Despite the federal emphasis on achieving timely permanency for children who enter foster care, some children continue to remain in foster care for extended periods, with approximately 20,000 children a year “aging out” of foster care into adulthood without having achieved permanency in a family setting (Dworsky, 2008). This literature review focuses on the federal Child and Family Services Review measure that measures agency performance related to achieving permanency for children in foster care for long periods of time. The review summarizes the factors associated with timely discharges from care; while each child and family involved with the child welfare system brings a set of unique challenges and characteristics, research points to common individual and system factors that may be associated with long stays in foster care. Broad strategies that may be linked to achieving permanency are described, followed by explicit models developed to increase the likelihood of timely permanency. [ABSTRACT FROM PUBLISHER]

In the context of child welfare, family reunification refers to the services that are provided for purposes of returning children who have been placed in out-of-home care to their families of origin. Family reunification is the primary permanency goal for the majority of children who have been placed temporarily outside of their homes. This literature review focuses on the federal Child and Family Services Review composite outcome indicator that seeks to measure agency performance related to family reunification. It describes factors at the level of the agency/system, the family, and the child that are associated with failure to reunify. Practices that have been linked with successful family reunification are described, with particular attention to the period between removal from the home and reunification. We report on a gain-coefficient switched Alexandrite laser. An electro-optic modulator is used to switch between high and low gain states by making use of the polarization dependent gain of Alexandrite. Foil and vibrating wire strain gauges have an optimum strain measurement accuracy of one microstrain. However, they can only provide discrete strain readings over a single fixed-gauge length. Digital image correlation (DIC) offers an alternative to conventional strain gauges because a two-dimensional (2D) surface strain field can be obtained from a single sensor (camera). However, the benefits of 2D strain measurements are only worthwhile if a similar level of measurement accuracy to conventional strain gauges can be achieved. This paper presents the results of an investigation into the optimum strain measurement accuracy that can be achieved by using the 2D technique on artificial images (which eliminate errors associated with cameras and lighting). The principle of the 2D DIC technique and its historical development will be introduced. Then, three potential techniques for taking strain measurements will be presented and compared: single readings, averaged linear readings, and an approach on the basis of Mohr's circle. The Mohr's circle approach was found to be the most accurate and was not susceptible to image misalignment. Strain measurement accuracy was also found to be affected by the bias error of the subpixel interpolation scheme, but the use of an 8 coefficient B-spline was found to produce satisfactory results within the error of conventional strain gauges. Gauge length was also found to have a significant effect on strain measurement accuracy, indicating that measuring strains in a material in which there are variations across the strain field could result in a loss of measurement accuracy. This study analyzes the crashes that occur at midblock called “midblock crashes” in an urban arterial road. The association of midblock crashes with various factors was examined using the 7-year (2000–2006) crash data on a section of a divided arterial road in Windsor, Ontario, Canada. To account for difference in traffic volume and road geometric factors between two directions of travel in a divided road, the data were collected for two directions separately. The results of log-linear models using these bidirectional data show that midblock crashes are more likely to occur on the road sections with access points and high percentage of trucks (>20%). The results of logistic regression models show that median opening, driver age/gender, lighting, time of day, and day of week are associated with different types of crashes classified by the vehicles involved in crashes. The study shows the importance of analyzing midblock crashes using the bidirectional data by vehicle type in urban divided arterial roads with high truck volume. [ABSTRACT FROM AUTHOR]

