Loss-of-function variant in the gene encoding the KCNQ4 potassium channel causes autosomal dominant nonsyndromic hearing loss (DFNA2), and no effective pharmacotherapeutics have been developed to reverse channel activity impairment. Phosphatidylinositol 4,5-bisphosphate (PIP2), an obligatory phospholipid for maintaining KCNQ channel activity, confers differential pharmacological sensitivity of channels to KCNQ openers. Through whole-exome sequencing of DFNA2 families, we identified three novel KCNQ4 variants related to diverse auditory phenotypes in the proximal C-terminus (p.Arg331Gln), the C-terminus of the S6 segment (p.Gly319Asp), and the pore region (p.Ala271_Asp272del). Potassium currents in HEK293T cells expressing each KCNQ4 variant were recorded by patch-clamp, and functional recovery by PIP2 expression or KCNQ openers was examined. In the homomeric expression setting, the three novel KCNQ4 mutant proteins lost conductance and were unresponsive to KCNQ openers or PIP2 expression. Loss of p.Arg331Gln conductance was slightly restored by a tandem concatemer channel (WT-p.R331Q), and increased PIP2 expression further increased the concatemer current to the level of the WT channel. Strikingly, an impaired homomeric p.Gly319Asp channel exhibited hyperactivity when a concatemer (WT-p.G319D), with a negative shift in the voltage dependence of activation. Correspondingly, a KCNQ inhibitor and chelation of PIP2 effectively downregulated the hyperactive WT-p.G319D concatemer channel. Conversely, the pore-region variant (p.Ala271_Asp272del) was nonrescuable under any condition. Collectively, these novel KCNQ4 variants may constitute therapeutic targets that can be manipulated by the PIP2 level and KCNQ-regulating drugs under the physiological context of heterozygous expression. A research team from South Korea led by Byung Yoon Choi from Seoul National University College of Medicine identified three new mutations in KCNQ4, a gene that encodes a potassium channel protein found in the inner ear, that can cause congenital deafness. Strikingly, the authors identified the first hyperactive KCNQ4 variant. They inserted these gene variants into human cells in culture, and found that drugs known to affect the activity of this channel protein had different effects on different mutations. Impairments caused by two of the variants were prevented by drug treatment; the third variant proved resistant to the same therapeutic approach. [Purpose] The purpose of this study was to assess changes in blood glucose level, blood pressure, and arterial stiffness after a 75 g oral glucose tolerance test (OGTT) in elderly women aged over 65 years with hypertension and either normal glycemic control, impaired fasting glucose tol erance, or diabetes mellitus. We also wished to investigate the relationship between stages of diabetes and physical fitness. [Methods] A total of 24 elderly women with hypertension were assigned to a control group (CON; n=7), impaired fasting glucose group (IFG; n=9), and diabetes mellitus group (DM; n=8). In each group, blood glucose level, bra chial ankle pulse wave velocity (PWV), and blood pressure were measured at baseline as well as 60 and 120 minutes after a 75 g OGTT. Physical fitness factors such as hand grip strength, balance test, 4 m gait speed test, chair stand test, short physical performance battery, and 6-minute walking test were subsequently assessed. [Results] In all three groups, blood glucose levels were significantly increased at 60 and 120 minutes after a 75 g OGTT. In the DM group, blood glucose levels were significantly higher before and after a 75 g OGTT than in the CON group. In the CON group, PWV was significantly increased at 60 minutes after a 75 g OGTT; however, there were no changes in other groups after glucose ingestion. In the CON group, systolic and diastolic blood pressures were significantly decreased at 60 and 120 minutes after a 75 g OGTT compared to baseline. However, there was no change in blood pressure after ingestion in the DM group. The IFG group had greater grip strength than the CON group; however, there were no differences in other variables between the groups. [Conclusion] After a 75 g OGTT, elderly women with hypertension and diabetes maintain higher blood glucose levels compared to those with hypertension alone. Un like elderly women with hypertension alone, those with hypertension and diabetes did not show changes in arterial stiffness and blood pressure after a 75 g OGTT. Therefore, elderly women with hypertension and diabetes may not be able to control their blood vessels following a 75 g OGTT due to impaired vascular endothelial function. Moreover, there was no association between diabetes stage and physical fitness in elderly women with hypertension.

