Abstract

International audience; Gas-phase studies of biomolecules are often difficult to initiate because of the thermolability of these systems. Such studies are nevertheless important to determine fundamental intrinsic properties of the molecules. Here we present the valence shell photoionization of gas-phase vitamins A and B1 close to their ionization threshold. The study was performed by means of an aerosol thermodesorption source coupled to an electron/ion coincidence spectrometer and synchrotron radiation (SOLEIL facility, France). Ion yield curves were recorded for both molecules over a few electronvolt energy range and the threshold photoelectron spectrum was also obtained for vitamin A. Some fundamental properties were extracted for both ions such as adiabatic and the three first vertical ionization energies of retinol (IEad = 6.8 ± 0.2 eV and IEvert = 7.4, 8.3, and 9.2 eV) and dissociation appearance energies for the main fragment ions of vitamin B1. Analysis of the data was supported by ab initio calculations which show a very good agreement with the experimental observations.

Abstract

RATIONALE High-energy collision-induced dissociation (CID) spectra of isomeric RNA/DNA chimeras using matrix-assisted laser desorption/ionization time-of-flight LIFT mass spectrometry (MALDI-LIFT-TOF/TOF) can potentially be applied for an exhaustive fragment characterization in a nucleic acid sequencing scheme. These chimeras contain deoxynucleotides and at the 3'-end a ribonucleotide with a 3'-phosphate group. METHODS Deprotonated RNA/DNA chimeras of 4-, 5-, 7- and 10-mers are analyzed by CID. This enhances consecutive dissociations from both the precursor and prompt product anions generated by MALDI and metastable fragmentations prior to entering the LIFT cell. RESULTS Gas-phase fragmentations of 4- and 5-mers produced many fragment ions, from base release prior to consecutive cleavage of the nucleotide phosphate bond linkage phosphate. The unusual a4− product ion is a specific and diagnostic dissociation of the 4-mer if the ribonucleotide contains cytosine. As the size of RNA/DNA chimeras increase, several abundant product ions are generated mainly from zwitterionic forms (deprotonated phosphate ester and protonated base sites): [(M-H)–BiH]−, [ai–BiH]−, wj−, [wj, (ai-BiH)]− (if Bi ≠ T) as internal product ion, and more rarely [wj–BiH]−. The absence of the majority of the [ai–BiH]− series although the wj− series suggested that the higher critical energy processes with a loose transition state are favored yielding the wj− series. A large number of abundant fragment ions are detected which enable each isomer to be sequenced. CONCLUSIONS This sequencing method is high-throughput, accurate and could be used to sequence isomers of up to 10-mers and also oligonucleotides of unknown sequence. However, RNA/DNA chimeras without thymine must be sufficiently concentrated to reach desorption of deprotonated molecular species to be selected in LIFT to produce all fragment ions within measurable abundances. Abstract

Photoelectron circular dichroism (PECD), a forward–backward asymmetry along the light propagation direction observed in the angular distribution of photoelectrons formed in the ionization of a chiral gas phase target with circularly polarized light, is becoming an established technique for chiral differentiation. In this work some of the fundamental and analytical properties of PECD are confirmed and explored further through a comparative study of the valence shell photoionization of enantiomerically pure trifluoromethyl-oxirane and methyl-oxirane, namely the sensitivity of PECD to the initial orbital and to chemical substitution. The recorded PECD experimental data and corresponding continuum multiple scattering calculations for the outermost orbitals obtained at various photon energies reveal the dramatic effect of substituting the CF3 and CH3 groups attached at the asymmetric chiral center. The previously unknown trifluoromethyl-oxirane ion spectroscopy and the fragmentation pattern measured by threshold electron/ion coincidence techniques over the first four eVs above the ionization threshold are also presented in this work and assigned through the use of ab initio calculations. The state-selected photochemistry and threshold electron spectroscopy of methyl-oxirane have additionally been recorded to complement previous spectroscopic studies.

