The scanning superconductivity quantum interference device (SQUID) microscope is widely used to detect and analyze weak magnetic fields above rock samples. To measure the saturation isothermal remanence curve, an extra magnetization and demagnetization system is needed. In this paper, we designed an in situ magnetization and demagnetization system that can be used in the scanning SQUID microscope. Measurement of the magnetic properties of the samples could be implemented without moving the samples manually. Magnetizing and demagnetizing fields were produced separately by activating different changeover switches to apply the corresponding power supply to the same coil. The effectiveness of this system was verified in the experiment by measuring different magnetic fields produced by this coil. Results showed that the design was reasonable and feasible. Rab GTPases are required for vesicle-vacuolar fusion during vacuolar biogenesis in fungi. To date, little is known about the biological functions of the Rab small GTPase components in Magnaporthe oryzae. In this study, we investigated MoYpt7 of M. oryzae, a homologue of the small Ras-like GTPase Ypt7 in Saccharomyces cerevisiae. Cellular localization assays showed that MoYpt7 was predominantly localized to vacuolar membranes. Using a targeted gene disruption strategy, a MoYPT7 mutant was generated that exhibited defects in mycelial growth and production of conidia. The conidia of the MoYPT7 mutant were malformed and defective in the formation of appressoria. Consequently, the MoYPT7 mutant failed to cause disease in rice and barley. Furthermore, the MoYPT7 mutant showed impairment in autophagy, breached cell wall integrity, and higher sensitivity to both calcium and heavy metal stress. Transformants constitutively expressing an active MoYPT7 allele (MoYPT7-CA, Gln67Leu) exhibited distinct phenotypes from the MoYPT7 mutant. Expression of MoYPT7-CA in MoYpt7 reduced pathogenicity and produced more appressoriaforming single-septum conidia. These results indicate that MoYPT7 is required for fungal morphogenesis, vacuole fusion, autophagy, stress resistance and pathogenicity in M. oryzae. [ABSTRACT FROM AUTHOR]

560. The Efficacy and Safety of the Probiotic Bacterium Lactobacillus reuteri DSM 17938 for Infantile Colic: A Meta-Analysis of Randomized Controlled Trials.

Objective: To evaluate the efficacy and safety of Lactobacillus reuteri DSM 17938 for treating infantile colic. Methods: A systematic literature retrieval was carried out to obtain randomized controlled trials of L. reuteri DSM 17938 for infantile colic. Trials were performed before May 2015 and retrieved from the PubMed, EMBASE, Cochrane library, CNKI, WanFang, VIP, and CBM databases. Data extraction and quality evaluation of the trials were performed independently by two investigators. A meta-analysis was performed using STATA version 12.0. Results: Six randomized controlled trials of 423 infants with colic were included. Of these subjects, 213 were in the L. reuteri group, and 210 were in the placebo group. Lactobacillus reuteri increased colic treatment effectiveness at two weeks (RR = 2.84; 95% CI: 1.24–6.50; p = 0.014) and three weeks (relative risk [RR] = 2.33; 95% CI: 1.38–3.93; P = 0.002) but not at four weeks (RR = 1.41; 95% CI: 0.52–3.82; P = 0.498). Lactobacillus reuteri decreased crying time (min/d) at two weeks (weighted mean difference [WMD] = –42.89; 95% CI: –60.50 to –25.29; P = 0.000) and three weeks (WMD = –45.83; 95% CI: –59.45 to –32.21; P = 0.000). In addition, L. reuteri did not influence infants’ weight, length or head circumference and was not associated with serious adverse events. Conclusions: Lactobacillus reuteri possibly increased the effectiveness of treatment for infantile colic and decreased crying time at two to three weeks without causing adverse events. However, these protective roles are usurped by gradual physiological improvements. The study is limited by the heterogeneity of the trials and should be considered with caution. BACKGROUND The root bark of Aralia is a rich source of bioactive components that may improve glycemic control and lipid status. In this study, 148 patients with type 2 diabetes mellitus ( T2DM) were assigned randomly to receive either glipizide alone or glipizide plus Aralia root bark extract ( ARBE) for 8 weeks to test the effects of ARBE plus glipizide therapy on glycemic control and lipid profiles in these patients. RESULTS Levels of HbA1c, fasting plasma glucose ( FPG) and 2 h postprandial plasma glucose (2-h PPG) in both groups significantly decreased from baseline. Glycated hemoglobin ( HbA1c) decreased marginally significantly in participants taking glipizide plus ARBE compared with the glipizide group ( P = 0.06). Participants in the combination group had significant decreases in total cholesterol ( TC) and low-density lipoprotein cholesterol ( LDL-C), and the between-group difference achieved statistical significance for LDL-C ( P = 0.04). Reduction in HbA1c in the combination group was significantly associated with changes in TC ( r = 0.32; P = 0.006) and LDL-C ( r = 0.34; P = 0.005), and the change in FPG was inversely correlated with LDL-C reduction ( r = 0.34; P = 0.004). CONCLUSIONS In patients with T2DM, combination therapy with glipizide and ARBE resulted in moderately lowering HbA1c and LDL-C levels compared with glipizide alone. © 2014 Society of Chemical Industry [ABSTRACT FROM AUTHOR]

