This study investigated how proteins of the insulin signaling cascade could modulate insulin resistance after dexamethasone (Dexa) treatment and aerobic training. Rats were distributed into 4 groups: sedentary control (SC), sedentary+Dexa (SD), trained control (TC), and trained+Dexa (TD), and underwent aerobic training for 70 days or remained sedentary. Dexa was administered during the last 10 days (1 mg · kg −1 per day i. p.). After 70 days, an intraperitoneal glucose tolerance test (ipGTT) was performed. Protein levels of IRS-1, AKT, and PKC-α in the tibialis anterior (TA) muscle were identified using Western blots. Dexa treatment increased blood glucose and the area under the curve (AUC) of ipGTT. Training attenuated the hyperglycemia and the AUC induced by Dexa. Dexa reduced IRS-1 (− 16%) and AKT (− 43%) protein level with no changes in PKC-α levels. Moreover, these effects on IRS-1 and AKT protein level were prevented in trained animals. These results show for the first time that aerobic exercise prevented reductions of IRS-1 and AKT level induced by Dexa in the TA muscle, suggesting that aerobic exercise is a good strategy to prevent Dexa-induced peripheral insulin resistance.

Results from our laboratory have suggested a pathway involving angiotensin II type 1 (AT1) receptors and vascular endothelial growth factor (VEGF) in angiogenesis induced by electrical stimulation. The present study investigated if similar mechanisms underlie the angiogenesis induced by short-term exercise training. Seven days before training and throughout the training period, male Sprague-Dawley rats received either captopril or losartan in their drinking water. Rats underwent a 3-day treadmill training protocol. The tibialis anterior and gastrocnemius muscles were harvested under anesthesia and lightly fixed in formalin (vessel density) or frozen in liquid nitrogen (VEGF expression). In controls, treadmill training resulted in a significant increase in vessel density in all muscles studied. However, the angiogenesis induced by exercise was completely blocked by either losartan or captopril. Western blot analysis showed that VEGF expression was increased in the exercised control group, and both losartan and captopril blocked this increase. The role of VEGF was directly confirmed using a VEGF-neutralizing antibody. These results confirm the role of angiotensin II and VEGF in angiogenesis induced by exercise.

Objective To investigate whether training changes skeletal muscle venular profile and hemodynamic responses to exercise we studied spontanesouly hypertensive rats (SHR) and Wistar-Kyoto (WKY) rats submitted to training programme (T = 50-60% of VO 2 max). Design Training (T) was performed on a treadmill over a period of 13 weeks. Age-matched control groups were kept sedentary (S). T and S rats were chronically instrumented for hindlimb flow (HLF) and arterial pressure (AP) measurements at rest, during dynamic exercise and recovery in two different situations: control and after extensive intravenous blockade (hexamethonium + losartan + N ω -nitro-L-arginine methyl ester + hydralazine). For morphometric analysis, skeletal muscle samples (gracilis) were obtained after transcardiac perfusion with fixative. Results T caused a significant reduction of resting mean arterial pressure (MAP) (-11%) only in the SHR group without changing basal HLF. In the sedentary SHR (SHR S ), basal relative hindlimb resistance was increased by 45%, but was significantly reduced after T (P < 0.05). During dynamic exercise, MAP increased similarly (10-20 mmHg) in all groups. HLF increases were similar for the four groups up to 0.8 km/h; at higher workloads, HLF was higher in trained SHR (SHR T ) versus trained WKY (WKY T ) (3.9- versus 2.9-fold increase over basal HLF, respectively). After blockade (and pressure correction with IV phenylephrine infusion), steady-state exercise was performed with similar hindlimb vasodilation in all groups and was accompanied by MAP reduction (-17 ± 8 mmH only in SHR T group. Skeletal muscle venular profile (density, diameter and lumen cross-sectional area) was similar in WKY T . WKY S and SHRs, but significantly increased in SHR T . In this group the two-fold increase in venule density was correlated with both the reduction in baseline MAP and the increase in HLF during dynamic exercise. Conclusions The results suggest that increased venule density is a specific adaptation of SHR skeletal muscle t training. Venular growth may contribute to both the pressure-lowering effect and the large HLF at high exercise intensities observed in the trained SHR.

Objective:Physiological angiogenesis in skeletal muscle is an adaptive response to physical training and electrical stimulation. This study investigated the role of angiotensin II (Ang II) in regulating both angiogenesis and vascular endothelial growth factor (VEGF) protein expression induced by electrical stimulation. Methods:The right tibialis anterior (TA) and extensor digitorum longus (EDL) muscles of Sprague-Dawley rats were stimulated for 8 hours per day for 7 days. This study investigated the potential protective effect of low-intensity resistance training (RT) against dexamethasone (DEX) treatment induced muscle atrophy. Rats underwent either an 8 week period of ladder climbing RT or remained sedentary. During the last 10 days of the exercise protocol, animals were submitted to a DEX treatment or a control saline injection. Muscle weights were assessed and levels of AKT, mTOR, FOXO3a, Atrogin-1 and MuRF-1 proteins were analyzed in flexor hallucis longus (FHL), tibialis anterior (TA), and soleus muscles. DEX induced blood glucose increase (+46%), body weight reduction (-19%) and atrophy in FHL (-28%) and TA (-21%) muscles, which was associated with a decrease in AKT and an increase in MuRF-1 proteins levels. Low-intensity RT prevented the blood glucose increase, attenuated the FHL atrophy effects of DEX, and was associated with increased mTOR and reductions in Atrogin-1 and MuRF-1 in FHL. In contrast, TA muscle atrophy and signaling proteins were not affected by RT. These are the first data to demonstrate that low-intensity ladder-climbing RT specifically mitigates the FHL atrophy, which is the main muscle recruited during the training activity, while not preventing atrophy in other limb muscle not as heavily recruited. The recruitment-dependent prevention of atrophy by low intensity RT likely occurs by a combination of attenuated muscle protein degradation signals and enhanced muscle protein synthesis signals including mTOR, Atrogin-1 and MuRF-1.

OBJETIVO: Comparar os limites inferiores (L.inf.) e superiores (L.sup.) da prescrição de treinamento físico aeróbio determinada pelo teste ergométrico convencional (60-70% do VO2máx estimado ou 70-85% da FCmáx atingida), com a prescrição obtida pelo teste ergoespirométrico [limiar anaeróbio (LA) e ponto de compensação respiratória (PCR)]. MÉTODOS: Realizaram teste ergoespirométrico progressivo até a exaustão 47 homens (30±5 anos), divididos em subgrupos, de acordo com a velocidade da esteira durante o teste (4 ou 5mph) e a capacidade física medida [baixa (BCF) e moderada (MCF)]. RESULTADOS: Os L.inf. de prescrição indireta apresentaram valores de VO2 e FC significantemente maiores que os valores de VO2 e FC no LA (4mph= 34,4±4,5 vs 19,6±4,6 e 5mph= 28,9±2 vs 18,9±5,4, e BCF= 32,0±4,1 vs 17,2±2,8 e MCF= 31,6±4,9 vs 21,1±5,7(mlO2.kg-1.min-1) e (4mph = 128,9±7,8 vs 113,1± 15,6 e 5mph= 130,3±5,2 vs 114,1± 18,9, e BCF= 127,6±7,2 vs 109,3±13,2 e MCF= 131,2± 5,7 vs 117,4± 19,2bpm). Os L.sup. de prescrição indireta no grupo de 4mph e BCF apresentaram valores de VO2 significantemente maiores que os valores medidos no PCR (40,1±5,3 vs 32,2±4,3 e 37,4±4,8 vs 30,6±2,5 mlO2.kg-1.min-1, respectivamente), e valores de FC semelhantes aos medidos no PCR. CONCLUSÃO: Os L.inf. da prescrição indireta de treinamento físico superestimam o LA, enquanto os L.sup. parecem adequados somente para indivíduos ativos com MCF. PURPOSE: To compare the lower (LL) and upper limits (UL) of exercise intensity prescription based on standard exercise test (60-70% of estimated VO2max or 70-85% of HRmax measured) with exercise intensity prescription based on cardiopulmonary exercise test [anaerobic threshold (AT) and respiratory compensation point (RCP)]. METHODS: Fourty seven men (30±5 years) who were submitted to a progressive cardiopulmonary exercise test until exhaustion were divided in subgroups according to treadmill speed during exercise test (4 or 5mph) and the physical capacity [lower (LPC) and moderate physical capacity (MPC)]. RESULTS: The LL of the indirect exercise intensity prescription showed VO2 and HR values significantly higher than VO2 and HR values measured at AT (4mph= 34.4±4.5 vs 19.6±4.6 and 5mph= 28.9±2 vs 18.9±5.4, and LPC= 32.0±4.1 vs 17.2±2.8 and MPC= 31.6±4.9 vs 21.1± 5.7mlO2.kg-1.min-1 ) and (4mph= 128.9±7.8 vs 113.1±15.6 and 5mph= 130.3±5.2 vs 114.1±18.9, and LPC= 127.6±7.2 vs 109.3±13.2 and MPC= 131.2±5.7 vs 117.4±19.2bpm) The UL of the indirect exercise intensity prescription in 4mph and LPC group showed VO2 values significantly higher than those measured at RCP (40.1±5.3 vs 32.2±4.3 and 37.4±4.8 vs 30.6±2.5mlO2.kg-1.min-1, respectively), but similar HR values to those obtained at RCP. CONCLUSION: The LL of prescription based on standard exercise test overestimate the AT, whereas the UL seem adequate only for subjects with moderate physical capacity.

Índice de massa corporal, pressão arterial (PA), circunferência abdominal (CA) e presença de lesões foram avaliadas em 94 corredores. Destes, 38,5% eram hipertensos auto-referidos, mas 41,9% estavam com a PA alterada no dia. Dentre os auto-referidos sem problemas de saúde, foram encontradas alterações na PA (42%), na CA (9,8%) e 6% apresentaram risco cardiovascular moderado. Verificou-se que 34% já sofreram lesão, sendo que 67,7% foram derivadas de treinos ou competições. Idade, distância da prova e realização de outras atividades foram associados com a ocorrência de lesão. Os resultados do presente estudo nos permitem concluir que os praticantes de pedestrianismo apresentam uma condição física propícia ao comprometimento cardiovascular durante uma prova e ao surgimento de lesões, sugerindo campanhas de conscientização sobre a condição de saúde neste público. This study investigated the anthropometric characteristics and the presence of cardiovascular risk factors (Part 1) as well as occurrence of injuries (Part 2) in road runners. Body mass index, blood pressure (BP), abdominal circumference (AC) and incidence of injuries were evaluated in 94 road runners. Among the participants, 38,5% self-reported as hypertensive, but 41,7% from total had altered BP. Among healthy runners, it was found alterations on BP (42%), AC (9.8%) and 6% presented moderated cardiovascular risk. It was observed that 34% already suffered some injuries and 67.7% derived from training and competition. Age, distance of the running and practice of other activities were associated with presence of injury. The results of the present study allow us to conclude that road runners have a physical condition that facilitates the cardiovascular event during the competition and that contributes to the injuries. Thus it suggests that campaigns need to be done in order to make the runner aware about its health condition. It represents the most important modifiable risk factor for cardiovascular diseases, which are the leading cause of morbidity and mortality for both men and women. In addition to involving sympathetic overactivity that alters hemodynamic parameters, hypertension is accompanied by several abnormalities in the skeletal muscle circulation including vessel rarefaction and increased arteriole wall-to-lumen ratio, which contribute to increased total peripheral resistance. Low-intensity aerobic training is a promising tool for the prevention, treatment and control of high blood pressure, but its efficacy may differ between men and women and between male and female animals. This review focuses on peripheral training-induced adaptations that contribute to a blood pressure-lowering effect, with special attention to differential responses in male and female spontaneously hypertensive rats (SHR). Heart, diaphragm and skeletal muscle arterioles (but not kidney arterioles) undergo eutrophic outward remodeling in trained male SHR, which contributed to a reduction of peripheral resistance and to a pressure fall. In contrast, trained female SHR showed no change in arteriole wall-to-lumen ratio and no pressure fall. on the other hand, training-induced adaptive changes in capillaries and venules (increased density) were similar in male and female SHR, supporting a similar hyperemic response to exercise. Modelo : Estudo de prevalência. Objetivos: Avaliar a presença de hipertensão arterial auto-referida e comparar com os níveis de pressão arterial aferidos. Além sso, o trabalho se propôs a avaliar o grau de informação dos funcionários da saúde e determinar se o setor onde trabalha o funcionário influencia a Pressão Arterial (PA). Métodos: Foram avaliados 349 funcionários (46±10 anos) das cidades de Bauru e Jaú, que responderam uma anamnese (dados pessoais, condição socioeconômica, presença de doenças, prática de exercícios físicos e setor ocupacional) e foram submetidos a uma avaliação da PA. Os avaliados se auto-referiram normotensos ou hipertensos. Valores de PA Sistólica >140 mmHg e/ou PA diastólica >90 mmHg foram considerados elevados. Neste sentido, procuramos descrever o perfil, avaliar a condição física e o risco cardiovasculares dos funcionários da saúde. Foram avaliados 198 funcionários (46 ± 10 anos) que responderam uma anamnese e foram submetidos a avaliações bioquímicas, antropométricas e funcionais. Os valores foram apresentados como média ± desvio padrão da média. Foram utilizados testes de qui-quadrado (x²), taxa de odds e correlações de Pearson (p < 0,05). Dos avaliados, 54% apresentaram peso corporal elevado, 33% pressão arterial (PA) elevada e 56% eram sedentários. Colesterol total (CT), lipoproteína de baixa densidade e triglicerídeos (TG) acima da normalidade foram encontrados em 49%, 41% e 24% dos servidores, respectivamente e, 75% apresentaram taxas reduzidas de lipoproteína de alta densidade. As análises de x² e taxa de odds apontaram que os valores de TG e PA foram significativamente maiores para homens comparados às mulheres. Capacidade cardiorrespiratória ruim foi verificada em 56% dos homens e 23% das mulheres, além disso, a flexibilidade apresentou-se com níveis insatisfatórios para homens e mulheres. Observou-se que o índice de massa corporal se correlacionou positivamente com PA, CT e TG. Constatou-se, também que a idade foi fator agravante para PA, flexibilidade, CT e glicemia nas mulheres. Dessa forma, a prevalência de múltiplos fatores de risco nestes trabalhadores demonstra tratar-se de população não assistida pela saúde, motivo pelo qual sugere-se conscientização sobre sua própria saúde evitando a aposentadoria precoce Several public health problems are due to environmental work, which, associated with sedentarism, contribute to reduce the quality of life of the worker. This study investigated whether exercise training could prevent the negative side effects of dexamethasone. Rats underwent a training period and were either submitted to a running protocol (60% physical capacity, 5 days/week for 8 weeks) or kept sedentary. After this training period, the animals underwent dexamethasone treatment (1 mg/kg per day, i.p., 10 days). Glycemia, insulinemia, muscular weight and muscular glycogen were measured from blood and skeletal muscle. Vascular endothelial growth factor (VEGF) protein was analyzed in skeletal muscles. Dexamethasone treatment evoked body weight loss (−24%), followed by muscular atrophy in the tibialis anterior (−25%) and the extensor digitorum longus (EDL, −15%). Dexamethasone also increased serum insulin levels by 5.7-fold and glucose levels by 2.5-fold compared to control. The exercise protocol prevented atrophy of the EDL and insulin resistance. Also, dexamethasone-treated rats showed decreased muscular glycogen (−41%), which was further attenuated by the exercise protocol. The VEGF protein expression decreased in the skeletal muscles of dexamethasone-treated rats and was unaltered by the exercise protocol. These data suggest that exercise attenuates hyperglycemia and may also prevent insulin resistance, muscular glycogen loss and muscular atrophy, thus suggesting that exercise may have some benefits during glucocorticoid treatment.

