Abstract

Abstract Background Sesquiterpene lactones (SL) are plant secondary metabolites that are known for their anti-fungal, anti-bacterial, anti-inflammatory, and anti-tumor properties. Considering that several SL-derived drugs are currently in cancer clinical trials, we have tested two SL molecules, 3-β-methoxy-iso-seco-tanapartholide (β-tan) isolated from Achillea falcata and salograviolide A (Sal A) isolated from Centaurea ainetensis, for their anti-tumor properties. We used the mouse epidermal JB6P + cells as a model for tumor promotion and cellular transformation. Key players that are involved in cellular transformation and tumorigenesis are the AP-1 and NF-κB transcription factors; therefore, we assessed how β-tan and Sal A modulate their signaling pathways in JB6P + cells. Methods The effects of β-tan and Sal A on the growth of normal and neoplastic keratinocytes and on the tumor promotion-responsive JB6P + cells were determined using the MTT assay. Anchorage-independent cell growth transformation assays were used to evaluate the anti-tumor promoting properties of these SL molecules in JB6P + cells and dual luciferase reporter assays and western blot analysis were used to investigate their effects on tumor promoter-induced AP-1 and NF-κB activities and protein levels of key AP-1 and NF-кB target genes. Results β-tan and Sal A selectively inhibited tumor promoter-induced cell growth and transformation of JB6P + cells at concentrations that do not affect JB6P + and primary keratinocytes basal cell growth. In addition, both molecules reduced basal and tumor promoter-induced NF-κB transcriptional activities, differentially regulated basal and tumor promoter-induced AP-1 transcriptional activities, and modulated key players of the AP-1 and NF-κB signaling pathways. Conclusions These results highlight the anti-tumor promoting properties of β-tan and Sal A. These SL molecules isolated from two plant species native to the Middle East may provide opportunities for complementary medicine practices.

Abstract

Abstract Background Medicinal plants represent alternative means for the treatment of several chronic diseases, including inflammation. The genus Ranunculus, a representative of the Ranunculaceae family, has been reported to possess anti-inflammatory, analgesic, antiviral, antibacterial, antiparasitic and antifungal activities, possibly due to the presence of anemonin and other. Different studies have shown the occurrence of unusual fatty acids (FAs) in Ranunculaceae; however, their therapeutic role has not been investigated. The purpose of this study is to characterize potential anti-inflammatory bioactivities in Ranunculus constantinopolitanus D'Urv., traditionally used in Eastern Mediterranean folk medicine. Methods The aerial part of R. constantinopolitanus was subjected to methanol (MeOH) extraction and solvent fractionation. The bioactive fraction (I.2) was further fractionated using column chromatography, and the biologically active subfraction (Y2+3) was identified using infrared (IR) spectroscopy, nuclear magnetic resonance (NMR) and gas chromatography-mass spectrometry (GC-MS). The effects of I.2 and Y2+3 on cell viability were studied in mouse mammary epithelial SCp2 cells using trypan blue exclusion method. To study the anti-inflammatory activities of I.2 and Y2+3, their ability to reduce interleukin (IL)-6 levels was assessed in endotoxin (ET)-stimulated SCp2 cells using enzyme-linked immunosorbent assay (ELISA). In addition, the ability of Y2+3 to reduce cyclooxygenase (COX)-2 expression was studied in IL-1-treated mouse intestinal epithelial Mode-K cells via western blotting. Data were analyzed by one-way analysis of variance (ANOVA), Student-Newman-Keuls (SNK), Tukey HSD, two-sample t-test and Dunnett t-tests for multiple comparisons. Results The chloroform fraction (I.2) derived from crude MeOH extract of the plant, in addition to Y2+3, a FA mix isolated from this fraction and containing palmitic acid, C18:2 and C18:1 isomers and stearic acid (1:5:8:1 ratio), reduced ET-induced IL-6 levels in SCp2 cells without affecting cell viability or morphology. When compared to fish oil, conjugated linoleic acid (CLA) and to individual FAs as palmitic, linoleic, oleic and stearic acid or to a mix of these FAs (1:5:8:1 ratio), Y2+3 exhibited higher potency in reducing ET-induced IL-6 levels within a shorter period of time. Y2+3 also reduced COX-2 expression in IL-1-treated Mode-K cells. Conclusion Our studies demonstrate the existence of potential anti-inflammatory bioactivities in R. constantinopolitanus and attribute them to a FA mix in this plant.

Abstract

Abstract Background Anti-inflammatory activities of medicinal plants have largely been attributed to their content of sesquiterpene lactones (SLs). SLs are predominantly found in the sunflower family Asteraceae and have been isolated from many plants of this family, particularly Centaurea. The anti-inflammatory activities of extract of Centaurea ainetensis, a Lebanese endemic plant, and the isolated active molecule were assessed for their potential ant-inflammatory activities. Methods Plant extract from Centaurea ainetensis, and the isolated active ingredient Salograviolide A (SA), a sesquiterpene lactones guaianolide, were used for the study. Western blotting and electrophoretic mobility shift assays were used to test the effects of the plant extract and SA on interleukin-1 (IL-1) induced increase in cyclooxygenase-2 (COX-2) levels and in nuclear factor-κB (NF-κB) translocation in an intestinal epithelial cell (IEC) of inflammation. Their effects on inflammation score and cytokine levels were also studied in an iodoacetoamide-induced rat model of inflammation. Results Plant extract and SA were shown to reverse the effects observed by IL-1 on COX-2 levels and NF-κB translocation in IEC. SA decreased the level of inflammatory cytokines and the level of inflammation in the animal model. Conclusion These findings suggest that SA may be useful in the development of natural therapies for inflammatory diseases.