PurposeThe study sought to explore how people's negative emotions change in a crisis situation when they get to know about the crisis and the corporate's socially responsible activities after crisis.Design/methodology/approachA 2 (crisis type: human error vs organization misdeed) × 2 (CSR fit: low vs high) × 3 (motive disclosure: no disclosure vs company-oriented disclosure vs society-oriented disclosure) between-subjects experiment was conducted online.FindingsMore anger was elicited toward organizational misdeed than human error from both within-person and between-persons perspectives. When using CSR as postcrisis strategy, within-person analyses revealed that high CSR fit in message helped to attenuate sadness (and potentially anger) to a greater extent than low CSR fit, whereas between-persons analyses did not find significant effects of either CSR fit or motive disclosure.Originality/valueOur findings demonstrate that situational dynamics in crisis situation constantly influence people's emotional states, suggesting a vertical investigation (e.g. within-individual analysis) of emotions may help both scholars and practitioners better understand the nature of crisis emotions and provide fresh insights on how to cope with them.

Airborne pollutants have detrimental effect on the human body and the environment. Diesel exhaust particles (DEPs) are known to be major component of particulate matter (PM) and cause respiratory diseases and neurotoxicity. However, the effects of air pollutants on the sensory nervous system, especially on the olfactory sense, have not been well studied. Herein, we aimed to explore DEP-induced changes in the olfactory perception process. Olfactory sensitivity test was performed after DEP inhalation in mice. Microarray was conducted to determine the differentially expressed genes, which were then utilized to build a network focused on neurotoxicity. Exposure to DEPs significantly reduced sniffing in mice, indicating a disturbance in the olfactory perception process. Through network analysis, we proposed five genes (Cfap69, Cyp26b1, Il1b, Il6, and Synpr) as biomarker candidates for DEP-mediated olfactory dysfunction. Changes in their expression might provoke malfunction of sensory transduction by inhibiting olfactory receptors, neurite outgrowth, and axonal guidance as well as lead to failure of recovery from neuroinflammatory damage through inhibition of nerve regeneration. Thus, we suggest the potential mechanism underlying DEPs-mediated olfactory disorders using genomic approach. Our study will be helpful to future researchers to assess an individual’s olfactory vulnerability following exposure to inhalational environmental hazards.

64. Total energy expenditure measured by doubly labeled water method in children and adolescents: a systematic review

Total energy expenditure (TEE) is essential for understanding children and adolescents' growth, development, and physical activities. This study aimed to summarize the existing evidence on TEE measured using the doubly labeled water (DLW) technique in children and adolescents aged 1-18 years. Furthermore, this review compared TEE between participants with obesity and normal weight. This systematic review was conducted using PubMed, ScienceDirect, Web of Science, and EBSCO. The studies were limited to those published in English between January 2000 and December 2021. Articles presenting objectively measured data on the TEE of children and adolescents aged 1-18 years, measured using the DLW method, were included. The physical activity level (PAL; TEE/basal metabolic rate [BMR]) and BMR data were also obtained. The search strategy identified 2,351 articles, of which 63 (n = 4,283 children and adolescents; 45.4% male) met the selection criteria. The participants in the 10 studies were overweight and obese (n = 413). In our results, TEE increased in men and women aged 1-18 years. PAL increased with age in males (y = 0.0272x + 1.3887, r2 = 0.511) and females (y = 0.0199x + 1.401, r2 = 0.335) and the slope of PAL with age did not show a difference between males and females. The TEE of participants with obesity and overweight was higher than that of participants with normal weight, but the slope of TEE did not show a difference between normal weight (y = 132.99x + 702.24, r2 = 0.877) and obesity (y = 136.18x + 1,037.9, r2 = 0.842). In conclusion, this review summarizes convincing evidence that daily TEE progressively increases with advancing growth in males and females aged 1-18 years.

