Edible plants such as sweet potato are sources of natural antioxidants that can be exploited in the management and treatment of insulin resistance. This present study investigated the effects of the extracts of an orange-fleshed sweet potato on oxidative stress biomarkers (glutathione status and lipid peroxidation) and activities of antioxidant enzymes (catalase, CAT and glutathione peroxidase, GPx) in palmitate-induced insulin resistant C2C12 cells. The intracellular antioxidant status of the cells was also measured using Ferric reducing antioxidant power (FRAP) and Trolox equivalent antioxidant capacity (TEAC) assays. Furthermore, this study determined the effect of the extracts on the regulation of some type 2 diabetes associated genes; glucose transporter 4 ( glut4 ), Nuclear respiratory factor 1 ( nrf1 ), Myocyte enhanced factor 2A ( mef2a ), Carnitine palmitoyltransferase 1 ( cpt1 ) and Acetyl-CoA carboxylase 2 ( acc2 ). The results showed a significant ( p < 0.05) increase in intracellular GSH level, a significant reduction in the level of malonaldehyde and a significant improvement in the intracellular antioxidant status upon treatment of the insulin resistant cells with the extracts. The extracts were also able to positively modulate the expression levels of the type 2 diabetes associated genes. On the other hand, HPLC-MS analysis of the extracts showed the presence of polyphenols which could have contributed to the bioactivity of the extracts through their antioxidant effects.

Context Although olive mill wastewater (OMWW) is a good source of bioactive phenolic compounds, disposing OMWW is a serious environmental challenge. Production of wine via fermenting OMWW may be a promising alternative to deal with OMWW. However, whether or not olive wine from OMWW still reserves its original bioactivities remains unclear. Objective This study examines antioxidant activity of olive wine fermented from OMWW. Materials and methods Hydroxytyrosol in olive oil was determined by HPLC. Total flavonoid, total polyphenol and in vitro antioxidant activities were measured by spectrophotometry. Aged mice were intragastricly administered 7, 14 and 28 mL/kg olive wine consecutively for 30 d. Afterward, levels of malonaldehyde (MDA), protein carbonyl, reduced glutathione (GSH) and activity of superoxide dismutase (SOD) were assayed in mouse plasma and liver. Results Contents of hydroxytyrosol, total flavonoid and total polyphenol in olive wine were 0.14 ± 0.01, 0.29 ± 0.06 and 0.43 ± 0.03 mg/mL, respectively. The IC50 value of olive wine to scavenge DPPH and hydroxyl free radicals was 2.5% and 3.2% (v/v), respectively. Compared with the solvent control group, olive wine with a dose of 28 mL/kg remarkably lowered mouse MDA concentration in liver, and reduced protein carbonyl level in plasma (p < 0.05). Meanwhile, olive wine at doses of 7 and 28 mL/kg notably enhanced SOD activity in both mouse plasma and liver (p < 0.05). The beneficial effect on liver was superior to that of γ-tocopherol. Conclusion The study demonstrated that olive wine from OMWW has potential for treating oxidative stress-associated diseases.

Objective: To investigate the association of sperm DNA integrity with semen routine parameters and seminal plasma oxidative stress and its influence on in vitro fertilization (IVF) in males with infertility., Methods: Using sperm chromatin dispersion (SCD), we detected sperm DNA damage in 433 infertile men undergoing IVF. Based on the sperm DNA fragmentation index (DFI), we divided the patients into a low DFI (lt;30%) and a high DFI ( ≥30%) group and then compared sperm concentration, the percentage of progressively motile sperm (PMS), the contents of malondialdehyde (MDA) and total antioxidant capacity (TAC) in the seminal plasma, and the rates of fertilization, cleavage and high-quality embryos between the two groups of patients., Results: Compared with the low DFI group, the high DFI group showed significantly decreased rates of PMS (［48.6±16.7］ vs ［29.2±16.8］%, P<0.01) and fast PMS ［19.0±9.1］ vs ［9.4±6.6］%, P<0.01), but no statistically significant difference in sperm concentration (［51.4±30.9］ vs ［52.3±32.4］ ×106/ml, P>0.05). The content of MDA in the seminal plasma was markedly higher in the high DFI than in the low DFI group (［2.28±0.26］ vs ［0.95±0.18］ nmol/L, P<0.01) but that of TAC remarkably lower in the former than in the latter (［10.2±3.5］ vs ［33.2±7.9］ U/L, P<0.01). The rate of fertilization was significantly lower in the high DFI than in the low DFI group (［58.9±30.0］ vs ［77.2±25.0］%, P<0.01), but there were no significant differences between the two groups in the rates of cleavage (［70.7±35.6］ vs ［80.4±15.6］%P>0.05) and high-quality embryos (［40.4±31.3］ vs ［41.7±29.4］%,P>0.05)., Conclusions: Sperm DNA damage is associated with seminal oxidative stress and may affect the outcomes of IVF by reducing the rate of fertilization.

