51. Glioblastoma multiforme recurrence on skin and subcutaneous tissue: a case report

Abstract: Glioblastoma multiforme (GBM) is the most common malignant primary central nervous system (CNS) tumor. It presents an aggressive pattern, with a tendency for intracranial progression despite optimal treatment. On the other hand, metastases and manifestations outside the CNS are exceptional, and very few cases have been described. The authors submitted a case of a 48-year-old male diagnosed with a left parietooccipital GBM isocitrate dehydrogenase (IDH) wild-type, non-methylated O6-methylguanine-DNA methyltransferase (MGMT). Six months after surgical treatment, followed by a chemotherapy (CT) and radiotherapy scheme, he developed a few subcutaneous erythematous nodules within the surgical scar. A punch biopsy confirmed the histopathology of GBM. The CT scan showed concomitant intracranial progression. We ruled out systemic metastases. As the performance status was good [Karnofsky Performance Status 90 (KPS 90)] and the subcutaneous implants were growing rapidly, limiting the quality of life, we decided to perform palliative surgery to remove the implants. The result was good. Unfortunately, the patient worsened during the following week, after ruling out medical complications, we attributed the worsening to cerebral tumor swelling, revealed in the CT scan, as unresponsive to steroids. He passed away a few days later. Based on the analysis of our case, we intend to provide useful information for the approach to this peculiar manifestation of GBM.

52. Beak and feather disease virus (BFDV) persists in tissues of asymptomatic wild Crimson Rosellas

Abstract: Abstract Infectious diseases can drive populations and species to extinction. Beak and feather disease virus (BFDV) is a circovirus of global conservation concern that can infect all Psittaciformes and some other species. Yet some parrot species, such as Crimson rosellas (Platycercus elegans), can live successfully with high BFDV prevalence (>40%) with no clinical signs reported in infected individuals. We assessed BFDV load in 10-12 tissues per bird, from n = 66 P. elegans, to reveal tissue tropism and BFDV persistence in tissues. Here we show that in 94% of individuals, BFDV was detected in one or more tissues. While BFDV replicated to high levels in subadults, in adults (some confirmed seropositive) the virus persisted in various tissues at much lower levels. Our findings reveal that BFDV is much more common in wild P. elegans than previously thought and suggest that current screening practices (mostly on blood) may substantially underestimate BFDV infection estimates, with implications for biosecurity and conservation programs globally.

53. Oculomic stratification of COVID-19 patients’ intensive therapy unit admission status and mortality by retinal morphological findings

Abstract: Abstract To investigate if retinal thickness has predictive utility in COVID-19 outcomes by evaluating the statistical association between retinal thickness using OCT and of COVID-19-related mortality. Secondary outcomes included associations between retinal thickness and length of stay (LoS) in hospital. In this retrospective cohort study, OCT scans from 230 COVID-19 patients admitted to the Intensive Care Unit (ITU) were compared with age and gender-matched patients with pneumonia from before March 2020. Total retinal, GCL + IPL, and RNFL thicknesses were recorded, and analysed with systemic measures collected at the time of admission and mortality outcomes, using linear regression models, Pearson’s R correlation, and Principal Component Analysis. Retinal thickness was significantly associated with all-time mortality on follow up in the COVID-19 group (p = 0.015), but not 28-day mortality (p = 0.151). Retinal and GCL + IPL layer thicknesses were both significantly associated with LoS in hospital for COVID-19 patients (p = 0.006 for both), but not for patients with pneumonia (p = 0.706 and 0.989 respectively). RNFL thickness was not associated with LoS in either group (COVID-19 p = 0.097, pneumonia p = 0.692). Retinal thickness associated with LoS in hospital and long-term mortality in COVID-19 patients, suggesting that retinal structure could be a surrogate marker for frailty and predictor of disease severity in this group of patients, but not in patients with pneumonia from other causes.

Abstract: The local Cosmic Ray (CR) energy spectrum exhibits a spectral softening at energies around 3~PeV. Sources which are capable of accelerating hadrons to such energies are called hadronic PeVatrons. However, hadronic PeVatrons have not yet been firmly identified within the Galaxy. Several source classes, including Galactic Supernova Remnants (SNRs), have been proposed as PeVatron candidates. The potential to search for hadronic PeVatrons with the Cherenkov Telescope Array (CTA) is assessed. The focus is on the usage of very high energy $\gamma$-ray spectral signatures for the identification of PeVatrons. Assuming that SNRs can accelerate CRs up to knee energies, the number of Galactic SNRs which can be identified as PeVatrons with CTA is estimated within a model for the evolution of SNRs. Additionally, the potential of a follow-up observation strategy under moonlight conditions for PeVatron searches is investigated. Statistical methods for the identification of PeVatrons are introduced, and realistic Monte--Carlo simulations of the response of the CTA observatory to the emission spectra from hadronic PeVatrons are performed. Based on simulations of a simplified model for the evolution for SNRs, the detection of a $\gamma$-ray signal from in average 9 Galactic PeVatron SNRs is expected to result from the scan of the Galactic plane with CTA after 10 hours of exposure. CTA is also shown to have excellent potential to confirm these sources as PeVatrons in deep observations with $\mathcal{O}(100)$ hours of exposure per source., Comment: 34 pages, 16 figures, Accepted for publication in Astroparticle Physics

57. Occurrence, co-occurrence and persistence of symptoms of depression and post-traumatic stress disorder in survivors of COVID-19 critical illness

