Anticoagulant rodenticides (AR) remain the most effective chemical substances used to control rodents in order to limit their agricultural and public health damage in both rural and urban environments. The emergence of genetically based resistance to AR worldwide has threatened effective rodent control. This study gives a first overview of the distribution and frequency of single nucleotide polymorphism in the vitamin K epoxide reductase subcomponent 1 (Vkorc1) gene in rodents in Lebanon. In the Mus genus, we detected two missense mutations Leu128Ser and Tyr139Cys, that confer resistance to anticoagulant rodenticides in house mice and a new missense mutation Ala72Val in the Mus macedonicus species, not previously described. In the Rattus genus, we found one missense mutation Leu90Ile in the roof rat and one missense mutation Ser149Ile in the Norway rat. This is the first study to demonstrate potential resistance to AR in Lebanese rodents and therefore it provides data to pest control practitioners to choose the most suitable AR to control rodents in order to keep their efficacy. BACKGROUND: Resistance to rodenticides has been reported globally and poses a considerable problem for efficacy in pest control. The most‐documented resistance to rodenticides in commensal rodents is associated with mutations in the Vkorc1 gene, in particular in codon 139. Resistance to anticoagulant rodenticides has been reported in the Netherlands since 1989. A study from 2013 showed that 25% of 169 Norway rats (Rattus norvegicus) had a mutation at codon 139 of the Vkorc1 gene. To gain insight in the current status of rodenticide resistance amongst R. norvegicus and house mice Mus musculus in the Netherlands, we tested these rodents for mutations in codon 139 of the Vkorc1 gene. In addition, we collected data from pest controllers on their use of rodenticides and experience with rodenticide resistance. RESULTS: A total of 1801 rodent samples were collected throughout the country consisting of 1404 R. norvegicus and 397 M. musculus. In total, 15% of R. norvegicus [95% confidence interval (CI): 13–17%] and 38% of M. musculus (95% CI: 33–43%) carried a genetic mutation at codon 139 of the Vkorc1 gene. CONCLUSION: This study demonstrates genetic mutations at codon 139 of the Vkorc1 gene in M. musculus in the Netherlands. Resistance to anticoagulant rodenticides is present in R. norvegicus and M. musculus in multiple regions in the Netherlands. BACKGROUND We conducted a vitamin K epoxide reductase subcomponent 1 (Vkorc1)‐based nonsynonymous Single Nucleotide Polymorphism (nsSNP) screen with focus on the house mouse (Mus musculus domesticus), but that also considered the Norway rat (Rattus norvegicus) and roof rat (R. rattus) in the USA. RESULTS: We detected six Vkorc1 nsSNPs underlying the amino‐acid polymorphisms Ala21Thr, Trp59Leu, Ile104Val, Val118Leu, Leu128Ser and Tyr139Cys in house mice (average coverage/SNP; n = 182 individuals), two nsSNPs underlying Arg35Pro and Gly46Ser in the Norway rat (n = 93), with the notable absence of Tyr139Cys (n = 179), and one nsSNP underlying Tyr25Phe in the roof rat (n = 27). Inferred resistance frequency is 29.1% for mice (variability of states 0–98.8%), 6.5% (0–33.3%) for the Norway rat, and 39.3% (0–52.6%) for the roof rat based on Tyr25Phe frequencies. CONCLUSIONS: Resistance detected in the USA in the 1980s likely was the consequence of Vkorc1 mutations in mice (Leu128Ser and Tyr139Cys), Norway rats (Arg35Pro) and roof rats (Tyr25Phe). Patterns of variant sharing between the USA and Europe indicate the importance of convergent evolution and gene flow in spreading resistance. The spread of nsSNPs in mice between continents appears to have been more effective than in Norway rats. We hypothesize that Arg35Pro may have originated in Norway rats in the USA, whereas Tyr139Cys variants originated in Europe. Tyr25Phe is the likely cause for resistance in roof rats. Summary: Polymorphisms in the vitamin K 2,3-epoxide reductase subcomponent 1 (vkorc1) of house mice (Mus musculus domesticus) can cause resistance to anticoagulant rodenticides such as warfarin []. Here we show that resistant house mice can also originate from selection on vkorc1 polymorphisms acquired from the Algerian mouse (M. spretus) through introgressive hybridization. We report on a polymorphic introgressed genomic region in European M. m. domesticus that stems from M. spretus, spans >10 Mb on chromosome 7, and includes the molecular target of anticoagulants vkorc1 []. We show that in the laboratory, the homozygous complete vkorc1 allele of M. spretus confers resistance