The industrial application of whey proteins as a food emulsifier is still limited due to its susceptibility towards heating. In this study, dry heat conjugation of whey protein and lactose was investigated to improve the heat stability of WPC-stabilised oil in water (O/W) emulsions. A commercially available whey protein concentrate (WPC), containing 80 % protein and 12 % lactose, was dry heat incubated. Hereby, the effect of incubation temperature (60, 70, and 80 °C), relative humidity (64, 74, and 79 %), and preconditioning pH (4, 6, 8, 10) were determined. Temperature variation showed a pronounced effect on the required duration to obtain conjugates with excellent emulsion heat stabilising capacity. Four hours of incubation at 80 °C was sufficient to retain the original particle size and consistency after heating the emulsions at 80 °C for 20 min. Whereas circular dichroism spectroscopy revealed that this dry heat incubation condition (80 °C, 4 h) maintained the secondary structure of WPC, pulsed-field gradient NMR revealed that about 40 % of the lactose present should become protein-bound to generate conjugates with the desired functionality. The results indicated that a heat stable WPC with about 40 % of bound lactose could be obtained by some hours of dry heat treatment at 80 °C.

Whey protein–lactose conjugates were prepared by dry heat incubation (60 °C, 74% relative humidity) at 1:1 M ratio of protein amino groups to reducing sugar. Pulsed-field gradient-nuclear magnetic resonance (PFG-NMR) was introduced as a new and absolute method to determine the degree of conjugation without any sample pretreatment, based on the different diffusion behaviour of free and protein-bound lactose. Whereas the TNBS method revealed 73% conjugation, PFG-NMR results indicated that only 37% of lactose was bound to the protein after 8 h incubation. Circular dichroism spectra showed no change of WPI secondary structure upon dry heat incubation, whereas the electrophoretic mobility shifted towards more negative values. The emulsifying and, more importantly, heat stabilising properties of the conjugates were subsequently evaluated in oil/water emulsions. Eight hours of incubation was sufficient to produce conjugates that successfully stabilised the emulsion against heating (80 °C, 20 min), regarding the particle size and viscosity.

Silica@zirconia core@shell oxide nanoparticles of approx. 50 nm diameter were developed in view of an application as a novel carrier with adjuvant activities for cancer vaccines. To this end, the aim was to cover the surface of the nanocarrier by potent single stranded DNA (ssDNA) type immune stimulators used in cancer vaccination. Prior to characterizing the binding of ssDNA to the nanocarrier surface, the adsorption of deoxynucleoside monophosphate building blocks (dNMPs) was studied. After optimization of conditions for synthesis, solvent exchange and surface modification, the effects and possible interference of several buffers, such as HEPES, PIPES, MOPS and MOPSO, were also investigated. Formation of a new compound was revealed by UV–VIS spectroscopy during the recording of zeta potential vs. pH titration curves in the presence of HEPES and PIPES buffer in the neutral-acidic pH region. We identified this new species as gold ions etched from the electrode of the zeta cuvette, stabilized by the buffers via chelation, which is then followed by their conversion into gold nanoparticles.

Pseudomonas species are metabolically robust, with capacity to produce secondary metabolites including cyclic lipopeptides (CLPs). Herein we conducted a chemical analysis of a crude CLP extract from the cocoyam rhizosphere-derived biocontrol strain Pseudomonas sp. COW3. We performed in silico analyses on its whole genome, and conducted in vitro antagonistic assay using the strain and purified CLPs. Via LC-MS and NMR, we elucidated the structures of four novel members of the bananamide group, named bananamides D-G. Besides variability in fatty acid length, bananamides D-G differ from previously described bananamides A-C and MD-0066 by the presence of a serine and aspartic acid at position 6 and 2, respectively. In addition, bananamide G has valine instead of isoleucine at position 8. Kendrick mass defect (KMD) allowed the assignment of molecular formulae to bananamides D and E. We unraveled a non-ribosomal peptide synthetase cluster banA, banB and banC which encodes the novel bananamide derivatives. Furthermore, COW3 displayed antagonistic activity and mycophagy against Pythium myriotylum, while it mainly showed mycophagy on Pyricularia oryzae. Purified bananamides D-G inhibited the growth of P. myriotylum and P. oryzae and caused hyphal distortion. Our study shows the complementarity of chemical analyses and genome mining in the discovery and elucidation of novel CLPs. In addition, structurally diverse bananamides differ in their antimicrobial activity.

While the surface termination of quasi-spherical metal chalcogenide nanocrystals or quantum dots has been widely investigated, it remains unclear whether the ensuing surface chemistry models apply to similar nanocrystals with anisotropic shapes. In this work, we report on the surface-chemistry of 2D CdSe nanoplatelets, where we make use of an improved synthesis strategy that yields stable and aggregation free nanoplatelet suspensions with a photoluminescence quantum yield as high as 55%. We confirm that such nanoplatelets are enriched in Cd and, by means of 1H nuclear magnetic resonance spectroscopy, we show that the Cd-rich surface is terminated by X-type carboxylate ligands. Not unlike CdSe quantum dots (QDs), entire cadmium carboxylate entities can be displaced by the addition of amines, and the desorption isotherm points toward a considerable binding site heterogeneity. Moreover, we find that even the slightest displacement of cadmium carboxylate ligands quenches the nanoplatelet photoluminescence. These experimental findings are further confirmed by density functional theory (DFT) calculations on a 5 monolayer model CdSe nanoplatelet. These simulations show that the most labile ligands are located in the vicinity of facet edges, and that the displacement of ligands from such edge sites creates midgap states that can account for the observed photoluminescence quenching. Next to extending surface chemistry insights from colloidal QDs to nanoplatelets, this work indicates that CdSe nanoplatelets constitute a unique nanocrystal model system to establish a comprehensive description of midgap trap states, which includes their structural, chemical, and electronic properties.