Abstract: Background: Intensive care unit (ICU) admission and invasive mechanical ventilation (IMV) are associated with psychological distress and trauma. The COVID-19 pandemic brought with it a series of additional long-lasting stressful and traumatic experiences. However, little is known about comorbid depression and post-traumatic stress disorder (PTSD).Objective: To examine the occurrence, co-occurrence, and persistence of clinically significant symptoms of depression and PTSD, and their predictive factors, in COVID-19 critical illness survivors.Method: Single-centre prospective observational study in adult survivors of COVID-19 with ≥24 h of ICU admission. Patients were assessed one and 12 months after ICU discharge using the depression subscale of the Hospital Anxiety and Depression Scale and the Davidson Trauma Scale. Differences in isolated and comorbid symptoms of depression and PTSD between patients with and without IMV and predictors of the occurrence and persistence of symptoms of these mental disorders were analysed.Results: Eighty-nine patients (42 with IMV) completed the 1-month follow-up and 71 (34 with IMV) completed the 12-month follow-up. One month after discharge, 29.2% of patients had symptoms of depression and 36% had symptoms of PTSD; after one year, the respective figures were 32.4% and 31%. Coexistence of depressive and PTSD symptoms accounted for approximately half of all symptomatic cases. Isolated PTSD symptoms were more frequent in patients with IMV (p≤.014). The need for IMV was associated with the occurrence at one month (OR = 6.098, p = .005) and persistence at 12 months (OR = 3.271, p = .030) of symptoms of either of these two mental disorders.Conclusions: Comorbid depressive and PTSD symptoms were highly frequent in our cohort of COVID-19 critical illness survivors. The need for IMV predicted short-term occurrence and long-term persistence of symptoms of these mental disorders, especially PTSD symptoms. The specific role of dyspnea in the association between IMV and post-ICU mental disorders deserves further investigation.Trial registration: ClinicalTrials.gov identifier: NCT04422444.

Abstract: Este trabalho avalia comparativamente o desempenho de dois sistemas de tradução automática, a saber, DeepL e Google Tradutor. O estudo parte de uma breve visão geral sobre traduções automáticas com a utilização de redes neurais, seguido de uma reflexão sobre os resultados da tradução automática ao utilizar a avaliação automática e humana. Subsequentemente, aborda a diferença de desempenho entre ambos os sistemas, além de identificar possíveis problemas linguísticos advindos dos resultados da tradução automática gerada por eles. Para a realização dessa comparação, o trabalho emprega um trecho da obra de Machado de Assis, Dom Casmurro, do capítulo “Olhos de Ressaca”. O estudo utiliza as traduções geradas de forma automática para analisar criticamente qual sistema apresenta mais qualidade no resultado da tradução e examina comparativamente os erros linguísticos gerados pelos resultados do DeepL e Google Tradutor. Os critérios de avaliação incluem a precisão da tradução, a capacidade de manter o significado e a estrutura da frase original, a fluência e adequação da tradução resultante.

59. The epsilon toxin from Clostridium perfringens stimulates calcium‐activated chloride channels, generating extracellular vesicles in Xenopus oocytes

Abstract: Abstract The epsilon toxin (Etx) from Clostridium perfringens has been identified as a potential trigger of multiple sclerosis, functioning as a pore‐forming toxin that selectively targets cells expressing the plasma membrane (PM) myelin and lymphocyte protein (MAL). Previously, we observed that Etx induces the release of intracellular ATP in sensitive cell lines. Here, we aimed to re‐examine the mechanism of action of the toxin and investigate the connection between pore formation and ATP release. We examined the impact of Etx on Xenopus laevis oocytes expressing human MAL. Extracellular ATP was assessed using the luciferin‐luciferase reaction. Activation of calcium‐activated chloride channels (CaCCs) and a decrease in the PM surface were recorded using the two‐electrode voltage‐clamp technique. To evaluate intracellular Ca2+ levels and scramblase activity, fluorescent dyes were employed. Extracellular vesicles were imaged using light and electron microscopy, while toxin oligomers were identified through western blots. Etx triggered intracellular Ca2+ mobilization in the Xenopus oocytes expressing hMAL, leading to the activation of CaCCs, ATP release, and a reduction in PM capacitance. The toxin induced the activation of scramblase and, thus, translocated phospholipids from the inner to the outer leaflet of the PM, exposing phosphatidylserine outside in Xenopus oocytes and in an Etx‐sensitive cell line. Moreover, Etx caused the formation of extracellular vesicles, not derived from apoptotic bodies, through PM fission. These vesicles carried toxin heptamers and doughnut‐like structures in the nanometer size range. In conclusion, ATP release was not directly attributed to the formation of pores in the PM, but to scramblase activity and the formation of extracellular vesicles.

Abstract: Line-like features in TeV $\gamma$-rays constitute a ''smoking gun'' for TeV-scale particle dark matter and new physics. Probing the Galactic Center region with ground-based Cherenkov telescopes enables the search for TeV spectral features in immediate association with a dense dark matter reservoir at a sensitivity out of reach for satellite $\gamma$-ray detectors, and direct detection and collider experiments. We report on 223 hours of observations of the Galactic Center region with the MAGIC stereoscopic telescope system reaching $\gamma$-ray energies up to 100 TeV. We improved the sensitivity to spectral lines at high energies using large-zenith-angle observations and a novel background modeling method within a maximum-likelihood analysis in the energy domain. No line-like spectral feature is found in our analysis. Therefore, we constrain the cross section for dark matter annihilation into two photons to $\langle \sigma v \rangle \lesssim 5 \times 10^{-28}\,\mathrm{cm^3\,s^{-1}}$ at 1 TeV and $\langle \sigma v \rangle \lesssim 1 \times 10^{-25}\,\mathrm{cm^3\,s^{-1}}$ at 100 TeV, achieving the best limits to date for a dark matter mass above 20 TeV and a cuspy dark matter profile at the Galactic Center. Abstract: The SNR G106.3+2.7, detected at 1--100 TeV energies by different $\gamma$-ray facilities, is one of the most promising PeVatron candidates. This SNR has a cometary shape which can be divided into a head and a tail region with different physical conditions. However, it is not identified in which region the 100 TeV emission is produced due to the limited position accuracy and/or angular resolution of existing observational data. Additionally, it remains unclear whether the origin of the $\gamma$-ray emission is leptonic or hadronic. With the better angular resolution provided by these new MAGIC data compared to earlier $\gamma$-ray datasets, we aim to reveal the acceleration site of PeV particles and the emission mechanism by resolving the SNR G106.3+2.7 with 0.1$^\circ$ resolution at TeV energies. We detected extended $\gamma$-ray emission spatially coincident with the radio continuum emission at the head and tail of SNR G106.3+2.7. The fact that we detected a significant $\gamma$-ray emission with energies above 6.0 TeV from the tail region only suggests that the emissions above 10 TeV, detected with air shower experiments (Milagro, HAWC, Tibet AS$\gamma$ and LHAASO), are emitted only from the SNR tail. Under this assumption, the multi-wavelength spectrum of the head region can be explained with either hadronic or leptonic models, while the leptonic model for the tail region is in contradiction with the emission above 10 TeV and X-rays. In contrast, the hadronic model could reproduce the observed spectrum at the tail by assuming a proton spectrum with a cutoff energy of $\sim 1$ PeV for the tail region. Such a high energy emission in this middle-aged SNR (4--10 kyr) can be explained by considering the scenario that protons escaping from the SNR in the past interact with surrounding dense gases at present., Comment: 13 pages, 7 figures, Accepted for publication in A&A