Background: Hydrogen sulfide (H2S), as a novel gaseous messenger molecule, plays an important role in signal transduction and biological modulation., Purpose: In the present study the effect of H2S after compression injury of cauda equina was studied., Study Design: The setting of this study is the laboratory investigation., Methods: A total of 162 rats were randomly allocated into three groups: sham group, compression group, and H2S group. Cauda equina compression (CEC) injury in rats was induced by implanting silicone gels (10×1×1 mm) into the epidural spaces L5 and L6; laminectomy was performed at the L4 level of the vertebra in the sham-operated group. The experimental group was treated with sodium hydrosulfide intraperitoneally (20 µmol/kg body weight), whereas the compression and sham groups received equal volumes of physiological saline. Levels of malonaldehyde (MDA) and glutathione (GSH) were determined immediately before CEC surgery, 12 h, 24 h, 48 h, and 72 h after CEC surgery. Furthermore, hematoxylin and eosin (H&E) staining and terminal deoxynucleotidyl transferase-mediated biotinylated UTP nick-end labeling (TUNEL) assay were performed 48 h after CEC., Results: Hematoxylin and eosin staining showed that myelin sheath and the cauda equina fibers in the compression group were less compact and highly degenerated compared with the sham group, and that H2S treatment could improve the status. Terminal deoxynucleotidyl transferase-mediated biotinylated UTP nick-end labeling staining exhibited that decreased number of TUNEL positive cells was found in the H2S group than in the compression group. The level of MDA was increased in the sham and H2S groups compared with the compression group (p<.05, p<.01), whereas the level of GSH was decreased (p<.05, p<.01)., Conclusions: With the above data, we conclude that H2S could reduce the oxidative stress and has neuroprotective effect in acute cauda equina syndrome., (Copyright © 2016 Elsevier Inc. Deoxynivalenol (DON) is one of the most common mycotoxins. The aim of this study consists in using diverse cellular and molecular assays to evaluate cytotoxicity, genotoxicity as well as oxidative damage and to investigate their mechanisms in human peripheral blood lymphocytes. The human lymphocytes were cultured in eight different doses of DON (0, 6.25, 12.5, 25, 50, 100, 250 and 500 ng/mL) during 6, 12 and 24 h. DON was able to decrease cell viability and cause damage to the membrane, the chromosomes or the DNA at all times of culture. It was also able to induce lipid peroxidation and raise the levels of 8-OHdG and ROS in 6, 12 and 24 h. The results of the RT-PCR and the Western Blot indicated that DON is able to enhance mRNA or protein expressions of DNA repair genes and HO-1 in 6 h and to inhibit these expressions in 24 h. DON potentially triggers genotoxicity in human lymphocytes. This mechanism is probably related to depletion of antioxidase and oxidative damage to the DNA that reduced expression of HO-1, thereby inhibiting the ability of DNA repair., (Copyright © 2013 Elsevier Ltd. We aimed to determine the oxidative stress status in placentas obtained from gestational (GDM) and type 1 (T1D) diabetic pregnancies. Malonaldehyde and protein carbonyls, two biomarkers of oxidative damage, were higher in T1D but not in GDM placentas. Also, higher reduced glutathione and lower oxidized glutathione levels and higher glutathione peroxidase activity were found in T1D but not in GDM placentas. Diabetic complications associated with increased oxidative stress can be suppressed by antioxidants. In the present study we investigated the antidiabetic and antioxidant effects of Kombucha (KT), a fermented black tea, in comparison to that of unfermented black tea (BT), in ALX-induced diabetic rats. ALX exposure lowered the body weight and plasma insulin by about 28.12% and 61.34% respectively and elevated blood glucose level and glycated Hb by about 3.79 and 3.73 folds respectively. The oxidative stress related parameters like lipid peroxidation end products (increased by 3.38, 1.7, 1.65, 1.94 folds respectively), protein carbonyl content (increased by 2.5, 2.35, 1.8, 3.26 folds respectively), glutathione content (decreased by 59.8%, 47.27%, 53.69%, 74.03% respectively), antioxidant enzyme activities were also altered in the pancreatic, hepatic, renal and cardiac tissues of diabetic animals. Results showed significant antidiabetic potential of the fermented beverage (150 mg lyophilized extract/kg bw for 14 days) as it effectively restored ALX-induced pathophysiological changes. Moreover, it could ameliorate DNA fragmentation and caspase-3 activation in the pancreatic tissue of diabetic rats. Although unfermented black tea is effective in the above pathophysiology, KT was found to be more efficient. Oxidative stress is one of the major mechanisms implicated in inorganic arsenic poisoning. Curcumin is a natural phenolic compound with impressive antioxidant properties. What's more, curcumin is recently proved to exert its chemopreventive effects partly through the activation of nuclear factor (erythroid-2 related) factor 2 (Nrf2) and its antioxidant and phase II detoxifying enzymes. In vivo, we investigated the protective effects of curcumin against arsenic-induced hepatotoxicity and oxidative injuries. Our results showed that arsenic-induced elevation of serum alanine amino transferase (ALT) and aspartate aminotransferase (AST) activities, augmentation of hepatic malonaldehyde (MDA), as well as the reduction of blood and hepatic glutathione (GSH) levels, were all consistently relieved by curcumin. We also observed the involvement of curcumin in promoting arsenic methylation and urinary elimination in vivo. Furthermore, both the hepatic Nrf2 protein and two typically recognized Nrf2 downstream genes, NADP(H) quinine oxidoreductase 1 (NQO1) and heme oxygenase-1 (HO-1), were consistently up-regulated in curcumin-treated mice. Our study confirmed the antagonistic roles of curcumin to counteract inorganic arsenic-induced hepatic toxicity in vivo, and suggested that the potent Nrf2 activation capability might be valuable for the protective effects of curcumin against arsenic intoxication. Aim of present study was to investigate the effect of NAC on experimental chronic hepatotoxicity models induced by carbon tetrachloride (CCl₄) and thioacetamide (TAA). CCl₄ toxicity was induced by administering 200 μl CCl₄ (diluted 2:3 in coconut oil)/100 g body weight, p.o., twice weekly for 8 weeks. TAA toxicity was induced by administering 150 mg/kg b. wt. of TAA i.p., twice weekly for 8 weeks. NAC treatment was started along with toxicants (CCl₄ and TAA) for 8 weeks and continued for further 4 weeks. Self reversal group was kept without any treatment for 4 weeks after completion of toxicant treatments. Alanine aminotransferase (ALT), Aspartate aminotransferase (AST), Alkaline phosphatase (ALP), Bilirubin were measured in serum. Hydroxyproline (HP), lipid peroxidation (LPO), catalase (CAT), Glutathione peroxidase (GPx) and Glutathione (GSH) were determined in liver samples by colorimetric methods. Cytochrome P450 2E1 (CYP 450 2E1), activity was determined as hydroxylation of aniline in liver microsomes. General examination and histological analysis were also performed. Serum markers of liver damage (AST, ALT, ALP