Abstract

Reproducibility among different types of excitation modes is a major bottleneck in the field of tandem mass spectrometry library development in metabolomics. In this study, we specifically evaluated the influence of collision voltage and activation time parameters on tandem mass spectrometry spectra for various excitation modes [collision-induced dissociation (CID), pulsed Q dissociation (PQD) and higher-energy collision dissociation (HCD)] of Orbitrap-based instruments. For this purpose, internal energy deposition was probed using an approach based on Rice-Rampserger-Kassel-Marcus modeling with three thermometer compounds of different degree of freedom (69, 228 and 420) and a thermal model. This model treats consecutively the activation and decomposition steps, and the survival precursor ion populations are characterized by truncated Maxwell-Boltzmann internal energy distributions. This study demonstrates that the activation time has a significant impact on MS/MS spectra using the CID and PQD modes. The proposed model seems suitable to describe the multiple collision regime in the PQD and HCD modes. Linear relationships between mean internal energy and collision voltage are shown for the latter modes and the three thermometer molecules. These results suggest that a calibration based on the collision voltage should provide reproducible for PQD, HCD to be compared with CID in tandem in space instruments. However, an important signal loss is observed in PQD excitation mode whatever the mass of the studied compounds, which may affect not only parent ions but also fragment ions depending on the fragmentation parameters. A calibration approach for the CID mode based on the variation of activation time parameter is more appropriate than one based on collision voltage. In fact, the activation time parameter in CID induces a modification of the collisional regime and thus helps control the orientation of the fragmentation pathways (competitive or consecutive dissociations).

Abstract

Glyceollin-related metabolites produced in rats following oral glyceollin administration were screened in plasma, feces, and urine, and these metabolites were identified by precursor and product ion scanning using liquid chromatography coupled online with electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS). Precursor ion scanning in the negative ion (NI) mode was used to identify all glyceollin metabolites based on production of a diagnostic radical product ion (m/z 148) upon decomposition. Using this approach, precursor peaks of interest were found at m/z 474 and 531. Tandem mass spectra of these two peaks allowed us to characterize them as byproducts of glutathione conjugation. The peak at m/z 474 was identified as the deprotonated cysteinyl conjugate of glyceollins with an addition of an oxygen atom, whereas m/z 531 was identified as the deprotonated cysteinylglyceine glyceollin conjugate plus an oxygen. These results were confirmed by positive ion (PI) mode analyses. Mercapturic acid conjugates of glyceollins were also identified in NI mode. In addition, glucuronidation of glyceollins was observed, giving a peak at m/z 513 corresponding to the deprotonated conjugate. Production of glucuronic acid conjugates of glyceollins was confirmed in vitro in rat liver microsomes. Abstract

According to the World Health Organization, food safety is an essential public health priority. In this context, we report a relevant proof of feasibility for the indirect specific detection of bacteria in food samples using unlabeled phage amplification coupled to ESI mass spectrometry analysis and illustrated with the model phage systems T4 and SPP1. High-resolving power mass spectrometry analysis (including bottom-up and top-down protein analysis) was used for the discovery of specific markers of phage infection. Structural components of the viral particle and nonstructural proteins encoded by the phage genome were identified. Then, targeted detection of these markers was performed on a triple quadrupole mass spectrometer operating in the selected reaction monitoring mode. E. coli at 1 × 10(5), 5 × 10(5), and 1 × 10(6) CFU/mL concentrations was successfully detected after only a 2 h infection time by monitoring phage T4 structural markers in Luria-Bertani broth, orange juice, and French bean stew ("cassoulet") matrices. Reproducible detection of nonstructural markers was also demonstrated, particularly when a high titer of input phages was required to achieve successful amplification. This strategy provides a highly time-effective and sensitive assay for bacterial detection.