Abstract: The BL Lac object 1ES 0647+250 is one of the few distant $\gamma$-ray emitting blazars detected at very high energies (VHE, $\gtrsim$100 GeV) during a non-flaring state. It was detected with the MAGIC telescopes during its low activity in the years 2009-2011, as well as during three flaring activities in the years 2014, 2019 and 2020, with the highest VHE flux in the latter epoch. An extensive multi-instrument data set was collected within several coordinated observing campaigns throughout these years. We aim to characterise the long-term multi-band flux variability of 1ES 0647+250, as well as its broadband spectral energy distribution (SED) during four distinct activity states selected in four different epochs, in order to constrain the physical parameters of the blazar emission region under certain assumptions. We evaluate the variability and correlation of the emission in the different energy bands with the fractional variability and the Z-transformed Discrete Correlation Function, as well as its spectral evolution in X-rays and $\gamma$ rays. Owing to the controversy in the redshift measurements of 1ES 0647+250 reported in the literature, we also estimate its distance in an indirect manner through the comparison of the GeV and TeV spectra from simultaneous observations with Fermi-LAT and MAGIC during the strongest flaring activity detected to date. Moreover, we interpret the SEDs from the four distinct activity states within the framework of one-component and two-component leptonic models, proposing specific scenarios that are able to reproduce the available multi-instrument data., Comment: 20 pages, 7 figures. Abstract: A novel instrumented baffle surrounding the suspended end mirror in the input mode cleaner cavity of the Virgo interferometer was installed in spring 2021. Since then, the device has been regularly operated in the experiment and the obtained results indicate a good agreement with simulations of the stray light inside the optical cavity. The baffle will operate in the upcoming O4 observation run, serving as a demonstrator of the technology designed to instrument the baffles in front of the main mirrors in time for O5. Abstract: Extended and delayed emission around distant TeV sources induced by the effects of propagation of gamma rays through the intergalactic medium can be used for the measurement of the intergalactic magnetic field (IGMF). We search for delayed GeV emission from the hard-spectrum TeV blazar 1ES 0229+200 with the goal to detect or constrain the IGMF-dependent secondary flux generated during the propagation of TeV gamma rays through the intergalactic medium. We analyze the most recent MAGIC observations over a 5 year time span and complement them with historic data of the H.E.S.S. and VERITAS telescopes along with a 12-year long exposure of the Fermi/LAT telescope. We use them to trace source evolution in the GeV-TeV band over one-and-a-half decade in time. We use Monte Carlo simulations to predict the delayed secondary gamma-ray flux, modulated by the source variability, as revealed by TeV-band observations. We then compare these predictions for various assumed IGMF strengths to all available measurements of the gamma-ray flux evolution. We find that the source flux in the energy range above 200 GeV experiences variations around its average on the 14 years time span of observations. No evidence for the flux variability is found in 1-100 GeV energy range accessible to Fermi/LAT. Non-detection of variability due to delayed emission from electromagnetic cascade developing in the intergalactic medium imposes a lower bound of B>1.8e-17 G for long correlation length IGMF and B>1e-14 G for an IGMF of the cosmological origin. Though weaker than the one previously derived from the analysis of Fermi/LAT data, this bound is more robust, being based on a conservative intrinsic source spectrum estimate and accounting for the details of source variability in the TeV energy band. Abstract: We study the broadband emission of Mrk 501 using multi-wavelength observations from 2017 to 2020 performed with a multitude of instruments, involving, among others, MAGIC, Fermi-LAT, NuSTAR, Swift, GASP-WEBT, and OVRO. Mrk 501 showed an extremely low broadband activity, which may help to unravel its baseline emission. Nonetheless, significant flux variations are detected at all wavebands, with the highest occurring at X-rays and very-high-energy (VHE) $\gamma$-rays. A significant correlation ($>$3$\sigma$) between X-rays and VHE $\gamma$-rays is measured, supporting leptonic scenarios to explain the variable parts of the emission, also during low activity. This is further supported when we extend our data from 2008 to 2020, and identify, for the first time, significant correlations between Swift-XRT and Fermi-LAT. We additionally find correlations between high-energy $\gamma$-rays and radio, with the radio lagging by more than 100 days, placing the $\gamma$-ray emission zone upstream of the radio-bright regions in the jet. Furthermore, Mrk 501 showed a historically low activity in X-rays and VHE $\gamma$-rays from mid-2017 to mid-2019 with a stable VHE flux ($>$0.2 TeV) of 5% the emission of the Crab Nebula. The broadband spectral energy distribution (SED) of this 2-year-long low-state, the potential baseline emission of Mrk 501, can be characterized with one-zone leptonic models, and with (lepto)-hadronic models fulfilling neutrino flux constraints from IceCube. We explore the time evolution of the SED towards the low-state, revealing that the stable baseline emission may be ascribed to a standing shock, and the variable emission to an additional expanding or traveling shock., Comment: 55 pages, 30 figures, 14 tables, accepted by APJS. Abstract: LHAASO J2108+5157 is one of the few known unidentified Ultra-High-Energy (UHE) gamma-ray sources with no Very-High-Energy (VHE) counterpart, recently discovered by the LHAASO collaboration. We observed LHAASO J2108+5157 in the X-ray band with XMM-Newton in 2021 for a total of 3.8 hours and at TeV energies with the Large-Sized Telescope prototype (LST-1), yielding 49 hours of good quality data. In addition, we analyzed 12 years of Fermi-LAT data, to better constrain emission of its High-Energy (HE) counterpart 4FGL J2108.0+5155. We found an excess (3.7 sigma) in the LST-1 data at energies E > 3 TeV. Further analysis in the whole LST-1 energy range assuming a point-like source, resulted in a hint (2.2 sigma) of hard emission which can be described with a single power law with photon index Gamma = 1.6 +- 0.2 between 0.3 - 100 TeV. We did not find any significant extended emission which could be related to a Supernova Remnant (SNR) or Pulsar Wind Nebula (PWN) in the XMM-Newton data, which puts strong constraints on possible synchrotron emission of relativistic electrons. The LST-1 and LHAASO observations can be explained as inverse Compton-dominated leptonic emission of relativistic electrons with a cutoff energy of $100^{+70}_{-30}$ TeV. The low magnetic field in the source imposed by the X-ray upper limits on synchrotron emission is compatible with a hypothesis of a PWN or a TeV halo. The lack of a pulsar in the neighborhood of the UHE source is a challenge to the PWN/TeV-halo scenario. The UHE gamma rays can also be explained as $\pi^0$ decay-dominated hadronic emission due to interaction of relativistic protons with one of the two known molecular clouds in the direction of the source. Abstract: Abstract Optical coherence tomography angiography (OCTA) is widely used for non-invasive retinal vascular imaging, but the OCTA methods used to assess retinal perfusion vary. We evaluated the different methods used to assess retinal perfusion between OCTA studies. MEDLINE and Embase were searched from 2014 to August 2021. We included prospective studies including ≥ 50 participants using OCTA to assess retinal perfusion in either global retinal or systemic disorders. Risk of bias was assessed using the National Institute of Health quality assessment tool for observational cohort and cross-sectional studies. Heterogeneity of data was assessed by Q statistics, Chi-square test, and I2 index. Of the 5974 studies identified, 191 studies were included in this evaluation. The selected studies employed seven OCTA devices, six macula volume dimensions, four macula subregions, nine perfusion analyses, and five vessel layer definitions, totalling 197 distinct methods of assessing macula perfusion and over 7000 possible combinations. Meta-analysis was performed on 88 studies reporting vessel density and foveal avascular zone area, showing lower retinal perfusion in patients with diabetes mellitus than in healthy controls, but with high heterogeneity. Heterogeneity was lowest and reported vascular effects strongest in superficial capillary plexus assessments. Systematic review of OCTA studies revealed massive heterogeneity in the methods employed to assess retinal perfusion, supporting calls for standardisation of methodology.