Background There are no standard diagnostic criteria or interventions for internet gaming addiction (IGA) even though IGA is one of the most pervasive public health issues among youth worldwide. Internet gaming reasons or motivations have been studied as a potential predictor of IGA, but the results have been inconsistent and biological indicators of gaming reasons have rarely been studied. We sought to (1) identify categories of internet gaming reasons, (2) examine the relationship of gaming reasons to risk of IGA, and (3) describe biological indicators associated with reasons for gaming. Methods We used a multi-phase cross-sectional design including individual interviews; focus group discussion; and descriptive, comparative analysis. Fifteen Korean adolescent male internet gamers participated in individual interviews and eight participated in a focus group aimed at identifying reasons for internet gaming. Using the identified gaming reasons from these sources we surveyed 225 adolescent game users using a self-report questionnaire. Participants provided blood samples for assessment of norepinephrine (NE) and serum cortisol. Results We identified four major categories of internet gaming reasons: entertainment, getting along with friends, stress relief, and habitual gaming. The habitual group showed significantly greater risk of IGA than the other groups (p p = .035), possibly indicating an alteration in autonomic function. Conclusion Health care providers are encouraged to screen adolescents for excessive internet gaming and to intervene with those who report habitual gaming behaviors. When feasible, assessment of biological indicators, such as plasma NE, may help to identify youth at greatest risk of IGA.

[Purpose] The purpose of this study was to investigate the effects of short-term creatine intake on muscle fatigue induced by resistance exercise in healthy adolescent men, i.e., lactic acid concentration and wrist and head tremor measured by an accelerometer. [Methods] Twelve healthy adolescent men who had no experience with creatine intake were included. The subjects were randomly assigned to the creatine group and the placebo group, followed by 5 days of creatine and placebo intake, and 5 times of 5 sets of leg press, leg extension, bench press, and arm curl exercises at 70% repetition maximum (RM). The lactic acid concentration before and after exercising, rate of perceived exertion (RPE), and accelerometer-based wrist tremor and head tremor during exercise were measured. Subsequently, after 7 days to allow for creatine washout, the same exercise treatment and measurement were performed in each group after switching drug and placebo between the groups. Objectives: Diesel exhaust particles (DEP)s are notorious ambient pollutants composed of a complex mixture of a carbon core and diverse chemical irritants. Several studies have demonstrated significant relationships between DEP exposure and serious nasal inflammatory response in vitro, but available information regarding underlying networks in terms of gene expression changes has not sufficiently explained potential mechanisms of DEP-induced nasal damage, especially in vivo. Methods: In the present study, we identified DEP-induced gene expression profiles under short-term and long-term exposure, and identified signaling pathways based on microarray data for understanding effects of DEP exposure in the mouse nasal cavity. Results: Alteration in gene expression due to DEP exposure provokes an imbalance of the immune system via dysregulated inflammatory markers, predicted to disrupt protective responses against harmful exogenous substances in the body. Several candidate markers were identified after validation using qRT-PCR, including S100A9, CAMP, IL20, and S100A8. Conclusions: Although further mechanistic studies are required for verifying the utility of the potential biomarkers suggested by the present study, our in vivo results may provide meaningful suggestions for understanding the complex cellular signaling pathways involved in DEP-induced nasal damages.

ImportanceA bayesian network meta-analysis (NMA) can help compare the various types of multifocal and monofocal intraocular lenses (IOLs) used in clinical practice.ObjectiveTo compare outcomes of presbyopia-correcting IOLs frequently recommended in clinical practice through a bayesian NMA based on a systematic review.Data SourcesMedline (PubMed) and the Cochrane Central Register of Controlled Trials (CENTRAL) were searched on May 15, 2021, from inception.Study SelectionBased on the research question, randomized clinical trials assessing multifocal IOLs in patients who underwent bilateral cataract extraction were searched. Nonrandomized studies, studies in patients with unilateral or contralateral cataract extractions, duplicated studies, conference abstracts, and nonpeer-reviewed articles were excluded.Data Extraction and SynthesisDescriptive statistics and outcomes were extracted. The NMA was conducted to compare different types of IOLs. The mean differences for continuous variables, odds ratios for binary variables, 95% credible intervals (CrIs), and ranks of interventions were estimated.Main Outcomes and MeasuresThe outcomes examined included binocular visual acuities by distance and optical quality, including glare, halos, and spectacle independence.ResultsThis NMA included 27 studies comprising 2605 patients. For uncorrected near visual acuity, trifocal IOLs (mean difference, −0.32 [95% CrI, −0.46 to −0.19]) and old bifocal diffractive IOLs (mean difference, −0.33 [95% CrI, −0.50 to −0.14]) afforded better visual acuity than monofocal IOLs. Regarding uncorrected intermediate visual acuity, extended depth-of-focus IOLs provided better visual acuity than monofocal IOLs. However, there were no differences between extended depth-of-focus and trifocal diffractive IOLs in pairwise comparisons. For uncorrected distant visual acuity, all multifocal IOLs were comparable with monofocal IOLs. There were no statistical differences between multifocal and monofocal IOLs regarding contrast sensitivity, glare, or halos.Conclusions and RelevanceFor patients considering a multifocal IOL due to presbyopia, bilateral implantation of a trifocal IOL might be an optimal option for patients without compromising distant visual acuity.

