Botulinum toxin (BoNT) has been known for over 50 years. It has conquered many areas of medicine and has become indispensable in contemporary medicine. Now, BoNT is used to treat at least 26 conditions in six medical specialties. Although the use of BoNT began in strabology, it became the gold standard for many ophthalmologic pathologies. The present review of the literature focuses on the use of BoNT in ophthalmology and treatment of the following conditions: blepharospasm, facial hemispasm, facial palsy, spastic entropion, strabismus, endocrine orbitopathy, convergence spasm, and facial trauma. We conclude that nearly half a century of experience in utilizing BoNT in ophthalmology ensured a satisfactory level of effectiveness and safety for patients with many pathologies. Areas of future research include the application of BoNT in new selected indications, the development of the route of application without injections, and the development of long-acting BoNT forms for patients who require repeated long-term treatment.

53. Effective Use of Textbooks by Early Education Teachers

Introduction: Schoolbooks are the basic didactic tool in early childhood education classes. As evidenced by numerous research reports, their content is significantly diversified in terms of quality. However, empirical research providing insight into teachers’ experiences of working with school textbooks is lacking. Research Aim: This study examines how teachers influence the use of a school textbook. It looks at their views on daily work with textbooks, the advantages and disadvantages they perceive, and their responses to these observations. Method: Six focus group interviews with 37 early childhood education teachers were conducted. The collected material was subsequently subjected to meaning-oriented content analysis. Results: On the basis of the collected data, it was established that teachers construe textbooks as: predestined fate, a stabilizing factor in the didactic process, a tool for everyday work, a subject-matter expert and a surety of their responsibility for the learning process. The work of educators with the school textbook is characterized by conformity, limited flexibility, adaptation to the range of children’s aptitudes, rationalization and, in rare cases, resistance. Conclusions: With respect to working with school textbooks, the agency of teachers manifests itself only as a superficial response to the identified shortcomings, without an autonomous approach to the content as well as tasks and instructions in school textbooks. Such a conclusion may offer grounds for a pedagogical discussion on the limitations and needs of training early education teachers in terms of enhancing their agency while working with a school textbook.

54. Subjectively salient faces differ from emotional faces: ERP evidence

Abstract The self-face is processed differently than emotional faces. A question arises whether other highly familiar and subjectively significant non-self faces (e.g. partner’s face) are also differentiated from emotional faces. The aim of this event-related potential (ERP) study was to investigate the neural correlates of personally-relevant faces (the self and a close-other’s) as well as emotionally positive (happy) and neutral faces. Participants were tasked with the simple detection of faces. Amplitudes of N170 were more negative in the right than in the left hemisphere and were not modulated by type of face. A similar pattern of N2 and P3 results for the self-face and close-other’s face was observed: they were associated with decreased N2 and increased P3 relative to happy and neutral faces. However, the self-face was preferentially processed also when compared to a close-other’s face as revealed by lower N2 and higher P3 amplitudes. Nonparametric cluster-based permutation tests showed an analogous pattern of results: significant clusters for the self-face compared with all other faces (close-other’s, happy, neutral) and for close-other’s face compared to happy and neutral faces. In summary, the self-face prioritization was observed, as indicated by significant differences between one’s own face and all other faces. Crucially, both types of personally-relevant faces differed from happy faces. These findings point to the pivotal role of subjective evaluation of the saliency factor.

55. The use of blackcurrant pomace and erythritol to optimise the functional properties of shortbread cookies

Abstract As a result of the production of blackcurrant juice, pomace is produced, which is a cheap, easy to further process raw material with high health benefits. The aim of the research was to develop a recipe for shortbread cookies based on blackcurrant pomace (0, 10, 30, 50%) and erythritol, and to assess their nutritional value (content of proteins, fats, sugars, dietary fibre, selected minerals and energy value), pro-health properties (antioxidant and anti-diabetic capacity) and sensory evaluation. The energy value of products with 50% of pomace sweetened with erythritol was nearly 30% lower compared to traditional cookies, while the content of dietary fibre was 10 times higher in products with the highest percentage of pomace. The antioxidant capacity and the total content of polyphenolic compounds increased with the increase in pomace content. The ability to inhibit α-amylase by shortbread cookies without pomace was about 400 times lower than those with 50% pomace. The results of the sensory evaluation showed that erythritol-sweetened cookies have more desirable characteristics compared to sucrose-sweetened cookies. Finally, it was proved that the proposed products are an excellent proposal for people struggling with food-dependent diseases, as well as being an opportunity to manage waste from the fruit industry.