Abstract: MAXI J1820+070 is a low-mass X-ray binary with a black hole as a compact object. This binary underwent an exceptionally bright X-ray outburst from March to October 2018, showing evidence of a non-thermal particle population through its radio emission during this whole period. The combined results of 59.5 hours of observations of the MAXI J1820+070 outburst with the H.E.S.S., MAGIC and VERITAS experiments at energies above 200 GeV are presented, together with Fermi-LAT data between 0.1 and 500 GeV, and multiwavelength observations from radio to X-rays. Gamma-ray emission is not detected from MAXI J1820+070, but the obtained upper limits and the multiwavelength data allow us to put meaningful constraints on the source properties under reasonable assumptions regarding the non-thermal particle population and the jet synchrotron spectrum. In particular, it is possible to show that, if a high-energy gamma-ray emitting region is present during the hard state of the source, its predicted flux should be at most a factor of 20 below the obtained Fermi-LAT upper limits, and closer to them for magnetic fields significantly below equipartition. During the state transitions, under the plausible assumption that electrons are accelerated up to ~ 500 GeV, the multiwavelength data and the gamma-ray upper limits lead consistently to the conclusion that a potential high-energy and very-high-energy gamma-ray emitting region should be located at a distance from the black hole ranging between 10^11 and 10^13 cm. Similar outbursts from low-mass X-ray binaries might be detectable in the near future with upcoming instruments such as CTA., Comment: 15 pages, 5 figures. Accepted for publication in MNRAS

Abstract: The problem of the electromagnetic radiation of an accelerated charged particle is one of the most controversial issues in Physics since the beginning of the last century, representing one of the most popular unsolved problems of the Modern Physics. Different equations of motion have been proposed throughout history for a point charge including the electromagnetic radiation emitted, but all these expressions show some limitations. An equation based on the principle of conservation of energy is proposed in this work for the ultra-relativistic motion. Different examples are analyzed showing that the energy lost by the charge agrees with the Li\'enard formula. Abstract: Abstract Despite the importance of protecting forests and woodlands to achieve global climate and biodiversity goals, logging impacts persist worldwide. Forestry advocates often downplay these impacts but rarely consider the cumulative threat deforestation and degradation has had, and continues to have, on biodiversity. Using New South Wales (Australia) as a case study, we quantify the extent of deforestation and degradation from 1788 (pre‐European colonization) to 2021. We used historical loss as a baseline to evaluate recent logging (2000–2022) and the condition of the remaining native forest and woodland. Condition was quantified by measuring the similarity of a current ecosystem to a historical reference state with high ecological integrity. Using these data, we measured the impacts on 269 threatened terrestrial species. We show that possibly over half (29 million ha) of pre‐1788 native forest and woodland vegetation in NSW has been lost. Of the remaining 25 million ha, 9 million ha is estimated to be degraded. We found recent logging potentially impacted 150 species that had already been affected by this historical deforestation and degradation, but the impacts varied across species. Forty‐three species that were identified as impacted by historical deforestation and degradation and continue to be impacted by logging, now have ≤50% of their pre‐1788 extent remaining that is intact and nine species now have ≤30%. Our research contextualizes the impact of current logging against historical deforestation and highlights deficiencies in environmental assessments that ignore historical baselines. Future land management must consider both the extent and condition of remaining habitat based on pre‐1788 extents.

