Summary: PARP inhibitors (PARPi) are efficacious in BRCA1-null tumors; however, their utility is limited in tumors with functional BRCA1. We hypothesized that pharmacologically reducing BRCA1 protein levels could enhance PARPi effectiveness in BRCA1 wild-type tumors. To identify BRCA1 downregulating agents, we generated reporter cell lines using CRISPR-mediated editing to tag endogenous BRCA1 protein with HiBiT. These reporter lines enable the sensitive measurement of BRCA1 protein levels by luminescence. Validated reporter cells were used in a pilot screen of epigenetic-modifying probes and a larger screen of more than 6,000 compounds. We identified 7 compounds that could downregulate BRCA1-HiBiT expression and synergize with olaparib. Three compounds, N-acetyl-N-acetoxy chlorobenzenesulfonamide (NANAC), A-443654, and CHIR-124, were validated to reduce BRCA1 protein levels and sensitize breast cancer cells to the toxic effects of olaparib. These results suggest that BRCA1-HiBiT reporter cells hold promise in developing agents to improve the clinical utility of PARPi.

IntroductionAlthough hereditary male neoplasms are quite rare, individuals harbouring germline BRCA1/2 pathogenic variants (PVs) may have a risk of developing tumours associated with Hereditary Breast and Ovarian Cancer (HBOC) syndrome, including male breast (MBC), prostate (PCa) and pancreatic (PC) cancers, and melanoma. Women and men showed a comparable genetic architecture of cancer susceptibility, but there are some gender-specific features. Since little is known about cancer genetic susceptibility in male population, our study was aimed at investigating the frequency of BRCA1/2 PVs in men with HBOC syndrome-associated tumors, in order to understand whether differences in gender may reflect in the prevalence and spectrum of germline alterations.Patients and methodsWe retrospectively collected and analysed clinical information of 352 HBOC-associated male cancer patients genetically tested for germline BRCA1/2 PVs by Next-Generation Sequencing analysis, enrolled, from February 2018 to January 2024, at the “Regional Center for the prevention, diagnosis and treatment of rare and heredo-familial tumors of adults” of the University-Hospital Policlinico “P. Giaccone” of Palermo (Italy).ResultsOur investigation revealed that 7.4% of patients was carrier of a germline BRCA PV, with an almost total prevalence of BRCA2 alterations. In particular, 65.4% of BRCA-positive patients developed MBC, 19.2% had PC, 11.6% developed PCa, and only 3.8% had melanoma. Specifically, MBC individuals showed a BRCA-associated genetic predisposition in 17% of cases, whereas patients with PCa or PC exhibited a lower frequency of BRCA2 PVs, taking into account the current national criteria for access to germline genetic testing.DiscussionOur study showed a high heterogeneity in prevalence of germline BRCA2 PVs among men which could reflect a potential gender-specific genetic heterogeneity. Therefore, BRCA-associated male tumours could be due to BRCA2 PVs different from those usually detected in women. In the event that it is demonstrated, in future, that male cancers are genetically distinct entities from those female this could improve personalized risk evaluation and guide therapeutic choices for patients of both sexes, in order to obtain a gender equality in cancer care.