and Bilirubin) were increased by CCl₄ and TAA intoxication (p<0.001), whereas co-treatment with NAC reversed such changes (p<0.001). HP was enhanced in toxicant groups (p<0.001 in CCl₄ and TAA), but inhibited by NAC (p<0.001). LPO was increased while as GSH, CAT and GPx decreased by the administration of CCl₄ and TAA (p<0.001); co-administration of NAC restored these liver markers to normal levels (p<0.001). Biochemical determinations were corroborated by general and histological findings. Keeping in view the biochemical and histopathological studies, it was concluded that CCl₄ and TAA are strong hepatotoxic agents that produce liver fibrosis with close proximity to human etiology (micronodular cirrhosis) and NAC has a significant protective activity against CCl₄ and TAA. This investigation sought to comprehensively evaluate the protective properties of Milk Thistle seeds against Supracideinduced oxidative stress in the Marbled Spinefoot fish. Liquid chromatography--mass spectrometry (LC--MS) was employed to identify metabolites present in Silybum marianum seeds. The LC50 value of organophosphorous insecticide was determined as 7.5 μg L-1 through a 24-hour bath exposure. The experimental setup involved a 14-day dietary supplementation of S. marianum seeds followed by exposure to the determined Supracide LC50. Various physiological parameters, including lipid degradation, Alanine aminotransferase (ALT), Lactate dehydrogenase (LDH), and Total cholesterol (T.ch) levels, were measured to assess fish health and stress. Additionally, histopathological examinations were conducted on the hepatopancreas tissues of both normal and stressed fish. The obtained results indicated significant increases (P<0.05) in serum ALT and LDH activities in the group of fish exposed to the toxin. Moreover, the levels of lipid peroxidation products in hepatopancreas homogenates were significantly higher (P<0.05) in the exposed fish group, except for total cholesterol, which exhibited a significant decrease. Histological analysis further revealed notable tissue modifications in the examined organs, while the control group of fish exhibited normal appearance. Taken together, these findings suggest that Milk thistle seeds may possess protective effects against Supracide-induced oxidative stress in challenged S. rivulatus fish. As a result, this study provides valuable insights into potential alternative ameliorating agents for addressing aquaculture contamination caused by organophosphate insecticides. Background: Stroke is a clinical syndrome consisting of rapidly developing clinical signs of focal or global disturbance of cerebral function. Antioxidants are exogenous or endogenous compounds acting by scavenging reactive oxygen species or their precursors, inhibiting reactive oxygen species formation and binding metal ions needed for the catalysis of its generation. Atherogenic index, a predictor of atherosclerosis, increases in dyslipidemia also shows a positive association with oxidative stress. Aims and Objectives: Our objective was to compare the lipid profile, the markers of lipid peroxidation (malonaldehyde and paraoxonase [PON]), and atherogenic index in non-diabetic stroke patients with healthy controls and to evaluate the correlation between the lipid parameters with markers of lipid peroxidation and atherogenic index in those patients. Materials and Methods: One hundred diagnosed stroke patients from the Neurology department were compared with 100 age-matched healthy controls. Estimation of lipid profile parameters was done by AutoAnalyzer using standardized kits. Estimation of serum malonaldehyde was done based on the principle of utilization of Thiobarbituric acid. Serum PON1 activity was estimated using paranitrophenyl acetate substrate. Results: Among the lipid profile parameters, high-density lipoprotein (HDL) was significantly lower (P<0.001) and low-density lipoprotein (LDL) was significantly higher among stroke patients. Serum malonaldehyde level was significantly increased and PON1 activity was significantly decreased in stroke patients in comparison to healthy individuals. A significant positive correlation between HDL and PON1 and a negative correlation between LDL and PON1 were found. Atherogenic index is negatively correlated with PON1. Conclusion: Low level of PON1 activity and an increased level of malonaldehyde were found in stroke patients indicating an imbalance between free radical generation and its scavenging action and also associated with more chance of atherosclerosis. Diabetes mellitus (DM) is a metabolic disease that causes high blood glucose which is called hyperglycemia. In addition to hyperglycemia, oxidative stress, which raises the risk of complications, is one of the many other important factors that contribute significantly to the pathogenesis of diabetes. The goal of the current study was to determine how well (Ganoderma lucidum) ethanol extract (GLE) intervenes against oxidative stress in living cells, with a focus on its mechanistic elements in the management of diabetes mellitus. Rats treated with alloxan showed a significant (p≤0.01) increase in serum glucose levels from 92.67 mg/dL in normal control rats to 311.24 mg/dL in treated rats. Serum glucose levels in rats fed GLE (200, 400, 600, and 800 mg/kg bw) for 28 days showed a significant (p≤0.05) decline, with values recorded at 294.26, 268.34, 210.17, and 192.78, respectively. With GLE intervention, there was a dose-dependent increase in the rate of serum glucose decrease. The biochemical indicators of oxidative stress levels in plasma, nitric oxide (NO), malonaldehyde (MDA), and reactive oxygen species (ROS), all exhibited the same conduct. However, there was a notable (P≤0.05) increase in the various antioxidant protective mechanisms in the serum, which included bioactive molecules (GSH fractions), antioxidant vitamins (A, C, and E), and antioxidant enzymes [glutathione peroxidase (GSH-Px), glutathione reductase (GSH-Rd) superoxide dismutase (SOD) and catalases (CAT)]. Ultimately, these results establish a foundation for the application of (Ganoderma lucidum) extracts in the prevention and/or treatment of T2D by inhibiting oxidative stress, which is one of the factors contributing to the illness and its consequent challenges. Background: Oral lichen planus is a chronic, mucocutaneous, inflammatory disease, with an unknown etiology. Reactive oxygen species and oxidative damage to the tissues might be the cause. Malonaldehyde (MDA), a low molecular weight end product of lipid peroxidation reaction is a suitable biomarker of endogenous DNA damage. monitoring the oxidant-antioxidant status of saliva may serve as an efficient marker of disease development in oral lichen planus patients. Aim and Objectives: To evaluate salivary oxidative stress in oral lichen planus subject using MDA and compare it with control subjects. Furthermore, to compare MDA levels in erosive and hypertrophic lichen planus. Materials and Methods: The current study is case-control study. Unstimulated salivary samples in the morning hours were taken from oral lichen planus subjects (n = 25) and controls subjects without any oral disease (n = 25). The saliva was centrifuged at 900 g for 10 min at a temperature of 4°C. Then, the entire filtrate was transferred to Eppendorf test tubes