Monofluorination at the proline 4-position results in conformational effects, which is exploited for a range of applications. However, this conformational distortion is a hindrance when the natural proline conformation is important. Here we introduce (3S,4R)-3,4-difluoroproline, in which the individual fluorine atoms instill opposite conformational effects, as a suitable probe for fluorine NMR studies.

Cyclic lipodepsipeptides (CLPs) are a group of metabolites produced by Pseudomonas bacteria, involved in various biological functions and displaying a wide range of properties, including antibacterial and antifungal activities. The white line-inducing principle (WLIP) is a member of the viscosin group featuring a Glu2 amino acid. Recently, a total synthesis of pseudodesmin A - the Gln2 counterpart of WLIP - was described, and we here expand this route to Glu2 containing CLPs. We report the first total synthesis of WLIP and at the same time establish that the Gln2 to Glu2 substitution has an adverse impact on the crude purity and overall yield. A comparative study of different CLP analogues reveals the importance of the nature of the Glx2 protecting group in determining these outcomes. Replacement of the conventional tBu protecting group by the larger benzyl group for the Glu residue in our synthesis strategy indeed resulted in an improved conversion. Next to achieving the first WLIP total synthesis, we thus show the importance of a careful choice of protecting groups for the success of this type of solid-phase synthesis approaches towards CLPs.

Seed survival in soil could be strongly influenced by habitat characteristics, but little is known about the behaviour of seeds sensitive to desiccation in seed banks installed in natural or disturbed habitats. Cryptocarya aschersoniana seeds disperse at the end of the rainy season but do not germinate immediately; thus, they may form seed banks in soil. This study evaluated the behaviour of C. aschersoniana seed banks induced in the natural environment of the species and in a disturbed area. Recently harvested C. aschersoniana germination units were characterized according to their water content, germination and viability. In 2011 and 2012, seed banks were established by burying samples of seeds in the understory of a semi-deciduous forest. In 2012, samples were also buried in a disturbed area. The seed banks were sampled at certain time intervals, and the samples were characterized as described above. Precipitation and air temperature data were collected. As a result, seeds in the seed bank established in the natural environment form a transient seed bank and showed the same behaviour in both years studied. A germination peak was observed starting 210 days after burial (coinciding with the onset of the rainy season) and reached germination percentages higher than 80% at the end of the experiment for both years. Seed mortality did not exceeded 28% in the natural environment. However, in the disturbed environment, the seeds lost their viability more rapidly, with 90% of the seeds becoming unviable 240 days after burial. Germinated seeds in the disturbed environment (maximum 21%) were not able to establish seedlings. These results underscore the importance of maintaining a natural, undisturbed forest for the conservation of this species.

Biocompatible inorganic nano- and microcarriers can be suitable candidates for protein delivery. This study demonstrates facile methods of functionalization by using nanoscale linker molecules to change the protein adsorption capacity of hydroxyapatite (HA) powder. The adsorption capacity of bovine serum albumin as a model protein has been studied with respect to the surface modifications. The selected linker molecules (lysine, arginine, and phosphoserine) can influence the adsorption capacity by changing the electrostatic nature of the HA surface. Qualitative and quantitative analyses of linker-molecule interactions with the HA surface have been performed by using NMR spectroscopy, zeta-potential measurements, X-ray photoelectron spectroscopy, and thermogravimetric analyses. Additionally, correlations to theoretical isotherm models have been calculated with respect to Langmuir and Freundlich isotherms. Lysine and arginine increased the protein adsorption, whereas phosphoserine reduced the protein adsorption. The results show that the adsorption capacity can be controlled with different functionalization, depending on the protein-carrier selections under consideration. The scientific knowledge acquired from this study can be applied in various biotechnological applications that involve biomolecule-inorganic material interfaces.

Ligand exchange is central in the processing of inorganic nanocrystals (NCs) and requires understanding of surface chemistry. Studying sterically stabilized HfO2 and ZrO2 NCs using (1) H solution NMR and IR spectroscopy as well as elemental analysis, this paper demonstrates the reversible exchange of initial oleic acid ligands for octylamine and self-adsorption of oleic acid at NC surfaces. Both processes are incompatible with an X-type binding motif of carboxylic acids as reported for sulfide and selenide NCs. We argue that this behavior stems from the dissociative adsorption of carboxylic acids at the oxide surface. Both proton and carboxylate moieties must be regarded as X-type ligands yielding a combined X2 binding motif that allows for self-adsorption and exchange for L-type ligands.

In the present work, gelatin type B is modified with highly reactive norbornene functionalities (Gel-NB) following a one-pot synthesis approach to enable subsequent thiol-ene photo-click crosslinking. The modification strategy displays close control over the amount of introduced functionalities. Additionally, Gel-NB exhibits considerably improved processing capabilities in terms of two-photon polymerization when benchmarked to earlier-reported crosslinkable gelatin derivatives (e.g., gelatin-methacrylamide (Gel-MOD) and gelatin-methacrylamide-aminoethylmethacrylate (Gel-MOD-AEMA)). The improvement is especially apparent in terms of minimally required laser power (20 mW vs >= 60 mW (Gel-MOD) vs >= 40 mW (Gel-MOD-AEMA) at 100 mm s(-1) scan speed) and processable concentration range (>= 5 w/v% vs >= 10 w/v% (Gel-MOD/Gel-MOD-AEMA)). Furthermore, the proposed functionalization scheme maintains the excellent biocompatibility and cell interactivity of gelatin. Additionally, the norbornene functionalities have potential for straightforward postprocessing thiol-ene surface grafting of active molecules. As a consequence, a very promising material toward tissue engineering applications and more specifically, biofabrication, is presented.

