enhance pollinator attraction to V. faba crops, particularly if paired with optimised rewards. [ABSTRACT FROM AUTHOR]

Insect pollination is fundamental for natural ecosystems and agricultural crops. The bumblebee species Bombus terrestris has become a popular choice for commercial crop pollination worldwide due to its effectiveness and ease of mass rearing. Bumblebee colonies are mass produced for the pollination of more than 20 crops and imported into over 50 countries including countries outside their native ranges, and the risk of invasion by commercial non‐native bumblebees is considered an emerging issue for global conservation and biological diversity. Here, we use genome‐wide data from seven wild populations close to and far from farms using commercial colonies, as well as commercial populations, to investigate the implications of utilizing commercial bumblebee subspecies in the UK. We find evidence for generally low levels of introgression between commercial and wild bees, with higher admixture proportions in the bees occurring close to farms. We identify genomic regions putatively involved in local and global adaptation, and genes in locally adaptive regions were found to be enriched for functions related to taste receptor activity, oxidoreductase activity, fatty acid and lipid biosynthetic processes. Despite more than 30 years of bumblebee colony importation into the UK, we observe low impact on the genetic integrity of local B. terrestris populations, but we highlight that even limited introgression might negatively affect locally adapted populations. The fat body has important functions in energy, fertility, and immunity. In female insects, mating stimulates physiological, behavioral, and gene expression changes. However, it remains unclear whether the metabolites in the fat body are affected after the bumblebee (Bombus terrestris) queen mates. Here, the ultrastructure and lipid metabolites in fat body of mated queens were compared with those of virgins. The fat body weight of mated bumblebee queens was significantly increased, and the adipocytes were filled with lipid droplets. Using LC-MS/MS-based untargeted lipidomics, 949 and 748 differential metabolites were identified in the fat body of virgin and mated bumblebee queens, respectively, in positive and negative ion modes. Most lipid metabolites were decreased, especially some biomembrane components. In order to explore the relationship between the structures of lipid droplets and metabolite accumulation, transmission electron microscopy and fluorescence microscopy were used to observe the fat body ultrastructure. The size/area of lipid droplets was larger, and the fusion of lipid droplets was increased in the mated queen's fat body. These enlarged lipid droplets may store more energy and nutrients. In agricultural landscapes, bees face a variety of stressors, including insecticides and poor-quality food. Although both stressors individually have been shown to affect bumblebee health negatively, few studies have focused on stressor interactions, a scenario expected in intensively used agricultural landscapes. Using the bumblebee Bombus terrestris, a key pollinator in agricultural landscapes, we conducted a fully factorial laboratory experiment starting at nest initiation. We assessed the effects of food quality and insecticides, alone and in interaction, on health traits at various levels, some of which have been rarely studied. Pollen with a diluted nutrient content (low quality) reduced ovary size and delayed colony development. Wing asymmetry, indicating developmental stress, was increased during insecticide exposure and interactions with poor food, whereas both stressors reduced body size. Both stressors and their interaction changed the workers' chemical profile and reduced worker interactions and the immune response. Our findings suggest that insecticides combined with nutritional stress reduce bumblebee health at the individual and colony levels, thus possibly affecting colony performance, such as development and reproduction, and the stability of plant–pollinator networks. The invasive hornet Vespa velutina nigrithorax is considered a proliferating threat to pollinators in Europe and Asia. While the impact of this species on managed honey bees is well-documented, effects upon other pollinator populations remain poorly understood. Nonetheless, dietary analyses indicate that the hornets consume a diversity of prey, fuelling concerns for at-risk taxa. Here, we quantify the impact of V. velutina upon standardised commercially-reared colonies of the European bumblebee, Bombus terrestris terrestris. Using a landscape-scale experimental design, we deploy colonies across a gradient of local V. velutina densities, utilising automated tracking to non-invasively observe bee and hornet behaviour, and quantify subsequent effects upon colony outcomes. Our results demonstrate that hornets frequently hunt at B. terrestris colonies, being preferentially attracted to those with high foraging traffic, and engaging in repeated—yet entirely unsuccessful—predation attempts at nest entrances. Notably however, we show that B. terrestris colony weights are negatively associated with local V. velutina densities, indicating potential indirect effects upon colony growth. Taken together, these findings provide the first empirical insight into impacts on bumblebees at the colony level, and inform future mitigation efforts for wild and managed pollinators. The study of animal behavior, particularly in insects, is crucial for understanding their biological and evolutionary aspects, with wide-ranging applications in agricultural science, pest management, conservation biology, and neuroscience. Investigating