Building on the literature related to evidence-based practice, knowledge management, and learning organizations, this cross-case analysis presents twelve works-in-progress in ten local public human service organizations seeking to develop their own knowledge sharing systems. The data for this cross-case analysis can be found in the various contributions to this Special Issue. The findings feature the developmental aspects of building a learning organization that include knowledge sharing systems featuring transparency, self-assessment, and dissemination and utilization. Background: Quality of attachment relations between adolescents and their parents and peers may be related to early maladaptive schemas as well as to symptoms of depression. Aims: The aim of the current study was to assess whether schema domains and maladaptive schemas mediate the relation between indices of quality of attachment relationships with parents and peers and symptoms of depression in non-clinical adolescents (N = 222). Method: A battery of questionnaires was completed, including measures of quality of attachment relations, maladaptive schemas, and depression. Results: The schema domains of disconnection and rejection and other-directedness mediated the relation between quality of attachment relationships and symptoms of depression. More precisely, the schemas’ mistrust/abuse and social isolation mediated the relation between trust in parents and depressive symptoms, whereas the schemas’ social isolation and self-sacrifice mediated the relation between alienation from peers and symptoms of depression. Conclusions: The mediation analyses suggest that treatment of adolescent depression could be focused on both attachment bonds and on changing maladaptive schemas. [ABSTRACT FROM PUBLISHER]

This study investigates the effects of the traffic flow parameters related to individual drivers' lane-changing and car-following behavior on the occurrence of sideswipe and rear-end crashes on freeways. A total of 184 sideswipe and 605 rear-end crashes on I-4 freeway in Orlando, Florida, with the loop detector data were used for the analysis. A set of the Bayesian logistic regression models were developed to estimate the likelihood of crashes in a specific lane compared to the adjacent lanes using the 5 min average and lane-by-lane traffic flow parameters. The analysis results showed that the significant traffic flow parameters affecting crash occurrence in a specific lane are distinctively different between sideswipe and rear-end crashes. The flow-related variables were significant in the sideswipe crash models whereas the speed-related variables were significant in the rear-end crash models. The results suggest that the lane-by-lane traffic flow parameters are considered as surrogate measures of lane-changing and car-following behavior that are associated with the occurrence of sideswipe and rear-end crashes. These parameters can potentially be used for real-time monitoring of the likelihood of crash occurrence by lane on instrumented freeways. [ABSTRACT FROM AUTHOR]

Plant-parasitic cyst nematodes penetrate plant roots and transform cells near the vasculature into specialized feeding sites called syncytia. Syncytia form by incorporating neighboring cells into a single fused cell by cell wall dissolution. This process is initiated via injection of esophageal gland cell effector proteins from the nematode stylet into the host cell. Once inside the cell, these proteins may interact with host proteins that regulate the phytohormone auxin, as cellular concentrations of auxin increase in developing syncytia. Soybean cyst nematode (Heterodera glycines) Hgl9C07 is a novel effector protein expressed specifically in the dorsal gland cell during nematode parasitism. Here, we describe its ortholog in the beet cyst nematode (Heterodera schachtii), Hsl9C07. We demonstrate that Hsl9C07 interacts with the Arabidopsis (Arabidopsis thaliana) auxin influx transporter LAX3. LAX3 is expressed in cells overlying lateral root primordia, providing auxin signaling that triggers the expression of cell wall-modifying enzymes, allowing lateral roots to emerge. We found that LAX3 and polygalacturonase, a LAX3-induced cell wall-modifying enzyme, are expressed in the developing syncytium and in cells to be incorporated into the syncytium. We observed no decrease in H. schachtii infectivity in auxl and Iax3 single mutants. However, a decrease was observed in both the auxllax3 double mutant and the auxllaxllax21ax3 quadruple mutant. In addition, ectopic expression of 19C07 was found to speed up lateral root emergence. We propose that Hsl9C07 most likely increases LAX3-mediated auxin influx and may provide a mechanism for cyst nematodes to modulate auxin flow into root cells, stimulating cell wall hydrolysis for syncytium development. •Soybean cyst nematodes ( Heterodera glycines) produce secreted effector proteins that function as peptide mimics of plant CLAVATA3/ESR (CLE)-like peptides probably involved in the developmental reprogramming of root cells to form specialized feeding cells called syncytia. •The site of action and mechanism of delivery of CLE effectors to host plant cells by the nematode, however, have not been established. In this study, immunologic, genetic and biochemical approaches were used to reveal the localization and site of action of H. glycines-secreted CLE proteins in planta. •We present evidence indicating that the nematode CLE propeptides are delivered to the cytoplasm of syncytial cells, but ultimately function in the apoplast, consistent with their proposed role as ligand mimics of plant CLE peptides. We determined that the nematode 12-amino-acid CLE motif peptide is not sufficient for biological activity in vivo, pointing to an important role for sequences upstream of the CLE motif in function. •Genetic and biochemical analysis confirmed the requirement of the variable domain in planta for host-specific recognition and