PURPOSE: Cancer survivors frequently report significant forgetfulness, but standard neuropsychological tests often fail to detect primary memory deficits. Past research has suggested survivors may misattribute forgetfulness to memory decay rather than impairments in initial encoding, but no studies have tested whether this pattern is evident in older survivors, who are more vulnerable to age-related memory difficulties. We examined whether long-term breast cancer survivors treated in older adulthood demonstrate deficits in initial encoding, as opposed to increased rates of memory decay, relative to non-cancer controls. METHODS: Three hundred twenty-eight breast cancer survivors age 60 and above, 5–15 years post-treatment, and 162 age-matched non-cancer controls completed list learning and narrative memory assessments at four time-points over 2 years. Performance on learning trials and delayed recall was analyzed at each time-point to assess group differences in memory encoding, and memory decay was assessed by analyzing changes in performance across delays. RESULTS: Univariate ANCOVAs correcting for age and education showed that survivors had worse initial encoding performance across multiple time-points, which were compensated for with multiple learning trials to produce recall performance comparable to controls. There were no significant group differences in memory decay. CONCLUSIONS: Older long-term breast cancer survivors exhibit a consistent pattern of initial encoding deficits, but memory retention was comparable to controls. Future research should consider the role of encoding deficits and age-related factors when evaluating cognitive function in older survivors. IMPLICATIONS FOR CANCER SURVIVORS: Commonly reported memory problems may stem from encoding deficits in older long-term cancer survivors.

From early to middle childhood, brain regions that underlie memory consolidation undergo profound maturational changes. However, there is little empirical investigation that directly relates age-related differences in brain structural measures to the memory consolidation processes. The present study examined system-level memory consolidations of intentionally studied object-location associations after one night of sleep (short delay) and after two weeks (long delay) in normally developing 5-to-7-year-old children (n = 50) and young adults (n = 39). Behavioural differences in memory consolidation were related to structural brain measures. Our results showed that children, in comparison to young adults, consolidate correctly learnt object-location associations less robustly over short and long delay. Moreover, using partial least squares correlation method, a unique multivariate profile comprised of specific neocortical (prefrontal, parietal, and occipital), cerebellar, and hippocampal subfield structures was found to be associated with variation in short-delay memory consolidation. A different multivariate profile comprised of a reduced set of brain structures, mainly consisting of neocortical (prefrontal, parietal, and occipital), and selective hippocampal subfield structures (CA1-2 and subiculum) was associated with variation in long-delay memory consolidation. BACKGROUND This study aimed to examine the effectiveness of microsurgery practice during an interval in robotic surgical training. METHODS Two participant groups with 30 trainees each practiced Thread the Rings 1 on a dV-Trainer. There were four 2-h training sessions, with a 72-h interval between each session. Group A received no additional training during the study period. During every interval, Group B practiced a similar 2-h ring penetration exercise using a microscope three times (once every 24 h). RESULTS The 72-h delay between two sessions for Group A caused the forgetting effect. For Group B, additional microscope-based training aided memory retention. After the training session, Group B presented a significantly higher score than Group A in Thread the Ring 1 learning curves. CONCLUSIONS Adding microsurgical skill training into the basic robotic-surgery training curriculum could be helpful because additional trainings with a binocular microscope could improve robotic surgical skill levels.

In infancy, sleep occurs in multiple nap and overnight bouts that change developmentally in quantity and distribution. Though studies suggest that infant memory benefits from a single nap, no work has assessed the relative benefits of different naps (morning vs. afternoon), nor how multiple naps support memory across the day. We investigated the memory benefit of a morning nap, relative to morning wake, and the effect of these intervals on afternoon nap function in 9-month-olds (n = 15). Infants participated in two within-subjects conditions (separated by 1-2 weeks). In the Nap-Nap condition, infants took their morning and afternoon naps; in the Wake-Nap condition, infants were kept awake during morning naptime, but napped unrestricted in the afternoon. Before each nap/wake interval, infants completed an imitation memory task, with memory assessed again shortly after the nap/wake interval. In the Nap-Nap condition, infants showed memory retention across morning and afternoon naps. In contrast, infants tended to forget items learned across morning wake in the Wake-Nap condition. Moreover, morning wake was associated with a significant decline in post-nap retention of items learned in the afternoon. Furthermore, relations between nap slow-wave activity (SWA) and memory varied across naps, with SWA either not predicting (morning naps) or positively predicting (afternoon naps) memory change in the Nap-Nap condition, but negatively predicting afternoon memory change in the Wake-Nap condition. We conclude that two naps per day (rather than one) aids memory at 9 months, and that skipping the morning nap may moderate relations between afternoon nap physiology and memory.

(1) Background: An age-related cognitive decline is commonly affecting the life of elderly with symptoms involved in progressive impairments to memory and learning. It has been proposed that probiotics could modulate age-related neurological disorders via the gut–brain axis. (2) Methods: To investigate the anti-aging effect of probiotic Lactobacillus plantarum GKM3, both survival tests and cognitive experiments were conducted in the SAMP8 mice model. The six-month-old SAMP8 (n = 20 in each gender) were fed with probiotic GKM3 at a dosage of 5.1 × 109 and 1.0 × 109 cfu/ kg B.W./day until their natural death. Then, the life span was investigated. Three-month-old SAMP8 (n = 10 in each gender) were administered GKM3 for 14 weeks. Then, the behavior tests and oxidation parameters were recorded. (3) Results: GKM3 groups showed significantly increased latency in the passive avoidance test and time of successful avoidance in the active avoidance test. The TBARS and 8-OHdG from mice brains also showed a significant reduction in the groups treated with GKM3. In addition, lower accumulation of the amyloid-β protein was found in SAMP8 mice brains with the supplement of GKM3. (4) Conclusions: These results indicated that L. plantarum GKM3 delayed the process of aging, alleviated age-related cognitive impairment, and reduced oxidative stress.

