The purpose of this study was to investigate the connection between the soundscape of a forest park, restorative perception, and health benefits. In order to assess sound source perception, soundscape perception, restorative perception, and health benefits, 10 forest park environments in Fuzhou National Forest Park were chosen for sound walks. Correlation analysis, structural equation modeling, and mediating effects were used to analyze the relationships between the variables. The results showed that (1) the majority of natural sounds, like birdsong, had a positive correlation with soundscape perception with respect to being perceived as pleasant, harmonious, varied, and fluctuating; however, human-related and traffic noises had a negative correlation with perceptions of being pleasant and harmonious, and a positive correlation with perceptions of roughness. (2) The sound of running water and wind-blown leaves had strong favorable connections with emotional, cognitive, and behavioral restorative perceptions. The sound of birdsong was strongly correlated with restorative perceptions regarding emotional dimensions. The sound of wind held substantial favorable associations with emotional and cognitive restorative perceptions. Natural sounds, except for the sound of cicada chirping, had positive correlations with health benefits. The associations between human-related and mechanical sounds and restorative perception and health benefits were not statistically significant. (3) Soundscape pleasantness had a significant positive effect on restorative perceptions, and restorative perceptions had a significant positive effect on health benefits. The effect of soundscape pleasantness on health benefits was fully communicated through restorative perceptions. The annoyingness of a soundscape had no effect on restorative perception or health benefits. In the future, forest recreation activities based on soundscape perception could be carried out through the considered use of natural soundscape resources to promote health benefits.

Mimicking the nitric oxide (NO)-release and glycocalyx functions of native vascular endothelium on cardiovascular stent surfaces has been demonstrated to reduce in-stent restenosis (ISR) effectively. However, the practical performance of such an endothelium-mimicking surfaces is strictly limited by the durability of both NO release and bioactivity of the glycocalyx component. Herein, we present a mussel-inspired amine-bearing adhesive coating able to firmly tether the NO-generating species (e.g., Cu-DOTA coordination complex) and glycocalyx-like component (e.g., heparin) to create a durable endothelium-mimicking surface. The stent surface was firstly coated with polydopamine (pDA), followed by a surface chemical cross-link with polyamine (pAM) to form a durable pAMDA coating. Using a stepwise grafting strategy, Cu-DOTA and heparin were covalently grafted on the pAMDA-coated stent based on carbodiimide chemistry. Owing to both the high chemical stability of the pAMDA coating and covalent immobilization manner of the molecules, this proposed strategy could provide 62.4% bioactivity retention ratio of heparin, meanwhile persistently generate NO at physiological level from 5.9 ± 0.3 to 4.8 ± 0.4 × 10−10 mol cm−2 min−1 in 1 month. As a result, the functionalized vascular stent showed long-term endothelium-mimicking physiological effects on inhibition of thrombosis, inflammation, and intimal hyperplasia, enhanced re-endothelialization, and hence efficiently reduced ISR.

Neointimal hyperplasia and stent thrombosis remain issues with vascular stents. Here, the authors report on the development of a nitric oxide releasing hydrogel which allows for endothelialisation of the stent surface and prevents smooth muscle cell growth reducing hyperplasia and thrombosis in in vivo models.

Endowing materials with catalytic activities analogous to those of the natural endothelium to thus enhance their biological performance has become an option for constructing advanced blood-contact materials. The electron transfer between Cu2+ and Cu+ in the porphyrin center can catalyze the reaction of GSH and GSNO to generate NO, and this electron transfer can also catalyze the decomposition of ROS. Based on this, we created a dual-catalytic surface possessing NO-generating and ROS-scavenging activities to better mimic the versatile catalytic abilities of the endothelium. Copper tetraphenylporphyrin/titanium dioxide nanoparticles (CuTPP/TiO2-NPs) exhibiting excellent NO-generating and ROS-scavenging activities were synthesized and immobilized on the material surface to form a dual-catalytic film (CuTPP/TiO2-film) with the help of the catechol chemistry technique. Unlike most single catalytic surfaces, the dual-catalytic CuTPP/TiO2-film effectively regulated the microenvironment surrounding the implanted device by releasing NO signaling molecules and scavenging harmful ROS. This dual-catalytic film exhibited excellent biosafety and biocompatibility with anti-thrombosis, vascular wall cells (ECs and SMCs) modulation, and anti-inflammatory properties. We envision that this dual-catalytic endothelial bionic strategy may provide a promising solution to the clinical problems plaguing blood-contact devices and provide a novel basis for the further development of surface catalytic-engineered biomaterials.

Chronic wounds and the accompanying inflammation are ongoing challenges in clinical treatment. They are usually accompanied by low pH and high oxidative stress environments, limiting cell growth and proliferation. Ordinary medical gauze has limited therapeutic effects on chronic wounds, and there is active research to develop new wound dressings. The chitosan hydrogel could be widely used in biomedical science with great biocompatibility, but the low mechanical properties limit its development. This work uses polyacrylamide to prepare double-network (DN) hydrogels based on bioadhesive catechol-chitosan hydrogels. Cystamine and N, N′-Bis(acryloyl)cystamine, which can be cross-linking agents with disulfide bonds to prepare redox-responsive DN hydrogels and pH-responsive nanoparticles (NPs) prepared by acetalized cyclodextrin (ACD) are used to intelligently release drugs against chronic inflammation microenvironments. The addition of catechol groups and ACD-NPs loaded with the Resolvin E1 (RvE1), promotes cell adhesion and regulates the inflammatory response at the wound site. The preparation of the DN hydrogel in this study can be used to treat and regulate the inflammatory microenvironment of chronic wounds accurately. It provides new ideas for using inflammation resolving factor loaded in DN hydrogel of good biocompatibility with enhanced mechanical properties to intelligent regulate the wound inflammation and promote the wound repaired.

Acute myocardial infarction (AMI) has the characteristics of sudden onset, rapid progression, poor prognosis, and so on. Therefore, it is urgent to identify diagnostic and prognostic biomarkers for it. Cuproptosis is a new form of mitochondrial respiratory-dependent cell death. However, studies are limited on the clinical significance of cuproptosis-related genes (CRGs) in AMI. In this study, we systematically assessed the genetic alterations of CRGs in AMI by bioinformatics approach. The results showed that six CRGs (LIAS, LIPT1, DLAT, PDHB, MTF1, and GLS) were markedly differentially expressed between stable coronary heart disease (stable_CAD) and AMI. Correlation analysis indicated that CRGs were closely correlated with N6-methyladenosine (m6A)-related genes through R language “corrplot” package, especially GLS was positively correlated with FMR1 and MTF1 was negatively correlated with HNRNPA2B1. Immune landscape analysis results revealed that CRGs were closely related to various immune cells, especially GLS was positively correlated with T cells CD4 memory resting and negatively correlated with monocytes. Kaplan–Meier analysis demonstrated that the group with high DLAT expression had a better prognosis. The area under curve (AUC) certified that GLS had good diagnostic value, in the training set (AUC = 0.87) and verification set (ACU = 0.99). Gene set enrichment analysis (GSEA) suggested that GLS was associated with immune- and hypoxia-related pathways. In addition, Gene Ontology (GO) analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis, competing endogenous RNA (ceRNA) analysis, transcription factor (TF), and compound prediction were performed to reveal the regulatory mechanism of CRGs in AMI. Overall, our study can provide additional information for understanding the role of CRGs in AMI, which may provide new insights into the identification of therapeutic targets for AMI.

