65. Identification of personalized neoantigen-based vaccines and immune subtype characteristic analysis of glioblastoma based on abnormal alternative splicing

Abstract

The development of personalized neoantigen-based vaccines in cancer immunotherapy has shown promise. In this study, a large-scale bioinformatics analysis was performed to identify potential GBM-associated neoantigens based on abnormal alternative splicing, and then screen suitable patients for vaccination. Gene expression profiles and clinical information were collected from TCGA. We filtered the percent-spliced-in (PSI) spectrum of alternative splicing events in the dataset to identify abnormal alternative splicing events. MAF package was used to identify and analyse tumour mutation burden (TMB) in cancer samples. Tumour Immune Estimation Resource (TIMER) was used to calculate and visualize the infiltration of antigen presenting cells (APCs). In addition, consistent clustering algorithm utilized to identify immune subtypes of GBM. Five potential tumour neoantigens (LRP1, TCF12, DERL3, WIPI2, and TSHZ3) were identified in GBM by selecting genes both with abnormal alternative splicing (upregulated) and gene frameshift mutations, in which LRP1 was significantly associated with APCs. According to the expressions of five potential tumour neoantigens, 160 patients with GBM were divided into three immune subtypes. Patients in cluster3 exhibited good prognoses. Furthermore, the characteristics, including TMB, abnormal alternative splicing events, immune activity, immune cells proportion, and association with tumour biomarkers, were unique in each immune subtypes. The characteristics of cluster3 illustrated that cluster3 participants were more suitable candidates for vaccination. LRP1 was identified as a potential neoantigen for immunotherapy against GBM, and patients in cluster3 were more suitable for vaccination. Our findings provide important guidance for the development of novel neoantigens and therapeutic targets in patients with GBM.

76. Inhibitory Mechanisms of Three Modified Small Molecules on the Misfolding of Cu2+‐Aβ42 Complex in Different pH Conditions: Insights from MD/QM Studies.

Abstract

The paper reports in silico three modified small molecules, two negatively charged E1C and E2C and one neutral TQ, which show more potent inhibitory effect than their parental molecules CQ3, TS0, and ER on the Cu2+ induced Aβ42 aggregation. In the acidic condition, TQ is the most powerful inhibitor, as it not only decreases the helix content of Cu2+‐Aβ42 complex but also increases the oxidizability; E1C/E2C however decreases the helix content of Cu2+‐Aβ42 complex but also with increased oxidizability, indicating that the inhibitory effect of the two charged molecules depends on the multiple factors, at least including the composite mechanism of helix decrease and oxidizability increase. In the basic condition, any of three modified molecules can increase the helix content of the Cu2+‐Aβ42 complex. Anyway, small molecules containing neutral and larger hydrophobic aromatic groups are always more effective in inhibiting Cu2+‐induced Aβ42 aggregation in both basic and acidic conditions. Abstract

Background: The infection rate of Coronavirus Disease 2019 (COVID-19) in children was less than that in adults. However, the underlining reason is not well known. Methods: Children with COVID-19 were recruited from two Children’s Hospitals in Wuhan and Shanghai in this case-control study. The associations of initial symptoms with age, vaccinations of Bacillus Calmette Guerin (BCG), and influenza and pathogens were determined by Chi-square t-test. Results: We evaluated 248 confirmed cases, and 56 suspected cases with COVID-19. The median age was 6.82 years old, and 118 cases (38.82%) were girls. Furthermore, 30.26% of all patients were asymptomatic cases. The percentage of asymptomatic cases vaccinated with BCG was not significantly higher than that without BCG vaccination [86/280 (30.71%) vs. 6/13 (46.15%), P=0.203], and initial symptoms were not related with immunized influenza vaccine (P=0.267). Compared to parameters in pediatric patients with normal body temperatures, patients with fever had higher C reactive protein (CRP) (P<0.001). Conclusions: Pediatric COVID-19 patients with BCG vaccinations exhibit similar clinical manifestations compared to those without BCG vaccinations, and the severity of symptoms in pediatric patients may be related to the maturity of immune function.