It remains unknown whether transcranial alternating current stimulation (tACS) affects episodic memory and the effect of gamma oscillations delivered to the left prefrontal cortex (PFC) on long-term memory retention has not been fully investigated. We examined whether tACS over the left PFC enhances recognition of episodic memory. The study enrolled 36 healthy young adult volunteers. The participants were randomly assigned to either a tACS group [n = 18; 14 females; mean age ± standard deviation (SD): 21.2 ± 0.4 years] or a sham-control group [n = 18; 14 females; mean age ± SD: 21.2 ± 0.4 years]. Participants received either tACS or sham stimulation both during the learning task that was conducted on day 1 and during a recognition task on day 2. The recognition task was also conducted on days 1 and 7, and response accuracy was measured at all three time points (days 1, 2, and 7). Patients in the tACS group were better able to retain long-term memory than those in the sham-control group. These findings suggest that tACS over the left PFC enhances recognition of episodic memory in healthy young adults.

When listening, familiarity with an attended talker’s voice improves speech comprehension. Here, we instead investigated the effect of familiarity with a distracting talker. In an irrelevant-speech task, we assessed listeners’ working memory for the serial order of spoken digits when a task-irrelevant, distracting sentence was produced by either a familiar or an unfamiliar talker (with rare omissions of the task-irrelevant sentence). We tested two groups of listeners using the same experimental procedure. The first group were undergraduate psychology students (N=66) who had attended an introductory statistics course. Critically, each student had been taught by one of two course instructors, whose voices served as familiar and unfamiliar task-irrelevant talkers. The second group of listeners were family members and friends (N=20) who had known either one of the two talkers for more than ten years. Students, but not family members and friends, made more errors when the task-irrelevant talker was familiar versus unfamiliar. Interestingly, the effect of talker familiarity was not modulated by the presence of task-irrelevant speech: students experienced stronger working-memory disruption by a familiar talker irrespective of whether they heard a task-irrelevant sentence during memory retention or merely expected it. While previous work has shown that familiarity with an attended talker benefits speech comprehension, our findings indicate that familiarity with an ignored talker deteriorates working memory for target speech. The absence of this effect in family members and friends suggests that the degree of familiarity modulates memory disruption.

Auditory closed-loop stimulation has gained traction in recent years as a means of enhancing slow oscillatory activity and, consequently, sleep-associated memory consolidation. Previous studies on this topic have primarily focused on the consolidation of semantically-congruent associations. In this study, we investigated the effect of auditory closed-loop stimulation on the overnight retention of semantically-incongruent associations. Twelve healthy males (age: M = 20.06, SD = 2.02 years) participated in two experimental conditions (simulation and sham). In the stimulation condition, clicks were delivered in phase with slow oscillation up-states, whereas in the sham condition no auditory stimuli were applied. Corroborating earlier work, stimulation (vs. sham) enhanced the slow oscillation rhythm, phase-coupled spindle activity and slow oscillation power. However, there was no benefit of stimulation on overnight memory retention. These findings suggest that closed-loop stimulation does not benefit semantically-incongruent associations.

Memory complaints are frequently reported by patients with epilepsy and are associated with seizure occurrence. Yet, the direct effects of seizures on memory retention are difficult to assess given their unpredictability. Furthermore, previous investigations have predominantly assessed declarative memory. This study evaluated within-subject effects of seizure occurrence on retention and consolidation of a procedural motor sequence learning task in patients with epilepsy undergoing continuous monitoring for five consecutive days. Of the total sample of patients considered for analyses (N = 53, Mage = 32.92 ± 13.80 y, range = 18–66 y; 43% male), 15 patients experienced seizures and were used for within-patient analyses. Within-patient contrasts showed general improvements over seizure-free (day + night) and seizure-affected retention periods. Yet, exploratory within-subject contrasts for patients diagnosed with temporal lobe epilepsy (n = 10) showed that only seizure-free retention periods resulted in significant improvements, as no performance changes were observed following seizure-affected retention. These results indicate general performance improvements and offline consolidation of procedural memory during the day and night. Furthermore, these results suggest the relevance of healthy temporal lobe functioning for successful consolidation of procedural information, as well as the importance of seizure control for effective retention and consolidation of procedural memory.

Senescence-accelerated mouse prone 10 (SAMP10) exhibits cerebral atrophy and depression-like behavior. A line of SAMP10 with spontaneous mutation in the Slc5a2 gene encoding the sodium-glucose cotransporter (SGLT) 2 was named SAMP10/TaSlc-Slc5a2slc (SAMP10-&Delta, Sglt2) and was identified as a renal diabetes model. In contrast, a line of SAMP10 with no mutation in SGLT2 (SAMP10/TaIdrSlc, SAMP10(+)) was recently established under a specific pathogen-free condition. Here, we examined the mutation effect in SGLT2 on brain function and longevity. No differences were found in the survival curve, depression-like behavior, and age-related brain atrophy between SAMP10-&Delta, Sglt2 and SAMP10(+). However, memory retention was lower in SAMP10-&Delta, Sglt2 mice than SAMP10(+). Amyloid beta (A4) precursor-like protein 1 (Aplp1) expression was significantly lower in the hippocampus of SAMP10-&Delta, SGLT2 than in SAMP10(+) at 2 months of age, but was similar at 12 months of age. CaM kinase-like vesicle association (Camkv) expression was remarkably lower in SAMP10(+). These genes have been reported to be involved in dendrite function. Amyloid precursor proteins have been reported to involve in maintaining homeostasis of glucose and insulin. These results suggest that mutation in SGLT2 results in down-regulation of Aplp1 in young age, which can lead to poor memory retention in old age.

We recently reported that constitutive ablation of cyclophilin-D (Cyp-D) in mice reduces oxygen consumption (VO2) while paradoxically increasing exercise endurance, thereby demonstrating increased O2 utilization efficiency. This response was associated with augmented glucose uptake and glucose utilization, in part mediated through adenosine monophosphate-activated kinase signaling. We now hypothesized that Cyp-D knock-out (KO) mice might also exhibit improved cognitive-behavioral performance and that these favorable adaptive responses may persist under hypoxic conditions. We therefore assessed under normoxic (20.9% O2, simulating ground O2 levels) and hypoxic (8% O2, simulating 7600 m altitude O2 levels) conditions exercise capacity and cognitive-behavioral performance. We used a treadmill test to assess exercise capacity, a pole-test to assess agility, an elevated-plus-maze test to assess anti-anxiety, and a passive avoidance test to assess learning and memory retention. Compared to wild type, Cyp-D KO mice showed comparable treadmill work under normoxia (48 ± 12 vs 47 ± 9 Joules) but increased treadmill work (12 ± 1 vs 8 ± 1 Joules; p = 0.02) under hypoxia. Cyp-D KO mice displayed increased pole-descending time (17 ± 3 vs 8 ± 2 s; p ≤ 0.05) under normoxia but shorter pole-descending time (21 ± 3 vs 37 ± 4 s; p ≤ 0.01) under hypoxia. In addition, the Cyp-D KO mice demonstrated increased elevated plus-maze open arm time (91 ± 31 vs 23 ± 12 s; p ≤ 0.05) under hypoxia and increased latency to enter dark chamber (261 ± 23 vs 185 ± 42 s; p ≤ 0.05) under normoxia. Thus, our experiments showed that under normoxia Cyp-D KO mice displayed anti-anxiety behavior and improved learning and memory retention. Under hypoxia, Cyp-D KO mice displayed increased exercise capacity, increased agility, and increased anti-anxiety consistent with our previously reported findings of increased O2 utilization efficiency. Identifying interventions to elicit these effects could be beneficial in a myriad of physiological and clinical conditions in which increasing O2 utilization efficiency would be advantageous.