ABSTRACTThe uptake of human papillomavirus (HPV) vaccine remains suboptimal despite being a part of routine vaccination within national immunization program(s). This indicates probable challenges with the implementation of HPV immunization program(s) in various countries. The objective of this systematic literature review (SLR) was to identify implementation strategies for HPV vaccination within national and regional immunization programs worldwide with an aim to provide guidance for countries targeting to increase their HPV vaccine coverage rate (VCR). A comprehensive literature search was conducted across Medline and Embase and included articles published between January 2012 and January 2022. Of the 2,549 articles retrieved, 168 met inclusion criteria and were included in the review. Strategies shown to improve HPV vaccination uptake in the reviewed literature include campaigns to increase community awareness and knowledge of HPV, health care provider trainings, integrating HPV vaccination within school settings, coordinated efforts via multi-sectoral partnerships, and vaccination reminder and recall systems. Findings may help national authorities understand key considerations for HPV vaccination when designing and implementing programs aiming to increase HPV VCR in adolescents.

Numerous strategies have been proposed to minimize obesity-associated health effects, among which phytocannabinoids appear to be effective and safe compounds. In particular, cannabigerol (CBG) emerges as a potent modulator of the composition of membrane phospholipids (PLs), which plays a critical role in the development of insulin resistance. Therefore, here we consider the role of CBG treatment on the composition of PLs fraction with particular emphasis on phospholipid subclasses (e.g., phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylserine (PS), and phosphatidylinositol (PI)) in the red gastrocnemius muscle of Wistar rats fed the standard or high-fat, high-sucrose (HFHS) diet. The intramuscular PLs content was determined by gas-liquid chromatography and based on the composition of individual FAs, we assessed the stearoyl-CoA desaturase 1 (SCD1) index as well as the activity of n-3 and n-6 polyunsaturated fatty acids (PUFAs) pathways. Expression of various proteins engaged in the inflammatory pathway, FAs elongation, and desaturation processes was measured using Western blotting. Our research has demonstrated the important association of obesity with alterations in the composition of muscular PLs, which was significantly improved by CBG supplementation, enriching the lipid pools in n-3 PUFAs and decreasing the content of arachidonic acid (AA), which in turn influenced the activity of PUFAs pathways in various PLs subclasses. CBG also inhibited the local inflammation development and profoundly reduced the SCD1 activity. Collectively, restoring the PLs homeostasis of the myocyte membrane by CBG indicates its new potential medical application in the treatment of obesity-related metabolic disorders.

Background: Our study explores the role of bats as reservoirs of coronaviruses. Methods: We conducted virological screening of bats hibernating in military bunkers at the Natura 2000 site “Nietoperek” in Western Poland collecting oral and anal swab samples from 138 bats across six species to apply a combination of pan-coronavirus and SARS-CoV-2 specific PCR assays. Results: Only one anal swab tested positive for coronavirus. No SARS-CoV-2 was detected in any of the samples. The low prevalence of coronavirus in the studied colony contrasts with higher rates found in other regions and may be influenced by hibernation. Conclusions: Hibernating bats may show a low prevalence of coronavirus, potentially due to the hibernation process itself. This finding indicates that hibernating bats may not be the most optimal subjects for screening zoonotic pathogens. However, biomonitoring of bats for emerging and reemerging diseases is recommended for comprehensive epidemiological insights.

According to the newest trends in food waste management, the priority for valorisation of oil and dairy industry by-products was recognised. New approach of blending side streams of plant and animal origin has been proposed for obtaining future foods with improved nutritional value. Major obstacle in valorising sunflower seed cake (SSC) and whey (W) is their perishability and a relatively high content of phenolics contributing to, among others, protein indigestibility. In the study, a new approach of microwave pre-treatment applied for SSC and W blends was used. Its influence on phenolic acids and antioxidant properties after fermentation with co-culture of lactic acid bacteria and yeast was examined. Microwave drying of blends was longer than that of SSC affecting phenolic acids content. Fermentation by L. lactis and K. lactis caused pH decrease and increase in total acidity in 48 h effectively limiting the growth of bacterial contaminants. Fermentation decreased the content of phenolic acids, especially in blend composed of SSC and W (8.75:1; w/w) with the highest content of these constituents (from 1611.3 down to 838.8 mg/100 g dry matter). The study showed the synergistic effect of pre-treatment method and fermentation in terms of reduction in phenolic acids in new food matrices.

Celem artykułu jest porównanie językowych obrazów deszczu występujących we frazeologii polskiej i chińskiej, ukazanie ich podobieństw i rozbieżności oraz przedstawienie wartościowania samego elementu deszczu. Praca stanowi również próbę odnalezienia przyczyn różnic w wykreowanych przez Polaków i Chińczyków w językowych interpretacjach rzeczywistości. Do analizy semantyczno-leksykalnej i komparatystycznej wykorzystane zostały założenia oraz narzędzia JOS, dzięki którym możliwe jest niepowierzchowne poznanie i zrozumienie kultury utrwalonej w języku danych narodów. W badaniu wykorzystane zostały dane leksykograficzne wyekscerpowane z polskich i chińskich słowników jednojęzycznych (ogólnych i frazeologicznych) oraz chińsko-angielskich słowników dwujęzycznych. Analiza wykazała między innymi, że obrazy te mają wspólne cechy wynikające z kwestii fizycznych, jak również z komunikatywnej funkcji języka. Rozbieżności są jednak bardziej uwidocznione, a językowe obrazy deszczu specyficzne dla języka polskiego i chińskiego uwydatniają różnice w postrzeganiu rzeczywistości przez użytkowników tych języków.

