Whether a family history of diabetes (FHD) and exposure to perfluoroalkyl acids (PFAAs) are correlated with an increased risk of developing arthritis remains unclear. This cross-sectional study was conducted to explore the correlations between FHD or exposure to PFAAs and arthritis as well as their interaction using the National Health and Nutrition Examination Survey (NHANES). In total, 6,194 participants aged ≥ 20 years from the 2011–2018 NHANES were enrolled. PFAAs are a cluster of synthetic chemicals, including perfluorononanoic acid (PFNA), perfluorooctanoic acid (PFOA), perfluorooctane sulfonic acid (PFOS), perfluorodecanoic acid (PFDA) and perfluorohexane sulfonic acid (PFHxS). FHD was evaluated using self-reported questionnaires. Arthritis was classified into three types, rheumatoid arthritis (RA), osteoarthritis (OA), and others, which were diagnosed using questionnaires. Generalized linear models (GLMs) were used to test the correlation between FHD and arthritis. To examine the joint effects of PFAAs and FHD on arthritis, interaction terms were applied in the GLM. Arthritis incidence was 26.7% among all participants. FHD was associated with both RA [OR = 1.70 (95% CI: 1.15–2.50)] and other types of arthritis [OR = 1.62 (95% CI: 1.21–2.16)]. However, the relationship between FHD and OA was not significant after adjustment (P = 0.18). Interaction outcomes indicated that higher PFDA levels increased the association between FHD and arthritis. FHD is associated with an increased incidence of arthritis, which may be increased by PFDA. Perfluoroalkyl acids (PFAA) in gas and particles were analyzed in southeast coastal and mountainous cities, including Fuzhou, Xiamen, Zhangzhou and Nanping, to study the pollution characteristics, particle size distribution, phase partitioning and atmospheric transport. PFAA ranged from 7.8 to 290 pg m−3 in gaseous phase, 27 − 1200 pg m−3 in particulate phase, and perfluorobutanoic acid (PFBA), perfluorooctanoic acid (PFOA) were main compounds. PFAA had the highest concentration in Nanping with perfluorohexanoic acid (PFHxA) dominant, which could be related to the emission of PFAS from local industrial plants. Perfluorocarboxylic acids (PFCAs) and perfluoroalkyl sulfonic acids (PFSAs) exhibited different particle size distribution characteristics, with PFSAs preferring to distribute on coarse particles, which could be affected by the salt, minerals and organic matter in different particle sizes. The gas − particle partitioning coefficient (KPA) had a line relationship with the fluorinated carbon chain length of PFAA, suggesting that long-chain PFAA tended to exist in particulate phase. The Winter Monsoon could transport to the study area and drive atmospheric PFAS to southern cities. Perfluoroalkyl acids (PFAAs) are emerging pollutants that endangers food safety. Developing methods for the selective determination of trace PFAAs in complex samples remains challenging. Herein, an ionic liquid modified porous imprinted phenolic resin-dispersive filter extraction-liquid chromatography-tandem mass spectrometry (IL-PIPR−DFE−LC-MS/MS) method was developed for the determination of perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) in eggs. The new IL-PIPR adsorbent was prepared at room temperature, which avoids the disorder and instability of the template at high temperatures. The imprinting factor of IL-PIPR for PFOA and PFOS exceeded 7.3. DFE, combined with IL-PIPR (15 mg), was used to extract PFOA and PFOS from eggs within 15 min. The established method exhibits low limits of detection (0.01–0.02 ng/g) and high recoveries (84.7%–104.7%), which surpass those of previously reported methods. This work offers a new approach to explore advanced imprinted adsorbents for PFAAs, efficient sample pretreatment technique, and analytical method for pollutants in foods. To study the contamination levels of perfluoroalkyl acids (PFAAs) in Chinese eggs and the effects of persistent organic pollutant (POP) amendments to the Stockholm Convention blacklist, 3200 eggs from 10 major producing areas were collected from June 2013 to May 2017. Seventeen PFAAs in eggs were analysed. Perfluorooctane sulphonic acid (PFOS), perfluoropentanoic acid (PFPeA) and perfluorooctanoic acid (PFOA) were the main PFAAs in eggs. Perfluoroalkyl carboxylic acids (PFCAs) and short-chain PFAAs levels in eggs decreased after the amendment was implemented in China (p <.05), but no significant difference was observed in PFOS. This study assessed the occurrence, origins, and potential risks of emerging perfluoroalkyl acids (PFAAs) for the first time in drinking water resources of Khyber Pakhtunkhwa, Pakistan. In total, 13 perfluoroalkyl carboxylic acids (PFCAs) with carbon (C) chains C4-C18 and 4 perfluoroalkyl sulfonates (PFSAs) with C chains C4-C10 were tested in both surface and ground drinking water samples using a high-performance liquid chromatography system (HPLC) equipped with an Agilent 6460 Triple Quadrupole liquid chromatography–mass spectrometry (LC–MS) system. The concentrations of ∑PFCAs, ∑PFSAs, and ∑PFAAs in drinking water ranged from 1.46 to 72.85, 0.30–8.03, and 1.76–80.88 ng/L, respectively. Perfluorobutanoic acid (PFBA), perfluorohexanoic acid (PFHxA), and perfluoropentanoic acid (PFPeA) were the dominant analytes in surface water followed by ground water, while the concentration of perfluorobutane sulfonate (PFBS), perfluorooctanoic acid (PFOA), perfluoroheptanoic acid (PFHpA), perfluorooctane sulfonate (PFOS), perfluorohexane sulfonate (PFHxS), perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), perfluoroundecanoic acid (PFUnDA), and perfluorododecanoic acid (PFDoDA) were greater than long-chain PFOA and PFOS. The correlation statistics, which showed a strong correlation (p < 0.05) between the PFAA analytes, potentially indicated the fate of PFAAs in the area's drinking water sources, whereas the hierarchical cluster analysis (HCA) and principal component analysis (PCA) statistics identified industrial, domestic, agricultural, and commercial applications as potential point and non-point sources of PFAA contamination in the area. From risk perspectives, the overall PFAA toxicity in water resources was within the ecological health risk thresholds, where for the human population the hazard quotient (HQ) values of individual PFAAs were < 1, indicating no risk from the drinking water sources; however, the hazard index (HI) from the ∑PFAAs should not be underestimated, as it may significantly result in potential chronic toxicity to exposed adults, followed by children. Perfluoroalkyl acids (PFAAs) are environmentally persistent components of surfactants that consist of fully fluorinated carbon chains and a terminal sulfonate or carboxylate polar head moiety. Due to their unique amphiphilic properties, PFAAs are used in the manufacturing of products such as aqueous film forming foams (AFFF). There is cause for concern for PFAA contamination resulting from runoff and groundwater infiltration of AFFF that were used during fire training. This study analyzed water and sediment samples that were collected over a 13-month sampling period from bayous upstream and downstream of two former fire training areas located near Barksdale Air Force Base (BAFB); the occurrence and magnitude of PFAAs supported an aquatic ecological risk assessment of potential impacts of PFAAs at the site. Liquid chromatography coupled with mass spectrometry was used for determination of 6 PFAAs listed under the third Unregulated Contaminant Monitoring Rule (UCMR 3). Total PFAA concentrations in surface water and sediment samples ranged from 0 (ND) 7.1 ng/mL and 0 (ND)31.4 ng/g, respectively. Perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) were the predominant PFAAs detected. In general, perfluorosulfonates were quantified more frequently and at higher concentrations than perfluorocarboxylates. The perfluoroalkyl chain length of PFAAs also showed significant influence on PFAA concentrations when analyzed by Spearman's rank correlation analysis. Some contamination we observed in surface water and sediment samples from reference locations could be a result of local runoff from the use of commercial products containing per- and