Abstract

RATIONALE The study of protein recognition sites is crucial for understanding the mechanisms of protein interaction. Mass spectrometry can be a method of choice for the investigation of the contact surface within the protein non-covalent complexes. METHODS Probing the reactivity of essential amino acid residues of soybean Bowman-Birk inhibitor (sBBI) within the non-covalent sBBI/bovine trypsin complex was performed using covalent labeling by the BS3 cross-linker and charge tag with a quaternary ammonium group in combination with matrix-assisted laser desorption/ionization (MALDI) mass spectrometry (MS) and tandem mass spectrometry (MS/MS) analysis. RESULTS Significant modulation of the reactivity of essential K16 and S17 residues in the sBBI molecule upon binding to trypsin was established. The studies of sBBI proteolytic peptides with the same structure but carrying different labels using metastable dissociation in LIFT mode demonstrated that fragmentation pathways were oriented by used modification (BS3 cross-linker or charge tag). CONCLUSIONS The effectiveness of the mass spectrometric approach including covalent modification for exploring protein-protein interaction sites has been demonstrated. The alteration of the reactivity of functionally important amino acid residues in the sBBI molecule is most likely related to changes in their microenvironment. It has been suggested that in the presence of charge tags fragmentation in LIFT mode proceeds through the formation of salt bridges between quaternary ammonium groups and acidic residues due to the occurrence of zwitterions (including basic and acidic residues). Despite the presence of one or several charge tags, fragmentation takes place yielding modulated bi/yj ion series depending on the positions of the tags. Copyright © 2014 John Wiley & Sons, Ltd.

Abstract

The "Best Match" model has been extended to account for the role that Na(+)/H(+) exchange plays on anion attachment in negative ion electrospray. Without any Na(+)/H(+) exchange on (Glu) fibrinopeptide B, the higher basicity anions F(-) and CH3COO(-) can hardly form observable adducts; however, after multiple Na(+)/H(+) exchanges, adduct formation is enabled. Moreover, dissociation pathways of CF3COO(-) adducts with singly deprotonated peptides that have undergone 0 to 3 Na(+)/H(+) exchanges exhibit a shift in CID product ions from losing predominately CF3COOH (case of 0 Na(+)/H(+) exchanges) to losing predominately CF3COO(-) (case of 3 Na(+)/H(+) exchanges). These phenomena can be rationalized by considering that Na(+) cations exchange at, and serve to "block", the most acidic sites, thereby forcing implicated anions to attach to lower acidity protons. In addition to forming ion pairs with carboxylate groups, Na(+) also participates in formation of tri-atomic ions of the form ANaA(-) during adduct dissociation. The fact that low gas-phase basicity (GB) anions preferentially form ANaA(-) species, even though high GB anions form more stable tri-atomic species, indicates that the monatomic ions were not in close contact in the initial adduct. The propensity for formation of stable anionic adducts is dependent on the degree of matching between anion GBs and GBapp of deprotonated sites on the peptide. The GBapp is raised dramatically as the charge state of the peptide increases via a through-space effect. The presence of Na(+) on carboxylate sites substantially decreases the GBapp by neutralizing these sites, while slightly increasing the intrinsic GBs by an inductive effect.

Abstract

Background: Horse urine is the medium of choice for the implementation of metabolomic approaches aimed at improving horse doping control. However, drug analysis in this biofluid is a challenging task due to the presence of large amounts of interfering compounds. Methodology & Results: A comparative study of sample preparation has been conducted to evaluate five sample-preparation methods, namely acetonitrile precipitation, proteinase K hydrolysis, membrane filtration and sample dilution with water by factors of five and 20, for metabolome analysis using liquid chromatography coupled to high resolution mass spectrometry. Assessment was performed at both global and targeted levels, by using a few thousand features obtained from peak detection software, and internal standards and 100 annotated or identified metabolites. Conclusion: By considering the number of detected signals, their intensity and their detection repeatability, acetonitrile precipitation was selected as the most efficient sample-preparation method for the analysis of horse urine metabolome in liquid chromatography coupled to high resolution mass spectrometry conditions.