To investigate the association of orthostatic hypotension (OH) with health-related quality of life (HRQoL) in older people living in the community. A cross-sectional design was used. A total of 217 participants aged 65 and older were classified as having OH if their systolic or diastolic blood pressure showed a drop of ≥ 20 mmHg systolic blood pressure or ≥ 10 mmHg diastolic blood pressure, respectively, within 3 min of standing. Participants provided demographic and medical information and responded to questionnaires about their HRQoL (EuroQoL-5D-3L), as well as depression, anxiety, cognitive function, and recent physical activities. Tight junctions (TJs) in the epithelium play a critical role in the formation of a paracellular epithelial barrier against the extracellular environment. Diesel exhaust particles (DEPs) disrupt the epithelial barrier. The aim of this study was to investigate how DEPs disrupt the epithelial barrier and whether Toll-like receptor 4 (TLR4) is involved in DEP-induced epithelial barrier dysfunction in primary human nasal epithelial (PHNE) cells.PHNE cells were cultured at an air-liquid interface (ALI) to create a fully differentiated in vivo-like model of the epithelium and then exposed to DEPs (particulate matter4 μm) or lipopolysaccharide (LPS) alone (mono-exposure) and DEPs plus LPS (co-exposure) at the apical side of the PHNE. TJ formation and integrity were monitored by measuring transepithelial electric resistance (TEER) and fluorescently labeled dextran permeability. The expression of TJ proteins was assessed by confocal microscopy and a biochemical assay.PHNE cell viability was reduced in a time- and dose-dependent manner following DEP exposure. TEER was significantly decreased at ALI day 20 but not at day 12 following DEP exposure. The dextran permeability of the PHNE was significantly increased at both ALI day 12 and day 20 following DEP exposure. The increased dextran permeability recovered to that of the control following co-exposure to DEPs plus LPS. In the presence of DEPs, the membrane expression of myosin light chain kinase (MLCK) was dramatically increased, and the expression of occludin, ZO1, claudin-1, and E-cadherin was significantly decreased. Co-exposure to DEPs plus LPS significantly reduced membrane MLCK, claudin-1, and E-cadherin but increased occludin and ZO1 expression at ALI day 12.The activation of TLR4 by LPS inhibits MLCK trafficking to the plasma membrane, and this increased during DEP exposure, resulting in increased occludin expression at the plasma membrane that partially recovered TJ barrier dysfunction following DEP exposure.

This study aims to provide roadmaps for the sustainable development of South Korea's energy system. To this end, this study developed transition scenarios toward renewable energy for both supply and demand. We use “Long-range Energy Alternatives Planning system (LEAP)” Model and a backcasting approach to illustrate pathways to achieve the goals set forth in each of the scenarios. The four scenarios developed are Business-As-Usual (BAU), Moderate Transition Scenario (MTS), Advanced Transition Scenario (ATS), and Visionary Transition Scenario (VTS). The scenarios are defined according to the differences in the level of final energy consumption and renewable energy share by 2050. Analysis shows an increase in the percentage of renewable energy and a decrease in energy demand lead to improved energy security, more jobs in the electricity generation sector, and a decrease in greenhouse gas emissions. The pathways represented by these scenarios can be regarded as strategic targets for the sustainable development of South Korea's energy system. A set of comprehensive and long-term policies will be needed to attain the goals set forth in the scenarios.

For minimizing patient inconvenience caused by intraoral sensor discomfort and unnecessary radiational exposure of diagnosis of dental caries, we developed a new system that insert the x-ray source tube (diameter: 3 mm, tube voltage: ~ 55 kV, tube current: ~ 300 uA) inside of the oral cavity and placed the the radiation acquisition device outside. The purpose of this study was to investigate the optimal acquisition conditions for the diagnosis of dental caries and to evaluate the image performance of the new intraoral x-ray imaging system. The 33 model sets were made with 3 teeth were randomly selected from 99 extracted human teeth, total of 5 conventional radiographic sets as control groups and 1 experimental set were prepared after cavity forming the 4 sites of artificial proximal caries(grade 1 to 4). Total 198 images were obtained and those were read by seven dentists of the grades and the correct answer rates were calculated and analyzed by one - way ANOVA. The new x-ray images were shown that there is no significant difference in reading for the diagnosis of proximal caries. Therefore the new x-ray system might be considerable for using in the clinical dental caries diagnosis, especially initial caries stage.