and frozen at-80°C until analysis. Salivary MDA was done through thiobarbituric acid reactive substance assay as per the protocol laid down by the manufacturer (Sigma Aldrich Lipid Peroxidation Assay Kit). Results: The data were expressed as the mean ± standard deviation and the statistical analysis was done using Student's t-test using SPSS version 21 IBM software. The salivary level of MDA was significantly higher than that of controls (P < 0.05). Conclusion: The higher level of MDA in patients with oral lichen planus suggests that free radicals and the resulting oxidative damage may be important in the pathogenesis of oral lichen planus lesions. Metal oxide nanoparticles (MONPs) are regarded as critical tools for overcoming ongoing and prospective crop productivity challenges. MONPs with distinct physiochemical characteristics boost crop production and resistance to abiotic stresses such as drought. They have recently been used to improve plant growth, physiology, and yield of a variety of crops grown in drought-stressed settings. Additionally, they mitigate drought-induced reactive oxygen species (ROS) through the aggregation of osmolytes, which results in enhanced osmotic adaptation and crop water balance. These roles of MONPs are based on their physicochemical and biological features, foliar application method, and the applied MONPs concentrations. In this review, we focused on three important metal oxide nanoparticles that are widely used in agriculture: titanium dioxide (TiO2), zinc oxide (ZnO), and iron oxide (Fe3O4). The impacts of various MONPs forms, features, and dosages on plant growth and development under drought stress are summarized and discussed. Overall, this review will contribute to our present understanding of MONPs’ effects on plants in alleviating drought stress in crop plants.

Study was carried out to assess supplemental effects of dietary reduced ubiquinol (rCoQ10) on lipid profile and serum antioxidant activity in broiler chicks fed diets with three energy levels. 270 broiler chicks divided into nine groups (3 energy levels x 3 levels of rCoQ10) with three replicates. Birds were fed with basal energy (BE), low energy (BE-100 (kcal)) and high energy (BE+100 (kcal.kg-1) feed) and rCoQ10 at 0, 20 and 40ppm. Broiler chicks were distributed in completely randomized design and reared for 42d and at end of study, serum and muscle samples were collected for antioxidant activities, lipid contents. The activity of superoxide dismutase, glutathione peroxidase, reduced glutathione, vitamin E were increased and serum malonaldehyde, total, LDL-cholesterol as well as muscle cholesterol were reduced by intake of rCoQ10. The consumption of rCoQ10 improved muscle oxidative stability than the control. Ubiquinol at 20ppm favourably altered lipid profile with reduced lipid peroxidation and improved serum antioxidants activities. [ABSTRACT FROM AUTHOR]

Abstract Background In diabetes mellitus, uncontrolled hyperglycemia has been reported to induce oxidative stress, which may lead to health complications. Vitamin D, however, acts as a non-enzymatic antioxidant to protect cells against oxidative stress and damage. Objective To investigate the antioxidative effect of vitamin D combined with calcium in streptozotocin (STZ)-induced diabetic rats. Methods Rats were divided into four groups (ten rats in each group). The first group (control) received a normal diet and water. The second group, including STZ-induced diabetic rats (diabetic controls), received a normal diet and water. The third group, also including STZ-induced diabetic rats, received vitamin D (2000 IU/day) with calcium (500 mg/kg/day) orally for 28 consecutive days. The fourth group consisted of STZ-induced diabetic rats that received insulin treatment for 28 consecutive days. Activities of superoxide dismutase (SOD), glutathione peroxidase (GPO) and catalase were measured in the liver tissues. The level of malonaldehyde (MDA) was measured in the plasma. Results Diabetic rats showed a significant decrease in the activities of SOD, GPO and catalase compared to normal rats. Oral administration of vitamin D with calcium to diabetic rats caused a significant increase in the activities of SOD, GPO and catalase compared with the untreated group. Furthermore, the plasma level of MDA was significantly elevated in diabetic rats compared to normal rats. Diabetic rats treated with vitamin D and calcium had a significantly reduced level of MDA, suggesting that vitamin D with calcium played a vital role in the protection of tissues from damage by free radicals. Conclusion Oral supplementation with vitamin D and calcium may be a useful treatment for diabetic patients to reduce/prevent the pathological complications of diabetes. [ABSTRACT FROM AUTHOR]

The development of nonalcoholic fatty liver disease (NAFLD) is caused by the steatosis of hepatocytes, which induces oxidative stress (OS). Thus, OS has an important role in the development of NAFLD. In the present study, the L‑02 hepatocyte cell line was used to develop a steatosis cell model. The best model was determined using an MTT assay and the triglyceride levels. Model cells were treated with high concentrations of uric acid (UA; 0, 5, 10, 20 and 30 mg/dl) for 24, 48, 72 and 96 h. Indicators of oxidation were then measured, which included total superoxide dismutase (SOD), malonaldehyde (MDA) and reduced glutathione (GSH), and the transcriptional and translational levels of SOD1 and γ‑glutamate‑cysteine ligase (γ‑GCLC) were also determined. In addition, the intracellular levels of aspartate aminotransferase and alanine aminotransferase (ALT) were detected. The activity of SOD1 decreased over time and the result was supported by the results of western blotting. The transcriptional levels of SOD1 in model cells was significantly higher than untreated cells at 48 h. With the decreased levels of SOD1 and GSH, MDA increased in a time‑dependent manner. The content of GSH decreased with time as well, which was also reflected in the results of western blotting. The transcriptional levels of γ‑GCLC in all UA‑treated groups were lower when compared with those observed in the model group. The activity of ALT tended to increase, depending on the duration of treatment. Treatment with 5 and 10 mg/dl UA had an antioxidative effect on the model cells, and 30 mg/dl UA treatment for 48 h increased OS in the cells. [ABSTRACT FROM AUTHOR]

Eight articles were finally selected that formed the base for this review. Results: The findings from the present review demonstrate that oxidative stress and consequently, elevated levels of malonaldehyde compared to controls, plays a role in the pathophysiology of oral lichen planus. In the eight relevant studies, estimation of the malonaldehyde in serum, saliva and tissue samples was done and its levels were found to be significantly higher than controls. Conclusion: This review reveals only 8 studies that demonstrate the elevated oxidative stress levels in oral lichen planus using malonaldehyde as a biomarker. Therefore, further studies need to be performed which would estimate the levels of MDA in serum, saliva and tissue samples of the same group of patients with oral lichen planus, so as to draw definitive conclusions and significant correlation between the MDA levels from the three samples.