Sleep is recognized as a physiological state associated with learning, with studies showing that knowledge acquisition improves with naps. Little work has examined sleep-dependent learning in people with developmental disorders, for whom sleep quality is often impaired. We examined the effect of natural, in-home naps on word learning in typical young children and children with Down syndrome (DS). Despite similar immediate memory retention, naps benefitted memory performance in typical children but hindered performance in children with DS, who retained less when tested after a nap, but were more accurate after a wake interval. These effects of napping persisted 24 h later in both groups, even after an intervening overnight period of sleep. During naps in typical children, memory retention for object-label associations correlated positively with percent of time in rapid eye movement (REM) sleep. However, in children with DS, a population with reduced REM, learning was impaired, but only after the nap. This finding shows that a nap can increase memory loss in a subpopulation, highlighting that naps are not universally beneficial. Further, in healthy preschooler’s naps, processes in REM sleep may benefit learning.

Evidence primarily exists in adults that engaging in task-related mental activity after new learning results in increased forgetting of learned information, compared with quietly resting in the minutes that follow learning, where less forgetting is observed. The current study investigated whether the beneficial effect of post-encoding rest can be observed in children aged 13-14 years. Each child (N = 102) encoded two word lists. After the presentation and immediate recall of one word list, children wakefully rested for 10 min (resting condition), after presentation and immediate recall of the other word list, they solved visuo-spatial problems for 10 min (problem-solving condition). Seven days later, a surprise free recall test for the two word lists took place. Our results showed that children retained more words over 7 days in the resting condition than with the problem-solving condition. Post-hoc analyses revealed that the resting effect was a function of the number of words recollected during the immediate recall. Specifically, those children who recalled fewest words (≤ 13/30 words) in the immediate recall showed a significant resting effect. There was no resting effect in those who recalled a mid-range (14-16/30 words) or a high number (>16/30 words) of words. These results provide new insights into the factors that influence memory in children, and suggest that a few minutes of wakeful rest benefits memory, relative to engaging in an ongoing task. Statement of contribution What is already known on this subject? Task-related mental activity after encoding weakens memory retention more than wakeful resting. Beneficial effect of resting after encoding was found primarily in younger and older adults. What does this study add? We investigated children at the age of 13-14 years. 8-min post-encoding wakeful resting supports memory retention over 7 days. Individuals differ in the impact of a brief period of wakeful resting after learning. Only children with lower immediate memory performances profited from wakeful resting.

Background High-fidelity simulation is known to improve participant learning and behavioral performance. Simulation scenarios generate stress that affects memory retention and may impact future performance. The authors hypothesized that more participants would recall three or more critical key messages at three months when a relaxation break was performed before debriefing of critical event scenarios. Methods Each resident actively participated in one scenario and observed another. Residents were randomized in two parallel-arms. The intervention was a 5-min standardized relaxation break immediately before debriefing; controls had no break before debriefing. Five scenario-specific messages were read aloud by instructors during debriefings. Residents were asked by telephone three months later to recall the five messages from their two scenarios, and were scored for each scenario by blinded investigators. The primary endpoint was the number of residents participating actively who recalled three or more messages. Secondary endpoints included: number of residents observing who recalled three or more messages, anxiety level, and debriefing quality. Results In total, 149 residents were randomized and included. There were 52 of 73 (71%) residents participating actively who recalled three or more messages at three months in the intervention group versus 35 of 76 (46%) among controls (difference: 25% [95% CI, 10 to 40%], P = 0.004). No significant difference was found between groups for observers, anxiety or debriefing quality. Conclusions There was an additional 25% of active participants who recalled the critical messages at three months when a relaxation break was performed before debriefing of scenarios. Benefits of relaxation to enhance learning should be considered for medical education.

In humans, memories for events happening early in life are forgotten more rapidly than those for events later in life. This form of accelerated forgetting in infancy is also observed in non-human species, and has been most extensively characterized in rats. Here we expand the characterization of infantile forgetting to mice, a species where a broader range of genetic tools can be used to understand the neurobiological mechanisms underlying this form of forgetting. Using a hidden platform version of the water maze task, we first assessed retention in mice that ranged in age from 15 to 150days-old at the beginning of training. All groups exhibited spatial memory when tested one day after training. However, only mice that were 20days or older at the time of training could remember one month later. Second, forgetting in younger cohorts of mice was not due to weaker encoding, since when younger mice were over-trained, such that their performance exceeded that of adult mice, they still exhibited forgetting. Third, in young mice, presentation of a reminder one month following training led to memory recovery, indicating that forgetting was due to a retrieval, rather than storage, deficit. Fourth, younger mice exhibited superior reversal learning compared to older mice, raising the possibility that a by-product of infantile forgetting might be greater flexibility.

The role of memory retention and attentional control on hemispheric asymmetry was investigated using a verbal dichotic listening paradigm, with the consonant–vowel syllables (/ba/,/da/,/ga/,/ka/,/pa/and/ta/), while manipulating the focus of attention and the time interval between stimulus and response. Attention was manipulated using three conditions: non-forced (NF), forced left (FL) and forced right (FR) attention. Memory involvement was varied using four delays (0, 1, 3 and 4 s) between stimulus presentation and response. Results showed a significant right ear advantage (REA) in the NF condition and an increased REA in the FR condition. A left ear advantage (LEA) was found in FL condition. The REA increased significantly in the NF attention condition at the 3-s compared to the 0-s delay and in the FR condition at the 1-s compared to the 0-s delay. No modulation of the left ear advantage was observed in the FL condition. These results are discussed in terms of an interaction between attentional processes and memory retention.