Transformer-based models have demonstrated excellent capabilities of capturing patterns and structures in natural language generation and achieved state-of-the-art results in many tasks. In this paper we present a transformer-based model for multi-turn dialog response generation. Our solution is based on a hybrid approach which augments a transformer-based generative model with a novel retrieval mechanism, which leverages the memorized information in the training data via k-Nearest Neighbor search. Our system is evaluated on two datasets made by customer/assistant dialogs: the Taskmaster-1, released by Google and holding high quality, goal-oriented conversational data and a proprietary dataset collected from a real customer service call center. Data-to-Text Generation (DTG) is a subfield of Natural Language Generation aiming at transcribing structured data in natural language descriptions. The field has been recently boosted by the use of neural-based generators which exhibit on one side great syntactic skills without the need of hand-crafted pipelines; on the other side, the quality of the generated text reflects the quality of the training data, which in realistic settings only offer imperfectly aligned structure-text pairs. Consequently, state-of-art neural models include misleading statements - usually called hallucinations - in their outputs. The control of this phenomenon is today a major challenge for DTG, and is the problem addressed in the paper. Previous work deal with this issue at the instance level: using an alignment score for each table-reference pair. In contrast, we propose a finer-grained approach, arguing that hallucinations should rather be treated at the word level. Specifically, we propose a Multi-Branch Decoder which is able to leverage word-level labels to learn the relevant parts of each training instance. These labels are obtained following a simple and efficient scoring procedure based on co-occurrence analysis and dependency parsing. Extensive evaluations, via automated metrics and human judgment on the standard WikiBio benchmark, show the accuracy of our alignment labels and the effectiveness of the proposed Multi-Branch Decoder. Our model is able to reduce and control hallucinations, while keeping fluency and coherence in generated texts. Globally, waterborne viral infections significantly threaten public health. While current European Union regulations stipulate that drinking water must be devoid of harmful pathogens, they do not specifically address the presence of enteric viruses in water used for irrigation or food production. Traditional virus detection methods rely on molecular biology assays, requiring specialized personnel and laboratory facilities. Here, we describe an electrochemical sandwich enzyme-linked immunomagnetic assay (ELIME) for the detection of the hepatitis A virus (HAV) in water matrices. This method employed screen-printed electrodes as the sensing platform and utilized commercially available pre-activated magnetic beads to provide a robust foundation for the immunological reaction. The ELIME assay demonstrated exceptional analytical performance in only 185 min achieving a detection limit of 0.5 genomic copies per milliliter (g.c./mL) and exhibiting good reproducibility with a relative standard deviation (RSD) of 7% in HAV-spiked drinking and processing water samples. Compared with the real-time RT-qPCR method described in ISO 15216-1, the ELIME assay demonstrated higher sensitivity, although the overall linearity of the method was moderate. These analytical attributes highlight the potential of the ELIME assay as a rapid and viable alternative for HAV detection in water used for agriculture and food processing.

We aim to demonstrate the scientific potential of the Gaia Early Data Release 3 (EDR3) for the study of the Milky Way structure and evolution. We used astrometric positions, proper motions, parallaxes, and photometry from EDR3 to select different populations and components and to calculate the distances and velocities in the direction of the anticentre. We explore the disturbances of the current disc, the spatial and kinematical distributions of early accreted versus in-situ stars, the structures in the outer parts of the disc, and the orbits of open clusters Berkeley 29 and Saurer 1. We find that: i) the dynamics of the Galactic disc are very complex with vertical asymmetries, and new correlations, including a bimodality with disc stars with large angular momentum moving vertically upwards from below the plane, and disc stars with slightly lower angular momentum moving preferentially downwards; ii) we resolve the kinematic substructure (diagonal ridges) in the outer parts of the disc for the first time; iii) the red sequence that has been associated with the proto-Galactic disc that was present at the time of the merger with Gaia-Enceladus-Sausage is currently radially concentrated up to around 14 kpc, while the blue sequence that has been associated with debris of the satellite extends beyond that; iv) there are density structures in the outer disc, both above and below the plane, most probably related to Monoceros, the Anticentre Stream, and TriAnd, for which the Gaia data allow an exhaustive selection of candidate member stars and dynamical study; and v) the open clusters Berkeley~29 and Saurer~1, despite being located at large distances from the Galactic centre, are on nearly circular disc-like orbits. Background Little is known about lived experience of synchronous telehealth in patients with musculoskeletal (MSK) disorders. Objective We conducted a rapid systematic review to answer: (1) what are the lived experiences and/or perspectives of people with MSK disorders receiving non-pharmacological interventions delivered through synchronous telehealth; and (2) what clinical implications can be inferred from qualitative studies focusing on lived experiences for how telehealth is delivered in the management of MSK disorders? Data sources A comprehensive search of MEDLINE, CINAHL, PsycINFO, ProQuest, and Google Scholar from June 2010 to July 2023. Eligible qualitative and mixed methods studies capturing lived experiences of adults with MSK disorders receiving non-pharmacological interventions via synchronous telehealth were included. Study methods Systematic rapid review conducted according to WHO guidelines. Titles and abstracts screened by reviewers independently, eligible studies critically appraised, and data was extracted. Themes summarized using the Consolidated Framework for Implementation Research (CFIR). GRADE-CERQual (Confidence in the Evidence from Reviews of Qualitative research) used to assess confidence in synthesis findings. Results We identified 9782 references, screened 8029, and critically appraised 22, and included 17 studies. There is evidence to suggest that the experience of telehealth prior to and during the pandemic was shaped by (1) patient perception of telehealth, (2) existing relationships with practitioners, (3) availability and accessibility of telehealth technologies, and (4) perceptions about the importance of the role of the physical exam in assessing and treating MSK disorders. Conclusion The five identified implications could be used to inform future research, policy, and strategy development.