BackgroundSubstance Use Disorder (SUD) persists as a significant public health challenge worldwide, with an estimated prevalence of approximately 10-15% across the global populace. This condition is characterized by a notably high risk of lapses and relapses, even subsequent to treatment interventions. Mobile health interventions, owing to their widespread accessibility, emerge as a promising approach to diminish the risk of relapse post-treatment and to broaden the scope of care, especially in regions with a scarcity of trained medical professionals.MethodThis study is designed to assess the effectiveness of mobile interventions in mitigating cravings and preventing lapses among individuals diagnosed with SUD. Employing a two-armed, randomized controlled trial framework, the study will evaluate a self-administered psychological intervention delivered through a mobile application, Nałogometr 2.0. Over a period of three months, participants will engage with intervention modules that primarily incorporate mindfulness techniques and Cognitive Behavioral Therapy (CBT) principles. Ecological Momentary Assessment (EMA) will be utilized to gather longitudinal data on a range of variables that are indicative of craving intensity and the risk of lapse. In addition to this, a monthly-administered battery of questionnaires will be employed to gauge the severity of substance dependence, as well as to measure levels of anxiety, depression, and overall life satisfaction.ResultsResults will be submitted for publication in peer-reviewed journals.Clinical trial registrationhttps://clinicaltrials.gov/, identifier [NCT05730504].

Background and Purpose: Radiation-induced lymphopenia (RIL) is a common side effect of radiotherapy (RT) that may negatively impact survival. We aimed to identify RIL predictors in patients with non-small-cell lung cancer (NSCLC) treated intensity modulated radiotherapy (IMRT) and volumetric modulated arc therapy (VMAT). Materials and Methods: We retrospectively analysed data of 306 patients who underwent radical RT for NSCLC. Absolute lymphocyte count (ALC) loss was evaluated for each patient by fitting an exponential decay curve to data from first 45 days since treatment start, and percentage ALC loss relative to baseline was calculated based on area under the decay curve and baseline ALC. We compared IMRT and VMAT treatment plans and used linear regression to predict ALC loss. Results: ALC decreased during RT in the whole patient group, while neutrophil counts remained stable and decreased only in those treated with concurrent chemoradiotherapy (CRT). Percentage ALC loss ranged between 11 and 78 % and was more strongly than lymphocyte nadir correlated with dose-volume metrics for relevant normal structures. We found evidence for the association of high radiation dose to the lungs, heart and body with percentage ALC loss, with lung volume exposed to 20–30 Gy being most important predictors in patients treated with IMRT. A multivariable model based on CRT use, baseline ALC and first principal component (PC1) of the dose-volume predictors showed good predictive performance (bias-corrected R2 of 0.40). Conclusion: Percentage lymphocyte loss is a robust measure of RIL that is predicted by baseline ALC, CRT use and dose-volume parameters to the lungs, heart and body.

In 2017 strategic lawsuits against public participation (SLAPPs) became an important topic on the EU level. As a result, the EU adopted the anti-SLAPP Directive, which shall protect journalists from abusive lawsuits that do not serve justice but only the sinister aim of silencing free press. However, there is important litigation as well. In 2024 the Real Madrid Club de Fútbol vs Le Monde case addressed the problem of exorbitant damages targeting press and introducing a deterrent effect on freedom of speech in transnational cases. From a rule of law and, especially, freedom of the press angle, the case is of paramount importance as it forwards a simple but groundbreaking argument: two of EU law’s most fundamental principles, mutual recognition and freedom of speech, are a strong basis to fight SLAPPs.