Alzheimer's disease is defined by abnormal levels of amyloid and tau biomarkers. Even cognitively normal older adults with clinically relevant amyloid and tau levels perform worse on memory tests. However, it is unclear if the relationship between biomarker level and memory extends below clinical thresholds. We hypothesized that even subclinical biomarker levels are associated with memory when measured with neuropsychological tests designed to detect dysfunction in preclinical disease states. In a group of cognitively normal, “biomarker-negative” older men and women, we investigated the relationship between cerebrospinal fluid biomarker levels and memory measured with the ModRey, a list-learning task designed to assess memory in preclinical and cognitively normal adults. Cerebrospinal amyloid levels were associated with ModRey memory retention, the proportion of information retained after a delay period. When older adults with mild impairment were included, cerebrospinal fluid tau levels were also associated with ModRey retention. The association of amyloid and tau levels with memory was independent of each other. These results suggest cognitive changes associated with Alzheimer's disease pathology might occur earlier than currently thought.

Introduction: Inhaler drugs are the basis of the treatment in asthma due to their local effects. Errors in inhaler techniques in asthma increase health care costs, hospitalization, and result in a lack of disease control. A brochure and a puzzle material were prepared as alternatives to face-to-face training in this study. It is aimed to evaluate the adequacy of these training materials in the diagnosis and the first month of treatment. Materials and Methods: Patients aged between 4-12 years diagnosed with asthma and their family members aged between 20-49 years who had not received inhaler drug training before were included in the study. A prospective, cross-sectional, quasi-experimental study was planned. Results: A total of 109 family members and patients were included. There was no difference between the groups in inhaler technique total scores in diagnosis. There was no difference in inhaler technique full score rates between the groups in the diagnosis and the first month. Conclusion: It was determined that puzzle games and brochures can be used as an alternative for treatment applications such as inhaler drugs with spacers. Further studies are needed to evaluate the effect of puzzle games and brochures with face-to-face training on long-term memory retention.

Summary Both sleep and future relevance influence memory consolidation; however, limited research has investigated their role in memory reconsolidation. We manipulated the future relevance of both stable and labile memories in need of reconsolidation. Two groups learned two blocks of syllable pairs on one evening and were told they would be tested on one of the blocks later. On the second evening, one group (Labile) received reminders designed to return their memories of syllable pairs to a labile state, while a second group (Stable) received reminders designed to leave these memories in a stable state. No significant differences in memory retention were found between blocks or groups the following morning. Frontal delta (0.5–4 Hz) electroencephalographic power during Stage 2 sleep correlated positively with retention of future-relevant material exclusively in the Labile group. Central theta (4–8 Hz) electroencephalographic power during rapid eye movement (REM) sleep correlated positively with the extent to which the Labile group selectively retained future-relevant material. These relationships suggest that sleep-dependent processes are involved in an economical reprocessing of memories beyond the initial stages of consolidation.

Wir stellen einen Patienten vor, der unter einer selektiven pathologischen Beeintrachtigung des verbalen Arbeitsgedachtnisses leidet („Phonological Short-Term Memory Syndrome“). Um die Lern- und Merkfahigkeit des Patienten zu uberprufen, haben wir mit ihm den Verbalen Lern- und Merkfahigkeitstest (VLMT) durchgefuhrt Unsere Hypothese war, dass der Patient bei den Anteilen der Aufgabe, die die unmittelbare Merkfahigkeit testen (das verbale Arbeitsgedachtnis), schlechter abschneidet als eine angepasste Gruppe von Kontrollpersonen. Bei den Lernaufgaben (die das Langzeitgedachtnis betreffen) sollte er hingegen Leistungen zeigen, die im Bereich des Abschneidens der Kontrollpersonen liegen. Die Hypothese konnte bestatigt werden. Unsere Studienergebnisse interpretieren wir als Beleg fur den Nutzen des Arbeitsgedachtnismodelle von A. Baddeley mit seiner funktionalen Aufteilung von Arbeits- und Langzeitgedachtnis fur die neuropsychologische Klinik und Forschung.

Poor nutrition, poor mobility, obesity and ageing increase the possibility of neuronal disorder and Alzheimer’s disease. This study examined effect of Cornus mas flavonoids on memory retention and the level of plasma glucose and lipids in an intra-cerebroventricular streptozotocin-induced experimental Alzheimer’s disease model in Wistar rats. Intracerebroventricular administration of streptozotocine (3 mg kg–1) was performed and the animals memory were evaluated through passive avoidance tasks. Five groups (saline-saline control, streptozotocine-saline, and streptozotocine with different flavonoid doses [5, 10, 20 mg kg –1 ]) were examined. Animals received different doses of C. mas flavonoids or saline for three weeks starting one day before surgery. Rat weight was measured at the beginning of each week. Injection of streptozotocine significantly reduced memory retention, compared to the control group. Flavonoid treatment increased memory retention in a dose-dependent manner, and triglyceride and glucose concentration decreased memory retention. At the same time, high-density lipoprotein concentration increased without any effect on low-density lipoprotein concentration. The dose of 10 mg kg –1 decreased rat weight significantly. Our findings show that C. mas flavonoids can optimize cognitive deficits caused by injections of streptozotocine and also have a few positive effects on reducing the risk factors in the serum.

Studies indicate that a brief period of wakeful rest after learning supports memory retention, whereas distraction weakens it. It is open for investigation whether advanced age has a significant effect on the impact of post-learning wakeful rest on memory retention for verbal information when compared to a cognitively demanding distraction task. In this study, we examined (1) whether post-learning rest promotes verbal memory retention in younger and older adults and (2) whether the magnitude of the rest benefit changes with increasing age. Younger adults and older adults learned and immediately recalled two consecutive word lists. After one word list, participants rested wakefully for 8 min; after the other list, they solved matrices. Memory performance was again tested in a surprise free recall test at the end of the experimental session. We found that, overall, younger adults outperformed older adults. Also, memory retention was higher following a wakeful rest phase compared to distraction. A detailed analysis revealed that this wakeful rest benefit was significant for the older adults group, whereas the younger adults group retained a similar amount of information in both post-encoding conditions. We assume that older adults can profit more from a wakeful rest phase after learning and are more prone to distraction than younger adults. With increasing age, a short break immediately after information uptake may help better retain the previously learned information, while distraction after learning tends to weaken memory retention.

