Residue of sulfamethazine (SMZ), a typical short-acting drug to prevent bacterial infections, in food is a threat to human health. A ternary heterogeneous metal-organic framework hybrid (Zn/Fe-MOF@PDANSs) of Zn-TCPP-MOF, MIL-101 (Fe) and polydopamine nanoparticles (PDANSs) was proposed to establish an aptasensor for the sensitive and selective detection of SMZ. In this sensor, Zn-TCPP-MOF and FAM emitted fluorescence at 609 nm and 523 nm, respectively, and the fluorescence of FAM-ssDNA could be quenched when it was adsorbed on the surface of MOF hybrid. In the presence of SMZ, the fluorescence of FAM-ssDNA recovered due to the dropping from MOF hybrid, while the fluorescence of MOF hybrid remained. With this strategy, a wide concentration range and high sensitivity of SMZ were detection. And the ternary Zn/Fe-MOF@PDANSs sensor exhibited more excellent performance than binary Zn/Fe-MOF aptasensor. The combination of carbon dots (CDs) with covalent organic frameworks (COFs) was used to design an innovative sensor based on fluorescence resonance energy transfer (FRET) for the detection of Escherichia coli O157:H7 (E. coli O157:H7) in food samples. Carbon dots were used as fluorescence donors, covalent organic frameworks as fluorescence acceptors. The antibody (Ab) specific to E. coli O157:H7 was used to form a CD-Ab-COF immunosensor by linking CDs and COFs. The antibody was specifically bound with E. coli O157:H7, which caused the connection between CDs and COFs to be interrupted, and the carbon dots exhibited fluorescence restoration. The sensor exhibited a linear detection range spanning from 0 to 106 CFU/mL, with the limit of detection (LOD) of 7 CFU/mL. Ferroptosis is a novel type of nonapoptotic programmed cell death involving the accumulation of lipid peroxidation (LPO) to a lethal threshold. Herein, we propose tunable zeolitic imidazolate framework (ZIFs)-engineered biodegradable nanozymes for ferroptosis mediated by both reactive oxygen species (ROS) and nitrogen species (RNS). l-Arginine is utilized as an exogenous nitric oxide donor and loaded into hollow ZIFs@MnO 2 artificial nanozymes, which are formed by etching ZIFs with potassium permanganate and simultaneously generating a MnO 2 shell in situ . The constructed nanozymes with multienzyme-like activities including peroxidase, oxidase, and catalase can release satisfactory ROS and RNS through a cascade reaction, consequently promoting the accumulation of LPO. Furthermore, it can improve the efficiency of ferroptosis through a three-step strategy of glutathione (GSH) depletion; that is, the outer MnO 2 layer consumes GSH under slightly acidic conditions and RNS downregulates SLC7A11 and glutathione reductase, thus directly inhibiting GSH biosynthesis and indirectly preventing GSH regeneration.

A novel ratiometric sensor using an electrochemically active metal-organic framework of Mo@MOF-808 and NH 2 -UiO-66 as response signals was developed to detect tetracycline (TET) in ultratrace quantities. To achieve the dual-response strategy, Mo@MOF-808, with a reduction peak at -1.06 V, and NH 2 -UiO-66, with an oxidation peak at 0.724 V, were used as signal probes directly. Concretely, Mo@MOF-808, single-stranded DNA (ssDNA), and complex system (Apt@NH 2 -UiO-66) of aptamer (Apt) and NH 2 -UiO-66 were sequentially immobilized on the electrode. With the addition of TET, Apt was hybridized with TET and Apt@NH 2 -UiO-66 was detached from the electrode, resulting in an increase in the current at -1.06 V and a decrease in the current at 0.724 V. Through this strategy, the sensor achieved a wide linear range (0.1-10000 nM) and a low limit of detection (0.009792 nM) for TET. Moreover, the ratiometric sensor exhibited better sensitivity, reproducibility, and stability than a single-signal sensor. Furthermore, the constructed sensor was successfully applied to detect TET in milk samples, suggesting excellent application prospects.