Vascular endothelial growth factor (VEGF) is important for angiogenesis of skeletal muscle. The present study evaluated the time course of VEGF and angiogenesis after short- and long-term exercise training of female SHR and Wistar Kyoto (WKY) rats, 8-9 weeks (200-250 g). Rats were allocated to daily training or remained sedentary for 3 days (N = 23) or 13 weeks (N = 23). After training, the carotid artery was catheterized for AP measurements. Locomotor (tibialis anterior and gracilis) and non-locomotor skeletal muscles (temporalis) were harvested and prepared for histologic and protein expression analyses. Training increased treadmill performance by all groups (SHR = 28%, WKY = 64%, 3 days) and (SHR = 141%, WKY = 122%, 13 weeks). SHR had higher values of AP than WKY (174 ± 4 vs 111 ± 2 mmHg) that were not altered by training. Three days of running increased VEGF expression (SHR = 28%, WKY = 36%) simultaneously with an increase in capillary-to-fiber ratio in gracilis muscle (SHR = 19%, WKY = 15%). In contrast, 13 weeks of training increased gracilis capillary-to-fiber ratio (SHR = 18%, WKY = 19%), without simultaneous changes in VEGF expression. Training did not change VEGF expression and capillarity of temporalis muscle. We conclude that training stimulates time- and tissue-dependent VEGF protein expression, independent of pressure levels. VEGF triggers angiogenesis in locomotor skeletal muscle shortly after the exercise starts, but is not involved in the maintenance of capillarity after long-term exercise in female rats. No. of bitstreams: 0 Previous issue date: 2007-03-01 Background and objective: It has been shown that aerobic exercise is useful to reduce arterial pressure, however, the effectiveness of an exercise program is still controversial and not very well analyzed among populations with low-income. The objective of the present study was to set up an individualized physical fitness program - Projeto Hipertensão - focused on hypertensive people, patients from a Health Basic Unit (HBU) and, after that, to investigate the effects of this program on physical fitness, metabolic profile and pressure levels. Methods: Sixteen hypertensive women (56 ± 3yrs) under regular pharmacological treatment underwent 4 months of a supervised aerobic and stretching exercise program (3 sessions/wk, 90 min/session, 60% of V̇O 2 max). Several physical and metabolic variables were compared before and after 4 months of training. Results: Training significantly reduced systolic arterial pressure (SAP, -6%), improved cardio-respiratory fitness (+42% of V̇O2max), flexibility (+11%) and plasma glucose content (-4%). BMI and % fat did not change. Besides modifying metabolic profile, it was found that training presented significant correlations between individual initial values of cholesterol total level (CT), high density lipoprotein (HDL-C) and low density lipoprotein (LDL-C) and its responses after exercise. Conclusions: The study shows that exercise programs can be personalized for hypertensive patients from a HBU and confirms the effectiveness of exercise on AP, physical fitness, flexibility and lipid profile on hypertensive patients. The expressive reduction of AP in hypertensive subjects suggests that this exercise intervention should be emphasized on other health centers which assist low-income population. Angiogenesis, under normal conditions, is a tightly regulated balance between pro- and antiangiogenic factors. The goal of this study was to investigate the mechanisms involved in the control of the skeletal muscle angiogenic response induced by electrical stimulation during the suppression of plasma renin activity (PRA) with a high-salt diet. Rats fed 0.4% or 4% salt diets were exposed to electrical stimulation for 7 days. The tibialis anterior (TA) muscles from stimulated and unstimulated hindlimbs were removed and prepared for gene expression analysis, CD31-terminal deoxynucleotide transferase-mediated dUTP nick-end labeling (TUNEL) double-staining assay, and Bcl-2 and Bax protein expression by Western blot. Rats fed a low-salt diet showed a dramatic angiogenesis response in the stimulated limb compared with the unstimulated limb. This angiogenesis response was significantly attenuated when rats were placed on a high-salt diet. Microarray analysis showed that in the stimulated limb of rats fed a low-salt diet many genes related to angiogenesis were upregulated. In contrast, in rats fed a high-salt diet most of the genes upregulated in the stimulated limb function in apoptosis and cell cycle arrest. Endothelial cell apoptosis, as analyzed by CD31-TUNEL staining, increased by fourfold in the stimulated limb compared with the unstimulated limb. There was also a 48% decrease in the Bcl-2-to-Bax ratio in stimulated compared with unstimulated limbs of rats fed a high-salt diet, confirming severe apoptosis. This study suggests that the increase in endothelial cell apoptosis in TA muscle might contribute to the attenuation of angiogenesis response observed in rats fed a high-salt diet.

Objective To investigate mechanisms underlying the training-induced blood pressure-lowering effect we analyzed the hemodynamic responses and morphometric changes of the skeletal muscle microcirculation of spontaneously hypertensive (SHR) and normotensive Wistar–Kyoto (WKY) rats during an exercise training program. Design Training (50–60% VO2 max) was performed on a treadmill for 13 weeks and control groups were kept sedentary over the same period of time. Trained and sedentary rats were chronically instrumented for hindlimb flow and arterial pressure (AP) recordings under conscious unrestrained conditions. Gracilis and myocardial muscle samples were obtained for morphometric analysis after transcardiac perfusion of fixative. Results SHR, when compared to WKY presented an elevated blood pressure, an increased relative hindlimb vascular resistance, capillary rarefaction in both gracilis and myocardium and an increased wall-to-lumen ratio of gracilis arterioles. Training increased significantly both capillary density and capillary/fiber ratio in the gracilis and myocardium of WKY and SHR groups, causing a complete reversal of capillary rarefaction in trained SHR. In SHR, training also reduced resting blood pressure and caused normalization of both relative hindlimb vascular resistance and gracilis arterioles wall-to-lumen ratio. Regression analysis revealed strong positive correlation between hindlimb vascular resistance and mean AP (MAP) and between arterioles wall-to-lumen ratio and MAP. Conclusions The results suggest that low-intensity training can significantly reduce pressure in SHR while normalizing both the arteriole morphology and the resistance of the skeletal muscle microcirculation.

The objective of the present study was to validate the transit-time technique for long-term measurements of iliac and renal blood flow in rats. Flow measured with ultrasonic probes was confirmed ex vivo using excised arteries perfused at varying flow rates. An implanted 1-mm probe reproduced with accuracy different patterns of flow relative to pressure in freely moving rats and accurately quantitated the resting iliac flow value (on average 10.43 +/- 0.99 ml/min or 2.78 +/- 0.3 ml min-1 100 g body weight-1). The measurements were stable over an experimental period of one week but were affected by probe size (resting flows were underestimated by 57% with a 2-mm probe when compared with a 1-mm probe) and by anesthesia (in the same rats, iliac flow was reduced by 50-60% when compared to the conscious state). Instantaneous changes of iliac and renal flow during exercise and recovery were accurately measured by the transit-time technique. Iliac flow increased instantaneously at the beginning of mild exercise (from 12.03 +/- 1.06 to 25.55 +/- 3.89 ml/min at 15 s) and showed a smaller increase when exercise intensity increased further, reaching a plateau of 38.43 +/- 1.92 ml/min at the 4th min of moderate exercise intensity. In contrast, exercise-induced reduction of renal flow was smaller and slower, with 18% and 25% decreases at mild and moderate exercise intensities. Our data indicate that transit-time flowmetry is a reliable method for long-term and continuous measurements of regional blood flow at rest and can be used to quantitate the dynamic flow changes that characterize exercise and recovery.

In a previous study, we demonstrated that Dahl S rats (SS group) have low plasma renin activity, whereas transfer of a region of chromosome 13 containing the renin gene from Dahl R onto a congenic strain of Dahl SS/Jr/Hsd/MCW rats (S/ren RR group) restores renin secretory responses. In the present study, we compared the angiogenic responses to electrical stimulation in the SS and S/ren RR groups to explore the hypotheses that the renin-angiotensin system is involved in vascular endothelial growth factor (VEGF) expression and angiogenesis in skeletal muscle. Congenic SS and S/ren RR rats fed a 0.4% or 4% salt diet were surgically prepared by chronic implantation of an electrical stimulator. Another group of S/ren RR rats was treated with lisinopril 2 days before the surgery and throughout the stimulation protocol. The right tibialis anterior (TA) and extensor digitorum longus (EDL) were stimulated for 8 hours per day for 7 days. The contralateral muscles served as controls. Western blot analysis was performed to identify VEGF protein expression in these muscles. Electrical stimulation produced no change in vessel density of the SS group fed a 0.4% salt diet (change 5.50% and 8.14% for EDL and TA, respectively). Transfer of a region containing the renin gene restored the angiogenic response (change 16% and 30% for EDL and TA, respectively) despite a significantly higher blood pressure. Blockade of the renin-angiotensin system by lisinopril or high salt restored the responses observed in the SS group fed a low salt diet. In addition, increases in VEGF expression to electrical stimulation were observed only in the S/ren RR group fed a low salt diet. These results suggest that renin gene transfer restores angiogenesis and VEGF expression in the skeletal muscle of Dahl S rats.

Study Objective: To characterize statin treatment management due to lipid alterations and side effects throughout statin treatment in basic healthcare unit. Methods: Medical reports of women from a basic healthcare unit were analyzed, obtaining: disease presence, regular medication prescription, statin type and dosage, biochemical exams results, musculoskeletal complaints, and statin use cessation, going back the information until the medical consultation of first prescription. Results: Prescribed statins were Simvastatin and Atorvastatin at low doses (10-20 mg). Dose (48,4%) and/or type (25,4%) alterations occurred for lipid profile adequacy. Lipid levels were reduced without creatine kinase elevation. Treatment withdrawn (30,6%) was mainly due to their own decision (74%), which was strongly associated with records of musculoskeletal complaints (Odds Ratio: 6,40[1,53-26,78]). Conclusion: Statin treatment was effective in reducing serum lipid levels and self-reported pain was underestimated, characterizing the major limiting factor for treatment adherence. Fluoride (F) is a potent anti-cariogenic element, but only an appropriate dose is effective to have therapeutic action, else systemic toxicity may be observed. Additionally, two factors, amount of F and time of exposure, drive its action. Surprisingly, the susceptibility to toxic effects of F is genetically determined. The present study identified the effects of F on the liver proteome of mice susceptible (A/J) or resistant (129P3/J) to the effects of F. Weanling male A/J (n=6) and 129P3/J mice (n=6) were housed in pairs and assigned to three groups given low-F diet and drinking water containing 0, 15 or 50 ppm F for 7 weeks. Liver proteome profiles were examined using nano-LC-ESI-MS/MS. Protein function was classified by GO biological process (Cluego v2.0.7 + Clupedia v1.0.8). Difference in expression among the groups was determined using the PLGS software. In the control group (0 ppm F), most proteins with fold change were increased in A/J mice. Precisely the proteins related to energy flux and oxidative stress were quite significant in this context, suggesting the high susceptibility of these mice to the effects of F, since the exposure also induces oxidative stress. Treatment with the lower F concentration provoked more pronounced alterations in fold change in liver proteins in comparison to the treatment with the higher F concentration. Strikingly, most of the proteins with fold change upon following 15 ppm F treatment, were increased in the A/J mice compared with their 129P3/J counterparts, thus suggesting attempt of the former to fight against the toxic effects of F. With respect to 50 ppm F, most proteins with fold change were decreased in the A/J mice compared with their 129P3/J counterparts, especially proteins related to oxidative stress and protein folding, which might be related to the higher susceptibility of the A/J animals to the deleterious effects of F. Our findings can provide new insights into the molecular mechanisms underlying genetic susceptibility to fluorosis by indicating key protein players which need to be better addressed in future experimental studies. O Fluoreto (F) é um potente elemento anti-cariogênico, mas é somente efetivo terapeuticamente em uma dose apropriada. Por outro lado, doses acima das recomendadas levam a toxicidade sistêmica. Em adição, dois fatores decidem sua efetividade de ação: quantidade de F e tempo de exposição. A suscetibilidade aos efeitos tóxicos do F é determinada geneticamente. O presente estudo avaliou os efeitos do F no proteoma do fígado de camundongos suscetíveis (A/J) ou resistentes (129P3/J) aos efeitos do F. Camundongos machos desmamados A/J (n=6) e 129P3/J (n=6) foram alojados em pares e divididos em três grupos tratados com ração com baixo teor de F e água contendo 0, 15, ou 50 ppm de F por 7 semanas. Perfis proteômicos do fígado foram examinados usando nano-LC-ESI-MS/MS. A função de proteínas foi classificada pelo processamento biológico GO (Cluego v2.0.7 + Clupedia v1.0.8). A diferença de expressão entre os grupos foi determinada usando o software PLGS. No grupo controle (0 ppm F), a expressão da maioria das proteínas foi aumentada nos camundongos A/J e precisamente as proteínas relacionadas ao fluxo de energia e estresse oxidativo foram significativas neste contexto, sugerindo portanto, a alta sucetibilidade destes camundongos aos efeitos do F, já que a exposição também induz o estresse oxidativo. O tratamento com baixa concentração de F provocou alterações mais pronunciadas em proteínas do fígado comparado ao tratamento com alta concentração de F. Notadamente, a maioria das proteínas encontradas no fígado dos animais tratados com 15 ppm de F foi aumentada em camundongos A/J comparados aos camundongos 129P3/J, demonstrando portanto, uma tentativa dos A/J de neutralizar os efeitos tóxicos do F. Já nos animais tratados com 50 ppm de F, a maioria das proteínas foi diminuída nos camundongos comparados aos seus pares 129P3/J, especialmente proteínas relacionadas ao estresse oxidativo e enovelamento de proteínas, o que pode estar relacionado à alta suscetibilidade dos animais A/J aos efeitos deletérios do F. Nossos achados podem fornecer novos insights que podem contribuir para a interpretação os mecanismos moleculares relacionados à suscetibilidade genética à fluorose, indicando proteínas chaves que precisam ser melhor estudadas em estudos futuros.

Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Hypertension (HT) is a multifactorial disease characterized by elevated and sustained blood pressure (BP), which can be primary or secondary. Dexamethasone (DEX) is a synthetic glucocorticoid and its chronic use causes hypertension. Hypertension exacerbation induced by DEX on spontaneously hypertensive rats (SHR) could increase mortality risk. Microvascular rarefaction is present in both primary and DEX-induced hypertension and it is associated with angiogenic/apoptotic pathway unbalance. More recently, microRNA has been considered important in determining the genesis of spontaneous hypertension, however, their role in DEX-induced hypertension in SHR has not been investigated. Conversely, physical training (T) has been recommended as a co-adjuvant for the HT treatment. Thus, the aim of the present study was to investigate whether physical training is able to attenuate blood pressure increase in SHR treated with DEX and whether if this effect is associated with changes in microRNA related to microcirculation . Normotensive (Wistar) and hypertensive (SHR) rats underwent 8 weeks of T on a treadmill (60% of maximum capacity, 5 days a week, 1 hour a day) or kept sedentary. During 14 days, after 8 weeks of physical training, animals were treated with DEX (50µg / kg of body weight per day, s.c.) or saline. Blood pressure measurement and spectral analysis were performed after DEX treatment. Subsequently, tibialis anterior muscle and left ventricle were removed and processed for capillary density and capillary-to-fiber ratio evaluation, as well as VEGF, AKT, p-AKT, eNOS, Bcl-2, capase-3-cleaved proteins levels and microRNA-16, 21, 126, 155, 221 and 222 expression. In Wistar rats, DEX increased blood pressure (+27%), caused an autonomic imbalance to the heart, reduced capillary density and capillary-to-fiber ratio on tibialis anterior muscle associated with VEGF, p-AKT / AKT, Bcl-2 reduction and caspase-3-cleaved increase. Training attenuated blood pressure (-10%) increase and autonomic imbalance to the heart and prevented capillary density and capillary-to-fiber ratio reduction on tibialis anterior muscle associated with microRNA 126 increase and with a better balance between VEGF, p-AKT/AKT, Bcl-2 and caspase-3-cleaved levels. In SHRs, training reduced blood pressure and increased tibialis anterior muscle and left ventricle capillary density and capillary-to-fiber ratio. Thus, exercise training attenuates hypertension induced by DEX in Wistar rats due to autonomic imbalance attenuation and microcirculation rarefaction prevention associated with microRNA 126 increase. T, per se, induced myocardium angiogenesis associated with microRNA 126 increase, regardless DEX treatment. In addition, T reduced H in SHR, regardless DEX treatment, due to autonomic balance and microcirculation improvement. DEX did not exacerbate hemodynamic and microcirculation alteration already present in SHR. A hipertensão arterial (HA) é uma doença multifatorial caracterizada por níveis elevados e sustentados de pressão arterial (PA) que pode ser dividida em HA primária e secundária. A dexametasona (DEX) é um glicocorticoide sintético, cujo uso crônico pode desencadear HA. O uso de DEX em ratos espontaneamente hipertensos (SHR) pode aumentar o risco de mortalidade pela hipertensão exacerbação. A rarefação da microcirculação está presente tanto na HA primária como na induzida por DEX e está associada com alterações nas vias angiogênicas e apoptóticas. Mais recentemente, os microRNAs vêm ganhando visibilidade e sendo considerados importantes na determinação da densidade capilar, contudo, seu papel na rarefação microvascular induzida pela DEX em SHR ainda não foi investigado. Inversamente, o treinamento físico (TF) tem sido recomendado como co-adjuvante no tratamento da HA. Assim, o objetivo do presente estudo foi investigar se o treinamento físico é capaz de atenuar aumento da pressão arterial em SHR tratados com DEX e se esse efeito está associado às alterações na expressão dos microRNAs relacionadas a microcirculação. Ratos normotensos (Wistar) e hipertensos (SHR) foram submetidos a 8 semanas de TF em esteira (60% da capacidade máxima, 5 dias por semana, 1 hora por dia) ou mantidos sedentários. Durante 14 dias, após as 8 semanas de treinamento físico, os animais foram tratados com DEX (50µg/kg de peso corporal por dia, s.c.) ou salina. A pressão arterial e a análise espectral foram aferidas após o tratamento com DEX. Posteriormente, o músculo tibial anterior e o ventrículo esquerdo foram retirados e processados para avaliação da densidade capilar e razão capilar fibra nessas musculaturas, bem como, para a avaliação da produção proteica do VEGF, AKT, p-AKT, eNOS, Bcl-2 e capase-3-clivada e da expressão gênica dos microRNAs-16, 21, 126, 155, 221 e 222. Nos ratos Wistar a DEX causou aumento da pressão arterial (+27%), desbalanço autonômico para o coração, redução da densidade capilar e da razão capilar fibra no músculo tibial anterior, associado à redução do VEGF, da p-AKT/AKT, da Bcl-2 e ao aumento da caspase-3-clivada. Já o treinamento físico atenuou o aumento da pressão arterial (-10%) e o desbalanço autonômico para o coração. Ainda, preveniu a redução da densidade capilar no músculo tibial anterior associada ao aumento da expressão do microRNA 126, atenuação da redução do VEGF, da p-AKT/AKT, da Bcl-2 e a atenuação do aumento da caspase-3-clivada. Além disso, o treinamento físico aumentou a densidade capilar no ventrículo esquerdo dos animais Wistar por aumentar a expressão do microRNA 126, a razão entre p-AKT/AKT e a eNOS. Nos SHR, a DEX, não acarretou aumento da pressão arterial. Já o treinamento físico reduziu a pressão arterial e aumentou a densidade capilar e a razão capilar fibra no músculo tibial anterior e no ventrículo esquerdo. Assim, pode-se concluir o pré-condicionamento físico atenua a hipertensão arterial induzida pela DEX em ratos Wistar, por prevenir o desbalanço autonômico para o coração, bem como a rarefação da microcirculação na musculatura esquelética, resposta esta que foi associada a maior expressão do microRNA 126. Além disso, o TF per se promove angiogênese no miocárdio associada ao aumento do microRNA 126, independente do tratamento com DEX. Ainda, o TF foi eficaz em reduzir a hipertensão arterial nos SHR, independente do tratamento com a DEX, por melhorar o balanço autonômico e a microcirculação dos músculos tibial anterior e cardíaco. A doença periodontal (DP) compreende um grupo de lesões que afeta os tecidos periodontais de proteção e de suporte e pode acarretar na perda dentária. Interações entre patógenos microbianos e vários sistemas desempenham um papel crítico no desenvolvimento e progressão da DP via liberação de vários mediadores inflamatórios e imunológicos. Experimentos recentes de nosso grupo de pesquisa mostraram que existe a expressão de RNAm para todos os componentes do Sistema Renina-Angiotensina (SRA), presença da renina e atividade da Enzima Conversora de Angiotensina (ECA) em tecido gengival de ratos e que o losartan inibe a perda óssea no modelo de DP induzida experimentalmente em ratos. Portanto, o presente trabalho propõe-se a investigar os mecanismos pelos quais o losartan (antagonista do receptor AT1) possui efeitos anti-inflamatórios no modelo da DP induzida experimentalmente em ratos. Para tanto, foi utilizado o modelo de indução da DP por colocação de ligadura ao redor do primeiro molar inferior de ratos divididos em treze grupos com 15 animais cada, que foram tratados com losartan (50 mg/kg/dia) via gavage. As técnicas utilizadas neste trabalho foram: indução da DP em ratos, a análise tomográfica da perda óssea alveolar (MicroCT), análise 2D com uso de software específico (ImageJ), RT-PCR e análise histológica do tecido periodontal. Após a coleta, os dados foram devidamente analisados por meio de gráficos e tabelas, sendo utilizado ANOVA, seguida de um pós-teste para determinar a significância da diferença entre os grupos dentro de um mesmo ensaio. Foi adotado nível de significância de 5%. O losartan foi capaz de atuar no aumento da expressão de enzimas, citocinas, quimiocinas e receptores, tais como: AT2, MAS, AT1a, AT1b, AGT, OPG, ALP, PHEX, RUNX2, BGLAP, IL-18, IL-21 e MMPs. Além disso, promoveu a diminuição de citocinas pró-inflamatórias como IL-6, IL-10 e INF, indicando assim seu papel anti-inflamatório, dados observados na análise molecular do tecido gengival e ósseo. Macroscopicamente foi confirmado que o tratamento com losartan foi capaz de atenuar a perda óssea. Os achados histológicos corroboram com as análises de MicroCT e RT-PCR. Concluiu-se então que o losartan possui efeito protetor na preservação do periodonto em animais submetidos à DP devido à diminuição de moléculas envolvidas na inflamação. Periodontal disease (PD) comprises a group of lesions that affect the periodontal tissues of protection and support and can lead to tooth loss. Interactions between microbial pathogens and various systems play a critical role in the development and progression of PD through release of various inflammatory and immunological mediators. Recent experiments from our research group have shown that mRNA expression exists for all components of the Renin-Angiotensin System (RAS), presence of renin and Angiotensin Converting Enzyme (ACE) activity in rat gingival tissue and that losartan inhibits the bone loss in the experimentally induced PD model in rats. Therefore, the present work aims to investigate the mechanisms by which losartan (AT1 receptor antagonist) has anti-inflammatory effects in the PD model induced experimentally in rats. For this, the PD induction model was used by placing ligation around the lower first molar of rats divided into thirteen groups with 15 animals each, which were treated with losartan (50mg / kg / day) via gavage. The techniques used in this work were: induction of PD in rats, tomographic analysis of alveolar bone loss (MicroCT), 2D analysis using specific software (ImageJ), RT-PCR and histological analysis of periodontal tissue. After the data collection, the data were analyzed by means of graphs and tables, using ANOVA, followed by a post-test to determine the significance of the difference between groups within the same trial. A significance level of 5% was adopted. Losartan was able to increase the expression of enzymes, cytokines, chemokines and receptors, such as AT2, MAS, AT1a, AT1b, AGT, OPG, ALP, PHEX, RUNX2, BGLAP, IL-18, IL-21 and MMPs. In addition, it promoted the reduction of proinflammatory cytokines such as IL-6, IL-10 and INF, thus indicating its anti-inflammatory role, data observed in the molecular analysis of gingival and bone tissue. Macroscopically it was confirmed that treatment with losartan was able to attenuate bone loss. Histological findings corroborate with MicroCT and RT-PCR analyzes. It was concluded that losartan has a protective effect on the preservation of the periodontium in animals submitted to PD due to the decrease of the molecules involved in the inflammation.