poly-fluoroalkyl substances (PFAS), but AFFF appears to be the primary source given the close proximity of the historical fire training areas. Per‐ and polyfluoroalkyl substances (PFAS) are organic pollutants that may have adverse effects on the ecosystem. Despite the global presence of PFAS, knowledge of PFAS on the African continent is limited because monitoring of PFAS is challenging and often not feasible owing to the lack of analytical capacity and high cost. However, it is necessary to understand the environmental risks posed by these chemicals in developing countries, because increasing urbanization will likely increase PFAS contamination in the environment. Although, as far as is known, PFAS concentrations in the African aquatic environment are generally lower than in more developed countries, exceedances of ecological quality standards (EQS) were reported in a few cases, providing evidence of potential ecological risks to these ecosystems. However, the number of ecosystems at risk will likely increase as urbanization and modernization increase in African countries. Therefore, environmental regulations should be updated and implemented to reduce further contamination of the aquatic environment with these chemicals. In addition, analytical laboratories in Africa should develop their capacity to detect PFAS and related compounds regularly and routinely. Local hot spots need to be identified, the influence of these hot spots on the PFAS burden in the environment should be investigated, and environmental regulations should be implemented for these hot spots to reduce their environmental impact. Therefore, we recommend a more routine monitoring of PFAS, including new PFAS that are currently used as perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) alternatives, which are not regulated and environmentally concerning. Data for 639 US children aged 3–11 years who participated in the National Health and Nutrition Examination Survey during 2013–2014 were analyzed by fitting regression models with log10-transformed values of blood lead and methyl and total mercury as dependent variables and log10-transformed values of perfluoroalkyl acids (PFAA) as one of the independent variables. PFAAs considered were 2-(N-methyl-perfluorooctane sulfonamido) acetic acid (MPAH), linear isomer of perfluorooctanoic acid (NPFOA), perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), perfluorohexane sulfonic acid (PFHxS), linear isomer of perfluorooctane sulfonic acid (NPFOS), and monomethyl branch isomer of perfluorooctane sulfonic acid (MPFOS). Adjusted regression slopes (β) indicating associations between the concentrations of PFAAs with blood lead and mercury were estimated. Statistically significant associations between concentrations of each PFAA and blood lead were observed. For 10% increases in concentrations of MPAH, NPFOA, PFNA, PFDA, PFHxS, NPFOS, and MPFOS, percent increases in the concentrations of blood lead were 0.45%, 1.59%, 0.78%, 0.32%, 0.65%, 1.32%, and 0.89% respectively. For 10% increases in concentrations of MPAH, PFNA, PFDA, and NPFOS, percent increases in the concentrations of total mercury in the blood were 1.62%, 1.44%, and 3.24% respectively. For 10% increases in concentrations of PFDA and NPFOS, percent increases in the concentrations of methyl mercury in the blood were 2.07% and 4.57% respectively. While concentrations of each of the seven PFAAs were positively associated with the concentrations of blood lead, concentrations of only PFDA and NPFOS were positively associated with increases in total and methyl mercury. PFAAs having positive associations with lead and mercury imply co-exposure and/or co-existence of high concentrations of PFAAs and lead/mercury. Since PFAAs as well as lead/mercury are known to be neurotoxic, nephrotoxic, and endocrine disruptors, their co-existence/co-exposure may lead to neurodevelopmental deficits that are additive/synergistic than neurodevelopmental deficits associated with exposures to PFAAs and lead/mercury alone. Data (N = 2552) from National Health and Nutrition Examination Survey for US adults aged ≥ 20 years for 2011–2016 were analyzed to estimate the associations between the concentrations of blood cadmium, lead, and total mercury and the concentrations of seven perfluoroalkyl acids (PFAA), namely, 2-(N-Methyl-perfluorooctane sulfonamido) acetic acid (MPAH), perfluorooctanoic acid (PFOA), perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), perfluoroundecanoic acid (PFUnDA), perfluorohexane sulfonic acid (PFHxS), and perfluorooctane sulfonic acid (PFOS). Concentrations of blood cadmium were negatively associated with the concentrations of PFHxS (β = − 0.05428, p < 0.01) and PFOS (β = − 0.0212, p = 0.02). Concentrations of blood lead were positively associated with the concentrations of MPAH (β = 0.03301, p < 0.01), PFOA (β = 0.04783, p = 0.01), PFNA (β = 0.11761, p < 0.01), PFDA (β = 0.08007, p < 0.01), PFUA (β = 0.11382, p < 0.01), and PFOS (β = 0.04996, p = 0.02). Percent increases in the concentration of blood lead were 0.32%, 0.46%, 1.13%, 0.77%, 1.09%, and 0.48% for 10% increases in the concentrations of MPAH, PFOA, PFNA, PFDA, PFUA, and PFOS, respectively. Concentrations of blood total mercury were positively associated with the concentrations of PFNA (β = 0.37105, p < 0.01), PFDA (β = 0.46875, p < 0.01), PFUA (β = 0.56934, p < 0.01), and PFOS (β = 0.17557, p < 0.01). Percent increases in the concentration of blood total mercury were 3.6%, 4.57%, 5.58%, and 1.69% for 10% increases in the concentrations of PFNA, PFDA, PFUA, and PFOS, respectively. Associations between the concentrations of PFAAs with blood total mercury were substantially stronger than the concentrations with blood lead. Perfluoroalkyl substances (PFAS) are surface‐active agents used in diverse industrial and commercial applications. They contaminate both freshwater and marine ecosystems, are highly persistent, and accumulate through trophic transfer. Seabirds are exposed to environmental contaminants due to their high trophic position in food webs and relatively long lifespan. We measured levels of 10 perfluoroalkyl acids (PFAAs) in egg yolks of yellow‐legged gulls (Larus michahellis) breeding in the northern Adriatic Sea (Northeast Italy). We examined variations in PFAAs within clutches (between eggs of different laying order) and among clutches. Perfluorooctane sulfonate (PFOS) was the most abundant yolk PFAA (mean = 42.0 ng/g wet wt), followed by perfluorooctanoic acid (PFOA; 3.8 ng/g wet wt) and perfluorododecanoic acid (PFDoDa; 2.8 ng/g wet wt). The ∑PFAAs averaged 57.4 ng/g wet weight, ranging between 26.5 and 115.0 ng/g wet weight. The PFAA levels varied substantially among clutches (0.29–0.79 of the total variation), whereas the effects of laying order were considerably weaker (0.01–0.13). Egg‐laying order effects were detected for ∑PFAAs, PFOS, perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), perfluoroundecanoic acid (PFUnA), and PFDoDa, whereby the last‐laid eggs exhibited lower PFAA concentrations than early‐laid eggs. Our results indicate that seagulls from the northern Adriatic basin deposit measurable amounts of PFAAs in their eggs. The large among‐clutches differences in PFAAs suggest that exposure of yellow‐legged gull females to these compounds is highly variable. Associations between selected perfluoroalkyl acids (PFAAs) and biomarkers of renal function were evaluated for US adult aged ≥ 20 years (N = 8220) in the National Health and Nutrition Examination Survey for 2005–2014. Glomerular filtration (GF) stage-stratified regression models were classified by estimated glomerular filtration rate (eGFR) with GF-1 (eGFR > 90 mL/min/1.73 m2), GF-2 (eGFR 60–89 mL/min/1.73 m2), GF-3A (45–59 mL/min/1.73 m2), and GF-3B/4 (15–44 mL/min/1.73 m2). For GF-1, PFOA, PFOS, and PFHxS were positively and significantly associated with serum creatinine. Serum albumin levels were positively associated with the PFAA considered at all stages and most associations were significant. Further, PFAS serum concentration associations to serum albumin were about 2–3 times stronger at GF-3B/4 than at GF-1. In contrast, urine albumin was negatively and significantly associated with PFOA