Abstract: Fundamento: el alcoholismo es una drogadicción que conduce a la dependencia de otros tipos de drogas peligrosas y dañinas para la salud humana, por lo que es un fenómeno que debe ser comprendido con objetividad para el desarrollo de acciones eficaces en su erradicación. Objetivo: exponer la efectividad de una intervención educativa para incrementar los conocimientos sobre el alcoholismo en adolescentes. Métodos: se realizó un estudio cuasiexperimental de intervención educativa, en el Policlínico Universitario “Ramón López Peñaˮ de Santiago de Cuba, en el período febrero-agosto de 2023. La población de estudio estuvo constituida por 45 adolescentes consumidores de riesgo, que cumplieron los criterios de inclusión, a los que se les aplicó un cuestionario inicial que permitió conocer los conocimientos antes y después de la intervención. Se utilizaron métodos teóricos: histórico-lógico, analítico-sintético e inductivo-deductivo para la recogida de datos y fundamentación de la investigación. Del nivel empírico: el cuestionario y la observación. Resultados: la intervención educativa aplicada en sesiones de trabajo grupal a través de técnicas participativas, favoreció la motivación de los adolescentes. Se modificaron los conocimientos relacionados con tipos de consumo, motivos y consecuencias, conducta a seguir ante el consumo excesivo y prevención del alcoholismo. Conclusiones: fue efectiva al incrementar los conocimientos de los adolescentes sobre el alcoholismo, con acciones para fortalecer la promoción y prevención de esta enfermedad, en el primer nivel de atención de salud.

75. Understanding how traumatic brain injury-related changes in fluid biomarkers affect quality of life outcomes in veterans: a prospective observational trial protocol (UNTANGLE)

Abstract: Introduction Traumatic brain injury (TBI) is a major cause of disability, with annual global incidence estimated as 69 million people. Survivors can experience long-term visual changes, altered mental state, neurological deficits and long-term effects that may be associated with mental illness. TBI is prevalent in military personnel due to gunshot wounds, and blast injury. This study aims to evaluate the relationship between evolving visual, biochemical and mental health changes in both military veterans and civilians, suffering from TBI, and detect preliminary indicators of prognosis for TBI recovery, and quality-of-life outcomes.Methods and analysis UNTANGLE is a 24-month prospective observational pilot study recruiting three patient groups: civilians with acute moderate-severe TBI, military veterans with diagnosis of a previous TBI and a control group of civilians or veterans with no history of a previous TBI. Patients will undergo visual, biochemical and mental health assessments, as well as patient-reported quality of life outcome measures over the course of a 1-year follow-up period.Ethics and dissemination Ethical approval has been obtained from the Health Research Authority and Health and Care Research Wales with a REC reference number of 23/NW/0203. The results of the study will be presented at scientific meetings and published in peer-reviewed journals, including both civilian and military-related publications. We will also present our findings at national and international meetings of learnt neuroscience and neuropsychiatry and military societies. We anticipate that our pilot study will inform a larger study on the long-term outcomes of TBI and quality of life, specific to military veterans, such that potential interventions may be accessed as quickly as possible.Trial registration number ISRCTN13276511.

Abstract: A recent pilot study in amyotrophic lateral sclerosis (ALS) patients analyzed the effect of a Mediterranean diet (MeDi) supplemented with nicotinamide riboside (NR, a NAD+ promoter), pterostilbene (PTER, a natural antioxidant) and/or coconut oil on anthropometric variables in ALS patients. The results suggested that the MeDi supplemented with NR, PTER and coconut oil is the nutritional intervention showing the greatest benefits at anthropometric levels. Over the last 30 years, glucose intolerance has been reported in ALS patients. Thus, suggesting that an alternative source of energy may be preferential for motor neurons to survive. Ketone bodies (KBs), provided through a MeDi with a lower carbohydrate content but enriched with medium chain triglycerides, could be a therapeutic alternative to improve the neuromotor alterations associated with the disease. Nevertheless, the use of a coconut oil-supplemented diet, as potentially ketogenic, is a matter of controversy. In the present report we show that a MeDi supplemented with coconut oil increases the levels of circulating KBs in ALS patients.

Abstract: Summary: Human genetic studies of critical COVID-19 pneumonia have revealed the essential role of type I interferon-dependent innate immunity to SARS-CoV-2 infection. Conversely, an association between the HLA-B∗15:01 allele and asymptomatic SARS-CoV-2 infection in unvaccinated individuals was recently reported, suggesting a contribution of pre-existing T cell-dependent adaptive immunity. We report a lack of association of classical HLA alleles, including HLA-B∗15:01, with pre-omicron asymptomatic SARS-CoV-2 infection in unvaccinated participants in a prospective population-based study in the United States (191 asymptomatic vs. 945 symptomatic COVID-19 cases). Moreover, we found no such association in the international COVID Human Genetic Effort cohort (206 asymptomatic vs. 574 mild or moderate COVID-19 cases and 1,625 severe or critical COVID-19 cases). Finally, in the Human Challenge Characterisation study, the three HLA-B∗15:01 individuals infected with SARS-CoV-2 developed symptoms. As with other acute primary infections studied, no classical HLA alleles favoring an asymptomatic course of SARS-CoV-2 infection were identified.

