Connected and automated vehicles (CAVs) willrevolutionize the way we travel; however, what impact this revolution will have on advancing broader societal goals is uncertain. To date, the private sector technology rollout has emphasized the automation side of CAVs and neglected the potentially transformative possibilities brought by a more collaborative notion of connectivity. This may have significant downsides from a broader societal perspective. For example, CAVs (including those on the road today) collect a vast amount of data gathered through onboard systems (e.g., radar, lidar, camera), however, this data is not typically shared with other vehicles, roadside infrastructure, or public transportation agencies. This lack of collaboration will likely make traffic worse and forfeit the opportunity to manage traffic at the systems-level, which is where significant gains can be made in terms of improving traffic flow and safety, reducing greenhouse gas emissions and vehicle energy use, and more.

The California Department of Transportation (Caltrans) collects megabytes of data every day using a dedicated traffic sensing infrastructure. The collected data provide support for traffic management and system performance monitoring activities that are crucial for supporting the agency’s mission, vision, and strategic goals to strengthen stewardship and drive efficiency. Operating this vast detection system requires extensive resources in the form of engineering and maintenance support, along with millions in capital funds to keep the system running. Within the above context, alternate hybrid data collection models utilizing purchased or third-party data to augment existing data collection system capabilities may enable a reduction in the number of physical detection stations required while maintaining suitable accuracy for Caltrans’ purposes. In addition to the potential for cost savings, the reliance on fewer physical sensors also offers the potential to reduce the exposure of Caltrans employees to the occupational hazard of maintaining roadside detection stations, in alignment with the agency’s “safety first” strategic goal.

Abstract Members of the leucine rich repeat (LRR) and PDZ domain (LAP) protein family are essential for animal development and histogenesis. Densin-180, encoded by LRRC7, is the only LAP protein selectively expressed in neurons. Densin-180 is a postsynaptic scaffold at glutamatergic synapses, linking cytoskeletal elements with signalling proteins such as the α-subunit of Ca2+/calmodulin-dependent protein kinase II. We have previously observed an association between high impact variants in LRRC7 and Intellectual Disability; also three individual cases with variants in LRRC7 had been described. We identify here 33 individuals (one of them previously described) with a dominant neurodevelopmental disorder due to heterozygous missense or loss-of-function variants in LRRC7. The clinical spectrum involves intellectual disability, autism, ADHD, aggression and, in several cases, hyperphagia-associated obesity. A PDZ domain variant interferes with synaptic targeting of Densin-180 in primary cultured neurons. Using in vitro systems (two hybrid, BioID, coimmunoprecipitation of tagged proteins from 293T cells) we identified new candidate interaction partners for the LRR domain, including protein phosphatase 1 (PP1), and observed that variants in the LRR reduced binding to these proteins. We conclude that LRRC7 encodes a major determinant of intellectual development and behaviour.

Background: The stunting prevalence in Central Java Province remains high. Six key messages, i.e. iron tablet supplementation, healthy latrines, exclusive breastfeeding, Integrated Health Care Post (Posyandu) visit, hand washing, and Antenatal Care visit are the efforts to reduce the prevalence of stunting. Objectives: This study aimed to analyse the relationship between the behaviour of the six key messages and the stunting prevalence in four districts of Central Java Province. Methods: This cross-sectional study was conducted in Brebes Regency, Tegal Regency, Banyumas Regency, and Semarang City with 424 mothers who have under two-year-old children randomly selected as sample. Data were collected by interview using a structured questionnaire with the results grouped into attitude and practices categories. The body’s weight and length were measured by a digital scale and infantometer. Data were analysed using the chi-square test. Results: The findings indicated that 21.5% of children under two had stunting. Besides, behaviour regarding six key messages were mostly good. There was no association between mothers' iron tablet consumption behaviour and the nutritional status of under-two children. There was an association between maternal attitudes about exclusive breastfeeding and knowledge about hand washing with length for age index (p=0.004). There was also an association between antenatal care and Posyandu visits practice with weight for age index (p=0.004 and 0.018). There was a relationship between attitude regarding the use of latrines and the weight for length index (p=0.001). Conclusions: Only the key messages of exclusive breastfeeding attitude, antenatal care visit practice, Posyandu visit practice, and hand washing attitude were associated with the nutritional status of under-two children.

