In this study, we used response surface methodology (RSM) to optimize the extraction conditions for recovering lutein from paprika leaves using accelerated solvent extraction (ASE). The lutein content was quantitatively analyzed using a UPLC equipped with a BEH C18 column. A central composite design (CCD) was employed for experimental design to obtain the optimized combination of extraction temperature (°C), static time (min), and solvent (EtOH, %). The experimental data obtained from a twenty sample set were fitted to a second-order polynomial equation using multiple regression analysis. The adjusted coefficient of determination (R(2)) for the lutein extraction model was 0.9518, and the probability value (p=0.0000) demonstrated a high significance for the regression model. The optimum extraction conditions for lutein were temperature: 93.26°C, static time: 5 min, and solvent: 79.63% EtOH. Unlabelled: We aimed to identify the optimum conditions for the extraction of lutein from green tea using accelerated solvent extraction, and achieve improved analytical resolution and sensitivity between lutein and zeaxanthin using an ultra performance liquid chromatography (UPLC) system. The optimized method employed 80% ethanol as the extraction solvent, 160 °C as the temperature, 2 static cycles, and 5 min of static time. In the validation of the UPLC method, recovery was found to be in the range approximately 93.73 to 108.79%, with a correlation coefficient of 0.9974 and a relative standard deviation of <9.29% in inter- and intraday precision analyses. Finally, the lutein contents of green tea and green tea by-products were measured as 32.67 ± 0.70 and 18.18 ± 0.68 mg/100g dw, respectively. Furthermore, we verified that green tea by-products, which are discarded after producing green tea beverages, might be used as a great resource for massive lutein production., Practical Application: We have demonstrated that the common problem of inadequate resolution between lutein and zeaxanthin during carotenoid analyses can be overcome by optimizing the combined techniques of accelerated solvent extraction and ultra performance liquid chromatography (UPLC). Lithospermum erythrorhizon, which has traditionally been used as a vegetable and to make the liquor Jindo Hongju, contains several naphthoquinone pigments, including shikonin. This study aimed to evaluate the antiobesity effects of Lithospermum erythrorhizon ethanol extract (LE) and elucidate the underlying mechanism. C57BL/6J mice were fed a normal or high-fat diet with or without LE supplementation for 8 weeks. LE reduced high-fat diet-induced increases in body weight, white adipose tissue mass, serum triglyceride and total cholesterol levels, and hepatic lipid levels while decreasing lipogenic and adipogenic gene expression. Furthermore, acetylshikonin suppressed adipocyte differentiation in a dose-dependent manner and significantly attenuated adipogenic transcription factor expression in 3T3-L1 cells. These findings suggest that Lithospermum erythrorhizon prevents obesity by inhibiting adipogenesis through downregulation of genes involved in the adipogenesis pathway and may be a useful dietary supplement for the prevention of obesity.

Unlabelled: American ginseng (Panax quinquefolius L.) root health benefits include treatment of type 2 diabetes and this study evaluated the hypoglycemic effects of American red ginseng (ARG). ARG roots have increased bioactive phenolic contents, such as cinnamic acid and ferulic acid during the steaming process. The antihyperglycemic effects of methanol fraction extract of ARG, ferulic acid, and cinnamic acid were examined using a type 2 diabetic mouse model. The ARG treated group presented relatively lower blood glucose levels than the control group (P < 0.05). In addition, the glycogen and high density lipoprotein (HDL) contents were significantly increased while levels of plasma cholesterol and low density lipoprotein (LDL) concentration were significantly decreased in the ARG treated group. The groups treated with ferulic and cinnamic acids showed similar effects as those found in the ARG treated group. Thus, it is suggested that ARG roots, ferulic acid, and cinnamic acid have hypoglycemic effects in an animal model., Practical Application: This study was conducted to elucidate the hypoglycemic effects of American red ginseng (ARG) using a type 2 diabetic mouse model. Seventeen edible mushrooms commercially available in Korea were analysed for their umami taste compounds (5′-nucleotides: AMP, GMP, IMP, UMP, XMP; free amino acids: aspartic, glutamic acid) and subjected to human sensory evaluation and electronic tongue measurements. Amanita virgineoides featured the highest total 5′-nucleotide content (36.9 ± 1.50 mg/g), while monosodium glutamate-like components (42.4 ± 6.90 mg/g) were highest in Agaricus bisporus . The equivalent umami concentration ( EUC ) ranged from 1.51 ± 0.42 to 3890 ± 833 mg MSG/g dry weight; most mushrooms exhibited a high umami taste. Pleurotus ostreatus scored the highest in the human sensory evaluation, while Flammulina velutipes obtained the maximum score in the electronic tongue measurement. The EUC and the sensory score from the electronic tongue test were highly correlated, and also showed significant correlation with the human sensory evaluation score. These results suggest that the electronic tongue is suitable to determine the characteristic umami taste of mushrooms. [ABSTRACT FROM AUTHOR]

Four types of powder were prepared from freeze-dried fruiting bodies of the edible mushroom Hericium erinaceum by 2 different methods-conventional roll milling and the superfine grinding method of jet milling-at 3 different speeds, to compare their physicochemical properties and extraction of proteins and polysaccharides. Jet-milled powders prepared at 16,000 rpm (JMP1), 9000 rpm (JMP2), and 5000 rpm (JMP3), and roll-milled powder (RMP) had an average particle size of 5.5, 12.9, 25.2, and 243.6 μm, respectively. The reduction of particle size resulted in increased specific surface area, porosity, and bulk density. Among the 4 powders, JMP1 showed the highest bulk density (0.17 g/ml). The water solubility index of H. erinaceum powder increased from 35.2% in RMP to 53.1% in JMP1. Similarly, the swelling capacity of JMP1, JMP2, JMP3, and RMP was 11.28 ml/g, 7.36 ml/g, 6.26 ml/g, and 5.9 ml/g, respectively. The extractions of proteins and polysaccharides from H. erinaceum powders were also improved by the application of jet milling. These results suggest that the superfine grinding method of jet milling could provide more opportunities for mushroom powders to be incorporated in a wider variety of functional food applications.