BackgroundSurgical interventions for tumors in the cervical esophageal region are complicated and laryngeal function is frequently sacrificed. Therefore, we attempted the tracheal transection approach to resect the tumor while preserving laryngeal function.MethodsThree patients with papillary thyroid cancer (PTC), six with cervical esophageal cancer (CEC), and four with CEC mixed with thoracic esophageal cancer (TEC) were enrolled. The esophagus was exposed after the trachea was transected between the second and third tracheal rings. CEC/TEC: Resection of the esophagus or/and a portion of the hypopharynx with acceptable safety margins and repair with free jejunum or tubular stomach. PTC: Suture the small esophageal incision immediately after removing the tumor. The tracheal dissection was repaired with interrupted sutures throughout the entire layer after the esophageal lesion was resected. The status of the recurrent laryngeal nerve (RLN) determined whether a tracheotomy was necessary.ResultsAll 13 patients had effective esophageal lesion excision, with six of them requiring intraoperative tracheotomy. Postoperative complications included a tracheoesophageal fistula (one case, 7.7%), postoperative RLN paralysis (two cases, 15.4%), and aspiration (three cases, 23.1%). Except for two patients with distant metastases, there was no recurrence in the remaining patients after 5–92 months of follow-up.ConclusionThe tracheal transection approach, as a new surgical technique, can retain laryngeal function while ensuring appropriate exposure and satisfactory surgical resection. Before surgery, the feasibility of this approach must be carefully assessed. The RLN should be protected during the procedure. The operation is both safe and effective, with a wide range of applications.

Long non-coding RNAs (lncRNAs) have been extensively studied as important regulators of tumor development in various cancers. Tumor protein 53 target gene 1 (TP53TG1) is a newly identified lncRNA in recent years, and several studies have shown that TP53TG1 may play oncogenic or anti-oncogenic roles in different cancers. Nevertheless, the role of TP53TG1 in the development of cervical cancer is unclear. In our study, pan-cancer analysis showed that high expression of TP53TG1 was significantly associated with a better prognosis. We then constructed a TP53TG1 overexpression model in HeLa cell line to explore its functions and molecular targets. We found that TP53TG1 overexpression significantly inhibited cell proliferation and induced apoptosis, demonstrating that TP53TG1 may be a novel anti-oncogenic factor in cervical cancer. Furthermore, overexpression of TP53TG1 could activate type I interferon signaling pathways and inhibit the expression of genes involved in DNA damage responses. Meanwhile, TP53TG1 could affect alternative splicing of genes involved in cell proliferation or apoptosis by regulating the expression of many RNA-binding protein genes. Competing endogenous RNA (ceRNA) network analysis demonstrated that TP53TG1 could act as the sponge of several miRNAs to regulate the expression level of target genes. In conclusion, our study highlights the essential role of lncRNA TP53TG1 in the development of cervical cancer and suggests the potential regulatory mechanisms.

Diabetic nephropathy (DN) is the leading cause of death and the greatest risk to the lives of people with advanced diabetes. Yet, the molecular mechanisms underlying its development and progression remain unknown. In this research, we studied the primary pathways driving DN using transcriptome sequencing and immune repertoire analysis. Firstly, we found that the diversity and abundance of the immune repertoire in late DN were significantly increased, while there was no significant change in early DN. Furthermore, B cell-mediated antibody responses may be the leading cause of DN progression. By analyzing master regulators, we found the key DN-driving transcription factors. In the late stage of DN, immune cells, fibroblasts, and epithelial cells were abundant, but other stromal cells were few. Early DN kidneys had a higher tissue stemness score than normal and advanced DN kidneys. We showed that DN progression involves proximal tubular metabolic reprogramming and stemness restoration using Monocle3. Through WGCNA, we found that co-expression modules that regulate DN progression and immune repertoire diversity mainly regulate immune-related signaling pathways. In addition, we also found that early DN had apparent activation of immune-related signaling pathways mainly enriched in immune cells. Finally, we found that activation of fibroblasts is typical of early DN. These results provide a research basis for further exploring the molecular biology and cellular mechanisms of the occurrence and development of DN and provide a theoretical basis for the prevention and treatment of DN.

The coronavirus disease 2019 pandemic has caused more than 532 million infections and 6.3 million deaths to date. The reactive and neutralizing fully human antibodies of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) are effective detection tools and therapeutic measures. During SARS-CoV-2 infection, a large number of SARS-CoV-2 reactive and neutralizing antibodies will be produced. Most SARS-CoV-2 reactive and neutralizing fully human antibodies are isolated from human and frequently encoded by convergent heavy-chain variable genes. However, SARS-CoV-2 viruses can mutate rapidly during replication and the resistant variants of neutralizing antibodies easily survive and evade the immune response, especially in the face of such focused antibody responses in humans. Therefore, additional tools are needed to develop different kinds of fully human antibodies to compensate for current deficiency. In this study, we utilized antibody humanized CAMouseHG mice to develop a rapid antibody discovery method and examine the antibody repertoire of SARS-CoV-2 RBD-reactive hybridoma cells derived from CAMouseHG mice by using high-throughput single-cell V(D)J sequencing analysis. CAMouseHG mice were immunized by 28-day rapid immunization method. After electrofusion and semi-solid medium screening on day 12 post-electrofusion, 171 hybridoma clones were generated based on the results of SARS-CoV-2 RBD binding activity assay. A rather obvious preferential usage of IGHV6-1 family was found in these hybridoma clones derived from CAMouseHG mice, which was significantly different from the antibodies found in patients with COVID-19. After further virus neutralization screening and antibody competition assays, we generated a noncompeting two-antibody cocktail, which showed a potent prophylactic protective efficacy against SARS-CoV-2 in cynomolgus macaques. These results indicate that humanized CAMouseHG mice not only provide a valuable platform to obtain fully human reactive and neutralizing antibodies but also have a different antibody repertoire from humans. Thus, humanized CAMouseHG mice can be used as a good complementary tool in discovery of fully human therapeutic and diagnostic antibodies.

Successful interaction in the target language requires L2 learners to use and understand the grammatically correct language. At the same time, the language used is expected to produce socioculturally appropriate utterances that it refers to their Pragmatic competence. The latter entails acquiring pragmatic competence, which has proved to be very challenging for L2 learners. This is because they gain limited exposure to the use of language for real communication in an authentic social setting. Moreover, instruction has been found to influence the functional abilities in L2 as it equips the learners with the ability to produce and comprehend L2 in different situations. Focusing on the research conducted on the role of explicit and implicit instruction on L2 pragmatic competence, this study aimed to give a summative description of the empirical studies carried out on teaching pragmatics. The investigation ends up with a conclusion, instructional implications, and suggestions for future research.

IntroductionThis study aimed to explore the novel classification of inpatients with new-onset diabetes in Eastern China by the cluster-based classification method and compare the clinical characteristics among the different subgroups.MethodsA total of 1017 Inpatients with new-onset diabetes of five hospitals in Eastern China were included in the study. Clustering analysis was used to cluster the data into five subgroups according to six basic variables. The differences in clinical characteristics, treatments, and the prevalence of diabetes-related diseases among the five subgroups were analyzed by multiple groups comparisons and pairwise comparisons. The risk of diabetes-related diseases in the five subgroups was compared by calculating odd ratio (OR). P value < 0.05 was considered significant.ResultsFive subgroups were obtained by clustering analysis with the highest proportion of patients with severe insulin-deficient diabetes (SIDD) 451 (44.35%), followed by patients with mild age-related diabetes (MARD) 236 (23.21%), patients with mild obesity-related diabetes (MOD) 207 (20.35%), patients with severe insulin-resistant diabetes (SIRD) 81 (7.96%), and patients with severe autoimmune diabetes (SAID) 42 (4.13%). Five subtypes had their own unique characteristics and treatments. The prevalence and risk of diabetes-related complications and comorbidities were also significantly different among the five subtypes. Diabetic kidney disease (DKD) was the most common in SIRD group. Patients in SIDD, SIRD, and MARD groups were more likely to develop cardiovascular disease (CVD) and/or stroke, diabetic peripheral vascular disease (DPVD), and diabetic distal symmetric polyneuropathy (DSPN). The prevalence and risk of metabolic syndrome (MS) were the highest in MOD and SIRD groups. Patients in SAID group had the highest prevalence and risk of diabetic ketoacidosis (DKA). Patients with MOD were more likely to develop non-alcoholic fatty liver disease (NAFLD).ConclusionsThe inpatients with new-onset diabetes in Eastern China had the unique clustering distribution. The clinical characteristics, treatments, and diabetes-related complications and comorbidities of the five subgroups were different, which may provide the basis for precise treatments of diabetes.