A novel ratiometric fluorescence sensor based on lanthanide-functionalized metal-organic frameworks (Ag + /Eu 3+ @UiO-66(COOH) 2 , AEUC) with intrinsic dual-emitting bands was fabricated to determine tetracycline (TC) residues by exploiting (UiO-66-(COOH) 2 , UC) as reference units, Eu 3+ as recognition units and Ag + as fluorescence enhancer. Benefiting from specific binding sites and functional adsorbent channels of AEUC, efficient capture and specific recognize toward TC could be achieved, remarkably enhancing the reliability. The synergistic effect of inner filter effect (IFE) and photoinduced electron transfer (PET) process between TC and AEUC was verified, enabling the detection limit and quantification limit of established strategy reaching as 12.8 nmol/L and 38.6 nmol/L and conspicuous fluorescence color gradation from red to blue. From the point of view of the ecological environment, contaminants such as heavy metal ions or toxic gases have caused harmful impacts on the environment and human health, and overcoming these adverse effects remains a serious and important task. Very recent, highly crystalline porous metal-organic frameworks (MOFs), with tailorable chemistry and excellent chemical stability, have shown promising properties in the field of removing various hazardous pollutants. This review concentrates on the recent progress of MOFs and MOF-based materials and their exploit in environmental applications, mainly including water treatment and gas storage and separation. A simple fluorescence and electrochemical dual-channel biosensor based on bifunctional Zr(IV)-based metal-organic framework (Zr-MOF) was proposed to detect Ochratoxin A (OTA). The bifunctional Zr-MOF, with photoluminescence properties and enormous electroactive ligands, was exploited to load OTA-specific aptamers for designing signal probes, greatly simplifying the probe-fabrication process and improving sensing reliability. Upon specific recognition of aptamer toward OTA, the anchored probe was released from the sensing interface into the reaction solution. In this circumstance, the increased amount of the signal probe in reaction solution led to an enhanced fluorescence response, while the decreased amount of the signal probe on the sensing interface resulted in a diminished electrochemical response. According to the dual-channel signal change with increasing OTA concentration, the visual fluorescence strategy was established for intuitive OTA detection, and meanwhile, sensitive electrochemical assay with a detection limit of 0.024 pg/mL was also achieved with the help of one-step electrodeposition as a sensing platform. Moreover, the proposed dual-channel assay has been successfully applied to determine OTA levels in corn samples with rapid response, superior accuracy, and high anti-interference capability, providing a promising method for food safety monitoring.

The discovery of CRISPR/Cas9 genome-editing technology enables the precise manipulation of mammalian DNA sequences for treating genetic disorders. Despite its high efficiency for genome editing, the introduction of CRISPR/Cas9 machinery, which is composed of Cas9 nuclease protein and guide RNA, into cells challenges its clinical translation potential. Therefore, the intracellular delivery of genome-editing machinery determines the efficacy of gene manipulation via the CRISPR/Cas9 technology. Recently, metallosupramolecules including metal-organic frameworks (MOFs) and metal-organic cages (MOCs) have been designed to selfassemble with Cas9 nuclease and guide RNA for CRISPR/Cas9 delivery and genome editing. Herein, we review the most recent advances and strategies of constructing metallosupramolecules for CRISPR/Cas9 delivery. In particular, we discuss nanoscale MOFs and MOCs that could be assembled and regulated by the intracellular environment for the spatiotemporal delivery of genome editing machinery. We also provide a perspective view of the future development of metallosupramolecules for genome editing and gene therapy in vivo .