Introduction: Mastocytosis is a rare hematologic neoplasm characterized by the abnormal proliferation and accumulation of mast cells in various tissues. Its clinical manifestations vary widely, ranging from cutaneous lesions to systemic involvement with potentially life-threatening symptoms. Understanding the pathophysiology, diagnosis, and management of mastocytosis is crucial for improving patient outcomes. Aim of the Study: The aim of this study was to provide a comprehensive overview of mastocytosis, including its epidemiology, risk factors, pathophysiology, clinical manifestations, diagnostic approaches, and management strategies. By synthesizing current knowledge on mastocytosis, this study aims to enhance understanding of the disease and guide clinical practice. Description of the State of Knowledge: Mastocytosis is classified into cutaneous and systemic forms, with various subtypes based on clinical and histopathological features. Diagnosis relies on a combination of clinical suspicion, serum tryptase levels, histological examination of bone marrow biopsies, and genetic testing. Management strategies include symptomatic treatment, avoidance of triggers, and targeted therapies such as monoclonal antibodies and tyrosine kinase inhibitors. Advanced forms may require cytoreductive therapy or allogeneic hematopoietic stem cell transplantation (alloHSCT). Conclusions: Despite advancements in diagnosis and treatment, achieving lasting remission in mastocytosis remains challenging, especially in advanced cases. Further research into the molecular mechanisms underlying the disease and the development of novel therapeutic modalities are needed to improve patient outcomes and quality of life.

Introduction: Adolescent pregnancy remains a contentious issue despite improvements in quality of life and increased access to sexual education. The average age of first pregnancy has shifted due to financial instability among young people, leading to delayed pregnancies until financial stability is achieved. Teenage mothers face social stigma, and early sexual initiation not only increases the risk of pregnancy but also sexually transmitted infections (STIs). Often, adolescent pregnancies are unwanted, resulting in delayed medical consultation due to fear or ignorance. Aim of the Study: This study aims to investigate the medical and social implications of adolescent pregnancy, emphasizing the importance of early sexual education and comprehensive healthcare support. Description of the State of Knowledge: Adolescent pregnancy, defined as pregnancy between ages 10 and 19, is a global concern with millions of cases annually. It poses significant health risks, including premature birth, low birth weight, and intrauterine growth restriction. Socially, it affects educational attainment and financial stability, with adolescents from lower socioeconomic backgrounds facing greater challenges. Early sexual initiation and risky sexual behaviors further compound the issue, increasing the risk of STIs. Lack of comprehensive sexual education exacerbates these risks, highlighting the need for multidisciplinary support from healthcare professionals. Conclusions: Adolescent pregnancy requires a holistic approach encompassing medical, social, and legal dimensions. Effective sexual education programs, involving gynecologists, pediatricians, and educators, are essential for prevention. Legal frameworks must protect adolescents and address issues of consent and sexual health. Additionally, access to comprehensive healthcare services, including prenatal care and contraception, is crucial for mitigating the adverse effects of adolescent pregnancy on both individuals and society.

Abstract This research examines the biological treatment of undiluted vegetable waste digestate conducted in a bubble column photobioreactor. Initially, the bioreactor containing 3N-BBM medium was inoculated with Microglena sp., Tetradesmus obliquus, and Desmodesmus subspicatus mixture with a density of 1.0 × 104 cells/mL and the consortium was cultivated for 30 days. Then, the bioreactor was semi-continuously fed with liquid digestate with hydraulic retention time (HRT) of 30 days, and the treatment process was continued for the next 15 weeks. The change in the microalgal and cyanobacterial species domination was measured in regular intervals using cell counting with droplet method on a microscope slide. At the end of the experiment, Desmonostoc sp. cyanobacteria (identified with 16S ribosomal RNA genetical analysis) as well as Tetradesmus obliquus green algae along with Rhodanobacteraceae and Planococcaceae bacteria (determined with V3–V4 16sRNA metagenomic studies) dominated the microbial community in the photobioreactor. The experiment demonstrated high treatment efficiency, since nitrogen and soluble COD were removed by 89.3 ± 0.5% and 91.2 ± 1.6%, respectively, whereas for phosphates, 72.8 ± 2.1% removal rate was achieved.

Additive manufacturing is a technology that can be successfully used in pharmacy and medicine. One of the examples of products that can be additively manufactured are microneedle systems. The specificity of these products, which are used for transdermal drug delivery, makes additive manufacturing a perfect choice for related research. However, the dimensions of microneedles usually do not exceed 2 mm, which means that manufacturing them using the most widely available additive manufacturing method, Fused Deposition Modelling (FDM), is problematic. In this study, the authors decided to investigate the possibilities of manufacturing microneedle systems using the FDM method in such a way as to minimize or exclude the need for post-processing. Five types of microneedle geometries were tested in four sizes, examining how changing the values of FDM process parameters would affect the accuracy of reproducing the digital geometry of the microneedles. From the point of view of the application of microneedle systems, it is not only necessary to obtain the designed shape of the microneedles, but also to maintain their appropriate strength. The study presents the results of the bending and compression strength of microneedles made of polylactic acid.