As vias suprabulbares assim como núcleos bulbares são de vital importância na modulação cardiovascular. As informações aferentes que chegam ao NTS ascendem a áreas suprabulbares por neurônios noradrenérgicos (NORérgicos) projetando-se principalmente a neurônios pré-autonômicos do núcleo paraventricular do hipotálamo (PVN) em sua maioria ocitocinérgicos (OTérgicos) e vasopressinérgicos (VPérgicos), os quais se projetam a áreas bulbares, fechando um pronto circuito modulatório. É nossa hipótese de trabalho que a hipertensão e/ou o treinamento possam alterar a plasticidade de vias NORérgicas ascendentes as quais, modificando a sinalização aferente, desencadeariam respostas plásticas e funcionais nos neurônios pré-autonômicos do PVN, alterando a modulação cardiovascular nestas situações. Objetivo: estudar o funcionamento da alça suprabulbar de modulação autonômica, avaliando os efeitos sequenciais da hipertensão e do treinamento sobre a sinalização NORérgica ascendente e sobre a plasticidade/atividade de neurônios OTérgicos e VPérgicos do PVN que se projetam a áreas bulbares de controle cardiovascular. Material e Métodos: Protocolo I - Ratos Wistar adultos foram submetidos a T de baixa intensidade (50-60% da capacidade máxima, 5 dias/semana, 1h/dia) ou foram mantidos sendentários (S) por 8 semanas. Após registros da hemodinâmica basal nas semanas 0 (S0=T0), 1 (T1), 2 (T2), 4 (T4) e 8 (T8 e S8) [protocolo Análise Temporal do Treinamento], após 8 semanas de T ou S [para protocolo de lesão seletiva dos neurônios NORérgicos ascendentes pela saporina], e, após 12 semanas de T ou S [para os protocolos SHAM & DAS] os ratos foram eutanasiados para perfusão e fixação do encéfalo. Cortes (30 &um;m) do PVN foram processados para imuno-histoquímica de dupla marcação para OT e dopamina beta-hidroxilase (DBH); imagens foram adquiridas pelo microscópio de fluorescência e confocal e analizadas pelo ImageJ e pelo Imaris, respectivamente. Protocolo II - SHR e WKY (4 semanas de idade no início dos protocolos) foram mantidos sedentários (S) por 8 semanas. Nas semanas experimentais 0, 1, 2, 4 e 8 avaliamos os parâmetros funcionais (valores basais PA e FC) e a imunoistoquímica para VP e DBH; em outro protocolo, realizamos cirurgia Sham ou DAS em animais SHR e Wistar de 41 dias e, após a 13ª semana realizamos estudos funcionais e a análise imunoistoquímica para DBH e VP. Resultados: S determinou no PVN diminuição idade-dependente da OTir em todos os subnúcleos magnocelular (mg), dorsal cap (dc), ventromedial (vm) e posterior (Po), sem alterações significativas de DBHir, e e sem alteração de OTir em aréas bulbares, como DMV, NTS, BVLc e BVLr. T induziu aumento a partir de T2 da síntese de DBH no NTS caudal, intermediário e BVLc, e o T induziu uma diminuição da de DBH no BVLr. No PVNvm e Po houve pronto aumento da OTir em T2 vs S0, e DBHir em T2 vs S0, e aumento de botões sinápticos NORérgicos em neurônios OTérgicos, e bradicardia de repouso entre T4 e T8, sem alterar a pressão arterial. A remoção dos barorreceptores diminuiu a densidade das projeções OTérgica no NTS intermediário, BVLr e no PVN mg, dc, vm e Po, além disso a desnervação sino-aórtica bloqueiou todos os efeitos induzidos pelo treinamento, como o aumento de DBHir no NTS caudal e intermediário, no caudal e rostral VLM e em todo PVN (mg, dc, vm e Po) e impediu o aumento de OTir em ambas as aréas do bulbo quando comparamos DAS -T vs. SHAM-T. No protocolo de saporina, a marcada redução das projeções NORérgicas induziu um significativa queda na síntese de OT em todo o PVN medial e posterior, e o T não foi capaz em aumentar a OTir. Em áreas bulbares, observamos que a DBHx induziu redução na DBHir e de neurônios NORérgicos no NTS caudal e intermediário e BVLc, porém não houve alteração no BVLr em animais S. No grupo DBHx-T houve aumento na síntese proteíca de DBH em todo o bulbo, com exceção do BVLr. Houve uma marcada redução das projeções Otérgicas em todo bulbo, mas não houve alteração da OTir em DBHx, e não foi observado bradicardia de repouso. No protocolo II, WKY e SHR, de 28 dias, possuem PA semelhantes, qual aumentou a partir de S2 apenas em SHR. Animais SHR em fase pré-hipertensiva já possuem FC aumentada, comparado com os WKY, permanecendo com valores superiores até o final do protocolo, em ambas as linhagens houve uma diminuição da FC basal idade depende. Com a técnica de imunoistoquímica de fluorescência, observamos que a VPir é maior em SHR (vs. WKY) durante as 8 semanas de avaliação apenas no PVNmg, no PVNvm e posterior o conteúdo de VP é semelhante entre as duas linhagens em S0 (4 semanas de vida), porém há um aumento significativo e progressivo a partir da S1, apenas em animais SHR. Com os experimentos DAS, observamos a eficácia da cirurgia da remoção dos barorreceptores, devido a ausência da resposta reflexa bradicardica e taquicardia em animais DAS. A DASimpediu o desenvolvimento da hipertensão em animais SHR, assim como houve um bloqueio do aumento de VP em todo PVN medial (mg, vm e dc) e PVN posterior. Conclusão: A observação de que a instalação da bradicardia de repouso (a partir de T4) ocorria após o aumento da densidade de neurônios OTérgicos do PVN que se projetam ao complexo solitário-vagal sugere que alterações induzidas pelo T no controle autonômico do coração são dependentes da maior modulação OTérgica ao complexo solitário-vagal desencadeado pela sinalização NORérgica ascendente. O bloqueio destes efeitos sugere ainda que a sinalização carreada pelos barorreceptores e quimirreceptores periféricos é um fator decisivo em desencadear os efeitos benéficos do T ao sistema cardiovascular. Podemos concluir também que o aumento da PA é acompanhado pelo aumento do conteúdo de VP em áreas que contém neurônios pré-autonômicos, e que os baro e quimiorreceptores desempenham papel chave no desenvolvimento da hipertensão. Suprabulbar pathways as well as brainstem nuclei are of vital importance in cardiovascular modulation. The afferent information arriving at the NTS ascends to suprabulbar areas by noradrenergic (NORergic) neurons projecting mainly to ocitocinergics (OTergics) and vasopressinergic (VPergicos) pre-autonomic neurons of the Paraventricular Nucleus of Hypothalamus (PVN), which they project to brainstem areas, closing a modulatory circuit. It is our hypothesis that hypertension and / or training may alter the plasticity of ascending NORergic pathways which, by modifying afferent signaling, would trigger plastic and functional responses in pre-autonomic PVN neurons, altering cardiovascular modulation in these situations. Aim: To study the functioning of suprabulbar pathway in autonomic modulation, evaluating the sequential effects of hypertension and training on ascending NORergic signaling and on the plasticity / activity of OTergic and VPergic neurons projecting to cardiovascular control brainstem areas. Methods: Protocol I -Adult Wistar rats were submitted to training (T) of low intensity (50-60% of the maximum capacity, 5 days / week, 1h / day) or were kept in the sententary (S) for 8 weeks. After recording baseline hemodynamics at weeks 0 (S0 and T0), 1 (T1), 2 (T2), 4 (T4) and 8 (T8 and S8) [Time course protocol]; after 8 weeks of T or S [ for protocol of selective lesion of ascending NORergic neurons by saporin]; and after 12 weeks of T or S [for the SHAM & Sino aortic denervation (SAD) protocols] the rats were euthanized for perfusion and fixation of the brain. Slices (30 m) of the PVN were processed for immunohistochemistry double-labelling for OT and dopamine beta-hydroxylase (DBH); images were acquired by fluorescence and confocal microscopy and analyzed by ImageJ and Imaris, respectively. Protocol II - SHR and WKY (4 weeks old at the beginning of protocols) were kept sedentary (S) for 8 weeks. In the experimental weeks 0, 1, 2, 4, 8 we evaluated the functional [parameters baseline arterial pressure (AP) and heart rate (HR)] and immunohistochemistry for VP and DBH; in another protocol, we performed Sham or SAD surgery in SHR and Wistar animals of 41 days. After the 13th week we performed functional studies and immunohistochemical analysis for DBH and VP. Results: Within PVN, there was age-dependent reduction in S group of OT immunoreactivity (ir) all magnocellular (mg), dorsal cap (dc), ventromedial (vm) and posterior (Po) subnuclei, without significant changes in DBHir, and a decrease of OTir in brainstem areas such as DMV, caudal and intermediate NTS, CVLM and RVLM. T induced increase from T2 in the synthesis of DBH in the NTS caudal, intermediate and CVLM, and T induced a decrease in DBH in RVLM. In PVNvm and Po, there was a rapid increase of OTir in T2 vs S0, and DBHir in T2 vs S0, an increase of NORergic synaptic boutons in OTergic neurons and resting bradycardia between T4 and T8, without altering blood pressure. Removal of the baroreceptors decreased the density of the OTergic projections in the NTS intermediate, RVLM and in the PVN mg, dc, vm and Po. Furthermore sino-aortic denervation blocked all training-induced effects, such as increased DBHir in the caudal and intermediate NTS, within caudal and rostral VLM and in the whole PVN (mg, dc, vm and Po) and prevented the increase of OTir in both areas of the brainstem when we compared SAD-T vs. SHAM-T. In the saporin protocol, the marked reduction of the NORergic projections induced a significant decrease in OT synthesis throughout the medial and posterior PVN, and the T was not able to increase the OTir. In the S group on the brainstem areas, we observed that DBHx induced reduction in DBHir and NORergic neurons in the caudal and intermediate NTS and CVLM, but there was no change in RVLM. In the DBHx-T group there was an increase in protein synthesis of DBH in the whole brainstem, with the exception of RVLM. There was a marked reduction of the OTergics projections in every brainstem, but there was no change of OTir in DBHx, and there was no resting bradycardia. In protocol II, WKY and SHR, of 28 days, have similar PA, which increased from S2 only in SHR. SHR animals in the prehypertensive phase already have HR increased, compared with WKY, remaining with higher values until the end of the protocol, in both strain there was a decrease in HR baseline age dependent. With the fluorescence immunohistochemistry technique, we observed that VPir is higher in SHR (vs. WKY) during the 8-week evaluation only in PVNmg, in PVNvm and posterior the VP content is similar between the two strains in S0 (4 weeks of life), but there is a significant and progressive increase from S1, only in SHR animals. With the SAD experiments, we observed the efficacy of baroreceptor removal surgery, due to the absence of bradycardic and tachycardia reflex response in SAD animals. SAD prevented the development of hypertension in SHR animals, as well as a blockade of the increase of VP in all medial PVN (mg, vm and dc) and posterior PVN. Conclusion: The observation that the installation of resting bradycardia (from T4) occurred after increasing the density of PVN OTergic neurons projecting to the solitary-vagal complex suggests that T-induced changes in the autonomic control of the heart are dependent of the greater modulation OTergic to the solitary-vagal complex triggered by ascending NORergic signaling. The blockade of these effects also suggests that the signaling carried by the baroreceptors and peripheral chemoreceptors is a decisive factor in triggering the beneficial effects of T on the cardiovascular system. We can also conclude that increased BP is accompanied by increased VP content in areas containing pre-autonomic neurons, and that baro and chemoreceptors play a key role in the development of hypertension.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Hypertension (HT) is a complex and multifactorial disease considered one of the main risk factor for cardiovascular diseases. It is maintained mainly by structural changes in vessels that contribute to increase vascular resistance. Changes in the intrinsic elastic properties of the vessel wall contribute to determine arterial stiffness and may compromise vessel functioning. Currently, arterial stiffness has been considered an important predictor of cardiovascular mortality. Among the antihypertensive treatments, angiotensin converting enzyme inhibitors (ACEi) have been used to improve vessel compliance, however, almost nothing is known about the use of ACEi, associated or not with aerobic TF, on the improvement of arterial stiffness in hypertensive animals. We hypothesized that T associated with perindopril could improve arterial stiffness and HT higher than isolated treatments. Therefore, the aim of this study was to investigate the effects of aerobic T, associated or not with perindopril treatment on arterial stiffness, arterial pressure and cardiac remodeling in spontaneously hypertensive rats (SHR). 49 SHR (200-300g) were randomly allocated into 4 groups: 1 / sedentary control (SC, n = 12) 2 / sedentary treated with perindopril (SP, n = 10, 3 mg / kg per day, by gavage) 3 / trained control (TC, n = 13, which underwent treadmill aerobic exercise per 60 days), 4 / trained perindopril (TP, n = 14, which underwent aerobic TF and were treated with perindopril). Ten wistar rats were used as normotensive control. Weekly tail pressure measurements were measured. At the end of experimental protocol, pulse wave velocity (PWV) was performed to analyze arterial stiffness. After measurement of direct arterial pressure (AP), autonomic balance and barroreflex analysis, aorta artery was removed for proteomic analysis and muscle tissues (soleus and left ventricle) for histological analyses. The sedentary hypertensive rats (SC) presented lower body weight (-26.51%) and lower SOL weight (-26%) compared to wistar rats. However, hemodynamic values of SBP, DBP and MAP were elevated (+ 69.61%, 62.33% and 63.53% respectively), as well as VOP (+ 49.35%) and sympathetic nerve activity for vessels (+ 64.33%) when compared to wistar. Spontaneous (non-pharmacological) barroreflex analysis of SC had values of reflex bradycardia (GAIN UP, -43.70%), as well as the sum of reduced reflex bradycardia and reflex tachycardia (GAIN ALL, -42.47%), when compared to wistar. The histological analysis of collagen density was increased (+ 136.80%) in SC rats, whereas C: F (+ 24.34%) was higher in wistar. PWV was higher and treatment with perindopril and / or T were able to reduce it in SHR (-36%, -21% and -46%, for SP, TF and TP, respectively), compared with SC. Media wall AST values were also reduced in the SP (-59%), TC (-55%) and TP (-44%) groups, compared with SC. In addition, LV collagen density was reduced after perindopril (-53%), T (-46%) and its association (-52%). Only trained groups presented higher values of capillary-to-fiber ratio (64% and 37%, for TC and TP respectively). Sympathetic nervous activity to the vessels was also reduced only after T. Proteomic analysis suggested that some proteins that were up regulated after perindopril, were contra balanced by T, through different pathways. Results of this study suggest that reduction of arterial stiffness induced by T maybe associated with renin angiontensin system, since reduction were similar to those after perindopril treatment. Besides functional responses were similar, proteomic results suggest that both strategies act through different protein pathway. Improvements of arterial stiffness may be also associated with local and neural skeletal muscle microcirculation alterationsand both treatments did not determine synergic responses. A hipertensão arterial (HA) é uma doença complexa e multifatorial, considerada um dos principais fatores de risco de doenças cardiovasculares. Em sua fase crônica, é mantida principalmente por alterações estruturais nos vasos que contribuem para aumentar a resistência vascular. As alterações nas propriedades elásticas intrínsecas da parede do vaso contribuem para determinar a rigidez arterial, a qual vem sendo considerada um importante preditor de mortalidade cardiovascular. Os inibidores da enzima conversora de angiotensina (iECA) vêm sendo utilizados para melhorar a complacência dos vasos, no entanto, pouco se sabe sobre o uso dos iECA, associados ou não ao treinamento físico (TF) aeróbio, na melhora da rigidez arterial em animais hipertensos. A hipótese deste estudo foi que o TF, associado ao tratamento com perindopril, poderia melhorar a rigidez arterial e a pressão arterial (PA) dos SHR, quando comparados aos tratamentos isolados. O objetivo geral desse trabalho foi verificar os mecanismos induzidos pelo TF aeróbio, associado ou não ao tratamento com perindopril que podem contribuir para o remodelamento da aorta e do VE de SHR e, consequentemente, melhorar rigidez arterial e pressão arterial. Foram utilizados 49 SHR (200-300g) separados aleatoriamente em 4 grupos: 1/ sedentário controle (SC, n=12) 2/ sedentário perindopril (SP, n=10, sedentários e tratados com perindopril, 3mg/kg por dia, via gavagem) 3/ treinados controle (TC, n=13, submetidos a TF aeróbio em esteira por 60 dias), 4/ treinado perindopril (TP, n=14, submetidos a TF aeróbio e tratados com perindopril). Todos os grupos controles receberam tratamento com água e 10 ratos Wistar foram usados com controle normotenso. Semanalmente foi aferida a pressão arterial de cauda dos ratos. A rigidez arterial foi avaliada pela medida de velocidade de onda de pulso (VOP). Após análises de PA direta, do balanço autonômico e barroreflexo espontâneo, a artéria aorta foi retirada para análise proteômica e os tecidos musculares (sóleo, SOL e ventrículo esquerdo, VE) para as análises histológicas. Os SHR sedentários (SC), apresentavam peso corporal (-26,51%) e o peso do SOL (-26%) menor quando comparados aos ratos wistar, entretanto os valores de PAS, PAD e PAM encontravam-se elevados (+69,61, 62,33% e 63,53% respetivamente). Da mesma forma, a VOP (+49,35%) e a atividade nervosa simpática para os vasos (+64,33%) era maior quando comparadas com os wistar. Os SC apresentavam menores valores de efetividade de bradicardia reflexa (GAIN UP , -43,70%) e da somatória das respotas de bradicardia e taquicardia reflexas (GAIN ALL, -42,47%), quando comparado com os wistar. A densidade de colágeno encontrava-se aumentada (+136,80%) nos ratos SC, enquanto que a razão C:F (+24,34%) se apresentava maior nos wistar. It has been shown that high arterial stiffness contributes to develop hypertension (HT) and may be determined by an unbalance between the main extracellular proteins, like collagen and elastin. Previous studies have demonstrated that dexamethasone (DEX) increases arterial pressure (AP) in rats and humans; however the DEX effects on arterial stiffness are unclear. On the other hand, combined physical training (CT) has been recommended to control hypertension, but almost nothing is known about its effects on mechanisms responsible to control arterial stiffness. We hypothesized that CT could control collagen and elastin levels and attenuate arterial stiffness and AP increase in DEX-treated rats. Therefore, the aim of this study was to evaluate the effects of CT on arterial stiffness and AP in DEX-treated rats. Thirty-seven Wistar rats (200-250g) were allocated into 4 groups: sedentary control (SC, n = 8), sedentary treated with DEX (SD, n = 9), trained control (TC, n=10) and trained and treated with DEX (TD, n = 10). All rats performed a maximum voluntary carrying capacity test (MVCC, on the ladder) and a maximal physical capacity test (TEM, on the treadmill) and underwent CT (60% maximum, 5d / week, 1h / on alternate days, for 74 days) or were kept sedentary. Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq) Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) It has been shown that chronic treatment with dexamethasone (DEX) promotes cardiac remodeling and hypertension (H) in normotensive rats, however little is known about the effects of DEX in spontaneously hypertensive animals (SHR). In contrast, aerobic physical training (T) has been used as a non-pharmacological treatment in hypertension and one of its mechanisms is the improvement on cardiac remodeling. Therefore, the objective of the present study was to investigate the effects of DEX treatment and aerobic training on AP and cardiac remodeling process in SHR. For this purpose, 64 SHR (12 weeks) underwent treadmill T (60% of maximal capacity, 5 days / week, 1 h / day, for 75 days) or remained sedentary. During the last 15 days, the animals were treated with low dose of DEX (50 μg / kg body weight per day, s.c.) or saline. A group of 20 sedentary Wistar rats (W, 12 weeks) were used as control. After the experimental protocol, all rats were submitted to echocardiography, fasting blood glucose, carotid artery catheterization for AP and heart rate (HR) measurements and analysis of the autonomic nervous system. Subsequently, heart was removed for analysis of cardiac mass, capillary density, myocyte diameter and left ventricle (LV) collagen density. Chronic treatment with DEX did not exacerbate mean AP (MAP) of SHR. However, T was effective in reducing MAP of SHR (~ 20%). Treatment with low dose of DEX did not alter systolic function, but increased LV diastolic diameter (LVDD / BW) and improved diastolic function, since it reduced the isovolumetric relaxation time (IVRT), increased capillary density (+ 19%), and reduced % of collagen in the LV (-22%), regardless of myocyte size. On the other hand, T in the SHR treated with DEX promoted increase in heart weight (11%), which suggests a beneficial mass increase, which was independent of myocyte size, increased posterior wall shortening velocity (PWSV), which also suggests an improvement in systolic function, as well as increased LVDD / BW and IVRT, as well as reduced % of collagen (-28%), indicating an improvement in diastolic function. In summary, the results of the present study indicate, for the first time, that chronic treatment with low dose of DEX does not exacerbate hemodynamic and autonomic variables in SHR, but improves LV diastolic function, increases angiogenesis and reduces % of collagen. Together with DEX treatment, T determined a significant reduction on AP and promoted cardiac remodeling that contributed to a significant improvement of LV systolic and diastolic function in SHR. These results may suggest that administration of DEX at low doses and at the time used in this study can be administered to the hypertensive population without major concerns. Tem sido demonstrado que o tratamento crônico com dexametasona (DEX) promove remodelamento cardíaco e hipertensão arterial (HA) em ratos normotensos, no entanto pouco se sabe sobre os efeitos da DEX em animais espontaneamente hipertensos (SHR). Em contrapartida, o treinamento físico (TF) aeróbio tem sido utilizado como tratamento não farmacológico na HA e um dos seus mecanismos é a melhora do remodelamento cardíaco. Diante disso, o objetivo do presente estudo foi investigar os efeitos do tratamento com a DEX e do treinamento físico aeróbio na PA e no processo de remodelamento cardíaco de ratos SHR. Para isso 64 ratos SHR (12 semanas) foram submetidos a um TF em esteira (60% da capacidade máxima, 5 dias/semana, 1 h/dia, por 75 dias) ou permaneceram sedentários. Durante os últimos 15 dias, os animais foram tratados com uma dosagem baixa de DEX (50 μg/kg de peso corporal por dia, s.c.) ou salina. Um grupo de 20 ratos Wistar (W, 12 semanas) sedentário foi usado como controle. Após o protocolo experimental, todos os ratos foram submetidos a um exame de ecocardiografia, glicemia de jejum, cateterização da artéria carótida para aferição da PA e frequência cardíaca (FC) e análise do sistema nervoso autonômico. Posteriormente, o coração foi retirado para análise de massa cardíaca, densidade capilar, diâmetro de miócitos e densidade de colágeno no ventrículo esquerdo (VE). O tratamento crônico com DEX não exacerbou PA média (PAM) dos SHR. Contudo, o TF foi eficaz em reduzir a PAM destes SHR (~20%). O tratamento com baixa dose de DEX não alterou a função sistólica, mas aumentou o diâmetro diastólico do VE (DDVE/PC) e melhorou a função diastólica, pois reduziu o tempo de relaxamento isovolumétrico (TRIV), além de aumentar densidade capilar (+19%) e reduzir a % de colágeno no VE (-22%), independente do tamanho de miócito. Já o TF nos SHR tratados com DEX promoveu aumento do peso do coração (11%) que sugere ser um aumento benéfico da massa, que foi independente do tamanho de miócito, aumentou a velocidade de encurtamento da parede posterior (VEPP), que sugere também uma melhora da função sistólica, bem como aumento do DDVE/PC e TRIV, assim como redução na % de colágeno (-28%), que indicam melhora na função diastólica. Em suma, os resultados do presente estudo indicam, pela primeira vez, que o tratamento crônico com baixas dose de DEX não exacerba as variáveis hemodinâmicas e autonômicas presentes nos SHR, mas melhora a função diastólica do VE, aumenta número de vasos e reduz % de colágeno. Em conjunto com a DEX, o TF determinou redução significativa da PA e promoveu um remodelamento cardíaco que contribuiu para melhorar significativamente a função sistólica e diastólica do VE nos SHR. Estes resultados podem sugerir que a administração de DEX em doses baixas e no tempo usado nesse estudo, possa ser administrada para a população hipertensa sem grandes preocupações. On the other hand the resistance training (RT) acts in opposition to atrophic situations, although its effects on muscle atrophy induced by treatment with dexamethasone (DEX) are poorly known.The purpose of this study was to determine how long RT is required to promote preventive effects in flexor halluces longus (FHL) muscle atrophy induced by dexamethasone (DEX). After maximal voluntary carrying capacity (MVCC), 100 rats were separated in sedentary (SED) or resistance trained for 10 (RT10), 40 (RT40), 70 (RT70) and 100 (RT100) days. Groups were divided as control (CTRL) or treated with DEX. RT was performed with 80% of maximal voluntary carrying capacity (MVCC). During the last 10 days, the animals either received DEX (0.5 mg/kg/day, i.p.) or vehicle (saline, same volume as DEX treatment, i.p.). Among the main factors that affect arterial stiffness are cross-sectional area, medial wall thickening, wall-to-lumen ratio and collagen density. Recently, a device using finger-toe pulse wave velocity was validated for humans study, but to better understand the mechanisms that change arterial stiffness, it is necessary to use the arterial vessel for genic, proteic and histological analysis. Therefore, a new device – popmètre® – was designed for measurements of PWV in animals and it was not validated yet. The aim of this study was to evaluate the validity, reliability and objectivity of a new device pOpmètri® 1.0 for animal studies. Forty-two Wistar rats were allocated into 3 groups of different ages: Young: 1 month; Adult: 4 months and Older: 12 months. Two photodiodes were positioned on the right thoracic and pelvic members for PWV measurement, on anesthetized rats. Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq) Hypertension (HA) is a complex and multifactorial disease, considered as an important risk factor for cardiovascular diseases. It is known that high blood pressure (BP) levels are maintained by increased Total peripheral resistance (TPR). Among the factors that contribute to the maintenance of high TPR, there are arterioles remodeling and rarefaction, which occurs in several tissues. Physical training (T) has been considered as an important adjunct to pharmacological treatment in the control and treatment of HA. In this sense, exercise has been effective in improving vascular remodeling and promoting angiogenesis. Among the pharmacological therapies used clinically, angiotensin II converting enzyme inhibitors (ACEi) have shown great effectiveness, mainly by reducing mortality. It has been suggested that due to some different biochemical and pharmacological characteristics between captopril and perindopril, there may be some differential effect on T-induced angiogenesis. It has been demonstrated that captopril may inhibit T-induced angiogenesis after acute exercise, but almost nothing is known about it effects after chronic exercise. Therefore, the objective of this study was to compare the effects of the chronic use of captopril and perindopril on angiogenesis induced by T in normotensive and spontaneously hypertensive rats and to investigate some of the possible mechanisms involved in these alterations. For this purpose, 48 normotensive Wistar rats and 48 spontaneously hypertensive rats (SHR) underwent T protocol (60% maximum physical capacity, for 8 weeks) or maintained sedentary. During the last 6 weeks, the animals were treated with captopril (25 mg / kg per day, i.p.) or perindopril (3 mg / kg per day, gavage) and the control animals received water. Body weight and baseline values of systolic blood pressure (SBP), diastolic blood pressure (DBP), mean (MAP) and heart rate (HR) were analyzed. The anterior tibial muscle (TA) and myocardium were removed for the morphometric analyzes of the capillary / fiber ratio (C: F) and analyzes of the protein production of vascular endothelial growth factor (VEGF), its receptor (VEGFR-2) and enzyme endothelial nitric oxide synthase (eNOS). As expected, the values of SBP, DBP and MAP were higher in the sedentary SHR (+ 30%, + 33% and + 32%, for SBP, DBP and MAP respectively) when compared to sedentary Wistar. Both T and pharmacological treatments reduced BP values by a similar magnitude. The C:F ratio in TA was reduced in all sedentary SHRS compared to sedentary Wistar, and this reduction was associated with lower levels of VEGF (~ -26%) and eNOS (~ -27%). Training increased C:F ratio in trained Wistar rats compared to sedentary Wistar because of increased production of VEGFR-2 (+ 17%) and eNOS (+ 31%). In addition, T prevented TA rarefaction in SHRs, mainly by preventing reductions in VEGF and eNOS production. Chronic treatment with captopril or perindopril significantly attenuated angiogenesis in Wistar animals, although this effect was greater after treatment with captopril (-19%) when compared to perindopril (-13%). In the SHR animals, perindopril did not attenuate T-induced angiogenesis. On the other hand, captopril attenuated this effect (18%). This response occurred due to normalization of eNOS levels after PT in trained Wistar and SHR rats treated with perindopril alone. The myocardial C: F ratio was reduced in all sedentary SHR compared to sedentary Wistar. T increased myocardial C: F ratio in trained Wistar rats (~ + 14%) in all groups compared to sedentary ones. Similarly, T increased C: F ratio in trained SHR compared to sedentary SHR and normalized to number of vessels compared to sedentary Wistar. This response was also mediated by increased production of eNOS in all trained groups. These findings allow us to conclude that T promoted angiogenesis in skeletal and cardiac musculature and chronic use of ACE inhibitors attenuated this response in the skeletal muscle, and perindopril did not attenuate T-induced angiogenesis in hypertensive animals. In this way perindopril may be suggested as the best drug of choice for hypertensive patients who do exercise. É sabido que os níveis pressóricos elevados são mantidos por aumento de resistência vascular periférica (RVP). Dentre os fatores que contribuem para a manutenção da elevada RVP pode-se citar o remodelamento das arteríolas e a rarefação, ou redução da densidade capilar, que ocorre em vários tecidos. O treinamento físico (TF) vem sendo considerado um importante coadjuvante ao tratamento farmacológico no controle e tratamento da HA. Neste sentido, o exercício tem sido eficaz em melhorar o remodelamento vascular e promover angiogênese. Dentre as terapias farmacológicas utilizadas clinicamente, os inibidores da enzima conversora de angiotensina II (iECA) têm demonstrado grande efetividade, principalmente em reduzir mortalidade. Tem sido sugerido que, devido a algumas características bioquímicas e farmacológicas diferentes entre captopril e perindopril, possa haver algum efeito diferencial na angiogênese induzida pelo TF. Já foi demonstrado que o uso de captopril pode inibir a angiogênese induzida pelo exercício agudo ou de curta duração, no entanto, quase nada se sabe sobre a associação de captopril e perindopril em hipertensos que praticam exercício de longa duração (TF). Portanto, o objetivo deste estudo foi comparar os efeitos do uso crônico do captopril e perindopril na angiogênese induzida pelo TF em ratos normotensos e espontaneamente hipertensos e investigar alguns dos possíveis mecanismos envolvidos nestas alterações. Para tanto 48 ratos Wistar normotensos e 48 ratos espontaneamente hipertensos (SHR) foram submetidos a um protocolo de TF (60% capacidade física máxima, por 8 semanas) ou mantidos como sedentários. Durante as últimas 6 semanas, os animais receberam tratamento com captopril (25 mg / kg por dia, i.p.) ou perindopril (3 mg / kg por dia, gavagem) e os animais controles receberam a água. Foram analisados o peso corporal e valores basais de pressão arterial (PA) sistólica (PAS), diastólica (PAD), média (PAM) e frequência cardíaca (FC). O músculo tibial anterior (TA) e miocárdio foram retirados para as análises morfométricas da razão capilar / fibra (C:F) e análises da produção proteica do fator de crescimento endotelial de vasos (VEGF), seu receptor (VEGFR-2) e da enzima óxido nítrico sintase endotelial (eNOS). Como o esperado, os valores de PAS, PAD e PAM estavam maiores nos SHR sedentários (+30%,+33% e +32%, para PAS, PAD e PAM, respectivamente), quando comparados aos Wistar sedentários. Tanto o TF, como ambos os tratamentos farmacológicos, reduziram de similar magnitude os valores de PA. A razão C:F no TA estava reduzida em todos os SHR sedentários, comparados aos Wistar sedentários, e esta redução estava associada aos menores níveis de VEGF (~ -26%) e eNOS (~ -27%). O TF aumentou a razão C:F nos ratos Wistar treinados comparados ao Wistar sedentário devido a um aumento na produção do VEGFR-2 (+17%) e da eNOS (+31%). Adicionalmente, o TF preveniu a raferação no TA dos SHR, principalmente por prevenir as reduções das produções do VEGF e eNOS. O tratamento crônico com captopril ou perindopril atenuou significativamente a angiogênese nos animais Wistar, embora este efeito foi maior após tratamento com captopril (-19%), quando comparado ao perindopril (-13%). Já, nos animais SHR, o perindopril não atenuou a angiogênese induzida pelo TF, diferente dos resultados apresentados pelo captopril, que atenuou a angiogênese em 18%. Esta resposta permitiu a normalização dos níveis da eNOS, após o TF, nos ratos treinados Wistar e SHR tratados com somente com perindopril. A razão C:F do miocárdio estava reduzida em todos o SHR sedentários comparado ao Wistar sedentário. O TF aumentou a razão C:F do miocárdio nos ratos Wistar treinados (~+14%) em todos os grupos comparados aos sedentários Da mesma forma, o TF aumentou a razão C:F nos SHR treinados, comparados aos SHR sedentários e normalizou a quantidade de vasos em relação aos Wistar sedentário. Esta resposta também foi mediada pela maior produção da eNOS em todos os grupos treinados. Estes achados nos permitem concluir que o TF promoveu a angiogênese na musculatura esquelética e cardíaca e o uso crônico dos fármacos iECA atenuou esta resposta na musculatura esquelética. O perindopril não atenuou a angiogênese induzido pelo TF nos animais hipertensos. Assim, apesar de não haver diferenças na redução da PA nos SHR tratados com captopril ou perindopril, os resultados do presente estudo sugerem que o perindopril possa ser o melhor fármaco de escolha para pacientes hipertensos que praticam exercício físico, uma vez que não atenua a angiogênese na musculatura esquelética induzida pelo TF. O íon fluoreto (F) provém do elemento flúor. Sua absorção é inversamente relacionada ao pH e ocorre rapidamente no estômago e posteriormente no intestino delgado. Após sua absorção, o F é distribuído pelos tecidos através da corrente sanguínea e armazenado nos tecidos calcificados e moles. Sua excreção acontece por via renal. Trata-se de um elemento relevante em termos de Saúde Pública, devido às suas propriedades de prevenir ou reverter lesões cariosas em indivíduos de todas as idades. No entanto, sua ingestão excessiva é capaz de afetar o metabolismo ósseo e desenvolvimento do esmalte dentário. Estudos sugerem que o F pode interferir em vias metabólicas, inibindo a ação de diversas enzimas. Entretanto, a literatura é conflitante em relação aos seus efeitos na homeostasia da glicose, o que poderia, talvez, ser explicado pela diferença genética entre as linhagens utilizadas. Sabe-se que camundongos da linhagem A/J são extremamente sensíveis aos efeitos do F, enquanto que os camundongos da linhagem 129P3/J são altamente resistentes ao tratamento com esse íon. Por este motivo, foi investigado se esses animais que sabidamente apresentam uma expressão proteica diferencial em função do F devido ao seu background genético apresentam também respostas diferentes em parâmetros bioquímicos (glicemia jejum, insulinemia, índice de resistência à insulina [HOMA2-IR] e teste de tolerância à insulina) e imunológicos (TNF-α). Após aprovação da Comissão de ética, 156 animais (78 da linhagem A/J e 78 da linhagem 129P3/J) foram divididos em 3 grupos para cada linhagem, e tratados por um período de 42 dias com doses de 0, 15 ou 50 ppm de F na água e ração com baixo teor de F. A doença periodontal (DP) corresponde a um grupo de doenças inflamatórias que acomete as estruturas periodontais de proteção e de suporte e pode levar à perda dentária. A etiologia está relacionada à placa dentobacteriana que leva à produção de grande quantidade de citocinas pró-inflamatórias importantes na destruição tecidual. A angiotensina (Ang) II também pode contribuir para a inflamação e destruição tecidual no periodonto agindo como mediador chave. A utilização de drogas que atuem na cascata do sistema renina-angiotensina (SRA) poderia interferir no estado de saúde ou inflamação do tecido mole, na perda óssea alveolar e na expressão gênica dos componentes do SRA e mediadores inflamatórios. Portanto, o objetivo do presente trabalho foi investigar se o ramipril, um inibidor da enzima conversora de angiotensina (ECA), altera a progressão da DP induzida experimentalmente em ratos. Foi utilizado o modelo de indução da DP por colocação de ligadura ao redor do primeiro molar inferior direito de ratos. Os grupos com 10 animais cada, foram divididos em tratados com ramipril (via gavagem 10 mg/kg/dia) ou água (veículo) durante 14 e 21 dias e o grupo Sham submetido à indução fictícia da DP. Outros quatro grupos foram submetidos ao pré-tratamento com ramipril durante os períodos de 7 e 14 dias e após a indução da DP e tratados por 14 ou 21 dias. As metodologias de avaliação foram: extração de RNA total, transcrição reversa seguida de reação em cadeia da polimerase quantitativa (RTqPCR), análises histológica e da perda óssea alveolar. Os dados foram analisados por meio de gráficos e os resultados foram submetidos à análise unidirecional de variância (ANOVA) e representaram médias e respectivos desvios-padrão. Diferenças entre os grupos foram consideradas estatisticamente significativas quando p < 0,05. Com base nos resultados obtidos pode-se concluir que o ramipril foi capaz de reduzir a progressão da perda óssea no grupo tratado por 21 dias (DP-21d-Rami), entretanto houve aumento do processo inflamatório, além de alteração da expressão de RNAm de ECA-2 e do receptor Mas, alguns mediadores do processo inflamatório, como COX2 e VEGF, e os receptores VEGF-R1 e VEGF-R2. Periodontitis (PD) consists of a group of inflammatory diseases that affect the protecting and supporting periodontal structures, and may lead to tooth loss. The etiology is related to the dentobacterial plaque that produces a large amount of proinflammatory cytokines with an important action on tissue destruction. Angiotensin (Ang) II may also contribute to the inflammation and periodontal tissue destruction by acting as a key mediator. The use of drugs that affect the cascade of the reninangiotensin system (RAS) might interfere with the healthy or inflammatory status of the soft tissue, alveolar bone loss and gene expression of RAS components and inflammatory mediators. Therefore, the aim of this work was to evaluate whether ramipril, an angiotensin-converting enzyme (ACE) inhibitor, alters the progression of experimentally-induced PD in rats. The model of PD induction by placement of a silk ligature around the right lower first molar was used. Groups with 10 animals each were divided into ramipril-treated (10 mg/kg/day, via gavage), water (vehicle) and the sham surgery group (sham) for 7 or 14 days previously to PD induction and after this period the drug was administered for 14 and 21 days. The techniques employed were: total RNA extraction, reverse transcription followed by quantitative polymerase chain reaction (RT-qPCR) as well as histological and alveolar bone loss analyses. Data were analyzed by means of graphs and the results submitted to unidirectional analysis of variance (ANOVA) and represent the means with respective standard deviations. Differences between groups were considered statistically significant when p < 0.05. Based on the results obtained in this work, it was concluded that ramipril was able to reduce the progression of alveolar bone loss in the group treated for 21 days (DP-21d-Rami), however, there was an increase in the inflammatory process, besides altering the expression of ACE-2, Mas receptor and some inflammatory mediators such as COX2, VEGF and the receptors VEGF-R1 and VEGF-R2.