Based on the theory of “Non-Economic Promotion Championship,” this paper takes Appraisal-And-Commendation (AAC) policy of National Civilised City (NCC) program in China as the research object, and analyses the influence and mechanism of the NCC on carbon emissions from the perspective of energy demand. We find that NCC reduces carbon emissions significantly, and this reduction effect continues to expand over time. Furthermore, the NCC reduces carbon emissions through two mechanisms: First, the NCC reduces carbon emissions by slowing urbanisation. This mechanism mainly functions in big cities, megacities, and super cities, and does not function in small and medium cities. Second, the NCC reduces carbon emissions by promoting industrial restructuring from secondary industry to tertiary industry. This carbon reduction effect is a pure structural adjustment effect, regardless of any effects on technological level and productivity. Moreover, there are regional differences in the reduction of carbon emissions by NCC through industrial restructuring in two dimensions: In the first dimension, compared with the western region and the northeast region, the eastern and central regions are more able and willing to reduce carbon emissions through industrial restructuring. In the second dimension, compared with the northern region, the southern region is more likely to reduce carbon emissions through industrial restructuring.

Previous studies suggest that allergic diseases may be a protective factor in SARS-CoV-2 infection. However, data regarding the impact of dupilumab, a widely used immunomodulatory medication, on COVID-19 in an allergic population are very limited. To investigate the incidence and severity of COVID-19 among moderate-to-severe atopic dermatitis (AD) patients treated with dupilumab, a retrospective cross-sectional survey was conducted among patients with moderate-to-severe AD who presented at the Department of Allergy of Tongji Hospital from 15 January 2023 to 31 January 2023. Healthy individuals matched for gender and age were also enrolled as a control. All subjects were asked about their demographic characteristics, past medical history, COVID-19 vaccination history, and medications, as well as the presence and duration of individual COVID-19-related symptoms. A total of 159 moderate-to-severe AD patients and 198 healthy individuals were enrolled in the study. Among the AD patients, 97 patients were treated with dupilumab, and 62 patients did not receive any biologicals or systemic treatments (topical treatment group). The proportions of people who were not infected with COVID in the dupilumab treatment group, topical treatment group and healthy control group were 10.31%, 9.68% and 19.19%, respectively (p = 0.057). There was no significant difference in COVID-19-related symptom scores among all groups (p = 0.059). The hospitalization rates were 3.58% in the topical treatment group and 1.25% in the healthy control group, and no patient was hospitalized in the dupilumab treatment group (p = 0.163). Compared with healthy control group and topical treatment group, the dupilumab treatment group had the shortest COVID-19-associated disease duration (dupilumab treatment group, 4.15 ± 2.85 d vs. topical treatment group, 5.43 ± 3.15 d vs. healthy control group, 6.09 ± 4.29 d; p = 0.001). Among the AD patients treated with dupilumab for different times, there was no appreciable difference (1 year group, 2.8 ± 1.32 d; p = 0.183). Dupilumab treatment shortened the duration of COVID-19 in patients with moderate-to-severe AD. AD patients can continue their dupilumab treatment during the COVID-19 pandemic.

Multiview stereo (MVS) achieves efficient 3D reconstruction on Lambertian surfaces and strongly textured regions. However, the reconstruction of weakly textured regions, especially planar surfaces in weakly textured regions, still faces significant challenges due to the fuzzy matching problem of photometric consistency. In this paper, we propose a multiview stereo for recovering planar surfaces guided by confidence calculations, resulting in the construction of large-scale 3D models for high-resolution image scenes. Specifically, a confidence calculation method is proposed to express the reliability degree of plane hypothesis. It consists of multiview consistency and patch consistency, which characterize global contextual information and local spatial variation, respectively. Based on the confidence of plane hypothesis, the proposed plane supplementation generates new reliable plane hypotheses. The new planes are embedded in the confidence-driven depth estimation. In addition, an adaptive depth fusion approach is proposed to allow regions with insufficient visibility to be effectively fused into the dense point clouds. The experimental results illustrate that the proposed method can lead to a 3D model with competitive completeness and high accuracy compared with state-of-the-art methods.

The bio-refining process of bamboo is more challenging compared to wood due to its dense and stabilized complex polymer structure, as well as its abundance of degradation-resistant components. Consequently, the bio-refining of bamboo requires more energy and time consumption compared to the bio-refining of wood. In this study, co-cultured microorganisms were utilized for the pretreatment of bamboo to improve pulping performance, reduce consumption, and increase efficiency. These microorganisms were constructed by combining environmental microorganisms found in bamboo pulp with Bacillus sp. that were self-screened. The results of 16S rRNA analysis showed that the genera Proteobacteria, Firmicutes, Bacteroidota, and Actinobacteriota gradually became dominant during the treatment process. Additionally, the PICRUSt results indicated that the co-culture microbial consortium C strategy strengthened key enzyme activities related to the degradation of bamboo lignocelluloses. The microbial consortium pretreatment resulted in removing lignin and hemicellulose at rates of 21.96% and 26.21%, respectively. This process also caused a decrease in the crystalline index, indicating the presence of disordered crystalline regions. This change was beneficial for the subsequent Kraft pulping process. Compared to the conventional bamboo pulp, the yield of pretreated bamboo pulp increased slightly, while the cellulose purity and paper properties were significantly superior. The obtained Kraft pulp, which underwent microbiological pretreatment, met the requirements for superior Kraft pulp products despite a 65 min decrease in cooking time and a 10 °C decline in maximum cooking temperature. This study proves that co-cultured microbial consortium used for pretreating bamboo are beneficial for bamboo Kraft pulping. This approach can be considered environmentally friendly and leads to energy saving and cost reduction in bamboo bio-refining processes.

Application of extracorporeal circuits and indwelling medical devices has saved many lives. However, it is accompanied with two major complications: thrombosis and infection. To address this issue, we apply therapeutic nitric oxide gas (NO) and antibacterial peptide for synergistically tailoring such devices for surface anti-thrombogenic and antifouling dual functions. Such functional surface is realized by stepwise conjugation of NO-generating compound of 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) chelated copper ions (Cu-DOTA) and dibenzylcyclooctyne- (DBCO-) modified antimicrobial peptide based on carbodiimide and click chemistry respectively. The integration of peptide and Cu-DOTA grants the modified surface the ability to not only efficiently inhibit bacterial growth, but also catalytically generate NO from endogenous s-nitrosothiols (RSNO) to reduce adhesion and activation of platelets, preventing the formation of thrombus. We envision that the stepwise synergistic modification strategy by using anticoagulant NO and antibacterial peptide would facilitate the surface multifunctional engineering of extracorporeal circuits and indwelling medical devices, with reduced clinical complications associated with thrombosis and infection.

Abstract Since the advances in protein engineering and manufacture, over the last 30 years, antibody-based immunotherapeutic has become a powerful strategy to treat diseases. The T-cell engaging bispecific antibody (BsAb) by combining the Fab binding domain of tumor antigens and Fab or single-chain variable fragments (scFvs) binding domain of CD3 molecules, could redirect cytotoxic T cells to kill tumor cells. The IgG-scFv format of BsAb is a dual bivalent and asymmetrical design, which adds the benefit of potent cytotoxicity and less complicated for manufacture but limits the stability and production. Here, we engineered a series of interchain disulfide bonds in the Fab region of IgG-svFv BsAbs and evaluated its biophysical and biological properties. We found that simultaneously replaced the position of VH44-VL100 and CH1126-CL121 residues with cysteine, to form two additional disulfide bonds, could markedly increase monomeric BsAb formation and yield. The thermostability and stability against aggregation and degradation also performed better than BsAbs without extra disulfide bonds introduction. Besides, the affinity of engineered BsAbs was maintained, and the h8B-BsAb antibody had a slight enhancement in an inhibitory effect on target cells.