Seven pairs of enantiomeric isoflavones (1a/1b–7a/7b) were obtained from the ethyl acetate extract of the fruits of Maclura tricuspidata (syn. Cudrania tricuspidata), and successfully separated by chiral high-pressure liquid chromatography (HPLC). The structures and absolute configurations of the enantiomeric isoflavones were established on the basic of comprehensive spectroscopic analyses and quantum chemical calculation methods. Compounds 1, 1a, and 1b exhibited neuroprotective activities against oxygen-glucose deprivation/reoxygenation (ODG/R)-induced SH-SY5Y cells death with EC50 values of 5.5 µM, 4.0 µM, and 10.0 µM, respectively. Furthermore, 1, 1a, and 1b inhibited OGD/R-induced reactive oxygen species generation in SH-5Y5Y cells with IC50 values of 6.9 µM, 4.5 µM, and 9.5 µM, respectively.

Young adults with type 1 diabetes mellitus (T1DM) often exhibit poor treatment adherence and suboptimal glucose control. Health-related stigma may adversely influence diabetes self-management behavior. The aim of the current study was to explore health-related stigma among young adults with T1DM using qualitative descriptive methods in focus groups. Data were collected from a purposive sample of 14 young adults ages 20 to 34 with T1DM. Using content analysis, transcripts from two focus groups were analyzed, and five main themes were identified: (a) Desire to be Seen as a Person, Not a Disease ; (b) Wanting to be “Normal” ; (c) Feeling Ashamed Managing Diabetes in Public ; (d) Struggling to Overcome Anger and Distress ; and (e) Feeling Distrusted by Others to Manage Their Condition . Young adults with T1DM reported experiencing a high degree of stigma in their daily lives, which negatively influenced their self-management and psychological well-being. Stigma led to delaying blood glucose monitoring and insulin dosing and feelings of anger and distress. 84. A Literature Review of Gustatory Over-Responsivity Among the Population with Autism Spectrum Disorders

- Abstract
Introduction Atypical response to sensory stimuli is common in the autism spectrum disorder (ASD) population. While sensory over-responsivity in the gustatory domain (GOR) among ASD has been frequently reported, previous reviews were not solely focused on GOR in the ASD population. This review sheds light on pertinent sensory functions in ASD, differences in GOR between those with and without ASD, and concepts related to GOR in ASD. Results Neurobiological mechanisms in GOR were less studied in the population with ASD. Also, it is not clear if GOR is from difficulties of differentiation, detection threshold, and multisensory integration. Among thirty-six studies using behavioral questionnaires, a greater proportion of ASD (36-72%) had GOR compared to typically developing (TD, 20%) individuals. Furthermore, the ASD population repeatedly showed severe GOR scores relative to the TD cohort. Brain imaging studies suggested aberrant neural responses to oral stimuli as a contributing factor. It is posited that GOR contributes to food selectivity and poor dental care among ASDs. Poor nutritional outcomes among ASDs are controversial. However, severe GOR and food selectivity in reported cases present life-threatening events like pulmonary hypertension mediated by vitamin deficiencies. In addition, being underweight or overweight was associated with GOR in youths with ASD, raising a chronic health concern for general health and well-being. Another emerging consideration is the role of gut microbiome disruption in individuals with ASD and its relationship with GOR. Finally, the ASD population showed more common gastrointestinal problems than their TD counterparts, implying that GOR could be a consequence of anxiety from prior gastrointestinal experiences. Conclusion To provide families with ASD with an environment that promotes healthy development and growth, we need further research, development of clinical management, and policy changes. Thus, we recommend the targeted study of the neural mechanisms, microbiome consequences, and evidence-based intervention for GOR in ASDs.