when introgressed into M. m. domesticus. Consistent with selection on the introgressed allele after the introduction of rodenticides in the 1950s, we found signatures of selection in patterns of variation in M. m. domesticus. Furthermore, we detected adaptive protein evolution of vkorc1 in M. spretus (Ka/Ks = 1.54–1.93) resulting in radical amino acid substitutions that apparently cause anticoagulant tolerance in M. spretus as a pleiotropic effect. Thus, positive selection produced an adaptive, divergent, and pleiotropic vkorc1 allele in the donor species, M. spretus, which crossed a species barrier and produced an adaptive polymorphic trait in the recipient species, M. m. domesticus. After the vkorc1 gene was first identified, changes occurring in the gene have been considered one of the main reasons of reduced susceptibility of house mice to anticoagulants. A no-choice feeding test was conducted according to standard EPPO (2004) methodology. Animals were fed for 21 days on baits containing difenacoum 0.005%. All animals were bromadiolone-resistant. Seven months earlier, all test animals had survived a 21-day bromadiolone (0.005%) no-choice feeding test, and sequencing of their vkorc1 gene had revealed the presence of Tyr139Cys and Leu1258Ser/Tyr139Cys VKOR variant. There were no survivors in the no-choice difenacoum feeding test. Consumption was not affected by VKOR variant, sex or genotype. A higher lethal dose was confirmed for Leu128Ser/Tyr139Cys than Tyr139Cys carriers, for females than males, and for homozygotes than heterozygotes. Our research showed that difenacoum 0.005% was effective against house mice carrying the Tyr139Cys VKOR variant, whether it occurred independently or in combination with the Leu128Ser variant in the Vkorc1 gene. Despite the widespread use of anticoagulant rodenticides in baits for controlling commensal rodent pests, their application is problematic due to secondary intoxication and increasing resistance. In contrast to studies on Western European house mice (Mus musculus domesticus), few resistance studies have focused on Eastern European house mice (M. musculus musculus), which have a western distribution boundary in the Czech Republic. This study newly analysed the VKORC1 gene in M. m. musculus field populations from Czech farms and grain stores and identified a nonsynonymous mutation Tyr139Phe. This mutation was common throughout the Czech Republic and was present in 80.2% of the 86 individuals sampled. Additionally, all individuals exhibited a genotype with three synonymous mutations specific to the subspecies M. m. musculus. The functional (mortality–survival) response of the Tyr139Phe mutation was validated in a laboratory choice feeding test using bromadiolone-based bait, where all resistant homozygous individuals survived, while all susceptible mice died, with a mean survival of 6.9 days. The management of invasive species has been greatly enhanced by population genetic analyses of multilocus single-nucleotide polymorphism (SNP) datasets that provide critical information regarding pest population structure, invasion pathways, and reproductive biology. For many applications there is a need for protocols that offer rapid, robust and efficient genotyping on the order of hundreds to thousands of SNPs, that can be tailored to specific study populations and that are scalable for long-term monitoring schemes. Despite its status as a model laboratory species, there are few existing resources for studying wild populations of house mice (Mus musculus spp.) that strike this balance between data density and laboratory efficiency. Here we evaluate the utility of a custom targeted capture genotyping-by-sequencing approach to support research on plaguing house mouse populations in Australia. This approach utilizes 3,651 hybridization capture probes targeting genome-wide SNPs identified from a sample of mice collected in grain-producing regions of southeastern Australia genotyped using a commercially available microarray platform. To assess performance of the custom panel, we genotyped wild caught mice (N = 320) from two adjoining farms and demonstrate the ability to correctly assign individuals to source populations with high confidence (mean >95%), as well as robust kinship inference within sites. Background: The house mouse (Mus musculus) is a globally distributed rodent pest species against which anticoagulant rodenticides are widely used for the protection of human and animal health and the conservation of threatened wildlife. Anticoagulant‐resistant house mice have been known for more than half a century. A