Coronary atherosclerotic lesions exhibit a low-pH chronic inflammatory response. Due to insufficient drug release control, drug-eluting stent intervention can lead to delayed endothelialization, advanced thrombosis, and unprecise treatment. In this study, hyaluronic acid and chitosan were used to prepare pH-responsive self-assembling films. The hydrogen sulfide (H2S) releasing aspirin derivative ACS14 was used as drug in the film. The film regulates the release of the drug adjusted to the microenvironment of the lesion, and the drug balances the vascular function by releasing the regulating gas H2S, which comparably to NO promotes the self-healing capacity of blood vessels. Drug releasing profiles of the films at different pH, and other biological effects on blood vessels were evaluated through blood compatibility, cellular, and implantation experiments. This novel method of self-assembled films which H2S in an amount, which is adjusted to the condition of the lesion provides a new concept for the treatment of cardiovascular diseases.

Abstract Background Allergen immunotherapy (AIT) is the only causal therapy for IgE-mediated allergy. There is less evidence about the safety and efficacy of AIT especially subcutaneous immunotherapy (SCIT) in children under 5 years old. We aimed to investigate the side effects and associated risk factors of house dust mite (HDM) SCIT in preschool children with respiratory allergic diseases. Methods The preschool children who had HDM-related allergic rhinitis with/without asthma were enrolled and undergone standardized HDM SCIT in our department from June 2013 to December 2019. Local reactions (LRs) and systemic reactions (SRs) were recorded and categorized according to World Allergy Organization recommendations. Demographic data and other therapeutic-related parameters were also recorded to investigate potential risk factors for these side effects. Results A total of 91 children (60 boys, 65.93%; 31 girls, 34.07%; mean age 4.13 years old) were included in the study. Among the 91 patients, 3109 SCIT injections were recorded, 62/91 (68.13%) experienced 186 immediate LRs, 4 /91(4.40%) experienced 6 delayed LRs, 11/91 (12.09%) children experienced 44 immediate SRs, 21/44 (47.73%) were grade 1 SRs, 21/44 (47.73%) were grade 2, 2/44 (4.55%) were grade 3, no grade 4 or 5 SRs occurred. Furthermore, 1/91 (1.10%) experienced 1 delayed SRs, manifested by urticaria 2 days later after allergen injection. 9/91 (9.89%) experienced 2 or more times SRs. Multivariable logistic regression analysis showed BMI (OR 1.506; 95%CI 1.091 to 2.079; p 0.05). Conclusions HDM subcutaneous immunotherapy is considered to be safe in preschool children with respiratory allergic diseases. Higher BMI and HDM sIgE level in children are risk factors for developing LRs. The incidence of SRs and the rate of severe SRs are low in preschool children.

Abstract Background Solute carrier family 7 member 11 (SLC7A11) is overexpressed in multiple human tumours and functions as a transporter importing cystine for glutathione biosynthesis. It promotes tumour development in part by suppressing ferroptosis, a newly identified form of cell death that plays a pivotal role in the suppression of tumorigenesis. However, the role and underlying mechanisms of SLC7A11‐mediated ferroptosis in hepatoblastoma (HB) remain largely unknown. Methods Reverse transcription quantitative real‐time PCR (RT‐qPCR) and western blotting were used to measure SLC7A11 levels. Cell proliferation, colony formation, lipid reactive oxygen species (ROS), MDA concentration, 4‐HNE, GSH/GSSG ratio and cell death assays as well as subcutaneous xenograft experiments were used to elucidate the effects of SLC7A11 in HB cell proliferation and ferroptosis. Furthermore, MeRIP‐qPCR, dual luciferase reporter, RNA pulldown, RNA immunoprecipitation (RIP) and RACE‐PAT assays were performed to elucidate the underlying mechanism through which SLC7A11 was regulated by the m6A modification in HB. Results SLC7A11 expression was highly upregulated in HB. SLC7A11 upregulation promoted HB cell proliferation in vitro and in vivo, inhibiting HB cell ferroptosis. Mechanistically, SLC7A11 mRNA exhibited abnormal METTL3‐mediated m6A modification, which enhanced its stability and expression. IGF2 mRNA‐binding protein 1 (IGF2BP1) was identified as the m6A reader of SLC7A11, enhancing SLC7A11 mRNA stability and expression by inhibiting SLC7A11 mRNA deadenylation in an m6A‐dependent manner. Moreover, IGF2BP1 was found to block BTG2/CCR4‐NOT complex recruitment via competitively binding to PABPC1, thereby suppressing SLC7A11 mRNA deadenylation. Conclusions Our findings demonstrated that the METTL3‐mediated SLC7A11 m6A modification enhances HB ferroptosis resistance. The METTL3/IGF2BP1/m6A modification promotes SLC7A11 mRNA stability and upregulates its expression by inhibiting the deadenylation process. Our study highlights a critical role of the m6A modification in SLC7A11‐mediated ferroptosis, providing a potential strategy for HB therapy through blockade of the m6A‐SLC7A11 axis.

Abstract Magnesium (Mg) and its alloys can be used as biomedical materials because of their excellent mechanical properties and biocompatibility. However, the rapid degradation rate of Mg‐based materials limits their application in biodegradable intravascular stents. To overcome this issue, we constructed a hydrophobic coating on magnesium. After pre‐treatments with alkali and a silane coupling agent of pure magnesium, 4,4’‐diphenylmethane‐diisocyanate (MDI) and amino‐terminated polydimethylsiloxane (H2N–PDMS–NH2) were stepwise deposited on the surface, forming an amino‐containing hydrophobic coating (–(M/P)3) to enhance the corrosion resistance. Furthermore, polypeptide TK14 was immobilised on the hydrophobic coating to promote vascular endothelial cell adhesion and proliferation. The electrochemical results revealed that the self‐corrosion current density (icorr) of –(M/P)3 decreased by approximately 4.5 orders of magnitude compared with that of pure Mg. After TK14 immobilisation, the number of endothelial cells adhering to the surface of –(M/P)3–T increased significantly. Although the corrosion resistance of –(M/P)3–T was slightly reduced, the subcutaneous implantation inflammatory response of the surrounding tissues was lower, showing suitable biocompatibility. Therefore, the polypeptide TK14 functionalised hydrophobic coating may be a promising candidate material for the interface of magnesium‐based cardiovascular implants.

Abstract Because of their good performance, including biocompatibility and mechanical properties, polyurethanes (PUs) are widely used in medical devices. However, undesired compatibility troubles, including thrombus, inflammation, and hyperplasia, still limit the applications of PUs. In this study, copper‐mediated polyurethane (PU‐Cu) materials with enzyme‐like catalysis were prepared. The PU‐Cu materials effectively catalysed the nitric oxide (NO) released from endogenous NO donors because of the glutathione peroxidase (GPx)‐like function of copper ion. The PU‐Cu materials were respectively evaluated via platelet adhesion and endothelial cell (EC), smooth muscle cell (SMC), and macrophage (MA) cultures. Scanning electron microscopy results showed that PU‐Cu materials significantly inhibited platelet adhesion and activation. Meanwhile, PU‐Cu materials not only promote the proliferation of EC but also inhibit SMC growth. Moreover, MA culture results intuitively stated the anti‐inflammatory ability of PU‐Cu. In addition, experimental samples were implanted into the subcutaneous tissue of Sprague Dawley rats. The anti‐inflammatory function of PU‐Cu was further confirmed by haematoxylin‐eosin staining results. With regard to their excellent biological performance, PU‐Cu materials are proposed for biocompatibility improvement of blood‐contacting materials, which should in turn provide new ideas for advanced medical devices.