and PFHxS serum concentrations at all stages of renal function, while, PFOS and PFNA were negatively and significantly associated to urine albumin at GF-3A and GF-3B/4. Urine albumin/creatinine ratios were negatively and significantly associated with PFOA, PFOS, and, and PFHxS serum concentrations at all stages of renal function, as well as with PFNA and PFDA at GF-3A and GF-3B/4. Recent work revealed that serum PFAAs have an inverted U-shaped association to the calculated stages of renal failure based on eGFR; this work adds that albuminuria makes additional negative contributions to already existing negative associations of PFAA to eGFR in advancing stages of renal failure. We hypothesize that both progressive renal failure per se and especially renal failure with albuminuria cause the kidneys to reabsorb less and to excrete more of the PFAAs studied. We suspect this finding may generalize to some other perfluoroalkyl substances (PFAS). Abstract Perfluoroalkyl acids (PFAAs), as a group of industrial chemicals, are characterized by persistence, long-distance transmission, bioaccumulation and toxicity, and have been recognized as persistent organic pollutants. However, PFAAs and their related products have been used daily and in industrially products over the past several decades, which resulting in ubiquitous presence in various environmental medias, biota and even in human body. Numerous studies have investigated the neurobehavioral deficit and molecular mechanism underlying those effects of PFAAs in the last decades. In the present review, we summarized the neurotoxic effects of some PFAAs, especially perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA). The potential molecular mechanism for PFAAs-induced neurotoxicity are mainly described in the following aspects: calcium homeostasis and its related signal pathway, synaptogenesis and synaptic plasticity, neurotransmitters, as well as the neural cells apoptosis. We analyzed paired serum, urine, and hair samples from 94 Korean children and adults to investigate levels of 11 perfluoroalkyl acids (PFAAs). The effects of demographic factors and dietary habits on PFAA exposure were also assessed based on the paired samples. The total PFAA concentrations were 2.4–31 ng/mL in serum, not detected–9.5 ng/mL in urine, and 0.48–15 ng/g in hair. Levels of perfluoropentanoic acid (PFPeA) and perfluorohexanoic acid (PFHxA), which have short carbon chains, were 1.5–5 fold higher in urine and hair than in serum. The PFAA concentrations in serum exhibited a decreasing trend with age from young childhood to adolescence, followed by an increasing trend after adolescence. For most PFAA species, concentrations in serum were higher in adult males than in adult females (p < 0.01). No sex difference was evident in the urine and hair samples. In addition, there was no age difference in the urine samples, but in the hair samples, we observed higher concentrations of PFAAs in children than in the other age groups (p < 0.01). The consumption rates of fish and water showed significant correlations with serum (positive correlation) and hair (negative) concentrations, respectively. No relationships between serum and hair/urine levels for most PFAAs were observed, except between serum and hair levels for perfluorooctanoic acid (PFOA). In this study, a molecular modeling approach was used to identify the most effective natural hydrophobic deep eutectic solvents (HDESs) for extracting the emerging pollutants, perfluorooctanoic acid (PFOA) and perfluorooctanesulfonic acid (PFOS) from synthetic wastewater. The use of HDESs for this purpose has not been reported in the literature, making this a pioneering contribution to the field. First, an extensive literature search was conducted to generate a list of 74 non-ionic HDES constituents. In the second step, a systematic screening of natural components was conducted to identify 55 new compounds that can be used for the synthesis of HDESs. The Conductor-like Screening Model for Real Solvents (COSMO-RS) was then used to screen the 129 potential HDES constituents based on three key performance indicators: distribution ratio, selectivity, and performance index at infinite dilution compared to the predicted performance of toluene as a benchmark, as the use of molecular solvents or DESs for this purpose has never been reported in the literature. The top 20 HDES constituents with the highest extraction performance were predominantly from the terpene and terpenoid families. The median lethal dose was also used to analyze toxicity. A multicriteria evaluation approach was adopted to evaluate several properties important to liquid–liquid extraction (LLE) processes, including the extraction performance, density, vapor pressure, water solubility, and leachability of DESs. Three HDESs, namely, trioctylphosphine oxide: 2-methylbenzothiazole (1:9), trioctylphosphine oxide: menthyl acetate (1:4), and 1,8-cineole: menthyl acetate (2.5:1), showed promising results. This study provides molecular insights on extracting PFOA/PFOS from wastewater using HDESs and emphasizes the need to evaluate LLE units using multicriteria. In addition, the results may serve as a starting point for further research, including the synthesis and characterization of novel HDES constituents and the development of optimized HDESs for the extraction of PFOA/PFOS from wastewater. This work demonstrates the extraction potential of carboxylated carbon nanospheres (CNSs-COOH) for solid-phase extraction of perfluoroalkyl acids (PFAAs) for the first time. Six PFAAs, including perfluorohexanesulfonate (PFHxS), perfluoroheptanoic acid (PFHpA), perfluorooctanoic acid (PFOA), perfluorooctanoic sulfonic (PFOS), perfluorononanoic acid (PFNA) and perfluorodecanoic acid (PFDA), were quantitatively adsorbed on a CNSs-COOH-packed cartridge; then, the PFAAs retained on the adsorbent were quantitatively eluted with an optimized volume of acetone (pH = 10). Finally, the desorbed PFAAs were determined by liquid chromatography–tandem mass spectrometry. Important relevant factors, such as the eluant and its volume, sample pH, amount of CNSs-COOH, sample flow rate and sample volume were optimized in detail. Under the optimized conditions, low limits of detection (0.01–1.2 ng L −1 ), wide linear range (0.50–200 ng L −1 ) and good repeatability (2.4–5.1%) and good reproducibility (3.7–8.8%) were obtained. The proposed method was applied to analyze the six PFAAs in real environmental water samples, and satisfactory results were achieved. All of these results showed that CNSs-COOH will be a good choice for the pre-concentration and analysis of target PFAAs at trace levels in environmental water samples in the future. Isomer-specific analysis of perfluoroalkyl acids (PFAAs) is important to accurately assess their environmental source, fate, and human risks. In this study, a method was developed for the determination of perfluorooctanoic acid (PFOA), perfluorooctane sulfonate (PFOS), and perfluorohexane sulfonate (PFHxS) isomers in biosolids, biosolids-amended soils and plants using ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC–MS/MS). The separation efficiencies of two chromatographic columns and extraction capacities of different methods were tested. Compared with the C18 column (ACQUITY UPLC BEH Shield RP18 column), the column with an alkyl perfluorinated C8 stationary phase (Epic FO LB column), in combination with the distinct MS/MS transitions of analytes, allowed better separation of most isomers. The ion-pair extraction method showed more effective matrix separation than that of the alkaline digestion method, with recoveries ranging from 79.6-105% for biosolids, 80.4-116% for soils, and 68.0-114% for plant tissues. The method detection limits ranged from 10 to 55, 3–13, and 8–58 pg/g dry weight for biosolids, soil, and plants, respectively. This method was applied successfully to quantify individual isomers in biosolids, biosolids-amended soils and plants. The Brick Township Municipal Utilities Authority (BTMUA), which relies on the Metedeconk River as its primary source of water supply, initiated a perfluoroalkyl acid (PFAA) source trackdown study in collaboration with the New Jersey Department of