72. Exploring lipophilic antioxidants accumulation in field-grown low temperature-stressed Ephedra monosperma

The seasonal patterns of changes in the content of lipophilic antioxidants β-carotene (β-Car), zeaxanthin (Zx), α-tocopherol (α-Toc), plastoquinone (PQ)/plastoquinol (PQH2) were studied in the assimilating shoots of evergreen shrub Ephedra monosperma J.G. Gmel ex C.A. Mey under natural conditions of Central Yakutia. The shortening of the photoperiod and the seasonal decrease in temperature induced a 1.4-fold increase in α-Toc content. The fall in the average daily temperature from 0.1 to -8.1°C in October led to a decrease in the content of β-Car as a result of the accumulation of rhodoxanthin (Rhd). In this period a sharp increase in the content of Zx retained overnight was also detected. In winter, elevated content of Zx and α-Toc persisted. During September, the content of PQH2 increased by 2.5 times and PQ by 1.4 times (compared to July). The beginning of exposure to freezing average daily temperatures from -3 to -5°C led to the depletion of the total PQ pool by 18%. However, the content of PQtot in the winter months was 1.5 times higher than at the end of July. The results revealed different timing and temperature ranges of variation for individual antioxidants during the development of frost resistance in ephedra.

Sorghum produces allelopathic compounds, including total phenolic compounds and sorgoleone, which exhibit a phytotoxic effect on weeds. The field study, carried out in 2016-2017, was designed as an one-factor experiment, in the randomized block design, in four replications, with Sucrosorgo 506, Rona 1, KWS Freya, KWS Juno, and KWS Sammos, to assess the impact of allelochemicals on weeds. Weed infestation was determined at the beginning of July. Individual weed species were collected from two random places in each plot and weighed. The aim of the laboratory study was to evaluate the total content of phenolic compounds, and sorgoleone in the early stages of plant development (5, 10, and 15 days after emergence) in varieties Rona 1, KWS Freya, KWS Juno, KWS Sammos, Farmsorgo 180, GK Aron, PR 845F, Sucrosorgo 506 and PR849F. The total content of phenolic compounds was determined using the colorimetric method, and the sorgoleone HPLC technique on a Flexar chromatographic set. The highest value of sorgoleone was observed in 15-day-old seedlings of KWS Juno, the lowest in 5-day-old seedlings of Sucrosorgo 506, the highest levels of total phenolic compounds in 5-day-old seedlings of PR 845F, the lowest in 15-day-old seedlings of Farmsorgo 180. The results do not fully confirm the beneficial effect of allelopathic compounds on reducing weed infestation, however, it is important to emphasize the diversity of cultivars used. The statistically insignificant results indicated that most varieties of sorghum plants do not exhibit a significant decrease in yield.

Abstract Introduction Chronic pruritus (CP) is a common symptom defined as a sensation that provokes the desire to scratch and which lasts for at least 6 weeks. CP remains a problem for up to 21.3% of renal transplant recipients (RTRs). Our research aimed to establish the possible association between serum levels of neurotrophin-4 (NT-4) and brain-derived neurotrophic factor (BDNF) and the presence and intensity of CP in RTR. Methods The study was performed on a group of 129 RTRs, who were divided according to the presence or absence of pruritus in the previous 3 days. The assessment of pruritus was performed with the use of a numeric rating scale (NRS), 4-Item Itch Questionnaire (4IIQ), and Itchy Quality of Life (Itchy QoL). A total of 129 blood samples with a volume of 9 ml were drawn from RTRs during the monthly routine control. Serum levels (pg/mL) of NT-4 and BDNF were measured by the ELISA. Results Pruritic RTRs have statistically significantly higher serum concentrations of NT-4 serum level compared to non-pruritic RTRs (229.17 ± 143.86 pg/mL and 153.08 ± 78.19 pg/mL [p = 0.024], respectively). Background: Alveolar bone augmentation is a complex process influenced by a multitude of factors. The materials applied in augmentation procedures must be confirmed as non-toxic, and their physicochemical properties should allow proper bone reconstruction. The specifics of oral surgical procedures require the use of regenerative biomaterials with antimicrobial properties. This study focuses on the physicochemical characteristics of chosen nanosized biomaterials, as well as their cytotoxicity and antimicrobial properties. Methods: nanosized hydroxyapatite and fluorapatite (abbreviated as nHAp and nFAp) pellets were manufactured using a microwave hydrothermal synthesis method. The impact on Candida albicans, Streptococcus mutans, and Lactobacillus rhamnosus strains activity and adherence to apatites was tested. Cytotoxic evaluation was performed based on the differentiation process of MC3T3 cells. The effectiveness of MC3T3 differentiation was confirmed by Alizarin Red staining. Results: Contact with both biomaterials caused a reduction in the mean microbial count of S. mutans and C. albicans strains, as observed. Studied biomaterials demonstrated enhanced proliferation of MC3T3 cells, with the exception of the 1:1 nFAp concentration. Conclusions: Both biomaterials enhance the proliferation of fibroblasts and limit the activity of specific oral pathogens in vitro. The research clearly demonstrates the advantage of nFAp over nHAp, with a notable reduction in microbial count of Candida albicans and Streptococcus mutans over time. The lowest microbial count reduction was observed in the case of L. rhamnosus. Further research is required in order to fully understand the specifics of nHAp and nFAp antimicrobial action. However, the results were found to be more favourable for nFAp biomaterial.

