Abstract

Adrenomedullin expression in rat uterus is correlated with plasma estradiol. Am J Physiol Endocrinol Metab 282: E139-E146, 2002.--Levels of expression of adrenomedullin (AM) in the uterus have been reported to vary with the reproductive cycle. This study examines the relationships among uterine AM mRNA, the stage of the estrous cycle, and circulating estradiol and progesterone in cycling rats and in ovariectomized (OVX) rats without or with estrogen replacement (ER). Strong AM mRNA, AM immunoreactivity, and pro-AM N[H.sub.2]-terminal 20 peptide (PAMP) immunoreactivity were observed in endometrial stroma by use of in situ hybridization and immunocytochemistry. Endometrial expression was particularly intense at proestrus and estrus, with weaker expression in the myometrium. By RNase protection assay, significant differences in AM mRNA between the stages of the estrous cycle could not be established. However, levels of AM mRNA were positively correlated with plasma estradiol in cycling rats (r = 0.56, P < 0.005) and in OVX and ER rats (r = 0.92, P < 0.001) and were not correlated with plasma progesterone. Levels of AM mRNA were significantly reduced after OVX compared with cycling rats, and ER restored AM mRNA to levels equivalent to those seen at the peak of the cycle (proestrus). In conclusion, although AM expression in the uterus varies throughout the estrous cycle, it is more closely correlated with circulating estradiol levels than with the stage of the cycle itself. adrenomedullin; proadrenomedullin N[H.sub.2]-terminal 20 peptide; estradiol; uterus; estrous cycle

Am J Physiol Regulatory Integrative Comp Physiol 281: R1887-R1894, 2001.--The role of adrenomedullin (ADM) in volume and pressure homeostasis remains undefined. Accordingly, we compared the biological responses to infusions of ADM and nitroprusside (NP; matched for reduction of arterial pressure) and assessed their effects on responses to ANG II and norepinephrine in eight conscious sheep. During matched falls in arterial pressure (8-10 mmHg, both P < 0.001) ADM and NP induced similar increases in heart rate. ADM increased cardiac output (P < 0.001), and the fall in calculated peripheral resistance was greater with ADM than NP (P = 0.013). ADM infusions raised plasma ADM levels (P < 0.001), plasma renin activity (P = 0.001), and ANG II (P < 0.001) but tended to blunt any concurrent rise in aldosterone compared with NP (P = 0.056). ADM maintained both urine flow (P < 0.001) and sodium excretion (P = 0.01) compared with falls observed with NP. ADM attenuated the vasopressor actions of exogenous ANG II (P = 0.006) but not norepinephrine. In addition, ADM antagonized the ANG II-induced, rise in plasma aldosterone (P < 0.001). In conclusion, ADM induces a different spectrum of hemodynamic, renal, and endocrine actions to NP. 82. Measurements on the growth of gaseous ionization at ultra high frequencies

Abstract

An apparatus has been built to study ionization by collision in a gas under ultra high frequency fields and especially the conditions covering the transition to the conducting state. The u.h.f. field is developed between two Rogowski profiled electrodes incorporated into a tuned quarter wave transmission line which is loosely coupled to the main oscillator. The u.h.f. field has a frequency of 105 Mc/sec. and its magnitude is measured by its effect on the frequency of oscillation of a small metal disc suspended in the gap. This instrument is used as a step-over so that a rectifying circuit loosely coupled to the transmission line can be calibrated. The disc can be removed from the gap during the experiments. Electrons are emitted from a thermionic cathode situated inside one of the electrodes and diffuse out into the gap through small holes in the electrode shell where they are moved across the gap by a small unidirectional field. The large u.h.f. field is superimposed on this field so that the electrons acquire enough energy to ionize the gas by collision; the resulting electron current to the far electrode is measured by a galvanometer. With this arrangement the amplification resulting from collision ionization can "be measured and its dependence on the lifetime of the electron in the gap can be studied. The measurements can. be carried right up to breakdown conditions, and the results can be expressed in terms of the number of ionizing collisions per second in the life of one electron, a coefficient closely related to Tosnsend's a. It is shown that the lifetime of the electrons not only depends on the drift under the direct field but also on the process of diffusion out of the gap and two methods of evaluating it are given. Although only preliminary results have been obtained these are sufficient to indicate that a useful contribution to ionization studies can be made by investigations with this apparatus.

Abstract

The biological actions of adrenomedullin in conscious sheep were analyzed to characterize the regulatory activity of the vasodilator peptide on renal performance, renal hemodynamics and endocrine activity. Infusions of adrenomedullin resulted in clear-cut renal hemodynamic effects such as reduced filling pressure and arterial pressure. The blood-pressure lowering effects of adrenomedullin was also coupled with increased cardiac output and renin stimulation in conscious sheep.

Abstract

Clearance receptor and endopeptidase 24.11 pathways play an equal role in the regulation of natriuretic peptide clearance in normal conscious sheep. The influence on hemodynamic and renal effects may also be significant. The administration of a natriuretic peptide receptor-C ligand or a endopeptidase 24.11 antagonist enhances plasma levels of atrial and brain natriuretic peptide and guanosine 3',5'-cyclic monophosphate. The same results are obtained when the two compounds are administered in combination.

Abstract

The effect of chronic dosing of the endogenous hormone ouabain on renal function was investigated by examining the role of ouabain in the development of hypertension in sheep. Ouabain infusion reduced heart rate and arterial pressure, and had no effect on pressor responsiveness to angiotensin II (ANG II). However, it reduced urine volume, urinary sodium excretion, plasma renin and ANG II concentration. These results provide evidence that ouabain does not cause hypertension, but it is both antidiuretic and antinatriuretic.