Background and Objectives:: Spexin is a neuropeptide involved in learning and memory. The aim of the present study was to investigate the effect of intrahippocampal CA3 injection of spexin on passive avoidance learning and memory in normal and castrated rats. Methods: A total of 42 adult male rats were divided into 6 groups. Sham 1 (received 0.5 µL ACSF); sham 2 (sham castrated rats that received 0.5 µL ACSF); experimental 1 and 2 (healthy rats that received 0.5 µL spexin 10 or 30 nmol/rat); experimental 3 and 4 (castrated rats that received 0.5 µL spexin 10 or 30 nmol/rat). In all groups, injection was done in the CA3 region of hippocampus. CA3 region of hippocampus was cannulated unilaterally by the stereotaxic procedure. Results: Present data showed that castrated significantly (P < 0.05) increased lighting time in comparison to sham 2 group in memory sessions. In normal rats intrahippocampal CA3 injection of spexin in 2 doses significantly (P < 0.05) increased lighting time in comparison to the sham group in learning and memory sessions. In castrated rats, intrahippocampal CA3 injection of spexin 30 nmole/rat significantly (P < 0.05) induced even further increase in lighting time as compared to the sham group in memory sessions. In memory consolidation and memory retention sessions lighting time was significantly (P < 0.05) higher in castrated rats than normal animals. Conclusions: According to the present results, castration and spexin at 2 doses in normal rats improve learning and memory; while in castrated rats spexin, in 30 nM/rat, produced more improvement in memory. Memory improvement in castrated rats was higher than that in normal rats.

We examined the enduring effects of predictable versus unpredictable fear conditioning early in life on memory and relearning in adulthood. At postnatal Day 17 or 25 (P17 or P25), rats either remained naive, or were fear conditioned using paired (predictable) or unpaired (unpredictable) presentations of white noise and foot shocks. At 2 months of age (adulthood), each group was fear conditioned (or reconditioned) with either paired or unpaired training, and then was tested for fear extinction the next day. Initial findings replicate previous work from our lab and others, demonstrating a difference in adult memory retention based on age of acquisition. Specifically, rats that received paired conditioning at P25, but not P17, show increased freezing to the cue when tested in adulthood. We further show that paired as well as unpaired conditioning at P17 potentiates paired conditioning in adulthood; however, paired, but not unpaired, conditioning at P25 potentiates paired and unpaired conditioning in adulthood. In the present study, we examined how the function relating continued retrieval practice (e.g., one, three, or five tests) and long-term memory retention is modulated by desirable difficulty (R. A. Bjork, 1994). Of particular interest was how retrieval difficulty differed across young and older adults and across manipulations of lag (Exp. 1) and spacing (Exp. 2). To extend on previous studies, the acquisition phase response latency was used as a proxy for retrieval difficulty, and our analysis of final-test performance was conditionalized on acquisition phase retrieval success, to more directly examine the influence of desirable difficulty on retention. The results from Experiment 1 revealed that continued testing in the short-lag condition led to consistent increases in retention, whereas continued testing in the long-lag condition led to increasingly smaller benefits in retention for both age groups. The results from Experiment 2 revealed that repeated spaced testing enhanced retention relative to taking one spaced test, for both age groups; however, repeated massed testing only enhanced retention over taking one test for young adults. Across both experiments, the response latency results were overall consistent with an influence of desirable difficulty on retention. The discussion focuses on the role of desirable difficulty during encoding in producing the benefits of lag, spacing, and testing.

Major Depression is a prevalent mental disorder that is characterized by negative mood and reduced motivation, and frequently results in social withdrawal and memory-related deficits. Repeated stressors, such as adverse life events, increase the risk for development of the disorder. Consequently, individual variability in stress response greatly weighs on depression-vulnerability and -resilience. Here, we employed the social defeat-induced persistent stress (SDPS) paradigm to identify depression-prone individuals and to examine the temporal development of depression in the months following exposure to brief defeat stress. Male Wistar rats were socially defeated (5 defeat episodes) and single-housed for a prolonged period of time (~24 weeks). We assessed the emergence of a sustained depressive-like state by repeatedly evaluating social motivation (social approach avoidance) and spatial memory (object place recognition) in SDPS rats during the isolation period. Individual variability in the effects of SDPS yielded two extreme subpopulations: an SDPS-prone group that showed gradual affective and cognitive deterioration in terms of social approach and memory retention, and a SDPS-resilient group that did not develop this phenotype. Notably, in SDPS-prone individuals, the affective deficits preceded later cognitive impairments, providing a novel temporal profile of the development of pathology in this preclinical model of sustained depression.

Background: This study provides further evidence for improvement of memory by oral consumption of nutmeg. The present study was undertaken with an objective to study the effects of oral administration of nutmeg on memory boosting and regaining. Methods: Here we investigate the influence of oral intake of nutmeg on behavioral task performance by using T-maze and radial arm maze and physiological measures relative to a milk control group. Results: We have observed significant memory boosting and memory regaining effects of nutmeg when administered orally. This effect may be due to facilitation of acetylcholine activity by decreasing acetylcholinesterase activity of nutmeg. Hence we recommend further research in this area by investigating compound metabolism to optimize quantification of memory performance following nutmeg consumption.

This study investigated the prenatal marginal vitamin A deficiency (mVAD)-related impairment in learning and memory and the interactions between RARα, Src and NR1. Learning and memory were assessed in adult rats that were exposed to prenatal mVAD with Morris water maze. The average escape time was longer in mVAD rats, and they passed the hidden platform fewer times during the memory retention test than normal vitamin A intake (VAN) rats. The mRNA and protein levels of RARα, Src and NR1 in mVAD rats were significantly lower than those in VAN rats. RARα and Src, but not NR1, were in the same protein complex. RA treatment-induced increase in RARα, Src and NR1 expressions in mVAD neurons was much lower than that in VAN neurons. Overexpression of RARα gene in VAN neurons induced an increase in RARα, Src and NR1 expressions, while silencing of RARα gene induced a decrease in expressions of RARα and Src, but not that of of NR1. In mVAD neurons, however, overexpression of RARα did not induce an increase in NR1 expression, while silencing of RARα gene had no effect on Src and NR1 expressions. Furthermore, inhibition of Src was associated with a decrease in NR1 expression but not that of RARα. Prenatal mVAD was associated with impaired learning and memory in adult rats. It is possible that mVAD-related decrease in RARα led to a decrease in Src expression, which in turn down-regulated NR1 expression and Ca2+ influx and eventually caused learning and memory deficits.