house mouse strain was developed in the laboratory that was homozygous resistant for the single nucleotide polymorphism (SNP) Tyrosine139Cysteine (Y139C) and, subsequently, heterozygous resistant animals were produced from this strain by crossing with the homozygous susceptible strain. Results: Using blood clotting response tests, resistance factors at the ED50 level in the homozygous resistant strain for the first‐generation anticoagulants warfarin, chlorophacinone, diphacinone and coumatetralyl were in the range 31.5 to 628.0 for males (M) and 21.6 to 628.0 for females (F), thus indicating that Y139C house mice are substantially resistant to all these substances. Resistance factors at the ED50 level for the homozygous strain generated against the second‐generation compounds were: brodifacoum (M, 1.7; F, 1.9), bromadiolone (M, 16.6; F, 21.0), difenacoum (M, 1.2; F, 2.7), difethialone (M, 1.5; F, 1.5), and flocoumafen (M, 0.9; F, 1.2). Equivalent values for the heterozygous strain were: brodifacoum (M, 1.6; F, 1.4), bromadiolone (M, 5.6; F, 6.5), difenacoum (M, 1.0; F, 1.3), difethialone (M, 1.1; F, 1.1), flocoumafen (M, 0.9; F, 1.1). Conclusion: Y139C SNP homozygous resistant mice are more resistant to anticoagulants than heterozygous resistant animals. House mouse (Mus musculus) is a cosmopolitan pest in agricultural facilities, commodity stores and rural and urban environments. It is regularly controlled by anticoagulant-based baits. Since the number of registered active ingredients is limited, the producers are trying to develop new formulations with increased tamper-resistance, attractiveness and palatability. Reliable and economical methods for laboratory screening/testing are needed. Therefore, this work compared simple no-choice with more economically demanding choice feeding tests for laboratory evaluation of efficacy of rodenticide anticoagulant-based preparations in wild populations of house mouse. We analyzed mice survival and bait intake of the various rodenticide commercial preparations differing in a toxic active substance (warfarin, bromadiolone, brodifacoum, difethialone) and nontoxic food lure components. In most cases, we found insignificant differences in survival of house mice treated with eight test baits within the no-choice and choice feeding tests. Background: Humans routinely attempt to manage pest rodent populations with anticoagulant rodenticides (ARs). We require information on resistance to ARs within rodent populations to have effective eradication programs that minimise exposure in non-target species. Mutations to the VKORC1 gene have been shown to confer resistance in rodents with high proportions of resistance in mice found in all European populations tested. We screened mutations in Mus musculus within Western Australia, by sampling populations from the capital city (Perth) and a remote island (Browse Island). These are the first Australian mouse populations screened for resistance using this method. Additionally, the mitochondrial D-loop of house mice was sequenced to explore population genetic structure, identify the origin of Western Australian mice, and to elucidate whether resistance was linked to certain haplotypes. Results: No resistance-related VKORC1 mutations were detected in either house mouse population. A genetic introgression in the intronic sequence of the VKORC1 gene of Browse Island house mouse was detected which is thought to have originated through hybridisation with the Algerian mouse (Mus spretus). Analysis of the mitochondrial D-loop reported two haplotypes in the house mouse population of Perth, and two haplotypes in the population of Browse Island. Conclusions: Both house mouse populations exhibited no genetic resistance to ARs, in spite of free use of ARs in Western Australia. Therefore weaker anticoagulant rodenticides can be employed in pest control and eradication attempts, which will result in reduced negative impacts on non-target species. The pet trade is one of the most important pathways by which small mammals are introduced to nonnative areas. To prevent the introduction and invasion of non-native pets, an impact assessment protocol is useful in understanding which pets might have potential negative impacts should they escape or be released from captivity. In this study, we used the Generic Impact Scoring System (GISS) to assess the potential effects associated with 24 non-native small mammal species sold in the South African pet trade. European rabbits Oryctolagus cuniculus, house mice Mus musculus, Norwegian rats Rattus norvegicus and eastern grey squirrels Sciurus carolinensis