Abstract Developmental and epileptic encephalopathies (DEEs) feature altered brain development, developmental delay and seizures, with seizures exacerbating developmental delay. Here we identify a cohort with biallelic variants in DENND5A, encoding a membrane trafficking protein, and develop animal models with phenotypes like the human syndrome. We demonstrate that DENND5A interacts with Pals1/MUPP1, components of the Crumbs apical polarity complex required for symmetrical division of neural progenitor cells. Human induced pluripotent stem cells lacking DENND5A fail to undergo symmetric cell division with an inherent propensity to differentiate into neurons. These phenotypes result from misalignment of the mitotic spindle in apical neural progenitors. Cells lacking DENND5A orient away from the proliferative apical domain surrounding the ventricles, biasing daughter cells towards a more fate-committed state, ultimately shortening the period of neurogenesis. This study provides a mechanism for DENND5A-related DEE that may be generalizable to other developmental conditions and provides variant-specific clinical information for physicians and families.

Abstract Importance Most patients presenting with a hip fracture regardless of their comorbidities are surgically treated. A growing body of research states that a certain type of elderly patient could benefit more from a palliative approach. Objective Identify the patient who would benefit most from a palliative care approach instead of a surgery. Design Exploratory-matched retrospective cohort study between 2015 and 2021. Setting Single Level 1 Trauma Center. Participants There were 2240 hip fracture patients admitted to our institution between 2015 and 2021. Patients over 65 years old with intertrochanteric or femoral neck fractures could be included. A total of 129 patients opted for palliative care (Palliative Group = PG). This cohort was compared to a matched cohort (for age, sex and fracture type) who underwent surgery but died within three months of the procedure (Surgery Deceased Group = SDG) and another matched cohort who survived more than three months (Surgery Alive Group = SAG) following surgery. Main outcomes and measures Medical charts were reviewed for patient demographics, autonomy level, level of care, neurocognitive disorders (NCD), fracture type, in-hospital data and outpatient death within three months of admission. Analysis was performed through univariate and multivariate models with SAS OnDemand for Academics (alpha 0.05). Results Patients in the PG (n = 129) were 88.2 ± 7.2 years old, 71.3% were females, and 61.2% had a femoral neck fracture. Patients in the SDG (n = 95) and SAG (n = 107) were well matched. Abstract Background Serum creatinine is used as initial test to derive eGFR and confirmatory testing with serum cystatin C is recommended when creatinine‐based eGFR is considered less accurate due to deviant muscle mass. Low muscle mass is associated with increased risk of premature mortality. However, the associations of serum creatinine and cystatin C with muscle mass and mortality remain unclear and require further investigation to better inform clinical decision‐making. Methods We included 8437 community‐dwelling adults enrolled in the Dutch PREVEND study and 5033 in the US NHANES replication cohort. Associations of serum creatinine and/or cystatin C with muscle mass surrogates and mortality were quantified with linear and Cox proportional hazards regression, respectively. Missing observations in covariates were multiply imputed using Substantive Model Compatible Fully Conditional Specification. Results Mean (SD) age of PREVEND and NHANES participants (50% and 48% male) were 49.8 (12.6) and 48.7 (18.7) years, respectively. Median (Q1–Q3) serum creatinine and cystatin C were 71 (61–80) and 80 (62–88) μmol/L and 0.87 (0.78–0.98) and 0.91 (0.80–1.10) mg/L, respectively. Higher serum creatinine was associated with greater muscle mass, while serum cystatin C was not associated with muscle mass. Adjusting both markers for each other strengthened the positive relationship between serum creatinine and muscle mass and revealed an inverse association between serum cystatin C and muscle mass. In the PREVEND cohort, 1636 (19%) deaths were registered over a median follow‐up of 12.9 (5.8–16.3) years with a 10‐year mortality rate (95% CI) of 7.6% (7.1–8.2%). In the NHANES, 1273 (25%) deaths were registered over a median follow‐up of 17.9 (17.3–18.5) years with a 10‐year mortality rate of 13.8% (12.8–14.7%). Both markers were associated with increased mortality. Notably, when adjusted for each other, higher serum creatinine was associated with decreased mortality, while the association between serum cystatin C and increased mortality strengthened. The shapes of the associations in the PREVEND study and NHANES were almost identical. Conclusions The strong association between serum creatinine and muscle mass challenges its reliability as GFR marker, necessitating a more cautious approach in its clinical use. The minimal association between serum cystatin C and muscle mass supports its increased use as a more reliable alternative in routine clinical practice.