Novel metal–organic framework-alginate composite beads (CA-MIL-53-AC) were synthesized using sodium alginate (gelling agent) and calcium chloride (complexing agent) via a one-pot and dropping method to selectively remove the dichlorodiphenyltrichloroethane (DDT), arsenate (As(V)), and fluoride in an aqueous solution. To evaluate the characteristics of the synthesized composite beads, scanning electron microscopy, X-ray diffraction, Fourier-transform infrared, X-ray photoelectron spectroscopy, thermal gravimetric analysis, zeta potential, and porosimetry techniques were used in this study. The adsorption kinetics showed that DDT, As(V), and fluoride removal could be described by the pseudo-first-order model. Also, the adsorption isotherms indicated that DDT and As(V)/fluoride removal followed the Langmuir and Freundlich models, respectively. Compared with granular AC and pristine CA, CA-AC, and CA-MIL-53, the CA-MIL-53-AC showed superior adsorption performance for all target contaminants (5.29, 4.89, and 3.18 mg/g for DDT, As(V), and fluoride, respectively). The adsorptive behavior for DDT was spontaneous, exothermic, and highly favorable based on thermodynamics calculations. Furthermore, the regenerated composite beads showed high removal capacity for DDT after the third recycle. The continuous DDT adsorption in the fixed-bed column at different flow rates were well fitted using Thomas, Adams–Bohart, and Yoon–Nelson models. The plausible mechanisms for the DDT, As(V), and fluoride removal by CA-MIL-53-AC were identified as hydrophobic and π–π interactions, ion exchange, surface complexation, and hydrogen bonding. These findings provide significant insight to remove DDT, As(V), and fluoride from water for practical applications.

In this study, we aimed to identify signaling alteration caused by exposure to diesel exhaust particles (DEPs) using primary human nasal epithelial cells (PHNECs). Global gene expression profiles in PHNECs following 50 and 200 μg/ml of DEP exposure were identified using microarray analysis. To cover the limitation of array-based mRNA expression analysis, text-mining-based software was used to analyze the integrative biological networks and relevant disease-focused functions among identified DEP-responsive genes. The confidence was valued based on the connectivity between the analyzed pathway and marker candidates. Through a literature-based pathway analysis, the stimulation of inflammation- and immune response-related processes mediated by TNF were predicted as major signaling alterations in PHNECs caused by DEP exposure. CSF3, CXCL8, MMP1, and VEGFA were identified as key hub genes in the predicted pathway. Significant expression level changes in the five key genes following DEP exposure were validated in terms of protein and mRNA expression. Although further studies are required, our toxicogenomic investigation provides key clues to the exact mechanism underlying DEP-induced nasal inflammatory damage. It also suggests an efficient approach for other research on adverse effects occurring in the upper respiratory tract following DEP exposure.

The active compounds of medicinal plants vary in composition and content depending on environmental factors, such as light, temperature, and soil. According to the Korean Pharmacopoeia standards for herbal medicine, the sum of nodakenin, decursin, and decursinolangelate, which are the marker components of Korean Angelica, should be at least 6.0 g/100 g. However, the content of the components in Korean Angelica cultivated in South Korea often fall below 6.0 g/100 g, due to weather conditions and cultivation site characteristics. This study aimed to gather information about environmental factors that affect the root growth and the content of active compounds. In total, 18 cultivation sites in Pyeongchang, Jecheon, and Bonghwa regions in Korea were investigated for this study. Environmental factors, such as the monthly mean temperature, mean relative humidity, duration of sunshine, total precipitation, soil acidity, and the characteristics of soil nutrient, were investigated over the growing season from April to October 2017. As for the growth characteristics, the dry weight of roots of Korean Angelica was measured. The sum of the contents of the three active compounds was 5.3&ndash, 7.0 g/100 g and the nodakenin content was 0.3&ndash, 1.3 g/100 g in the cultivation sites. This study concludes that the root yields in the cultivation sites would be improved if weather conditions are maintained with similar levels as those in their natural habitats. Additionally, the environment that improves root growth did not increase the content of active compounds, however, when there was a lot of gravel or high temperatures during the growth period, the content of active compounds was relatively high.

Dramatic increases in the release of anthropogenic CO2 and global temperatures have resulted in alterations to seawater carbonate chemistry and metabolisms of marine organisms. There has been recent interest in the effects of these stressors on crustose coralline algae (CCA) because photosynthesis and calcification are influenced by all components of carbonate chemistry. To examine this, a mesocosm experiment was conducted to evaluate photosynthesis, calcification and growth in the temperate CCA Chamberlainium sp. under acidification (doubled CO2), warming (+5 °C), and greenhouse (doubled CO2 and +5 °C) conditions compared to present-day conditions. After 47 days of acclimation to these conditions, productivity was lowest under acidification, although photochemical properties were improved, while respiration was highest under warming. Likewise, growth was lowest under acidification, but this negative response was offset by elevated temperature under greenhouse. Together, these results suggest that warming offsets the negative effects of acidification by creating more suitable conditions for photosynthesis and growth.