Abstract

This work aims at evaluating the relevance and versatility of liquid chromatography coupled to high resolution mass spectrometry (LC/HRMS) for performing a qualitative and comprehensive study of the human serum metabolome. To this end, three different chromatographic systems based on a reversed phase (RP), hydrophilic interaction chromatography (HILIC) and a pentafluorophenylpropyl (PFPP) stationary phase were used, with detection in both positive and negative electrospray modes. LC/HRMS platforms were first assessed for their ability to detect, retain and separate 657 metabolite standards representative of the chemical families occurring in biological fluids. More than 75% were efficiently retained in either one LC-condition and less than 5% were exclusively retained by the RP column. These three LC/HRMS systems were then evaluated for their coverage of serum metabolome. The combination of RP, HILIC and PFPP based LC/HRMS methods resulted in the annotation of about 1328 features in the negative ionization mode, and 1358 in the positive ionization mode on the basis of their accurate mass and precise retention time in at least one chromatographic condition. Less than 12% of these annotations were shared by the three LC systems, which highlights their complementarity. HILIC column ensured the greatest metabolome coverage in the negative ionization mode, whereas PFPP column was the most effective in the positive ionization mode. Altogether, 192 annotations were confirmed using our spectral database and 74 others by performing MS/MS experiments. This resulted in the formal or putative identification of 266 metabolites, among which 59 are reported for the first time in human serum.

Abstract

An hapten 1 designed for the production of catalytic antibodies was synthesized after coupling its precursor 2 to bovine serum albumin (BSA). The conjugates BSA- 2 and BSA- 1 were characterized by MALDI mass spectrometry. This paper shows that besides the average number of molecules bound to the protein deduced from the molecular ion peak shift, the MALDI technique can also give access to their distribution, bv simulation of the peak.

Abstract

The reactivity of the explosive tetryl (N-methyl-N,2,4,6-tetranitroaniline; Mw = 287 u) was studied using electrospray ionization in negative mode. The main species detected in the spectrum corresponds to the ion observed at m/z 318 (previously assumed to be the odd-electron ion [tetryl + HNO]-•, C7H6O9N6). In this study, we show using D-labeling combined with high-resolution mass spectrometry that this species corresponds to an even-electron anion (i.e. C8H8O9N5), resulting from the formation of a Meisenheimer complex between tetryl and the methanol used as the solvent. Fragmentation of this complex under CID conditions revealed an unexpected fragment: the formation of a 2,4,6-trinitrophenoxide anion at m/z 228. 18O-labeling combined with quantum chemical calculations helped us better understand the reaction pathways and mechanisms involved in the formation of this product ion. This occurs via a transition state leading to a SN2-type reaction, consequently evolving toward an ion-dipole complex. The latter finally dissociates into deprotonated picric acid.

Abstract

International audience; The transcriptional activity of human estrogen receptor ERα is modulated by a number of coregulatory proteins among which calmodulin (CaM). Segment 295-311 in the hinge region of ERα has previously been proposed to be the CaM binding site. In this work, we investigate the molecular mechanism of the interaction of CaM with peptides derived from the hinge region of ERα, using a biophysical approach combining isothermal titration calorimetry, fluorescence, CD and NMR. The ERα17p peptide, corresponding to the previously identified 295-311 region of ERα, recruits mainly the C-terminal domain of Ca(4)CaM, as shown by NMR spectroscopy. In contrast, a longer peptide, ERα25p, extended on the N-terminal side (residues 287-311) interacts with both N- and C-terminal domains of Ca(4)CaM. These results lead to a new delineation of the CaM binding site, encompassing residues 287-294. In particular, fluorescence spectroscopy reveals that the conserved W(292) residue is engaged within hydrophobic pockets on Ca(4)CaM. ITC results show that ERα25p binds Ca(4)CaM with an atypical 2:1 stoichiometry and a dissociation constant in the micromolar range. Based on the NMR titration of Ca(4)CaM by ERα25p showing a biphasic behavior for several residues, we suggest that concerted conformational changes of CaM domains may be required to accommodate the binding of a second peptide. CD spectra indicate that ERα25p partially folds into an α-helix upon binding to Ca(4)CaM. Hence, ERα25p is a new CaM-binding ligand that could be appropriate for the synthesis of derivatives able to control ER-dependent transcription, particularly in the context of hormone-dependent breast tumors.