Gaia Early Data Release 3 (Gaia EDR3) contains results for 1.812 billion sources in the magnitude range G = 3 to 21 based on observations collected by the European Space Agency Gaia satellite during the first 34 months of its operational phase. We describe the input data, the models, and the processing used for the astrometric content of Gaia EDR3, as well as the validation of these results performed within the astrometry task. The processing broadly followed the same procedures as for Gaia DR2, but with significant improvements to the modelling of observations. For the first time in the Gaia data processing, colour-dependent calibrations of the line- and point-spread functions have been used for sources with well-determined colours from DR2. In the astrometric processing these sources obtained five-parameter solutions, whereas other sources were processed using a special calibration that allowed a pseudocolour to be estimated as the sixth astrometric parameter. Compared with DR2, the astrometric calibration models have been extended, and the spin-related distortion model includes a self-consistent determination of basic-angle variations, improving the global parallax zero point. Gaia EDR3 gives full astrometric data (positions at epoch J2016.0, parallaxes, and proper motions) for 1.468 billion sources (585 million with five-parameter solutions, 882 million with six parameters), and mean positions at J2016.0 for an additional 344 million. Solutions with five parameters are generally more accurate than six-parameter solutions, and are available for 93% of the sources brighter than G = 17 mag. The median uncertainty in parallax and annual proper motion is 0.02-0.03 mas at magnitude G = 9 to 14, and around 0.5 mas at G = 20. Extensive characterisation of the statistical properties of the solutions is provided, including the estimated angular power spectrum of parallax bias from the quasars., Comment: 35 pages, 26 figures. We produce a clean and well-characterised catalogue of objects within 100\,pc of the Sun from the \G\ Early Data Release 3. We characterise the catalogue through comparisons to the full data release, external catalogues, and simulations. We carry out a first analysis of the science that is possible with this sample to demonstrate its potential and best practices for its use. The selection of objects within 100\,pc from the full catalogue used selected training sets, machine-learning procedures, astrometric quantities, and solution quality indicators to determine a probability that the astrometric solution is reliable. The training set construction exploited the astrometric data, quality flags, and external photometry. For all candidates we calculated distance posterior probability densities using Bayesian procedures and mock catalogues to define priors. Any object with reliable astrometry and a non-zero probability of being within 100\,pc is included in the catalogue. We have produced a catalogue of \NFINAL\ objects that we estimate contains at least 92\% of stars of stellar type M9 within 100\,pc of the Sun. We estimate that 9\% of the stars in this catalogue probably lie outside 100\,pc, but when the distance probability function is used, a correct treatment of this contamination is possible. We produced luminosity functions with a high signal-to-noise ratio for the main-sequence stars, giants, and white dwarfs. We examined in detail the Hyades cluster, the white dwarf population, and wide-binary systems and produced candidate lists for all three samples. We detected local manifestations of several streams, superclusters, and halo objects, in which we identified 12 members of \G\ Enceladus. Context. Gaia Early Data Release 3 (Gaia EDR3) provides accurate astrometry for about 1.6 million compact (QSO-like) extragalactic sources, 1.2 million of which have the best-quality five-parameter astrometric solutions. Aims. The proper motions of QSO-like sources are used to reveal a systematic pattern due to the acceleration of the solar system barycentre with respect to the rest frame of the Universe. Apart from being an important scientific result by itself, the acceleration measured in this way is a good quality indicator of the Gaia astrometric solution. Methods. The effect of the acceleration is obtained as a part of the general expansion of the vector field of proper motions in Vector Spherical Harmonics (VSH). Various versions of the VSH fit and various subsets of the sources are tried and compared to get the most consistent result and a realistic estimate of its uncertainty. Additional tests with the Gaia astrometric solution are used to get a better idea on possible systematic errors in the estimate. Results. Our best estimate of the acceleration based on Gaia EDR3 is $(2.32 \pm 0.16) \times 10^{-10}$ m s${}^{-2}$ (or $7.33 \pm 0.51$ km s$^{-1}$ Myr${}^{-1}$) towards $\alpha = 269.1^\circ \pm 5.4^\circ$, $\delta = -31.6^\circ \pm 4.1^\circ$, corresponding to a proper motion amplitude of $5.05 \pm 0.35$ $\mu$as yr${}^{-1}$. This is in good agreement with the acceleration expected from current models of the Galactic gravitational potential. We expect that future Gaia data releases will provide estimates of the acceleration with uncertainties substantially below 0.1 $\mu$as yr${}^{-1}$., Comment: A&A, accepted