cognitive characteristics, specifically aversive learning, plays a pivotal role in comprehending the success of insects. This adaptive ability enables animals to efficiently cope with the stressful factors in their environment. In this study, we investigated the aversive learning capabilities of Bombus terrestris Audax workers, crucial pollinators across diverse ecosystems. Bees were trained and tested in a flight arena using artificial flowers equipped with electric shocks to simulate conditions wherebees could associate punishment alongside food resources with available cues. The result suggested that bees possess the ability to simultaneously detect potential threats and food resources, indicating a dual aversive-appetitive memory. Furthermore, comparing groups trained with aversive (electric shock) and neutral (distilled water) stimuli showed that danger cues led to faster learning and stronger memory formation. This conditioning setup aimed to simulate real-life foraging situations, exploring bee responses when confronted with potential dangers. Several studies on floral nectar demonstrated that the behaviour of visit performed by pollinators is influenced by nectar chemistry. Biogenic amines act as neurotransmitters in invertebrates and recently have been reported in the floral nectar of 15 plant species for the first time. However, both their occurrence in floral nectar and the effects of their nectar-like concentrations on bee behaviour remains largely unsubstantiated. To increase knowledge on the role of biogenic amines on plant–pollinator interactions, here we (i) investigated the biogenic amine composition of Echium vulgare nectar in relation to its floral sexual phases, and (ii) studied how an artificial solution enriched with nectar-like concentrations of tyramine affects the visit on flowers of bumblebees under semi-controlled conditions. The chemical analysis reported the presence of tyramine in E. vulgare nectar and no difference in concentration between the two sexual phases. To explore potential effects of tyramine on bee behaviour, we designed a new method consisting in zucchini flowers emptied of their natural nectar and refilled with artificial tyramine-enriched nectar, and we used bumblebee workers as pollinator model. We found that bees fed tyramine-enriched solution spent less time foraging on a single flower than those fed control solution, suggesting that their behaviour of visit was overall more dynamic. Our results highlight the importance of addressing further investigations on this emerging class of nectar compounds on insect cognition and behaviour, other than on its occurrence and distribution in nectar of other species. Many pollinators, including bumble bees, are in decline. Such declines are known to be driven by a number of interacting factors. Decreases in bee populations may also negatively impact the key ecosystem service, pollination, that they provide. Pesticides and parasites are often cited as two of the drivers of bee declines, particularly as they have previously been found to interact with one another to the detriment of bee health. Here we test the effects of an insecticide, sulfoxaflor, and a highly prevalent bumble bee parasite, Crithidia bombi, on the bumble bee Bombus terrestris. After exposing colonies to realistic doses of either sulfoxaflor and/or Crithidia bombi in a fully crossed experiment, colonies were allowed to forage on field beans in outdoor exclusion cages. Foraging performance was monitored, and the impacts on fruit set were recorded. We found no effect of either stressor, or their interaction, on the pollination services they provide to field beans, either at an individual level or a whole colony level. Further, there was no impact of any treatment, in any metric, on colony development. Our results contrast with prior findings that similar insecticides (neonicotinoids) impact pollination services, and that sulfoxaflor impacts colony development, potentially suggesting that sulfoxaflor is a less harmful compound to bee health than neonicotinoids insecticides. Background: Bees provide essential pollination services for many food crops and are critical in supporting wild plant diversity. However, the dietary landscape of pollen food sources for social and solitary bees has changed because of agricultural intensification and habitat loss. For this reason, understanding the basic nutrient metabolism and meeting the nutritional needs of bees is becoming an urgent requirement for agriculture and conservation. We know that pollen is the principal source of dietary fat and sterols for pollinators, but a precise understanding of what the essential nutrients are and how much is needed is not yet clear. Sterols are key for producing the hormones that control development and may be present in cell membranes, where fatty-acid-containing species are important structural and signalling molecules (phospholipids) or to supply, store and distribute energy (glycerides). Aim of the review: In this critical review, we examine the current general understanding of sterol and lipid metabolism of social and solitary bees from a variety of literature sources and discuss implications for bee health. Key scientific concepts of review: We found that while eusocial bees are resilient to some dietary variation in sterol supply the scope for this is limited. The evidence of both de novo lipogenesis and a dietary need for particular fatty acids (FAs) shows that FA metabolism in insects is analogous to mammals but with distinct features. Hemoglobins are mainly functional in the transport and storage of O2, and are present in aerobic, lung respiratory metazoans. Based on recent studies, it has been