had the highest potential impacts for both socio-economic and environmental categories. We found no statistically significant difference between the overall environmental and socio-economic impact scores. Impacts on agricultural and animal production (livestock) were the main mechanisms in the socio-economic category, while the impacts on animals (predation), competition and hybridisation prevailed for environmental impacts. Small mammals such as rodents can to carry zoonotic pathogens. Currently, there is impaired knowledge on zoonotic pathogens in rodents and insectivores in the Netherlands. This limits opportunities for preventive measures and complicates risk‐assessments for zoonotic transmission to humans. Leptospira spp. and Toxoplasma gondii are present on a list of prioritized emerging pathogens in the Netherlands and were therefore the focus of this study. Both pathogens have the ability to survive under moist environmental conditions. In total, a group of 379 small mammals (rodents & insectivores) were tested on pathogenic Leptospira spp., and 312 on T. gondii. Rodents and insectivores were trapped at various sites, but mostly on pig and dairy farms throughout the country. Over five percent of the animals (5.3%, n = 379) tested positive for Leptospira DNA, and five of the animals (1.6%, n = 312) tested were positive for T. gondii DNA. The animals positive for T.gondii were all brown rats and the ones for Leptospira spp. were various species. Our results show that insectivores and rodents might be used as an indicator for the environmental contamination and/or the contamination in wildlife for Leptospira spp. [ABSTRACT FROM AUTHOR]

Impacts of alien invasive species on island communities and ecosystems may be even more detrimental than on the mainland. Therefore, since the 1950s, hundreds of restoration projects have been implemented worldwide, with the aim of controlling or eradicating alien species from islands. To date, no review has been focused on eradication on Mediterranean islands. To fill the gap, I reviewed the available information concerning mammal eradications so far carried out on Mediterranean islands, examining the details of several aspects of project implementation and monitoring.I obtained data for 139 attempted eradications on 107 Mediterranean islands in eight countries, with Greece, Italy, and Spain accounting for the highest number. Eradication projects targeted 13 mammal species. The black rat Rattus rattus was the target of over 75% of the known attempted eradications in the Mediterranean Basin; other species targeted were feral goat Capra hircus, house mouse Mus musculus, European rabbit Oryctolagus cuniculus, and domestic cat Felis catus. The most widely adopted technique was poisoning (77% of all eradications), followed by trapping (15%) and hunting (4%). However, techniques were largely target‐specific.The average failure rate was about 11%. However, this percentage varied according to the specific mammalian order, and eradications of Carnivora failed more often than those of other mammals. Among rodents, house mouse eradication attained a very high failure rate (75%). Reinvasion occurred after 15% of successful eradications.A better understanding of the motivations of animal rights activists may improve the chance of success when eradicating charismatic or domesticated species. Furthermore, it is crucial to collect data and case studies about reinvasions, in order to strengthen biosecurity programmes following eradication. Mus musculus (house mouse) is a serious rodent pest in poultry farms of Argentina. In these habitats, bromadiolone is the most frequently used anticoagulant. In spite of this control, more than 90% of the farms are infested with rodents. The frequent application of anticoagulants and the permanent presence of M. musculus suggest that there are resistant rodent populations. The goal of this study was to assess bromadiolone susceptibility through a blood clotting response (BCR) test in M. musculus populations inhabiting poultry farms of Exaltación de la Cruz, Buenos Aires, Argentina. Results showed that 56.6% of wild M. musculus were responders to the BCR test. We found non-responder individuals in all the poultry farms studied. Non-responder females had at least one non-responder offspring. Since the percentages of resistant animals per poultry farm were high and considering that bromadiolone is extensively used for rodent control in these habitats, there is a potential risk of an increase in the proportion of resistant individuals and a decrease in the effectiveness of bromadiolone