Multivariate logistic regression has shown that the following variables are independent risk factors for preeclampsia: age, BMI, parity, and twin pregnancy. Moreover, preeclampsia is significantly associated with the examined clinical effects, confirming itself as a leading cause of maternal-fetal morbidity.

Abstract

Objectives The spread of carbapenem-resistant Enterobacteriaceae (CRE) represents one of the most worrisome problems for clinical medicine worldwide. In Italy, the Antibiotic-Resistance-Istituto Superiore di Sanita surveillance network, in collaboration with the Committee for Antimicrobial Agents of the Italian Society of Clinical Microbiologists, promoted a study to investigate the carbapenem-resistance mechanisms, clonal relatedness and capsular typing of a recent collection of carbapenem-resistant Klebsiella pneumoniae (CR-KP). Methods A total of 17 laboratories distributed across Italy collected all consecutive non-replicate CR-KP isolated from invasive infections during two different study periods (2011-12 and 2013). Carbapenemase genes were searched for by filter hybridization and confirmed by PCR and sequencing. KPC-producing K. pneumoniae (KPC-KP) were typed by PFGE and MLST. Capsular types were identified by wzi gene typing. Results Of the collected K. pneumoniae isolates (n = 461), the overall proportion of CR-KP was 36.2% (n = 167). The majority (97%) of the CR-KP were positive for the blaKPC gene. Among the KPC-KP population, nine different STs were detected with the majority of isolates (94%) belonging to the clonal group (CG) 258. A subpopulation that belonged to ST512 and showed an identical PFGE profile represented the majority (57%) of KPC-KP strains, with a countrywide distribution. Capsular characterization showed the predominance of the wzi154, cps-2 capsular type (88.8% of all CG258 strains). ST258 strains were associated with both cps-1 and cps-2 capsular types, while ST512 was associated with cps-2 only. Conclusions Although a trend to a polyclonal evolution of the Italian KPC-KP was noted, this study showed that the KPC-KP population remained largely oligoclonal with the wide diffusion of an ST512 lineage carrying cps-2 capsular type and producing the KPC-3 enzyme.

Abstract

Invasive Group A Streptococcus disease is a severe and sometimes life-threatening infection with only few cases reported in literature. We describe the case of a 49-day-old male infant with invasive Group A Streptococcus infection characterized by acute otitis media and development of septicemia within a probably community-acquired cluster. The causative agent resulted to be a rare emm-89 genotype of Streptococcus pyogenes. Group A Streptococcus must be considered responsible for sepsis in newborns and young infants.

Abstract

Studio di fattibilità a Mazara del Vallo: questo studio descrive la valutazione geotermica effettuata nel sito di Mazara del Vallo e la proposta tecnico-economica per lo sviluppo di un progetto impiantistico relativo a risorse geotermiche a media entalpia e la realizzazione di un impianto pilota per alimentare una rete di teleriscaldamento per il riscaldamento ed il raffrescamento di edifici pubblici presenti nel Comune di Mazara del Vallo. La verifica della possibilità di utilizzo della risorsa geotermica per la realizzazione dell'impianto, e la valutazione quindi della sua realizzabilità, è avvenuta mediante indagini geologiche, idrogeologiche, geochimiche, geofisiche e la realizzazione di una specifica simulazione numerica dinamica dei parametri fisico-chimici del serbatoio. L'impianto proposto consiste nella produzione centralizzata di energia termica (acqua calda a 90 °C) e la successiva produzione decentralizzata di acqua fredda mediante gruppi frigoriferi ad assorbimento localizzati presso singole e/o gruppi di utenze.

Abstract

Consecutive non-replicate clinical isolates (n=191) of carbapenem non-susceptible Enterobacteriaceae were collected from 21 hospital laboratories across Italy from November 2013 to April 2014 as part of the European Survey on Carbapenemase-producing Enterobacteriaceae (EuSCAPE) project. Klebsiella pneumoniae carbapenemase-producing K. pneumoniae (KPC-KP) represented 178 (93%) isolates with 76 (43%) respectively resistant to colistin, a key drug for treating carbapenamase-producing Enterobacteriaceae. KPC-KP colistin-resistant isolates were detected in all participating laboratories. This underscores a concerning evolution of colistin resistance in a setting of high KPC-KP endemicity.