determined that globin-like proteins are also found in invertebrate groups which have tracheal respiratory systems. Thus, it has been claimed that the globin molecules may have housekeeping functions beyond their known functions. In this study, the presence of a hemoglobin gene was investigated in a natural pollinator, Bombus terrestris. Bombus terrestris (Hymenoptera: Apidae: Bombinae) is an ecologically and economically important species, it has a wide distribution in high and low altitude habitats, can live in different localities in terms of altitude and temperature. The expression of globin gene was determined using real time qPCR and RACE system, and confirmed by SRA data. The potential globin ORF sequence was 516 bp nucleotide and 171 bp amino acid long. This data was deposited in the GenBank database under accession number OP312897. On the other hand, expression profiles were investigated in different hypoxic conditions at different developmental stages (larva, pupae and adult) and in different tissue types (abdomen, thorax, head and legs). It was determined that the globin gene is transcribed in all developmental stages, but only in abdomen; and that globin expression decreases with decreasing amount of oxygen. [ABSTRACT FROM AUTHOR]

Entomopathogenic fungi have a high potential as microbial control agents of insect pests, both through direct application and through endophytic colonisation and the subsequent effects on the phytophagous insects. Recently, such symbiotic relationships with their host plants and compatibility with other natural enemies of insect pests have been characterized, including the modification of plant volatile emissions and the consequent effect on the insects. The present research aimed at investigating whether systemic Beauveria bassiana (Balsamo) Vuillemin colonisation of melon plants reached the floral tissues and affected flowering, pollen production and pollinator behaviour. Melon plants sprayed with B. bassiana conidial suspensions were endophytically colonised throughout the entire plant growth cycle and reached the floral tissues. In addition, the endophytically-colonized plants produced a significantly higher number of pollen grains, and flowered earlier than control plants. A foraging preference of honeybees and bumblebees for the control plants in choice assays was detected, likely due to the advanced maturation stage of colonised plants flowers. Noteworthy, changes in the emitted blend of volatiles were observed in the endophytically-colonised plants, which emitted more of the pollinator attractants D-limonene and benzeneacetaldehyde than control plants. Future research should consider the mechanisms for endophytemediated early flowering and variations in volatile profiles throughout the blooming process, together with their impact on pollination under greenhouse conditions. Body size is a trait that can affect plant–pollinator interaction efficiency and plant reproductive success. We explored the impact of intraspecific size shifts on the interactions between pollinators and flowering plants under controlled conditions. We considered two development conditions leading to the production of large and small individual flowers of Borago officinalis and Echium plantagineum. We also used the natural variability of worker size within bumblebee colonies to isolate small and large workers. We performed a fully crossed experiment with the two flower sizes of each plant species and the two sizes of bumblebee workers. Our results show that the size of both partners did not affect bee foraging behavior in most of the evaluated parameters and both bee sizes were equally efficient in depositing pollen. Significant differences were found only in pollen deposition across the life of a flower in small flowers of B. officinalis, with the greatest quantity of pollen deposited by small bees. We did not find a relationship between pollinator size and plant fitness. Our results suggest that generalist plant–pollinator interactions may be resilient to future potential mismatches in the size of the partners but remain to be tested if they are still resilient under the new environmental conditions resulting from global changes. Abstract Background Pesticides are identified as one of the major reasons for the global pollinator decline. However, the sublethal effects of pesticide residue levels found in pollen and nectar on pollinators have been studied little. The aim of our research was to study whether oral exposure to the thiacloprid levels found in pollen and nectar affect the learning and long-term memory of bumble bees. We tested the effects of two exposure levels of thiacloprid-based pesticide (Calypso SC480) on buff-tailed bumble bee (Bombus terrestris) in laboratory utilizing a learning performance and memory tasks designed to be difficult enough to reveal large variations across the individuals. Results The lower exposure level of the thiacloprid-based pesticide impaired the bees’ learning performance but not long-term memory compared to the untreated controls. The higher exposure level caused severe acute symptoms, due to which we were not able to test the learning and memory. Conclusions Our results show that oral exposure to a thiacloprid-based pesticide, calculated based on residue levels found in pollen and nectar, not only causes sublethal effects but also acute lethal effects on bumble bees. Our study underlines an urgent demand for better understanding of pesticide residues in the environment, and of the effects of those residue levels on pollinators. These findings fill the gap in the existing knowledge and help the scientific community and policymakers to enhance the sustainable use of pesticides.