revealed a novel role in trafficking cytoplasmically delivered CLEs to the apoplast in order to function as ligand mimics. Fourteen of the most commonly used textbooks in human behavior and social environment (HB&SE) courses in graduate social work education were examined for content related to theories of immigration. To what extent are social work students informed in theories of immigration to enhance their understanding and inform their practice with immigrant populations? In an attempt to address this issue, a brief overview of some of the major theories of immigration is provided as a context for the analysis of HB&SE textbooks. The 14 texts were assessed for immigration-related content according to three criteria: (a) the manner in which immigration was addressed, (b) the extent to which immigration was described in terms of human behavior, the social environment, and/or the interaction between the two, and (c) the context in which immigration was presented. The findings from this analysis indicate that immigration is not thoroughly or consistently addressed in these HB&SE texts. This literature review examines major theoretical frameworks utilized by sociologists in examining the processes by which immigrants are incorporated into a host society. Beginning with classical assimilation theory in the 1920s, theorists defined the incorporation process as a linear path for all immigrants, regardless of group characteristics or social context. The responsibility for change was placed solely on the immigrant group. By the 1960s, theorists had redefined assimilation to acknowledge different paths of integration influenced by interactions of immigrant group characteristics with the host society context. Modern theories branching from classic assimilation theory include segmented assimilation and spatial assimilation and theories focusing on ethnic group influences. The review concludes with implications for better understanding influences of interactions between human behavior and the social environment as they relate to immigrant populations. Crashes that occur on freeway ramps and in the vicinity of ramps were analyzed using a variety of statistical methods. The data used in this study are the 6 year records of crashes that occurred on 98 ramps of a 25 mile section of Interstate-4 freeway in the Orlando, Florida, metro area. Log linear models were used to identify the relationship among the frequency of ramp crashes, ramp geometry, and ramp traffic volume. Binary logit models and bivariate probit models were developed to observe the propensity of specific type of ramp crashes in terms of curvature of ramps, number of vehicles involved in crashes, and collision type. The binary logit model was also used to identify the factors correlated with injury severity of drivers involved in ramp crashes. The results of model estimates showed that crash characteristics differ by the location of crashes with respect to ramps: ramp related and ramp intersection related crashes. It was found that the presence of advisory speed signs potentially reduces the likelihood of the occurrence of crashes on off ramps and rear-end crashes on ramps. Finally, it was found that female drivers, single vehicle crashes, and shorter ramp length are correlated with higher probability of fatal and severe crashes on ramps. Abstract: The deposition of carbon layers on the surfaces of optics exposed to extreme ultraviolet (EUV) radiation has been observed in EUV lithography. It has become of critical importance to detect the presence of the carbon layer in the order of nanometer thickness due to carbon''s extremely strong absorption of EUV radiation. Furthermore, the development of efficient cleaning strategies requires that the nature of these carbon layers is well understood. Here, we present experimental results on the detection and characterization of carbon layers, grown on Mo/Si EUV reflecting optics, by laser-generated surface acoustic waves (LG-SAW). It was found that SAW pulses with a frequency bandwidth of more than 220 MHz can be generated and detected for multilayer mirrors and LG-SAW is sensitive enough to detect EUV induced carbon layers less than 5 nm thick. It was inferred from the low Young''s modulus (<100 GPa) that the carbon layer induced by EUV illumination in these vacuum conditions is mechanically soft and polymeric in nature with a high percentage of hydrogen. Objectives: Proposals to make decisions about coverage of new technology by comparing the technology's incremental cost-effectiveness with the traditional benchmark of dialysis imply that the incremental cost-effectiveness ratio of dialysis is seen a proxy for the value of a statistical year of life. The frequently used ratio for dialysis has, however, not been updated to reflect more recently available data on dialysis. Methods: We developed a computer simulation model for the end-stage renal disease population and compared cost, life expectancy, and quality-adjusted life expectancy of current dialysis practice relative to three less costly alternatives and to no dialysis. We estimated incremental cost-effectiveness ratios for these alternatives relative to the next least costly alternative and no dialysis and analyzed the population distribution of the ratios. Model parameters and costs were estimated using data from the