86. Clouds and 'throat hit': Effects of liquid composition on nicotine emissions and physical characteristics of electronic cigarette aerosols

Abstract

Electronic cigarettes (ECIGs) heat and vaporize a liquid mixture to produce an inhalable aerosol that can deliver nicotine to the user. The liquid mixture is typically composed of propylene glycol (PG) and vegetable glycerin (VG), in which are dissolved trace quantities of flavorants and, usually, nicotine. Due to their different chemical and thermodynamic properties, the proportions of PG and VG in the liquid solution may affect nicotine delivery and user sensory experience. In social media and popular culture, greater PG fraction is associated with greater “throat-hit,” a sensation that has been attributed in cigarette smokers to increased presence of vapor-phase nicotine. VG, on the other hand, is associated with thicker and larger exhaled “clouds.” In this study, we aim to investigate how PG/VG ratio influences variables that relate to nicotine delivery and plume visibility. Aerosols from varying PG/VG liquids were generated using a digitally controlled vaping instrument and a commercially available ECIG, and analyzed for nicotine content by GC-MS. Particle mass and number distribution were determined using a six-stage cascade impactor and a fast particle spectrometer (TSI EEPS), with tightly controlled dilution and sampling biases. A Mie theory model was used to compute the aerosol scattering coefficients in the visible spectrum. Decreasing the PG/VG ratio resulted in a decrease in total particulate matter (TPM) and nicotine yield (R2 > 0.9, p < .0001). Measured particle count median diameter ranged between 44 and 97nm, and was significantly smaller for PG liquids. Although the particle mass concentration was lower, aerosols produced using liquids that contained VG had an order of magnitude greater light scattering coefficients. These findings indicate that PG/VG ratio is a strong determinant of both nicotine delivery and user sensory experience. Copyright © 2017 American Association for Aerosol Research

Abstract

The lack of scientific evidence on the constituents, properties, and health effects of second-hand waterpipe smoke has fueled controversy over whether public smoking bans should include the waterpipe. The purpose of this study was to investigate and compare emissions of ultrafine particles (UFP, <100 nm), carcinogenic polyaromatic hydrocarbons (PAH), volatile aldehydes, and carbon monoxide (CO) for cigarettes and narghile (shisha, hookah) waterpipes. These smoke constituents are associated with a variety of cancers, and heart and pulmonary diseases, and span the volatility range found in tobacco smoke. Sidestream cigarette and waterpipe smoke was captured and aged in a 1 m3 Teflon-coated chamber operating at 1.5 air changes per hour (ACH). The chamber was characterized for particle mass and number surface deposition rates. UFP and CO concentrations were measured online using a fast particle spectrometer (TSI 3090 Engine Exhaust Particle Sizer), and an indoor air quality monitor. Particulate PAH and gaseous volatile aldehydes were captured on glass fiber filters and DNPH-coated SPE cartridges, respectively, and analyzed off-line using GC–MS and HPLC–MS. PAH compounds quantified were the 5- and 6-ring compounds of the EPA priority list. Measured aldehydes consisted of formaldehyde, acetaldehyde, acrolein, methacrolein, and propionaldehyde. We found that a single waterpipe use session emits in the sidestream smoke approximately four times the carcinogenic PAH, four times the volatile aldehydes, and 30 times the CO of a single cigarette. Accounting for exhaled mainstream smoke, and given a habitual smoker smoking rate of 2 cigarettes per hour, during a typical one-hour waterpipe use session a waterpipe smoker likely generates ambient carcinogens and toxicants equivalent to 2–10 cigarette smokers, depending on the compound in question. Abstract

The impact of the form of nicotine, i.e. free base or salt, on its delivery from tobacco products and electronic nicotine delivery systems (ENDS) has been debated by scientists and regulators. In this paper, we briefly discussed the various ways of modifying the ratio of nicotine forms in tobacco products and ENDS. We focus on partitioning of nicotine forms in ENDS liquids, especially in the recently introduced pod-based ENDS. We discuss the influence of various parameters on nicotine delivery from ENDS, such as the form of nicotine, counter-anions in nicotine salts, the power output of devices and user puffing topography. Recommendations are made on means to avoid capping nicotine concentrations in ENDS liquids as the sole measure for regulating nicotine delivery from ENDS and on adoption of "nicotine flux" as a regulatory tool that accounts for all the parameters that affect nicotine delivery from ENDS. We highlight the importance of including the form of nicotine in constructing a nicotine flux model and of minimizing possible customization of ENDS by users. While research is still necessary on methods for testing nicotine forms in ENDS liquids and aerosols and on absorption of the different forms by the body in the presence or absence of flavours, it is recommended that WHO urge countries to include nicotine flux and form in ENDS regulation to better inform users about nicotine delivery from their devices.