The conception of “floral strips” is a strategy to provide more and different food sources for pollinators. The impact of “homogeneous” Phacelia tanacetifolia (“Phacelia”) and “heterogeneous” (flower mix) food sources on the enzyme activity of bumblebees (Bombus terrestris) and honeybees (Apis mellifera) under urban conditions has not been reported. Organisms responding to challenging environmental conditions are known to exhibit increases in oxidative stress parameters which in turn affect both physiological and metabolic parameters. A field study was conducted in Berlin-Dahlem, Germany, using the response of the “marker” enzyme superoxide dismutase (SOD) on food sources for assessment. SOD data is also shown from the wild bee Megachile rotundata (Fabricius 1787), obtained from three different locations in the federal state Brandenburg, Germany. The results demonstrate that the enzyme activity of SOD significantly increased in bumblebees visiting the flower mix compared to the Phacelia. The experimental approach had individual effects at the level of the species, bumblebees and honeybees, respectively. The activity of the biomarker SOD could be successfully used to assess the effects of the compositions of homogeneous and heterogeneous flower fields.

Neonicotinoids harm especially bumblebees and other species in agricultural areas all over the world. The toxic effect of thiamethoxam from the neonicotinoid group has been little studied especially on bees. This research aimed to evaluate the effects of thiamethoxam on the immune system cells of Bombus terrestris workers. Different ratios of 1/1000, 1/100 and 1/10 of the recommended maximum application dose of thiamethoxam formed the experimental groups. Ten foraging workers were used for each dose and control groups. Contamination was ensured by spraying the prepared suspensions at different ratios to the bees for 20 s at a pressure of 1 atm. The effects of thiamethoxam on the structures of immune system cells of bumblebees and the amount of these cells were investigated after 48 h of exposure. In general, anomalies such as vacuolization, cell membrane irregularities and cell shape changes were detected in prohemocyte, plasmatocyte, granulocyte, spherulocyte and oenocytoid in each dose group. Hemocyte area measurements in all groups were examined comparatively between groups. In general, granulocyte and plasmatocyte sizes were decreased, while spherulocyte and oenocytoid were increased. It was also determined that there was a significant decrease in the amount of hemocytes in the 1 mm3 hemolymph as dose increased. The results of the study revealed that sublethal doses of thiamethoxam negatively affected hemocytes and their amounts of B. terrestris workers. [ABSTRACT FROM AUTHOR]

Simple Summary: Several lab studies have shown that pesticides are causing sublethal effects on bees; however, little is known about their impact in actual field conditions. Continuous surveillance of the bees in the field will help monitor their pesticide exposure and better understand their impact on bee health. This study investigated the potential use of a new optical sensor, coupled with machine learning, to automatically identify flying insects, as an approach to monitoring the sublethal effects of pesticides on insects. To do so, bumblebees, Bombus terrestris, were exposed to field-realistic doses of a neonicotinoid pesticide, and their flight activity was recorded by the sensor. Through the data acquired, the investigation focused on whether the machine learning algorithm could distinguish the flight events that were created by the healthy bumblebees and the bumblebees that had been exposed to sublethal doses of the pesticide. The results showed that the algorithm could differentiate flight events, indicating the promise of the sensor as a continuous monitoring tool for bee health in the field. Among insects, bees are important pollinators, providing many vital ecosystem services. The recent pollinator decline is threatening both their diversity and abundance. One of the main drivers of this decline is the extensive use of pesticides. Neonicotinoids, one of the most popular groups of pesticides, can be toxic to bees. In fact, numerous studies have found that neonicotinoids can cause sublethal effects, which can impair the biology, physiology, and colony survival of the bees. Yet, there are still knowledge gaps, and more research is needed to better understand the interaction between neonicotinoids and bees, especially in the field. A new optical sensor, which can automatically identify flying insects using machine learning, has been created to continuously monitor insect activity in the field. This study investigated the potential use of this sensor as a tool for monitoring the sublethal effects of pesticides on bumblebees. Bombus terrestris workers were orally exposed to field-realistic doses of imidacloprid. Two types of exposures were tested: acute and chronic. The flight activity of pesticide-exposed and non-exposed bumblebees was recorded, and the events of the insect flights recorded by the sensor were used in two ways: to extract the values of the wingbeat frequency and to train machine learning models. The results showed that the trained model was able to recognize differences between the events created by pesticide-exposed bumblebees and the control bumblebees. This study demonstrates the possibility of the optical sensor for use as a tool to monitor bees that have been exposed to sublethal doses of pesticides.
