It is well known that β-defensins are key components of the host innate immune response against pathogens and potentially provide a link between innate and adaptive immunity. In zebrafish (Danio rerio), a vertebrate model species in numerous biomedical fields, three β-defensin isoforms were recently identified. To our knowledge, however, studies describing antimicrobial or immunomodulatory properties of any of the zebrafish β-defensins isoforms are absent today. Since it is indubitable that deepening the study of zebrafish β-defensins would be of interest in this work we investigated whether or not the zebrafish β-defensin 2 (zfBD2) has the antiviral properties described for their vertebrate counterparts. Our in vitro and in vivo studies showed that zfBD2 has antiviral activity, immunomodulatory properties and, most importantly, is a potent viral DNA vaccine molecular adjuvant. In addition, a potential relationship between zfBD2 activity and the NF-κB signaling pathway is suggested. Altogether these results show that the zebrafish could be a suitable in vivo animal model to study the roles played by β-defensin 2 in viral diseases, vaccinology and even in clinical dermatology. The rainbow trout (Oncorhynchus mykiss) is one of most significant model in fish immunology, as well as a key species in European aquaculture. Two well-known Novirhabdoviruses, the viral haemorrhagic septicemia virus (VHSV) and the infectious hematopoietic necrosis virus (IHNV) cause serious damage in fish farms and represent a significant threat for aquaculture in a number of countries. A significant variability of resistance to these infections - intra- and inter-populations - has been described in rainbow trout. This genetic variability is not only relevant for the selection of fish naturally resistant to these viruses, but also for the dissection of the mechanisms of fish antiviral immunity. The methods used to analyze the genetic bases of the trout resistance to VHSV, and primary results are reported here from the identification of a major QTL to the impact of type I interferons.

Australian bat lyssavirus (ABLV) is a rhabdovirus of the lyssavirus genus capable of causing fatal rabies-like encephalitis in humans. There are two variants of ABLV, one circulating in pteropid fruit bats and another in insectivorous bats. Three fatal human cases of ABLV infection have been reported with the third case in 2013. Importantly, two equine cases also arose in 2013; the first occurrence of ABLV in a species other than bats or humans. We examined the host cell entry of ABLV, characterizing its tropism and exploring its cross-species transmission potential using maxGFP-encoding recombinant vesicular stomatitis viruses that express ABLV G glycoproteins. Results indicate that the ABLV receptor(s) is conserved but not ubiquitous among mammalian cell lines and that the two ABLV variants can utilize alternate receptors for entry. Proposed rabies virus receptors were not sufficient to permit ABLV entry into resistant cells, suggesting that ABLV utilizes an unknown alternative receptor(s)., (Published by Elsevier Inc.)

Question: How does virulence evolve in the Drosophila melanogaster /sigma virus (DMelSV) system?, Organisms: Drosophila melanogaster (host) and DMelSV (parasite)., Empirical Methods: Artificial selection on whole-carcass viral titre of infected flies, including two selection regimes (maternal and biparental transmission) and three treatments within each regime (increased titre, decreased titre, and control). The maternal transmission selection regime lasted for six generations, while the biparental transmission selection regime lasted for twelve generations. We further quantified virulence by estimating the fecundity, viability, and development time of infected flies. Finally, we sequenced virus strains at the end of selection., Predictions and Conclusions: Titre is defined here as the number of viral genomes inside a single fly, while virulence is defined as harm to host. We predicted that titre would respond to both increased and decreased selection, that virulence would evolve as a positively correlated response, and that sequence evolution in the viruses would be responsible for these changes. Titre did respond to selection in the biparental regime, although both high and control lines both demonstrated increased titre, while the titre of the low lines did not change. One component of virulence, development time, was positively correlated with titre in the biparental transmission lines (maternal transmission lines were not scored for virulence). However, we detected few (and in some cases, no) genomic changes in the virus, making viral evolution unlikely to be responsible for the response to selection and the association between development time and titre.

Epizootic ulcerative syndrome is one of the most serious infections noticed in the finfish of Asia-Pacific during the last three decades. Different viral agents and a consistent fungus were isolated from the EUS infected fishes from various countries. Rhabdoviruses are by far the most isolated group of viruses followed by birnaviruses. One reovirus and a ranavirus have also been isolated from ulcerated fishes of which the ranavirus was capable of reproducing the clinical signs of the disease. Albeit heterogenic, due to frequent isolation, range of areas from which isolates have been obtained and ability to induce dermal lesions in experimental infectivity studies make rhabdoviruses one of the likely primary aetiological agents that could be triggering the initiation to EUS. However, further investigations may be required to fully establish the role of these viral agents in the induction of EUS. Viruses observed as persistent infections in fishes evidenced by their presence in cell cultures also require further investigation for their likely role in predisposing the fishes to EUS.