Abstract

Three epidemiologically independent Pseudomonas aeruginosa isolates, representative of the first VIM-1 metallo-beta-lactamase producers detected at three different hospitals in northern Italy, were investigated to determine their genomic relatedness and to compare the structures of the genetic supports for the VIM-1 determinants. The three isolates, all of serotype O11, appeared to be clonally related according to the results of genotyping by macrorestriction analysis of genomic DNA by pulsed-field gel electrophoresis and random amplification of polymorphic DNA. Investigation of the genetic support for the bla(VIM-1) determinant revealed that it was carried on identical or almost identical integrons (named In70.2 and In70.3) located within a conserved genomic context. The integrons were structurally related to In70 and In110, two plasmid-borne bla(VIM-1)-containing integrons from Achromobacter xylosoxidans and Pseudomonas putida isolates, respectively, from the same geographic area (northern Italy) and were found to be inserted close to the res site of a Tn5051-like transposon, different from any of those described previously, that was apparently carried on the bacterial chromosome. The present findings suggest that the three VIM-1-producing isolates are members of the same clonal complex which have been spreading in hospitals in northern Italy since the late 1990s and point to a common ancestry of their bla(VIM-1)-containing integrons.

Abstract

The production of proinflammatory cytokines is likely to play a major pathophysiological role in meningitis and other infections caused by Haemophilus influenzae type b (Hib). Previous studies have shown that Hib porin contributes to signaling of the inflammatory cascade. We examined here the role of Toll-like receptors (TLRs) and the TLR-associated adaptor protein MyD88 in Hib porin-induced production of tumor necrosis factor alpha (TNF-*) and interleukin-6 (IL-6). Hib porin-induced TNF-* and IL-6 production was virtually eliminated in macrophages from TLR2- or MyD88-deficient mice. In contrast, macrophages from lipopolysaccharide (LPS)-hyporesponsive C3H/HeJ mice, which are defective in TLR4 function, responded normally to Hib porin. Moreover anti-TLR2 antibodies but not anti-TLR4 antibodies significantly reduced Hib porin-stimulated TNF-* and IL-6 release from the human monocytic cell line THP-1. These data indicate that the TLR2/MyD88 pathway plays an essential role in Hib porin-mediated cytokine production. These findings may be useful in the development of alternative therapies aimed at reducing excessive inflammatory responses during Hib infections.

Abstract

Fragile X syndrome (FXS) is the most common form of inherited mental retardation and autism and is caused by the lack of Fragile X Mental Retardation Protein (FMRP), an RNA binding protein involved in RNA metabolism and protein synthesis. Several studies report that FMRP is associated with the pool of mRNAs that go into stress granules (SGs) upon cellular stress. Stress granules are cytoplasmic aggregates that are formed under stress conditions such as exposure to heat, oxidative agents, UV irradiation; these foci are thought to serve as sites of mRNA storage or triage during the cell stress response. SGs contain translation initiation components and many additional proteins affecting mRNA function. After environmental stress, mRNAs stored in this abortive translation initiation complex can be routed to either translation initiation or degradation. The link between SG formation and cell survival after stress is poorly understood. In this study we tested the appearance of SGs and the cell survival in wild type (WT) and Fmr1 knockout (KO) cultured astrocytes after exposure to sodium arsenite (200 uM for 1 hour or 500 µM for 30 minutes) and hydrogen peroxide (500 uM for 1 hour), by immunocytochemistry using an anti-TIA antibody and MTT assay, respectively. We also tested the effect of (S)-3,5-Dihydroxyphenylglycine (DHPG, 100 uM for 5 minutes), a selective agonist of group-I metabotropic glutamate (mGlu) receptor subtype1/5 on both SG formation and oxidative stress induced damage in both WT and Fmr1 KO astrocytes. We observed an increased number of cells with granules after exposure to oxidative stress, as expected; however, we detected a lower number of cells with granules in Fmr1 KO than WT astrocytes both under basal condition and after exposure to oxidative stress. Interestingly, under the same oxidative stress conditions cell survival was lower in Fmr1 KO than WT astrocytes. The activation of mGlu receptors before oxidative stress reduced both SG formation and cell survival in WT but had no effect in Fmr1 KO astrocytes. These results indicate that the lack of FMRP impairs the formation of SGs and sensitize to oxidative stress-induced damage. Furthermore, the observed differential effect of group-I mGlu receptors activation on both SG formation and cell survival in the presence and absence of FMRP suggests an interesting signalling interplay among SGs and mGlu receptors which may be related to their involvement in FMRP-mediated RNA transport and protein synthesis.