O músculo estriado esquelético é uma entidade extremamente versátil, capaz de alterar seus padrões e características fenotípicas sob diversas condições, tais como, atividade neuromuscular, estimulação elétrica, idade, atividade hormonal e exercício físico. Sabe-se também que o balanço entre estímulos atróficos e hipertróficos controlam diretamente a massa muscular do indivíduo e estas variações implicam diretamente não apenas sobre o volume muscular, mas também conteúdo de proteínas e produção de força. Neste sentido, a atrofia muscular (perda de massa muscular) é caracterizada tanto pela diminuição na área de secção transversa das fibras musculares, como também pelo decréscimo no conteúdo de proteínas miofibrilares e consequente redução do volume muscular. Esta atrofia muscular pode ocorrer sob diversas condições clinicas e/ou patológicas, acarretando na diminuição ou perda da massa magra levando à uma consequente diminuição da atividade física, da qualidade de vida e inclusive pior resposta ao tratamento, ocasionando consequentemente aumento da mortalidade. Fica claro desta maneira, que, os métodos que visam a prevenção ou tratamento à atrofia muscular tem importante relevância clínica em muitos grupos de pacientes, além de ser um importante fator contribuinte na qualidade de vida e autonomia destes indivíduos. Com isso o estudo de determinados tratamentos que combatam a atrofia muscular torna-se de vital importância, dentre os quais, vem ganhando destaque o βHidroxi βMetilbutirato (HMB). Deste modo teve-se como objetivo neste trabalho verificar a efetividade do HMB em prevenir a atrofia muscular induzida por dexametasona (DEXA). Para isto, foram utilizados 32 animais da linhagem Wistar com idade de 60 dias, distribuídos nos seguintes grupos: Grupo Experimental Placebo (GEP), n=8, tratados por 10 dias consecutivos com gavagem e injeção intraperitoneal, ambas contendo apenas solução salina; Grupo Experimental Dexametasona (GED), n=8, tratados por 10 dias consecutivos com gavagem contendo solução salina e injeção intraperitoneal contendo dexametasona; Grupo Experimental HMB (GEH), n=8, tratados por 10 dias consecutivos com gavagem contendo HMB e injeção intraperitoneal contendo solução salina; e Grupo Experimental Dexametasona + HMB (GEDH), n=8, tratados por 10 dias consecutivos com gavagem contendo HMB e injeção intraperitoneal contendo DEXA. Os animais foram acondicionados em caixas coletivas com 4 animais por caixa, com comida e agua à vontade em sala climatizada com temperatura de 22o e respeitando o ciclo de 12 horas claro/escuro. Finalizados os dez dias de tratamento, os animais foram eutanasiados para a coleta do material. O processo inflamatório pode ser iniciado pela presença de patógenos (PAMPs) ou de moléculas associadas ao dano tecidual (DAMPs), estes sinais são reconhecidos por células do sistema imune via receptores de reconhecimento de padrão localizados na membrana ou no citoplasma destas células. A ativação da família de receptores intracitoplasmáticos do tipo NOD (NLR) pode levar à formação de complexos proteicos denominados inflamassomas, entre eles, o inflamassoma NLRP3, formado por três estruturas básicas: receptores do tipo NOD, proteína adaptadora (ASC) e pró-caspase 1. Essas caspases ativas são capazes de induzir a maturação proteolítica de interleucina-1 beta (IL-1b) e IL-18, que então são liberadas para o meio extracelular. É sabido que a hipertensão é uma doença inflamatória associada à presença de DAMPs em regiões de controle da pressão arterial, como o núcleo paraventricular do hipotálamo (PVN) bem como, que o treinamento aeróbio é uma conduta eficaz em reverter muitos dos efeitos deletérios da hipertensão. Desta forma, é nossa hipótese de trabalho que inflamassomas NLRP3 possam estar ativados na hipertensão e que o treinamento aeróbio corrija muitos déficits do controle cardiovascular por modular a resposta imune inata. Em ratos machos adultos Wistar Kyoto e ratos espontaneamente hipertensos (SHR), foram feitas análises de parâmetros cardiovasculares, além da expressão gênica dos constituintes do inflamassoma e citocinas, e proteica de citocinas no PVN de animais sedentários ou treinados por quatro semanas de ambas as linhagens. Nossos resultados mostram que animais hipertensos se caracterizam por valores aumentados de pressão arterial e frequência cardíaca de repouso, associados à disfunção autonômica ao coração e vasos. SHR apresentam inflamação no PVN, o que foi evidenciado pelo elevado perfil de expressão dos constituintes da via do inflamassoma NLRP3, TLR-4 e citocinas pró-inflamatórias. O treinamento aeróbio de baixa a moderada intensidade reduziu a expressão gênica desses parâmetros e proteica de citocinas pró-inflamatórias, bem como a disfunção autonômica, além de determinar a instalação da bradicardia de repouso. Nossos dados sugerem que a elevada expressão de citocinas pró-inflamatórias no PVN de SHR seja devido à ativação do inflamassoma NLRP3, numa via dependente de TLR-4. Nosso trabalho é o primeiro a investigar o papel de inflamassomas em regiões autonômicas em hipertensos, bem como o efeito do treinamento físico sobre este perfil inflamatório. The inflammatory process can be triggered by the presence of pathogens (PAMPs) or damaged-associated molecules pattern (DAMPs), this danger signals are recognized by immune system via pattern recognition receptors (PRR) in membrane or in the cytoplasm cells. The activation of NOD like receptors in the cytoplasm can trigger the formation of protein complex called inflammasomes, such as the NLRP3 inflammasome, which is comprised of three basics structures: NOD like receptor, adaptor protein and pro-caspase-1. This complex can induce the proteolytic maturation of pro-inflammatory cytokines (PICs), like interleukin 1 beta (IL-1b) and IL18, which are then released into the extracellular medium. It is known that hypertension is an inflammatory disease associated with DAMPs in blood pressure controlling areas in hypothalamus, like paraventricular nucleus (PVN) and that aerobic training can reverse the deleterious effects of hypertension. So, is our hypothesis that NLRP3 inflammasomes can be active in hypertension and that the exercise training restored the cardiovascular deficits by modulate the innate response. In adult male rats Wistar Kyoto and spontaneously hypertensive rats (SHR), cardiovascular parameters, gene expression of inflammasomes members or cytokines and cytokines protein contents were measured in the PVN of sedentary or four weeks trained groups from both lineages. Our results show that hypertensive animals are characterized by enhanced blood pressure and resting heart rate values, associated with autonomic dysfunction to heart e vessels. SHR had PVN inflammation, what is evidenced by the enhanced inflammasome NLRP3, TLR-4 and PICs expression in this area. The aerobic training with low or moderate intensity reduce the gene expression of these parameters and the protein expression of PICs, as well as, the autonomic dysfunction, in addition to determining the installation of resting bradycardia. Our data suggest that the elevated expression of PICs in SHR PVN is due to the activation of NLRP3 inflammasomes, in a TLR-4 dependent pathway. Our work is the first to investigate the role of inflammasomes in autonomic brain areas in hypertensive rats, as well as, the effect of exercise training on this inflammatory profile.