We compare the Gaia DR2 and Gaia EDR3 performances in the study of the Magellanic Clouds and show the clear improvements in precision and accuracy in the new release. We also show that the systematics still present in the data make the determination of the 3D geometry of the LMC a difficult endeavour; this is at the very limit of the usefulness of the Gaia EDR3 astrometry, but it may become feasible with the use of additional external data. We derive radial and tangential velocity maps and global profiles for the LMC for the several subsamples we defined. To our knowledge, this is the first time that the two planar components of the ordered and random motions are derived for multiple stellar evolutionary phases in a galactic disc outside the Milky Way, showing the differences between younger and older phases. We also analyse the spatial structure and motions in the central region, the bar, and the disc, providing new insights into features and kinematics. Finally, we show that the Gaia EDR3 data allows clearly resolving the Magellanic Bridge, and we trace the density and velocity flow of the stars from the SMC towards the LMC not only globally, but also separately for young and evolved populations. This allows us to confirm an evolved population in the Bridge that is slightly shift from the younger population. We present the early installment of the third Gaia data release, Gaia EDR3, consisting of astrometry and photometry for 1.8 billion sources brighter than magnitude 21, complemented with the list of radial velocities from Gaia DR2. Gaia EDR3 contains celestial positions and the apparent brightness in G for approximately 1.8 billion sources. For 1.5 billion of those sources, parallaxes, proper motions, and the (G_BP-G_RP) colour are also available. The passbands for G, G_BP, and G_RP are provided as part of the release. For ease of use, the 7 million radial velocities from Gaia DR2 are included in this release, after the removal of a small number of spurious values. New radial velocities will appear as part of Gaia DR3. Finally, Gaia EDR3 represents an updated materialisation of the celestial reference frame (CRF) in the optical, the Gaia-CRF3, which is based solely on extragalactic sources. The creation of the source list for Gaia EDR3 includes enhancements that make it more robust with respect to high proper motion stars, and the disturbing effects of spurious and partially resolved sources. The source list is largely the same as that for Gaia DR2, but it does feature new sources and there are some notable changes. The source list will not change for Gaia DR3. Gaia EDR3 represents a significant advance over Gaia DR2, with parallax precisions increased by 30 percent, proper motion precisions increased by a factor of 2, and the systematic errors in the astrometry suppressed by 30--40 percent for the parallaxes and by a factor ~2.5 for the proper motions. The photometry also features increased precision, but above all much better homogeneity across colour, magnitude, and celestial position. The management of unruptured brain arteriovenous malformations (ubAVMs) is a complex challenge to neurovascular practitioners. This meta-analysis aimed to identify the optimal management of ubAVMs comparing conservative management, embolization, radiosurgery, microsurgical resection, and multimodality. The search strategy was developed a priori according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. We searched the Ovid Medline, Embase, Web of Science, and Cochrane Library databases to identify relevant papers. Using R version 4.1.1., a frequentist network meta-analysis was conducted to compare different management modalities for the ubAVMs. Overall, the conservative group had the lowest risk of rupture (P-score=0.77), and the lowest rate of complications was found in the conservative group (P-score=1). Among different interventions, the multimodality group had the highest rupture risk (P-score=0.34), the lowest overall complications (P-score=0.75), the best functional improvement (P-score=0.65), and the lowest overall mortality (P-score=0.8). However, multimodality treatment showed a significantly higher risk of rupture (odds ratio [OR]=2.13; 95% confidence interval [95% CI]=1.18–3.86) and overall complication rate (OR=5.56; 95% CI=3.37–9.15) compared to conservative management; nevertheless, there were no significant differences in overall mortality or functional independence when considered independently. Conservative management is associated with the lowest rupture risk and complication rate overall. A multimodal approach is the best option when considering mortality rates and functional improvement in the context of existing morbidity/symptoms. Microsurgery, embolization, and radiosurgery alone are similar to the natural history in terms of functional improvement and mortality, but have higher complication rates.