Nanocrystal-ligand interactions and ligand exchange processes are usually described by a uniform distribution of equal binding sites. Here, we analyze this assumption by a quantitative study of the displacement of Z-type cadmium oleate ligands from CdSe nanocrystals by addition of L-type ligands. First, we determined the stoichiometry of the displacement reaction by analyzing the equilibrium upon dilution using solution nuclear magnetic resonance spectroscopy. We found that 1 equiv of tetramethylethylene-1,2-diamine (TMEDA) or 2 equiv of n-butylamine or benzylamine bind to the displaced cadmium oleate. We only reached a comprehensive description of the displacement isotherm by including two types of binding sites with a different equilibrium constant. We corroborated this finding by density functional theory calculations on a CdSe model nanocrystal, which show that even single facets contain a broad variety of binding sites. Finally, we analyzed the thermodynamics of the displacement equilibrium for the weaker binding sites by constructing van't Hoff plots for the different displacers. Whereas displacement with TMEDA appears to be enthalpically neutral, it is entropically favorable. In contrast, displacement with the primary amines is entropically unfavorable but is associated with a negative change in enthalpy. Since the distribution of binding energy emanates from the large fraction of edge and vertex sites on a nanocrystal facet, these findings are most likely inherent to nanocrystals in general and should be considered when analyzing surface reactions on such materials.

The present work investigates the electrochemical determination of cocaine in the presence of levamisole, one of the most common adulterants found in cocaine street samples. Levamisole misleads cocaine color tests, giving a blue color (positive test) even in the absence of cocaine. Moreover, the electrochemical detection of cocaine is also affected by the presence of levamisole, with a suppression of the oxidation signal of cocaine. When levamisole is present in the sample in ratios higher than 1:1, the cocaine signal is no longer detected, thus leading to false negative results. Mass spectrometry and nuclear magnetic resonance were used to investigate if the signal suppression is due to the formation of a complex between cocaine and levamisole in bulk solution. Strategies to eliminate this suppressing effect are further suggested in this manuscript. In a first approach, the increase of the pH of the sample solution from pH 7 to pH 12 allowed the voltammetric determination of cocaine in the presence of levamisole in a concentration range from 10 to 5000 μM at nonmodified graphite disposable electrodes with a detection limit of 5 μM. In a second approach, the graphite electrode was cathodically pretreated, resulting in the presence of oxidation peaks of both cocaine and levamisole, with a detection limit for cocaine of 3 μM over the linear range of concentrations from 10 to 2500 μM. Both these strategies have been successfully applied for the simultaneous detection of cocaine and levamisole in three street samples on unmodified graphite disposable electrodes.

Ten new crystal structures of cis and trans bicyclic diketopiperazines (DKPs) of thia-pipecolic acid (with sulfur in the β, γ or δ position) or thia-proline (with sulfur in the β or γ position) and N-methyl phenylalanine [(NMe)Phe]: cyclo[(β-S)Pip-(NMe)Phe], cyclo[(γ-S)Pip-(NMe)Phe], cyclo[(δ-S)Pip-(NMe)Phe], cyclo[(β-S)Pro-(NMe)Phe] and cyclo[(γ-S)Pro-(NMe)Phe] were determined with X-ray crystallography. Density functional theory calculations of these molecules in the gas phase succeed in reproducing the observed molecular conformations in the crystal remarkably well. This illustrates the weak to moderate impact of intermolecular packing forces in the absence of classical N—H...O hydrogen bonds. The effect of sulfur on the geometry of the DKP ring and details of amide bond non-planarity are discussed. Molecular flexibility of the DKP ring, as estimated from the calculated deformation energies of its endocyclic ring torsion angles, is not in general the decisive factor for the occurrence of multiple symmetry independent molecules in the unit cell (Z′ > 1), though in some cases a correlation is observed.

International audience; G protein-coupled receptors (GPCRs) play an important role in many cellular responses; as such, their mechanism of action is of utmost interest. To gain insight into the active conformation of GPCRs, the X-ray crystal structures of nanobody (Nb)-stabilized β2 -adrenergic receptor (β2 AR) have been reported. Nb80, in particular, is able to bind the intracellular G protein binding site of β2 AR and stabilize the receptor in an active conformation. Within Nb80, the complementarity-determining region 3 (CDR3) is responsible for most of the binding interactions. Hence, we hypothesized that peptidomimetics of the CDR3 loop might be sufficient for binding to the receptor, inhibiting the interaction of β2 AR with intracellular GPCR interacting proteins (e.g., G proteins). Based on previous crystallographic data, a set of peptidomimetics were synthesized that, similar to the Nb80 CDR3 loop, adopt a β-hairpin conformation. Syntheses, conformational analysis, binding and functional in vitro assays, as well as internalization experiments, were performed. We demonstrate that peptidomimetics can structurally mimic the CDR3 loop of a nanobody and its function by inhibiting G protein coupling as measured by partial inhibition of cAMP production.

The association of kinetic studies, DFT calculations and 1H–7Li NMR analyses allowed the control of the cyclo-ATRP of PEG9DMA and the production of polymethacrylate pseudo crown-ethers of various molar masses. Their potential to act as a solid-state polymer electrolyte in Li-ion batteries has been highlighted and may come from the supramolecular organization of the cyclo-PEG forming a Li+ diffusion channel.