Titanium dioxide (TiO2) has a long history of application in blood contact materials, but it often suffers from insufficient anticoagulant properties. Recently, we have revealed the photocatalytic effect of TiO2 also induces anticoagulant properties. However, for long-term vascular implant devices such as vascular stents, besides anticoagulation, also anti-inflammatory, anti-hyperplastic properties, and the ability to support endothelial repair, are desired. To meet these requirements, here, we immobilized silver nanoparticles (AgNPs) on the surface of TiO2 nanotubes (TiO2-NTs) to obtain a composite material with enhanced photo-induced anticoagulant property and improvement of the other requested properties. The photo-functionalized TiO2-NTs showed protein-fouling resistance, causing the anticoagulant property and the ability to suppress cell adhesion. The immobilized AgNPs increased the photocatalytic activity of TiO2-NTs to enhances its photo-induced anticoagulant property. The AgNP density was optimized to endow the TiO2-NTs with anti-inflammatory property, a strong inhibitory effect on smooth muscle cells (SMCs), and low toxicity to endothelial cells (ECs). The in vivo test indicated that the photofunctionalized composite material achieved outstanding biocompatibility in vasculature via the synergy of photo-functionalized TiO2-NTs and the multifunctional AgNPs, and therefore has enormous potential in the field of cardiovascular implant devices. Our research could be a useful reference for further designing of multifunctional TiO2 materials with high vascular biocompatibility.

Thanks to its simplicity, versatility, and secondary reactivity, dopamine self-polymerized coatings (pDA) have been widely used in surface modification of biomaterials, but the limitation in secondary molecular grafting and the high roughness restrain their application in some special scenarios. Therefore, some other catecholamine coatings analog to pDA have attracted more and more attention, including the smoother poly-norepinephrine coating (pNE), and the poly-levodopa coating (pLD) containing additional carboxyl groups. However, the lack of a systematic comparison of the properties, especially the biological properties of the above three catecholamine coatings, makes it difficult to give a guiding opinion on the application scenarios of different coatings. Herein, we systematically studied the physical, chemical, and biological properties of the three catecholamine coatings, and explored the feasibility of their application for the modification of biomaterials, especially cardiovascular materials. Among them, the pDA coating was the roughest, with the largest amount of amino and phenolic hydroxyl groups for molecule grafting, and induced the strongest platelet adhesion and activation. The pLD coating was the thinnest and most hydrophilic but triggered the strongest inflammatory response. The pNE coating was the smoothest, with the best hemocompatibility and histocompatibility, and with the strongest cell selectivity of promoting the proliferation of endothelial cells while inhibiting the proliferation of smooth muscle cells. To sum up, the pNE coating may be a better choice for the surface modification of cardiovascular materials, especially those for vascular stents and grafts, but it is still not widely recognized.

Pre-pregnancy body mass index and weight gain management are associated with pregnancy outcomes in expecting women. Poor gestational weight gain (GWG) management could increase the risk of adverse complications. These risks can be alleviated by lifestyle-based interventions if an undesired GWG trend is detected early on in the pregnancy. Current literature lacks analysis of gestational weight gain data and tracking the pregnancy over time. In this work, we collected longitudinal gestational weight gain data from women during their pregnancy and model their weight measurements to predict the end-of-pregnancy weight gain and classify it in accordance with the medically recommended guidelines. The measurement frequency of the weights is often very variable such that segments of data can be missing and the need to predict early utilising few data points complicates data modelling. We propose a Bayesian approach to forecast weight gain while effectively dealing with the limited data availability for early prediction. We validate on diverse populations from Europe and China. We show that utilising individual’s data only up to mid-way through the pregnancy, our approach produces mean absolute errors of 2.45 kgs and 2.82 kgs in forecasting end-of-pregnancy weight gain on these populations respectively, whereas the best of state-of-the-art yields 8.17 and 6.60 kgs on respective populations. The proposed method can serve as a tool to keep track of an individual’s pregnancy and achieve GWG goals, thus supporting the prevention of excessive or insufficient weight gain during pregnancy.

With the development of science and technology, the function of waterborne coatings has been advanced to a higher standard, which requires researchers to innovate and expand the research on them. Aloe-emodin is a natural material with antibacterial properties. Applying its antibacterial effect to the coating can enrich its function and meet the diversified needs of consumers. In this study, the urea-formaldehyde resin was used as the wall material and the aloe-emodin as the core material to prepare the microcapsules. The coating rate, yield, and morphology of the microcapsules were characterized. Through an orthogonal experiment and a single factor experiment, the optimization scheme of microcapsule preparation was explored. The results indicated that the optimum preparation process of aloe-emodin microcapsules was as follows: the mass ratio of core material to wall material was 1:15, the molar ratio of urea to formaldehyde was 1:1.2, the temperature of microencapsulation was 50 °C, and the stirring speed of microencapsulation was 600 rpm. On this basis, the aloe-emodin microcapsules with 0%, 1.0%, 3.0%, 6.0%, 9.0%, and 12.0% contents were added to the waterborne coating to prepare the paint films, and their influence on the antibacterial and optical properties of the waterborne paint films was explored. The results demonstrated that the aloe-emodin microcapsules had antibacterial activity. When the content was 6.0%, the comprehensive performance of the film was better. The antibacterial rate of the film against Escherichia coli was 68.1%, and against Staphylococcus aureus it was 60.7%. The color difference of the film was 59.93, and the glossiness at 60° was 7.8%. In this study, the microcapsules that can improve the antibacterial performance of water-based coatings were prepared, which can expand the application of water-based coatings and provide a reference for the study of the functionalization of water-based coatings.

This report describes maxillary antral pseudocyst drift after maxillary sinus floor augmentation through osteotome sinus floor elevation with simultaneous implant placement. 3D Slicer was used to measure the pseudocyst and maxilla for the placement of the implants; follow-up visits were scheduled at 6, 12, and 22 months. No adverse effects were observed during or after surgery, and all implants exhibited osseointegration without mobility. At 6 months after surgery, the pseudocyst had moved posterolaterally from the preoperative position near the anterior medial maxillary sinus, then returned to its original position at 12 months. However, it had remigrated to the posterolateral position at 22 months. The preoperative volume of the pseudocyst was 3.795 mm3; it was 2.370, 3.439, and 2.930 mm3 at 6, 12, and 22 months after surgery, respectively. The changes in pseudocyst drift and volume did not have a substantial negative influence on the implants, presumably because of cystic attachment and the recurrence of multiple pseudocysts at different locations. The risks associated with changes in a pseudocyst can be avoided, if an appropriate treatment plan is selected.

Abstract Background Influenza virus remains a continuous and severe threat to public health worldwide, and its prevention and treatment have always been a major international issue. Because of its ability to evade immune surveillance through rapid antigenic drift and antigenic shift, broad-spectrum vaccines seem increasingly important. Methods A mAb named 3C12 from an immortalized hybrid cell was generated via immunizing mice with HA2 protein from A/chicken/Anhui/BRI99/2016 (AH/BRI99/16, H9N2) generated by prokaryotic expression. Then, its broad-spectrum activity was analyzed by WB and IFA. Next, the minimal linear epitope was identified via analyzing the reaction of a series of HA truncations with 3C12. Finally, the protective effects of 3C12 were evaluated in vitro and in vivo infection experiments. Results The mAb could react with the viruses of subtypes H1, H2, H5, H8, H9, H12, H13, H16, and HA protein of H18 in group 1, but failed to react with viruses in group 2. The minimal linear epitope targeted by the mAb was 433NAELLVL439 in full length of HA and localized in the C-helix region of HA2 (residue 95-101, HA2 numbering). What’s more, the mAb 3C12 inhibited H1, H2, H5, H8, H9, H12, H13 and H16 virus-replication in vitro and also has shown effectiveness in preventing and treating disease in mice challenged with lethal dose of AH/BRI99/16 (H9N2) virus in vivo. These results suggested that the broadly reactive anti-HA stem mAb 3C12 exhibited prophylactic and therapeutic efficacy. Conclusions Here, we have demonstrated that the linear epitope identified in this study could be a novel target for developing broad-spectrum influenza diagnostics or vaccine design, and the HA2-based monoclonal antibody is indeed a promising strategy for broad-spectrum protection against seasonal and pandemic influenza viruses.