Interleukin-6 (IL-6) plays a crucial role in autoimmunity and chronic inflammation. This study aims to develop a low-cost, simple-to-manufacture, and user-friendly label-free electrochemical point-of-care device for the rapid detection of IL-6 in patients with psoriasis. Precisely, a sandwich-based format immunosensor was developed using two primary antibodies (mAb-IL6 clone-5 and clone-7) and screen-printed electrodes modified with an inexpensive recycling electrochemical enhancing material, called biochar. mAb-IL6 clone-5 was used as a covalently immobilized capture bioreceptor on modified electrodes, and mAb-IL6 clone-7 was used to recognize the immunocomplex (Anti-IL6 clone-5 and IL-6) and form the sandwich. Cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were used to conduct electrochemical characterization of the layer-by-layer assembly of the immunosensor, while square wave voltammetry (SWV) was used to perform the sensing. Background For acute proximal intracranial artery occlusions, contact aspiration may be more effective than stent‐retriever for first‐line reperfusion therapy. Due to the lack of data regarding medium vessel occlusion thrombectomy, we evaluated outcomes according to first‐line technique in a large, multicenter registry. Methods Imaging, procedural, and clinical outcomes of patients with acute proximal medium vessel occlusions (M2, A1, or P1) or distal medium vessel occlusions (M3, A2, P2, or further) treated at 37 sites in 10 countries were analyzed according to first‐line endovascular technique (stent‐retriever versus aspiration). Multivariable logistic regression and propensity‐score matching were used to estimate the odds of the primary outcome, expanded Thrombolysis in Cerebral Infarction score of 2b–3 (“successful recanalization”), as well as secondary outcomes (first‐pass effect, expanded Thrombolysis in Cerebral Infarction 2c‐3, intracerebral hemorrhage, and 90‐day modified Rankin scale, 90‐day mortality) between treatment groups. Introduction Dual antiplatelet therapy (DAPT) is necessary to minimize the risk of periprocedural thromboembolic complications associated with aneurysm embolization using Pipeline embolization device (PED). We sought to assess the impact of platelet function testing (PFT) on reducing periprocedural thromboembolic complications associated with PED flow diversion in patients receiving aspirin and clopidogrel. Methods Patients with unruptured intracranial aneurysms requiring PED flow diversion were identified from 13 centers for retrospective evaluation. Clinical variables including the results of PFT prior to treatment, periprocedural DAPT regimen, and intracranial complications occurring within 72‐hours of embolization were identified from the medical record. Complication rates were compared between PFT and non‐PFT groups. Differences between groups were tested for statistical significance using the Wilcoxon rank sum, Fisher exact, or Chi‐square tests. A p‐value 0.9). Conclusion Pre‐procedural PFT prior to PED treatment of intracranial aneurysms in patients pre‐medicated with an aspirin and clopidogrel DAPT regimen may not be necessary to significantly reduce the risk of procedure‐related intracranial complications.