International audience; The structure and behavior of the catalytic species involved in the PAPd- and PAPt-catalyzed formal [2 + 1] cycloaddition between an alkyne and norbornadiene have been studied by combined mass spectrometry experiments and density functional theory calculations. The first step of this reaction, the coordination of the catalyst with the alkyne, is also investigated under collision-activated reaction conditions. Two different structures are proposed for the key intermediates: an acetylide structure for Pd and a metallacarbene for Pt.

Abstract

Metabolomics opens new perspectives for biomarker discovery in the field of nutrition and heath, and also in the development of system biology. The metabolic profiles of biofluids obtained by mass spectrometry or nuclear magnetic resonance contain a few hundreds to thousands of signals. However, a major part of this information remains unknown, or at least not characterized in the analytical systems, thus hampering the obtention of biologically meaningfull data. In this context, we here report on the development of an electrospray (ESI) spectral database for the annotation of high resolution mass spectrometry based metabolomics data sets and on its application to the analysis of urine samples from a cohort of healthy volunteers.

Abstract

The intermediacy of gold acetylides in the gold(I)-catalyzed cycloisomerization of enynes was questioned. While dinuclear gold complexes are observed under electrospray ionization conditions, the solution reactivity of gold acetylides also leads to the conclusion of their high affinity for the second coordination of a gold moiety, leading to dinuclear gold complexes. However, the involvement of gold acetylides and the corresponding diaurated species in the elementary steps of cycloisomerization mechanisms of enynes appears unlikely.

Abstract

Electrospray ionization mass spectrometry was used to investigate the intrinsic chemical properties of β-carbolines: Eudistomin U (as EU) and 5-Br-Eudistomin U (as BrEU) yielding self-association in negative ion mode. It was observed that the deprotonated homodimer of EU was more stable in the gas phase than BrEU. Indeed, BrEU presents a higher polarizability that could influence the dimerization energy. On the contrary, the stability energies calculated at B3LYP/6-31+G*//AM1 level showed a small difference for these two dimers. To understand the experimental stability variation, the experimental and theoretical gas phase acidities of these two β-carbolines have been estimated. It has been shown that EU is less acidic than BrEU in the gas phase, which could explain the lower stability of the [2BrEU-H]− dimer. In addition, the investigation of non-covalent interaction of β-carbolines and nucleobases has been carried out. It was experimentally demonstrated that the deprotonated heterodimer's relative stability scale was [Gua + M−H]− >> [Thy + M−H]− > [Ade + M−H]−, but the Cyt/β-carboline complexes were not observed. However, the absence of Cyt/β-carboline complexes could be explained by the great acidity difference between Cyt and β-carbolines, yielding unstable non-covalent complexes. The studied β-carbolines showed a particular affinity with guanine which suggested a strong interaction, from a structural and reactivity point of view. Calculated reaction pathways rationalize the set of experimental results concerning the heterodimer stabilisation reflected by the V1/2 value of breakdown curves of deprotonated heterodimers.

Abstract

PtII-catalyzed cycloisomerization of a 1,6-enyne proceeds quickly in CHCl3 at room temperature, and also in the gas phase. However, calculations predict a slow reaction, because of the formation of a very stable chelate that undergoes oxidative cyclization at a high energy cost. The electrophilic activation of the alkyne followed by nucleophilic attack of the double bond to generate a cyclopropyl carbene would lead to a faster reaction, but the decomplexation of the double bond to generate an unsaturated platinum species is also energetically disfavored. However, decomplexation can be envisaged if an adventitious molecule of water enters the coordination sphere of the metal. This crucial role of water in PtII-catalyzed cycloisomerization of enynes, initially sensed some years ago (Echavarren et al., J. Am. Chem. Soc.2001, 123, 10511) is now supported by mass spectrometry studies.