Acoustic stimulation methods applied during sleep in young adults can increase slow wave activity (SWA) and improve sleep-dependent memory retention. It is unknown whether this approach enhances SWA and memory in older adults, who generally have reduced SWA compared to younger adults. Additionally, older adults are at risk for age-related cognitive impairment and therefore may benefit from non-invasive interventions. The aim of this study was to determine if acoustic stimulation can increase SWA and improve declarative memory in healthy older adults. Thirteen participants 60–84 years old completed one night of acoustic stimulation and one night of sham stimulation in random order. During sleep, a real-time algorithm using an adaptive phase-locked loop modeled the phase of endogenous slow waves in midline frontopolar electroencephalographic recordings. Pulses of pink noise were delivered when the upstate of the slow wave was predicted. Each interval of five pulses (“ON interval”) was followed by a pause of approximately equal length (“OFF interval”). SWA during the entire sleep period was similar between stimulation and sham conditions, whereas SWA and spindle activity were increased during ON intervals compared to matched periods during the sham night. The increases in SWA and spindle activity were sustained across almost the entire five-pulse ON interval compared to matched sham periods. Verbal paired-associate memory was tested before and after sleep. Overnight improvement in word recall was significantly greater with acoustic stimulation compared to sham and was correlated with changes in SWA between ON and OFF intervals. Using the phase-locked-loop method to precisely target acoustic stimulation to the upstate of sleep slow oscillations, we were able to enhance SWA and improve sleep-dependent memory storage in older adults, which strengthens the theoretical link between sleep and age-related memory integrity.

Background. Alzheimer’s disease (AD) is a progressive degenerative disease which causes memory disorders, decreases cognitive functions and abilities, and results in behavioural changes. Some studies have indicated that the flavonoids are able to cross the blood–brain barrier and have a positive effect on the reduction of neuronal damage disorders in the brain such as Alzheimer’s disease. Materials and Methods. ICV administration of streptozotocin (3 mg/kg) was done on the first and the third day of the surgery and the animals’ memory was evaluated through passive avoidance tasks. Animals were divided into five groups: Salin-Salin, STZ-Salin, and STZ- different kaempferol doses (5, 7/5, 10 mg/kg). All animals received different doses of kaempferol or saline for 3 weeks starting one day before the surgery. Later, they were put into a learning test. After the memory test, the animals were killed and their brains were fixed with Paraformaldehyde 4%, and tissue processing was done. Finally, density of intact neurons in the CA1 area of the hippocampus in the brains of all groups was investigated. Results. The ICV injections of STZ significantly reduced memory retention and intact pyramidal cells compared to the control group. The kaempferol improved the effects of STZ. Conclusion. Our findings show that kaempferol can optimize cognitive deficits caused by injections of STZ and also has some useful impacts on hippocampal CA1 pyramidal neurons.

Rotation of a visual image in mind is associated with a slow posterior negative deflection of the event-related potential (ERP), termed rotation-related negativity (RRN). Retention of a visual image in short-term memory is also associated with a slow posterior negative ERP, termed negative slow wave (NSW). We tested whether short-term memory retention, indexed by the NSW, contributes to the RRN. ERPs were recorded in the same subjects in two tasks, a mental rotation task, eliciting the RRN, and a visual short-term memory task, eliciting the NSW. Over both right and left parietal scalp, no association was found between the NSW and the RRN amplitudes. Furthermore, adjusting for the effect of the NSW had no influence on a significant association between the RRN amplitude and response time, an index of mental rotation performance. Our data indicate that the RRN reflects manipulation of a visual image but not its retention in short-term memory.

Spaced training produces gains in performance associated with memory consolidation, which develops between sessions (offline gain). Learning motor skills that require many repetitions may generate a delay in memory formation and in offline gain. We tested the presence of this delay by studying a precision throwing task. Sixteen participants performed 1020 underarm precision throws distributed over four sessions. Eight participants spaced the training by 40-min between-session intervals, while the remaining subjects distributed the practice with 1-day intervals. Memory retention was tested 15 days after training. Differences in accuracy over groups, sessions, directions of throwing movements and blocks of throws were evaluated by analysis of variance. The 40-min group had better performance than the 1-day group after the first two sessions. As the level of skill stabilized, the 1-day group exhibited offline gains, with significant performance improvements during the fourth and retention session. Both medial-lateral and antero-posterior movement directions of throwing contributed to the performance. Initial decrements in performance appeared within sessions for both groups. Overall, when learning a precision throwing task, benefits from spaced training is delayed and occurs as the skill stabilizes. These findings may help to optimize training distribution schedules, particularly for precision motor skills requiring extensive practice.

Objectives: Because of the well-known neurochemical interactions between the septum and hippocampus during memory processes, we investigated the effect of amyloid-beta (A-beta) injection into the medial septum (MS) on the behavior in Wistar rats. We also assessed whether the observed effects were functional or due to apoptosis.Methods: Specific A-beta oligomers (beta1-40:beta1-42 = 10:1) were injected into MS for seven consecutive days. Behavior was assessed with the Morris water maze task.Results: Compared with the control group, rats that received A-beta oligomers exhibited significant memory retention impairment (P < 0·05) without apoptosis in the cornu ammonis (CA)1 and CA3 regions of the dorsal hippocampus.Discussion: These data indicate that septal injection of A-beta impairs memory retention, even in the absence of hippocampal apoptosis. This result might bring new insight to spatial memory-related disorders like Alzheimer’s disease (AD).