Purpose: The aim of this study is to measure the output factor (OF) and profile of surface dose in regular and small radiation therapy fields using Cherenkov imaging (CI). Methods: A medical linear accelerator (linac) was employed to generate radiation fields, including regular open photon field (ROPF), regular wedge photon field (RWPF), regular electron field (REF) and small photon field (SPF). The photon beams consisted of two filter modes including flattening filter (FF) and flattening filter free (FFF). All fields were delivered to a solid water phantom. Cherenkov light was captured using a charge-coupled device system during phantom irradiation. The OF and profile of surface dose measured by CI were compared with those determined by film measurement, ionization chamber measurement and treatment planning system calculation in order to examine the feasibility of measuring surface dose OF and profile using CI. Results: The discrepancy between surface dose OF measured by CI and that determined by other methods is less than 6% in ROPFs with size less than 10 × 10 cm 2 , REFs with size less than 10 × 10 cm 2 , and SPFs except for 1 × 1 cm 2 field. In the flat profile region, the discrepancy between surface dose profile measured by CI and that determined by other methods is less than 4% in REFs and less than 3% in ROPFs, RWPFs, and SPFs except for 1 × 1 cm2 field. The discrepancy of the surface dose profile is in compliance with the recommendation by IAEA TRS 430 reports. The discrepancy between field width measured by CI and that determined by film measurement is equal to or less than 2 mm, which is within the tolerance recommend by the guidelines of linac quality assurance in regular open FF photon fields, SPFs, and REFs with cone size of 10 × 10 cm 2 in area. Conclusion: CI can be used to quantitatively measure the OF and profile of surface dose. It is feasible to use CI to measure the surface dose profile and field width in regular open FF photon fields and SPFs except for 1 × 1 cm 2 field.

Silver nanoparticles (AgNPs) are widely used because of their excellent antimicrobial properties. However, the poor hemocompatibility limits the application of AgNPs in blood contact materials. General approaches to improve the hemocompatibility of AgNPs-containing surfaces are to construct barrier layers or co-immobilize anticoagulant biomolecules. But such modification strategies are often cumbersome to prepare and have limited applications. Therefore, this study proposes a simple UV-photo-functionalization strategy to improve the hemocompatibility of AgNPs. We loaded AgNPs onto titanium dioxide (TiO2) nanoparticles to form a composite nanoparticles (Ag@TiO2NPs). Then, UV treatment was performed to the Ag@TiO2NPs, utilizing the diffusible photo-induced anticoagulant properties of TiO2 nanoparticles to enhance the hemocompatibility of AgNPs. After being deposited onto the PU surface, the photo-functionalized Ag@TiO2NPs coating showed excellent antibacterial properties against both Gram-positive/Gram-negative bacteria. Besides, In vitro and ex-vivo experiments demonstrated that the photo-functionalized Ag@TiO2NPs coating had desirable hemocompatibility. This modification strategy can provide a new solution idea to improve the hemocompatibility of metal nanoparticles.

Thrombosis and infections are the two major complications associated with extracorporeal circuits and indwelling medical devices, leading to significant mortality in clinic. To address this issue, here, we report a biomimetic surface engineering strategy by the integration of mussel-inspired adhesive peptide, with bio-orthogonal click chemistry, to tailor the surface functionalities of tubing and catheters. Inspired by mussel adhesive foot protein, a bioclickable peptide mimic (DOPA)4-azide-based structure is designed and grafted on an aminated tubing robustly based on catechol-amine chemistry. Then, the dibenzylcyclooctyne (DBCO) modified nitric oxide generating species of 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) chelated copper ions and the DBCO-modified antimicrobial peptide (DBCO-AMP) are clicked onto the grafted surfaces via bio-orthogonal reaction. The combination of the robustly grafted AMP and Cu-DOTA endows the modified tubing with durable antimicrobial properties and ability in long-term catalytically generating NO from endogenous s-nitrosothiols to resist adhesion/activation of platelets, thus preventing the formation of thrombosis. Overall, this biomimetic surface engineering technology provides a promising solution for multicomponent surface functionalization and the surface bioengineering of biomedical devices with enhanced clinical performance.

Objective: Hereditary angioedema (HAE) is a rare, life-threatening autosomal dominant disorder. We aimed to investigate the prevalence of HAE in a Chinese population with a decreased Complement 4 (C4) level. Methods: All the patients present in Tongji Hospital with C4 below lower normal range were included from January 2019 to June 2020. The individual data were extracted from the database and categorized by diagnosis. Patients suspected of HAE were further evaluated by C1 inhibitor level and function test to confirm the HAE diagnosis. Results: A total of 8226 patients were enrolled in our study, among whom 18 had symptoms similar to HAE and received C1 inhibitor level and function tests. Two (1 male and 1 female) of the 18 patients were identified as HAE patients. This means the prevalence of HAE was 2.43/10 000 among the C4-decreased population and 10.1/10 000 in the C4-decreased population with etiology undetermined. The 2 HAE patients had experienced skin and oropharynx edema attack and received tracheotomy. The female patient had a family history. Laboratory tests showed significant decrease of C4 and C1 inhibitor levels in the 2 patients, both of whom were diagnosed as type 1 HAE. Conclusion: The prevalence of HAE is low in C4-decreased patients. In a large cohort, C4 level can serve as a practical indicator to screen the HAE patients, but further testing of C1 inhibitor activity and levels is needed to confirm the diagnosis of HAE.

PURPOSEWe aimed to explore the imaging findings of computed tomography (CT) in diagnosing coronavirus disease 2019 (COVID-19) and its clinical value for further evaluation of suspected cases.METHODSFiles of 155 patients visiting the fever clinics at our hospital and affiliated hospitals from January 20th to February 9th, 2020 were searched. Among them, 140 cases (including 82 males and 58 females) were included as suspected COVID-19 cases based on clinical and epidemiological history; the CT image features of 70 cases with suggestive findings on CT, confirmed by positive nucleic acid test were analyzed and evaluated. The sensitivity and specificity of CT in diagnosing COVID-19 were evaluated in patients with epidemiological history.RESULTSOf the 70 patients, 84.3% showed bilateral lung involvement on CT; 27 cases (38.6%) showed ground-glass opacity (GGO), which was mostly distributed in the subpleural area (55.7%), and this sign was mainly observed in early COVID-19 patients. In addition, 41 cases (58.6%) manifested GGO combined with focal consolidation opacity, 2 (2.8%) had flake-like consolidation opacity, with involvements of the periphery of lung field and the central zone (44.3%), and this sign was mostly observed in severe or critical patients. Concomitant signs such as pleural effusion and mediastinal lymph node enlargement were rare. Among patients with epidemiological history, the sensitivity of CT in diagnosing COVID-19 was 89.7% (70/78), and the specificity was 88.7% (55/62).CONCLUSIONCT shows high sensitivity and specificity in diagnosing COVID-19. CT is an important examination method in evaluation of suspected cases and assessment of disease severity.

