Valuable precursors of popular chiral drugs propranolol and pindolol, 3-(1-naphthyloxy)-propane-1,2-diol 3 and 3-(4-indolyloxy)-propane-1,2-diol 4 were investigated by IR spectroscopy, DSC, and X-ray diffraction methods. Both compounds, crystallizing from enantiopure feed material, form “guaifenesin-like” crystal packing in which the classic H-bonded bilayers, framed in both sides by hydrophobic fragments of the molecules, acts as the basic crystal-forming motif. Diol 4 prone to spontaneous resolution and conserves its packing pattern crystallizing from racemate. Under the same conditions, diol 3 forms weakly stable solid racemic compound. Some reasons for such a behavior are identified and discussed.

An I2–NaOAc mediated one-pot process for the oxidative dimerization of indolizines under mild conditions is described. The reaction afforded a variety of products in good to excellent yields from indolizines with acceptor groups. Macrocyclic systems with the biindolizine fragment(s) were also synthesized from monoindolizinylquinoxaline podands under the same conditions.

A highly efficient and versatile method for the synthesis of quinoxaline derivatives with two benzimidazole fragments have been developed on the basis of the ring contraction of 3-(benzimidazo-2-yl)quinoxalin-2(1H)-one with 1,2-diaminobenzene and its various types of substituted and condensed derivatives. Owing to the inter- and intramolecular processes, involving self association, proton exchange, conformational, and/or tautomeric exchanges between several forms for most of the bis-benzimidazolylquinoxalines signals of bridged and neighboring carbon atoms and the hydrogen atoms of the neighboring carbon atoms of benzimidazole fragments in the NMR spectra are broadened. The conjugation between the benzimidazole fragments and the quinoxaline core of the molecules is increased from the quinoxaline derivative (10c) to its thiadiazol[f]- (17) and pyrrolo[a]-(19) annulated derivatives, resulting in a greater planarity of the molecule as a whole.

A reaction of 3-(α-aminobenzyl)quinoxalin-2(1H)-one with ethyl acetoacetate in boiling acetic acid is accompanied by contraction of the pyrazine ring. A rearrangement involving the fragment C(2)-C(3)-C(NH2)Ph of the quinoxaline system and the fragment C(2)-C(3) of ethyl acetoacetate yields 2-(4-ethoxycarbonyl-5-methyl-2-phenylpyrrol-3-yl)benzimidazole. Possible pathways of this reaction are considered.

The reaction of substrates (I) and (III) proceeds under extremely mild conditions to give target compounds of type (II) and macrocyclic ring systems (IV) including biindolizine fragments.

For a family of chiral terminal ortho-alkylphenyl glycerol ethers [alkyl = Me (1), Et (2), n-Pr (3), i-Pr (4), t-Bu (5)] nine different crystalline modifications have been obtained and investigated by X-ray analysis. All possible types for chiral compound crystallization, including a very rare type of false conglomerate (polymorph of rac-4.1) and extremely rare case of anomalous conglomerate (metastable form of rac-5) have been found within this set. Three tested substances crystallize as normal conglomerates, while two are capable of forming normal racemic compounds. Centrosymmetrical dimers of enantiomers, which are typical for most racemates, are absent in all the investigated crystals. The samples of the enantiopure compound 2 and 3 form stable supramolecular gels in hydrocarbon solvents. Some assumptions have been made linking the features of the molecular and crystal structures in the investigated series.