Abstract

The Linear Collider Forum is an initiative born within the Helmholtz-Alliance ”Physics at the Terascale”, a network of German Universities and Research Centers working in high-energy particle physics. Yearly dedicated meetings focus on the physics case and the technologies at a future linear collider, covering both the ILC and CLIC concepts. In February 2012 an extended meeting with significant international participation was dedicated to the discussion of physics scenarios to strengthen and update the case for a linear e+e- collider in light of new developments and the latest LHC results. Contributions to LC Forum sessions were collected on the DESY LC-note server with a dedicated mark ‘LC-REPyear- number’. These Proceedings comprise all contributions that have been submitted with the dedicated mark in 2012 and 2013. Therefore also contributions are included that actually have not been presented on a LC Forum meeting but are strongly related to the subject.

84. Surface expression and axon/dendritic distribution of mGlu5 receptors in hippocampal wild type and Fmr1 knockout neurons

Abstract

Fragile X Syndrome (FXS) is a leading genetic cause of intellectual disability. It is caused by the lack of expression of Fragile X Mental Retardation Protein (FMRP), an RNA-binding protein involved in mRNA transport, stability and translation. A growing number of studies supports a role of group-I mGlu receptors (mGlu1 and mGlu5) in the pathophysiology of FXS. However, the expression of mGlu5 receptors have not been systematically investigated in FXS mouse model. In FXS a reduced number of mGlu5 receptors are associated to Homer proteins, a class of scaffolding proteins which anchor mGlu receptors to post-synaptic density (Giuffrida et al., J Neurosci., 2005). Considering that mGlu5-Homer interaction regulates the surface expression and axonal/dendritic distribution of mGlu5 receptors (Ango et al., J Neurosci., 2000; Mol Cell Neurosci 2002), we hypothesised that mGlu5 receptors might be differently targeted to cell surface of Fmr1 knock out (KO) neurons. We studied the expression of mGlu5 receptors in hippocampal synaptic proteins prepared from mice of different ages by Western blot analysis and the surface expression of mGlu5 receptors in hippocampal cultured neurons by confocal analysis of surface fluorescence-labeled receptors (kind gift of Prof. Shigemoto). Hippocampal cell cultures were prepared from wild type (WT) and Fmr1 KO newborn mice (P0-P1) and immunocytochemistry was performed at different days in vitro (3, 7, 12 DIV). Double- or triple-labelling experiments with anti-MAP2 and/or anti-tau1 antibodies were carried out to visualize the presence of mGlu5 receptors in dendrites and axons, respectively. Our results show that: a) mGlu5 receptors are more expressed in hippocampal synaptic proteins at postnatal day (P) 21 and 45, but not at 15 month of age; b) mGlu5 receptors are more expressed on the cell surface in Fmr1 KO than in WT cultured neurons. Interestingly, mGlu5 receptors were clearly detectable in the cellular body and dendrites in WT neurons, whereas in Fmr1 KO neurons a strong staining was also observed in the axons. Our data suggest that mGlu5 receptors are differently targeted to the cell surface and differently distributed in dendrites and axons in WT and Fmr1 KO mice.

86. Molecular Testing for Trichomonas vaginalis in Women: Results from a Prospective U.S. Clinical Trial

Abstract

Trichomoniasis is a common sexually transmitted disease associated with preterm birth, low birth weight, and increased susceptibility to infection with other pathogenic sexually transmitted microorganisms. Nucleic acid amplification tests for Trichomonas vaginalis have improved sensitivity for detecting infected individuals compared to existing culture-based methods. This prospective, multicenter U.S. clinical trial evaluated the performance of the automated Aptima T. vaginalis assay for detecting T. vaginalis in 1,025 asymptomatic and symptomatic women. Vaginal swab, endocervical swab, ThinPrep PreservCyt, and urine specimens were collected. Subject infection status was determined by wet-mount microscopy and culture. Aptima T. vaginalis assay performance was determined for each specimen type by comparison to subject infection status. Of 933 subjects analyzed, 59.9% were symptomatic. Aptima T. vaginalis clinical sensitivity and specificity were, respectively, 100% and 99.0% for vaginal swabs, 100% and 99.4% for endocervical swabs, 100% and 99.6% in ThinPrep samples, and 95.2% and 98.9% in urine specimens. Aptima T. vaginalis performance levels were similar in asymptomatic and symptomatic subjects. This study validates the clinical performance of the Aptima T. vaginalis assay for screening asymptomatic and symptomatic women for T. vaginalis infection.