Deltamethrin (DM) is being used as a substitute for organochlorines and organophosphates in pest control because of its low environmental persistence and toxicity. But it has become an environmental contaminant as it has been used widely. In this study, we investigated the effect of DM (technical grade) on the antioxidant system of adult zebrafish. For this, six-month-old fish were exposed to 2, 4 and 6 μg/L of DM for 96 h. The tissues selected were liver and ovary. Our data showed that exposure to DM increases CAT (catalase), SOD (superoxide dismutase), GPx (glutathione peroxidase, antioxidant enzymes), LPO (lipid peroxidation, non-enzymatic antioxidant) and GST (glutathioneS-transferase, detoxifying enzyme) in liver and ovary. Increased GST could detoxify the toxicant; still there could be enough DM to cause oxidative stress. It appears from our study that zebrafish used compensatory mechanisms in eliminating reactive oxygen species. PURPOSE: To evaluate the effects of the oxidative stress in an experimental model of torsion/detorsion of the spermatic cord and the legitimacy of this model for oxidative stress studies. METHODS: Forty-eight male Wistar rats were randomized in two groups (n=24): G-1 (Sham) and G-2 (Ischemia/Reperfusion). All rats received intraperitoneal saline injections (2.0 ml), at 21, 9, and 1 h before right spermatic cord torsion or first sham operation. Detorsion or second sham operation was carried out 3 h later followed by testis and blood samples collection (T-0). Additional samples were collected at 1-3-6 h time-points for assessment of testis malonaldehyde, glutathione, and plasma total antioxidant power (TAP). RESULTS: Spermatic cord torsion/detorsion induced a significant increase in testicular malonaldehyde contents and a significant decrease in glutathione concentrations in ischemic rats compared with sham animals. Additional increase in malonaldehyde levels occurred during reperfusion in G-2 rats. TAP was similar in both groups denoting absence of systemic effects in this study. CONCLUSION: Torsion/detorsion of the spermatic cord for 3 h induces significant lipid peroxidation and reduction in glutathione content of the testis and is, therefore, a valid model for studying the oxidative stress effects of the ischemia/reperfusion injury in young rat testis.OBJETIVO: Investigar os efeitos do estresse oxidativo utilizando um modelo experimental de torção/destorção do cordão espermático e a aplicabilidade do modelo para estudo do estresse oxidativo. MÉTODOS: Foram utilizados 48 ratos distribuídos aleatoriamente em dois grupos (n=24): G-1 (Simulado) e G-2 (Isquemia/Reperfusão). Todos os animais receberam injeções intraperitoniais de solução salina (2,0 ml) 21-9-1 horas antes da torção ou primeira operação simulada. A destorção/segunda operação simulada (T-0) foi realizada após 3 horas, com coleta das amostras (testículo e sangue arterial) nesta ocasião e após 1-3-6 horas para determinação das concentrações de malonaldeído, glutationa e capacidade antioxidante total do plasma (CAT). RESULTADOS: a torção do cordão espermático induziu aumento significante das concentrações de malonaldeído e redução simultânea das concentrações de glutationa nos ratos do grupo G-2, comparados ao grupo simulado. Ocorreu aumento adicional das concentrações de malonaldeído durante a reperfusão (G-2). A capacidade antioxidante total do plasma foi semelhante nos dois grupos estudados, denotando ausência de efeitos sistêmicos. CONCLUSÃO: A torção durante 3 horas seguida de destorção do cordão espermático induz um nível significante de peroxidação lipídica e redução da concentração de glutationa constituindo-se um modelo válido para o estudo dos efeitos do estresse oxidativo decorrente da lesão de isquemia/reperfusão no testículo de ratos jovens.