Calendula officinalis L. has numerous health-promoting properties due to the presence of a large number of lipophilic compounds. Their effective delivery to the body requires the use of an appropriate technique such as emulsification. So, the main purpose of this study was to understand how the profile of lipophilic compounds from pot marigold and the pro-health potential are shaped by different types of protein, oil, and drying techniques in o/w nanoemulsion. To obtain this, the profiles of carotenoid compounds and tocols were measured. Additionally, antioxidant potential and the ability to inhibit α-amylase and α-glucosidase were measured. Pea protein emulsion exhibited a higher final content of carotenoid compounds (23.72–39.74 mg/100 g), whereas those with whey protein had stronger α-amylase inhibition (487.70 mg/mL). The predominant compounds in the studied nanoemulsions were β-carotene (between 19% and 40%), followed by α-tocopherol/γ-tocopherol. The type of proteins shaped the health-promoting properties and determined the content of health-promoting compounds.

79. Black and Red Currant Pomaces as Raw Materials to Create Smoothies with In Vitro Health-Promoting Potential

Pomace is a by-product resulting from the pressing of fruits and vegetables into juices, and it is typically treated as waste. Interestingly, pomace contains minimal amounts of protein and fat but is characterized by its high polyphenol and dietary fiber contents, which may have health benefits for human physiology. Therefore, they are a potentially attractive raw material for the food industry, but to our knowledge, no smoothies with their addition have been prepared and described so far. Consequently, products derived from apple juice, incorporating different doses of fresh (6% and 12%) and dried (3% and 6%) black or red currant pomace, were formulated, and their physical properties, chemical composition, bioactive compound content, and health-promoting potential (in vitro antioxidant and antidiabetic activity) were evaluated. Additionally, the products underwent sensory assessment by consumers. The fortified beverages exhibited different physical characteristics and chemical compositions than apple juice. All smoothies were characterized by higher concentrations of anthocyanins, flavonols, and procyanidin polymers compared to the base product. Moreover, 75% of them exhibited a significantly elevated phenolic acid content as well as a higher concentration of flavan-3-ols. The majority of fresh smoothies exhibited significantly higher in vitro antioxidant capacities and increased in vitro α-amylase and α-glucosidase inhibitory effects compared to the base product. The highest ABTS activity was recorded in the variant with 6% dried black currant pomace. In turn, the smoothie with 3% dried red currant pomace had the most effective FRAP effect and, together with the product containing 12% fresh black currant pomace, ORAC antioxidant activity and α-glucosidase inhibition also. The introduction of 6% dried red currant pomace led to the creation of a beverage that most effectively inhibited α-glucosidase. The study showed that the application of various types of pomace, mainly that of black currant, into apple juice enables the development of new functional products with sensory attributes that are favorably evaluated by consumers.

80. Influence of Microwave-Assisted Chemical Thermohydrolysis of Lignocellulosic Waste Biomass on Anaerobic Digestion Efficiency

To date, microwave radiation has been successfully used to support the chemical hydrolysis of organic substrates in the laboratory. There is a lack of studies on large-scale plants that would provide the basis for a reliable evaluation of this technology. The aim of the research was to determine the effectiveness of using microwave radiation to support the acidic and alkaline thermohydrolysis of lignocellulosic biomass prior to anaerobic digestion on a semi-industrial scale. Regardless of the pretreatment options, similar concentrations of dissolved organic compounds were observed, ranging from 99.0 ± 2.5 g/L to 115.0 ± 3.0 in the case of COD and from 33.9 ± 0.92 g/L to 38.2 ± 1.41 g/L for TOC. However, these values were more than twice as high as the values for the substrate without pretreatment. The degree of solubilisation was similar and ranged between 20 and 28% for both monitored indicators. The highest anaerobic digestion effects, ranging from 99 to 102 LCH4/kgFM, were achieved using a combined process consisting of 20 min of microwave heating, 0.10–0.20 g HCl/gTS dose, and alkaline thermohydrolysis. For the control sample, the value was only 78 LCH4/kgFM; for the other variants, it was between 79 and 94 LCH4/kgFM. The highest net energy gain of 3.51 kWh was achieved in the combined alkaline thermohydrolysis with NaOH doses between 0.10 and 0.20 g/gTS. The use of a prototype at the 5th technology readiness level made it possible to demonstrate that the strong technological effects of the thermohydrolysis process, as demonstrated in laboratory tests to date, do not allow for positive energy balance in most cases. This fact considerably limits the practical application of this type of solution.

