Mischief Reef is located in the eastern Nansha Islands of the South China Sea. With increasingly intense anthropogenic disturbance, Pinctada fucata, a previously unrecorded species in the reef, has occurred in the region. In this study, we identified and described the occurrence of P. fucata in Mischief Reef based on morphology and molecular markers. Furthermore, we performed a population genetics analysis of seven P. fucata populations of the South China Sea. All P. fucata populations showed significant high-level genetic diversity, but the differentiation among P. fucata populations was small. There was an FST value close to zero (−0.0083) between the Lingshui and Mischief Reef populations. Our results hint that Lingshui may be one of the potential sources of P. fucata to Mischief. In addition, we discussed the possible cause of the mass occurrence of P. fucata. The Luciolinae genus Emeia Fu, Ballantyne & Lambkin, 2012 is reviewed. Phylogenetic relationships based on cox1 DNA barcoding sequences from 42 fireflies and 2 outgroup species are reconstructed. The dataset included three main Lampyridae subfamilies: Luciolinae, Photurinae and Lampyrinae, and Emeia was recovered within Luciolinae. A new species, Emeia pulchra Zhu & Zhen sp. nov., is described from the wetland of Lishui, Zhejiang, China. Emeia pulchra is sister species to E. pseudosauteri from Sichuan, which is supported by morphological characters and a phylogeny based on DNA barcoding sequences. Artificial breeding of the iron clam, Cyclina sinensis, was successful for the first time in Taiwan in 2020. To prevent potential inbreeding depression and genetic incompatibility, the genetic diversity and structure of wild populations of C. sinensis must be assessed. The genetic diversity of 153 iron clams from 7 sample populations in Taiwan, including the Penghu and Kinmen Islands, was examined in this study. A total of 658 base pairs in the mitochondrial DNA cytochrome c oxidase subunit I sequence revealed only one phylogenetically informative site, and 15 haplotypes were identified. Compared with populations on the Chinese coast, those in the south of and in Taiwan Strait exhibited much lower nucleotide diversity (0.0003 vs. 0.0022), which may cause difficulties in the artificial breeding of C. sinensis in Taiwan. Compared with other bivalves, the C. sinensis of northern China did not exhibit lower nucleotide diversity (0.0022 vs. 0.002--0.006). Demographic analyses of C. sinensis in East Asia revealed no pattern of decline. Although Taiwan is closer to southern China (FST = 0.000), the introduction of broodstock from northern China should be evaluated for viability. In addition, our study revealed a difference in the genetic composition and genetic diversity of C. sinensis in Taijiang National Park before and after the enactment of the annual catch limit policy, which warrants further study. For more than a decade, various research groups have tracked the population genetics of the oriental fruit fly, Bactrocera dorsalis (Hendel) in China and neighboring countries using mitochondrial cytochrome c oxidase subunit I (COI) DNA. Although most research has reported high levels of mtDNA variation, to date no efforts have been made to integrate and compare the results from these studies simultaneously. Here, we show that: 1) despite the fact that a large portion of the sampling effort has focused on the Yunnan province beginning in 2005, each subsequent study recovers only a small number of previously sampled haplotypes; 2) new haplotypes of B. dorsalis remain to be found, a projection of new haplotypes versus the number of individuals sampled suggest that sampling the species mtDNA diversity is far from reaching an asymptote; 3) it is unlikely that the observed genetic variation is the result of NUMTs (nuclear mitochondrial DNA), as most differences between haplotypes are silent substitutions; and 4) although all studies employed the 3′ end of COI, the length of COI fragment sequenced differs among studies, making comparisons challenging. Neuroptera (lacewings) is one of the ancient holometabolous insect groups, but some extant species stand as important natural enemies for biological control. As the capital city of China, Beijing has a rich fauna of Neuroptera, previously with 47 species recorded and sorted in 32 genera of seven families. In this study, DNA barcoding based on sequences of COI gene fragments is used to discriminate lacewing species from Beijing. 