While the adverse effects of raising a child with autism are well demonstrated, there have been few reports of the post-traumatic growth of mothers of children with autism. The purpose of this research was to explore dimensions of post-traumatic growth in this population in Mainland China and identify the factors facilitating post-traumatic growth. A total of 11 mothers of pre-school children were recruited from five rehabilitation centres of children with disabilities in Shanghai. The semi-structured interviews were conducted between August 2012 and October 2012 and analysed using thematic analysis. The data partially confirmed Tedeschi and Calhoun’s model of post-traumatic growth. A new philosophy of life, appreciation of life, relating to others, personal strength and spiritual change were five domains of post-traumatic growth in mothers of children with autism. Perceived social support, peer example, effective coping style and self-efficacy enhancement were facilitating factors of post-traumatic growth. By lead the warp yarn production process of fancy yarn in continuous fiber production process, the ramie/PP warp yarn textile composite has been prepared. The appearance, hemp content, mechanical property, structure of the warp yarn textile composite were texted. The result shows that along with the increase of the hollow in got parameters, the fabric hemp content decreased. When the warp yarn textile composite stretched, it turns out that the fracture is multi-storey in high power. The fracture of heart yarn was related to coating tightness. - Abstract
The pathogenic fungus, Magnaporthe oryzae, has emerged as a model system for dissecting fungal-plant interactions. Considerable progress has been made in identifying genes necessary for the regulation of pre- and post-penetration events in the development and pathogenicity of the fungus, such as the appressorium (a dome-shaped infection structure) mediated penetration and invasive growth of the pathogen. However, the detailed molecular mechanisms of these developmental stages, especially appressorium morphogenesis, are still relatively poorly understood. Currently, we isolated the MNH6 gene for a nonhistone chromosomal protein and 24 autophagic genes from the genome of this fungus. These genes are highly conserved among other eukaryotes, including humans and plants, and they control the capability of surviving starvation, conidiation, conidial germination, lipid turnover, and appressorium turgor generation. Clarification of the functions of these autophagy genes and their interaction networks will allow us to understand the role of autophagic genes in fungal pathogenesis. [ABSTRACT FROM AUTHOR]

- Abstract
Rice blast is a devastating disease worldwide, threatening rice production and food security. The blast fungus Magnaporthe oryzaeinvades the host via the appressorium, a specialized pressure-generating structure that generates enormous turgor pressure to penetrate the host cuticle. However, owing to ongoing evolution of fungicide resistance, it is vitally important to identify new targets and fungicides. Here, we show that Trs85, a subunit of the transport protein particle III complex, is essential for appressorium-mediated infection in M. oryzae. We explain how Trs85 regulates autophagy through Ypt1 (a small guanosine triphosphatase protein) in M. oryzae. We then identify a key conserved amphipathic α helix within Trs85 that is associated with pathogenicity of M. oryzae. Through computer-aided screening, we identify a lead compound, SP-141, that affects autophagy and the Trs85–Ypt1 interaction. SP-141 demonstrates a substantial capacity to effectively inhibit infection caused by the rice blast fungus while also exhibiting wide-ranging potential as an antifungal agent with broad-spectrum activity. Taken together, our data show that Trs85 is a potential new target and that SP-141 has potential for the control of rice blast. Our findings thus provide a novel strategy that may help in the fight against rice blast.