Environmental Protection (NJDEP) after discovering that the concentration of one PFAA, perfluorooctanoic acid (PFOA), was elevated at their raw surface water intake. Water samples were collected over eight sampling events between September 2011 and July 2014. Samples included surface water, groundwater, stormwater, sanitary sewer water, and commercial/industrial process water. Each sample was analyzed for ten PFAAs. Results from a set of samples collected from the 80 km South Branch Metedeconk River watershed directed the focus of this study to a 7.5-km area of interest. Within this area, a high concentration of PFAA contamination was documented in a localized zone. Subsequent groundwater sampling led to the identification of a plume of groundwater contamination emanating from an industrial/business park. The suspected source of PFAA detected in the river and drinking water intake was identified to a small industrial facility that used materials containing PFOA. Groundwater PFOA concentrations as high as 70,000 ng/L were found in samples taken within 200 m of the parcel and surface water concentrations as high as 130 ng/L were observed in the river. While various PFAAs were detected in the samples, particularly in groundwater samples, PFOA was identified as the primary contaminant of concern with respect to the river and the BTMUA water supply. The occurrence of 17 target PFAA analytes was determined in surface sediments ( n = 37) of the East China Sea and potential influencing factors were examined. ΣPFAAs ranged from 0.41 ng/g dw to 3.06 ng/g dw, with perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) as the most abundant perfluorocarboxylic acid and perfluoroalkyl sulfonate, respectively. PFAAs in the sediments were strongly influenced by terrigenous input. Analysis of the relationship between dynamic influence factors and PFAA concentrations showed that the characteristics of PFAA distribution were rather complex. ΣPFAA concentrations and TOC were positively correlated ( p < 0.0001). Circumfluence also influenced the whole PFAA distribution and seasonal variation. In addition, correlation analysis suggested that log K oc values increased with increasing perfluoroalkyl chain length. Bench- and pilot-scale sorption tests were used to probe the performance of several biochars at removing perfluoroalkyl acids (PFAA) from field waters, compared to granular activated carbon (GAC). Screening tests using organic matter-free water resulted in hardwood (HWC) ( K d = 41 L g −1 ) and pinewood (PWC) ( K d = 49 L g −1 ) biochars having the highest perfluorooctanoic acid (PFOA) removal performance that was comparable to bituminous coal GAC ( K d = 41 L g −1 ). PWC and HWC had a stronger affinity for PFOA sorbed in Lake Mead surface water (K F = 11 mg (1−n) L n g −1 ) containing a lower (2 mg L −1 ) dissolved organic carbon (DOC) concentration than in a tertiary-filtered wastewater (K F = 8 mg (1−n) L n g −1 ) with DOC of 4.9 mg L −1 . A pilot-scale study was performed using three parallel adsorbers (GAC, anthracite, and HWC biochar) treating the same tertiary-filtered wastewater. Compared to HWC, and anthracite, GAC was the most effective in mitigating perfluoropentanoic acid (PFPnA), perfluorohexanoic acid (PHxA), PFOA, perfluorooctane sulfonic acid (PFOS), and DOC (45–67% removed at 4354 bed volumes) followed by HWC, and then anthracite. Based on bench- and pilot-scale results, shorter-chain PFAA [perfluorobutanoic acid (PFBA), PFPnA, or PFHxA] were more difficult to remove with both biochar and GAC than the longer-chain, PFOS and PFOA. Perfluoroalkyl acid (PFAA) pollution of freshwaters has attracted a great deal of attention and poses a challenge to the restoration of river ecosystems. However, detailed studies on the impact of PFAAs and their alternatives on microbial plankton communities are lacking, and few studies have examined the adaptability of those communities to PFAAs. In this study, water samples were obtained from water bodies in the vicinity of the largest perfluorinated industrial park in Asia for comparative analysis. The spatial distribution of 19 PFAAs and 4 alternatives as well as their effect on microbial community structure were investigated through field sampling. The maximum total concentration of perfluoroalkyl carboxylic acids (PFCAs) at an industrial wastewater treatment plant was 2423.48 ng/L. The primary components included perfluorooctanoic acid (PFOA, 43.15%, 1045.82 ng/L), perfluorohexanoic acid (PFHxA, 27.10%, 656.70 ng/L) and perfluorobutanoic acid (PFBA, 20.13%, 487.85 ng/L). The maximum total concentration of PFAA alternatives was 48.59 ng/L, with hexafluoropropylene oxide dimer acid (HFPO-DA) accounting for 97.43%. The microbial community was mainly composed of Proteobacteria , Actinobacteriota , Bacteroidota , and Cyanobacteria. While Verrucomicrobiota tolerated PFAAs, significant effects on microbial community structure were determined for PFCAs and PFSAs, albeit with clear differences. Short-chain PFAAs had a similar impact on the microbial community as the restricted long-chain PFAAs. Thus, our study demonstrated the potentially negative impact of PFAAs on the microbial plankton community in the natural environment and the combined effect of poor water quality. The result provides new directions for investigations of PFAA pollution in freshwater environments. Perfluoroalkyl acids (PFAAs), a subset of per- and poly-fluoroalkyl substances (PFAS), are environmentally stable, mobile and bioaccumulative compounds. This leads to high concentrations in wildlife species essential to the cultural identity and subsistence of Arctic populations. Our objective was to characterize the distribution and exposure determinants of PFAAs among Nunavik Inuit adults. The study included up to 1322 Nunavik residents aged 16–80 years who participated in the Qanuilirpitaa? 2017 Nunavik Inuit Health Survey (Q2017). Plasma concentrations were compared to those the general Canadian population using data from the Canadian Health Measures Survey Cycle 5 (2016–2017). Associations between plasma concentrations of nine PFAAs, determined by liquid chromatography-tandem mass spectrometry, and sociodemographic factors and traditional activity participation were examined using multiple linear regression models. Overall exposure to PFAAs was twice as high compared to the general Canadian population and less regulated perfluorononanoic acid (PFNA) and perfluoroundecanoic acid (PFUnDA) concentrations were 7-fold higher, and perfluorodecanoic acid (PFDA) concentrations were 4-fold higher. Males had higher concentrations of perfluorooctanoic acid (PFOA) and perfluorohexane sulfonate (PFHxS), whereas females had higher concentrations of PFDA and PFUnDA. PFAAs concentrations increased with age and were highest among those aged 60+ years. PFNA and PFOA concentrations followed a J-shaped pattern: those aged 16–29 years had higher concentrations than those aged 20–29 and 30–39 years. Ungava Bay generally had lower concentrations of all PFAAs congeners compared to Hudson Bay and Hudson Strait, with the exception of PFNA, which tended to have the lowest concentration in Hudson Strait. PFAAs concentrations were highly associated with hunting activity, omega-3 polyunsaturated fatty acids, and drinking water from environmental sources. The results highlight the importance of characterizing PFAAs exposure sources in Arctic communities and provide further evidence for the long-range transport of long-chain PFAAs and their precursors that necessitate international action. Evidence on the associations of perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) with hypertension or blood pressure (BP) levels was limited and inconsistent. The present prospective study aims to evaluate the longitudinal associations of serum levels of PFOA and PFOS with incident hypertension risk and change of blood pressure levels. At baseline 1080 participants (mean age 62 years, 58.9% females) free of hypertension, cardiovascular disease, diabetes, and cancer were followed up for nearly 5 years. Baseline serum levels of PFOA and PFOS were measured with ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS). Hypertension was defined as any of (1) self-reported physician-diagnosed hypertension (2) use of hypotension drugs (3) measured systolic BP ≥ 140 mmHg or diastolic BP ≥ 90 mmHg. Change of BP was evaluated as a difference between twice measurements (BP at follow-up visit－BP at baseline). After adjustment for multiple covariates, serum PFOS levels were negatively correlated with risk of hypertension [RR per lg-unit = 0.94 (95% CI: 0.88, 0.99)] and change of systolic BP [β = −1.48 (95% CI: −2.56, −0.41)]. The highest vs lowest quartiles of PFOS concentration was negatively associated with hypertension risk. Compared with Q1, the RRs (95% CIs) for Q2, Q3, and Q4 were 0.83 (0.67–0.98), 0.81 (0.67–0.97), and 0.81(0.67–0.97), respectively (p for trend = 0.016). The negative associations remained in females but not in males (p for interaction = 0.44). No significant association of PFOA with hypertension risk was observed. Background In some cross-sectional epidemiologic studies the shape of the association between serum concentrations of perfluoroalkyl acids (PFAAs) and lipids suggests departures from linearity. Objectives We used statistical approaches allowing for non-linearity to determine associations of prenatal exposures of perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) with lipid concentrations. Methods PFAAs were measured in serum from pregnant women collected in 1991–1992 at enrollment in the Avon Longitudinal Study of Parents and Children and lipids in serum from their daughters at ages 7 (n = 111) and 15 (n = 88). The associations of PFAAs with lipids were first explored by cubic splines, followed by piecewise linear regressions by tertiles to obtain regression coefficients (β) and their 95% confidence limits (95% CL) (in mg/dL per 1 ng/mL). Results At age 7, total cholesterol was positively associated with prenatal PFOA concentrations in the lower tertile (β = 15.01; 95% CL = 2.34, 27.69) but not with PFOA concentrations in the middle (β = − 3.63; 95% CL = − 17.43, 10.16) and upper (β = − 1.58; 95% CL = − 4.58, 1.42) tertiles. At age 15, a similar pattern was noted as well. Positive associations between LDL-C and prenatal PFOA concentration in the lower tertile were observed in daughters at ages 7 (β = 14.91; 95% CL = 3.53, 28.12) and 15 (β = 13.93; 95% CL = 0.60, 27.26). LDL-C was not associated with PFOA concentrations in the middle or upper tertile at any age. Neither HDL-C nor triglycerides was associated with prenatal PFOA exposure. Non-linear patterns of association of total cholesterol and LDL-C with prenatal PFOS were less consistently noted. Conclusion Exposure to low levels of PFOA during prenatal development may alter lipid metabolism later in life. After global commercialization of short-chain perfluoroalkyl acids (PFAAs) as substitutes to conventional long-chain PFAAs by the major manufacturers, two fluorine industry parks for production of short-chain PFAAs located in the Daling River Basin of northern China have developed rapidly in the last few years. This study provides a systematic assessment of sources, emissions, transportation, and potential risks of the PFAAs in this area. The C4 perfluorobutane sulfonic acid (PFBS) and perfluorobutanoic acid (PFBA) were the predominant short-chain PFAAs in river water, with maximum concentrations of 2.90 and 1.35 μg/L, respectively. Park 1 equipped with a telomerization process was identified to be the source of linear and branched mixtures of PFBS, PFBA, and perfluorooctanoic acid (PFOA), while park 2 with an electrochemical fluorination process (ECF) was identified to be the source of linear and branched mixtures of PFBS and PFOA. Partition coefficients between water and sediment were consistent for C4-C8 perfluoroalkyl carboxylic acids (PFCAs) but directly proportional to C9-C11 PFCAs and perfluoroalkyl sulfonic acids (PFSAs). This is the first nationwide study of perfluoroalkyl acids (PFAAs) in environmental waters in Vietnam. Twenty-eight river water and 22 groundwater samples collected in four major cities and 14 river water samples from the Red River were screened to investigate the occurrence and sources of 16 PFAAs. Perfluorooctane sulfonic acid (PFOS), perfluorooctanoic acid (PFOA), and perfluorononanoic acid (PFNA) were the most prevalent of 11 detected PFAAs with maximum concentrations in urban river water of 5.3, 18 and 0.93 ng L −1 , respectively, and in groundwater of 8.2, 4.5 and 0.45 ng L −1 , respectively. PFAAs in the Red River water were detected at low levels. PFAA concentrations in river water were higher in the rainy season than in the dry season, possibly due to storm water runoff, a common phenomenon in Southeast Asian countries. The highest concentrations of PFAAs in river water were observed in samples from highly populated and industrialized areas, perhaps sourced from sewage. The PFAA concentrations observed were similar to those in other Southeast Asian countries, but lower than in developed nations. From the composition profiles of PFAAs, industrial products containing PFAAs imported from China and Japan might be one of the major sources of PFAAs in the Vietnamese aquatic environment. According to the health-based values and advisory issued by the United States Environmental Protection Agency (USEPA), the concentrations of detected PFAAs in this study do not pose an immediate health risk to humans and aquatic organisms. Perfluoroalkyl acids (PFAAs) have been used in a variety of products for many years and have been detected worldwide in human serum. Previous studies have suggested the potential effects of PFAAs on serum lipids. To investigate the associations between serum concentrations of PFAAs and serum lipid levels, 133 participants were randomly selected from the people coming for health check-up in Yuanyang Red Cross Hospital of Henan, China. Linear regression analysis revealed that perfluoro-octanoic acid (PFOA), perfluorononanoic acid (PFNA), and perfluorodecanoic acid (PFDA), with a median concentration of 1.43, 0.37, and 0.19ng/mL, respectively, were positively associated with total cholesterol (TC). Those in the highest quartile of PFOA exposure had ln-TC levels 0.24mmol/L higher than those in the lowest quartile. For PFNA and PFDA, effect estimates were 0.25 and 0.16mmol/L, respectively. A positive association between high-density lipoprotein cholesterol (HDLC) and PFDA was found, and there was a 0.18mmol/L increase of HDLC for the top PFDA quartile compared with the lowest quartile. PFOA and PFNA were positively associated with low-density lipoprotein cholesterol (LDLC). Ln-LDLC levels of people in both top PFOA and PFNA quartiles were 0.33mmol/L higher than those in the lowest quartiles. Logistic regression analysis indicated that increased PFOA and PFOS quartiles were positively associated with an increased risk of abnormal TC and LDLC when controlling for no confounding factors. Abstract: Perfluoroalkyl acids (PFAAs) are persistent organic pollutants that are detected in humans worldwide. Laboratory animal studies have shown that PFAAs are associated with immunotoxic effects. However, epidemiological studies investigating the role of PFAAs, in particular PFAAs with longer chains than perfluorooctanoic acid, are scarce. We investigated associations between prenatal exposure to PFAAs, including long-chain compounds, and infant allergic diseases at 12 and 24months in a large study population. The participants included mothers and their infants who enrolled in the Hokkaido Study on Environment and Children's Health 2003–2009. Eleven PFAAs were measured in maternal plasma taken at 28–32weeks of gestation using ultra-performance liquid chromatography coupled to triple quadrupole tandem mass spectrometry. Characteristics of participants and information on infant allergic diseases were obtained from self-administered questionnaires and medical records. At 24months, the adjusted odds ratio (OR) (first vs. fourth quartiles) for eczema in association with higher maternal perfluorotridecanoic acid (PFTrDA) levels was 0.62 (95% confidence interval (CI) 0.45, 0.86). After stratification by gender, the adjusted ORs in female infants from mothers with higher maternal perfluoroundecanoic acid (PFUnDA) and PFTrDA levels were also statistically significant (PFUnDA: OR=0.50; 95% CI, 0.30, 0.81; PFTrDA: OR=0.39; 95% CI, 0.23, 0.64). Abstract: Perfluoroalkyl acids (PFAAs) are found globally in the environment, detected in humans and wildlife, and are typically present as mixtures of PFAA congeners. Mechanistic studies have found that responses to PFAAs are mediated in part by PPARα. Our previous studies showed that individual PFAAs activate PPARα transfected into COS-1 cells. The goal of the current study was to determine if binary combinations of perfluorooctanoic acid (PFOA) and another PFAA act in an additive fashion to activate PPARα in the mouse one-hybrid in vitro model. COS-1 cells were transiently transfected with mouse PPARα luciferase reporter construct and exposed to either vehicle control (0.1% DMSO or water), PPARα agonist (WY14643, 10μM), PFOA at 1–128μM, perfluorononanoic acid (PFNA) at 1–128μM, perfluorohexanoic acid (PFHxA) at 8–1024μM, perfluorooctane sulfonate (PFOS) at 4–384μM or perfluorohexane sulfonate (PFHxS) at 8–2048μM to generate sigmoidal concentration–response curves. In addition, cells were exposed to binary combinations of PFOA + either PFNA, PFHxA, PFOS or PFHxS in an 8×8 factorial design. The concentration–response data for individual chemicals were fit to sigmoidal curves and analyzed with nonlinear regression to generate EC50s and Hillslopes, which were used in response-addition and concentration–addition models to calculate predicted responses for mixtures in the same plate. All PFOA+PFAA combinations produced concentration–response curves that were closely aligned with the predicted curves for both response addition and concentration addition at low concentrations. However, at higher concentrations of all chemicals, the observed response curves deviated from the predicted models of additivity. Per- and polyfluoroalkyl substances (PFAS) have received continuous attention; however, there is limited understanding of their sources in the atmosphere and related human exposure risks. This study measured PFAS in the atmospheric total suspended particles collected from Karachi, Pakistan, during the winter. Among the quantified PFAS, perfluorobutanoic acid (PFBA) showed the highest average concentration (3.11 ± 2.64 pg/m3), accounting for 32% of the total PFAS. Wind speed was positively correlated with perfluorohexanoic acid (PFHxA) and N- ethyl perfluorooctanesulfonamide (N -EtFOSA), while relative humidity was negatively correlated with perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA). Weighted potential source contribution function (WPSCF) and concentration weighted trajectory (WCWT) analyses suggested that northwestern Pakistan and western Afghanistan areas were highly associated with the long-range atmospheric transport of PFAS. We also calculated the daily intake of PFAS via inhalation, which were in the range of 0.07–3.98 and 0.01–0.33 pg/kg bw/d for children and adults, respectively. The calculated hazard quotient (HQ) of PFOS and PFOA was significantly lower than 1, indicating less or unlikely to cause non-carcinogenic effect via inhalation exposure. Overall, this study contributes to the understanding of geographic origins and human inhalation risks of airborne PFAS on a regional scale. Perfluoroalkyl acids (PFAAs) have been ubiquitously detected in water bodies and are a cause of great public concern due to their adverse effects. This study investigated the long-term temporal-spatial trends of PFAAs in the water bodies of the entire Taihu Lake, and predicted PFAA concentrations for 2024. A field investigation conducted in 2021 and previous data allowed to derive trends over a broad temporal-spatial scale, which is often not feasible in short-term studies. In the 2009–2021 period, the most quantifiable PFAAs increased, among which perfluorooctanoic acid and perfluorohexanoic acid were predominant. As of 2021, the mean total concentration of ten PFAAs (∑ 10 PFAA) showed a distinct spatial decreasing trend, moving from north to south within the lake, and similar spatial distribution patterns were also noted in other years. The main PFAA input and most serious contamination were concentrated in the northern region, due to the riverine inputs and clustering of PFAA-related industries. The ∑ 10 PFAA concentration in the wet season was greater and presented a more uniform distribution pattern than that in the dry season, possibly due to the combined effects of the degradation of PFAA precursors, water inflow, rainfall, shipping activities, and a shallow water column. From 2009 to 2021 the ∑ 10 PFAA concentration of the entire lake showed an increasing trend, but the rate of increase was significantly reduced. In addition, a grey model predicted that the mean ∑ 10 PFAA concentration in the entire Taihu Lake will reach 431 ng/L in 2024, and the northern region will be affected by a more serious PFAA pollution in the future because it exhibited a high mean ∑ 10 PFAA concentration of 426 ng/L in 2021. These findings provide novel insights into the temporal-spatial distribution of PFAAs in Taihu Lake, and could help regulators to formulate policy decisions in response to PFAA pollution. Concerns have been raised about per- and polyfluoroalkyl substances (PFAS) in sewage sludge given the urgent need of finding suitable disposal methods for sludge. In this study, we evaluated the effect of ultrasonication on PFAS changes in sewage sludge. It was revealed that although ultrasonication at 20 kHz increased the soluble chemical oxygen demand (SCOD) of treated sewage sludge, this technique was ineffective for degrading perfluoroalkyl acids (PFAAs) and their precursors. Ultrasonic treatment for longer time (>15 min) led to concentration increase of perfluorooctanoic acid (PFOA), perfluoroheptanoic acid (PFHpA), and perfluorohexanoic acid (PFHxA) in the liquid phase, possibly due to their release from disrupted sludge flocs during cavitation. Adding permanganate (10 mM) to the ultrasonic system could also enhance the disruption of sludge particles, resulting in higher concentrations of PFOA and PFHxA in the solid phase and PFOA, PFHpA, PFHxA, and perfluorobutanesulfonic acid (PFBS) in the liquid phase. Overall, ultrasonic pretreatment at 20 kHz and 0.7 W/mL is unlikely to remove PFAS from sewage sludge. Instead, it could increase the risk of PFAS pollution upon final sludge disposal. Effective treatment technologies are thus demanded if PFAS in sludge are regulated. Abstract: This study investigates the influence of biological and environmental factors on the concentrations of perfluoroalkyl acids (PFAAs) in a top predator; the American mink. Perfluorobutane sulfonate (PFBS), perfluorohexane sulfonate (PFHxS), perfluorooctane sulfonate (PFOS) and perfluoroalkyl carboxylates (PFCAs) with C8–C13 perfluorinated carbon chains were analyzed in livers from wild male mink liver (n =101) from four areas in Sweden representing two inland environments (rural and highly anthropogenic, respectively) and two different coastal environments. Mean PFOS concentrations were 1250ng/g wet weight and some mink from the urban inland area had among the highest PFOS concentrations ever recorded in mink (up to 21 800ng/g wet weight). PFBS was detected in 89% of the samples, but in low concentrations (mean 0.6ng/g ww). There were significant differences in PFAA concentrations between the geographical areas (p<0.001–0.01). Age, body condition and body weight did not influence the concentrations significantly, but there was a seasonal influence on the concentrations of perfluorodecanoic acid (PFDA) and perfluoroundecanoic acid (PFUnDA) (p<0.01 and p<0.05, respectively), with lower concentrations in autumn samples than in samples taken in the winter and spring. It is thus recommended to take possible seasonal differences into account when using mink exposure data. Abstract: In Canada, perfluoroalkyl acids (PFAAs) have been the focus of several monitoring programs and research and surveillance studies. Here, we integrate recent data and perform a multi-media assessment to examine the current status and ongoing trends of PFAAs in Canada. Concentrations of perfluorooctane sulfonate (PFOS), perfluorooctanoate (PFOA), and other long-chain perfluorocarboxylates (PFCAs) in