through time in the study area. Mus musculus, Rattus rattus and Rattus norvegicus are pests in poultry farms, causing economic losses and transmitting diseases. Control is commonly conducted through anticoagulant rodenticides, but this control is not effective through time. We aimed to assess the effect of rodent exclosure on long-term success of rodent control in poultry farms of the pampean region, Argentina, and to evaluate indirect estimators of rodent abundance. In both exclosure and non-exclosure sheds rodent abundance decreased significantly after eradication but mice populations showed a recovery, suggesting that the eradication was not complete. Strong-smelling plant extracts, such as essential oils, have a variety of feeding effects on mammals. Considering current concerns over long-term health issues and environmental effects of chemicals, plant-based products with repellent or antifungal activities may represent good solutions for improvement of rodent pest control programs. The present study was therefore focused on examining the effects of bergamot, lavender, and thyme essential oils as additional bait components on daily intakes of cereal-based baits by wild house mice. Lavender essential oil, containing linalool and linalyl acetate as main components, and thyme essential oil with a prevailing thymol component had no effects on house mice diet. Bergamot essential oil, whose main components were linalool, limonene, and linalyl acetate, showed a repellent effect on house mouse diet. [ABSTRACT FROM AUTHOR]

Non-synonymous mutations in the gene encoding vitamin K epoxide reductase (VKORC1) are responsible for resistance to anticoagulant rodenticides in rodents. Detection of resistant individuals is crucial to implement effective management plans and guarantee the success of eradication campaigns. Resistant individuals of the house mouse Mus musculus have been reported in several European countries and in North America. However, to date no data is available for M. musculus domesticus in Italy. In this study, we sequenced the three exons of the VKORC1 gene in 30 mice from the Pontine Archipelago, currently subject to an eradication campaign of invasive rodents. We recorded the presence of a mutation known to confer resistance to rodenticides (Tyr139Cys) in one individual (3.3% of the total samples set). BACKGROUND After reports of management problems in practice, a survey was conducted to determine the presence of bromadiolone-resistant animals in different house mouse ( Mus musculus L.) populations in Serbia. A 21 day no-choice feeding test was carried out to examine the resistance of house mice to bromadiolone. Eighty house mice collected from four locations (ten males and ten females per location) were tested for bromadiolone tolerance. Surviving animals and their F1 offspring were screened for mutations. The influence of VKOR variant, zygosity and sex on bromadiolone tolerance was analysed. RESULTS Bait intake and changes in body weight revealed different animal responses regarding susceptibility or resistance. Leu128Ser, Tyr139Cys and a new Ala21Thr polymorphism were detected in wild-born survivors and their F1 generation. However, not every individual with the polymorphisms Leu128Ser and Tyr139Cys survived the feeding test. VKOR variants and sex caused variations in bromadiolone tolerance. CONCLUSION For the first time it was shown that the VKOR variant, along with sex, is responsible for bromadiolone tolerance in house mice. Other factors influencing bromadiolone tolerance, including sex-specific factors, cannot be excluded. The competence of a species for a zoonotic disease can be broadly defined as its ability to cause new infections, thereby increasing the fitness of the causative parasite (Merrill and Johnson 2020). Although scant evidence from amphibian disease systems suggests that evolved tolerance to pesticides could modify disease competence (Hua et al. 2017), less is known about zoonotic disease systems from mammals. Although chemical contaminants can alter disease competence (e.g., pesticides via suppressed immunity), there has been less focus on whether adaptive responses to contaminants can modify competence. The article presents a study which examines the susceptibility of bromadiolone in wild and laboratory Mus musculus L. or house mice in Buenos Aires, Argentina. It involves animals that were captured in poultry farms and a laboratory strain and were fed with bromadiolone. It concludes that there is a presence of anticoagulant resistant Mus musculus L. which may be influenced by the feeding behaviour of animals.