Research to date has mainly focused on the properties and efficiency of the production of selected, individual types of biofuels from microalgae biomass. There are not enough studies investigating the efficiency of the production of all energy sources synthesised by these microorganisms in a single technological cycle. The aim of this research was to determine the possibilities and efficiency of the production of hydrogen, bio-oil, and methane in the continuous cycle of processing T. subcordiformis microalgae biomass. This study showed it was feasible to produce these three energy carriers, but the production protocol adopted was not necessarily valuable from the energy gain standpoint. The production of bio-oil was found to be the least viable process, as bio-oil energy value was only 1.3 kWh/MgTS. The most valuable single process for microalgae biomass conversion turned out to be methane fermentation. The highest specific gross energy gain was found after applying a protocol combining biomass production, hydrogen biosynthesis, and subsequent methane production from T. subcordiformis biomass, which yielded a total value of 1891.4 kWh/MgTS. The direct methane fermentation of T. subcordiformis biomass enabled energy production at 1769.8 kWh/MgTS.

The study investigated the impact of Lonicera caerulea L. juice matrix modification and drying techniques on powder characteristics. The evaluation encompassed phenolics (514.7–4388.7 mg/100 g dry matter), iridoids (up to 337.5 mg/100 g dry matter), antioxidant and antiglycation capacity, as well as anti-ageing properties of powders produced using maltodextrin, inulin, trehalose, and palatinose with a pioneering role as a carrier. Spray drying proved to be competitive with freeze drying for powder quality. Carrier application influenced the fruit powder properties. Trehalose protected the phenolics in the juice extract products, whereas maltodextrin showed protective effect in the juice powders. The concentrations of iridoids were influenced by the matrix type and drying technique. Antiglycation capacity was more affected by the carrier type in juice powders than in extract products. However, with carrier addition, the latter showed approximately 12-fold higher selectivity for acetylcholinesterase than other samples. Understanding the interplay between matrix composition, drying techniques, and powder properties provides insights for the development of plant-based products with tailored attributes, including potential health-linked properties.

Backgrounds: The titanium–aluminum–vanadium alloy (Ti-6Al-4V) is frequently used in implantology due to its biocompatibility. The use of 3D printing enables the mechanical modification of implant structures and the adaptation of their shape to the specific needs of individual patients. Methods: The titanium alloy plates were designed using the 3D CAD method and printed using a 3D SLM printer. Qualitative tests were performed on the material surface using a microcomputed tomography scanner. The cytotoxicity of the modular titanium plates was investigated using the MTT assay on the L929 cell line and in direct contact with Balb/3T3 cells. Cell adhesion to the material surface was evaluated with hFOB1.19 human osteoblasts. Microbial biofilm formation was investigated on strains of Lactobacillus rhamnosus, Staphylococcus epidermidis, Streptococcus mutans and Candida albicans using the TTC test and scanning electron microscopy (SEM). Results: The surface analysis showed the hydrophobic nature of the implant. The study showed that the titanium plates had no cytotoxic properties. In addition, the material surface showed favorable properties for osteoblast adhesion. Among the microorganisms tested, the strains of S. mutans and S. epidermidis showed the highest adhesion capacity to the plate surface, while the fungus C. albicans showed the lowest adhesion capacity. Conclusions: The manufactured modular plates have properties that are advantageous for the implantation and reduction in selected forms of microbial biofilm. Three-dimensional-printed modular titanium plates were investigated in this study and revealed the potential clinical application of this type of materials, regarding lack of cytotoxicity, high adhesion properties for osteoblasts and reduction in biofilm formation. The 3D CAD method allows us to personalise the shape of implants for individual patients.

CRNDE is an oncogene expressed as a long non-coding RNA. However, our team previously reported that the CRNDE gene also encodes a micropeptide, CRNDEP. The amino acid sequence of CRNDEP has recently been revealed by other researchers, too. This study aimed to investigate genetic alterations within the CRNDEP-coding region of the CRNDE gene, methylation profiling of this gene, and CRNDEP expression analysis. All investigations were performed on clinical material from patients with ovarian tumors of diverse aggressiveness. We found that CRNDEP levels were significantly elevated in highly aggressive tumors compared to benign neoplasms. Consistently, a high level of this micropeptide was a negative, independent, prognostic, and predictive factor in high-grade ovarian cancer (hgOvCa) patients. The cancer-promoting role of CRNDE(P), shown in our recent study, was also supported by genetic and epigenetic results obtained herein, revealing no CRNDEP-disrupting mutations in any clinical sample. Moreover, in borderline ovarian tumors (BOTS), but not in ovarian cancers, the presence of a single nucleotide polymorphism in CRNDE, rs115515594, significantly increased the risk of recurrence. Consistently, in BOTS only, the same genetic variant was highly overrepresented compared to healthy individuals. We also discovered that hypomethylation of CRNDE is associated with increased aggressiveness of ovarian tumors. Accordingly, hypomethylation of this gene’s promoter/first exon correlated with hgOvCa resistance to chemotherapy, but only in specimens with accumulation of the TP53 tumor suppressor protein. Taken together, these results contribute to a better understanding of the role of CRNDE(P) in tumorigenesis and potentially may lead to improvements in screening, diagnosis, and treatment of ovarian neoplasms.