There is a delayed increase in arterial stiffness after eccentric exercise that is possibly mediated by the concurrent delayed increase in circulating oxidative stress. Taurine has anti-oxidant action, and taurine supplementation may be able to attenuate the increase in oxidative stress after exercise. The purpose of the present study was to investigate whether taurine supplementation attenuates the delayed increase in arterial stiffness after eccentric exercise. In the present double-blind, randomized, and placebo-controlled trial, we divided 29 young, healthy men into 2 groups. Subjects received either 2.0 g of placebo ( n = 14) or taurine ( n = 15) 3 times per day for 14 days prior to the exercise, on the day of exercise, and the following 3 days. The exercise consisted of 2 sets of 20 maximal-effort eccentric repetitions with the nondominant arm only. On the morning of exercise and for 4 days thereafter, we measured serum malondialdehyde (MDA) and carotid-femoral pulse wave velocity (cfPWV) as indices of oxidative stress and arterial stiffness, respectively. On the third and fourth days after exercise, both MDA and cfPWV significantly increased in the placebo group. However, these elevations were significantly attenuated in the taurine group. The increase in MDA was associated with an increase in cfPWV from before exercise to 4 days after exercise ( r = 0.597, p < 0.05) in the placebo group, but not in the taurine group. Our results suggest that delayed increase in arterial stiffness after eccentric exercise was probably affected by the exercise-induced oxidative stress and was attenuated by the taurine supplementation. Objective To observe the effects of lead exposure through drinking water on the expression of APE1 protein in cerebral cortex, cerebellum and hippocampus of rats and its relationship with oxidative stress. Methods Forty weaned male SD rats were randomly assigned to five groups (control group and four exposure groups) according to body weights of rats. Rats in control group were given deionized water as drinking water. Rats in four exposure groups were given 100 mg/L, 200 mg/L, 400 mg/L and 800 mg/L lead acetate solution for 60 days. The activity of superoxide dismutase (SOD), the contents of hydrogen peroxide (H2O2) and malondialdehyde (MDA) in cortex, cerebellum and hippocampus were measured using kits. The protein level of APE1 in cortex, cerebellum and hippocampus were detected by Western blotting assay. Results After being exposed to lead, the APE1 protein levels were significantly decreased in cortex, cerebellum and hippocampus (P < 0.05). The protein level showed a trend of gradual decline with the increase of exposed lead (P < 0.05). With the increase of dye lead dose, the activity of SOD in cortex, cerebellum and hippocampus showed a downward trend, while the contents of H2O2 and MDA showed a rising trend. The activity of SOD was positively correlated with APE1 protein level in cortex, cerebellum and hippocampus (r=0.619, 0.380 and 0.375，P < 0.05). While the contents of H2O2 and MDA were negatively correlated with APE1 protein level in cortex, cerebellum and hippocampus (r=-0.472,-0.535,-0.436,-0.514,-0.486 and -0.316，P < 0.05). Rheumatoid arthritis (RA) is a debilitating autoimmune disease whose etiology remains unknown, but studies have consistently implicated a plethora of inflammatory mechanisms culminating in chronic symmetric and erosive synovitis. Importantly, reactive oxygen species (ROS) have been attributed to directly contribute towards the destructive, proliferative synovitis evident in RA. Accordingly, this study aimed to establish whether the degree of oxidative stress and disease activity score (DAS28) correlated with the downstream effects of oxidative damage. The redox status of neutrophils sourced from synovial fluid (SF) was measured by flow cytometry in terms of total ROS and hydroxyl radicals. Among the molecular damage markers, protein carbonylation and lipid peroxidation were detected by spectrophotometry and S-nitrosothiols by fluorimetry. Neutrophils constituted the major cellular component of the SF of patients with RA and their levels of ROS and hydroxyl radicals correlated strongly with protein carbonylation and lipid peroxidation. However, all the oxidative damage markers correlated positively with DAS28. Taken together, in patients with RA, the strong correlation between levels of ROS and DAS28 with markers of oxidative damage suggests that measurement of oxidative stress could serve as a biomarker for monitoring disease severity in RA. Numerous experimental and epidemiologic studies shown that oxygen-free-radicals are elevated in uncontrolled diabetes mellitus (DM). Paraoxonase-1 (PON1) has been reported to confer antioxidant activities by decreasing the accumulation of lipid peroxidation products. This study was designed to investigate the role of PON1 activity, glutathione reductase (GR), and lipid peroxidation in patients with diabetic nephropathy (DN). 70 subjects were included in the study: 30 as a control, 20 with type 2 DM (T2DM) with nephropathy (DN), 20 T2DM without nephropathy. All studied groups are subjected to the following laboratory investigations after their consents: fasting and postprandial blood glucose, Serum triglycerides (TG), total cholesterol (TC), high density lipoprotein (HDL) and low density lipoprotein (LDL), HbA1c, blood urea, serum creatinine, urine microalbumin, serum PON1 activity, serum malondialdehyde (MDA), and serum GR activity. Fasting and postprandial blood glucose levels, and HbA1c was significantly higher in the DN group than the diabetic or control group (p=0.0001). Serum creatinine show a significant rise in the DN group than diabetic or control group (p=0.0001). Microalbuminuria show a significant rise in the DN group than the control or diabetic group as well as in the diabetic group than control group (p=0.0001). Serum PON1 activity and GR level showed significant decrease in diabetic patients with or without nephropathy compared to the control group with a significant decrease in its activity in the DN group while there was a significant a rise of MDA in diabetic groups than control with a significant rise in DN. The decreased antioxidant enzyme activities in DN patients, suggesting that oxidative stress may contribute to the development of DN and other microvascular complications beside chronic hyperglycemia and dyslipidemia. Oxidative stress has been associated with mild cognitive impairment (MCI) and Alzheimer's disease (AD). However, little is known about oxidative stress in postoperative cognitive dysfunction (POCD) in aging. The aim of this study was to investigate urinary excretion rate of 8-isoprostane:creatinine (U8-isoPG:Cr) and malonaldehyde:creatinine (UMDA:Cr) to predict short-term POCD in elderly patients undergoing general and orthopedic surgery. 