air, water, sediment, fish, and birds across Canada are generally related to urbanization, with elevated concentrations observed around cities, especially in southern Ontario. PFOS levels in water, fish tissue, and bird eggs were below their respective Draft Federal Environmental Quality Guidelines, suggesting there is low potential for adverse effects to the environment/organisms examined. However, PFOS in fish and bird eggs tended to exceed guidelines for the protection of mammalian and avian consumers, suggesting a potential risk to their wildlife predators, although wildlife population health assessments are needed to determine whether negative impacts are actually occurring. Long-term temporal trends of PFOS in suspended sediment, sediment cores, Lake Trout (Salvelinus namaycush), and Herring Gull (Larus argentatus) eggs collected from Lake Ontario increased consistently from the start of data collection until the 1990s. However, after this time, the trends varied by media, with concentrations stabilizing in Lake Trout and Herring Gull eggs, and decreasing and increasing in suspended sediment and the sediment cores, respectively. For PFCAs, concentrations in suspended sediment, sediment cores, and Herring Gulls generally increased from the start of data collection until present and concentrations in Lake Trout increased until the late 1990s and subsequently stabilized. A multimedia comparison of PFAA profiles provided evidence that unexpected patterns in biota of some of the lakes were due to unique source patterns rather than internal lake processes. High concentrations of PFAAs in the leachate and air of landfill sites, in the wastewater influent/effluent, biosolids, and air at wastewater treatment plants, and in indoor air and dust highlight the waste sector and current-use products (used primarily indoors) as ongoing sources of PFAAs to the Canadian environment. The results of this study demonstrate the utility of integrating data from different media. Simultaneous evaluation of spatial and temporal trends in multiple media allows inferences that would be impossible with data on only one medium. As such, more co-ordination among monitoring sites for different media is suggested for future sampling, especially at the northern sites. We emphasize the importance of continued monitoring of multiple-media for determining future responses of environmental PFAA concentrations to voluntary and regulatory actions. Abstract: The sources and pathways of human exposure to perfluoroalkyl acids (PFAAs) are not well characterized, particularly in China where many perfluorinated substances are now manufactured. Here, isomer-specific PFAA analysis was used for the first time to evaluate exposure sources for Chinese people, by applying the method to 129 serum samples collected in two typical cities (Shijiazhuang and Handan) in North China. Among all samples, total perfluorooctanesulfonate (∑PFOS, mean 33.3ng/ml) was the predominant PFAA followed by perfluorohexanesulfonate (2.95ng/ml), total perfluorooctanoate (∑PFOA, 2.38ng/ml), and perfluorononanoate (0.51ng/ml). The level of ∑PFOS was higher than in people from North America in recent years. The mean concentrations of ΣPFAAs in the participants living in urban Shijiazhuang (59.0ng/ml) and urban Handan (35.6ng/ml) were significantly higher (p<0.001 and p=0.041, respectively) than those living in the rural district of Shijiazhuang (24.3ng/ml). The young female sub-population had the lowest ΣPFAA concentrations compared with older females and all males. On average, the proportion of linear PFOS (n-PFOS) was only 48.1% of ∑PFOS, which is much lower than what was present in technical PFOS from the major historical manufacturer (ca. 70% linear), and which is also lower than data reported from any other countries. Moreover, the proportion of n-PFOS decreased significantly with increasing ∑PFOS concentration in the serum samples (r=−0.694, p<0.001). Taken together, the data lend weight to previous suggestions that i) high branched PFOS content in serum is a biomarker of exposure to PFOS-precursors, and ii) that people with the highest ∑PFOS concentrations are exposed disproportionately to high concentrations of PFOS-precursors. On average, linear PFOA (n-PFOA) contributed 96.1% of ∑PFOA, significantly higher than in technical PFOA (ca. 75–80% linear), but lower than in Americans, suggesting higher exposure to electrochemically fluorinated PFOA than in other countries, including the United States. Abstract: Dietary intake has been hypothesized to be the major pathway of human exposure to perfluoroalkyl acids (PFAAs). However, difficulties associated with the analysis of PFAAs at ultra trace levels in food samples have prevented the confirmation of this hypothesis. In this study, the dietary intake of PFAAs for the general Swedish population was estimated by applying a highly sensitive analytical method to a set of archived food market basket samples from 1999, 2005 and 2010. Dietary exposure to perfluorooctane sulfonic acid (PFOS) (860–1440pgkg−1 day−1), perfluoroundecanoic acid (PFUnDA) (90–210pgkg−1 day−1), perfluorodecanoic acid (PFDA) (50–110pgkg−1 day−1) and perfluorononanoic acid (PFNA) (70–80pgkg−1 day−1) was dominated by the consumption of fish and meat. In contrast, dietary exposure to PFOA (350–690pgkg−1 day−1) originated from low levels (8–62pgg−1) found in several high consumption food categories including cereals, dairy products, vegetables and fruit. The dietary intakes of PFOS and PFOA estimated in this study were 4 to 10 times lower compared to previous exposure modeling studies. Nevertheless, the dietary intake of PFOS and PFOA was still a factor of 6 to 10 higher than exposure through ingestion of household dust and drinking water estimated for the general Swedish population. For perfluorohexanoic acid (PFHxA), perfluoroheptanoic acid (PFHpA) and perfluorohexane sulfonic acid (PFHxS) drinking water intake was the major exposure pathway (36–53% of the total exposure) whereas dust ingestion made a significant contribution (27–49%) to the total exposure for PFHxA, PFHpA, PFNA, perfluorotridecanoic acid (PFTrDA) and perfluorotetradecanoic acid (PFTeDA). Dietary intakes varied by less than a factor of three for all PFAAs during the different sampling years which demonstrates that dietary intake has been fairly constant over the past decade when many manufacturing changes occurred. Abstract: In recent exposure modeling studies diet has been identified as the dominant pathway of human exposure to perfluorooctanoic acid (PFOA) and perfluorooctane sulfonic acid (PFOS). However, the paucity of highly sensitive and accurate analytical data to support these studies means that their conclusions are open to question. Here a novel matrix effect-free method is described for ultra-trace analysis of perfluoroalkyl carboxylic acids and perfluoroalkane sulfonic acids in dietary samples of varied composition. The method employs ion pair extraction of the analytes into methyl tert-butyl ether and subsequent solid phase extraction clean-up on Florisil and graphitized carbon. The target compounds are separated and detected using ultra performance liquid chromatography coupled to tandem mass spectrometry. Special care was taken to avoid procedural blank contamination and potential contamination sources were elucidated. The performance of the method was validated for five different food test matrices including a duplicate diet sample. Method detection limits in the low to sub pgg−1 range were obtained for all target analytes, which is 5–100 times more sensitive than previously reported for duplicate diet samples. Total method recoveries were consistently between 50 and 80% for all analytes in all tested food matrices and effects of co-extracted matrix constituents on ionization of the target compounds were found to be negligible. The precision of the method (defined as percentage relative standard deviation) at concentrations close to the respective method limits of quantification was <15% for all analytes. Accurate quantification at ultra-trace levels was demonstrated by laboratory control spike experiments. For the first time the presence of long-chain PFCAs in duplicate diet samples is