Resistance to anticoagulant rodenticides in Rattus norvegicus is now estimated to cover an area of roughly 26 000 km2 in northwest Germany. The chemical compounds involved are warfarin, coumatetralyl, bromadiolone and, to a lesser extent, difenacoum. Recent investigations revealed that the resistance area is extending considerably further east than was previously recorded. The easternmost populations containing resistant individuals are now present in the city of Hannover, whereas it seems that on the southern edge the urbanised Ruhr Basin remains mostly unaffected by resistance problems. Reported control failures with house mice (Mus musculus/domesticus) suggest that resistance to high potency compounds may occur, but these await scientific confirmation. We propose that molecular genetic testing methodology should be exploited to improve monitoring of anticoagulant resistance and provide more accurate information to users of rodenticides. In populations that are distributed across steep environmental gradients, the potential for local adaptation is largely determined by the spatial scale of fitness variation relative to dispersal distance. Since altitudinal gradients are generally characterized by dramatic ecological transitions over relatively short linear distances, adaptive divergence across such gradients will typically require especially strong selection to counterbalance the homogenizing effect of gene flow. Here we report the results of a study that was designed to test for evidence of adaptive divergence across an altitudinal gradient in a natural population of deer mice, Peromyscus maniculatus. We conducted a multilocus survey of allozyme variation across a steep altitudinal gradient in the southern Rocky Mountains that spanned several distinct biomes, from prairie grassland to alpine tundra. As a control for the effects of altitude, we also surveyed the same loci in mice sampled along a latitudinal transect through the prairie grassland that ran perpendicular to the east-west transect. We used a coalescent-based simulation model to identify loci that deviated from neutral expectations, and we then assessed whether locus-specific patterns of variation were nonrandom with respect to altitude. Results indicated that the albumin locus (Alb) reflects a history of diversifying selection across the altitudinal gradient. This conclusion is supported by two main lines of evidence: (1) Alb was characterized by levels of divergence across the altitudinal transect that exceeded neutral expectations in two consecutive years of sampling (in contrast to the spatial pattern of variation across the latitudinal transect), and (2) levels of divergence at the Alb locus exhibited a positive association with altitudinal distance in both years (in contrast to the pattern observed at unlinked loci). Simple Summary: The presence of pest rodents around food production and storage sites is one of many underlying problems contributing to food contamination and loss, particularly influencing food and nutrition security in low-income countries. By reducing harvest losses by rodents, millions of food-insecure people would benefit. As there are limited data on post-harvest rice losses due to rodents, our objectives were to assess stored rice losses in local households and two rice milling factories in Bangladesh. We also wanted to monitor the effect of different rodent control strategies on stored rice losses over a period of two years (2016 and 2017). Four control strategies were tested, (i) untreated control, (ii) use of domestic cats, (iii) use of rodenticides, (iv) use of snap-traps. In total, 210 rodents were captured from inside people's homes, with Rattus rattus trapped most often, followed by Mus musculus and Bandicota bengalensis. In the milling stations, 68 rodents were trapped, of which 21 were M. musculus, 19 R. rattus, 17 B. bengalensis, 8 Rattus exulans, and 3 Mus terricolor. In 2016, losses from standardised baskets of rice within households were between 13.6% and 16.7%. In 2017, the losses were lower, ranging from 0.6% to 2.2%. Daily rodent removal by trapping proved to be most effective to diminish stored produce loss. The effectiveness of domestic cats was limited.