72 patients aged above 65 years were enrolled in this prospective observational study. Each patient underwent cognitive testing to determine POCD performed by an investigator before surgery and 1 week after surgery. Morning urine was collected at baseline, 1, 2, and 7 days postoperatively. U8-isoPG was performed using enzymelinked immunosorbent assay (ELISA), and UMDA levels were measured by chemiluminescence detection. Creatinine levels were also analyzed if differences in the oxidative biomarkers were observed in the urine creatinine concentration. (1). Of 72 patients who completed cognitive testing, postoperative cognitive dysfunction was detected in 29.2 % (n = 21) of patients in 7 days. (2) U8-isoPG:Cr levels in 7 days postoperatively were significantly higher in POCD patients compared with the non-POCD group ( p = 0.01). When measuring change from baseline, U8-isoPG:Cr levels were higher than that of control groups ( p = 0.01). (3) UMDA:Cr levels were significantly elevated in 1 and 2 days postoperatively in both groups ( p < 0.05). U8-isoPG:Cr level seems to be a valuable marker to detect lipid peroxidation early in POCD patients. However, it will also be important to take into account or reduce potential confounders to improve the identification of changes in the status of oxidative stress as a marker for POCD. This review will discuss the concept of reproductive aging, which includes the definition of menopause, its symptoms, and predisposing conditions. It will elaborate upon the contributory factors implicated in the pathogenesis of menopause, focusing most prominently on oxidative stress. Specifically, this paper will explain how oxidative stress, in the form of free radicals and antioxidant deficiencies, has been directly linked to the decline of estrogen during reproductive aging. Additionally, this paper will elaborate upon the treatment options aimed at mitigating the menopausal symptoms and hormonal deficiencies that can lead to various disease processes. Treatment options such as hormonal therapy, antioxidant supplementation, and lifestyle modification have been explored for their effectiveness in treating and preventing the symptoms and sequelae of menopause. Malondialdehyde (MDA) is used for the estimation of damage by reactive oxygen species. MDA is a major reactive aldehyde resulting from the peroxidation of biological membranes. The most common method used to assess MDA production is the thiobarbituric acid (TBARS) assay. However, the value of this method is curbed by low specificity and has been criticized for its use in human studies. In the present study we have used an alternative method for the estimation of MDA production i.e. reaction of MDA with a chromogenic agent 1-methyl-2-phenylindole at 45°C. The paper describes the method of preparing standards for the estimation of MDA (lipid peroxidation) after the treatment with an oxidative stress inducing agent hydrogen peroxide (H2O2). In the present study, the treatments of 1, 5, 10, 20, 50, 100, 150 and 200 μM of H2O2 induced significant increase in lipid peroxidation as compared to the untreated ones. The results suggest that the present method can be used to measure the lipid peroxidation in cultured human peripheral blood lymphocytes and is specific for MDA estimation. [ABSTRACT FROM AUTHOR]

Oxidative stress has been associated with normal aging and Alzheimer's disease (AD). However, little is known about oxidative stress in mild cognitive impairment (MCI) patients who present a high risk for developing AD. The aim of this study was to investigate plasma production of the lipid peroxidation marker, malonaldehyde (MDA) and to determine, in erythrocytes, the enzymatic antioxidant activity of catalase, glutathione peroxidase (GPx), glutathione reductase (GR), and glutathione S-transferase (GST) in 33 individuals with MCI, 29 with mild probable AD and 26 healthy aged subjects. GR/GPx activity ratio was calculated to better assess antioxidant defenses. The relationship between oxidative stress and cognitive performance was also evaluated by the Mini Mental State Examination (MMSE). AD patients showed higher MDA levels than both MCI and healthy elderly subjects. MCI subjects also exhibited higher MDA levels compared to controls. Catalase and GPx activity were similar in MCI and healthy individuals but higher in AD. GR activity was lower in MCI and AD patients than in healthy aged subjects. Additionally, GR/GPx ratio was higher in healthy aged subjects, intermediate in MCI and lower in AD patients. No differences in GST activity were detected among the groups. MMSE was negatively associated with MDA levels (r = -0.31, p = 0.028) and positively correlated with GR/GPx ratio in AD patients (r = 0.68, p < 0.001). MDA levels were also negatively correlated to GR/GPx ratio (r = -0.31, p = 0.029) in the AD group. The aim of our study was to evaluate the markers of oxidative status of erythrocyte during general anesthesia and compare the markers of oxidative status of erythrocyte in both sevoflurane and desflurane. Venous blood samples of patients were collected the following time intervals; initial time (IT) and first hour (1st h), first (1st day) and third days (3rd day) after anesthesia (sevoflurane and Desflurane). The levels of magnesium (Mg), zinc (Zn) as a cofactor of these enzymes, malondialdehyde (MDA) and the activities of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) enzymes were also determined. No significant changes were observed in these measurements when the patients were exposed to desflurane anesthesia. On the other hand, the levels of Zn on erythrocytes were significantly increased at 1st hour and 1st and 3rd days compared to initial time in sevoflurane group (p < 0.01, p < 0.01, and p < 0.05, respectively). The activity of GSH-Px was significantly increased (p = 0.05) while the activity of SOD was significantly decreased (p < 0.01) at 1st hour after administration of sevoflurane compared to the initial time. There were no changes on the levels of Mg and MDA. Our results showed that sevoflurane has more impacts on the antioxidant status of erythrocytes than desflurane. [ABSTRACT FROM PUBLISHER]