Integration of two or more biomolecules with synergetic and complementary effects on a material surface can help to obtain multi-functions for various biomedical applications. However, the amounts of biomolecules integrated and their physiological functions are compromised due to the limited surface anchoring sites. Herein, we propose a novel concept of film engineering strategy “from surface to bulk synergetic modification”. This new concept is realized by employing the surface amine groups of plasma polymerized allylamine (PPAm) film for grafting a molecule e.g., thrombin inhibitor, bivalirudin (BVLD), meanwhile its bulk amine groups is used as a universal depot for storing and releasing therapeutic nitric oxide (NO) gas as supplement to the functions of BVLD. It is demonstrated that such a “from surface to bulk synergetic modification” film engineering can impart the modified-substrates with anti-platelet and anti-coagulant dual functions, giving rise to a highly endothelium-mimetic thromboresistant property. We believe that our research provides a very promising strategy to deliver multifunctional surface versatilely that require NO release in combination with other properties, which will find broad biomedical applications in blood-contacting devices, and et al. Moreover, it also provides a brand-new film engineering strategy for tailoring surface multi-functionalities of a wide range of materials. Optical image computing, such as spatial differentiation, logical transform, and addition-subtraction operation, is of great importance to signal enhancement, transform, and extraction, but such nonlinear methods are restrained in incoherent or scattering imaging. Here, the logical white-light image reconstruction, as an optical image computing method for incoherent and scattered images, is proposed by monitoring a seeded potential in variable detection planes. The weak white-light signals collectively seed a potential at the expense of scattering noise based on seeded modulation instability in a photorefractive crystal. Then the logical image operation is achieved by sensitively probing the seeded potential and recording the intensity distribution of the probe beam in different detection planes. The proposed method has a strong ability of ballistic signal recording, reproduction, and reversal. The numerical and experimental results show a new physical phenomena in which the indirect interaction between light waves through a seeded potential is presented in different forms. The nonlinear method is proved to effectively operate pure images, the images scattered by a rotating diffuser, and the images scattered by fogs whether under back lighting or under front lighting. Our work suggests an alternative method for all-optical image processing, recovery, and computing.

Abstract Background N6-Methyladenosine (m6A) modification has been implicated in many biological processes. It is important for the regulation of messenger RNA (mRNA) stability, splicing, and translation. However, its role in cancer has not been studied in detail. Here we investigated the biological role and underlying mechanism of m6A modification in hepatoblastoma (HB). Methods We used Reverse transcription quantitative real-time PCR (RT-qPCR) and Western blotting to determine the expression of m6A related factors. And we clarified the effects of these factors on HB cells using cell proliferation assay, colony formation, apoptotic assay. Then we investigated of methyltransferase-like 13 (METTL3) and its correlation with clinicopathological features and used xenograft experiment to check METTL3 effect in vivo. m6A-Seq was used to profiled m6A transcriptome-wide in hepatoblastoma tumor tissue and normal tissue. Finally, methylated RNA immunoprecipitation (MeRIP) assay, RNA remaining assay to perform the regulator mechanism of MEETL3 on the target CTNNB1 in HB. Results In this research, we discovered that m6A modifications are increased in hepatoblastoma, and METTL3 is the main factor involved with aberrant m6A modification. We also profiled m6A across the whole transcriptome in hepatoblastoma tumor tissues and normal tissues. Our findings suggest that m6A is highly expressed in hepatoblastoma tumors. Also, m6A is enriched not only around the stop codon, but also around the coding sequence (CDS) region. Gene ontology analysis indicates that m6A mRNA methylation contributes significantly to regulate the Wnt/β-catenin pathway. Reduced m6A methylation can lead to a decrease in expression and stability of the CTNNB1. Conclusion Overall our findings suggest enhanced m6A mRNA methylation as an oncogenic mechanism in hepatoblastoma, METTL3 is significantly up-regulated in HB and promotes HB development. And identify CTNNB1 as a regulator of METTL3 guided m6A modification in HB.

Abstract Fetal bovine serum (FBS) is widely used in cell cultures due to its high stability and easy access. It was also used as a substitute for porcine follicular fluid (PFF) in previous studies. However, FBS components are unclear, and the presence of FBS in culture media may introduce a variation from batch to batch. This study aimed to establish an effective method to screen FBS in place of PFF in the culture media for porcine oocytes in vitro. We screened FBS from different sources by using porcine fetal fibroblast cells. The effects of six FBS samples on porcine fetal fibroblast cell growth were tested via frozen cell survival assay, cell clone formation assay, cell growth curve, and cell passage activity assay. The best serum that we called GFBS (heat‐inactivated FBS, cat. no. 10500‐64; Gibco) showed a similar effect on the maturation and development of porcine oocytes to that of PFF and can be used as a good substitute for PFF. These results suggested that the porcine fetal fibroblast cell culture test can be used as a valuable method to screen FBS for porcine oocyte maturation and embryonic development in vitro.

Grassland restoration can play a critical role in regulating ecosystem carbon cycling, especially for changes in methane (CH4) flux. However, how grassland restoration measures affect soil CH4 flux remains poorly understood. Here, we investigated the relationships between soil CH4 flux and properties of both plant communities and soil under different grassland restoration measures (shallow ploughing, harrowing and natural recovery) in a field experiment conducted during the growing season in a semiarid grassland in northern China using the static chamber method. Soil was a CH4 sink across all three restoration measures. Compared to natural recovery, shallow ploughing significantly enhanced and harrowing significantly decreased soil CH4 uptake. The rates of soil CH4 uptake were positively associated with litter production and belowground biomass and negatively associated with plant taxonomic and phylogenetic diversity. Soil temperature, NH4+-N concentration, and soil organic matter quality had positive effects on CH4 uptake rate, but soil moisture and NO3−-N concentration had negative effects on CH4 uptake rate. Our results elucidate the pathway through which shallow ploughing facilitates soil CH4 uptake in a semiarid grassland and thus highlights its role in sustaining ecosystem multifunctionality as a measure for grassland restoration.

Predictive current control (PCC) is an advanced control strategy for permanent magnet synchronous motors (PMSM). When the motor drive system is undisturbed, predictive current control exhibits a good dynamic response speed and steady−state performance, but the conventional PCC control performance of PMSM that depends on the motor body model is vulnerable to parameter perturbation. Aiming at this problem, an improved model−free predictive current control (IMFPCC) strategy based on a high−gain disturbance observer (HGDO) is proposed in this paper. The proposed strategy is introduced with the idea of model−free control, relying only on the system input and output to build an ultra−local current prediction model, which gets rid of the constraints of the motor body parameters. In the paper, the ultra−local structure is optimized by comparing and analyzing the equation of the state of the classical ultra−local structure and PMSM system. The system’s current state variables are incorporated into the ultra−local system modeling, as a result, the current estimation errors existing in the classical ultra−local structure are eliminated. For the unmodeled and parametric perturbation part of the ultra−local system, a high−gain disturbance observer is designed to estimate it in real time. Finally, the proposed IMFPCC strategy is compared with the conventional model−based predictive current control (MPCC) and the conventional model−free predictive current control (CMFPCC) in simulation and experiment. The results show that the current steady−state error of the IMFPCC strategy in the case of parameter variation is only 50% of the MPCC method, which proves the effectiveness and correctness of the proposed strategy.

Influenza A virus (IAV) infection has traditionally been a serious problem in animal husbandry and human public health security. Recently, many studies identified that long noncoding RNAs play an important role in the antiviral immune response after the infection of the influenza virus. However, there are still lots of IAV-related lncRNAs that have not been well-characterized. Using RNA sequencing analysis, we identified a lncRNA, named Serpina3i Activation Associated lncRNA (SAAL), which can be significantly upregulated in mice after IAV infection. In this study, we found that overexpression of SAAL inhibited the replication of A/WSN/33(WSN). SAAL upregulated Serpina3i with or without WSN infection. Overexpression of Serpina3i reduced influenza virus infection. Meanwhile, knockdown of Serpina3i enhanced the replication of WSN. Furthermore, knockdown of Serpina3i abolished the SAAL-mediated decrease in WSN infection. Overexpression of SAAL or Serpina3i positively regulated the transcription of interferon β (IFN-β) and several critical ISGs after WSN infection. In conclusion, we found that the novel lncRNA SAAL is a critical anti-influenza regulator by upregulating the mRNA level of Serpina3i.