Abstract: Resumen: La medicina familiar y comunitaria reúne buena parte de los elementos y valores que caracterizan la profesión de médico. Esto hace que pueda aportar unos fundamentos sólidos, útiles y duraderos al estudiante de Medicina sea cual sea la especialidad que elija en un futuro. Es necesario formar a profesionales capaces de ver al paciente desde un punto de vista holístico en su contexto personal y social, así como comprender los factores determinantes de la salud individual y comunitaria. Que comprendan al paciente y su sufrimiento, y que empaticen, para acompañarle durante su proceso de enfermedad. También se nos plantea el reto de situar a la atención primaria en el centro del sistema sanitario y hacer frente a una demanda creciente de médicos especialistas en Medicina Familiar y Comunitaria.Para la consecución de estos objetivos se hace estrictamente necesaria la integración de la Atención Primaria en los planes de estudios de las universidades, con intervención de médicos de familia en diferentes asignaturas, aportando una visión más transversal y holística en el abordaje de las distintas enfermedades y con más presencia en las actividades docentes y en las prácticas.El modelo propuesto por la Universidad Pompeu Fabra se inspira en la metodología de la mentoría en la que al estudiante se le asignará un médico de familia como mentor durante toda su formación de grado, y este le acompañará en sus estancias en el centro de atención primaria y en actividades relacionadas con diferentes asignaturas y velará por el cumplimiento de los objetivos de formación. También participará en su evaluación al finalizar cada curso. Este modelo pretende mejorar la calidad de las actividades vinculadas a la asistencia clínica y de las prácticas, y facilitará el conocimiento de la atención primaria y de la profesión de médico a través del contacto precoz y mantenido con la atención primaria. Abstract: Family and community medicine brings together many of the elements and values that characterize the medical practice. This means that it can provide solid, useful and lasting foundations to the medical student, whatever specialty they choose in the future. It is necessary to train professionals capable of seeing the patient from a holistic point of view in their personal and social context, as well as understanding the determining factors of individual and community health. And also understanding the patient and their suffering, and that they empathize, to accompany them during their illness process. We are also faced with the challenge of placing Primary Care at the center of the health system and facing a growing demand for doctors specializing in Family and Community Medicine.To achieve these objectives, it is strictly necessary to integrate Primary Care into the university curricula, with the intervention of family doctors in different subjects, providing a more transversal and holistic vision in the approach to different pathologies and with more presence in teaching activities and practices.The model proposed by the Pompeu Fabra University is inspired by the mentoring methodology in which the student will be assigned a family doctor as a mentor throughout their undergraduate training, and this will accompany them during their stays at the Primary Care center and in activities related to different subjects and will ensure compliance with the training objectives. They will also participate in evaluation at the end of each course. This model aims to improve the quality of activities linked to clinical care and practices, and will facilitate knowledge of Primary Care and the profession of physician with an early and maintained contact with Primary Care.

Abstract: In this paper we propose a generative adversarial network (GAN) framework to enhance the perceptual quality of compressed videos. Our framework includes attention and adaptation to different quantization parameters (QPs) in a single model. The attention module exploits global receptive fields that can capture and align long-range correlations between consecutive frames, which can be beneficial for enhancing perceptual quality of videos. The frame to be enhanced is fed into the deep network together with its neighboring frames, and in the first stage features at different depths are extracted. Then extracted features are fed into attention blocks to explore global temporal correlations, followed by a series of upsampling and convolution layers. Finally, the resulting features are processed by the QP-conditional adaptation module which leverages the corresponding QP information. In this way, a single model can be used to enhance adaptively to various QPs without requiring multiple models specific for every QP value, while having similar performance. Experimental results demonstrate the superior performance of the proposed PeQuENet compared with the state-of-the-art compressed video quality enhancement algorithms.

Abstract: Neural video compression has emerged as a novel paradigm combining trainable multilayer neural networks and machine learning, achieving competitive rate-distortion (RD) performances, but still remaining impractical due to heavy neural architectures, with large memory and computational demands. In addition, models are usually optimized for a single RD tradeoff. Recent slimmable image codecs can dynamically adjust their model capacity to gracefully reduce the memory and computation requirements, without harming RD performance. Abstract: Video frame interpolation involves the synthesis of new frames from existing ones. Convolutional neural networks (CNNs) have been at the forefront of the recent advances in this field. One popular CNN-based approach involves the application of generated kernels to the input frames to obtain an interpolated frame. Despite all the benefits interpolation methods offer, many of these networks require a lot of parameters, with more parameters meaning a heavier computational burden. Reducing the size of the model typically impacts performance negatively. This paper presents a method for parameter reduction for a popular flow-less kernel-based network (Adaptive Collaboration of Flows). Through our technique of removing the layers that require the most parameters and replacing them with smaller encoders, we reduce the number of parameters of the network and even achieve better performance compared to the original method. This is achieved by deploying rotation to force each individual encoder to learn different features from the input images. 82. Cloudy and Cloud-free Thermal Phase Curves with PICASO: Applications to WASP-43b

Abstract: We present new functionality within PICASO, a state-of-the-art radiative transfer model for exoplanet and brown dwarf atmospheres, by developing a new pipeline that computes phase-resolved thermal emission (thermal phase curves) from three-dimensional (3D) models. Because PICASO is coupled to Virga, an open-source cloud code, we are able to produce cloudy phase curves with different sedimentation efficiencies ($f_{sed}$) and cloud condensate species. We present the first application of this new algorithm to hot Jupiter WASP-43b. Previous studies of the thermal emission of WASP-43b from Kataria et al. found good agreement between cloud-free models and dayside thermal emission, but an overestimation of the nightside flux, for which clouds have been suggested as a possible explanation. We use the temperature and vertical wind structure from the cloud-free 3D general circulation models of Kataria et al. and post-process it using PICASO, assuming that clouds form and affect the spectra. We compare our models to results from Kataria et al., including Hubble Space Telescope Wide-Field Camera 3 (WFC3) observations of WASP-43b from Stevenson et al. In addition, we compute phase curves for Spitzer at 3.6 and 4.5 $\mu m$ and compare them to observations from Stevenson et al. We are able to closely recover the cloud-free results, even though PICASO utilizes a coarse spatial grid. We find that cloudy phase curves provide much better agreement with the WFC3 and Spitzer nightside data, while still closely matching the dayside emission. Abstract: El objetivo de este artículo parte del problema de la autoridad en la educación a fin de proporcionar algunas claves que permitan construir una posición de deseo en el educador y despertar la pasión y el deseo de saber en el sujeto de la educación. La crisis de la autoridad nos toca de lleno a todas aquellas personas que, como maestros, profesores y educadores representamos algo del sistema simbólico y el orden institucional que hace posible el lazo social. Así pues, desde el punto de vista educativo cabe preguntarse por el tipo de autoridad que habría que poder construir para que los agentes de la educación puedan seguir desempeñando su función educativa. El conocido ensayo en torno a «La crisis de la educación» de la filósofa alemana Hannah Arendt nos permite situar y actualizar, en una primera línea reflexiva, los cambios sociales, culturales y pedagógicos que han debilitado la autoridad en el campo educativo. Posteriormente, centramos el análisis en el cuestionamiento y la reformulación de los saberes que hay que transmitir en la escuela, un cuestionamiento que ha desplazado el rol docente hacia funciones más borrosas y confusas. La pregunta en torno a los actos pedagógicos que permitirían recuperar el vínculo y la autoridad educativa sitúa el deseo de saber del agente de la educación en el centro de la discusión. En este contexto, nuestra propuesta explora cómo asumir la responsabilidad de la función educativa mediante el compromiso decidido de transmitir un testimonio de deseo, es decir, una forma de relacionarse con el mundo y con el saber, buscando así una vía que permita revitalizar el vínculo educativo entre el educador y el sujeto de la educación y, con él, el lazo social entre las generaciones.