Objective: Elderly patients with obstructive sleep apnea/hypopnea syndrome (OSAHS) has a higher risk of cardiovascular and cerebrovascular disease. However, changes of homocysteine (Hcy) as markers of cardiovascular and cerebrovascular disease associated with OSAHS and their mechanism have not been elucidated so far. This study aims to investigate the changes of both serum Hcy and oxidative stress and their possible links with OSAHS in elderly patients. Methods: Based on polysomnogram (PSG) and age, 83 patients with OSAHS were recruited and divided into elderly-OSAHS (n=32) and non-elderly OSAHS groups (n=51). Fifty two subjects without OSAHS were divided into elderly control (n=29) and non-elderly control groups (n=23). A total of 135 subjects were included in the present study. All subjects were recorded for PSG variables and the contents of homocysteine (Hcy), malonaldehyde (MDA), and glutathione (GSH) which were detected after sleep. Serum homocysteine was measured by cyclophorase. MDA and GSH were measured by spectrophotometer. Results: (1) The serum levels of Hcy showed significant difference among the four groups (P<0.05). The concentrations of Hcy in elderly OSAHS patients were higher than in other groups, while those in the elderly control group were higher than in the non-elderly control; the concentrations in the non-elderly OSAHS group were higher than in the non-elderly control. (2) The concentrations of MDA and GSH changed at an equal pace with Hcy in the four groups. (3) Multielement linearity regression analysis indicated a statistically significant relationship between Hcy concentration and age, MDA, GSH, and apnea hypopnea index (AHI). Conclusions: (1) The concentrations of Hcy and oxidative stress have increased with advancing age. (2) The concentrations of Hcy and oxidative stress have further increased in the elderly patients with OSAHS. (3) Oxidative stress might cause high-level serum Hcy in the elderly patients with OSAHS. Many of the biological actions of resveratrol have been attributed to its antioxidant properties. In this work, we subjected human erythrocytes to in vitro oxidative stress by incubating them with tert-butylhydroperoxide (t-BHP). This caused a significant increase in the malondialdehyde (MDA) level and the protein carbonyl group content above the basal values. The presence of trans-resveratrol at micromolar concentrations in the incubation medium protected the erythrocytes from t-BHP-induced oxidative stress, as evidenced by the decrease in the MDA level and the protein carbonyl group content. The effect of resveratrol was concentration and time-dependent. Our findings help to explain some of the beneficial effects of resveratrol. [ABSTRACT FROM AUTHOR]

The article examines the hypothesis that hexavalent chromium Cr(VI)-induced DNA-protein crosslinks (DPCs) could be formed via generation of protein carbonyls and malonaldehyde (MDA) through protein oxidation and lipid peroxidation respectively. Treatment of human leukemic T-lymphocyte MOLT4 cells with potassium chromate induced the formation of protein carbonyls and DPC within 2 h. However, the increased level of MDA only after 4 h in a dose-dependent manner. Chromate treatment of MOLT4 cell homogenates also resulted in augmented formation of MDA and protein carbonyls in a dose-dependent manner. Electron paramagnetic resonance (EPR) spectrometry in combination with spin trapping techniques showed that reaction of Cr(VI) with biological reductants.

Abstract: Conditional double knock-out of presenilin-1 (PS1) and presenilin-2 (PS2) (PS cDKO) in forebrain of mice led to progressive memory dysfunction and forebrain degeneration. These changes in the brain recapitulated most of the neurodegenerative phenotypes of Alzheimer''s disease (AD). Oxidative stress in brain tissues is intimately related to AD. In this report, we examined oxidative stress status in cerebral cortex in 2-, 4- and 7-month PS cDKO and the age- and gender-matched control mice (WT). Lipid peroxidation (MDA as the measure) and protein oxidation (protein carbonyl as the measure) were found to be significantly increased in PS cDKO mice over the age points examined, notably in those at 2-month, suggesting that oxidative stress is an early event in response to PS loss-of-function. The oxidative modification of cortical proteins was further confirmed by Oxyblot assay. The investigations into endogenous antioxidant defense (CAT, SOD and GSH-px as measures) revealed a compensatory defense against oxidative stress, particularly at the early age stage, in PS cDKO mice. The expression level of cortical glial fibrillary acidic protein (GFAP) increased in an age-related manner, in particular in 2-month PS cDKO mice, suggesting that the interaction relationship between oxidative stress and inflammatory response may be closely associated with the underlying loss-of-function pathogenesis of AD. Abstract: Nitric oxide (NO) plays critical role in endothelial dysfunction and oxidative stress in COPD, pointing to the significance of endothelial nitric oxide synthase gene (eNOS) variants. We investigated the association of −786T/C, −922A/G, 4B/4A, and 894G/T polymorphisms of eNOS with the disease and its impact on nitrite and malonaldehyde levels in 190 COPD patients and 134 healthy controls, all smokers. The −786C, −922G and 4A alleles were significantly over-represented in patients (p =0.02, p =0.02, and p =0.03, respectively). The haplotypes, −786C:4A, 4A:894G, −786C:894G, and −786C:4A:894G were significantly over-represented in patients (p <0.0001, p =0.02, p =0.02, and p <0.0001, respectively), whereas, haplotypes, −786T:4B, 4B:894G, −786T:894G, and −786T:4B:894G were significantly under-represented in the patients (p <0.0001). The patients had significantly increased levels of nitrite (p =0.003) and malonaldehyde (p <0.0001). Combination of genotypes containing −786C and 4A alleles were greater in patients (p ⩽0.05), and these combinations associated with decreased FEV1 value and nitrite level (p =0.03 and p =0.04, respectively) and with increased malonaldehyde levels (p =0.02). The eNOS −786C, −922G, and 4A alleles, these alleles associated haplotypes and genotype combinations were over-represented in patients. The variants and their combinations of four polymorphisms of eNOS contribute to disturbed pulmonary function and oxidative stress in COPD. [Copyright &y& Elsevier]

Abstract: The effect of Centella asiatica extract and powder in reducing oxidative stress in SpraqueDawley rats was evaluated. Lipid peroxidation was monitored by measuring malonaldehyde (MDA) level in blood. Activities of free radical-scavenging enzymes (superoxide dismutase and catalase) were determined using H2O2 decomposition and nitrobluetetrazolium reduction, respectively. Results showed that administration of H2O2 (0.1%) in drinking water of the rats, for 25 weeks, increased the malonaldehyde levels in erythrocytes of all the rats. However, rats receiving C. asiatica extract, powder and α-tocopherol had lower MDA levels than did the other rats, which indicates, decrease lipid peroxidation in these rats. Increase in catalase activity of the rats appears to be a response to H2O2 accumulation. The decrease in the activity of superoxide dismutase in C. asiatica- and α-tocopherol supplemented rats suggested a lower requirement for the enzyme and this indicates the protective effect of the plant in combating oxidative stress undergone by the rats. Results revealed that C. asiatica extract and powder may ameliorate H2O2-induced oxidative stress by decreasing lipid peroxidation via alteration of the antioxidant defence system of the rats.