O presente trabalho teve como objetivo avaliar e comparar a reatividade vascular de agentes vasoconstritores presentes nas soluções anestésicas locais (Adrenalina - vasoconstrição e vasodilatação; Felipressina - vasoconstrição), nas doses de 80, 160, 320, 640 e 1280ng (adrenalina) ou 0,25; 0,5; 1; 2 e 4 x10-3UI (felipressina), em leito arterial mesentérico deratos normotensos, diabéticos, hipertensos renais um-rim, um-clip (1R-1C) e hipertensos1R-1C-diabéticos. E correlacionar tal reatividadecom expressão de RNAm dos receptores 1A e 2- adrenérgicos, V1A para vasopressina e AT1A, AT1Be AT2 para angiotensina II visando verificar se a hipertensão arterial e o diabetes mellitus provocam alteração em modelo indutivo e isogênico. Ratos Wistar pesando 110-160g, foram anestesiados com mistura de quetamina e xilazina (50+10mg/ml/kg de peso), tiveram seu abdômen aberto e receberam um clip de prata com abertura 0,25mm na artéria renal esquerda, removendo-se cirurgicamente o rim direito (ratos 1R-1C). Após 14 dias, receberam injeção subcutânea de estreptozotocina (50 e 60mg/kg de peso) para indução do diabetes mellitus sendo a glicemia testada pela veia caudal previamente aos experimentos (diabéticos). Após 30-42 dias da implantação do clip, todos os grupos foram novamente anestesiados e implantou-se cânula de polietileno (PE-50) na artéria carótida esquerda para registro direto da pressão arterial. Após registro da pressão os animais tiveram a artéria principal mesentérica exposta e canulada. O leito arterial mesentérico foi então isolado e colocado em banho com solução nutritiva de Krebs a 37ºC. O cateter foi conectado ao sistema de registro computadorizado (PowerLab®) utilizando software específico (Chart 5Pro ®). Analisaram-se: a pressão máxima (vasoconstrição) e mínima (vasodilatação), o tempo necessário para atingir esse valor, duração total da resposta, integral e integral sobre a linha de base. Os dados foram submetidos à análise de variância de medidas repetidas (ANOVA), seguida do teste de Holm-Sidak (distribuição normal) ou de Mann-Whitney (nãoparamétrico), quando apropriado, nível de significância de 5%. Além das alterações autonômicas, a síndrome metabólica (SMet) causa disfunção vascular relacionada a eventos cardiovasculares e morte. Considerando que a resistência à insulina é associada à hiperativação simpática, testamos a hipótese de que a presença da glicemia de jejum alterada (GLI) é a principal causa das alterações estruturais e funcionais em grandes e pequenos vasos, via aumento do tônus simpático nesses pacientes. Foram avaliados pacientes com SMet recém diagnosticados (ATP-III) e não medicados, divididos em: glicemia de jejum alterada >=100mg/dL (SMet+GLI, n=35; 50±1 anos) e glicemia normal < 100mg/dL (SMet-GLI, n=24, 46 ± 1). Um grupo de indivíduos saudáveis foi estudado como controle (CS, n=17, 50±1 anos). Foram medidas rigidez arterial (velocidade de onda de pulso, VOP), atividade nervosa simpática muscular (ANSM, microneurografia) fluxo sanguíneo muscular (FSM, pletismografia), pressão arterial média (PAM, oscilométrico), resistência vascular periférica (RVP=PAM/FSM) e dimetilarginina assimétrica (ADMA). SMet+GLI apresentou maior VOP que SMet-GLI e CS (8,0[7,2-8,6], 7,3[6,9-7,9] e 6,9[6,6-7,2]m/s, respectivamente, P=0,001), não havendo diferença entre SMet-GLI e CS. Além disso, SMet+GLI foi similar ao SMet-GLI mas teve maior RVP que CS (P=0,008), não havendo diferença entre SMet-GLI e CS. Adicionalmente, SMet+GLI teve maior ANSM que SMet-GLI e CS; enquanto SMet-GLI teve maior ANSM que CS (31+-1; 26+-1; 19+-1 disparos /min, P < 0,001). ADMA foi similar entre os grupos (0,62 [0,56-0,71], 0,67 [0,59-0,92] e 0,60 [0,54-1,43] umol/L). Dentre os fatores de risco da SMet, GLI foi preditor do aumento da ANSM. ANSM foi associada à VOP (R=0.39; P=0,002) e à RVP (R=0,30, P=0,034). Em conclusão, a hiperativação simpática, que está aumentada na presença da glicemia alterada, é o mecanismo básico que pode explicar, pelo menos em parte, o aumento na VOP e na RVP. Introdução: A hipertensão arterial (HA) tem tomado proporções significativas nas sociedades atuais, sendo uma das principais causas de mortalidade na população idosa. Alguns estudos têm mostrado que é devido ao alto grau de complexidade da HA e partindo do pressuposto que a variação interindividual dos valores da pressão arterial (PA) é, em parte, determinada geneticamente, algumas abordagens vêm sendo utilizadas para identificar os genes que participam da etiologia da HA. Essa estratégia baseia-se no princípio de que alterações genéticas, envolvidas em funções fisiológicas específicas, contribuem para a variação da PA. Alguns polimorfismos que têm sido estudados são os relacionados com os sistemas reninaangiotensina-aldosterona e o da sintase endotelial do óxido nítrico. Contrapondo-se aos efeitos dos polimorfismos, o exercício físico é considerado umas das principais ferramentas não farmacológicas para o controle da hipertensão. Objetivo: Desta forma, o objetivo deste estudo é investigar de forma integrada a relação dos polimorfismos sistemas renina-angiotensina e da síntese endotelial do óxido nítrico no controle da PA de idosos normotensos e hipertensos e, como esta relação pode ser modulada pelo nível de condicionamento físico (CF). Método: Trata-se de um estudo transversal realizado em Bauru, SP o qual participaram do estudo 155 idosos com idade média 66,94 (6,83) anos, que realizaram os seguintes testes: Bateria de testes para determinar nível de CF por meio do índice de aptidão funcional geral (IAFG), pressão arterial sistólica (PAS) e diastólica (PAD), coleta de sangue para extração do DNA (QIAamp DNA Mini Kit) e posterior análise dos polimorfismos do gene da eNOS -786T>C; INTRON 4 e 894G>T e M235T do Gene do Angiotensinogênio; D/I do Gene da ECA; A1166C Gene do Receptor AT1 do SRAA e, quantificações da atividade da ECA (Ensaio fluorimétrico) e nitrito (Método da quimioluminescência). Resultado e Conclusão: O IAFG é a variável que parece exercer maior influência nas variáveis antropométrica, hemodinâmicos e humorais. Notou-se também uma associação entre os haplótipos dos genes do SRAA, MDA, TIA e TIC, ao risco de HA. E ainda, identificou-se um modelo de interação significativo entre o gene eNOS e AGT em hipertensos comparados a normotensos obtidos pelo método MDR. Background: Hypertension (HA) has taken significant proportions in contemporary societies, one of the leading causes of mortality in the elderly population. Some studies have shown that it is due to the high complexity of HA, assuming that the interindividual variation in blood preswsure (BP) are in part genetically determined. There are some approaches which have been used to identify genes that participate in the origin of HA. This strategy is based on the principle of genetic alterations involved in specific physiological functions that contribute to the variation of the BP. Some polymorphisms that have been studied are related to the renin-angiotensinaldosterone system (RAAS) and the syntase of endothelial nitric oxide (eNOS). Opposed to the effects of polymorphisms, physical exercise is considered one of the main tools for non-pharmacological management for HA. Objectives: The purpose of this study was to investigate the relationship between eNOS and RAAS polymorphisms and the effect of these interaction in the BP control in hypertensive and normotensive elderly people, and, how this relationship can be modulated by the level of physical fitness. Methods: This is a cross-sectional study developed in Bauru-SP. Participated in this study 155 elderly (66.94±6.83 years old), who underwent the following tests: battery test to determine the fitness level through the general functional fitness index (GFFI), systolic and diastolic blood pressure (SBPand DBP) , blood collection for DNA extraction (QIAamp DNA Mini Kit) and subsequent analysis of polymorphisms of the eNOS gene -786T> C; INTRON 4 and 894G> T and M235T of angiotensinogen gene; D / I of the ACE gene; A1166C Gene AT1 Receptor RAAS and quantifications of ACE activity (fluorimetric assay) and nitrite concentration (chemiluminescence Method). Results and Conclusion: The GFFI is the variable that appears to exert greater influence on anthropometric, hemodynamic and humoral values. It was noted an association between the haplotypes of RAAS genes, MDA, TIA and TIC and the risk of hypertension. Additionally, it was identified a significant interaction model between eNOS and AGT gene in hypertensive compared to normotensive obtained by MDR method.