Two new compounds, including a nor-pimarane diterpenoid (continentanol, 1) and a phenolic derivative (aralianic acid, 2), along with the known diterpenoids (3–11), polyacetylenes (12–15), phenolic components (16–28), and phytosterols (29 and 30), were isolated from roots of Aralia continentalis. The structures of the new compounds were established by spectroscopic data interpretation, particularly HRESIMS, 1 D and 2 D NMR data including HSQC and HMBC. Also, those of the known compounds were identified by spectral comparison with those of the reported values.

Background Androgen hormone levels are strongly associated with obesity in adult mammals, especially with advanced age. We investigated androgen receptor inhibition on fat metabolism and long-chain fatty acid (LCFA) transport proteins in skeletal muscle during exercise. Methods Male ICR mice were randomly divided into three groups: CON (control), EX (exercise), and EXIN (exercise + androgen receptor inhibition). EX and EXIN groups were trained on a treadmill five times a week. After 4 weeks, the fat metabolism of each group was measured using open-circuit calorimetry during 1 hour of exercise. After the metabolism measurement, the expression levels of LCFA transport proteins (FAT/CD36, CPTI) were analyzed in skeletal muscle. Results Weight gain and final body weight were significantly lower in the EX group than in either the CON or EXIN groups. Conversely, food intake was significantly higher in the EX group than it was in the CON and EXIN groups. The total weight (CON; 2.07 ± 0.6, EX; 1.64 ± 0.2, EXIN; 1.95 ± 0.2) of the abdominal adipose tissue were significantly lower in the EX group than in the CON and EXIN groups (P P P P P P Conclusion Exercise training increases the expression of LCFA transport proteins (FAT/CD36, CPTI). Blocking androgen receptors can decreases the expression of CPTI in the skeletal muscle, which reduces fat metabolism. Thus, reducing sex hormones or suppressing the sensitivity of AR receptors can inhibit energy efficiency and fat metabolism by suppressing CPTI.

BackgroundThe prevalence of overactive bladder syndrome (OAB) increases with age. Sleep disturbances in elderly individuals with OAB is a common problem. The purpose of this study was to examine the effects of a biofeedback-based sleep improvement (BBSI) program on urinary symptoms and sleep patterns in elderly Korean women with OAB.MethodsA non-equivalent control group pre−/post-test design was used. Elderly women with OAB were assigned to an intervention group (n = 20) or a control group (n = 18). The BBSI program was implemented in the intervention group for 12 weeks, while two educational sessions of general sleep hygiene and lifestyle modification were provided to the control group. Using SPSS 23.0, the data were analyzed by descriptive analysis using the chi-square test, Fisher’s exact test, Mann-Whitney test, and Wilcoxon test.ResultsAfter the 12-week BBSI program, significant improvements were found in the intervention group’s the square root of the mean squared differences of successive R-R intervals (p = 0.025), low frequency/high frequency ratio (p = 0.006), and epinephrine (p = 0.039). We also observed a significant difference in urinary symptoms, sleep efficiency, wake after sleep onset, number of awakenings, and number of awakenings within 3 h after sleep onset (p p = 0.004,p = 0.001,p = 0.001, andp = 0.048, respectively). However, no significant changes were found in these variables in the control group.ConclusionsThe BBSI program effectively improved urinary symptoms and sleep patterns of elderly Korean women with OAB. Further longitudinal research is required to investigate the sustainability and effects of the BBSI program.Trial registrationKCT0003882. Date of registration: 02/05/2019. Retrospectively registered.

Cybersecurity breaches have rapidly become a high-impact crisis for many corporations. Thus, it is critically important for corporations to know how to protect their reputation through effective crisis communication. Considering the scarcity of empirical research on cybersecurity breaches in crisis communication, the current study attempts to fill this research gap. This study compared 108 official statements issued by organizations in the United States and South Korea when cybersecurity breaches threatened the reputations of various corporations. The characteristics of an apology (responsibility admittance, sympathetic expression, reassurance, compensation) and other features of crisis response (use of excuses, functions of apology, and organizational representation) were examined. This study found that the features of the official statements differed by cultural dimension (individualism vs. collectivism, small vs. large power distance) and by communication style (low-context vs. high-context communication).