Morphological control of metal-organic frameworks (MOFs) at the micro/nanoscopic scale is critical for optimizing the electrochemical properties of them and their derivatives. In this study, manganese organic phosphate (Mn-MOP) with three distinct two-dimensional (2D) morphologies was synthesized by varying the molar ratio of Mn2+ to phenyl phosphonic acid, and one of the morphologies is a unique palm leaf shape. In addition, a series of 2D Mn-MOP derivatives were obtained by calcination in air at different temperatures. Electrochemical studies showed that 2D Mn-MOP derivative calcined at 550 °C and exhibited a superior specific capacitance of 230.9 F g−1 at 0.5 A g−1 in 3 M KOH electrolyte. The aqueous asymmetric supercapacitor and the constructed flexible solid-state device demonstrated excellent rate performance. Based on 3‐(2,4‐dicarboxylphenyl)‐6‐carboxylpyridine ligand (H3dpcp), a lanthanide metal–organic framework with the porous three‐dimensional structure was successfully synthesized, namely, [Eu (dpcp)(H2O)]n (1). The experimental results indicated that complex 1 exhibited stable luminescence in the range of pH = 2–11 in aqueous solution. It is noteworthy that complex 1 could recognize tryptophan (Trp) and trans, trans‐muconic acid (tt‐MA) molecules through the fluorescence quenching effect, and showed high sensitivity, selectivity, recoverability, anti‐interference, and low detection limits for the identification of Trp and tt‐MA in aqueous solution. In this study, we propose a versatile method for synthesizing uniform three‐dimensional (3D) metal carbides, nitrides, and carbonitrides (MXenes)/metal–organic frameworks (MOFs) composites (Ti3C2TX/Cu‐BTC, Ti3C2TX/Fe,Co‐PBA, Ti3C2TX/ZIF‐8, and Ti3C2TX/ZIF‐67) that combine the advantages of MOFs and MXenes to enhance stability and improve conductivity. Subsequently, 3D hollow Ti3C2TX/ZIF‐67/CoV2O6 composites with excellent electron‐ and ion‐transport properties derived from Ti3C2TX/ZIF‐67 were synthesized. The specific capacitance of the Ti3C2TX/ZIF‐67/CoV2O6 electrode was 285.5 F g−1, which is much higher than that of the ZIF‐67 and Ti3C2TX/ZIF‐67 electrode. This study opens a new avenue for the design and synthesis of MXene/MOF composites and complex hollow structures with tailorable structures and compositions for various applications. Herein, a ratiometric fluorescent probe was developed for sensitive detection of salicylic acid (SA) in rice using silk-derived carbon quantum dots @ Curcumin @ iron-based metal organic framework (SCQDs@Cur@Fe-MOFs). Fe-MOFs with porous structure not only provided holes for SCQDs to evade self-aggregation of SCQDs, but Fe2+ ions from MOFs was ingeniously employed to capture active sites of Cur, solving the problem of lacking sufficient specificity of Cur to SA while converting weak response signal to amplified "turn on" mode. Upon exposed to SA, the probe interacted with SA to form Cur-Fe2+-SA ternary complex, which inhibited the internal filtration effect between Cur and SCQDs, and triggered a cascade of response signaling. With this strategy, the proposed probe achieved sensitive determination of salicylic acid in rice with detection limit as low as 0.14 μmol/L. In this work, PtPd nanoparticles functionalized porphyrin metal-organic framework nanoenzymes (PtPd@PCN-224 nanoenzymes) are exploited as signal amplification tags to fabricate a rapid and ultrasensitive sensitive CRISPR/Cas14a-based electrochemical biosensor for Burkholderia pseudomallei (B. pseudomallei) specific DNA sequences detection. The prepared PtPd@PCN-224 nanoenzymes not only catalyze the reduction peak current of H 2 O 2 to obtain a strong electrochemical signal output, but also provide massive active sites for the assembly of nucleic acids by Zr–O–P bonds. Besides, the designed target-activated CRISPR/Cas14a is able to recognize the target DNA sequences and further trigger the trans -cleavage of ssDNA for signal amplification. Benefiting from the target-activated CRISPR/Cas14a and PtPd@PCN-224 nanoenzymes, the developed electrochemical biosensor for B. pseudomallei DNA detection exhibits high sensitivity with detection of limit down to 12.8 aM and excellent specificity for distinguishing non-targeted bacteria. Moreover, the CRISPR/Cas14a-based electrochemical detection platform can also apply for other pathogenic bacteria diagnostic by well-designing sgRNA for target sequence recognition, possessing high flexibility and versatility in clinical diagnosis. This work presents an electrochemical device based on a composite modification of amine functionalized Zr(IV) metal–organic framework (UiO-66-NH 2) and multi-walled carbon nanotubes (MWCNTs) for voltammetry determination of cadmium ions (Cd2+). The UiO-66-NH 2 @MWCNTs composites were prepared by one-pot hydrothermal reaction. The prepared sensor performs excellent performance, which was attributed to the synergism between UiO-66-NH 2 with a special octahedral structure and enlarged surface area and MWCNTs with outstanding conductivity. Under optimal experiment condition, the fabricated sensor showed good linear relationship from 0.5 to 170 μg/L, with a detection limit of 0.2 μg/L. Finally, the sensor was successfully applied to detect Cd2+ in meat samples (N = 21) with relative standard deviation (RSD) lower than 4.5% and recovery of 95.1–107.5%, and the results were compared with certified method, there was no statistical significance difference between the developed sensor and certified method at a 95% confidence level.