Medicare population and a large integrated health-care delivery system between 1996 and 2003. The sensitivity of results to model assumptions was tested using 38 scenarios of one-way sensitivity analysis, where parameters informing the cost, utility, mortality and morbidity, etc. components of the model were by perturbed +/−50%. Results: The incremental cost-effectiveness ratio of dialysis of current practice relative to the next least costly alternative is on average $129,090 per quality-adjusted life-year (QALY) ($61,294 per year), but its distribution within the population is wide; the interquartile range is $71,890 per QALY, while the 1st and 99th percentiles are $65,496 and $488,360 per QALY, respectively. Higher incremental cost-effectiveness ratios were associated with older age and more comorbid conditions. Sensitivity to model parameters was comparatively small, with most of the scenarios leading to a change of less than 10% in the ratio. Conclusions: The value of a statistical year of life implied by dialysis practice currently averages $129,090 per QALY ($61,294 per year), but is distributed widely within the dialysis population. The spread suggests that coverage decisions using dialysis as the benchmark may need to incorporate percentile values (which are higher than the average) to be consistent with the Rawlsian principles of justice of preserving the rights and interests of society's most vulnerable patient groups. Abstract: This study examines the impact of passengers on the driver''s crash potential on freeways. To estimate the impact, a set of bivariate probit models were developed using the 5-year (1999–2003) crash records on a 36.3-mile stretch of Interstate-4 freeway (I-4) in Orlando, Florida. Bivariate probit models identify the correlation between potentially inter-related choices of three passenger characteristics and three crash characteristics. The analysis using bivariate probit models showed that there exist strong correlations between passenger and crash characteristics. It was found that drivers generally display safer driving behavior when they are accompanied by passengers, and more passengers reduce driver''s crash potential. It was also found that younger driver''s crash potential increases with the presence of a younger passenger only. In addition, the analysis of crash type using traffic flow parameters at the time of crashes showed that young drivers with only younger passengers are more likely to be involved in single-vehicle crashes in high-speed and low-volume conditions. The findings in this study provide insight into how the presence of passengers has an impact on driver behavior and traffic safety in various conditions. [Copyright &y& Elsevier]

This study was conducted to assess outcomes after percutaneous transluminal septal myocardial ablation (PTSMA) treatment in 131 patients (mean age 56 ± 16 years) with obstructive hypertrophic cardiomyopathy. In-hospital and follow-up complications as well as late PTSMA failure (defined as unsatisfactory clinical outcome and a significant residual outflow tract gradient, necessitating reintervention) were noted. Baseline clinical, echocardiographic, and PTSMA characteristics were examined as determinants of outcomes. Also, the effect of ethanol volume and the role of a learning curve were investigated. PTSMA was successful in 90% of the patients. In-hospital and follow-up cardiac events were noted in 20 patients, including cardiac death (in-hospital n = 4, follow-up n = 1), acute myocardial infarction due to ethanol leakage (n = 1), coronary dissection (n = 2), nonfatal cardiac tamponade (n = 1), and permanent pacemaker (n = 6) or cardiac defibrillator (in-hospital n = 4, follow-up n = 1) implantation. Late PTSMA failure was noted in 12 patients. All baseline characteristics were comparable between successful and failed PTSMA. Ethanol volume was related to peak creatinine kinase value (p <0.0001) but not to late PTSMA failure or greater need for pacemaker implantation. Late PTSMA failure occurred more frequently in PTSMA procedures performed in the early, less experienced time period (p <0.001). In conclusion, this study confirms that PTSMA, although effective, has a relatively high complication rate. Late PTSMA failure could not be predicted by baseline characteristics but could partially be explained by a learning-curve effect. Abstract: Background: The national recommendation for the management of localized T2 gallbladder cancer (GBCA) is radical cholecystectomy. Although reported survival for localized T2 disease has been poor, groups have documented improvement with radical resection. We hypothesized that a discrepancy exists between national recommendations and current practice patterns. Methods: Patients diagnosed with localized T2 GBCA between 1988 and 2002 were identified from the Surveillance, Epidemiology, and End Results registry. Age, sex, race, ethnicity, extent of surgery, and overall survival were assessed. Surgical procedure was categorized as cholecystectomy alone (CS), cholecystectomy plus lymph node dissection (CS+LN), radical cholecystectomy (RCS), or other. Survival calculations were made using the Kaplan-Meier method and compared with the log-rank test. Results: Of 382 patients with pathologically confirmed T2 GBCA, 280 were women. The median patient age was 75 