In the last few years, many different methods have been focusing on using deep recurrent neural networks for natural language generation. The most widely used sequence-to-sequence neural methods are word-based: as such, they need a pre-processing step called delexicalization (conversely, relexicalization) to deal with uncommon or unknown words. These forms of processing, however, give rise to models that depend on the vocabulary used and are not completely neural. In this work, we present an end-to-end sequence-to-sequence model with attention mechanism which reads and generates at a character level, no longer requiring delexicalization, tokenization, nor even lowercasing. Moreover, since characters constitute the common "building blocks" of every text, it also allows a more general approach to text generation, enabling the possibility to exploit transfer learning for training. These skills are obtained thanks to two major features: (i) the possibility to alternate between the standard generation mechanism and a copy one, which allows to directly copy input facts to produce outputs, and (ii) the use of an original training pipeline that further improves the quality of the generated texts. We also introduce a new dataset called E2E+, designed to highlight the copying capabilities of character-based models, that is a modified version of the well-known E2E dataset used in the E2E Challenge. Two new supramolecular photocatalysts made of covalently linked Ru(II) polypyridine chromophore subunits ([Ru(bpy)3]2+-type species; bpy = 2,2′-bipyridine) and [RuL(pic)2] (L = 2,2′-bipyridine-6,6′-dicarboxylic acid; pic = 4-picoline) water oxidation catalyst subunits have been prepared. The new species, 1 and 2, contain chromophore and catalyst subunits in the molecular ratios 1:1 and 1:2, respectively. The model chromophore species [Ru(bpy)2(L1)]2+ (RuP1; L1=4-[2-(4-pyridyl)-2-hydroxyethyl]-4-methyl-2,2′-bipyridine) and [Ru(bpy)2(L2)]2+ (RuP2; L2 = 4,4′-bis[2-(4-pyridyl)-2-hydroxyethyl]-2,2′-bipyridine) have also been prepared. The absorption spectra, oxidation behavior, and luminescent properties of 1 and 2 have been studied, and the results indicate that each subunit largely maintains its own properties in the supramolecular species. However, the luminescence of the chromophore subunits is significantly quenched in 1 and 2 in comparison with the luminescence of the respective model species. Both 1 and 2 exhibit catalytic water oxidation in the presence of cerium ammonium nitrate, exhibiting an I2M mechanism, with a better efficiency than the known catalyst [RuL(pic)2] under the same experimental conditions. Upon light irradiation, in the presence of persulfate as a sacrificial acceptor agent, 1 and 2 are more efficient photocatalysts than a system made of separated [Ru(bpy)3]2+ and [RuL(pic)2] species, highlighting the advantage of using multicomponent, supramolecular species with respect to isolated species. The O–O bond formation step is I2M, even in the photo-driven process. The photocatalytic process of 2 is more efficient than that of 1, with the turnover frequency reaching a value of 1.2 s−1. A possible reason could be an increased local concentration of catalytic subunits in the needed bimolecular assembly required for the I2M mechanism in 2 with respect to 1, a consequence of the presence of two catalytic subunits in each multicomponent species 2. A key component for landslide early warning systems (LEWSs) is constituted by thresholds providing the conditions above which a landslide can be triggered. Traditionally, thresholds based on rainfall characteristics have been proposed, but recently, the hydrometeorological approach, combining rainfall with soil moisture or catchment storage information, is becoming widespread. Most of the hydrometeorological thresholds proposed in the literature use the soil moisture from a single layer (i.e., depth or depth range). On the other hand, multi-layered soil moisture information can be measured or can be available from reanalysis projects as well as from hydrological models. Approaches using this multi-layered information are lacking, perhaps because of the need to keep the thresholds simple and two-dimensional. In this paper, we propose principal component analysis (PCA) as an approach for deriving two-dimensional hydrometeorological thresholds that use multi-layered soil moisture information. To perform a more objective assessment we also propose a piecewise linear equation for the identification of the threshold's shape, which is more flexible than traditional choices (e.g., power law or bilinear). Comparison of the receiver operating characteristic (ROC) (true skill statistic, TSS) of thresholds based on single- and multi-layered soil moisture information also provides a novel tool for identifying the significance of multi-layered information on landslide triggering in a given region. Results for Sicily island, considering the ERA5-Land reanalysis soil moisture data (available at four different depth layers), corroborate the advantages of the hydrometeorological approach gained in spite of the coarse spatial resolution and the limited accuracy of reanalysis data. Specifically, the TSS of traditional precipitation intensity–duration thresholds is equal to 0.5, while those of the proposed hydrometeorological thresholds is significantly higher (TSS=0.71). For the analyzed region, however, multi-layered information seems not to be relevant, as performances in terms of TSS are similar to those obtained with single-layer soil moisture at the upper depths, namely 0–7 and 7–28 cm, which can imply that in Sicily landslide phenomena are mainly influenced by soil moisture in most shallow soil layers.