593. Effects of back fat, growth rate, and age at first mating on Yorkshire and Landrace sow longevity in China

- Abstract
In the present study, genetic parameters for longevity and genetic correlations with back fat (BF), average daily gain (ADG), and first mating age (FMA) were estimated from 19 300 Yorkshire and 18 378 Landrace sows from 12 farms derived from one breeding system. General linear models (GLM) were employed to estimate genetic correlations with heritability (h2). Longevity was defined by six different parameters (i.e., traits) and heritability ranged from h2=0.13 to 0.16 (P<0.05) in both swine breeds. Genetic correlations between different longevity definitions were positive and high (r=0.79 to 0.99; P<0.05). BF exhibited low positive genetic correlations (r=0.12 to 0.15; P<0.05) with longevity traits identified in both pig breeds which were dependent on trait definition, whereas ADG and FMA showed weak negative genetic correlations with longevity traits. The influence of BF, ADG, and FMA on longevity traits was examined by dividing the production and reproductive traits into several numerical groups; the influence of each longevity trait was subsequently analyzed among the different groups. The results indicated BF was the best trait to improve longevity among BF, ADG, and FMA, which showed positive effects for almost all longevity traits. For Yorkshire sows, gilts with over 18 mm BF were most desirable; Landrace with 10.6–13 mm BF were suitable for longevity. ADG was not favorable for gilt selection, because the correlation between growth rate and longevity very low. For FMA, our results indicated gilts mating before 230 days exhibited a longer life span. Therefore, we suggest farmers should select for thicker back fat and mate at an earlier age to improve sow longevity.

Background: Recently, Sigma nonopioid intracellular receptor 1 (SIGMAR1) variants have been shown harboring C9orf72 pathogenic repeat expansions in some frontotemporal dementia (FTD) cases. However, no SIGMAR1 genotype analysis has been reported in a cohort absent of C9orf72 pathogenic repeat expansions to date. Objective: The present study investigated the contribution of SIGMAR1 independent of C9orf72 gene status to FTD spectrum syndromes. Methods: We directly sequencing the entire coding region and a minimum of 50 bp from each of the flanking introns of SIGMAR1 gene in 82 sporadic FTD patients (female: male = 42 : 40) and 417 controls. For the patient carrying SIGMAR1 variant, a follow-up 3T MR imaging was performed in the study. Results: Gene sequencing of SIGMAR1 revealed a rare 3′UTR nucleotide variation rs192856872 in a male patient with semantic dementia independent of C9orf72 gene status. The MR imaging showed asymmetrical atrophy in the anterior temporal lobes and the degeneration extends caudally into the posterior temporal lobes as the disease progresses. ESEFinder analysis showed new SRSF1 and SRSF1-IgM-BRCA1 binding sites with significant scores, which is predicted to affect normal splicing. Conclusion: We found a novel SIGMAR1 variant independent of C9orf72 gene status associated with semantic dementia phenotype. [ABSTRACT FROM AUTHOR]

The study evaluated the impact of solution-focused brief therapy (SFBT) group-work on the post-traumatic growth (PTG) of mothers who have a child with ASD. A quasi-experimental design was used in which 43 mothers participated. 18 mothers in 2 SFBT groups (n = 9 in each group) received a 6-session SFBT group therapy while 25 mothers in a control group received no treatment. The Post-traumatic Growth Inventory was used to measure the PTG levels of the participants at baseline, post-treatment and 6-month follow-up assessments. Mothers who attended SFBT group-work reported higher PTG scores both at post-treatment (t = 4.065, p = .001) and 6-month follow-up (t = 2.980, p = .006) assessments. Further investigations to prove whether SFBT in groups can increase the positivity of clients would promote the use of SFBT. [ABSTRACT FROM AUTHOR]