Abstract: The coronavirus disease 2019 (COVID-19) pandemic had a meaningful impact on several areas of human activity. With respect to psychological assessment, the requirements to study the fear of vaccination as a means to diminish negative behaviour towards vaccination had been reported. This study aimed to evaluate the factorial invariance of the six-item Vaccination Fear Scale (VFS-6) across individuals and cultures. To achieve this goal, a sample of university students was recruited (n = 2535; mean age = 20.59, SD = 2.04; males: 26.75%, females: 73.25%) from Spain (n = 388; 15.3%), Italy (n = 376; 14.83%), Lebanon (n = 487; 19.21%), Nigeria (n = 561; 22.13%), Turkey (n = 410; 16.17%), and Ukraine (n = 313; 12.34%). The results showed that the most appropriate factorial structure, exhibiting excellent fit indices, was a model with two correlated factors (cognitive symptoms: items 1, 2, and 4; somatic symptoms: items 3, 5, and 6) for both the total sample and individual samples from each country and language (Spanish, Italian, Arabic, English, Turkish, and Ukrainian). Notably, the VFS-6 demonstrated configural, metric, scalar, and strict invariance across sex. Regarding countries and languages, configural invariance was observed between them. Also, metric invariance was observed between Spain, Italy, and Ukraine and between Lebanon, Nigeria, and Turkey, which indicates the presence of two well-differentiated groups of countries and the possibility of inferential analysis between them. Item Response Theory analysis suggested an appropriate level of discrimination and difficulty of the test. These significant findings lay the groundwork for future investigations into vaccination fear across diverse cultural backgrounds, providing valuable insights for addressing vaccination-related concerns worldwide.

Abstract: Abstract Background The Lactobacillaceae family comprises many species of great importance for the food and healthcare industries, with numerous strains identified as beneficial for humans and used as probiotics. Hence, there is a growing interest in engineering these probiotic bacteria as live biotherapeutics for animals and humans. However, the genetic parts needed to regulate gene expression in these bacteria remain limited compared to model bacteria like E. coli or B. subtilis. To address this deficit, in this study, we selected and tested several bacteriophage-derived genetic parts with the potential to regulate transcription in lactobacilli. Results We screened genetic parts from 6 different lactobacilli-infecting phages and identified one promoter/repressor system with unprecedented functionality in Lactiplantibacillus plantarum WCFS1. The phage-derived promoter was found to achieve expression levels nearly 9-fold higher than the previously reported strongest promoter in this strain and the repressor was able to almost completely repress this expression by reducing it nearly 500-fold. Conclusions The new parts and insights gained from their engineering will enhance the genetic programmability of lactobacilli for healthcare and industrial applications.

87. Pleiotropic contribution of rbfox1 to psychiatric and neurodevelopmental phenotypes in two zebrafish models

Abstract: Abstract RBFOX1 is a highly pleiotropic gene that contributes to several psychiatric and neurodevelopmental disorders. Both rare and common variants in RBFOX1 have been associated with several psychiatric conditions, but the mechanisms underlying the pleiotropic effects of RBFOX1 are not yet understood. Here we found that, in zebrafish, rbfox1 is expressed in spinal cord, mid- and hindbrain during developmental stages. In adults, expression is restricted to specific areas of the brain, including telencephalic and diencephalic regions with an important role in receiving and processing sensory information and in directing behaviour. To investigate the contribution of rbfox1 to behaviour, we used rbfox1 sa15940 , a zebrafish mutant line with TL background. We found that rbfox1 sa15940 mutants present hyperactivity, thigmotaxis, decreased freezing behaviour and altered social behaviour. We repeated these behavioural tests in a second rbfox1 mutant line with a different genetic background (TU), rbfox1 del19 , and found that rbfox1 deficiency affects behaviour similarly in this line, although there were some differences. rbfox1 del19 mutants present similar thigmotaxis, but stronger alterations in social behaviour and lower levels of hyperactivity than rbfox1 sa15940 fish. Taken together, these results suggest that mutations in rbfox1 lead to multiple behavioural changes in zebrafish that might be modulated by environmental, epigenetic and genetic background effects, and that resemble phenotypic alterations present in Rbfox1-deficient mice and in patients with different psychiatric conditions. Our study, thus, highlights the evolutionary conservation of rbfox1 function in behaviour and paves the way to further investigate the mechanisms underlying rbfox1 pleiotropy on the onset of neurodevelopmental and psychiatric disorders.

89. Land use change alters carbon composition and degree of decomposition of tropical peat soils

Abstract: Drainage associated with land use change in tropical peatlands has increased the rate of decomposition of peat soils and contributed to CO2 emissions. Increased decomposition may result in changes in the composition of the soil organic carbon (SOC). We examined the carbon functional group composition and degree of decomposition of peat soils under five different land uses to understand the effects of changing management intensity on tropical peatland soils. Samples were collected from seven sites spanning five different land uses (forest, shrubland, fernland, revegetation, smallholder oil palm) at the Pedamaran peatland in South Sumatra, Indonesia. SOC composition, measured by Solid-state 13C Nuclear Magnetic Resonance (NMR) spectroscopy, was dominated by the alkyl carbon (C) functional group in managed peatlands. However, in the forest far from drainage canals, the SOC comprised predominantly O-alkyl C. The contributions of the functional groups ketone C, carbonyl C and O-aryl C were low and tended to occur in stable proportions throughout the soil profiles. Drainage and land use change significantly affected peat carbon chemistry. The effects were greatest under oil palm, where O-alkyl C had been depleted rapidly under aerobic conditions leading to a change in the dominant carbon functional group from O-alkyl C to alkyl C. Furthermore, our results indicate that the alkyl C:O-alkyl C ratio is a more useful and informative indicator of the degree of decomposition of peat soil than the traditionally used C:N ratio. This more nuanced understanding of the different types of carbon that make up tropical peat soils under different land uses can be applied to support peatland restoration. In particular, nutrient cycling and water availability are likely to be influenced by carbon functional group and degree of decomposition. In order to reduce fire risk and support Indonesia’s aspirations to manage the national forest estate as a net carbon sink, further research into the links between peat soil organic carbon chemistry, revegetation performance and new peat accumulation is recommended.