Abstract

Palladium(II) acetate complexes with phosphinous acids such as tert-butyl(phenyl)phosphinous acid (PAPd) have been successfully applied in a new [2 + 1] cycloaddition reaction between norbornadiene (NBD) and phenylacetylene (PhCCH) to produce exo-phenylmethylidenecyclopropane in the solution phase. In the present study, collision-activated reaction (CAR)/collision-induced dissociation (CID) mass spectral experiments were performed on a cationic Pd(II) complex (Pd(OPPhtBu)2H ion, [a]+). These experiments were useful for the identification of the reactive intermediates in the gas phase. On the basis of the results of the collision-activated reactions on this particular Pd(II) complex ion with phenylacetylene and norbornadiene, we demonstrated the essential role of the order of addition of organic partners on the efficiency to form the diadduct ion [a + PhCCH + NBD]+. Abstract

International audience; Covalent protein-nucleic acid conjugates form an original class of compounds that occur in nature or can be generated in vitro through cross-linking to investigate domains involved in protein/nucleic acid interactions. Their mass spectrometry fragmentation patterns are poorly characterized. We have used electrospray-ionization mass spectrometry (ESI-MS) combined with collision-induced dissociation (CID) to characterize microcin C7-C51, an antimicrobial nucleotide peptide that targets aspartyl-tRNA synthetase and inhibits translation. The fragments of microcin C7-C51 were analyzed in positive- and negative-ion modes and compared with those of the corresponding unmodified heptapeptide and to the derived aspartyl-adenylate. The positive- and negative-ion mode fragments of microcin C7-C51 provided information on both the nucleotide and peptide moieties. Accurate mass measurement obtained using an LTQ Orbitrap instrument was a key factor for a comprehensive interpretation of the fragments. The experimental results obtained permitted the proposal of stepwise fragmentation pathways involving ion-dipole complexes. The data provide a better understanding of nucleotide peptide fragmentation in the gas phase.

Abstract

International audience; Guanine tetraplexes are biological non-covalent systems stabilized by alkali cations. Thus, self-clustering of guanine, xanthine and hypoxanthine with alkali cations (Na(+), K(+) and Li(+)) is investigated by electrospray ionization mass spectrometry (ESI-MS) in order to provide new insights into G-quartets, hydrogen-bonded complexes. ESI assays displayed magic numbers of tetramer adducts with Na(+), Li(+) and K(+), not only for guanine, but also for xanthine bases. The optimized structures of guanine and xanthine quartets have been determined by B3LYP hybrid density functional theory calculations. Complexes of metal ions with quartets are classified into different structure types. The optimized structures obtained for each quartet explain the gas-phase results. The gas-phase binding sequence between the monovalent cations and the xanthine quartet follows the order Li(+) > Na(+) > K(+), which is consistent with that obtained for the guanine quartet in the literature. The smallest stabilization energy of K(+) and its position versus the other alkali metal ions in guanine and xanthine quartets is consistent with the fact that the potassium cation can be located between two guanine or xanthine quartets, for providing a [gua(or (xan))(8)+K](+) octamer adduct. Even if an abundant octamer adduct with K(+) for xanthine was detected by ESI-MS, it was not the case for guanine.