Aged dogs spontaneously develop progressive decline in both cognitive and behavioral function, in addition to neuropathological changes, that collectively parallel several aspects of human aging and Alzheimer's disease progression and likely contribute to the development of canine cognitive dysfunction syndrome. In the current study, ethologically relevant spatial learning, retention, and reversal learning tasks were conducted, with the goal of expanding canine neuropsychological testing to pet dogs. Initially, dogs (N = 44, aged 7.8 ± 2.8 years, mean ± SD) had to learn which of two alternative routes successfully led out of a T-maze. Two weeks later, long-term memory retention was assessed, immediately followed by a reversal learning task in which the previously correct route out of the maze was reversed compared with the initial learning and memory retention tasks. No effects of age were evident on the learning or retention tasks. However, older (≥ 8 years) dogs were significantly impaired on the reversal learning task compared with younger ones (8 years). Moreover, trial response latency was significantly increased in aged dogs across both the initial and reversal learning tasks but not on the retention task, which suggests that processing speed was impaired by increasing age during the acquisition of novel spatial information but not during performance of previously learned responses. Overall, the current study provides a framework for assessing cognitive function in pet dogs, which should improve understanding of the effects of aging on cognition in the dog population.

Many studies have shown that memory is enhanced when study sessions are spaced apart rather than massed. This spacing effect has been shown to have a lasting benefit to long-term memory when the study phase session follows the encoding session by 24 hours. Using a spacing paradigm we examined the impact of sleep and spacing gaps on long-term declarative memory for Swahili-English word pairs by including four spacing delay gaps (massed, 12 hours same-day, 12 hours overnight, and 24 hours). Results showed that a 12-hour spacing gap that includes sleep promotes long-term memory retention similar to the 24-hour gap. The findings support the importance of sleep to the long-term benefit of the spacing effect.

Adolescence is a critical developmental stage during which substantial remodeling occurs in brain areas involved in emotional and learning processes. Although a robust literature on the biological effects of extremely low frequency magnetic fields (ELF-MFs) has been documented, data on the effects of ELF-MF exposure during this period on cognitive functions remain scarce. In this study, early adolescent male mice were exposed from postnatal day (P) 23-35 to a 50 Hz MF at 2 mT for 60 min/day. On P36-45, the potential effects of the MF exposure on spatial memory performance were examined using the Y-maze and Morris water maze tasks. The results showed that the MF exposure did not affect Y-maze performance but improved spatial learning acquisition and memory retention in the water maze task under the present experimental conditions.

Apolipoprotein E, which plays an important role in lipid transport and metabolism and neuronal repair, might modulate the CNS risk following 56Fe irradiation exposure during space missions. In this study, we investigated this risk by behavioral and cognitive testing male E2, E3, and E4 mice 3 months following cranial 56Fe irradiation. In the open field, mice irradiated with 2 Gy showed higher activity levels than sham-irradiated mice or mice irradiated with 1 Gy. In addition, E2 mice showed higher activity and lower measures of anxiety than E3 and E4 mice in the open field and elevated zero maze. During hidden platform training, sham-irradiated mice showed most robust learning, 1 Gy irradiated mice reduced learning, and 2 Gy irradiated mice no improvement over the four sessions. In the water maze probe trials, sham-irradiated E2, E3, and E4 mice and E2 and E4 mice irradiated with 1 Gy showed spatial memory retention, but E3 mice irradiated with 1 Gy, and E2, E3, and E4 mice irradiated with 2 Gy did not. Thus, cranial 56Fe irradiation increases activity levels in the open field and impairs spatial learning and memory in the water maze. E3 mice are more susceptible than E2 or E4 mice to impairments in spatial memory retention in the water maze, indicating that apoE isoform modulates the CNS risk following space missions.

The formation of long-term memories for food sources is essential for the survival of most animals. Long-term memory formation in mammalian species has been demonstrated through a variety of conditioning tasks, however, the nature of long-term memory in fish is less known. In the current study, we explored whether African cichlids (Labidochromis caeruleus) could form memories for food-reinforced stimuli that last for 12 days. During the training sessions, fish were reinforced for approaching an upward drifting line grating. After a rest period of 12 days, fish demonstrated a significant preference for the upward drifting grating. To determine whether this preference could also be reversed, fish were then reinforced for approaching a downward drifting line grating after a 20-day rest period. When tested 12 days later, there were no significant differences in preference for either stimulus; however, following a second training period for the downward stimulus, there was a significant preference for the downward drifting grating. This suggests that cichlids are able to form reversible discrimination-based memories for food-reinforced stimuli that remain consolidated for at least 12 days.

Introduction: The effects of electromagnetic fields on biological organisms have been a controversial and also interesting debate over the past few decades, despite the wide range of investigations, many aspects of extremely low frequency electromagnetic fields (ELF/EMFs) effects including mechanism of their interaction with live organisms and also their possible biological applications still remain ambiguous. In the present study, we investigated whether the exposures of ELF/EMF with frequencies of 3 Hz and 60 Hz can affect the memory, anxiety like behaviors, electrophysiological properties and brain’s proteome in rats. Methods: Male rats were exposed to 3 Hz and 60 Hz ELF/EMFs in a protocol consisting of 2 cycles of 2 h/day exposure for 4 days separated with a 2-day interval. Short term memory and anxiety like behaviors were assessed immediately, 1 and 2 weeks after the exposures. Effects of short term exposure were also assessed using electrophysiological approach immediately after 2 hours exposure. Results: Behavioral test revealed that immediately after the end of exposures, locomotor activity of both 3 Hz and 60 Hz exposed groups significantly decreased compared to sham group. This exposure protocol had no effect on anxiety like behavior during the 2 weeks after the treatment and also on short term memory. A significant reduction in firing rate of locus coeruleus (LC) was found after 2 hours of both 3 Hz and 60 Hz exposures. Proteome analysis also revealed global changes in whole brain proteome after treatment.Conclusion: Here, some evidence regarding the fact that such exposures can alter locomotor activity and neurons firing rate in male rats were presented.

Twenty-five years after the FIGO Workshop that produced the "Guidelines for the use of fetal monitoring," these remain the only broad international consensus effort in this field. Documents of a similar nature have been produced by national institutions, with subsequent updates of the initial concepts. The 3-class classification system has now been adopted by all the major guidelines, and while there are numerous similar features, and indeed many ideas were inspired by the FIGO guidelines, many key aspects still lack consensus. Making guidelines simpler and more objective may be an important step to guarantee a wide application and assimilation of the recommendations, as well as an enhanced reproducibility and increased memory retention.