years. A total of 238 patients underwent CS, 76 underwent CS+LN, and 14 underwent RCS. The remaining 54 patients underwent a lesser or no procedure and were excluded from comparative analysis. The median survival was 14 months for all patients and 14, 14, and 8 months for subgroups treated with CS, CS+LN, and RCS, respectively. Rates of 5-year survival were 23%, 24%, and 36% for CS, CS+LN, and RCS subgroups, respectively. There was no significant difference in survival rates between RCS and CS+LN, or between RCS and CS. This paper considers the value of a specific non-punitive approach to dealing with bullying. It is based upon an exploratory project undertaken in a Year 3 class in a primary school which has not been identified by name. The project involved working with pupils involved in bullying using the stages and principles of the Support Group Method (SGM) and collecting data for analysis through interviews, observations and field notes. Abstract: This paper examines automated control strategies of variable speed limits that aim at reducing crash potential on instrumented freeways. A real-time crash prediction model was developed to estimate crash potential based on short-term variation of traffic flow characteristics. A microscopic traffic simulation model was used to realistically simulate changes in traffic conditions as an effect of variable speed limits and combined with the crash prediction model for the evaluation of control logics. Within this integrated evaluation framework, the study investigated the effect of strategy control factors on the crash potential reduction and total travel time. The study results indicated that variable speed limits could reduce crash potential by 5–17%, by temporarily reducing speed limits during risky traffic conditions when crash potential exceeded the pre-specified threshold. In this paper, we report improvements to a previously developed method based on optical nonlinearity for characterizing polymorphism and concentration of pharmaceuticals in powdered and tablet form. An apparatus that measures the nonlinear optical response of a sample through second harmonic generation (SHG) is described. The response of several enalapril maleate-polyvinylpyrrolidone (PVP) tablets was measured, the results of which were used to determine the concentration of the drug. The current limit of detection of the apparatus was found to be ∼1%–2%. Ranitidine hydrochloride (RN) polymorph forms I and II were also characterized using SHG. It was found that pure samples of forms I and II could be clearly and rapidly distinguished. Mixtures consisting of 50% form I and 50% form II were also distinguishable from the respective pure forms. An investigation was performed into the size dependence of the SHG response for crystalline lactose. It was found that the SHG response was a slowly decreasing monotonic function of particle size. Additional investigation into the angular dependence of the scattered SHG light was also undertaken for crystalline lactose. Abstract: This study presents a real-time crash prediction model and uses this model to investigate the effect of the local traffic-responsive ramp metering strategy on freeway safety. Safety benefits of ramp metering are quantified in terms of the reduced crash potential estimated by the real-time crash prediction model. Driver responses to ramp metering and the consequent traffic flow changes were observed using a microscopic traffic simulation model and crash potential was estimated for a 14.8km section of I-880 in Hayward, California and a hypothetical isolated on-ramp network. The results showed that ramp metering reduced crash potential by 5–37% compared to the no-control case. It was found that safety benefits of local ramp metering strategy were only restricted to the freeway sections in the vicinity of the ramp, and were highly dependent on the existing traffic conditions and the spatial extent over which the evaluation was conducted. The results provide some insight into how a local ramp metering strategy can be modified to improve safety (by reducing total crash potential) on longer stretch of freeways over a wide range of traffic conditions. Abstract: This study analyzes vehicle–pedestrian crashes at intersections in Florida over 4 years, 1999–2002. The study identifies the group of drivers and pedestrians, and traffic and environmental characteristics that are correlated with high pedestrian crashes using log-linear models. The study also estimates the likelihood of pedestrian injury severity when pedestrians are involved in crashes using an ordered probit model. To better reflect pedestrian crash risk, a logical measure of exposure is developed using the information on individual walking trips in the household travel survey. Lastly, the impact of average traffic volume on pedestrian crashes is examined. As a result of the analysis, it was found that pedestrian and driver demographic factors, and road geometric, traffic and environment conditions are closely related to the frequency and injury severity of pedestrian crashes. Higher average traffic volume at intersections increases the number of pedestrian crashes; however, the rate of increase is steeper at lower values of average traffic volume. This paper makes use of a probabilistic model that predicts the likelihood of crashes (crash potential) on freeways on the basis of traffic flow conditions, in real-time crash prevention. The model was developed using incident logs and loop detector data collected over a 13-month period on the Gardiner Expressway in Toronto. Previous work suggested that an increase in levels of traffic turbulence generally yields high crash potential. Traffic turbulence was defined in terms of a series of crash precursors that represent traffic conditions that were present prior to crash occurrence. To apply the model in crash prevention, the link needs to be established between crash potential and real-time safety intervention. The objective of this paper is to explore this link for different thresholds of crash potential. The paper discusses the guidelines for evaluating the safety benefit of one crash prevention strategy (variable speed limits) and suggests the risk-based evaluation framework for real-time traffic control. • The current paper provides an overview of human evaluation practices in NLG. • The current paper gives an overview of the steps necessary to undertake a human evaluation study. • Building on findings from NLG, but also statistics and the behavioral sciences, the current paper provides a set of recommendations and best practices for human evaluation in NLG. Currently, there is little agreement as to how Natural Language Generation (NLG) systems should be evaluated, with a particularly high degree of variation in the way that human evaluation is carried out. This paper provides an overview of how (mostly intrinsic) human evaluation is currently conducted and presents a set of best practices, grounded in the literature. These best practices are also linked to the stages that researchers go through when conducting an evaluation research (planning stage; execution and release stage), and the specific steps in these stages. In a 'projective' test of the golden section hypothesis (Benjafield & Adams- Webber, 1976). 24 Canadian highschool students (9 girls and I5 boys), ages 18-19, completed a repertory grid in which they categorized themselves and ten comic strip characters (e.g., Fred Flintstone) on the basis of twelve bipolar constructs (e.g., generous-mean). The overall of cartoon figures which they assigned to the positive poles of constructs (e.g., generous) was 0.615, which is very close to the golden section (approximately 0.618). • Perception-reaction time at various spacing was determined using a visual variable. • Drivers generally reacted when the accumulation of evidence reached a threshold. • Magnitude of deceleration depends on the level of urgency at the start of braking. • High urgency and insufficient brake force are related to high crash risk. Prediction of driver reaction to the lead vehicle motion based on the perception-reaction time (PRT) is critical for prediction of rear-end crash risk. This study determines PRT at various spacings in approaching and braking conditions, and examines the association of PRT and deceleration rate with crash risk. For these tasks, a total of 50 drivers' behavior was observed in a driving simulator experiment with 4 different scenarios – reaction to a decelerating lead vehicle, reaction to a stopped lead vehicle, perception of a lead vehicle's speed change, and perception of a slow-moving lead vehicle. The study tested three hypotheses of PRT including perception and reaction thresholds and the evidence accumulation framework using a visual variable (tau-inverse). It was found that the drivers neither reacted after a specific PRT from the start of perception nor reacted at a specific value of tau-inverse. Rather, the drivers generally reacted when the accumulation of evidence (tau-inverse) over time reached a threshold. It was also found that the magnitude of deceleration rate depends on the tau-inverse at the start of braking and hence, higher crash risk was associated with higher level of urgency and insufficient brake force rather than longer PRT. This study demonstrates that the evidence accumulation framework is a promising method of predicting driver reaction in approaching and braking conditions for different types of lead vehicle, and the level of urgency is important for predicting the probability of crash. In this paper, we present an algorithm developed to detect Earth-orbiting object streaks within astronomical images. The NASA Wide-field Infrared Survey Explorer (WISE) database of imagery was selected, in order to assess the viability of infrared wavelengths for the detection and analysis of satellite, rocket body and debris objects. The algorithm was designed to scan a high volume of images for the presence of streaks and then to correlate these to known Earth-orbiting objects without the use of a-priori knowledge of position or orbit parameters of the detected object. As part of the streak analysis process, the algorithm performed a photometric characterisation of the detected objects in all four infrared bands observed by the WISE satellite. Accumulation of amyloid-b peptide (Aβ) aggregates in synapses may contribute to the profound synaptic loss characteristic of Alzheimer's disease (AD). The origin of synaptic Aβ aggregates remains elusive, but loss of endosomal proteostasis may trigger their formation. In this study, we identified the synaptic compartments where Aβ accumulates, and