reported. The method presented here can thus support an improved assessment of human exposure from dietary intake for a range of PFCA and PFSA homologues. Re-analysis of duplicate diet samples, which had been analyzed earlier using another analytical methodology, indicated that dietary intake of PFOA and PFOS may previously have been overestimated. Abstract: Perfluorooctanoate (PFOA) and perfluorooctanesulfonate (PFOS) are surface active fluorochemicals that, due to their exceptional stability to degradation, are persistent in the environment. Both PFOA and PFOS are eliminated slowly in humans, with geometric mean serum elimination half-lives estimated at 3.5 and 4.8 years, respectively. The biological activity of PFOA and PFOS in rodents is attributed primarily to transactivation of the nuclear receptor peroxisome proliferator activated receptor alpha (PPARA), which is an important regulator of lipid and carbohydrate metabolism. However, there are significant species-specific differences in the response to PFOA and PFOS exposure; non-rodent species, including humans, are refractory to several but not all of these effects. Many of the metabolic effects have been attributed to the activation of PPARA; however, recent studies using PPARα knockout mice demonstrate residual PPARA-independent effects, some of which may involve the activation of alternate nuclear receptors, including NR1I2 (PXR), NR1I3 (CAR), NR1H3 (LXRA), and NR1H4 (FXR). The objective of this investigation was to characterize the activation of multiple nuclear receptors and modulation of metabolic pathways associated with exposure to PFOA and PFOS, and to compare and contrast the effects between rat and human primary liver cells using quantitative reverse transcription PCR (RT-qPCR). Our results demonstrate that multiple nuclear receptors participate in the metabolic response to PFOA and PFOS exposure resulting in a substantial shift from carbohydrate metabolism to fatty acid oxidation and hepatic triglyceride accumulation in rat liver cells. This shift in intermediary metabolism was more pronounced for PFOA than PFOS. Furthermore, while there is some similarity in the activation of metabolic pathways between rat and humans, particularly in PPARA regulated responses; the changes in primary human cells were more subtle and possibly reflect an adaptive metabolic response rather than an overt metabolic regulation observed in rodents. Abstract: 11β-Hydroxysteroid dehydrogenase 2 (11β-HSD2) regulates active glucocorticoid access to glucocorticoid and mineralocorticoid receptors by metabolizing it to an inactive form. Perfluoroalkylated substances (PFASs) are man-made polyfluorinated compounds that are widely used and persistent in the environment. We tested the inhibitory potencies of four PFASs including perfluorooctanoic acid (PFOA), perfluorooctane sulfonate (PFOS), perfluorohexanesulfonate (PFHxS) and perfluorobutane sulfonate (PFBS) on human and rat 11β-HSD2. PFOS was a potent inhibitor of both human (IC50 =48nM) and rat (IC50 =293nM) 11β-HSD2 activities. The potencies for the inhibition of human and rat 11β-HSD2 activities were PFOS>PFOA>PFHxS>PFBS. PFASs showed competitive inhibition of both human and rat 11β-HSD2 activities. This observation indicates that PFOS is a potent endocrine disruptor for glucocorticoid metabolism. Abstract: Polyfluoroalkyl chemicals (PFCs) comprise a group of man-made organic compounds, some of which are persistent contaminants with developmental toxicity shown in laboratory animals. There is a paucity of human perinatal exposure data. The US EPA conducted a pilot study (Methods Advancement for Milk Analysis) including 34 breastfeeding women in North Carolina. Milk and serum samples were collected at 2–7 weeks and 3–4 months postpartum; 9 PFCs were assessed in milk and 7 in serum. Perfluorooctane sulfonic acid (PFOS), perfluorooctanoic acid (PFOA), perfluorononanoic acid (PFNA), and perfluorohexane sulfonic acid (PFHxS) were found in nearly 100% of the serum samples. PFOS and PFOA were found at the highest concentrations. PFCs were below the limit of quantification in most milk samples. Serum concentrations of PFOS, PFOA and PFHxS were lower (p <0.01) at the second visit compared to the first visit. Living in North Carolina 10 years or longer was related to elevated PFOS, PFOA and PFNA (p ≤0.03). These pilot data support the need to further explore perinatal PFC exposures and potentially related health effects, as planned in the upcoming National Children''s Study which provided the framework for this investigation. Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) are environmentally widespread and persistent chemicals with multiple toxicities reported in experimental animals and humans. These compounds can trigger biological activity by activating the alpha isotype of peroxisome proliferator-activated receptors (PPARs), ligand-activated transcription factors that regulate gene expression; however, some biological effects may occur independently of the receptor. Activation of the peroxisome proliferator-activated receptor alpha (PPARα) modulates lipid and glucose homeostasis, cell proliferation and differentiation, and inflammation. Reported immunomodulation in experimental animals exposed to PFOA and PFOS has included altered inflammatory responses, production of cytokines and other proteins, reduced lymphoid organ weights, and altered antibody synthesis. Mounting experimental animal evidence suggests PPARα independence of some immune effects. This evidence originates primarily from studies with PPARα knockout models exposed to PFOA that demonstrate hepatic peroxisome proliferation, reduced lymphoid organ weights, and altered antibody synthesis. As human PPARα expression is significantly less than that of rodents, potential PPARα independence indicates that future research must explore mechanisms of action of these compounds, including PPARα -dependent and -independent pathways. This multiauthored review contains brief descriptions of current and recently published work exploring immunomodulation by PFOA and PFOS, as well as a short overview of other PPARα ligands of therapeutic and environmental interest. Per- and polyfluoroalkyl substances (PFAS) represent a large class of structurally diverse chemicals of increasing public concern, mostly due to their chemical stability and undetermined toxicity profiles. In laboratory animals, adverse effects implicated for certain PFAS, perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) in particular, include liver toxicity and the associated metabolic dysregulation, immune and thyroid alterations, reproductive toxicity, and selected tumors. The broad commercialization and environmental distribution of PFAS has drawn attention to the need for understanding risks associated with combined exposure to multiple PFAS in complex mixtures. The purpose of this investigation is to determine whether binary combinations of PFAS elicit a molecular response that is either greater than or less than the sum of the individual responses. Exposure of FaO rat hepatoma cells for 24 h to 25 μM–200 μM of the 4- and 8-carbon perfluorocarboxylic acids (PFBA and PFOA) or the 4, 6, and 8-carbon perfluorosulfonic acids (PFBS, PFHxS, and PFOS, respectively) individually caused a dose-dependent increase in PPARα-regulated expression of peroxisomal bifunctional enzyme (Ehhadh). Potency increased with carbon number, with the carboxylates eliciting a greater transcriptional response than the corresponding sulfonates. Combined exposure to PFOA and PFBA produced an effect that was significantly less than the sum of the individual responses. The response to the combination of PFOA and PFOS produced a summative effect at concentrations that were not cytotoxic. Combined exposures to PFOS and either PFBS or PFHxS at low noncytotoxic concentrations produced a transcriptional effect that was significantly less than the sum of the individual effects. The results demonstrate that among the five structurally related perfluoroalkyl acids included in this investigation, PPARα transcriptional activation in response to combined binary exposures is consistently at or below that predicted by the sum of the individual effects.