The major house dust mite allergen, Der p 1, is a papain-like cysteine protease expressed as an inactive precursor, proDer p 1, carrying an N-terminal propeptide with a unique structure. The maturation of the zymogen into an enzymatically-active form of Der p 1 is a multistep autocatalytic process initiated under acidic conditions through conformational changes of the propeptide, leading to the loss of its inhibitory ability and its subsequent gradual cleavage. The aims of this study were to characterize the residues present in the Der p 1 propeptide involved in the initiation of the zymogen maturation process, but also to assess the impact of acidic pH on the propeptide structure, the activity of Der p 1 and the fate of the propeptide. Using various complementary enzymatic and structural approaches, we demonstrated that a structural triad K17p-D51p-Y19p within the N-terminal domain of the propeptide is essential for its stabilization and the sensing of pH changes. Particularly, the protonation of D51p under acidic conditions unfolds the propeptide through disruption of the K17p-D51p salt bridge, reduces its inhibition capacity and unmasks the buried residues K17p and Y19p constituting the first maturation cleavage site of the zymogen. Our results also evidenced that this triad acts in a cooperative manner with other propeptide pH-responsive elements, including residues E56p and E80p, to promote the propeptide unfolding and/or to facilitate its proteolysis. Furthermore, we showed that acidic conditions modify Der p 1 proteolytic specificity and confirmed that the formation of the first intermediate represents the limiting step of the in vitro Der p 1 maturation process. Altogether, our results provide new insights into the early events of the mechanism of proDer p 1 maturation and identify a unique structural triad acting as a stabilizing and a pH-sensing regulatory element.

Many Pseudomonas spp. produce cyclic lipodepsipeptides (CLPs), which, besides their role in biological functions such as motility, biofilm formation and interspecies interactions, are antimicrobial. It has been established that interaction with the cellular membrane is central to the mode of action of CLPs. In this work, we focus on the CLPs of the so-called viscosin group, aiming to assess the impact of the main structural variations observed within this group on both the antimicrobial activity and the interaction with model membranes. The antimicrobial activity of viscosin, viscosinamide A, WLIP and pseudodesmin A were all tested on a broad panel of mainly Gram-positive bacteria. Their capacity to permeabilize or fuse PG/PE/cardiolipin model membrane vesicles is assessed using fluorescent probes. We find that the Glu2/Gln2 structural variation within the viscosin group is the main factor that influences both the membrane permeabilization properties and the minimum inhibitory concentration of bacterial growth, while the configuration of the Leu5 residue has no apparent effect. The CLP-membrane interactions were further evaluated using CD and FT-IR spectroscopy on model membranes consisting of PG/PE/cardiolipin or POPC with or without cholesterol. In contrast to previous studies, we observe no conformational change upon membrane insertion. The CLPs interact both with the polar heads and aliphatic tails of model membrane systems, altering bilayer fluidity, while cholesterol reduces CLP insertion depth.

To address the need for highly potent, metabolically stable, and selective agonists, antagonists, and inverse agonists at the melanocortin receptor subtypes, conformationally constrained indolo- and benzazepinone residues were inserted into the α-MSH pharmacophore, His(6)-Phe(7)-Arg(8)-Trp(9)-domain. Replacement of His(6) by an aminoindoloazepinone (Aia) or aminobenzazepinone (Aba) moiety led to hMC4R and hMC5R selective agonist and antagonist ligands, respectively (tetrapeptides 1 to 3 and 4, respectively). In peptides 1 to 3 and depending on the para-substituent of the d-Phe residue in position 2, the activity goes from allosteric partial agonism (1, R = H) to allosteric full agonism (2, R = F) and finally allosteric partial agonism (3, R = Br).

The predictable 3D structure of double-stranded DNA renders it ideally suited as a template for the bottom-up design of functionalized nucleic acid-based active sites. We here explore the use of a 14mer DNA duplex as a scaffold for the precise and predictable positioning of catalytic functionalities. Given the ubiquitous participation of the histidine-based imidazole group in protein recognition and catalysis events, single histidine-like modified duplexes were investigated. Tethering histamine to the C5 of the thymine base via an amide bond, allows the flexible positioning of the imidazole function in the major groove. The mutual interactions between the imidazole and the duplex and its influence on the imidazolium pKa(H) are investigated by placing a single modified thymine at four different positions in the center of the 14mer double helix. Using NMR and unrestrained molecular dynamics, a structural motif involving the formation of a hydrogen bond between the imidazole and the Hoogsteen side of the guanine bases of two neighboring GC base pairs is established. The motif contributes to a stabilization against thermal melting of 6 degrees C and is key in modulating the pKa(H) of the imidazolium group. The general features, prerequisites and generic character of the new pKa(H)-regulating motif are described.

We analyze the surface chemistry of CuInS2 nanocrystals synthesized in the presence of amines. Using solution NMR spectroscopy and elemental analysis, we come to the conclusion that as-synthesized CuInS2 nanocrystals have charge neutral inorganic cores and are stabilized by a layer of tightly bound L-type amines. In situ NMR heating-up experiments show that desorption of amines can be induced by increasing the temperature, which makes the partial exchange of amines for thiols possible. On the other hand, we find that carboxylic acids are unable to bind as L-type ligands to the CuInS2 surface. In addition, we demonstrate that the use of technical oleylamine in the synthesis of CuInS2 nanocrystals leads to nonstoichiometric nanocrystals which have, next to oleylamine ligands, also X-type impurities on the surface that can be exchanged for carboxylic acids.

In an attempt to decrease the degree of heat-induced intermolecular protein interactions in recombined concentrated milk, the effect of sunflower lecithin, a natural source of phospholipids, was examined. The obtained results proved that lecithin supplemented milk emulsions demonstrated a less pronounced increase in viscosity as well as particle size, upon severe heating. Additionally, enzymatic hydrolysis was performed using a phospholipase A1. Phosphorus nuclear magnetic resonance and high performance liquid chromatography revealed that the lysophospholipid content reached a maximum after 20 min of hydrolysis and gradually declined as the hydrolysis reaction duration was extended. This work showed that sunflower lecithin largely improves the heat stability of whey protein containing solutions and emulsions due to its high phospholipid content. As the phospholipid molecular structure is important, it follows that the heat stability of whey protein containing products may be optimised by appropriate processing, such as enzymatic hydrolysis of intact lecithins.