En 2002, le Prix Nobel de Chimie a été décerné à trois grands scientifiques, John B. Fenn et Koichi Tanaka pour la spectrométrie de masse et Kurt Wüthrich pour la résonance magnétique nucléaire. Il récompense ainsi les idées innovantes qui ont conduit à développer, à l’interface chimie-biologie, des méthodologies spectroscopiques majeures initialement introduites en chimie analytique. La découverte de nouvelles méthodes douces d’ionisation en spectrométrie de masse, et la conception d’une stratégie d’attributions séquentielles des signaux de résonance magnétique nucléaire de biopolymères, ont permis l’application de ces deux méthodologies puissantes et complémentaires à la détermination de structures de macromolécules du vivant, protéines, acides nucléiques (ADN, ARN) et polysaccharides, etc. Par l’élucidation de leurs édifices (structures planes et tridimensionnelles) et des mécanismes moléculaires régissant leurs interactions, ces travaux fournissent des clés précieuses pour la compréhension des mécanismes moléculaires du vivant., The Nobel Prize in Chemistry for 2002 has been awarded to two powerful spectroscopic methodologies through three valorous scientists, John Fenn and Koichi Tanaka, for mass spectrometry and Kurt Wüthrich for nuclear magnetic resonance. These techniques were previously known for their intensive use in chemical analysis. They are now developed at the chemistry/biology interface. Two new methods of soft ionization in mass spectrometry and a strategy of sequential assignment of nuclear magnetic resonance signals of biopolymers now allow the use of these powerful and complementary methodologies for the structural analysis of biological macromolecules, proteins, nucleic acids (DNA, RNA) and polysaccharides. Through the elucidation of their planar and three-dimensional structures and of the molecular mechanisms that govern their interactions, these techniques now may afford precious clues for understanding the molecular mechanisms of life.

Abstract

Estrogen 2,3- and 3,4-quinones are reactive species toward nucleophiles and Michael acceptors. As such, they can bind to DNA and induce cellular damages. As an alkylation model, reactions of estradiol-2,3-quinone with deoxyribonucleosides were previously studied by mass spectrometry. In this work, estrogen-deoxyribonucleoside adducts were synthesized by reaction of 17beta-estradiol-2,3-quinone with deoxyguanosine or deoxyadenosine and analyzed by NMR and LC-MS(n)() in order to determine the structure and the stereochemistry of the resulting covalent adducts. Although estradiol- and estrone-2,3-quinones were previously thought to give mainly stable adducts, identification of depurinating adducts with both nucleosides, i.e., 2-OHE(2)-6(alpha,beta)-N7Gua and 2-OHE(2)-6(alpha,beta)-N7Ade, was unambiguously obtained. This is of particular interest since depurinating adducts are generated from DNA, and therefore, their amount should be correlated to the parallel formation of apurinic sites, which might play an important role in the cancer initiation process. Besides, a byproduct, i.e., 2-hydroxy-11-oxo-estradiol, corresponding to an unstable alkylation product of 2-hydroxyestradiol has been unambiguously identified and is indicative of a plausible addition process at the C9 position of catechol estrogens. The synthetic adducts will be useful as reference compounds to further elucidate the structure of adducts formed by reaction of estrogen metabolites with DNA or oligonucleotides.

Abstract

The synthesis and characterisation of symmetrical glycosylated neutral and cationic porphyrin dimers linked at the meso -position via a flexible hydrocarbon chain to improve targeting on malignant cells is reported. Photocytotoxicity of these compounds against the K562 leukemia cell line compared to the effect of hematoporphyrin is also presented.

Abstract

High‐resolution mass spectrometry (HRMS) has become a vital tool for dissolved organic matter (DOM) characterization. The upward trend in HRMS analysis of DOM presents challenges in data comparison and interpretation among laboratories operating instruments with differing performance and user operating conditions. It is therefore essential that the community establishes metric ranges and compositional trends for data comparison with reference samples so that data can be robustly compared among research groups. To this end, four identically prepared DOM samples were each measured by 16 laboratories, using 17 commercially purchased instruments, using positive‐ion and negative‐ion mode electrospray ionization (ESI) HRMS analyses. The instruments identified ~1000 common ions in both negative‐ and positive‐ion modes over a wide range of m/z values and chemical space, as determined by van Krevelen diagrams. Calculated metrics of abundance‐weighted average indices (H/C, O/C, aromaticity, and m/z) of the commonly detected ions showed that hydrogen saturation and aromaticity were consistent for each reference sample across the instruments, while average mass and oxygenation were more affected by differences in instrument type and settings. In this paper we present 32 metric values for future benchmarking.