Abstract

Air-cooled frost-free household refrigerators have become popular in the market because of their large storage volume and frost-free compartments. However, the large amount of power consumed by the defrosting process of the evaporator restricts the widespread application of such refrigerators. In this study, a new type of defrosting system using outdoor air (DSUOA) is developed. The operation modes (refrigeration and defrosting cycles) of the overall system are proposed. An experimental prototype is built, and an experimental study is conducted for different temperature ranges (12–22 °C, 22–45 °C, and 0–12 °C). Results show that the power consumption of DSUOA is 77.6–98.2% less than that of the electric heater defrosting system under a similar defrosting time.

Abstract

A series of 3-[2-(2-aminoethylamino)ethylamino]propyl-trimethoxysilane (PAPTS) grafted on Materials of Institute Lavoisier-101(Cr) [MIL-101(Cr)] porous adsorbent with varied PAPTS loadings were synthesized and characterized by powder X-ray diffraction patterns (PXRD), N2 adsorption-desorption measurement at 77 K, Fourier transform infrared spectroscopy (FTIR), and thermal gravimetric analysis (TGA). The water vapor adsorption isotherms were obtained by gravimetric method. Water adsorption isotherms and dynamic on MIL-101 as well as PAPTS- MIL-101(Cr) are discussed simultaneously. Results indicated that even though the Brunauer-Emmett-Teller (BET) surface areas of PAPTS-MIL-101(Cr)-0.5 and PAPTS-MIL-101(Cr)-0.75 were reduced by approximately 50% compared with that of the original MIL-101(Cr), which have slightly higher water uptake capacities in a region of 0.30

Abstract

This paper analyzes the air quality monitoring data of Qingyang, Tianshui and Pingliang in northwestern China in 2015 by time series analysis. The research results are as follows: (1) the concentrations of PM2.5, PM10 and SO2 in the three cities began to decline in March, reached a minimum in September before rising gradually; (2) concentrations of PM2.5, PM10 and SO2 in the three cities were low in summer and autumn, but high in winter and spring; (3) the coefficient of correlation among PM2.5, PM10 and SO2 in the three cities was greater than 0.589, indicating a strong correlation.

Abstract

In this article, MIL-101(Cr) powder material was successfully prepared by hydrothermal synthesis and combined with different mass fractions of CaCl2 by immersion method to obtain MIL-101(Cr)/CaCl2 composite materials. The physical properties of the adsorbent were determined by X-ray powder diffraction, N2 adsorption/desorption isotherm, TGA and FTIR. The water vapor adsorption performance of MOFs was tested by a gravimetric instrument to analyze the underlying mechanism. Results indicated that the specific surface area, total pore volume, and pore size distribution of micropores and mesopores of MIL-101(Cr)/CaCl2 composites decreased with increasing CaCl2 content. When P/P0 = 0.9, the water vapor equilibrium adsorption capacities of MIL-101(Cr), MIL-101(Cr)/CaCl2-10%, MIL-101(Cr)/CaCl2-20%, and MIL-101(Cr)/CaCl2-30% were 1.05, 0.88, 1.13, and 1.35 g/g, respectively. MIL-101(Cr)/CaCl2-30% agglomerated after water absorption, so MIL-101(Cr)/CaCl2-20% was selected as the final experimental material. The water vapor adsorption heat of MIL-101(Cr)/CaCl2-20% was 42–58 kJ/mol, which is close to the evaporation enthalpy of water (44 kJ/mol at 25 °C). The analysis of the adsorption kinetics showed the relatively uniform adsorption rate of MIL-101(Cr)/CaCl2-20%. Overall, MIL-101(Cr)/CaCl2-20% exhibited superior water vapor adsorption performance and have great potential for application to adsorption heat pumps.