[Purpose] Brown adipose tissue (BAT) plays an important role in metabolizing different substances, including androgens. The aim of this study was to determine whether a single bout of aerobic exercise would increase the androgen hormone concentration in mouse BAT and whether its increase was associated with uncoupling protein-1 (UCP-1), protein kinase A (PKA)-related mechanism in BAT. [Methods] Twenty, 9-week-old ICR adult male micewere randomly divided into three groups: Control (n=6, CON), Exercise (n=7, EX), and Exercise + SRD5A1A2 inhibitor (n=7, EXIN). SRD5A1A2 is an enzyme needed when free testosterone is metabolized to dihydrotestosterone (DHT). SRD5A1A2 was administered intraperitoneally in the EXIN group, while the CON and EX groups were treated with the vehicle only. One hour later, exercise was performed at 60–70% V˙O2max for 30minutes. The levels of testosterone and DHT in BAT were determined by ELISA, and UCP-1 mRNA level was examined by RT-PCR. UCP-1 and PKA protein levels were determined by western blotting. [Results] After a single period of exercise, testosterone and DHT concentrations in BAT were significantly higher in EX than those in CON, and lower in EXIN than those in EX. The ratio of phosphorylated PKA to total PKA in BAT was significantly higher in EX than that in CON, and lower in EXIN than that in EX. UCP-1 levels in BAT were not different in the three groups. [Conclusion] Aerobic exercise increased bioactive androgen hormone levels in BAT in association with the increase in phosphorylated PKA levels. In contrast, 30minutes of treadmill exercise did not affect UCP-1 expression.

A 64-word-line-stacked 512-Gb 3-b/cell 3-D NAND flash memory is presented. After briefly examining the challenges that occur to a stack, several technologies are suggested to resolve the issues. For performance enhancement, a novel program method hiding two-page data loading time is presented. This paper also discusses an electrical annealing improving reliability characteristic by removing holes in shallow traps. In addition, a valley tracking read for reducing timing overhead at a read retry is introduced by fast finding optimal read levels. Finally, a high-speed self-test mode for IO operation is presented. The chip, designed with the fourth generation of V-NAND technology, achieved an areal density of 3.98 Gb/mm2 and operated up to 1 Gb/s at 1.2 V.

[Purpose] Dihydrotestosterone (DHT) plays an important role in various processes that utilize energy, including fat and carbohydrate oxidation. The purpose of this study was to determine the effect of inhibiting DHT formation during mid-intensity running exercise on energy expenditure and fat and carbohydrate oxidation in the whole body using a calorimetric chamber. [Methods] Twelve ICR adult male mice, 9 weeks of age, were randomized into two groups: CON (n = 6, no treatment with exercise) and CONIN (n = 6, DHT inhibitor treatment with exercise, SRD5A1A2 is an enzyme involved in the metabolism of free testosterone into DHT). Inhibitor was administered to the CONIN group intraperitoneally, while the CON group was treated with vehicle (corn oil 2 mg/kg). After 3 days of administration of the inhibitor or vehicle, exercise was performed at 60-70% VO2max for 30 min on a treadmill in a calorimetric chamber. The O2 uptake, CO2 production, carbohydrate and fat oxidation, and respiratory exchange ratio (RER) during 30-min exercise were measured using a calorimeter. [Results] During a single bout of exercise, the CONIN group showed a significantly higher area under the curve (AUC) of O2 uptake and CO2 production from 20 min into exercise than the CON group (p < 0.001). The CONIN group showed a significantly higher AUC for carbohydrate oxidation from 20 min into exercise than the CON group (p < 0.001), whereas no difference was found in fat oxidation between groups (p = 0.067). The CONIN group had a significantly higher AUC of RER from 20 min into exercise than the CON group (p < 0.001). [Conclusion] We observed increased energy consumption at the later phase of 30-min moderate-intensity treadmill running when DHT production was inhibited. Furthermore, when DHT production during exercise was inhibited, whole-body fat utilization was inhibited and carbohydrate oxidation was substantially increased at the later phase of exercise compared to in the control group. Therefore, changes in DHT concentration in the body during exercise may be involved in whole-body fat utilization, suggesting that DHT may be an important factor affecting endurance exercise capacity.