Abstract The contraction of CAG/CTG repeats is an attractive approach to correct the mutation that causes at least 15 neuromuscular and neurodegenerative diseases, including Huntington’s disease and Myotonic Dystrophy type 1. Contractions can be achieved in vivo using the Cas9 D10A nickase from Streptococcus pyogenes (SpCas9) using a single guide RNA (sgRNA) against the repeat tract. One hurdle on the path to the clinic is that SpCas9 is too large to be packaged together with its sgRNA into a single adeno-associated virus. Here we aimed to circumvent this problem using the smaller Cas9 orthologue, SlugCas9, and the Cas9 ancestor OgeuIscB. We found them to be ineffective in inducing contractions, despite their advertised PAM sequences being compatible with CAG/CTG repeats. Thus, we further developed smaller Cas9 hybrids, made of the PAM interacting domain of S. pyogenes and the catalytic domains of the smaller Cas9 orthologues. We also designed the cognate sgRNA hybrids using molecular dynamic simulations and binding energy calculations. We found that the four Cas9/sgRNA hybrid pairs tested in human cells failed to edit their target sequences. We conclude that in silico approaches can identify functional changes caused by point mutations but are not sufficient for designing larger scale complexes of Cas9/sgRNA hybrids.

This report describes how current methods of estimating truck traffic volumes from existing fixed roadway sensors could be improved by using tracking data collected from commercial truck fleets and other connected technology sources (e.g., onboard GPS-enabled navigation systems and smartphones supplied by third-party vendors). Using Caltrans District 1 in Northern California as an example, the study first reviews existing fixed-location data collection capabilities and highlights gaps in the ability to monitor truck movements. It then reviews emerging data sources and analyzes the analytical capabilities of StreetLight 2021, a commercial software package. The study then looks at the Sample Trip Count and uncalibrated Index values obtained from three weigh-in-motion (WIM) and twelve Traffic Census stations operated by Caltrans in District 1. The study suggests improvements to StreetLight’s “single-factor” calibration process which limits its ability to convert raw truck count data into accurate traffic volume estimates across an area, and suggests how improved truck-related calibration data can be extracted from the truck classification counts obtained from Caltrans’ WIM and Traffic Census stations. The report compares uncalibrated StreetLight Index values to observed truck counts to assess data quality and evaluates the impacts of considering alternate calibration data sets and analysis periods. Two test cases are presented to highlight issues with the single-factor calibration process. The report concludes that probe data analytical platforms such as StreetLight can be used to obtain rough estimates of truck volumes on roadway segments or to analyze routing patterns. The results further indicate that the accuracy of volume estimates depends heavily on the availability of sufficiently large samples of tracking data and stable and representative month-by-month calibration data across multiple reference locations.

Electro-Fenton process could remove pollutant in in tannery industry wastewater. The objectives of this is used to know the effect of electro-Fenton on, ammonia (NH3), chromium (III) (Cr(III), total suspended solid (TSS), biological oxygen deman d (BOD), and chemical oxygen demand (COD) removal in tannery wastewater treatment. The voltage variation used to trigger the electro-Fenton reaction was 6V, 8V, and 10V. Observation time at 0, 30, 60, 90, and 120 mins was used to see changes in the sample. The results obtained are electro-Fenton method has an effective time of 30 mins to removal efficiency of 60.7%, 32.9%, 72.8%, 53.4%, and 53.4% for NH3, Cr(III), TSS, BOD, and COD respectively. Pollution from tannery effluent can be eliminated to a sufficient extent using the electro-Fenton technique.