O Sistema renina-angiotensina (SRA) tem sido relatado como um importante modulador de processos inflamatórios e imunológicos, incluindo a doença periodontal (DP). Estudos sugerem neste sistema um eixo alternativo (ECA-2 /ANG(1-7) /MAS) que atuaria como um contra-regulador de efeitos mediados pelo clássico eixo (ECA /ANGII /AT1). Sabe-se que bactérias periodontopatogênicas, como a Porphyromonas gingivalis (Pg), possuem componentes bioativos de membrana (ex. lipopolissacarídeos-LPS) capazes de induzir uma forte resposta imune no hospedeiro devido à liberação de citocinas nas células, entre elas Interleucina (IL)- 1β. Neste contexto, fibroblastos são as células mais abundantes nos tecidos periodontais e possuem em sua superfície celular receptores necessários para o reconhecimento da invasão bacteriana, ativando cascatas intracelulares, que levam à produção de citocinas. O objetivo deste estudo foi verificar se os eixos ECA/ ANGII/ AT1 e ECA-2/ ANG(1-7)/ MAS contribuem para a produção e/ ou regulação de citocinas inflamatórias (CI) por fibroblastos de gengiva humana (HGF) e ligamento periodontal humano (HPLF) estimulados por IL-1β. Após o pré-tratamento com Losartan e Ang (1-7) ou silenciamento mediado por RNA de interferência (RNAi) de AT1, HGF e HPLF foram estimulados por IL-1β por 3 horas (RNAm) ou 24 horas (proteína). Expressão de RNAm para AT1, MAS, ECA, ECA-2, IL-1β, TNF-α, IL-6, IL-8, IL-10, TGF-β, CXCL12, RANK-L e OPG foram avaliados por RT-qPCR e das proteínas IL-6, IL-8, ECA e ECA-2 por ELISA. Foi realizado também Western Blot para detecção de AT1 e ECA nos extratos celulares e dosagem de nitrito no sobrenadante das culturas. Ambos os subtipos de fibroblastos mostraram aumento da expressão de RNAm para AT1, IL-1β, IL-6, IL-8, TNF-α e OPG, quando estimulados por IL-1β. No entanto, apenas em HPLF foi observado aumento para MAS, ECA e TGF-β. Losartan e Ang (1-7) não modularam o transcrito, a secreção de CI e nem a produção de nitrito no sobrenadante das culturas, tanto em HGF como em HPLF. O silenciamento do receptor AT1 reduziu a secreção de IL-6 e IL-8 induzida por IL-1β em cultura de HGF e HPLF e aumentou a expressão gênica de OPG somente em HGF. Estes resultados sugerem que o silenciamento de AT1, mas não o bloqueio farmacológico deste receptor pelo antagonista Losartan, em HGF e HPLF, pode controlar a produção de IL-6 e IL-8, que por sua vez contribuem para a patogênese periodontal. The renin-angiotensin system (RAS) has been reported as an important modulator of inflammatory and immune responses, including periodontal disease (PD). Studies suggest an alternative axis as part of this system (ACE-2 / ANG (1-7) / MAS) that would act as counter-regulatory to the classical axis (ECA / ANGII / AT1). It is known that periodontal bacteria such as Porphyromonas gingivalis (Pg) have bioactive components in their membrane (such as lipopolysaccharide-LPS) capable of inducing a strong immune response in the host due to the release of cytokines in cells, including interleukin (IL) - 1β. In this regard, fibroblasts are the most abundant cells in periodontal tissues and receptors needed for the recognition of bacterial invasion by activating intracellular cascades that lead to cytokine production. The aim of this study was to determine whether the axes ACE / ANGII / AT1 and ACE-2 / ANG (1-7) / MAS contribute to the production and / or regulation of inflammatory cytokines (IC) by fibroblasts of human gingiva (HGF) and human periodontal ligament (HPLF) stimulated IL-1β. After pre-treatment with Losartan, Ang (1-7) or silencing mediated by RNA interference (RNAi) of AT1, HGF and HPLF were stimulated by IL-1β for 3 hours (RNAm) or 24 hours (protein). Expression mRNA for AT1, MAS, ACE, ACE-2, IL-1β, TNF-α, IL-6, IL-8, IL-10, TGF-β, CXCL12, RANK-L and OPG was assessed by RT- qPCR and proteins IL-6, IL-8, ACE and ACE-2 by ELISA. Western Blot for the detection of AT1 and ECA and dosage of nitrite was also performed. Experiments stimulated by IL-1β showed a positive control for gene expression AT1, IL-1β, IL-6, IL-8, TNF-α and OPG in HGF and HPLF and MAS, ACE and TGF-β only HPLF. Losartan and Ang (1-7) did not modulate the transcription and secretion of IC and no nitrite production in the culture supernatant of HGF and HPLF. The silencing AT1 reduced IL-6 secretion and IL-8 induced by IL- β in cultured HGF and HPLF and increased OPG gene expression only HGF. These results suggest that silencing AT1, but not pharmacological blockade of this receptor by Losartan in HPLF and HGF, can control the production of IL-6 and IL-8, which in turn contribute to the pathogenesis of periodontal disease.

Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Dexamethasone (DEX) is widely used due to its potent anti-inflammatory effect, although its chronic use may cause unwanted effects, including arterial hypertension (H) and muscle atrophy. The mechanisms responsible for these effects remain unclear. Nervous system, microcirculation and oxidative stress may be involved in H. In addition, oxidative stress may also be responsible for muscle atrophy. On the other hand, exercise training (T) its recommended for H treatment and it is an oxidative stress modulator. The aim of this study was to investigate if DEX-induced H is associated with changes in the autonomic nervous system, microcirculation and oxidative stress. Moreover, it evaluated if arterial pressure (AP) reduction induced by T is associated with a better balance of these mechanisms. Still, it investigated if muscle atrophy was also involved with oxidative stress and if T could improve this response. Rats were submitted to T for 8 weeks or kept sedentary and then treated or not with DEX (50μg/kg/day for 14 days), composing four groups: sedentary control (SC), sedentary DEX (SD), trained control (TC) and trained DEX (TD). Body weight (BW), basal AP, autonomic balance to the heart and sympathetic nerve activity to the vessels were analyzed. Aorta was collected for evaluation of oxidative stress and tibialis anterior (TA) and soleus (SOL) muscles were collected for microcirculation histological analysis and for gene and protein evaluations of gp91phox, p47phox, SOD-1, SOD-2 and CAT. DEX decreased BW (-29.1g), increased AP (+12.1%), TA muscle atrophy (- 8.2%), increased low-frequency waves to the heart (+58.5%) and vessels (+96.6%), decreased high-frequency waves to the heart (-12.0%), reduced capillary density and capillary-fiber-ratio (C:F ratio) in TA (respectively -21.9%, -11.1%) and SOL (respectively -25.9%, -22.2%) muscles. In contrast, T mitigated AP increase (-10.0%), atennuated low-frequency waves increase to the heart (-21.5%) and vessels (-8.4%) and also high-frequency waves decrease to the heart (+8.4%). Moreover, T prevented capillary density and C:F ratio reductions in TA (respectively, +46.0% +31.6%) and SOL (respectively, +44.4% +37.5%) muscles and increased SOD-2 and CAT protein level in TA (respectively + 37.4%, + 20.9%) and SOL (respectively +15.4%, +18.0%) muscles. The results allow us to suggest that DEX-induced H may be associated with changes in sympathetic nervous system to the heart and vessels, parasympathetic nervous system to the heart and also microcirculation rarefaction. In the other hand, T was able to mitigate AP increase due to autonomic balance improvement and rarefaction prevention. Since T increased rat´s antioxidant capacity independent of attenuation of muscle atrophy, we may suggest that oxidative stress is not involved in DEX-induced muscle atrophy. A dexametasona (DEX) é amplamente utilizada devido ao seu potente efeito antiinflamatório, contudo seu uso crônico pode acarretar efeitos deletérios, dentre eles a hipertensão arterial (HA) e a atrofia muscular. Os mecanismos responsáveis por esses efeitos permanecem incompreendidos. O sistema nervoso, a microcirculação e o estresse oxidativo parecem estar envolvidos com a HA. O estresse oxidativo também pode ser responsável pela atrofia muscular. Por outro lado, o treinamento físico (TF) é recomendado no tratamento da HA e é considerado moderador do estresse oxidativo. O objetivo do estudo foi investigar se a HA induzida pela DEX está associada com alterações no sistema nervoso autônomo, na microcirculação e ao estresse oxidativo. Além disso, avaliar se a diminuição da PA induzida pelo TF está associada a um melhor ajuste nestes mecanismos. Ainda, investigar se a atrofia muscular também estaria envolvida com o estresse oxidativo e se o TF poderia contribuir para melhora dessa alteração. Ratos Wistar foram submetidos a um protocolo de TF ou mantidos sedentário por 8 semanas. Os animais foram tratados ou não com DEX (50μg/kg, por dia, por 14 dias) compondo 4 grupos: sedentário controle (SC), sedentário DEX (SD), treinado controle (TC) e treinado DEX (TD). Foram analisados peso corporal (PC), pressão arterial em repouso, balanço autonômico para o coração e atividade simpática para os vasos. A aorta foi coletada para avaliação do estresse oxidativo e os músculos tibial anterior (TA) e sóleo (SOL) coletados para realização das análises histológicas na microcirculação e avaliações gênicas e proteicas da gp91phox, p47phox, SOD-1, SOD- 2 e CAT. A DEX provocou redução do PC (-29,1g), aumento da PA (+12,1%), atrofia muscular no TA (-8,2%), aumento das ondas de baixa frequência para o coração (+58,5%) e para os vasos (+96,6%), diminuição das ondas de alta frequência para o coração (-12,0%), redução da densidade capilar e da razão capilar fibra nos músculos TA (respectivamente, -21,9%, -11,1%) e SOL (respectivamente -25,9%, -22,2%). O TF atenuou o aumento da PA (-10,0%), o aumento das ondas de baixa frequência para o coração (-21,5%) e para os vasos (-8,4%) e a diminução das ondas de alta frequência para o coração (+8,4%), além de prevenir a redução da densidade capilar e da razão capilar fibra, tanto no TA (respectivamente, +46,0%, +31,6%) como no SOL (respectivamente, +44,4%, +37,5%) e aumentar a produção protéica da SOD-2 e CAT no TA (respectivamente, +37,4%, +20,9%) e no SOL (respectivamente, +15,4%, +18,0%). Assim, podemos concluir que a HA induzida por DEX pode estar relacionada tanto com alterações no sistema nervoso autônomo para o coração e no sistema simpático para os vasos, como com a rarefação encontrada na microcirculação e que o TF foi capaz de atenuar esse aumento da PA devido a uma melhora no balanço autonômico para o coração e a prevenção da rarefação na microcirculação. O aumento da capacidade antioxidante induzido pelo TF independente da atenuação da atrofia muscular sugere que o estresse oxidativo parece não influenciar a atrofia muscular induzida pela DEX.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) Dexamethasone (DEX) is widely used in clinic due to its effectiveness as anti- inflammatory. Nevertheless, its chronic use may cause unwanted metabolic and hemodynamic alterations, which contribute for the development of hypertension. Arterial hypertension may be determined by increases in cardiac output or peripheral resistance and rarefaction may be associated with this response. Microcirculation maintenance is dependent on the balance between anti-apoptotic and apoptotic proteins and vascular endothelial growth factor (VEGF), known to be a key-regulator protein of the physiological angiogenesis, helps to maintain this balance. We recently demonstrated that high doses with DEX-treatment reduce VEGF protein levels, but the mechanisms involved in this response were not evaluated. In the other hand, it has been shown that aerobic training (T) is a good strategy in preventing rarefaction and/or stimulating angiogenesis, however almost nothing is known about the effects of T on microcirculation and hypertension induced by DEX. Therefore, the aim of this study was to investigate the mechanisms induced by T that can contribute to attenuate DEX-induced rarefaction. Wistar rats were subjected to an aerobic exercise protocol on the treadmill or kept sedentary for 8 weeks. Additionally, animals were treated with DEX or saline (50μg/kg, s.c. for 14 days). Groups were: sedentary control (SC), DEX sedentary (SD), trained control (TC) and trained DEX (TD). Body weight (BW) and arterial pressure (AP) were analyzed. After euthanasia, adrenal gland, myocardium, SOL and TA muscles were weighted and normalized by tibia. The cross-sectional area (CSA), capillary:fiber ratio (C:F ratio), capillary density (CD) and protein levels were evaluated in SOL and TA. Treatment with DEX caused reduction in BW and in muscle weight (MW) in TA. DEX treatment also determinated decrease in CSA (TA). Further, C/F and CD were also reduced (-41 and -43%, SOL) and (-30 and 68.6%, TA). Training was able to prevent C:F ratio and CD reduction (72.7 and 81.0%) and (32.9 and 54.2%) induced by DEX- treatment. Furthermore, DEX significantly reduced protein levels in SOL and TA muscles VEGFR-2 (-14.6% and -20.1%), VEGF (-15.6 and -19%), Bcl-2 (-18.4 and20.5%), Bcl-2/Bax ratio (-29.0 and -13.7%) and p-Bax/Bax (-25.4 and -20%), beyond COX-2 in TA (-22.8%). DEX also promoted increase in caspase-3 cleaved (25 and 24.1%, SOL and TA). Moreover, training was able to prevent reduction in proteins levels in DEX-treated groups in SOL and TA: VEGFR-2 (14.7 and 25.2%), VEGF (15.3 and 25.3%), Bcl-2 (21.6 and 35.5%), Bcl-2/Bax ratio (26.1 and 19.9%), p-Bax/Bax (23.7 and 32.1%) and COX-2 (31.5%) and the increase in caspase-3 cleaved (16.0 and 17.8%). In conclusion, these results showed that DEX-induced rarefaction promoted imbalance between apoptotic and angiogenic factors, become one possible causes of hypertension. However, also showed that aerobic training is a good strategy to attenuate DEX-induced rarefaction and this response may involve a better balance between apoptotic and angiogenic factors, which contribute for the attenuation of hypertension. A dexametasona (DEX) é amplamente utilizada em vários casos clínicos devido a sua eficácia como fármaco anti-inflamatório. Por outro lado, a utilização crônica deste medicamento pode causar alterações metabólicas e hemodinâmicas que contribuem para o desenvolvimento da hipertensão. A hipertensão arterial pode ser determinada pelo aumento no débito cardíaco ou da resistência periférica e a rarefação pode estar associada a esta resposta. A manutenção da microcirculação é dependente do equilíbrio entre proteínas anti e apoptóticas e o fator de crescimento endotelial vascular (VEGF) contribui para a manutenção deste equilíbrio. Demonstramos recentemente que altas doses de DEX reduz a produção do VEGF, no entanto, os mecanismos envolvidos nesta resposta não foram avaliados. Por outro lado, tem sido mostrado que o treinamento físico (TF) aeróbio é uma ferramenta importante na prevenção da rarefação e/ou promoção angiogênese, no tratamento da hipertensão, no entanto, quase nada se sabe sobre os efeitos do TF na microcirculação e hipertensão induzida pela DEX. Portanto, o objetivo deste estudo foi investigar se o tratamento crônico com DEX compromete a densidade de vasos por alterar o balanço angiogênico/apoptótico na musculatura esquelética e se o pré-condicionamento físico aeróbio atenua esta resposta. Para isso, 60 ratos wistar foram submetidos a um protocolo de TF aeróbio na esteira ou mantidos sedentários por 8 semanas. Além disso, os animais foram tratados com DEX ou salina (50μg/kg, s.c. por 14 dias). Os grupos foram separados em: sedentário controle (SC), sedentário e tratado com DEX (SD), treinado controle (TC) e treinado tratado com DEX (TD). O peso corporal (PC) e a pressão arterial (PA) foram analisados. Após a eutanásia, a glândula adrenal, miocárdio e os músculos SOL e TA foram pesados e normalizados pela tíbia. Foram avaliados também, área de secção transversa (AST), razão capilar/fibra (C/F), densidade capilar (DC) e análise da produção proteica dos músculos SOL e TA. O tratamento com DEX causou redução tanto no PC quanto no peso muscular do TA, assim como da AST. Além disso, a DEX reduziu significativamente C/F e DC nos músculos (-41 e -43%, SOL) e (- 30 e 68,6%, TA). Por outro lado, o TF aeróbio preveniu a redução da C/F e DC causado pela DEX (72,7 e 81,0%, SOL) e (32,9 e 54,2%, TA). Já, os níveis proteicos no SOL e TA foram reduzidos significativamente pela DEX: VEGFR-2 (-14,6% e -20,1%), VEGF (-15,6 e -19%), Bcl-2 (-18,4 e -20,5%), razão Bcl-2/Bax (-29,0 e -13,7%) e p-Bax/Bax (- 25,4 e -20%), além da COX-2 no TA (-22,8%). A caspase-3 clivada estava aumentada (16,0 e 17,8%, SOL e TA respectivamente). Em contrapartida, o TF aeróbio foi capaz de prevenir a redução dos níveis proteicos causado pela DEX nos músculos SOL e TA, além de prevenir o aumento da caspase-3 clivada. Em conclusão, os resultados deste presente estudo mostrou que a rarefação induzida DEX ocorreu por promover o desbalanço entre fatores angiogênicos e apoptóticos,sendo esse um dos possíveis mecanismos da hipertensão, no entanto, demonstrou que o TF aeróbio é uma boa estratégia para manter o balanço entre fatores angiogênicos e apoptóticos, o qual contribui para a manutenção da microcirculação na musculatura esquelética e, portanto, pode contribuir para a atenuação da hipertensão induzida pela DEX.

A doença periodontal corresponde a um grupo de doenças inflamatórias que resultam na destruição das estruturas de suporte dental. São doenças infecciosas e possuem etiologia relacionada a microrganismos gram-negativos podendo manifestar-se de inúmeras maneiras. Estes possuem uma variedade de fatores que permitem o aumento de sua virulência e capacidade de se multiplicarem e persistirem no periodonto. Experimentos recentes de nosso laboratório mostraram que no tecido gengival de rato existe a expressão de RNAm para todos os componentes do Sistema Renina-Angiotensina (SRA), presença da renina e atividade da Enzima Conversora de Angiotensina I (ECA) em tecido gengival de ratos, sugerindo, assim, possível correlação entre o SRA e a doença periodontal (DP). Portanto, o objetivo do presente trabalho foi investigar se o captopril, um inibidor da ECA, altera a progressão da DP induzida experimentalmente em ratos. Para tanto, foi utilizado o modelo de indução da DP por colocação de ligadura ao redor do primeiro molar inferior de ratos divididos em grupos com 10 animais cada, que foram tratados com captopril (via gavagem, 30 mg/kg/dia) ou água (veículo). Foi realizado pré-tratamento com esta droga por 7 ou 14 dias previamente à indução da DP e após este período, o captopril foi administrado por 14 e 21 dias. Além disso, foi realizada cirurgia fictícia para indução da DP (grupo SHAM). As técnicas utilizadas neste trabalho foram: indução da DP em ratos, extração de RNA total, transcrição reversa seguida de reação em cadeia da polimerase quantitativa (RT-qPCR) e análise de perda óssea alveolar. Os dados foram analisados por meio de gráficos. Todos os resultados foram submetidos à análise unidirecional de variância (ANOVA) e representaram médias e respectivos desvios-padrão. Diferenças entre os grupos foram consideradas estatisticamente significativas quando p < 0,05. Com base nos resultados obtidos neste trabalho, foi possível concluir que o captopril não foi capaz de diminuir a perda óssea na doença periodontal induzida experimentalmente em ratos, apesar desta droga ter alterado a expressão de RNAm para um alvo do RAS (AT1a) e alguns mediadores do processo inflamatório no tecido periodontal, tais como COX-2, ECA-2, IL-6, RANKL, VEGF-R1 e VEGF-R2. Periodontal disease (PD) consists of a group of inflammatory diseases which result in the destruction of tooth supporting structures. They are of infectious nature, with etiological factors related to gram-negative microorganisms, and may have manifestations in several ways. These comprise a variety of factors that allow the increase in its virulence and ability to multiply and persist in the periodontal tissue. Recent findings form our laboratory showed that mRNA expression exists in rat gingival tissue for all components of the Renin-Angiotensin System (RAS), the presence of renin as well as Angiotensin Converting Enzyme I (ACE) activity in rat gingival tissue, thus suggesting a possible correlation between the RAS and periodontal disease. Therefore, the aim of this study was to investigate whether captopril, an ACE inhibitor, alters the progression of experimentally-induced PD in rats. Thus, the model of PD induction by ligature placement around rat lower first molar was used. Animals were divided groups of 10 animals each, which were treated with captopril (via gavage, 30 mg/kg/day) or water (vehicle). Pre-treatment with this drug during 7 or 14 days was performed previously to PD induction, and after this period captopril was administered during 14 or 21 days.