Abstract: Dermatomal analgesia achieved with quadratus lumborum blocks is site-dependent and inconsistent. Cadaveric and clinical studies reveal multiple mechanisms of action. We dissected six fresh human cadavers bilaterally and thoroughly studied their neurological linkages to the quadratus lumborum muscle (QLM) to identify neural structures and block targets. At the end of the investigation, only the subcostal nerve (anterolateral) and the ilioinguinal nerves were found near the QLM in all specimens. The iliohypogastric nerve was found in only two specimens. No further neural targets were found in the fascial planes before and posterior to the QLM.

Abstract: This study aimed to assess the reliability and validity of the Polar V800 to measure vertical jump height. Twenty-two physically active healthy men (age: 22.89 +- 4.23 years; body mass: 70.74 +- 8.04 kg; height: 1.74 +- 0.76 m) were recruited for the study. The reliability was evaluated by comparing measurements acquired by the Polar V800 in two identical testing sessions one week apart. Validity was assessed by comparing measurements simultaneously obtained using a force platform (gold standard), high-speed camera and the Polar V800 during squat jump (SJ) and countermovement jump (CMJ) tests. In the test-retest reliability, high intraclass correlation coefficients (ICCs) were observed (mean: 0.90, SJ and CMJ) in the Polar V800. There was no significant systematic bias +- random errors (p > 0.05) between test-retest. Low coefficients of variation (<5%) were detected in both jumps in the Polar V800. In the validity assessment, similar jump height was detected among devices (p > 0.05). There was almost perfect agreement between the Polar V800 compared to a force platform for the SJ and CMJ tests (Mean ICCs = 0.95; no systematic bias +- random errors in SJ mean: -0.38 +- 2.10 cm, p > 0.05). Mean ICC between the Polar V800 versus high-speed camera was 0.91 for the SJ and CMJ tests, however, a significant systematic bias +- random error (0.97 +- 2.60 cm; p = 0.01) was detected in CMJ test. Abstract: The Cherenkov Telescope Array (CTA) is the next-generation ground-based very-high-energy gamma-ray observatory. The Large-Sized Telescope (LST) of CTA is designed to detect gamma rays between 20 GeV and a few TeV with a 23-meter diameter mirror. We have developed the focal plane camera of the first LST, which has 1855 photomultiplier tubes (PMTs) and the readout system which samples a PMT waveform at GHz with switched capacitor arrays, Domino Ring Sampler ver4 (DRS4). To measure the precise pulse charge and arrival time of Cherenkov signals, we developed a method to calibrate the output voltage of DRS4 and the sampling time interval, as well as an analysis method to correct the spike noise of DRS4. Since the first LST was inaugurated in 2018, we have performed the commissioning tests and calibrated the camera. Abstract: Classical novae are cataclysmic binary star systems in which the matter of a companion star is accreted on a white dwarf (WD). Accumulation of hydrogen in a layer eventually causes a thermonuclear explosion on the surface of the WD, brightening the WD to ~10^5 solar luminosities and triggering ejection of the accumulated matter.They provide extreme conditions required to accelerate particles, electrons or protons, to high energies. Here we present the detection of gamma rays by the MAGIC telescopes from the 2021 outburst of RS Ophiuchi (RS Oph), a recurrent nova with a red giant (RG) companion, that allowed us, for the first time, to accurately characterize the emission from a nova in the 60 GeV to 250 GeV energy range. The theoretical interpretation of the combined Fermi-LAT and MAGIC data suggests that protons are accelerated to hundreds of GeV in the nova shock. Abstract: The blazar TXS 0506+056 got into the spotlight of the astrophysical community in September 2017, when a high-energy neutrino detected by IceCube (IceCube-170922A) was associated at the 3 $\sigma$ level to a $\gamma$-ray flare from this source. This multi-messenger photon-neutrino association remains, as per today, the most significant one ever observed. TXS 0506+056 was a poorly studied object before the IceCube-170922A event. To better characterize its broad-band emission, we organized a multi-wavelength campaign lasting 16 months (November 2017 to February 2019), covering the radio-band (Mets\"ahovi, OVRO), the optical/UV (ASAS-SN, KVA, REM, Swift/UVOT), the X-rays (Swift/XRT, NuSTAR), the high-energy $\gamma$ rays (Fermi/LAT) and the very-high-energy (VHE) $\gamma$ rays (MAGIC). In $\gamma$ rays, the behaviour of the source was significantly different from the 2017 one: MAGIC observations show the presence of flaring activity during December 2018, while the source only shows an excess at the 4$\sigma$ level during the rest of the campaign (74 hours of accumulated exposure); Fermi/LAT observations show several short (days-to-week timescale) flares, different from the long-term brightening of 2017. No significant flares are detected at lower energies. The radio light curve shows an increasing flux trend, not seen in other wavelengths. We model the multi-wavelength spectral energy distributions in a lepto-hadronic scenario, in which the hadronic emission emerges as Bethe-Heitler and pion-decay cascade in the X-rays and VHE $\gamma$ rays. Abstract: In this paper, we propose a deformable convolution-based generative adversarial network (DCNGAN) for perceptual quality enhancement of compressed videos. DCNGAN is also adaptive to the quantization parameters (QPs). Compared with optical flows, deformable convolutions are more effective and efficient to align frames. Deformable convolutions can operate on multiple frames, thus leveraging more temporal information, which is beneficial for enhancing the perceptual quality of compressed videos. Instead of aligning frames in a pairwise manner, the deformable convolution can process multiple frames simultaneously, which leads to lower computational complexity.