A rapid and efficient total synthesis is reported for the cyclic lipodepsipeptide pseudodesmin A. This member of the Pseudomonas viscosin group is active against Gram-positive bacteria and features self-assembling properties. A conserved serine residue within the lactone macrocycle is exploited for initial immobilization on 2-chlorotrityl chloride resin through ether formation with the side-chain alcohol. Subsequent elongation proceeds through Fmoc solid-phase peptide synthesis, including automated incorporation of the enantioselectively synthesized (R)-3-hydroxydecanoic acid lipid tail. Following esterification to generate the incipient lactone bond, the macrocycle is formed by on-resin head-to-tail macrolactamization and cleaved from the resin to give the desired compound in good purity. The short and efficient synthesis route allows rapid generation of analogues by facile variation of both the peptide and lipid moieties with good control of epimerization while maximizing automation. Synthesis of the pseudodesmin A enantiomer yields identical self-assembly and biological activity to that observed for the natural compound, showing that activity is not mediated by chiral interactions. A D-Asn8 analogue developed en route retains self-assembly, but loses activity. The synthesis strategy should be generally applicable for the rapid generation of analogues from various cyclic lipodepsipeptide groups, allowing an investigation of their self-assembling properties and structure-activity relationships.

The non-polar extract of the tubers of Lepidium meyenii Walpers yielded two benzylated alkamides (macamides), N-(3,4-dimethoxybenzyl)-hexadecanamide (1) and N-benzyltetracosanamide (2). The structure elucidation of the compounds was based on 1D and 2D NMR spectroscopic analyses, including 1H–1H COSY, 1H–13C HSQC, 1H–13C HMBC, 1H–15N HSQC and HMBC experiments. Due to the weak regeneration potential of cartilage, there is a high clinical incidence of articular joint disease, leading to a strong demand for cartilaginous tissue surrogates. The aim of this study was to evaluate a gelatin-based hydrogel for its suitability to support chondrogenic differentiation of human mesenchymal stem cells. Gelatin-based hydrogels are biodegradable, show high biocompatibility, and offer possibilities to introduce functional groups and/or ligands. In order to prove their chondrogenesis-supporting potential, a hydrogel film was developed and compared with standard cell culture polystyrene regarding the differentiation behavior of human mesenchymal stem cells. Cellular basis for this study were human adipose tissue-derived mesenchymal stem cells, which exhibit differentiation potential along the adipogenic, osteogenic and chondrogenic lineage. The results obtained show a promotive effect of gelatin-based hydrogels on chondrogenic differentiation of mesenchymal stem cells in vitro and therefore encourage subsequent in vivo studies.

The saikosaponins comprise oleanane- and ursane-type triterpene saponins that are abundantly present in the roots of the genus Bupleurum widely used in Asian traditional medicine. Here we identified a gene, designated CYP716Y1, encoding a cytochrome P450 monooxygenase from Bupleurum falcatum that catalyzes the C-16α hydroxylation of oleanane- and ursane-type triterpenes. Exploiting this hitherto unavailable enzymatic activity, we launched a combinatorial synthetic biology program in which we combined CYP716Y1 with oxidosqualene cyclase, P450, and glycosyltransferase genes available from other plant species and reconstituted the synthesis of monoglycosylated saponins in yeast. Additionally, we established a culturing strategy in which applying methylated β-cyclodextrin to the culture medium allows the sequestration of heterologous nonvolatile hydrophobic terpenes, such as triterpene sapogenins, from engineered yeast cells into the growth medium, thereby greatly enhancing productivity. Together, our findings provide a sound base for the development of a synthetic biology platform for the production of bioactive triterpene sapo(ge)nins.

This study aimed at the interception of flavour deviations of salt-reduced Gouda-type cheeses, salted in sodium-reduced brines (NaCl + KCl brine) compared with a reference brine (NaCl brine), by the addition of adjunct strains of Lactobacillus helveticus and Lactobacillus paracasei , next to the common mixed-strain starter culture for Gouda. A multiphasic analysis of carbohydrates, organic acids, amino acids, and volatile flavour compounds was performed. In this paper, the formation of ZrO2 and yttria-stabilised-zirconia (YSZ) aqueous colloidal systems via microwave assisted hydrothermal synthesis is studied. Microwave synthesis allows a fast screening of the influence of different parameters such as time and temperature. The temperature varied from 140 degrees C up to 180 degrees C and the used reaction time varied from 5 min up to 1 h. The synthesised zirconia nanoparticles have a particle size of 50 nm confirmed by TEM. A H-1 NMR (nuclear magnetic resonance) study helped to understand the stabilization mechanism of the synthesised particles. By the addition of ytrrium ions into the zirconia colloidal solution, YSZ could be formed via an additional thermal treatment. Hereby, the samples are heated up to 400 degrees C for 1 h. YSZ colloidal solutions are synthesised by making use of complexing agents such as nitrilotriacetic acid, ethylenediaminetetraacetic acid and citric acid to control the hydrolysis and condensation of both ions to avoid non-stoichiometric phases. The ratio of Zr/Y in the particles is quantified by XRF. The amorphous structure of those particles necessitates an additional thermal treatment up to 600 degrees C during 1 h in order to obtain crystalline YSZ.

The synthesis and evaluation of gelatin acrylamide as an alternative to the well-established gelatin methacrylamide are reported. High-resolution magic angle spinning NMR is used for the quantitative determination of the chemical cross-linking density. The gelatin acrylamide precursors reveal enhanced cross-linking in terms of reactivity and double bond conversion, resulting in stronger networks. Remarkably, even at very low double bond conversions, ≈5%, gel fractions of ≈40% are obtained. The cross-linked networks are also reviewed in the framework of the rubber elasticity and thermodynamic swelling theories to estimate important nanostructural properties. Preliminary cell tests revealed that highly viable (>90%) cell-laden constructs are obtained.