We highlight the power of the Gaia DR2 in studying many fine structures of the Hertzsprung-Russell diagram (HRD). Gaia allows us to present many different HRDs, depending in particular on stellar population selections. We do not aim here for completeness in terms of types of stars or stellar evolutionary aspects. Instead, we have chosen several illustrative examples. We describe some of the selections that can be made in Gaia DR2 to highlight the main structures of the Gaia HRDs. We select both field and cluster (open and globular) stars, compare the observations with previous classifications and with stellar evolutionary tracks, and we present variations of the Gaia HRD with age, metallicity, and kinematics. Late stages of stellar evolution such as hot subdwarfs, post-AGB stars, planetary nebulae, and white dwarfs are also analysed, as well as low-mass brown dwarf objects. The Gaia HRDs are unprecedented in both precision and coverage of the various Milky Way stellar populations and stellar evolutionary phases. Many fine structures of the HRDs are presented. The clear split of the white dwarf sequence into hydrogen and helium white dwarfs is presented for the first time in an HRD. The relation between kinematics and the HRD is nicely illustrated. Two different populations in a classical kinematic selection of the halo are unambiguously identified in the HRD. Membership and mean parameters for a selected list of open clusters are provided. They allow drawing very detailed cluster sequences, highlighting fine structures, and providing extremely precise empirical isochrones that will lead to more insight in stellar physics. Gaia DR2 demonstrates the potential of combining precise astrometry and photometry for large samples for studies in stellar evolution and stellar population and opens an entire new area for HRD-based studies., Comment: Published in the A&A Gaia Data Release 2 special issue. Gaia Data Release 2 (Gaia DR2) contains results for 1693 million sources in the magnitude range 3 to 21 based on observations collected by the European Space Agency Gaia satellite during the first 22 months of its operational phase. We describe the input data, models, and processing used for the astrometric content of Gaia DR2, and the validation of these results performed within the astrometry task. Some 320 billion centroid positions from the pre-processed astrometric CCD observations were used to estimate the five astrometric parameters (positions, parallaxes, and proper motions) for 1332 million sources, and approximate positions at the reference epoch J2015.5 for an additional 361 million mostly faint sources. Special validation solutions were used to characterise the random and systematic errors in parallax and proper motion. For the sources with five-parameter astrometric solutions, the median uncertainty in parallax and position at the reference epoch J2015.5 is about 0.04 mas for bright (G<14 mag) sources, 0.1 mas at G=17 mag, and 0.7 mas at G=20 mag. In the proper motion components the corresponding uncertainties are 0.05, 0.2, and 1.2 mas/yr, respectively. The optical reference frame defined by Gaia DR2 is aligned with ICRS and is non-rotating with respect to the quasars to within 0.15 mas/yr. From the quasars and validation solutions we estimate that systematics in the parallaxes depending on position, magnitude, and colour are generally below 0.1 mas, but the parallaxes are on the whole too small by about 0.03 mas. Significant spatial correlations of up to 0.04 mas in parallax and 0.07 mas/yr in proper motion are seen on small (<1 deg) and intermediate (20 deg) angular scales. Background: Despite large-scale interventions aimed at developing literacy skills, children’s reading competence levels in South Africa continue to remain an area of concern. In addition, the need to prepare learners for the increased demands of a fast-changing world of learning and working is gaining attention in educational policy and practice. Aim: Using a qualitative multi-site case study research design, the authors aimed to explore teachers’ understanding and enactment of scripted literacy lessons that are designed to promote 21st-century competencies. Setting: Five Grade 1 teachers were selected from four schools, three of which are in peri-urban and the other in a township area. Methods: Data were generated in two phases across three teaching cycles. In the first phase, lessons were observed and recorded on video. The second phase consisted of stimulated recall interviews (SRIs) in which teachers commented on their recorded lessons. Results: The findings showed that when teachers had relevant prior knowledge of specific competencies, they were better positioned to leverage these as a basis for their lessons. However, the findings also indicated that teachers’ entrenched methods and ways of thinking were obstacles for change. Conclusion: Training and development opportunities should pay sufficient attention to the practicalities of changing pedagogies and using scripted lesson plans. Contribution: The findings contribute to knowledge on the affordances and challenges when designing training opportunities aimed at integrating 21st-century competencies into scripted literacy lessons.