performed a longitudinal analysis of synaptosomes isolated from brains of TgCRND8 APP transgenic mice of either sex. To evaluate the specific contribution of Aβ-degrading protease endothelin-converting enzyme (ECE-1) to synaptic/endosomal Aβ homeostasis, we analyzed the effect of partial Ece1 KO in brain and complete ECE1 KO in SH-SY5Y cells. Global inhibition of ECE family members was used to further assess their role in preventing synaptic Aβ accumulation. Results showed that, before extracellular amyloid deposition, synapses were burdened with detergent-soluble Aβ monomers, oligomers, and fibrils. Levels of all soluble Aβ species declined thereafter, as Aβ42 turned progressively insoluble and accumulated in Aβ-producing synaptic endosomal vesicles with characteristics of multivesicular bodies. Accordingly, fibrillar Aβ was detected in brain exosomes. ECE-1-deficient mice had significantly increased endogenous synaptosomal Aβ42 levels, and protease inhibitor experiments showed that, in TgCRND8 mice, synaptic Aβ42 became nearly resistant to degradation by ECE-related proteases. Our study supports that Aβ accumulating in synapses is produced locally, within endosomes, and does not require the presence of amyloid plaques. ECE-1 is a determinant factor controlling the accumulation and fibrillization of nascent Aβ in endosomes and, in TgCRND8 mice, Aβ overproduction causes rapid loss of Aβ42 solubility that curtails ECE-mediated degradation. Aims: Changes in serotonergic sensory modulation associated with overexpression of 5‐HT3 receptors in the central nervous system (CNS) have been implicated in the pathophysiology of neuropathic pain after peripheral nerve damage. 5‐HT3 receptor antagonists such as ondansetron can potentially alleviate neuropathic pain, but have limited effectiveness, due potentially to limited CNS access. However, there is currently limited information on CNS disposition of systemically‐administered 5‐HT3 receptor antagonists. This study evaluated the cerebrospinal fluid (CSF) disposition of ondansetron, as a surrogate of CNS penetration. Methods: Fifteen patients were given a single 16 mg intravenous 15 minute infusion of ondansetron, followed by serial blood and a single CSF sampling. Population pharmacokinetic (PK) modelling was implemented to describe the average and individual plasma and CSF profiles of ondansetron. A two‐compartmental model was used to capture ondansetron plasma PK with a single CSF compartment to describe distribution to the CNS. Results: The individual model‐estimated CSF to plasma partition coefficients of ondansetron were between 0.09 and 0.20. These values were mirrored in the calculated CSF penetration ratios, ranging from 0.08 to 0.26. Conclusions: After intravenous administration, CSF concentrations of ondansetron were approximately 7‐fold lower than those observed in the plasma. A model could be developed to describe individual CSF concentration–time profiles of ondansetron based on a single CSF data point. The low CSF penetration of ondansetron may explain its limited analgesic effectiveness, and affords an opportunity to explore enhancing its CNS penetration for targeting conditions such as neuropathic pain. Shrimp is the most-consumed seafood product in the United States; however, there is a lack of research into the extent of short-weighting and mislabeling of shrimp in the commercial marketplace. The objective of this study was to investigate frozen shrimp for Country of Origin Labeling (COOL) compliance, species authentication, acceptable market names, net weights, and percent glaze. A total of 106 frozen shrimp packages were purchased from grocery stores in Southern California. Samples were considered COOL compliant if both the procurement method and country of origin were reported at the point of sale. Species authentication and acceptable market names were determined by comparing the species identification based on DNA barcoding to the acceptable market names on the FDA Seafood List. Net weights and percent glaze were determined by recording the weight of each sample before and after deglazing according to AOAC methods. The measured net weight of each product was compared to the declared net weight to determine if samples had been short-weighted, taking into account the maximum allowable variation (MAV) by the National Institute of Standards and Technology (NIST). Overall, 94% of samples were compliant with COOL. The average percent glaze was 16.6%, with 26% of samples having >20% glaze. Short-weighting was detected in 37% of samples, with the greatest proportion of incidents recorded for the super/extra colossal shrimp category (57.1%). Species labeling errors were observed in 37% of samples due to conflicting market names, species substitution, and/or use of unacceptable market names. The results of this study indicate a high level of COOL compliance but suggest a need for increased scrutiny of species mislabeling and short-weighting of frozen shrimp. • Shrimp was examined for net weight, species authentication, and labeling compliance. • COOL compliance was identified in 94% of samples. • Short-weighting was detected in 37% of samples and 26% of samples were overglazed. • Overall, 37% of samples had species labeling errors. • Species substitution was detected in 21% of products identified with DNA barcoding.