Because most dairy processes require heat treatment to ensure microbiological safety during storage, dairy industry has invested a lot of effort to acquire knowledge related to all the changes that may take place during heating. More precisely, measures have to be taken to avoid undesired changes that can badly affect the quality of the final product. Whey protein denaturation is one of the major changes that take place upon sterilization. The purpose of the present study was to investigate and compare the degree and type of interaction that occurs between whey protein isolate and different kinds of dialkyl phospholipids, as a function of their molecular structure, upon heating at different temperatures. Circular Dichroism (CD) experiments were performed to assess the extent of secondary structural changes of β-lactoglobulin upon incubation in the presence of both anionic and zwitterionic phospholipids. The far-UV spectra of the former revealed a significant change in ellipticities in the region between 208 and 222 nm, which represents the α-helical content of peptides and proteins upon incubation in the presence of anionic phospholipids at temperatures above their phase transition temperature. On the other hand, these changes were not evident upon incubation with phospholipids in the gel state or when applying zwitterionic phospholipids. Nuclear Magnetic Resonance (NMR) has been used to investigate the whey protein–surfactant interaction into more detail. The chemical shift of the surfactant methyl groups toward a more hydrophobic region in the NMR spectra revealed a hydrophobic whey protein–phospholipid interaction. This finding was further supported by Saturation Transfer Difference-NMR (STD-NMR) measurements. The NMR experiments also revealed that the lack of protein structural reorganization upon incubation in the presence of zwitterionic (lyso)-phospholipids could not be ascribed to a lack of interaction, but was caused by the fact that the interaction did not affect the protein's structure. Overall, this work indicated that phospholipids can modify the secondary structure of whey proteins (i.e. when using anionic phospholipids in the liquid crystalline state) due to hydrophobic interactions. In addition, the results prove that the heat stability of whey protein containing products may be optimized by appropriate selection of the phospholipid composition.

The structure of a pyoverdine produced by Pseudomonas putida, W15Oct28, was elucidated by combining mass spectrometric methods and bioinformatics by the analysis of non-ribosomal peptide synthetase genes present in the newly sequenced genome. The only form of pyoverdine produced by P. putida W15Oct28 is characterized to contain α-ketoglutaric acid as acyl side chain, a dihydropyoverdine chromophore, and a 12 amino acid peptide chain. The peptide chain is unique among all pyoverdines produced by Pseudomonas subspecies strains. It was characterized as -L-Asp-L-Ala-D-AOHOrn-L-Thr-Gly-c[L-Thr(O-)-L-Hse-D-Hya-L-Ser-L-Orn-L-Hse-L-Ser-O-]. The chemical formula and the detected and calculated molecular weight of this pyoverdine are: C65H93N17O32, detected mass 1624.6404 Da, calculated mass 1624.6245. Additionally, pyoverdine structures from both literature reports and bioinformatics prediction of the genome sequenced P. putida strains are summarized allowing us to propose a scheme based on pyoverdines structures as tool for the phylogeny of P. putida. This study shows the strength of the combination of in silico analysis together with analytical data and literature mining in determining the structure of secondary metabolites such as peptidic siderophores.

Solution nuclear magnetic resonance (NMR) spectroscopy is used to analyze the surface chemistry of CdTe quantum dots (QDs) synthesized in the presence of tetradecylphosphonic acid and oleylamine. The resulting CdTe QDs have a mixed capping, composed of tightly bound phosphonate anhydride and amine ligands. The overall ligand density is relatively low, which concurs with a low Cd excess in the QDs. We find that upon addition of oleic acid, neither amine or phosphonate anhydride ligands are released from the QDs. On the other hand, the addition of phosphonic acids to CdTe QDs partially capped by oleic acid moieties through a high temperature ligand exchange process leads to the ready, one-to-one release of oleic acid. A full thermodynamic analysis of the phosphonate/carboxylate exchange is however not possible since the exchange appears to be modified by the forced ligand exchange used to replace phosphonate by carboxylate ligands.

Polysaccharides, and especially alginate, can be useful for self-healing of cracks in concrete. Instead of weak electrostatic bonds present within calcium alginate, covalent bonds, by methacrylation of the polysaccharides, will result in mechanically stronger superabsorbent polymers (SAPs). These methacrylated alginate chains as backbone are combined with two acrylic monomers in a varying molar fraction. These SAPs show a moisture uptake capacity up to 110% their own weight at a relative humidity of 95%, with a negligible hysteresis. The swelling capacity increased (up to 246 times its own weight) with a decreasing acrylic acid/2 acrylamido-2-methylpropane sulfonic acid ratio. The SAPs also showed a thermal stability up to 200 degrees C. Interestingly, the SAP composed of alginate and acrylic acid exerted a very limited decrease in compressive strength (up to 7% with addition of 1 wt% SAP) rendering this material interesting for the envisaged self-healing application. Copper-based chalcogenides that comprise abundant, low-cost, and environmental friendly elements are excellent materials for a number of energy conversion applications, including photovoltaics, photocatalysis, and thermoelectrics (TE). In such applications, the use of solution-processed nanocrystals (NCs) to produce thin films or bulk nanomaterials has associated several potential advantages, such as high material yield and throughput, and composition control with unmatched spatial resolution and cost. Here we report on the production of Cu3SbSe4 (CASe) NCs with tuned amounts of Sn and Bi dopants. After proper ligand removal, as monitored by nuclear magnetic resonance and infrared spectroscopy, these NCs were used to produce dense CASe bulk nanomaterials for solid state TE energy conversion. By adjusting the amount of extrinsic dopants, dimensionless TE figures of merit (ZT) up to 1.26 at 673 K were reached. Such high ZT values are related to an optimized carrier concentration by Sn doping, a minimized lattice thermal conductivity due to efficient phonon scattering at point defects and grain boundaries, and to an increase of the Seebeck coefficient obtained by a modification of the electronic band structure with Bi doping. Nanomaterials were further employed to fabricate ring-shaped TE generators to be coupled to hot pipes, which provided 20 mV and 1 mW per TE element when exposed to a 160 ºC temperature gradient. The simple design and good thermal contact associated with the ring geometry and the potential low cost of the material solution processing may allow the fabrication of TE generators with short payback times.