Abstract Background Coronary heart disease (CHD) is the leading cause of deaths and disability worldwide. Cardiac rehabilitation (CR) effectively reduces the risk of future cardiac events and is strongly recommended in international clinical guidelines. However, CR program quality is highly variable with divergent data systems, which, when combined, potentially contribute to persistently low completion rates. The QUality Improvement in Cardiac Rehabilitation (QUICR) trial aims to determine whether a data-driven collaborative quality improvement intervention delivered at the program level over 12 months: (1) increases CR program completion in eligible patients with CHD (primary outcome), (2) reduces hospital admissions, emergency department presentations and deaths, and costs, (3) improves the proportion of patients receiving guideline-indicated CR according to national and international benchmarks, and (4) is feasible and sustainable for CR staff to implement routinely. Methods QUICR is a multi-centre, type-2, hybrid effectiveness-implementation cluster-randomized controlled trial (cRCT) with 12-month follow-up. Eligible CR programs (n = 40) and the individual patient data within them (n ~ 2,000) recruited from two Australian states (New South Wales and Victoria) are randomized 1:1 to the intervention (collaborative quality improvement intervention that uses data to identify and manage gaps in care) or control (usual care with data collection only). This sample size is required to achieve 80% power to detect a difference in completion rate of 22%. Outcomes will be assessed using intention-to-treat principles. Mixed-effects linear and logistic regression models accounting for clusters within allocated groupings will be applied to analyse primary and secondary outcomes. Discussion Addressing poor participation in CR by patients with CHD has been a longstanding challenge that needs innovative strategies to change the status-quo. This trial will harness the collaborative power of CR programs working simultaneously on common problem areas and using local data to drive performance. The use of data linkage for collection of outcomes offers an efficient way to evaluate this intervention and support the improvement of health service delivery. Ethics Primary ethical approval was obtained from the Northern Sydney Local Health District Human Research Ethics Committee (2023/ETH01093), along with site-specific governance approvals. Sengketa tanah akhir-akhir ini menjadi isu yang sangat sentral yang menjadikan skala prioritas segera bisa terselesaikan, ketidakberdayaan lembaga peradilan untuk menyelesaikan penyelesaian sengketa tanah diberbagai wilayah, menjadikan kepercayaan masyarakat berkurang. Oleh karena itu diperlukan penyelesaian lain (opsi lain), untuk mengatasi berbagai kasus penyelesaian pertanahan di negerini ini. Yaitu penyelesaiannya di luar pengadilan (Non Litigasi) Kota Depok, daerah Propinsi Jawa Barat, berbagai persoalan pertanahan banyak sekali yang tidak bisa diselesaikan, ketidakmampuan lembaga pemerintah Kantor Pertanahan Kota Depok, untuk segera mengatasi mungkin persoalan tersebut biar tidak menambah persoalan menjadi semakin sulit. Permasalahan bidang pertanahan di wilayah Kota Depok Jawa Barat mempengaruhi berbagai faktor, karena wilayah depok yang bersinggungan langsung dengan pusar pemerintahan DKI Jakarta, sehingga kebutuhan tanah menjadi hal yang tidak bisa dihindari, hal ini menjadi permasalahn dibidang pertanahan menjadi eskalali tinggi. Untuk mencari solusi agar permasalahan tersebut dapat diatasi, harapan permasalahan selesai maka tanah dapat dimanfaatkan secara ekonomi. Salah satu alternatif solusi untuk menyelesaikan penyelesaian pertanahan dengan melalui Mediasi (Non Litigasi), disisi lain hal tersebut dapat dilakukan secara efektif dan efisien, dengan biaya murah, waktu singkat, dengan sebuah syarat bahwa para pihak dalam penyelesaian pertanahan dapat menerima dengan rasa keadilan. Melalui kegiatan pengabdian kepada masyarakat ini dalam bentuk penyuluhan hukum yang bertemakan “Penyuluhan Hukum Penyelesaian Sengketa Tanah Atau Lahan Melalui Jalur Non Litigasi atau Diluar Ranah Pengadilan Diwilayah Kelurahan Pangkalan Jati Baru Kecamatan Cinere Kota Depok” diharapkan bisa memberikan solusi atau opsi bagi permasalahan yang saat ini masih terjadi di Kelurahan Pangkalan Jati Baru Kecamatan Cinere Kota Depok. Kegiatan pengabdian kepada masyarakat ini sudah sebelumnya pernah dilaksanakan dua kali tahun 2021 dan tahun ini 2022 dengan tema berbeda. Kegiatan Abdimas ini dilakukan dengan menggunakan metode ceramah, diskusi dengan menghadirkan perangkat kelurahan, tokoh masyarakat, ketua RT, RW yang ada dilingkungan kelurahan Pangkalan Jati Baru. Kesimpulan Dari hasil kegiatan Abdimas masyarakat sangat antusias banyaknya pertanyaan-pertanyaan yang dilontarkan kepada penyaji dan keingintahuan tentang pertanahan sangat serius, peserta Abdimas menyarankan kegiatan pengabdian masyarakat ini bisa berkesinambungan memberikan wacana atau gambaran hal hal yang berkaitan dengan penebangan lahan atau dengan tema yang lain