Bimatoprost is a prostaglandin analog used in cosmetic products designed to stimulate hair growth, including eyebrows. However, limited study-based evidence confirming its efficacy, safety, and patient satisfaction is available. The aim of this study was to investigate whether, and to what extent, bimatoprost affects the density and length of eyebrow hair in young women in comparison to sweet almond oil. The study group included 27 healthy women who used bimatoprost cosmetic preparation once daily for 5 weeks and then crossed over to use sweet almond oil once daily for 5 weeks. Bimatoprost preparation, in comparison to sweet almond oil, improved eyebrow density (70% vs. 30%; p = 0.003) and eyebrow hair elongation (59% vs. 26%; p = 0.014), but not eyebrow hair hydration (59% vs. 89%; p = 0.001) and hair darkness (22% vs. 11%; p = 0.278). The comfort of use was comparable for both preparations (63% vs. 67%; p = 0.996), but treatment satisfaction was significantly higher with bimatoprost (66% vs. 22%; p < 0.001). All reported adverse events were minor, transient, and resolved spontaneously. We conclude that the bimatoprost preparation was significantly more effective than sweet almond oil in improving the density and length of eyebrow hair, with a similar level of safety. Therefore, bimatoprost can be considered an ingredient in cosmetics designed to enhance eyebrow growth; however, further larger studies with extended follow-ups are needed.

The aim of the study was to investigate the effect of ethanol on the properties of acrylic-urethane resin products obtained by vat photopolymerization using the masked stereolithography method. The effect of alcohol at concentrations of 15, 25, and 35% in the resin on the chemical structure, weight, thickness of the samples, and mechanical properties in static tabltensile tests performed immediately after printing and one month later were studied. The results obtained were evaluated in terms of the use of ethanol as a cosolvent to help load the resin with agomelatine for the potential of obtaining microneedle transdermal systems. It was shown that in terms of stability of properties, the most favorable system was resin with the addition of alcohol at a concentration of 15%. The greatest changes induced by the presence of the solvent in the resin were observed in the case of tensile properties, where the alcohol caused a decrease in the plasticity of the material, reducing the relative elongation at break from 14% for the pure resin to 4% when the alcohol concentration was 35%. Young’s modulus and tensile strength also decreased with the addition of alcohol by 18% and 31%, respectively, for testable samples with the maximum amount of alcohol in the resin. The deterioration in properties is most likely related to the effect of the solvent on the radical polymerization process of the resin, particularly the phenomenon of chain transfer to the solvent, which is important in view of the intended application of the developed material.

BackgroundPatients with inborn errors of immunity (IEI) experience recurrent infections, autoimmunity, and malignancies. Owing to repeated medical procedures, the need for constant treatment and surveillance, and the unpredictable course of the disease, patients with IEI are prone to develop mental health disorders, including anxiety. In this study, we aimed to assess the prevalence and level of anxiety symptoms in adult Polish patients with IEI and explore the determinants of anxiety in this group of patients.MethodsData from 105 Polish patients with IEI were collected via the hospital anxiety and depression scale (HADS), brief illness perception questionnaire (B-IPQ), illness cognition questionnaire (ICQ), Pittsburgh sleep quality index (PSQI), and a questionnaire on general health and demographic data. For statistical analyses of data, the normality of distribution of quantitative data was assessed, and internal consistency of tests was investigated using Cronbach’s alpha coefficient; moreover, we performed the analysis of correlations and between-group differences, and path analysis to explore causal relationships. Significance was considered at p < 0.050.ResultsThirty-eight (36.2%) patients had anxiety symptoms (HADS-A ≥ 8); 14 (13.3%) patients had severe anxiety (score ≥ 11), and 24 (22.9%) had moderate anxiety (score of 8–10). Patients with poor sleep quality, higher pain frequency, younger age, and no fixed income had higher anxiety scores than others. Emotional and cognitive representations of illness were positively correlated with anxiety levels. Intense anxiety was related to more negative illness perception, higher helplessness, lower illness acceptance, and lower perceived benefits.DiscussionAnxiety is common in patients with IEI. However, results indicate that it is not related to a more severe course of IEI or several comorbidities, whereas, pain frequency and poor sleep quality were identified to be important clinical factors for anxiety. Because anxiety was related to negative illness perception, psychological therapy may apply to this group of patients.