Aluminium doped zinc oxide is considered an interesting, earth abundant alternative to the transparent conductor indium tin oxide. In this respect, we synthesized Al-ZnO (AZO) thin films by a chemical solution deposition approach. Using water-based inks, the AZO thin films were printed in one single step on glass substrates in non-vacuum conditions. The drying process and the humidity of the atmosphere proved to be crucial during the thermal processing of the wet deposited thin films. To improve the understanding of the phenomena involved, the influence of these processing parameters on the final properties of the AZO thin films are investigated by FTIR, XRD, SEM, 1H and 27Al NMR and resistivity measurements. Optimal conditions were obtained by drying the wet thin film at 120 °C, followed by heating in a tube furnace until 500 °C under a 200 ppm O2/N2 atmosphere with a humidity controlled by bubbling the gas flow through 2 glass bottles containing water at 20 °C. AZO thin films with an resistivity of 2.54 × 10−2 Ω cm were obtained.

PubMed ID: 29058690, Steroid-resistant nephrotic syndrome (SRNS) is a frequent cause of chronic kidney disease. Here, we identified recessive mutations in the gene encoding the actin-binding protein advillin (AVIL) in 3 unrelated families with SRNS. While all AVIL mutations resulted in a marked loss of its actin-bundling ability, truncation of AVIL also disrupted colocalization with F-actin, thereby leading to impaired actin binding and severing. Additionally, AVIL colocalized and interacted with the phospholipase enzyme PLCE1 and with the ARP2/3 actin-modulating complex. Knockdown of AVIL in human podocytes reduced actin stress fibers at the cell periphery, prevented recruitment of PLCE1 to the ARP3-rich lamellipodia, blocked EGF-induced generation of diacylglycerol (DAG) by PLCE1, and attenuated the podocyte migration rate (PMR). These effects were reversed by overexpression of WT AVIL but not by overexpression of any of the 3 patient-derived AVIL mutants. The PMR was increased by overexpression of WT Avil or PLCE1, or by EGF stimulation; however, this increased PMR was ameliorated by inhibition of the ARP2/3 complex, indicating that ARP-dependent lamellipodia formation occurs downstream of AVIL and PLCE1 function. The possibilities offered by 1H solution NMR for the study of colloidal nanocrystal ligands are reviewed. Using CdSe and PbSe nanocrystals with tightly bound oleate ligands as examples, the solution NMR toolbox for ligand analysis is introduced, highlighting 1D 1H, diffusion ordered (DOSY) and nuclear Overhauser effect (NOESY) spectroscopy as NMR techniques that enable bound ligands to be distinguished from free ligands. For each of the toolbox techniques, it is outlined how dynamic stabilization in the fast exchange regime affects the spectra obtained. Next, it is shown how the perturbation of a purified dispersion by dilution or the addition of excess ligands can be used to analyze the binding of ligands to a nanocrystal. Finally, saturation transfer difference (STD) spectroscopy is presented as an NMR technique that may complement the established toolbox.

Poly(styrene-maleic anhydride) or SMA are versatile copolymers available in different grades of molecular weight (Mw = 5500–170,000 g/mol) and molar percentage of maleic anhydride (22–50 mol-%). As specific grades can be used for the synthesis of nanoscale structures in paper industry, a better understanding of the molecular structure was needed. Therefore, a broad range of SMA grades was analyzed by means of size exclusion chromatography, infrared spectroscopy, Raman spectroscopy and nuclear magnetic resonance spectroscopy. While the nominal composition could be determined by 1H NMR and Raman spectroscopy, the combination with 13C NMR and FTIR allowed a more detailed characterization of the molecular structure. The high-molecular weight SMA grades show a heterogeneous molecular structure with segments including high styrene and maleic anhydride moieties, respectively. The formation of film patterns was evaluated by deposition from an acetone solution, resulting in homogeneous films, fibrous structures, or spherical shapes with decreasing diameter as a function of maleic anhydride content including primary particle sizes of 100 nm. This study illustrates intrinsic molecular features of SMA grades that are capable for self-assembly, and can provide a selection tool for future nanoparticle formulation.

We present a scalable synthesis of a versatile MTX reagent with an azide ligation handle that allows rapid γ-selective conjugation to yield MTX fusion compounds (MFCs) appropriate for MASPIT, a three-hybrid system that enables the identification of mammalian cytosolic proteins that interact with a small molecule of interest. We selected three structurally diverse pharmacologically active compounds (tamoxifen, reversine, and FK506) as model baits. After acetylene functionalization of these baits, MFCs were synthesized via a CuAAC reaction, demonstrating the general applicability of the MTX reagent. In analytical mode, MASPIT was able to give concentration-dependent reporter signals for the established target proteins. Furthermore, we demonstrate that the sensitivity obtained with the new MTX reagent was significantly stronger than that of a previously used non-regiomeric conjugate mixture. Finally, the FK506 MFC was explored in a cellular array screen for targets of FK506. Out of a pilot collection of nearly 2000 full-length human ORF preys, FKBP12, the established target of FK506, emerged as the prey protein that gave the highest increase in luciferase activity. This indicates that our newly developed synthetic strategy for the straightforward generation of MFCs is a promising asset to uncover new intracellular targets using MASPIT cellular array screening.