217 DNA barcode sequences belonging to 49 species were successfully obtained. The COI barcode data worked well for identification of almost all lacewing species herein examined except Pseudomallada prasinus (Burmeister), in which cryptic species may exist. Twenty species of Neuroptera are newly recorded from Beijing. Besides, Nothochrysinae is first recorded from Beijing. The genetic diversity and population genetic structure ofOnchidium “struma”were investigated using mitochondrial cytochrome c oxidase subunit I (COI) gene sequences. A total of 240 individuals representing 10 collection sites from across a large portion of its known range were included in the analysis. Overall, 42 haplotypes were defined and 97 polymorphic sites were observed. TheO. “struma”populations had high haplotype diversity (0.9280) and nucleotide diversity (0.0404). We inferred that the early maturity and extensive survival habitat led to high genetic diversity ofO. “struma”populations in China. Bayesian analysis and SAMOVA analysis showed significant genetic differentiation among populations and all populations were divided into two groups, (HK and HN) versus (GY, DF, CX, CN, ND and XM). The Mantel test revealed no significant correlation between geographic distance and genetic distance (r = 0.251;p = 0.058). Restricted gene flow caused by a shorter term pelagic veliger stage and limited dispersal potential were inferred to result in genetic differentiation among populations based on nested analysis. Common economic aquatic products are important contributors to the human food supply. However, with the rapid globalization of the aquatic products industry, aquatic products market has become increasingly disordered. Therefore, an accurate and convenient method for identifying common economic aquatic products is important and necessary in many areas. DNA barcoding, which constitutes the analysis of a short fragment of the mitochondrial cytochrome c oxidase subunit I (COI) sequence, has been widely used in species identification. To discriminate common economic aquatic species using DNA barcoding, we collected 534 COI barcode sequences of 66 common species consisting of 39 fish, 9 crustaceans and 18 mollusks. The intraspecific genetic distances based on the Kimura 2-parameter (K2P) model were less than 1.37% for fish, 7.32% for crustaceans and 3.40% for mollusks, whereas the intragenus distances ranged from 3.91% to 13.82% for fish, 14.99%–16.17% for crustaceans and 14.82%–16.36% for mollusks. The average intraspecific K2P distance was also compared with the average intragenus distance. The taxonomic resolution ratio values obtained for fish, crustaceans and mollusks were 58.50, 21.59 and 27.63 respectively, which are higher than the threshold of (10×). A neighbor-joining (NJ) tree based on the K2P distance, and a maximum likelihood (ML) tree, based on the GTR + I + G model, were constructed, and all of the species could be identified unambiguously in the trees with several branches exhibiting high bootstrap values. Our results demonstrated high efficiency of DNA barcoding as an efficient molecular tool for the identification of common economic aquatic products, and 8 substitute species were successfully detected in 66 species. Our analyses also indicated that the common aquatic products trade industry could be effectively monitored and managed by DNA barcoding. Cicadidae periostracum (CP), the medicinal name of cicada exuviae, is well-known insect-derived traditional medicine with various pharmacological effects, e.g., anticonvulsive, anti-inflammatory, antitussive, and anticancer effects; it is also beneficial for the treatment of Parkinson's disease. For appropriate CP application, accurate species identification is essential. The Korean pharmacopoeia and the pharmacopoeia of the People's Republic of China define Cryptotympana atrata as the only authentic source of CP. Species identification of commercially distributed CP based on morphological features, however, is difficult because of the combined packaging of many cicada exuviae in markets, damage during distribution, and processing into powder form. DNA-based molecular markers are an excellent alternative to morphological detection. In this study, the mitochondrial cytochrome c oxidase subunit I sequences of C. atrata, Meimuna opalifera, Platypleura kaempferi, and Hyalessa maculaticollis were analyzed. On the basis of sequence alignments, we developed sequence-characterized amplified-region (SCAR) markers for efficient species identification. These markers successfully discriminated C. atrata from the three other cicada species, and detected the adulteration of market CP samples. This SCAR assay is a rapid, simple, cheap, reliable, and reproducible method for species identification, regardless of sample form and status, and contributes to CP quality control.