Potato yellow dwarf virus (PYDV) is the type species of the genus Nucleorhabdovirus and, like all members of this genus, replication and morphogenesis occurs inside the nuclei of infected cells. Protein localization prediction algorithms failed to identify a nuclear localization signal (NLS) in PYDV nucleocapsid (N) protein, although PYDV-N has been shown to localize exclusively to the nucleus when expressed as a green fluorescent protein (GFP):N fusion in plant cells. Deletion analysis using fragments of PYDV-N identified a karyophilic region in the carboxy-terminal 122 amino acids. Alanine-scanning mutagenesis was performed across this region in the context of the full-length N protein. Mutants were assayed for their ability to nuclear localize using live-cell imaging and a yeast-based assay. Two amino acid motifs, (419)QKR(421) and (432)KR(433) were shown to be essential for nuclear import and interaction with importin-α. Additional bimolecular fluorescence complementation showed that the PYDV-N-NLS mutants cannot be ferried into the nucleus via interaction with PYDV-P or -M. In contrast, interaction with N-NLS mutants appeared to retard the nuclear import of PYDV-P. GFP fused to aa 419-434 established that the PYDV-N-NLS can function outside the context of this protein. Taken together, it was determined that PYDV-N contains the bipartite NLS (419)QKRANEEAPPAAQKR(433).

Rhabdoviruses enter the cell via the endocytic pathway and subsequently fuse with a cellular membrane within the acidic environment of the endosome. Both receptor recognition and membrane fusion are mediated by a single transmembrane viral glycoprotein (G). Fusion is triggered via a low-pH induced structural rearrangement. G is an atypical fusion protein as there is a pH-dependent equilibrium between its pre- and post-fusion conformations. The elucidation of the atomic structures of these two conformations for the vesicular stomatitis virus (VSV) G has revealed that it is different from the previously characterized class I and class II fusion proteins. In this review, the pre- and post-fusion VSV G structures are presented in detail demonstrating that G combines the features of the class I and class II fusion proteins. In addition to these similarities, these G structures also reveal some particularities that expand our understanding of the working of fusion machineries. Combined with data from recent studies that revealed the cellular aspects of the initial stages of rhabdovirus infection, all these data give an integrated view of the entry pathway of rhabdoviruses into their host cell.

The family Rhabdoviridae contains important pathogens of humans, livestocks or even crops. The matrix protein of rhabdovirus is a constituent of the virion, forming a layer between the viral envelope and the nucleocapsid. It is crucial for the bullet-like morphology of the virion. There is a strong structural convergence between the lyssavirus and vesiculovirus matrix proteins. These matrix proteins are able to self assemble to form non-covalent polymers, acting as a scaffold for the other components of the virion. The matrix protein of rhabdovirus also has non-structural functions within the viral cycle: it is involved in the expression and replication of the viral genome, and in the virion budding. Moreover, it is a key mediator of the interactions between the virus and the infected cell, even involved in the death of the cell. To conclude, the repertoire of the matrix protein of rhabdovirus functionalities keeps on expanding.

Like many animal viruses, those of the Rhabdoviridae family, are able to antagonize the type I interferon response and cause disease in mammalian hosts. Though these negative-stranded RNA viruses are very simple and code for as few as five proteins, they have been seen to completely abrogate the type I interferon response early in infection. In this review, we will discuss the viral organization and type I interferon evasion of rhabdoviruses, focusing on vesicular stomatitis virus (VSV) and rabies virus (RABV). Despite their structural similarities, VSV and RABV have completely different mechanisms by which they avert the host immune response. VSV relies on the matrix protein to interfere with host gene transcription and nuclear export of anti-viral mRNAs. Alternatively, RABV uses its phosphoprotein to interfere with IRF-3 phosphorylation and STAT1 signaling. Understanding the virus-cell interactions and viral proteins necessary to evade the immune response is important in developing effective vaccines and therapeutics for this viral family.

Glycoprotein G of the rhabdoviruses is involved in both receptor recognition at the host cell surface and membrane fusion via a low pHinduced structural rearrangement. G is an atypical fusion protein as there is a pH-dependent equilibrium between the pre- and post-fusion conformations of the protein. The atomic structures of the pre- and post-fusion conformations reveal that G is homologous to both glycoprotein gB of herpesviruses and gp64 of baculovirus and also that it combines features of the previously characterized class I and class II fusion proteins. Comparison of the structures of G pre- and post-fusion states reveals an extensive structural reorganization of the molecule that resembles that of paramyxovirus fusion protein F. It also allows to localize the fusion loops and to identify conserved key residues that constitute pH-sensitive molecular switches. Finally, the fusion properties and the structures of G also reveal some particularities that invite us to reconsider a few dogmas concerning fusion proteins.

Mosquito-borne infections cause some of the most debilitating human diseases, including yellow fever and malaria, yet we lack an understanding of how disease risk scales with human-driven habitat changes. We present an approach to study variation in mosquito distribution and concomitant viral infections on the landscape level. In a pilot study we analyzed mosquito distribution along a 10-km transect of a West African rainforest area, which included primary forest, secondary forest, plantations, and human settlements. Variation was observed in the abundance of Anopheles, Aedes, Culex, and Uranotaenia mosquitoes between the different habitat types. Screening of trapped mosquitoes from the different habitats led to the isolation of five uncharacterized viruses of the families Bunyaviridae, Coronaviridae, Flaviviridae, and Rhabdoviridae, as well as an unclassified virus. Polymerase chain reaction screening for these five viruses in individual mosquitoes indicated a trend toward infection with specific viruses in specific mosquito genera that differed by habitat. Based on these initial analyses, we believe that further work is indicated to investigate the impact of anthropogenic landscape changes on mosquito distribution and accompanying arbovirus infection., Human Evolutionary Biology