Background and Objectives: Current programs in medical education technology concentrate mainly upon “how-to-teach.” The focus is needed on learner's memory retention too. An innovative strategy like concept mapping might be a way forward. The study was carried out to assess its effectiveness and to know students' perceptions. Materials and Methods: During community medicine classes, a student-group was sensitized on how to make and use concept maps out of taught contents. At the end of epidemiology exercises sessions, this group was given additional minutes to prepare concept maps, interact, and brainstorm followed by quick QA session. Others were taught same contents in conventional way. Performances of both groups were assessed in one immediate (term-ending) and one distant (preliminary) exam. Feedback was also taken from study group. Results: Study group consistently scored higher in both exams. Difference in scored mean marks was highly significant in term-ending-examination (P < 0.0001, t = 5.754, df = 121.9). Largely positive feedback was received on utility of concept maps in memorizing, confidence-boosting, and understanding subject. Felt need for innovations in conventional teaching-learning (T-L) was palpable. Conclusion: Time has come to start focusing on enhancing students' learning. Performance-enhancing utility of concept maps is proven and should be integrated in regular T-L.

Mice deficient in apoE (Apoe-/-) can be used to assess the potential role of apoE in the effects of cranial irradiation on hippocampal function. Radiation-induced impairments in hippocampal function may be more pronounced in female Apoe-/- mice and more pronounced in mice irradiated and tested cognitively later in life. To assess this possibility, female wild-type and Apoe-/- mice were irradiated at 6 months of age with 10 Gy 137Cs gamma rays and tested cognitively 3 months later. Sham-irradiated wild-type female mice showed enhanced hippocampal-dependent novel location recognition compared to sham-irradiated Apoe-/- female mice. However, cranial irradiation impaired novel location recognition similarly in both genotypes. Cranial irradiation also impaired hippocampal-dependent spatial memory retention similarly in wild-type and Apoe-/- female mice in the water maze. Because novel location recognition was not affected after 137Cs gamma irradiation in younger mice, these data support the possibility that older mice are more susceptible to the effects of gamma radiation on novel location recognition. Together with the impairments in spatial memory retention in the water maze after irradiation, these data support the existence of detrimental effects of cranial irradiation on hippocampal function. In addition, compared to wild-type female mice, Apoe-/- female mice showed enhanced levels of anxiety, and in Apoe-/-, but not in wild-type, female mice, radiation decreased levels of anxiety. Because levels of anxiety during the hidden session of the water maze were associated with ability to locate the hidden platform, assessments of anxiety need to be considered in evaluating the effects of cranial irradiation on cognitive performance after cranial irradiation.

The behavioral phenotypes of out-bred ICR mice were compared with those of in-bred C57BL/6 and BALB/c mice. In particular, this study examined the locomotor activity and two forms of hippocampus-dependent learning paradigms, passive avoidance and object recognition memory. The basal open-field activity of the ICR strain was greater than that of the C57BL/6 and BALB/c strains. In the passive avoidance task, all the mice showed a significant increase in the cross-over latency when tested 24 hours after training. The strength of memory retention in the ICR mice was relatively weak and measurable, as indicated by the shorter cross-over latency than the C57BL/6 and BALB/c mice. In the object recognition memory test, all strains had a significant preference for the novel object during testing. The index for the preference of a novel object was lower for the ICR and BALB/c mice. Nevertheless, the variance and the standard deviation in these strains were comparable. Overall, these results confirm the strain differences on locomotor activity and hippocampus-dependent learning and memory in mice.

High sustained positive Gz (+Gz) exposures have been shown to have a pathophysiological impact on the brain in rats. However, the consequence for brain function such as learning and memory remains elusive. In the present study, we investigated locomotor activity, learning ability and memory in rats over 6 days (d) following +6 Gz/3 min and +10 Gz/3 min exposures. In an open field test, rats treated with +10 Gz showed a significant increase in the time spent in the center square immediately and 2d after exposure compared with control rats. In contrast, the number of grid crosses and number of rears of rats decreased significantly immediately after +6 Gz and +10 Gz exposures. In addition, a Y-maze test revealed that +Gz exposures reduced the number of correct responses and increased total reaction time in rats, and the number of correct responses was negatively correlated with the total reaction time in all groups throughout the period of study. Moreover, the passive avoidance test exhibited that the latency increased significantly on 0 d and decreased significantly on 6 d after +Gz exposures compared with control. Thus, +6 Gz/3 min and +10 Gz/3 min exposures may transiently decrease locomotor activity, impair learning, and induce a deficit in memory retention in rats.

Infants show robust ability to track transitional probabilities within language and can use this information to extract words from continuous speech. The degree to which infants remember these words across a delay is unknown. Given well-established benefits of sleep on long-term memory retention in adults, we examine whether sleep similarly facilitates memory in 6.5month olds. Infants listened to an artificial language for 7minutes, followed by a period of sleep or wakefulness. After a time-matched delay for sleep and wakefulness dyads, we measured retention using the head-turn-preference procedure. Infants who slept retained memory for the extracted words that was prone to interference during the test. Infants who remained awake showed no retention. Within the nap group, retention correlated with three electrophysiological measures (1) absolute theta across the brain, (2) absolute alpha across the brain, and (3) greater fronto-central slow wave activity (SWA).

Spontaneous movement tempo (SMT) was a popular field of study of the Gestalt psychologists It can be determined from subjects freely tapping out a rhythm with their finger, and it has been found to average about 2 Hz. A previous study showed that SMT changed after the observation of rhythmical movements performed at frequency different from the SMT. This effect was long-lasting only when movement execution immediately followed action observation (AO). We recently demonstrated that only when AO was combined with peripheral nerve stimulation (AO-PNS) was it possible to induce plastic changes in the excitability of the motor cortex, whereas AO and PNS alone did not evoke any changes. Here we investigated whether the observation of rhythmical actions at a frequency higher than the SMT combined with PNS induced lasting changes in SMT even in absence of immediate movement execution. Forty-eight participants were assigned to four groups. In AO-PNS group they observed a video showing a right hand performing a finger opposition movement sequence at 3 Hz and contemporarily received an electrical stimulation at the median nerve; in AO group and PNS group participants either observed the same video or received the same electrical stimulation of the AO-PNS group, respectively; in LANDSCAPE group subjects observed a neutral video. Participants performed a finger opposition movement sequence at spontaneous movement rate before and 30 min after the conditioning protocols. Results showed that SMT significantly changed only after AO-PNS. This result suggested that the AO-PNS protocol was able to induce lasting changes in SMT due to neuroplasticity mechanisms, indicating possible application of AO-PNS in rehabilitative treatments.