Smart pH-responsive superabsorbent polymers (SAPs) could be useful for self-healing of cracks in mortar. They will swell minimally during the alkaline conditions of mixing, leading to only small macro-pores but will swell stronger with a lower pH when water enters the cracks. As such, polysaccharides (alginate, chitosan and agarose) were methacrylated and cross-linked with amine-based monomers (dimethylaminoethyl methacrylate and dimethylaminopropyl methacrylamide) to induce a varying pH-sensitivity. These materials showed a strong cross-linking efficiency and induced moisture uptake capacities up to 122% at 95% relative humidity with a negligible hysteresis. Additionally, interesting pH-responsive swelling capacities were obtained, especially for SAPs based on chitosan and agarose with values up to 110gwater/gSAP. Most of these materials showed limited hydrolysis in cement filtrate solutions, making them very promising for use in mortar.

Bola-amphiphilic surfactants are molecules with fascinating properties. Their unique configuration consisting of a long hydrophobic spacer connecting two hydrophilic entities renders the molecule more water soluble than the average surfactant, but still allows formation of supramolecular structures. These properties make them extremely suitable for applications in in nanotechnology, electronics, and gene and drug delivery. In general, these compounds are obtained by chemical synthesis. We report here an efficient microbial production process for the fully green synthesis of bolaform surfactants. A sophorolipid-producing Starmerella bombicola yeast strain was disabled in its sophorolipid acetyltransferase and lactone esterase, which should logically result in synthesis of non-acetylated acidic sophorolipids; molecules with the classic amphiphilic structure. Surprisingly, also bolaform glycolipids were obtained, with an additional sophorose linked to the free carboxyl end of the acidic sophorolipids as confirmed by MS and NMR analysis. The obtained titers of 27.7 g/L total product are comparable to wild type values, and the novel molecules account for at least 74% of this. Bola-amphiphile biosynthesis proved to be attributed to the promiscuous activity of both UDP-glucosyltransferases UGTA1 and UGTB1 from the core sophorolipid pathway, displaying activity toward non-acetylated intermediates. The absence of acetyl groups seems to trigger formation of bolaform compounds starting from acidic sophorolipids. Hence, wild type S. bombicola produces these compounds only at marginal amounts in general not reaching detection limits. We created a strain knocked-out in its sophorolipid acetyltransferase and lactone esterase able to produce these novel compounds in economical relevant amounts, opening doors for the application of biological-derived bolaform structures. Orfamide type cyclic lipopeptides (CLPs) are biosurfactants produced by Pseudomonas and involved in lysis of oomycete zoospores, biocontrol of Rhizoctonia and insecticidal activity against aphids. In this study, we compared the biosynthesis, structural diversity, in vitro and in planta activities of orfamides produced by rhizosphere-derived Pseudomonas protegens and related Pseudornonas species. Genetic characterization together with chemical identification revealed that the main orfamide compound produced by the P. protegens group is orfamide A, while the related strains Pseudomonas sp. CMR5c and CMR12a produce orfamide B. Comparison of orfamide fingerprints led to the discovery of two new orfamide homologs (orfamide F and orfamide G) in Pseudornonas sp. CMR5c. The structures of these two CLPs were determined by nuclear magnetic resonance (NMR) and mass spectrometry (MS) analysis. Mutagenesis and complementation showed that orfamides determine the swarming motility of parental Pseudomonas sp. strain CMR5c and their production was regulated by luxR type regulators. Orfamide A and orfamide B differ only in the identity of a single amino acid, while orfamide B and orfamide G share the same amino acid sequence but differ in length of the fatty acid part. The biological activities of orfamide A, orfamide B, and orfamide G were compared in further bioassays. The three compounds were equally active against Magnaporthe oryzae on rice, against Rhizoctonia solani AG 4-HGI in in vitro assays, and caused zoospore lysis of Phytophthora and Pythium. Furthermore, we could show that orfamides decrease blast severity in rice plants by blocking appressorium formation in M. oryzae. Taken all together, our study shows that orfamides produced by P protegens and related species have potential in biological control of a broad spectrum of fungal plant pathogens.

Aerobic oxidative dehydrogenative polycondensation of the new monomer 4,4'-(3-hexylthiophene-2,5-diyl)bis(2-octylaniline) has proved very effective under mild conditions using an inexpensive copper bromide/pyridine complex and dioxygen acting as catalyst and oxidant, respectively. Such kinetically controlled process allowed preparing elec-troconjugated photosensitive polymers with high molar weight (Mn > 15000 g.mol-1).

The secondary metabolite mediating the GacS-dependent growth-inhibitory effect exerted by the rice rhizosphere isolate Pseudomonas putida RW10S2 on phytopathogenic Xanthomonas species was identified as white-line-inducing principle (WLIP), a member of the viscosin group of cyclic lipononadepsipeptides. WLIP producers are commonly referred to by the taxonomically invalid name “ Pseudomonas reactans ,” based on their capacity to reveal the presence of a nearby colony of Pseudomonas tolaasii by inducing the formation of a visible precipitate (“white line”) in agar medium between both colonies. This phenomenon is attributed to the interaction of WLIP with a cyclic lipopeptide of a distinct structural group, the fungitoxic tolaasin, and has found application as a diagnostic tool to identify tolaasin-producing bacteria pathogenic to mushrooms. The genes encoding the WLIP nonribosomal peptide synthetases WlpA, WlpB, and WlpC were identified in two separate genomic clusters ( wlpR-wlpA and wlpBC ) with an operon organization similar to that of the viscosin, massetolide, and entolysin biosynthetic systems. Expression of wlpR is dependent on gacS , and the encoded regulator of the LuxR family (WlpR) activates transcription of the biosynthetic genes and the linked export genes, which is not controlled by the RW10S2 quorum-sensing system PmrR/PmrI. In addition to linking the known phenotypes of white line production and hemolytic activity of a WLIP producer with WLIP biosynthesis, additional properties of ecological relevance conferred by WLIP production were identified, namely, antagonism against Xanthomonas and involvement in swarming and biofilm formation.

A small series of 5-(hetero)aryl-modified nucleoside phosphonates was synthesized via an 8-step procedure including a Wittig reaction and Suzuki-Miyaura coupling.