Objective: To compare the prognostic predictive performance of six lymph node (LN) staging schemes: American Joint Committee on Cancer (AJCC) N stage, number of retrieved lymph nodes (NRLN), number of positive lymph nodes (NPLN), number of negative lymph nodes (NNLN), lymph node ratio (LNR), and log odds of positive lymph nodes (LODDS) among node-positive endometrioid endometrial cancer (EEC) patients.Methods: A total of 3,533 patients diagnosed with node-positive EEC between 2010 and 2016 from the Surveillance, Epidemiology, and End Results (SEER) database were retrospectively analyzed. We applied X-tile software to identify the optimal cutoff value for different staging schemes. Univariate and multivariate Cox regression models were used to assess the relationships between different LN schemes and survival outcomes [disease-specific survival (DSS) and overall survival (OS)]. Moreover, Akaike information criterion (AIC) and Harrell concordance index (C-index) were used to evaluate the predictive performance of each scheme in both continuous and categorical patterns.Results: N stage (N1/N2) was not an independent prognostic factor for node-positive EEC patients based on multivariate analysis (DSS: p = 0.235; OS: p = 0.145). Multivariate model incorporating LNR demonstrated the most superior goodness of fit regardless of continuous or categorical pattern. Regarding discrimination power of the models, LNR outperformed other models in categorical pattern (OS: C-index = 0.735; DSS: C-index = 0.737); however, LODDS obtained the highest C-index in continuous pattern (OS: 0.736; DSS: 0.739).Conclusions: N stage (N1/N2) was unable to differentiate the prognosis for node-positive EEC patients in our study. However, LNR and LODDS schemes seemed to have a better predictive performance for these patients than other number-based LN schemes whether in DSS or OS, which revealed that LNR and LODDS should be more helpful in prognosis assessment for node-positive EEC patients than AJCC N stage.

(1) Background: The prevalence of allergic rhinitis (AR) and asthma has increased rapidly in China. However, perceptions of respiratory allergies and barriers to their management have not attracted enough attention. (2) Objective: To investigate the prevalence of, parents’ perceptions of and their unmet needs for information concerning respiratory allergies in a 3- to 16-year-old children population. (3) Methods: A cross-sectional survey was conducted from June to July 2021 in three schools in Wuhan, China. A total of 1963 participants were recruited through cluster sampling for their parents to complete an online questionnaire regarding respiratory allergic symptoms. The diagnosis of respiratory allergies was based on self-reported symptoms and face-to-face physician evaluation. All the participants with respiratory allergies were asked to complete the Brief Illness Perception Questionnaire (B-IPQ), the Asthma Knowledge Questionnaire (AKQ) and a questionnaire regarding their unmet needs for disease management. (4) Results: The prevalence of respiratory allergies was 29.3% (576/1963) in the 3- to 16-year-old population, among whom AR accounted for 25.7%; asthma, 1.8% and AR-complicated asthma (AR&Asthma), 1.9%. The total B-IPQ score was 40.2 ± 10.9 in the participants with respiratory allergies, and there were no differences among the AR, asthma and AR&Asthma groups (all p > 0.05). The B-IPQ score correlated significantly with symptom onset time and a history of atopic dermatitis (p < 0.01). Nearly one fifth, 18.9%, of the participants with respiratory allergies never went to hospital for treatment, but those with higher B-IPQ scores were more likely to seek professional treatment (p < 0.001). The accuracy rates of AKQ were 72.5% in the participants with asthma and 76.7% in those without asthma (p = 0.147). Among the 576 participants with respiratory allergies, 568 (98.6%) had tried to obtain disease-management information from online platforms, and 55.5% (315/568) were dissatisfied with current platforms; the reasons included incomprehensive contents of illness (45.7%), lack of voice from leading experts (40.3%), too many advertisements (37.5%) and similar contents on different platforms (36.8%). (5) Conclusions: The prevalence of respiratory allergies is high in the 3- to 16-years old population in Wuhan, China. Yet the parents’ perceptions of respiratory allergies and knowledge of asthma are insufficient. It is crucial to increase parents’ awareness of the illness and facilitate their access to truly informative and professional platforms.

Underwater acoustic signal separation is a key technique for underwater communications. The existing methods are mostly model-based, and cannot accurately characterize the practical underwater acoustic communication environment. They are only suitable for binary signal separation and cannot handle multivariate signal separation. However, recurrent neural networks (RNNs) show a powerful ability to extract the features of temporal sequences. Inspired by this, in this paper, we present a data-driven approach for underwater acoustic signal separation using deep learning technology. We use a bidirectional long short-term memory (Bi-LSTM) approach to explore the features of a time–frequency (T-F) mask, and propose a T-F-mask-aware Bi-LSTM for signal separation. Taking advantage of the sparseness of the T-F image, the designed Bi-LSTM network is able to extract the discriminative features for separation, which further improves the separation performance. In particular, this method breaks through the limitations of the existing methods and not only achieves good results in multivariate separation but also effectively separates signals when they are mixed with 40 dB Gaussian noise signals. The experimental results show that this method can achieve a 97% guarantee ratio (PSR), and the average similarity coefficient of the multivariate signal separation is stable above 0.8 under high noise conditions. It should be noted that our model can only handle known signals such as test signals for calibration.

Agricultural terraces, as typically artificial landforms, have maintained an important role throughout human history. Terrace ridges, which form the main constructive part of terraces, face the greatest risk from gully or gravity erosion. Mapping terrace ridges can benefit agricultural production management and soil-and-water conservation. This study presents a contour-directional detection method that automatically maps terrace ridges by considering the image features and topographic features of terraces. First, the terrace areas were extracted via object-based image analysis. Second, the Canny edge detector was employed to extract all edges from the image, and a digital elevation model (DEM) was used to generate the contour direction. The contour direction and edge image were then overlaid to implement the contour-directional detection method, which detects ridges along the contour direction. Finally, those candidate terrace ridges which lengths exceed an appropriate threshold were identified as terrace ridges. Five study areas located in the Loess Plateau of China were used to validate the proposed method. The results showed that the correctness, completeness, and quality ranged from 82.57% to 94.92%, from 82.51% to 88.39%, and from 70.28% to 84.40%, respectively. However, the proposed method is widely available and can be easily applied by using open global images and DEMs, such as Google Earth images and ASTER GDEM.

Abstract Background By combining regional homogeneity (ReHo) and functional connectivity (FC) analyses, this study aimed to explore brain functional alterations in Attenuated Psychosis Syndrome (APS), which could provide complementary information for the neurophysiological indicators for schizophrenia (SZ) associated brain dysfunction. Methods Twenty-one APS subjects and twenty healthy controls were enrolled in the data acquisition of demographics and clinical characteristics as well as structural and resting-state functional magnetic resonance imaging (rs-fMRI). ReHo analysis was conducted to determine the peak coordinate of the abnormal regional brain activity. Then, identified brain regions were considered as seed regions and were used to calculate FC between reginal brain voxels and whole brain voxels. Finally, potential correlations between imaging indices and clinical data were also explored. Results Four APS and two HC subjects were excluded because the largest dynamic translation or rotation had exceeded 2 mm / 2°. Compared with healthy controls (HCs), APS subjects exhibited higher ReHo values in the right middle temporal gyrus (MTG) and lower ReHo values in the left middle frontal gyrus (MFG), left superior frontal gyrus (SFG), left postcentral gyrus (PoCG), and left superior frontal gyrus, medial (SFGmed). Considered these areas as seed regions, the APS subjects showed abnormal enhancement in functional brain connections, predominantly in the frontal and temporal lobes. Conclusions We concluded that the APS subjects had spatially regional dysfunction and remoted synchronous dysfunction in the frontal and temporal lobes of the brain, and changes in ReHo and FC patterns may reveal the mechanism of brain dysfunctions and may serve as an imaging biomarker for the diagnosis and evaluation of SZ.

Metal-organic frameworks based on metal-formates have emerged as a intriguing class of multiferroics with wide range of applications. In this work, we present magneto-Raman spectroscopic investigations on [(CH3)2NH2]Co(HCOO)3 belonging to this family. The spectroscopic studies were performed at magnetic fields up to 31 T at the temperature of 2.3 K. It was observed that the formate bending mode at around 798 cm−1 shifts to higher energies with increasing magnetic field. The magneto-response of the phonon also exhibits anomalies at magnetic fields of 14.5 T and 23.5 T corresponding to magnetic phase transitions. Based on our results, we conclude that the formate bending mode does play a role in facilitating the saturation of magnetic states similar to its Mn and Ni analogs.
